The fruit ofMorus albaL. (MAF) has been consumed as a food worldwide. MAF has also been widely used in traditional medicine for thousands of years in East Asia, and its diverse bioactivities have been reported in numerous publications. However, no prokinetic activity has been reported for MAF or its components. In the present study, therefore, we investigated the effects of MAF on gastrointestinal motor function by measuring the intestinal transit rate (ITR) of Evans blue in mice in vivo. The ITR values accelerated by MAF were significantly higher than those accelerated by cisapride or metoclopramide, suggesting that MAF has potential as a new prokinetic agent to replace cisapride and metoclopramide. We also investigated the effects of MAF on myogenic and neurogenic contractions in human intestinal smooth muscles by measuring spontaneous contractions of smooth muscle strips, smooth muscle contractions induced by neural stimulation, and migrating motor complexes from intestinal segments in the human ileum and sigmoid colon in situ. MAF increased both myogenic and neurogenic contractions to enhance ileal and colonic motility in the human intestine. Taken together, these results indicate that MAF enhanced intestinal motility by increasing both myogenic and neurogenic contractions, thereby accelerating the ITR.Keywords:intestinal transit;Morus alba;mulberry fruit;myogenic contraction;neurogenic contraction;prokinetic;smooth muscle

Fluorinated amino acids play a crucial role as fundamental components in drug synthesis owing to their favorable attributes that enhance the pharmacokinetic properties of drugs. Nevertheless, the conventional chemical synthesis of fluorinated amino acids faces challenges in achieving stereoselectivity control. Additionally, the high cost of substrates and the intricate cofactor recycling process in enzymatic synthesis have limited their industrial production and applications. In this study, we developed a three-step chemoenzymatic cascade for the asymmetric synthesis of chiral fluorinated amino acids. The methodology involves the utilization of a variety of cost-effective aldehydes as starting materials, enabling economical aldol reactions catalyzed by aldolase. Subsequent chemical decarboxylation produces substrates for reductive amination through phenylalanine ammonia lyase, resulting in a diverse library of fluorinated aromatic l-α-amino acid products with a relative high yield and high enantiomeric purity (up to 99%). Furthermore, we synthesized three pharmaceutically relevant fluorinated l-α-amino acids on a preparative scale using whole-cell biocatalysts, demonstrating the versatility of the approach. This study has introduced a simplified, efficient, and cost-effective route for synthesizing fluorinated l-α-amino acids under mild conditions, offering insights into the production of valuable fluorine-containing pharmaceuticals and other biologically active compounds.

Monitoring nitrite in water and food is critical for ensuring human health. Herein, an electrochemical sensor was constructed for determining nitrite based on iodide probe and gold nanoparticles-poly(L-arginine)-poly(L-lysine) (AuNPs-PArg-PLys) modified electrode. The AuNPs-PArg-PLys nanocomposites were fabricated by one-step in-situ electrochemical synthesis method. Electrochemical detection of nitrite was carried out using square-wave voltammetry. With the assistance of iodide probe, the minimum detectable nitrite concentration reduced remarkably by five orders of magnitude when compared to that without this probe. The iodide-assisted AuNPs-PArg-PLys-based electrochemical sensor could determine nitrite from 1.0 nM to 10.0 μM with a detection limit of 271.8 pM. This sensor possessed the advantages of simplicity, rapidness, low detection limit, wide response range, excellent reproducibility, and high selectivity towards nitrite, which was successfully utilized to measure nanomolar-scale nitrite in tap water and beef. The proposed sensor offered potential applications in environmental protection and food safety.

The flowers ofSophora japonicaL. (SJF) andRobinia pseudoacaciaL. (RPF) are edible and similar in appearance. The chemical constituents of SJF and RPF were compared by UPLC-Q-TOF-MS/MS and HPLC analysis in this study. A total of 29 and 19 constituents were identified in SJF and RPF, respectively. Flavonoid glycosides were the main constituents found in both flowers. The main aglycon moieties found in SJF were quercetin, kaempferol and isorhamnetin, whereas acacetin and kaempferol were the main ones found in RPF. Additionally, the content of flavonoids in SJF was significantly higher than that in RPF, as determined by HPLC. Rutin was the most dominant flavonoid in SJF with a content range of 72.31~88.15 mg/g, followed by quercetin (13.05~20.30 mg/g). Kaempferol-di(rhamnoside)-hexoside was the most dominant flavonoid in RPF with a content range of 25.94~30.00 mg/g. The distinct flavonoid profiles indicated the chemical non-equivalence of SJF and RPF. Therefore, RPF should not be considered a direct substitute for SJF in herbal medicine without further pharmacological and clinical validation.

