Oral multilayer nanomedicine for gut barrier repair and immune–microbiota reprogramming in inflammatory bowel disease and inflammatory arthritis

Man Li,Yonghe Jiang,Sheng Lin,Yibo An,Xinyu Tan,Lijuan Yang,Yubo Tan,Jingwen Zheng,Wenxiang Zhang,Wenjie Ni,Yixin Chen,Yaohui He,Gang Liu,Zhixiang Lu

Inflammatory bowel disease (IBD) emerges from a self-reinforcing triad of epithelial barrier failure, oxidative stress, and immune dysregulation, while concomitants dysbiosis amplifies mucosal inflammation and— via the gut–joint axis—drives systemic autoimmunity such as rheumatoid arthritis (RA). Here, we report a triple-action multilayer oral nanoplatform, LBL-BM, that co-delivers berberine (BBR), myricetin (MYR), and TNF- α -targeting small interfering RNA (siTNF- α ). A chitosan/siTNF- α /alginate shell over a supramolecular BBR/MYR core overcomes gastrointestinal instability, poor lesion targeting, and fleeting siRNA exposure, enabling redox control and epithelial support coupled with pro-inflammatory cytokine silencing. This platform reduces reactive oxygen species (ROS), attenuates oxidative injury and apoptosis, and preserves tight-junction proteins. In dextran sulfate sodium (DSS)-induced colitis, it significantly improves clinical and histological outcomes, skews macrophage polarization toward an M2 phenotype, and reprograms the microbiota toward short-chain fatty acids (SCFAs)-associated, inflammation-attenuating communities. Consistent with modulation of the gut–joint axis, benefits extended beyond the intestine: in collagen antibody-induced arthritis (CAIA) it lowers clinical scores and paw swelling, attenuates synovitis and bone erosion, preserves cartilage, and reduces systemic cytokines (TNF- α , IL-17A, and IL-1 β ). Collectively, these findings establish LBL-BM as a multifunctional oral nanomedicine that restores intestinal homeostasis and holds translational promise for treating both IBD and RA.

Metabolomic profiling and bioactive potential of Tetragonula gressitti and Lepidotrigona flavibasis propolis

Jiao Wu,Xiang-Tan Xu,Bing-Jun Han,Yan He,Yan-Biao He,Shan Zhao,Shi-Jie Wang

The metabolism of two stingless bee propolis revealed a high flavonoids content. • Isorhamnetin was main compound in Lepidotrigona flavibasis propolis (LFP). • Eriodictyol was main metabolite in Tetragonula gressitti propolis (TGP). • LFP exhibited stronger antioxidant activity compared to TGP. • LFP demonstrated significant anti-inflammatory potential. Propolis composition and bioactivity reflect bee species and local flora, yet comparative data from stingless bees in China are scarce. We combined physicochemical profiling, UPLC-MS/MS metabolomics, and in vitro assays to compare Tetragonula gressitti propolis (TGP) and Lepidotrigona flavibasis propolis (LFP). LFP showed higher loss on drying and beeswax fraction but lower ash content compared with TGP. We detected 768 metabolites in TGP and 841 metabolites in LFP, with species-specific enrichment of flavonoid, phenylpropanoid, and isoflavonoid pathways. Quantification revealed elevated isorhamnetin in LFP (up to 4643.75 μg/g in 70% ethanol). 70% ethanolic extracts of LFP had higher total phenolics/flavonoids, stronger antioxidant activity (ABTS IC 50 68.76 μg/mL; FRAP 199.00 μmol TE/g), and greater suppression of LPS-induced inflammation in RAW 264.7 macrophages. Although these in vitro findings require in vivo validation, they reveal pronounced chemical and functional differences between the two propolis types and highlight LFP as a rich source of bioactive compounds with potential for functional food applications. Comparative Analysis of the In Vitro Fermentation Characteristics of Polysaccharides from Polygonatum cyrtonema Hua with Different Growth Years Using Human Fecal Microbiota

