Characteristics of myoglobin degradation by cold plasma and its pro-oxidative activity on lipid in washed fish muscle
In this study, the degradation of myoglobin both in solution and in tuna muscle by cold plasma (CP) was investigated through ultraviolet–visible and fluorescence spectroscopy analysis, and the effect of the degradation products on lipid oxidation in washed turbot muscle (WTM) was explored. Results showed that heme in myoglobin was degraded upon CP treatment, from which a fluorescent product was formed and iron was released. The degradation products promoted lipid oxidation in WTM during cold storage, in which the released iron played an important role. Results of gas chromatography-ion mobility spectrometry analysis showed that the contents of 37 volatile compounds in WTM were increased by heme degradation products induced lipid oxidation, including 20 aldehydes, 9 alcohols, 5 ketones and 3 furans. This study provides a new insight to the mechanism of lipid oxidation in CP-treated fish during cold storage, as well as guidance for preserving myoglobin-containing foods with CP.
Efficient production of ε-poly-l-lysine using cassava starch and fish meal by Streptomyces albulus FQC-24
ε-Poly-l-lysine (ε-PL) is used as a natural food preservative which consists of l-lysine units connected. However, due to the expensive culture medium, the production cost of ε-PL remains high. In this study, cheap raw materials cassava starch (CS) and fish meal (FM) were employed by S. albulus FQC-24 for ε-PL production. In the single factor experiment, the maximum ε-PL production reached 0.97 g/L at 60 g/L CS and 15 g/L FM. The results of screening experiments by Plackett–Burman design showed that three main components affecting ε-PL production were CS, FM, and (NH4)2SO4. And the standardized effects of CS, FM, and (NH4)2SO4 were 0.13, −0.22, and −0.2, respectively. The optimum fermentation medium developed by response surface methodology for ε-PL production contained (g/L) CS, 67.56; FM, 14.70 and (NH4)2SO4, 5.41. Under the optimum conditions, the ε-PL production was achieved 1.30 g/L, with 34.02% higher than that before optimization. Moreover, ε-PL productions of batch and fed-batch fermentation in a 7-L fermentor were improved to 2.13 and 17.17 g/L respectively, which increased by 0.64 and 12.2 times compared with the shake flask culture. The results indicated that FM and CS are promising substrates for the efficient production of ε-PL.
Preparation of magnetic mesoporous metal-phenolic coordination spheres for extraction of crystal violet and leuco-metabolites in fish
To address the undesirably low porosity of phenolic resin, a new material termed magnetic mesoporous metal-phenolic coordination spheres (Fe 3 O 4 @Co-TA) was synthesized by chelating tannic acid (TA) with metal ions. Fe 3 O 4 @Co-TA was characterized by transmission electron microscopy, scanning electron microscopy, hysteresis loop (B-H) analysis, Fourier-transform infrared spectroscopy, and N 2 adsorption-desorption. The results indicated that the new material comprises mesopores (2 nm and 3 nm) and exhibits a good magnetic response (44 emu/g). Combined with high-performance liquid chromatography (HPLC), a novel method for the detection of crystal violet (CV) and leucocrystal violet (LCV) by magnetic solid-phase extraction was established. Under the optimal extraction conditions, the linear ranges of CV and LCV detection were 0.2‒60 μg/L and 0.04‒40 μg/L, the detection limits were 0.04 μg/L and 0.008 μg/L, and the enrichment factors were 435 and 460, respectively. Fe 3 O 4 @Co-TA was reused ten times without significant reduction of the extraction ability. This method was successfully used for the detection of CV and LCV in fish samples, providing an effective technique for food safety monitoring and quality control.
