New features of edge-selectively hydroxylated graphene nanosheets as NIR-II photothermal agent and sonothermal agent for tumor therapy

Wenqian Zhang,Man Fan,Ruchao Yang,Zhihao Li,Yuzhi Qiu,Mengna Dong,Peng Song,Nan Wang,Yajiang Yang,Qin Wang

Second near-infrared (NIR-II) laser-mediated photothermal therapy and sonothermal therapy using low-intensity focused ultrasound exposure for tumors have attracted increasing attention owing to their ability to penetrate deep tissues and provide noninvasive ablation with high therapeutic efficacy. However, their applications were limited by the shortness of optimal NIR-II photothermal agents and sonothermal agents. In this study, we discovered that the edge-selectively hydroxylated graphene nanosheets (EHG NSs) with excellent water dispersibility and an “intact conjugated plane” were not only an outstanding NIR-II photothermal agent but also an effective sonothermal agent for tumor therapy. EHG NSs were incorporated into an injectable adhesive thermosensitive hydrogel with a characteristic sol–gel phase transition behavior. EHG NSs endowed the injectable hydrogel with an exceptional photothermal effect under the laser irradiation (1064 nm, 1.0 W cm−2) as well as an effective sonothermal effect under ultrasonic exposure (3.0 MHz, 2.1 W cm−2), effectively killing tumor cells in vitro and inhibiting tumor growth after intratumoral injection. Especially, the NIR-II photothermal therapy based on the hybrid hydrogel completely ablated the primary tumors and effectively activated systemic anti-tumor immune responses benefiting from the protein adsorption capacity of the injectable hydrogel, significantly inhibiting the growth of the distal tumors. Collectively, EHG nanosheets loaded in the injectable hydrogel will be a promising “all-rounder” for noninvasive deep penetrating thermotherapy and a potent platform that integrates various therapies.

Growth of NIR-II-Responsive Gold Nanobipyramids with Improved Yield and a Study of Their Application in Cancer Therapy

Yujie Ma,Liangbin Xiong,Wenbin Qiu,Zhixin Dong,Xindong Wei,Jilei Li,Yi Fu,Jie Xiong,Hailong Wu

Gold nanobipyramids (Au NBPs), as a novel and emerging plasmonic nanomaterial, have shown larger local electric field enhancements, larger optical cross-sections, and higher refractive index sensitivity compared to other plasmonic nanoparticles. Besides, their localized surface plasmon resonance (LSPR) absorption can be tailored to cover the entire NIR-II light region, which is crucial for clinical cancer treatment. However, strategies to synthesize Au NBPs with an NIR-II light response still have several disadvantages, such as tedious procedures and the use of hazardous chemical substances. In this work, we used a seed-mediated growth method and explored new strategies to synthesize high-yield Au NBPs with an NIR-II light response, especially in the region above 1030 nm, by regulating the conditions during Au seed growth. The yield of Au NBPs with a response at 1060 nm has been improved by 5-fold and 6-fold compared to previous seed-mediated growth methods when 230 μL of sodium citrate or a controlled temperature of 45 °C were used for Au seed growth with an Au seed solution containing 125 μL of NaBH4. The influence mechanism of these factors on Au seed growth and the yield of Au NBPs has been explored in detail. Furthermore, the photodynamic and photothermal behaviors of Au NBPs with LSPR absorption at 1060 nm have been investigated, and the results indicate that Au NBPs have great potential in photothermal therapy. Besides, Au NBPs have been proven to induce immunogenic cell death (ICD), indicating their potential in immunotherapy. This work may provide a new idea for the synthesis and biomedical applications of Au NBPs with an NIR-II response.

A hemostatic carbon foam prevents postoperative tumor recurrence and metastasis via NIR-II light triggered drug release and hyperthermia

Xiuxia Wang,Shide Dong,Xinxi Li,Min Wang,Chunjie Yan,Huaming Yang,Yuting Gao

Postoperative tumor recurrence and metastasis have long been challenging issues for cancer patients. In this work, we have developed an implantable carbon foam that uniquely integrates second near-infrared (NIR-II) light triggered drug release, photothermal therapy, and rapid hemostasis by cross-linking a drug-loaded agarose gel with porous carbon foam (DOX@AG/CF). The functionalized carbon foam possessed outstanding photothermal conversion performance, achieving a rapid temperature rise exceeding 108 °C in just 4 s under a low power intensity of 0.25 W cm −2 with 1064 nm laser irradiation, thereby minimizing the risk of severe skin damage. Furthermore, DOX@AG/CF can regulate the release of the loaded drug though the temperature-responsive agarose gel, which synergistically facilitates postoperative therapy. As a result, such all-in-one gel/carbon foam composite, integrating postoperative hemostasis, photothermal therapy and photocontrolled chemotherapy, can effectively eradicate residual tumor cells and prevent the recurrence and metastasis of cancer in vivo . More importantly, the implanted carbon foam demonstrated excellent degradability and biocompatibility, presenting great potential in clinical application.

