Electrochemiluminescence “turn-off” detection of curcumin via energy transfer using luminol-doped silica nanoparticles

Zhao Maoyu,Qi Wenjing,Fu Yuling,He Hongkun,Wu Di,Qi Lin,Li Rong

A method is presented for electrochemiluminescent (ECL) detection of the food additive curcumin via an energy transfer strategy and by using luminol-doped silica nanoparticles (luminol-NPs). The ECL emission of the luminol-NPs (peaking at 425 nm) is reduced in the presence of curcumin due to spectral overlap. The assay can be performed within 1 min, response is linear in the 0.1 to 100 µM curcumin concentration range, and the limit of detection is 32 nM. The method is selective over many ions, adenosine triphosphate, ascorbic acid, cysteine and folic acid. It was successfully applied to the determination of curcumin in spiked human serum and urine. The average recoveries range from 99.0 to 102.6%. Electrochemiluminescence (ECL) “turn-off” detection of curcumin at levels as low as 32 nM via energy transfer using luminol-doped silica nanoparticles. No hydrogen peroxide (H 2 O 2 ) is used in ECL detection which makes the luminol-NPs ECL system more stable than the conventional luminol-H 2 O 2 ECL system.

Reinforcement of Polycaprolactone/Chitosan with Nanoclay and Controlled Release of Curcumin for Wound Dressing

Yanping Huang,Nianhua Dan,Weihua Dan,Weifeng Zhao

A novel clay-reinforced polycaprolactone/chitosan/curcumin (PCl/CS/Clay/Cur) composite film was fabricated in this study. The prepared Cur-loading composite films were characterized with attenuated total reflection Fourier transformed infrared spectroscopy, scanning electron microscopy, atomic force microscopy, water contact angle, differential scanning calorimetry, thermogravimetric analysis, and X-ray diffraction, and the results showed good dispersion of clay in the composite films. The addition of nanoclay was found to significantly increase the tensile strength. Also, the clay-enhanced drug-loading films exhibited better controlled-release profiles of Cur than those membranes without clay. Skin disinfection test demonstrated that the curcumin-loaded film could protect wound from bacterial infection. Cytotoxicity analysis proved the good biocompatibility of the composite films. The clay-enhanced Cur-loading films might be promising candidates for wound care.

Oral Curcumin via Hydrophobic Porous Silicon Carrier: Preparation, Characterization, and Toxicological Evaluation In Vivo

Yue Zhang,Wei Li,Di Liu,Yafang Ge,Mengyuan Zhao,Xuerui Zhu,Weiwei Li,Longfeng Wang,Tiesong Zheng,Jianlin Li

Curcumin has antioxidant, anti-inflammatory, antimicrobial, and anticarcinogenic activities. However, the clinical application of curcumin has been restricted by the poor water solubility and low bioavailability of this molecule. In this work, hydrophobic porous silicon (pSi) particles were prepared by electrochemical etching method and grafted with the different hydrophobic groups on their surfaces. The loading efficiency of curcumin in pSi has been investigated. The properties of pSi particles have been characterized by scanning electron microscopy (SEM) and Fourier transform-infrared spectroscopy (FTIR). The highest loading efficiency of curcumin can be obtained with pSi surface modified with the octadecyl silane group. The release properties of curcumin in hydrophobic pSi have been researched in vitro and in vivo. The curcumin in the hydrophobic pSi surface keeps a high antioxidant bioactivity. The toxicological evaluation of the hydrophobic pSi particles indicates they have a high in vivo biocompatibility within the observed dose ranges. The hydrophobic pSi particles could provide an effective and controlled release delivery carrier for curcumin, which may provide a new tool platform for the further development of curcumin.