A novel colorimetric sensing platform was developed for rapid detection of L-cysteine (L-Cys) in human serum and milk samples, utilizing L-glutamic acid-coated Fe 3 O 4 quantum dots (Glu-Fe 3 O 4 QDs) with enhanced peroxidase-like activity. The Glu-Fe 3 O 4 QDs were synthesized via a two-step hydrothermal method, where L-glutamic acid modification significantly improved both catalytic performance and stability compared to unmodified Fe 3 O 4 nanoparticles. Characterization by TEM, XPS, and FTIR confirmed the ultrasmall size (2.57 ± 0.78 nm) and successful surface functionalization with carboxyl/amino groups. The nanozymes catalyzed the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) in the presence of H 2 O 2 , generating a blue color change measurable at 652 nm. L-Cys suppressed this reaction by competitively consuming hydroxyl radicals, enabling quantitative detection with a linear range of 5–50 µM (R 2 = 0.997) and a low detection limit of 0.73 µM. The method exhibited high selectivity against interfering amino acids and ions, and achieved recoveries of 96.99–107.59% in spiked serum and milk samples with RSDs < 3.86%. This work provides a simple, cost-effective tool for monitoring L-Cys in complex biological matrices, with potential applications in food safety and clinical diagnostics. Biodegradable poly-(lactide-coε-caprolactone) (PLCL) scaffolds have opened new perspectives for tissue engineering due to their nontoxic and fascinating functionality. Herein, a black phosphorus-based biodegradable material with a combination of promising enhanced hydrophilicity , shape recovery and osteodifferentiation properties was proposed. First, amino black phosphorous (BP-NH 2 ) was prepared by a simple ball milling method. Then, L -lysine-modified black phosphorous ( L -NH-BP) was formed by hydrogen bonding between L -lysine and amino BP and integrated into PLCL to form PLCL/ L -NH-BP composite fibers . The scaffolds had excellent shape recovery and shape fixity properties. Moreover, based on gene expression and protein level assessment, the scaffolds could enhance the expression of alkaline phosphatase (ALP) and bone morphogenetic protein 2 (BMP2), simultaneously improving the mineralization ability of bone mesenchymal stem cells. Specifically, this new composite material was experimentally verified to be degradable under mild conditions. This strategy provided new insight into the design of multifunctional materials for diverse applications.

Toxicity of engineered nanoparticles on organisms is of concern worldwide due to their extensive use and unique properties. The impacts of ZnO nanoparticles (ZnO NPs) on seed germination and root elongation of corn ( Zea mays L.) and cucumber ( Cucumis sativus L.) were investigated in this study. The role of seed coats of corn in the mitigation toxicity of nanoparticles was also evaluated. ZnO NPs (1,000 mg L −1 ) reduced root length of corn and cucumber by 17 % ( p < 0.05) and 51 % ( p < 0.05), respectively, but exhibited no effects on germination. In comparison with Zn 2+ , toxicity of ZnO NPs on the root elongation of corn could be attributed to the nanoparticulate ZnO, while released Zn ion from ZnO could solely contribute to the inhibition of root elongation of cucumber. Zn uptake in corn exposed to ZnO NPs during germination was much higher than that in corn exposed to Zn 2+ , whereas Zn uptake in cucumber was significantly correlated with soluble Zn in suspension. It could be inferred that Zn was taken up by corn and cucumber mainly in the form of ZnO NPs and soluble Zn, respectively. Transmission electron microscope confirmed the uptake of ZnO NPs into root of corn. Although isolation of the seed coats might not be the principal factor that achieved avoidance from toxicity on germination, seed coats of corn were found to mitigate the toxicity of ZnO NPs on root elongation and prevent approximately half of the Zn from entering into root and endosperm.

