(Pc)Eu(Pc)Eu[trans-T(COOCH3)2PP]/GO Hybrid Film-Based Nonenzymatic H2O2 Electrochemical Sensor with Excellent Performance

Zhenning Yu,Lei Zou,Yanli Chen,Jianzhuang Jiang

A facile approach was developed for preparing the multilayer hybrid films of mixed (phthalocyaninato) (porphyrinato) europium(III) triple-decker compound (Pc)Eu(Pc)Eu[trans-T(COOCH3)2PP] (1) and graphene oxide (GO) using the solution-processing QLS method. The combination of the nature of relatively high conductivity and great surface area for GO with the electroactive and semiconductive triple-decker compound in ITO electrode renders the hybrid film excellent sensing property for H2O2, due to the optimized triple-decker molecular packing in the uniform-sized nanoparticles (ca. 70 nm) formed on the GO surface. The amperometric responses are linearly proportional to the concentration of H2O2 in the range of 0.05–1800 μM with a fast response time of 0.03 s μM–1, a low detection limit of 0.017 μM, and good sensitivity of 7.4 μA mM–1. The present work represents the best result of tetrapyrrole-based nonenzymatic electrochemical sensor for H2O2. Nevertheless, the triple-decker/GO/ITO also shows excellent stability, reproducibility, and selectivity, indicating the great potential of electroactive tetrapyrrole rare earth sandwich compounds in combination with GO in the field of nonenzymatic electrochemical sensors.

Trace-Dissolved S(-II) Triggers the Fe(III)-Activated H2O2 Process for Organic Pollutant Degradation by Promoting the Fe(III)/Fe(II) Cycle: Kinetics, Toxicity, and Mechanisms

Kunjie Hou,Zhoujie Pi,Fei Chen,Li He,Fubing Yao,Shengjie Chen,Xiaoming Li,Haoran Dong,Qi Yang

The practical application of the traditional Fenton system (Fe(II)/H2O2) is seriously hampered by the sluggish Fe(III)/Fe(II) cycle and the stringent acidic reaction condition. In this work, we found that trace-dissolved S(-II) (DS(-II), 5 μM) could significantly accelerate the Fe(III)/Fe(II) cycle and trigger a rapid H2O2 activation process in the Fe(III)/H2O2 system. Thus, more hydroxyl radicals (·OH) were generated for the rapid removal of various organic pollutants such as bisphenol A (BPA) and sulfamethoxazole (SMX) from water. Besides, the additive DS(-II) effectively broadened the application pH range of the Fenton or Fenton-like system. The constructed Fe(III)/DS(-II)/H2O2 system showed extensive applicability to different water matrices and maintained high BPA removal in the actual water sources. Furthermore, the toxicity assessment revealed that the toxicity of the target contaminant was diminished. The harmless SO42– was the final product of DS(-II) in the Fe(III)/DS(-II)/H2O2 system, and the residual DS(-II) was less than the threshold limit value for fresh or saltwater (0.5 mg/L). This discovery is expected to promote the large-scale practical application of iron-based Fenton or Fenton-like systems in practical organic wastewater purification.

Biodegradable Nanocatalyst with Self-Supplying Fenton-like Ions and H2O2 for Catalytic Cascade-Amplified Tumor Therapy

Wenting Li,Xinglu Zhou,Shikai Liu,Jialing Zhou,He Ding,Shili Gai,Rumin Li,Lei Zhong,Huijie Jiang,Piaoping Yang

Therapeutic nanosystems triggered by a specific tumor microenvironment (TME) offer excellent safety and selectivity in the treatment of cancer by in situ conversion of a less toxic substance into effective anticarcinogens. However, the inherent antioxidant systems, hypoxic environment, and insufficient hydrogen peroxide (H2O2) in tumor cells severely limit their efficacy. Herein, a new strategy has been developed by loading the chemotherapy prodrug disulfiram (DSF) and coating glucose oxidase (GOD) on the surface of Cu/ZIF-8 nanospheres and finally encapsulating manganese dioxide (MnO2) nanoshells to achieve efficient DSF-based cancer chemotherapy and dual-enhanced chemodynamic therapy (CDT). In an acidic TME, the nanocatalyst can biodegrade rapidly and accelerate the release of internal active substances. The outer layer of MnO2 depletes glutathione (GSH) to destroy the reactive oxygen defensive mechanisms and achieves continuous oxygen generation, thus enhancing the catalytic efficiency of GOD to burst H2O2. Benefiting from the chelation reaction between the released Cu2+ and DSF, a large amount of cytotoxic CuET products is generated, and the Cu+ are concurrently released, thereby achieving efficient chemotherapy and satisfactory CDT efficacy. Furthermore, the release of Mn2+ can initiate magnetic resonance imaging signals for the tracking of the nanocatalyst.