Determination of Nitrofuran Metabolites in Fish by Ultraperformance Liquid Chromatography-Photodiode Array Detection with Thermostatic Ultrasound-Assisted Derivatization
Nitrofuran (NF) is a class of broad-spectrum antibiotics that are used illegally in animal feeding. NF and its metabolites have proven to pose potential risk to human health. To address the current analytical needs to quantify low levels of NF metabolites in animal foods, a sensitive method was developed for simultaneous detection of four NF metabolites in fish products by an ultraperformance liquid chromatography-diode array detector (UPLC-DAD). With 2-nitrobenzaldehyde (2-NBA) as the derivatizing reagent, the metabolites were hydrolyzed and derivatized under the assistance of thermostatic ultrasound. Compared with the current detection methods, the time of the derivatization reaction has been shortened from 16 to 2 h. The relative coefficient of four NF metabolite derivatives reached more than 0.998, with excellent linear relationship. The limits of detection (LODs) and limits of quantification (LOQs) of six repeated determinations reached 0.25–0.33 and 0.80–1.10 μg/kg, respectively. For all four NF metabolites, the limit of detection of the method was below the minimum required performance limit (MRPL) of 1.0 μg/kg, which makes it compatible with the EU requirements. The recoveries ranging from 89.8 to 101.9% with relative standard deviation below 6.5% were obtained for all of the NF metabolites. What’s more, this method was successfully applied for the determination of four NF metabolites in the fish products. As a promising approach, this method could also be extended for the quantitation of NF metabolites in aquaculture and poultry products.
Determination of Ciprofloxacin in Fish by Surface-Enhanced Raman Scattering Using a Liquid-Liquid Self-Assembled Gold Nanofilm
Trace detection of antibiotic ciprofloxacin by surface-enhanced Raman scattering (SERS) remains a challenge, because this compound is water-insoluble and amphoteric and it has no functional groups to combine with gold nanoparticles. Herein, ciprofloxacin was physically adsorbed on a self-assembly gold nanofilm for realizing quantitative SERS detection of ciprofloxacin in fish. A gold nanofilm containing ciprofloxacin spontaneously formed at the interface of gold colloids/n-hexane by vortex mixing in a few seconds without ethanol or other organic additives. The density of the gold nanoparticles on the interface which influences the SERS effect was controlled by varying the vortex mixing time and the volume of n-hexane. Furthermore, the proposed substrates with an enhancement factor of 2.7 × 106 were successfully applied for the determination of ciprofloxacin in fish samples with a linear range from 0.5 to 10.0 μg/mL and a detection limit of 0.19 μg/mL. The recovery measurements showed that the proposed SERS method had satisfactory precision and accuracy.
Preparation of aminated fish scale collagen and oxidized sodium alginate hybrid hydrogel for enhanced full-thickness wound healing
Acute full-thickness wounds require a more extended healing period, thus increasing the risk of infection. Severe infection frequently resulted in wound ulceration, necrosis, and even life-threatening complications. Here, a hybrid hydrogel comprising aminated collagen (AC), oxidized sodium alginate (OSA), and antimicrobial peptides (polymyxin B sulfate and bacitracin) was developed to enhance full-thickness wound healing. The AC with low immunogenicity and high biocompatibility was made from marine fish scales, which are eco-friendly, low-cost, and sustainable. The cross-linked hydrogel was formed by a Schiff base reaction without any catalysts and additional procedures. As expected, the presented hybrid hydrogel can effectively against E. coli and S. aureus , as well as promote cell growth and angiogenesis in vitro . In addition, the hydrogel can promote full-thickness wound healing in a rat model through accelerating reepithelialization, collagen deposition, and angiogenesis. Our work demonstrated that the hybrid hydrogel has promising applications in the field of wound healing, which would prompt the utilization of marine fish resources during food processing.
Fish oil-loaded emulsions stabilized by synergetic or competitive adsorption of gelatin and surfactants on oil/water interfaces
The simultaneous application of proteins and surfactants for emulsion preparation and stabilization is a research hotspot in the field of emulsions, and their interfacial adsorption mechanisms remain unclear. In order to analyze the interaction mechanism of gelatin with low-molecular-weight surfactants, we mainly explored the preparation and storage of fish oil-loaded gelatin/surfactant-stabilized emulsions in this work. The results demonstrated that gelatin and four types of surfactants were synergetically (Span 80 and soybean lecithin) or competitively (Tween 80 and SDS) adsorbed on the oil/water interfaces in the emulsions. The adsorption behaviors affected emulsion stability (creaming, liquid-gel transformation, and droplet coalescence behaviors) and size distribution of emulsion droplets. These fish oil-loaded gelatin/surfactant-stabilized emulsions have the potential to reduce the disadvantages of fish oils and to provide multiple prospective applications in beverages, hard foods, and alkaline foods. It would also be beneficial to the basic understanding of protein/surfactant interfacial adsorption in emulsion formation.