Dose-reduction antiangiogenic curcumin-low molecular weight heparin nanodrugs for enhanced combinational antitumor therapy

Yan Xiao,Cheng Xu,Hui Xiong,Shi Du,Jianping Zhou,Lifang Yin,Jing Yao

Curcumin (CUR) is a natural diketone with diverse bioactivities of inhibiting angiogenesis and tumor growth. However, its clinical application for cancer treatment was severely hindered by poor aqueous solubility and chemical instability. To overcome these drawbacks and achieve enhanced antitumor efficiency, low molecular weight heparin (LMWH) was conjugated to CUR via the one-step esterification reaction to yield LMWH-CUR (LCU) nanodrugs with the size of 180  nm , which exhibited enhanced accumulation within tumor site by EPR effect and long circulating capacity by LMWH hydrophilic shell. The solubility of conjugated CUR was increased to 0.12 mg/mL (equivalent of CUR) in comparison with 0.006 mg/mL of free CUR. The bioactivities of CUR were guaranteed because of the improved stability of LCU nanodrugs in low pH condition. Moreover, the stronger anti-angiogenesis efficacy of LCU nanodrugs than LMWH monotherapy was also verified. Notably, at a rather low dose of equivalent LMWH (5 mg/kg) and CUR (0.3 mg/kg), the tumor inhibition rate of LCU nanodrugs were much higher than that of LMWH (10 times) and LMWH plus CUR mixture (3.8 times) respectively, indicating its excellent in vivo antitumor efficacy. Overall, our study managed to obtain the novel nanodrugs with potent anti-angiogenesis and antitumor effects whereas avoiding tedious and complicated synthetic procedures. These results also suggested that LCU nanodrugs could be considered as a promising targeted delivery system for cancer treatment.

Fabrication of ovalbumin/κ-carrageenan complex nanoparticles as a novel carrier for curcumin delivery

Hujun Xie,Chuyue Xiang,Yang Li,Lihong Wang,Yutong Zhang,Zhijun Song,Xiangjuan Ma,Xiaoxing Lu,Qunfang Lei,Wenjun Fang

Natural biopolymers are biocompatible and biodegradable, and have been a focus on the research of biologically active carrier materials. In this manuscript, the nanoparticles of ovalbumin/ κ -carrageenan (OVA-CRG) complex were fabricated to encapsulate and protect curcumin under moderate conditions, forming the ovalbumin/ κ -carrageenan/curcumin (OVA-CRG-CUR) complex nanoparticles. The OVA-CRG complex nanoparticles show good curcumin entrapping performance, and the addition of curcumin does not affect the emulsifying stability (ES) and emulsifying activity (EA) of OVA-CRG complex nanoparticles. The maximum ES of OVA-CRG-CUR is reached at 36 h with the value of 84.6 min, and the maximum EA is reached at 48 h with the value of 74.2 m 2 /g. The encapsulated curcumin demonstrates high antioxidation activity, while its thermal and light stabilities of curcumin are both increased. The release behavior of curcumin is further studied in simulated gastric and intestinal fluids. The lower release rate of curcumin in simulated gastric fluid with the final value of 12.9% after 3 h indicates that the OVA-CRG complex nanoparticles are good carriers for improving the stability of curcumin in acidic environment, and are able to enhance the bioavailability of curcumin by a gradual release in intestine. Present researches elucidate that proteins-polysaccharides complex nanoparticles are feasible to solubilize and protect sensitive amphiphilic bioactive compounds and have extensive potential in food and medicinal applications.

Self-Assembled Nanoparticles of Glycyrrhetic Acid-Modified Pullulan as a Novel Carrier of Curcumin

Roufen Yuan,Fuchun Zheng,Shuping Zhong,Xiaojun Tao,Yanmei Zhang,Fenfei Gao,Fen Yao,Jiaxiong Chen,Yicun Chen,Ganggang Shi

Glycyrrhetic acid (GA)-modified pullulan nanoparticles (GAP NPs) were synthesized as a novel carrier of curcumin (CUR) with a degree of substitution (DS) of GA moieties within the range of 1.2–6.2 groups per hundred glucose units. In the present study, we investigated the physicochemical characteristics, release behavior,in vitrocytotoxicity and cellular uptake of the particles. Self-assembled GAP NPs with spherical shapes could readily improve the water solubility and stability of CUR. The CUR release was sustained and pH-dependent. The cellular uptake of CUR-GAP NPs was confirmed by green fluorescence in the cells. An MTT study showed CUR-GAP NPs with higher cytotoxicity in HepG2 cells than free CUR, but GAP NPs had no significant cytotoxicity. GAP is thus an excellent carrier for the solubilization, stabilization, and controlled delivery of CUR.Keywords:pullulan;glycyrrhetic acid;self-aggregated nanoparticle;curcumin;drug delivery