Rongguang Yang,Luning Zhao,Ying Zhu,Yansheng Zhao,Juan Bai,Xiang Xiao

Polygonatum cyrtonemaHua polysaccharides (PCPs) are bioactive components with antioxidant and immunomodulatory properties. However, whether the polysaccharides derived from elderPolygonatum cyrtonemaexhibit superior health benefits remains unclear. This study systematically compared the in vitro fermentation characteristics, gut microbiota-modulating effects, and short-chain fatty acid (SCFA) production of PCPs extracted from three-year-old (TPP), five-year-old (FPP), and eight-year-old (EPP) plants using a human fecal fermentation model. The fermentation dynamics were evaluated by monitoring pH, OD600, and the consumption of total and reducing sugars, while the structural degradation and compositional differences in PCPs were tracked via molecular weight distribution and monosaccharide composition analysis. The results showed that all three PCPs were degraded and utilized by gut microbiota, accompanied by decreased pH, increased OD600, and enhanced antioxidant activities. High-throughput 16S rDNA sequencing revealed that, at the phylum level, all PCPs increased the Firmicutes/Bacteroidetes ratio. At the genus level, they reduced the abundance of harmful bacteria such asSutterellaand increased beneficial bacteria includingBifidobacteriumandMegasphaera. Furthermore, gas chromatography (GC) analysis demonstrated that, compared with FPP, TPP and EPP significantly promoted the production of SCFAs. In summary, this study indicates that the in vitro fermentation characteristics and prebiotic properties of PCPs vary with growth years, and a comprehensive evaluation suggests that three-year-oldPolygonatum cyrtonemaHua represents a promising raw material for the development of functional foods.

High pressure processing combined with Phyllanthus emblica juice effectively inhibits enzymatic browning and enhances nutritional quality of NFC apple juice

Xueyan Zhang,Zi Li,Lixiang Zhu,Junjie Yi,Linyan Zhou

This study investigated the combined effects of high-pressure processing and Phyllanthus emblica (PE) juice supplementation on enzymatic browning and nutritional quality of apple juice. All PE-treated groups delayed enzymatic browning. After 35 d, the 10% PE group showed a 0.17-fold higher L* value, and 1.27- and 0.29-fold lower a* and b* values, respectively. The HPP-PE treatment suppressed polyphenol oxidase (PPO) and peroxidase (POD), with 10% PE reducing initial PPO and POD activities by 28% and 24%, respectively, and remaining 23.81% and 26.25% lower by day 35. PE supplementation enhanced biofunctional activities. In the 10% PE group, total phenol and flavonoid contents increased by 13.5- and 3.78-fold on day 0, and by 28.75- and 6.4-fold on day 35, respectively. Correspondingly, ABTS and DPPH radical scavenging activities increased by 18.23- and 15.35-fold on day 0, and by 20.56- and 33.81-fold on day 35. Twenty phenols were identified in PE juice, predominantly ellagic acid, trigalloyl-hexoside, ellagic acid 4-O-β-D-glucoside, and geraniin. Molecular docking indicated that hydrogen bonding and ionic interactions were the key forces inhibiting PPO activity. By integrating high-resolution metabolite profiling with molecular docking, this study elucidated the structure–function relationships underlying PE-derived browning inhibition, offering a mechanistically grounded, clean-label strategy for NFC juice preservation.

Triphasic sequential gas delivery via a photo-instructive hydrogel for enhanced diabetic skin wound healing

Jingwen Dong,Junjie Zhou,Shuguang Huang,Junmei Mu,Ye Tian,Zhifei Ding,Yihui Li,Aiping Wang,Zhanwei Zhou,Zeyu Xu,Minjie Sun

Near infrared light-triggered three-dimensional adaptive release in depth, time and dose for the treatment of diabetic foot ulcers. • A triphasic sequential NO release hydrogel achieves antibacterial, angiogenic and remodeling effects. • Triphasic sequential strategy for adapting NO dosage enhances the NO metabolic microenvironment at the wound condition, ultimately expediting wound healing. Diabetic foot ulcer (DFU) requires spatiotemporal coordinated modulation of the inflammatory response and tissue regeneration process, which is still the bottleneck. Here, a triphasic-sequential hydrogel was explored to tackle the infection treatment and tissue regeneration in DFU. The PDA@B6/L-Arg hydrogel was loaded with near infrared-responsive polydopamine nanoparticles, carrying the nitric oxide (NO) donors for different healing stages. Upon exposure to laser irradiation, 1) the PDA@B6/L-Arg hydrogel rapidly released a substantial amount of nitric oxide (NO) (μM) from N , N ′-di-sec-butyl-N,N′-dinitroso-1,4-phenylenediamine (BNN6), thereby synergistically achieving effective anti-bactericidal and anti-inflammatory effects; 2) L-Arg, as the substrate for nitric oxide synthases NOS, provided the support for deep angiogenesis; 3) the hydrogel degraded, allowing the released L-Arg to penetrate the wound and promote additional extracellular matrix remodeling. Compared to non-sequential NO therapy, PDA@B6/L-Arg hydrogel exhibited spatiotemporal modulation, enhancing the biofilm inhibition and angiogenesis. Furthermore, PDA@B6/L-Arg hydrogel, as a reactive oxygen species-scavenging agent, simultaneously attenuating inflammatory to improve the healing efficiency of chronic wounds, achieving 97.9% closure of infected wounds. It offers a promising therapeutic option for managing diabetic foot ulcers.