A Three-Dimensional Electrochemiluminescence Sensor Integrated with Peptide Hydrogel for Detection of H2O2 Released from Different Subtypes of Breast Cancer Cells

Yunfan Zhou,Xue Wei,Jun Chen,Hanzhi Xiong,Dandan Sui,Xu Chen,Wensheng Yang

Breast cancer is a malignant tumor, with various subtypes showing different behaviors. Endogenous H2O2 is an important marker of tumor progression, which makes it important to study the relationship between breast cancer subtypes and H2O2 for pathogenesis and treatment strategies, but this has rarely been reported so far. In this work, we constructed a three-dimensional (3D) electrochemiluminescence (ECL) sensing platform for the detection of H2O2 released from two typical subtypes of breast cancer cells (MCF-7 cells for luminal A-type and MDA-MB-231 cells for three negative breast cancers, TNBCs). To adequately replicate the tumor microenvironment, the peptide hydrogel was introduced as a scaffold for 3D cell culture. The titanium foam (TF) was used as a 3D electrode to better match the 3D culture substrate. N-(4-Aminobutyl)-N-ethylisoluminol (ABEI) was selected as the ECL emitter and assembled into the peptide hydrogel by hydrogen bonding and π-stacking, which resulted in a stable and homogeneous distribution of ABEI along the hydrogel fibers. Furthermore, basic amino acids were introduced to provide alkaline microenvironment for ABEI. Therefore, ABEI exhibited high ECL efficiency, resulting in a high sensitivity with an ultralow detection limit of 0.023 nM (S/N = 3) for H2O2 of the proposed ECL biosensor. MCF-7 and MDA-MB-231 cells were cultured in a 3D peptide hydrogel/ABEI/TF electrode, respectively, and endogenous H2O2 was successfully monitored. A notably significant difference of H2O2 released between MDA-MB-231 cells and MCF-7 cells without stimulation but similar extra release with stimulation were observed. These findings may help understand the physiological mechanisms behind the various subtypes and reactive oxygen species (ROS)-related treatment for breast cancer.

Cu-Doped black phosphorus quantum dots as multifunctional Fenton nanocatalyst for boosting synergistically enhanced H2O2-guided and photothermal chemodynamic cancer therapy

Haimei Li,Yaofa Liu,Shulan Li,Silong Zhang,Biao Huang,Ran Cui,Yi Liu,Peng Jiang

Chemodynamic therapy (CDT) is a cancer treatment that converts endogenous H2O2 into hydroxyl radicals (˙OH) through Fenton reaction to destroy cancer cells. However, there are still some challenges in accelerating the Fenton reaction of CDT and improving the biodegradability of nanocatalysts. Herein, a multifunctional biomimetic BPQDs-Cu@GOD (BCG) Fenton nanocatalyst for boosting synergistically enhanced H2O2-guided and photothermal CDT of cancer is reported. Cu2+ in BCG can be reduced to Cu+ by black phosphorus quantum dots (BPQDs), triggering a Cu+-mediated Fenton-like reaction to degrade H2O2 and generate abundant ˙OH for cancer CDT. The loaded glucose oxidase (GOD) can consume the glucose in the tumor to produce abundant H2O2 for Fenton-like reaction. In addition, Cu2+ in BCG can react with GSH in tumor cells to alleviate the antioxidant capacity of tumor tissues, further improving the CDT efficacy. Furthermore, the photothermal performance of BPQDs can be enhanced by capturing Cu2+, improving the photoacoustic imaging and photothermal therapy (PTT) functions. More importantly, the enhanced photothermal performance can rapidly accelerate the Fenton-like reaction under NIR irradiation. Finally, Cu2+ can accelerate the degradation of BPQDs, which can reduce the retention of reagents. As a novel multifunctional biocompatible Fenton nanocatalyst, BCG have great potential in cancer therapy.