Control of dimensional stability is crucial for using electrospun membranes in biomedical and tissue engineering applications. In this study, the contraction of electrospun membranes of poly( l -lactide-co-glycolide) (PLLGA) after stretch was investigated. Then, by adding PLLGA-b-polyethylene glycol (PLLGA-PEG), the contraction of the membranes was controlled. In addition, the mechanical properties of the membranes were investigated. On the basis of the strain–stress behavior of the membranes, an elongation of 50 % was used to investigate the orientation degrees of the membrane fibers. On the basis of the morphology of the stretched membranes, the contraction was found to depend on the orientation degree of fibers after stretch. A linear relationship between the contraction and degree of orientation after stretch was confirmed. Two-dimensional fast Fourier transform (2D FFT) analysis of the scanning electron microscopy (SEM) images of the membranes was used to quantify the orientation degree. It was surmised that the different orientation degrees were responsible for the different morphologies and thermal behaviors of the membranes.

Chronic obstructive pulmonary disease (COPD), a condition primarily linked to oxidative stress, poses significant health burdens worldwide. Recent evidence has shed light on the association between the dysbiosis of gut microbiota and COPD, and their metabolites have emerged as potential modulators of disease progression through the intricate gut-lung axis. Here, we demonstrate the efficacy of oral administration of the probiotic Pediococcus pentosaceus SMM914 (SMM914) in delaying the progression of COPD by attenuating pulmonary oxidative stress. Specially, SMM914 induces a notable shift in the gut microbiota toward a community structure characterized by an augmented abundance of probiotics producing short-chain fatty acids and antioxidant metabolisms. Concurrently, SMM914 synthesizes L-tryptophanamide, 5-hydroxy-L-tryptophan, and 3-sulfino-L-alanine, thereby enhancing the tryptophan-melatonin pathway and elevating 6-hydroxymelatonin and hypotaurine in the lung environment. This modulation amplifies the secretion of endogenous anti-inflammatory factors, diminishes macrophage polarization toward the M1 phenotype, and ultimately mitigates the oxidative stress in mice with COPD. The demonstrated efficacy of the probiotic intervention, specifically with SMM914, not only highlights the modulation of intestine microbiota but also emphasizes the consequential impact on the intricate interplay between the gastrointestinal system and respiratory health.

In this work, we proposed one simple and effective chiral-sensing method for the discrimination of carnitine enantiomers. The gold nanorods (AuNRs) were used as colorimetric probes to recognize carnitine enantiomers in this method. l -Carnitine can induce rapidly the blue-to-gray change of AuNR solution color, whereas d -carnitine cannot induce the obvious change of the color. Based on this distinct difference of color, a visual method for recognition of carnitine enantiomers was suggested. This method has been applied to detect the enantiometric excess of l -carnitine in the range of − 100~100%. The presented chiral measurement can be performed with a simple ultraviolet-visible spectrometer and even the naked eyes. In this method, AuNRs need no modification with chiral molecule, and the chiral recognition is based on the inherent chirality of AuNRs. This new method opens up possibility for fast and easy assay of carnitine enantiomers.

Learning from nature emerges as one of the most promising ways to develop advanced functional materials and biodevices. Here, inspired by melanin formation, we report the tyrosinase (Tyr)-catalyzed polymerization of L-DOPA (versus L-tyrosine and dopamine) to immobilize enzymes for amperometric biosensing. The enzymatic polymerization is examined by UV-vis spectrophotometry, scanning electron microscopy and electrochemical methods. A poly(L-DOPA) (PD)-Tyr/glassy carbon electrode (GCE) prepared by casting an aqueous mixture of L-DOPA and Tyr on a GCE exhibits a linear cathodic amperometric response to catechol concentration from 0.4 to 57 μM (R2 = 0.997) with a sensitivity of 4.29 mA mM−1 cm−2 and a limit of detection (LOD) of 70 nM (S/N = 3). A PD-glucose oxidase (GOx)-Tyr/Pt electrode prepared by casting an aqueous mixture of L-DOPA, GOx and Tyr on a Pt electrode exhibits a linear anodic amperometric response to glucose concentration from 2 to 5700 μM (R2 = 0.998) with a sensitivity of 78.6 μA mM−1 cm−2 and a LOD of 0.1 μM (S/N = 3). The PD-based enzyme electrode shows better biosensing performance and higher bioactivity of the immobilized Tyr than those based on similarly biosynthesized poly(L-tyrosine) and polydopamine as well as well-established chitosan and Nafion systems, implying that the biosynthesized PD as a melanin-biomimetic material has promising application potential for biomacromolecular immobilization and biosensing.