A Perioperative Hydrogel Platform for Integrated Endoscopic Submucosal Dissection Management

Zizhao Wang,Yang Yang,Hongxin Sun,Peng Fan,Xiaoqing Jia,Liang Zhang,Ting Wang,Jiulong Zhao,Qian Chen,Shige Wang

Endoscopic submucosal dissection (ESD), a minimally invasive procedure, is constrained by the rapid dissipation of conventional submucosal cushions and the absence of sustained postoperative therapeutic support. Herein, we develop a multifunctional hydrogel integrating mucosal elevating, sustained local analgesia, wound protection, antibacterial and antioxidant capabilities for perioperative management of ESD. The hydrogel is formed via Schiff base reactions between carboxymethyl chitosan and oxidized dextran, incorporates methylene blue for intraoperative visualization, and integrates mesoporous silica nanoparticles loaded with Bupivacaine (BUP@MSNs) to enable local sustained-release analgesia. Targeting critical limitations of conventional mucosal elevating agents, the hydrogel rapidly forms a stable submucosal cushion through in situ gelation, enhancing ESD procedural efficiency and safety while enabling immediate wound sealing and protection. Under physiological conditions, the hydrogel exhibits excellent injectability, self-healing capacity, and tissue adhesion at the lesion site. It also exerts potent antioxidant and antibacterial activities, fulfilling requirements for perioperative microenvironment modulation. Furthermore, the hydrogel delivers early postoperative analgesia that persists for over 48 h, effectively covering the peak pain phase post-ESD. Comprehensive in vitro and in vivo safety evaluations confirm its favorable biocompatibility and tolerability. Collectively, the hydrogel represents an integrated, clinically operable, and cost-effective candidate for gastrointestinal endoscopic minimally invasive therapies.

An Integrated Autolysis Process Utilizing Endogenous Enzymes for Enhanced Astaxanthin Extraction and De-Esterification From Antarctic Krill

Kexin Wang,Yurui Zhang,Wenzheng Shi,Donglei Luan

Natural astaxanthin is a high-value carotenoid abundantly found in Antarctic krill, the efficient extraction and bioactivity enhancement of natural astaxanthin are of significant importance for its application in nutraceuticals and functional foods. However, this process faces dual challenges: the robust structural matrix of krill impedes solvent penetration, and astaxanthin predominantly exists in esterified forms with lower bioavailability. To address this, we developed an integrated autolysis process that utilizes the krill's own endogenous enzymes (proteases, lipases, and chitinases) for simultaneous matrix disruption and astaxanthin de-esterification. Compared to conventional methods that rely on exogenous enzymes or chemicals, our approach offers distinct advantages: it is environmentally benign, cost-effective, and enables simultaneous matrix disruption and astaxanthin de-esterification in a single step. Through experimental design, the optimal autolysis conditions were identified as pH 7.5, 60 ℃C, and 2 h. Under these optimized conditions, the process achieved three key outcomes: (1) An astaxanthin yield of 91.18 µg/g was achieved, representing a significant increase compared to conventional extraction methods; (2) A protein hydrolysate was simultaneously produced, with its proportion of essential amino acids substantially elevated from 12.276 ± 0.013 to 34.368 ± 0.042 mg/100 g, thereby significantly enhancing its nutritional value; (3) Effective de-esterification raised the proportion of free astaxanthin from 18.11% to 30.50%. The resulting extract demonstrated superior antioxidant activity, which was positively correlated with its free astaxanthin content. This green and cost-effective strategy represents a promising approach for the comprehensive, high-value utilization of Antarctic krill biomass in nutraceutical and functional food applications.