Preparation of a colon-specific sustained-release capsule with curcumin-loaded SMEDDS alginate beads

Yuxia Zhang,Yan Bai,Huali Chen,Yuanrui Huang,Pei Yuan,Liangke Zhang

A colon-specific capsule with alginate beads containing a self-microemulsifying drug delivery system (SMEDDS) was developed and evaluated. The SMEDDS technique was used to improve the solubility of curcumin (Cur). After encapsulating the Cur-loaded SMEDDS, the alginate beads were placed inside an impermeable capsule body. A konjac glucomannan/lactose/hydroxypropyl methylcellulose (KG/Lac/HPMC) plug tablet was then prepared and placed in the mouth of the capsule. The capsule demonstrated a pulsatile drug-release profile with a specific lag time and subsequent sustained-release phase. The lag time was modified by changing the type of HPMC and the ratio of KG/Lac/HPMC. In addition, 0.5% β-mannase solution and 5% rat cecal solution were used to simulate the colon fluid, significantly decreasing the lag time of the capsule. The results show that the capsule has potential for use in colon-specific drug delivery and exhibits a sustained-release characteristic.

Preparation of curcumin-loaded PCL-PEG-PCL triblock copolymeric nanoparticles by a microchannel technology

Fangyuan Guo,Dingjia Guo,Wei Zhang,Qinying Yan,Yan Yang,Weiyong Hong,Gensheng Yang

Biodegradable polymeric nanoparticles (NPs) have potential therapeutic applications; however, preparing NPs of a specific diameter and uniform size distribution is a challenge. In this work, we fabricated a microchannel system for the preparation of curcumin (Cur)-loaded NPs by the interfacial precipitation method, which rapidly and consistently generated stable NPs with a relatively smaller diameter, narrow size distribution, and higher drug-loading capacity and entrapment efficiency. Poly(ε-caprolactone)-poly(ethylene glycol)-poly (ε-caprolactone) triblock copolymers(PCEC) used as the carrier material was synthesized and characterized. Cur-loaded PCEC NPs had an average size of 167.2   nm with a zeta potential of −   29.23   mV, and showed a loading capacity and drug entrapment efficiency of 15.28%   ±   0.23% and 96.11%   ±   0.13%, respectively. Meanwhile, the NPs demonstrated good biocompatibility and bioavailability, efficient cellular uptake, and long circulation time and a possible liver targeting effect in vivo. These results indicate that the Cur-loaded PCEC NPs can be used as drug carriers in controlled delivery systems and other biomedical applications.

Synthesis and Cytotoxic Evaluation of Monocarbonyl Analogs of Curcumin as Potential Anti-Tumor Agents

Zheer Pan,Chengwei Chen,Yeli Zhou,Feng Xu,Yaozeng Xu

Preclinical Research A series of mono-carbonyl curcumin analogs with different substituents at the 4/4’-position of the phenyl group were synthesized and screened for in vitro cytotoxicity against a panel of human cancer cell lines using a methyl thiazolyl tetrazolium assay. Several of the curcumin analogs, especially B114, exhibited a wide-spectrum of anti-tumor properties in all tested cell lines, indicating their potential in as anti-cancer lead compounds. Further toxicity testing in the NRK-52E kidney cell line revealed that the analogs A111, A113, and B114 had comparable or higher safety than curcumin. These data suggested that the introduction of appropriate substituents in the 4/4’-positions could be a promising approach for curcumin-based drug design. Enhancement of Anti-Inflammatory Activity of Curcumin Using Phosphatidylserine-Containing Nanoparticles in Cultured Macrophages