DNA-enhanced peroxidase-like activity of layered double hydroxide nanosheets and applications in H2O2 and glucose sensing

Lijian Chen,Kaifang Sun,Peipei Li,Xianzhong Fan,Jianchao Sun,Shiyun Ai

LDH nanosheets were obtained via continuous impaction and exfoliation by herring sperm DNA molecules using a constant vibration method. DNA–LDH nanohybrids were composed by electrostatic forces and they exhibited DNA-enhanced peroxidase-like activity. The morphology and structure of DNA–LDH nanohybrids were analyzed by transmission electron microscopy (TEM), selected-area electron diffraction (SAED), X-ray diffraction (XRD), and atomic force microscopy (AFM) characterization. On the basis of the high catalytic activity of DNA/CuAl–LDH nanosheets, a rapid, sensitive, and convenient approach was developed for colorimetric detection of H2O2 and blood glucose. This method can be potentially applied in medical diagnostics and biotechnology fields.

Polydopamine-coated bimetallic ZIF derivatives as a H2O2-free oxidase mimetic for the colorimetric sensing of L-cysteine

Ran Zhang,Shaohong Zhang,Junhao Lu,Ying Wu,Jinjin Zhao,Zhijuan Wang

Nanozymes have attracted much attention as a new type of artificial enzyme with great development potential in recent years. Here, a cubic Co and N co-doped porous carbon material (Co-HPNC@NC) was synthesized through the calcination of PDA-modified bimetallic Co–Zn ZIFs. Co-HPNC@NC exhibits exceptionally high oxidase-mimicking activity and effectively oxidizes the colorless 3,3′,5,5′-tetramethylbenzidine (TMB) to blue-colored oxidized TMB (oxTMB) without H2O2. However, the formation of oxTMB was inhibited by the addition of L-cysteine (L-Cys). Therefore, the blue color faded to colorless, causing a notable decrease in absorbance. Furthermore, Co-HPNC@NC showed good selectivity and anti-interference ability toward the detection of L-Cys. As a result, a sensitive and efficient colorimetric method has been developed to determine L-Cys with a low detection limit of 0.17 μM (3σ). It is believed that this colorimetric method for the detection of L-Cys based on Co-HPNC@NC will show potential applications in biosensors and medical diagnosis.

Hydrangea-like AuPtRu/ZnO-rGO Nanocomposites with Enhanced Peroxidase Mimiking Activity for Senitive Colorimetric Determination of H2O2

Jie Dong,Laixi Zhang,Wei Li,Xin Hu,Anyi Chen,Chaorui Li

In this study, a facile and cost-effective hydrothermal synthesis method was used to synthesize zinc oxide nanoflowers modified by reduced graphene oxide, and subsequently, trimetallic AuPtRu nanoparticles(AuPtRuNPs) were supported via the reduction method for high-sensitivity colorimetric detection of H2O2 in weakly acidic solutions. Compared to monometallic and bimetallic nanoparticles, trimetallic nanoparticles exhibit significant synergistic effects and enhanced catalytic activity. After providing a three-dimensional structure with multiple pores by zinc oxide and enhancing electron transfer ability by reduced graphene, the trimetallic nanocomposites (AuPtRu/ZnO-rGO) exhibited excellent peroxidase-mimicking activity, which can effectively catalyze 3,3′,5,5′-tetramethylbenzidine (TMB) to produce a blue oxidation product (oxTMB) in the presence of H2O2. Compared to horseradish peroxidase (HRP), AuPtRu/ZnO-rGO demonstrated significantly enhanced catalytic velocity (Vmax = 6.16 × 10–8 M/s) and affinity (Km = 0.02) for H2O2. The study of the catalytic mechanism showed that trimetallic Au, Pt, and Ru could effectively catalyze H2O2 to produce hydroxyl radicals (•OH) to accelerate the oxidation of TMB and enhance the peroxidase-mimicking activity of the AuPtRu/ZnO-rGO nanocomposites. The results showed that the as-synthesized hydrangea-like AuPtRu/ZnO-rGO nanocomposites showed enhanced peroxidase-mimicking activity. It could be used for the colorimetric detection of H2O2 in the range 5–1000 μM with a LOD of 3.0 μM (S/N = 3), and the recoveries are 93.0–101.7%. In addition, the AuPtRu/ZnO-rGO nanocomposites have good applicability for sensitive colorimetric determination of H2O2 in milk, and it has broad application prospects as a multifunctional sensing platform in the food processing industry.