Synthesis and Characterization of Argan Oil-Loaded Polyacrylamide Hydrogels: A Multifunctional Approach

Ahmad Ali,Jiaqi Yu,Qasim Qasim,Faizan Ikhlaq,Yangxufei Huang,Shun Yao

This study presents the pioneering development and comprehensive characterization of argan oil (AO)-loaded polyacrylamide (PAm) hydrogel (AO-PAm), establishing its multifunctional potential for biomedical applications. Systematic evaluation revealed that the AO-PAm hydrogel combines excellent mechanical properties (90% transparency, > 25 cm stretchability) with remarkable biological activity, demonstrating: (1) potent antibacterial effects against E. coli (1.2 ± 0.2 mm inhibition zone), P. aeruginosa (0.7 ± 0.2 mm), and S. aureus (0.6 ± 0.2 mm); (2) exceptional antioxidant capacity (97.6% DPPH scavenging at 75 μg/mL); and (3) pH-responsive swelling behavior. GC–MS analysis identified key bioactive compounds (β-pinene, camphene, undecanal, β-bisabolene) in AO that contribute to these properties, while microscopic characterization confirmed successful AO encapsulation within the PAm matrix. The hydrogel exhibited controlled drug release following Higuchi kinetics ( R 2 = 0.9547), with enhanced performance at physiological pH (6.5–6.8). These findings position AO-PAm hydrogel as a promising platform for advanced wound care, transdermal drug delivery, and daily healthcare applications, combining the therapeutic benefits of AO with the structural advantages of a synthetic hydrogel matrix.

Application of Subcritical Water Extraction and Enzymatic Hydrolysis as a Combination Green Extraction Technique for Rambutan (Nephelium lappaceum L.) Peel Polyphenol

Qiuming Liu,Qingyu Ma,Jiao Li,Liping Sun,Yongliang Zhuang,Yuanlong Tang

This study aimed to develop a green and efficient technique for extracting rambutan peel polyphenols, while systematically evaluating chemical components, the water solubility, storage stability, in vitro antioxidant activity, and digestive enzyme inhibitory potential of the extracts. Subcritical water (SW) alone and SW followed by enzymatic hydrolysis (SW-EH) were employed to extract rambutan peel polyphenols. The optimal conditions of SW extraction were temperature of 130°C, time of 30 min, and liquid–solid ratio of 80:1 mL/g, which yielded a total phenol content (TPC) of 226.54 mg gallic acid equivalents per gram of dry weight powder. A total of 39 phenolics were identified with ellagic acid as the predominant compound. SW-EH extract maintained similar phenolic profiles but contained less soluble dietary fiber compared to SW extract. The transmittance and polyphenol solubility of SW-EH extract were higher than those of SW extract in the concentration range of 5 to 100 mg/mL. The ellagic acid content in the SW and SW-EH extracts decreased by 19.05% and 1.71%, respectively, after 40 days of storage. Both extracts demonstrated comparable antioxidant activities superior to ascorbic acid, while EH treatment greatly enhanced α-glucosidase inhibitory activity compared to SW extract. With superior water solubility, storage stability, antioxidant activity, and digestive enzyme inhibitory potential, SW-EH extract is a promising product for a wide range of applications.

Carbon-Dot-Zn2+ Assembly Hybrid Multifunctional Hydrogels for Synergistically Monitoring Local Glucose and Repairing Diabetic Wound

Xu Yang,Xinmei Duan,Li Zhu,Xiong Zhao,Shirong Zhang,Sheng Ni,Shuai Wu,Zhiqiang Han,Wuquan Deng,Da Sun,Qiao Chen,Wei Wu