Long-Lived Plasma Cells Secrete High-Affinity Antibodies Responding to a T-Dependent Immunization in a Teleost Fish
The recent discovery of long-lived plasma cells (LLPCs) in mammals, which provide a constant expression of specific high-affinity antibodies that mediate humoral memory, has caused a dramatic paradigm shift in the study of immunity and vaccine development. In teleost fish, there are few studies regarding the association between LLPCs and antibody production, and the affinity of the antibodies secreted by the LLPCs is poorly understood. In the present study, channel catfish (Ictalurus punctatus) were immunized with trinitrophenylated-keyhole limpet hemocyanin (TNP-KLH) to examine TNP-specific antibody titers, affinities, antibody-secreting cell (ASC) dynamic changes, and especially the affinity of secreted antibodies by LLPCs post-immunization. We demonstrated that TNP-specific LLPCs were generated starting at week 4 post-immunization, achieved a maximal number at week 8, and maintained a comparable level throughout the 18-week post-immunization period, which was correlated with the dynamics of serum antibody titers and affinity maturation in the response. The LLPCs appeared to mostly reside within, or migrate to, the anterior kidney (bone marrow-like tissue in mammals), but a small portion was also located in the spleen and peripheral blood. The antibodies produced by the LLPCs possessed high affinities, indicating that the generation and development of LLPCs were driven by affinity selection in teleosts. Collectively, the results of this study provide insights toward the evolutionary understanding of the affinity-dependent mechanism of LLPC generation and development.
The inhibitory effect of large yellow croaker roe phospholipid as a potential antioxidant on fish oil oxidation stability
Large yellow croaker roe phospholipid (LYCRPLs) is a potential antioxidant. In this study, the inhibition of LYCRPLs on fish oil (FO) oxidation was systematically investigated by evaluating peroxide value , acid value, p -anisidine value, thiobarbituric acid value, and volatile compounds in the accelerated oxidation reaction . The results showed that LYCRPLs presented remarkable antioxidant activity and extended FO shelf-life compared with soybean phospholipids. The analysis of volatile compounds elucidated that LYCRPLs mainly inhibited the formation of aldehyde volatile compounds and thus improved the oxidative stability of FO. Moreover, LYCRPLs could effectively improve the thermal stability of FO. This research provides new insights into LYCRPLs as a promising natural antioxidant in the food industry.
Flavor and sensory profile of Chinese traditional fish noodles produced by different silver carp (hypophthalmichthys molitrix) mince ingredients
This study employs sensory evaluation, headspace gas chromatography–ion mobility spectrometry (HS-GC-IMS), and headspace solid-phase microextraction gas chromatography–mass spectrometry (HS-SPME-GC-MS) techniques to investigate the effect of different pretreatment of fresh silver carp mince (running water rinsing 0, 1, or 2 times) and commercially frozen surimi on the odor characteristics of fish noodles. The free choice profiling (FCP) and check all that apply (CATA) sensory analysis methods were utilized to identify 10 characteristic descriptors, which include “grass, fish fragrance, unpleasant fishy, fatty, roast, ammonia, caramel, warmed-over, earthy, and mushroomy”. HS-GC-IMS and HS-SPME-GC-MS detected 80 and 37 volatile compounds (VCs) in fish noodles. The 1-Penten-3-ol, (E)-2-pentenal-D, hexanal-D, pentanal-D, (E,E)-2, 4-heptadienal-D contents were significantly correlated with “fish fragrance” and “unpleasant fishy”, and octanal, nonanal, heptanal, 2-methylpyrazine contents were significantly correlated with “warmed-over” flavor. The results of this study can be helpful for fish noodle quality improvement and industrial production.
Development of tilapia muscle-based Shanghai smoked fish and the effect of salt amounts in the preparation process
Frozen tilapia muscle-based Shanghai smoked fish without fish bones was developed using a three-stage method (salt pickling, frying, and sugar smoking), and the effect of varying salt amounts (0.5%, 1.0%, and 1.5% of muscles) was studied. The muscle with 1.0% salt had the best sensory evaluation score. Salt pickling significantly increased and then frying significantly decreased the numbers and concentrations of the volatile organic compounds (VOCs) in the muscles. Compared with the others, 1.0% salt resulted in the lowest numbers and concentrations of the VOCs in the frying stage and the highest VOC concentrations in the sugar smoking stage. Statistical analyses of VOC concentrations suggested that two VOC classes (esters and aldehydes) and 42 VOC individuals could be used to differentiate the treated tilapia muscles with different salt amounts. This work provided a basic knowledge of the effects of food processing stages and salt amounts on fish quality.