Ji Wang,Yu-Xia Kang,Wen Pan,Wan Lei,Bin Feng,Xiao-Juan Wang

Macrophages are one kind of innate immune cells, and produce a variety of inflammatory cytokines in response to various stimuli, such as oxidized low density lipoprotein found in the pathogenesis of atherosclerosis. In this study, the effect of phosphatidylserine on anti-inflammatory activity of curcumin-loaded nanostructured lipid carriers was investigated using macrophage cultures. Different amounts of phosphatidylserine were used in the preparation of curcumin nanoparticles, their physicochemical properties and biocompatibilities were then compared. Cellular uptake of the nanoparticles was investigated using a confocal laser scanning microscope and flow cytometry analysis in order to determine the optimal phosphatidylserine concentration. In vitroanti-inflammatory activities were evaluated in macrophages to test whether curcumin and phosphatidylserine have interactive effects on macrophage lipid uptake behavior and anti-inflammatory responses. Here, we showed that macrophage uptake of phosphatidylserine-containing nanostructured lipid carriers increased with increasing amount of phosphatidylserine in the range of 0%–8%, and decreased when the phosphatidylserine molar ratio reached over 12%. curcumin-loaded nanostructured lipid carriers significantly inhibited lipid accumulation and pro-inflammatory factor production in cultured macrophages, and evidently promoted release of anti-inflammatory cytokines, when compared with curcumin or phosphatidylserine alone. These results suggest that the delivery system using PS-based nanoparticles has great potential for efficient delivery of drugs such as curcumin, specifically targeting macrophages and modulation of their anti-inflammatory functions. Keywords:phosphatidylserine;curcumin;nanostructured lipid carriers;anti-inflammatory;macrophages

Self-assembled nanoparticles based on amphiphilic chitosan derivative and arginine for oral curcumin delivery

Mazhar Ali Raja,Shah Zeenat,Muhammad Arif,Chenguang Liu

Curcumin (Cur) is a striking anticancer agent, but its low aqueous solubility, poor absorption, hasty metabolism, and elimination limit its oral bioavailability and consequently hinder its development as a drug. To redress these limitations, amphiphilic chitosan (CS) conjugate with improved mucoadhesion and solubility over a wider pH range was developed by modification with hydrophobic acrylonitrile (AN) and hydrophilic arginine (Arg); the synthesized conjugate (AN–CS–Arg), which was well characterized by Fourier transform infrared and 1H nuclear magnetic resonance spectroscopy. Results of critical aggregation concentration revealed that the AN–CS–Arg conjugate had low critical aggregation concentration and was prone to form self-assembled nanoparticles (NPs) in aqueous medium. Cur-encapsulated AN–CS–Arg NPs (AN–CS–Arg/Cur NPs) were developed by a simple sonication method and characterized for the physicochemical parameters such as zeta potential, particle size, and drug encapsulation. The results showed that zeta potential of the prepared NPs was 40.1±2.81 mV and the average size was ~218 nm. A considerable improvement in the aqueous solubility of Cur was observed after encapsulation into AN–CS–Arg/Cur NPs. With the increase in Cur concentration, loading efficiency increased but encapsulation efficiency decreased. The in vitro release profile exhibited sustained release pattern from the AN–CS–Arg/Cur NPs in typical biological buffers. The ex vivo mucoadhesion study revealed that AN–CS–Arg/Cur NPs had greater mucoadhesion than the control CS NPs. Compared with free Cur solution, AN–CS–Arg/Cur NPs showed stronger dose-dependent cytotoxicity against HT-29 cells. In addition, it was observed that cell uptake of AN–CS–Arg/Cur NPs was much higher compared with free Cur. Furthermore, the in vivo pharmacokinetic results in rats demonstrated that the AN–CS–Arg/Cur NPs could remarkably improve the oral bioavailability of Cur. Therefore, the developed AN–CS–Arg/Cur NPs might be a promising nano-candidate for oral delivery of Cur.

Curcumin-Based Fluorescent and Colorimetric Probe for Detecting Cysteine in Living Cells and Zebrafish

Lanfang Pang,Yanmei Zhou,Wenli Gao,Junli Zhang,Haohan Song,Xiao Wang,Yong Wang,Xiaojun Peng

We synthesized a new curcumin-based fluorescent and colorimetric probe (CAC) and described its application for the specific detection of cysteine (Cys), including in solution, in living cells, and in a living vertebrate organism. The probe CAC itself showed a strong fluorescence emission, the conjugate addition–cyclization reaction with Cys resulted in the fluorescence quenching at 490 nm and the absorbance decreasing at 400 nm, accompanied by the appearance of a new absorption band at 427 nm. The probe CAC exhibited higher properties such as acting as a “naked eye” probe, readily synthesized, highly selective for Cys, and a lower detection limit of 0.19 μM. In addition, the probe CAC can be successfully applied to detect Cys in living cells and zebrafish with good cell and organism permeability, indicating its significant potential in living biological systems.