The prolonged healing period and increased rates of amputation and mortality are significant challenges for diabetic patients’ wound care in clinical practice. Therefore, there is an urgent need to develop timely and efficient wound management strategies to monitor the local glucose while promoting rapid wound healing. In this study, a multifunctional hydrogel (PF127@Zn/C-G) was constructed by coloading the monitoring and therapeutic carbon dot (CD)-based agents (Zn/C) and glucose oxidase (GOx). For applications, the ″off–on″ characteristic in fluorescence (FL) signal of the Zn/C assemblies enabled the real-time monitoring of the local glucose levels in diabetic wounds. Moreover, the synergistic effect of Zn2+ and CDs was developed to promote wound healing. In vitro and in vivo results showed that the PF127@Zn/C-G hydrogel exhibited a good linear relationship between FL intensity and glucose concentration, and the FL signal intensity in the wound environment in vivo correlated well with the blood glucose, suggesting its function for timely reflection of the blood glucose content. In addition, the PF127@Zn/C-G hydrogel could accurately fit the wound contour to form a protective film, while effectively promoting cell proliferation and migration, and scavenging the accumulated ROS at the local wound, exhibiting an excellent angiogenic capacity and collagen deposition for significantly accelerating the wound healing during tissue remodeling. Therefore, the multifunctional hydrogel lPF127@Zn/C-G provides an effective and feasible candidate for improving the diabetic wound management.

Electrospun Nanofiber Membranes Based on Sodium Alginate and Multi-Ether Fullerene Derivative as Wound Dressings with the Antioxidant Property

Xiaoxu Li,Yuchen Dong,Zhangzun Guo,Xiaoyu Huang,Xiaoyan Zhang

Excessive high levels of reactive oxygen species (ROS) at the wound site are also one of the main reasons for difficulty in wound healing. Herein, a kind of antioxidant nanofiber membranes (NFMs) was fabricated by electrospinning sodium alginate (SA) and multi-ether fullerene derivative (mono-C60-O) solutions, which combined the good biocompatibility of SA and excellent antioxidant property of fullerene. The molecular structure of mono-C60-O was characterized by 1H NMR, 13C NMR, and MALDI-TOF mass spectrometry. The morphology of the produced SA/mono-C60-O-X NFMs was homogeneous and consistent, as characterized by scanning electron microscopy (SEM). The scavenging efficiencies of SA/mono-C60-O-25 NFMs were high, about 93.3%, 91.1%, and 92.2% for DPPH, ABTS, and ·OH, respectively. The efficiency value is also significantly higher than that of the commercial product Tegaderm. Good cell and blood compatibility exhibited a potential use of SA/mono-C60-O-25 NFMs in wound healing. In vivo wound healing studies showed that the wound of the SA/mono-C60-O-25 NFM-treated mice group was almost completely healed, while the SA NFM-treated and control groups still obviously healed on day 14. Hematoxylin and eosin (H&E), Masson, and immunohistochemical staining indicated that the SA/mono-C60-O-25 NFMs had a rapid wound healing effect due to the excellent antioxidant property of fullerene, which could reduce inflammatory factors, accelerate angiogenesis, increase collagen deposition, and exert anti-inflammatory effects.

Interface-Engineered Polystyrene-Divinylbenzene Microspheres with Polyethylenimine and Metal-Phenolic Networks for Multifunctional Sepsis Hemoperfusion

Xingqiang Zhu,Wenjing Song,Ye Tian,Zhuhao Tan,Baolei Huang,Hailin Zhang,Anmin Wang,Wenfang Liu,Wei Zhai,Li Ren

Sepsis, a life-threatening organ dysfunction from a dysregulated infection response, causes many deaths, especially in the elderly. Aging population exacerbates the sepsis burden, requiring cost-effective prevention, detection, and treatment strategies. Hemoperfusion has emerged as an effective clinical treatment for sepsis. Herein, polystyrene-divinylbenzene microspheres functionalized with metal-phenolic networks and polyethylenimine (P-PEI Cu/TAn microspheres) with endotoxin and cell-free DNA (cfDNA) adsorption, oxidative stress relief and antibacterial activity were constructed as a multifunctional adsorbent for hemoperfusion therapy toward sepsis. The adsorbent exhibited good blood compatibility, high-efficiency adsorption of endotoxin (with an adsorption capacity of 1239.6 EU/g) and cfDNA (with a clearance ratio of 74.8%), robust scavenging of reactive oxygen/nitrogen species, and antibacterial activity against Escherichia coli (84.6%) and Staphylococcus aureus (68.1%) within 4 h. In simulated hemoperfusion, P-PEI Cu/TA0.5 microspheres achieved rapid endotoxin removal (75.3% clearance in 15 min) and effectively mitigated oxidative stress induced by endotoxin, as evidenced by preserved blood oxidoreductase activity. This work introduced a novel therapeutic approach that combines toxin adsorption, antioxidative protection, and antibacterial effects, holding significant promise for enhancing clinical sepsis management through extracorporeal blood purification.