Copper peroxide coated upconversion nanoparticle modified with glucose oxidase for H2O2 self-supplying starvation-enhanced chemodynamic therapy in vitro

Yale Hong,Qinfeng Tao,Yuan-Yuan Liu,Zhuo Wang,Haifang Wang,Lining Sun

Chemodynamic therapy (CDT) can convert endogenous hydrogen peroxide (H2O2) to highly reactive hydroxyl radical (˙OH) through Fenton or Fenton-like reaction to kill tumor cells, which is a promising anticancer strategy. However, the limited H2O2 and overexpressed glutathione (GSH) in tumor cells make CDT ineffective. Here, an efficient nanocomposite, UCN@CuO2–GOx (UCCuG), was synthesized, realizing both starvation therapy and H2O2 self-supplying CDT in vitro. In this case, the glucose oxidase (GOx) of the nanocomposite could consume glucose for starvation therapy after the UCCuG nanocomposite entered tumor cells. In addition, the acidic environment of the lysosome triggered the release of Cu2+ and H2O2 by the decomposition of UCCu; then, Cu2+ was reduced to Cu+ by GSH in tumor cells; and finally, Cu+ catalyzed the released H2O2 to generate ˙OH for CDT. The in vitro experiments demonstrated starvation-enhanced CDT with remarkable results. Meanwhile, under 980 nm laser irradiation, the upconversion luminescence signal of UCN in the UCCuG nanocomposite was reduced due to the CuO2–GOx coating, while it gradually recovered after the UCCuG nanocomposite reacted with glucose and GSH under the tumor microenvironment (TME). Such a luminescent intensity recovery process is expected to monitor the TME-activated therapeutic effect in real time. This strategy may solve the problem of insufficient CDT efficacy caused by limited endogenous H2O2 and overexpressed GSH in tumor cells. This multifunctional nanocomposite demonstrates the promising application of starvation-enhanced CDT in tumor treatment.

A tunable bifunctional hollow Co3O4/MO3 (M = Mo, W) mixed-metal oxide nanozyme for sensing H2O2 and screening acetylcholinesterase activity and its inhibitor

Xiaodan Zhang,Yuwan Lu,Qiumeng Chen,Yuming Huang

A self-templated strategy was adopted to design hollow Co3O4/MO3 (M = Mo, W) mixed-metal oxides via the Mo or W doping of ZIF-67, and subsequent pyrolysis under an atmosphere of air at a low temperature of 450 °C. The hollow Co3O4/MO3 (M = Mo, W) mixed-metal oxides displayed tunable oxidase-like and peroxidase-like activities able to efficiently catalyze the oxidation of TMB to generate a deep blue color in the absence or presence of H2O2. Relative to that of the un-doped Co3O4, the oxidase mimic activity of the Mo-doped Co3O4 increased to 1.3 to 2.1-fold, while its peroxidase mimic activity increased to 7.1 to 19.9-fold, depending on different Mo doping amounts. The oxidase mimic activity of the W-doped Co3O4 increased to 2.1 to 2.3-fold, while its peroxidase mimic activity increased to 4.8 to 5.9-fold, depending on the different W doping amounts. The Mo- and W-doped Co3O4 nanohybrid exhibited both higher O2 and H2O2 activating capability, and their H2O2 activating capacity was superior to the O2 activating capability. Furthermore, the Mo- and W-doped Co3O4 nanohybrids exhibited similar O2 activating abilities, while the Mo-doped one displayed a higher H2O2 activating capability than the W-doped one. The discrepant peroxidase-like nature of Mo- and W-doped Co3O4 nanohybrids is likely attributed to their different catalytic mechanisms. The peroxidase-like activity of Mo-doped Co3O4 is highly related to the ˙OH free radical, while that of W-doped Co3O4 is likely ascribed to the electron transfer between TMB and H2O2. The Km values of Co3O4/MoO3 for TMB and H2O2 were 0.0352 mM and 0.134 mM, which were 3.2- and 1.9-fold lower than that of pure Co3O4, respectively. A Co3O4/MoO3-based colorimetric platform was developed for the determination of H2O2 in the 0.1–200 μM range, with a limit of detection of 0.08 μM (3σ). Based on the thiocholine (TCh) inhibition of the excellent peroxidase-like activity of Co3O4/MoO3 and the TCh generation via acetylcholinesterase (AChE) catalyzed hydrolysis of acetylthiocholine chloride (ATCh), the colorimetric platform was extended to screen AChE activity and its inhibitor.

3D DNAzyme Motor Nanodevice With Self-Powered FRET Amplifier and Self-Supplied H2O2 for Enhancing Human Neutrophil Elastase Profiling and Chemodynamic Therapy in Lung Tumor

Huiyan Du,Ensheng Xu,Yihan Xu,Qingwang Xue,Hongxia Xu,Jibin Song

The development of theragnostic nanosystems integrating FRET (fluorescence resonance energy transfer) imaging and chemodynamic therapy (CDT) for accurate diagnosis and effective treatment of lung tumors is still a big challenge. Herein, a peptide-assembled 3D DNAzyme motor nanodevice is engineered for a self-powered FRET amplifier profiling human neutrophil elastase (HNE) and self-supplied H 2 O 2 enhancing CDT. The nanodevice is prepared by depositing AuNPs on ZIF-8, in which ZIF-8 co-loaded the lysosomal targeting peptide-modified copper peroxides (PCPs) and hairpins (H1, H2, and H3), AuNPs are co-labeled by DNAzyme-peptide (DP) conjugate and H3. In the tumor micro-environment, HNE driven 3D DNAzyme walker followed by an exponential amplification constructed by a synergistic cross-activation between hybridization chain reaction and DNAzyme, generating a self-powered FRET amplifier. The FRET amplifier specifically measures HNE with a sensitivity of 0.026 pM, and successfully images exogenous HNE in living cells and monitors HNE in mouse models. Moreover, the PCPs can target lysosomes, reducing lysosome escape. The self-supplying H 2 O 2 undertaken by PCPs improves the Cu (II)-catalyzed Fenton-like reaction, effectively causing cell apoptosis to inhibit tumor growth. Significantly, the nanodevice successfully screens inhibitors and discriminates the HNE level in normal and lung cancer tissues, suggesting that the nanodevice provides an effective tool for the diagnosis and treatment of lung tumors.

A H2O2-activatable nanoprobe for diagnosing interstitial cystitis and liver ischemia-reperfusion injury via multispectral optoacoustic tomography and NIR-II fluorescent imaging

Chen Junjie,Chen Longqi,Wu Yinglong,Fang Yichang,Zeng Fang,Wu Shuizhu,Zhao Yanli

Developing high-quality NIR-II fluorophores (emission in 1000–1700 nm) for in vivo imaging is of great significance. Benzothiadiazole-core fluorophores are an important class of NIR-II dyes, yet ongoing limitations such as aggregation-caused quenching in aqueous milieu and non-activatable response are still major obstacles for their biological applications. Here, we devise an activatable nanoprobe to address these limitations. A molecular probe named BTPE-NO 2 is synthesized by linking a benzothiadiazole core with two tetraphenylene groups serving as hydrophobic molecular rotors, followed by incorporating two nitrophenyloxoacetamide units at both ends of the core as recognition moieties and fluorescence quenchers. An FDA-approved amphiphilic polymer Pluronic F127 is then employed to encapsulate the molecular BTPE-NO 2 to render the nanoprobe BTPE-NO 2 @F127. The pathological levels of H 2 O 2 in the disease sites cleave the nitrophenyloxoacetamide groups and activate the probe, thereby generating strong fluorescent emission (950~1200 nm) and ultrasound signal for multi-mode imaging of inflammatory diseases. The nanoprobe can therefore function as a robust tool for detecting and imaging the disease sites with NIR-II fluorescent and multispectral optoacoustic tomography (MSOT) imaging. Moreover, the three-dimensional MSOT images can be obtained for visualizing and locating the disease foci.