Ultrasound-propelled Janus Au NR-mSiO2 nanomotor for NIR-II photoacoustic imaging guided sonodynamic-gas therapy of large tumors

Ye Jiamin,Fu Qinrui,Liu Luntao,Chen Lanlan,Zhang Xuan,Li Qingqing,Li Zhi,Su Lichao,Zhu Rong,Song Jibin,Yang Huanghao

Varieties of contrast agents have been developed for photoacoustic (PA) and ultrasound (US) imaging of cancers in vivo . However, access of traditional contrast agents into the sites of tumors has been principally through passive infiltration without any external force, preventing their deep penetration into the tissues of the tumors, and hindering the use of PA and US for deep tumor imaging. The concept of micro/nanomotors has been the focus of increasing attention as active theranostic agents due to their active movement in particular fluids, thereby conducting assigned tasks. Herein, US-propelled Janus mesoporous SiO 2 partially coated gold nanorods (Au NR-mSiO 2 ) were fabricated for deep tumor NIR-II PA imaging and synergistic sonodynamic-gas therapy. Following US irradiation, 2,2-azobis[2-(2-imidazolin-2-yl) propane] dihydrochloride (AIPH) loaded in mSiO 2 (Au NR-mSiO 2 /AIPH) generated N 2 microbubbles with high efficiency to achieve nanomotor drive. Due to the deep penetration of US, the nanomotors exhibited a capability to travel deep within sites of tumors, providing enhanced PA/US imaging inside the tumors. Furthermore, the nanomotor based cancer therapy was demonstrated through synergistic N 2 gas and sonodynamic therapy. The US-propelled nanomotors demonstrated a novel strategy for the simultaneous PA/US dual imaging deep within tumor tissues and precise therapy of large tumors.

NIR-II Regulation of Mitochondrial Potassium Channel with Dual-Targeted Conjugated Oligomer Nanoparticles for Efficient Cancer Theranostics In Vivo

Chaoqun Li,Dong Gao,Yijian Gao,Ran Zhang,Xiongwei Qu,Shengliang Li,Chengfen Xing

Cell fate can be efficiently modulated by switching ion channels. However, the precise regulation of ion channels in cells, especially in specific organelles, remains challenging. Herein, biomimetic second near-infrared (NIR-II) responsive conjugated oligomer nanoparticles with dual-targeted properties are designed and prepared to modulate the ion channels of mitochondria to selectively kill malignant cells in vivo. Upon 1060 nm laser irradiation, the mitochondria-located nanoparticles photothermally release a specific ion inhibitor of the potassium channel via a temperature-sensitive liposome, thus altering the redox balance and pathways of mitochondria. NIR-II responsive nanoparticles can effectively regulate the potassium channels of mitochondria and fully suppress tumor growth. This work provides a new modality based on the NIR-II nanoplatform to regulate ion channels in specific organelles and proposes an effective therapeutic mechanism for malignant tumors.

A novel TMTP1-modified theranostic nanoplatform for targeted in vivo NIR-II fluorescence imaging-guided chemotherapy for cervical cancer

Nuernisha Alifu,Rong Ma,Lijun Zhu,Zhong Du,Shuang Chen,Ting Yan,Gulinigaer Alimu,Linxue Zhang,Xueliang Zhang

Near-infrared II (NIR-II, 900–1700 nm) fluorescence bioimaging with advantages of good biosafety, excellent spatial resolution, high sensitivity, and contrast has attracted great attention in biomedical research fields. However, most of the nanoprobes used for NIR-II fluorescence imaging have poor tumor-targeting ability and therapeutic efficiency. To overcome these limitations, a novel NIR-II-emissive theranostic nanoplatform for fluorescence imaging and treatment of cervical cancer was designed and prepared. The NIR-II-emissive dye IR-783 and chemotherapy drug doxorubicin (DOX) were encapsulated into liposomes, and the tumor-targeting peptide TMTP1 (a polypeptide with a sequence of cyclic ASN Val Val Arg Gln Cys) was conjugated to the surface of the liposomes to form IR-783-DOX-TMTP1 nanoparticles (NPs) via self-assembly methods. The IR-783-DOX-TMTP1 NPs showed strong NIR-II emission, excellent biocompatibility and a long lifetime in vivo. Furthermore, high-definition NIR-II fluorescence microscopy images of ear blood vessels and intratumoral blood vessels were obtained from IR-783-DOX-TMTP1 NP-stained mice with high spatial resolution under 808 nm laser excitation. Moreover, IR-783-DOX-TMTP1 NPs showed strong tumor-targeting ability and highly efficient chemotherapeutic characteristics towards cervical tumors. The novel targeting and NIR-II-emissive IR-783-DOX-TMTP1 NPs have great potential in diagnosis and therapy for cervical cancer.

Self-Calibrated Identification of Metastatic Lymph Nodes Using Full-NIR-II Tunable Ag2Se Quantum Dots Engineered by Short-Chain Phosphines

Wei Ge,Beibei Gao,Shuxian Li,Guangbo Yu,Lu Gao,Zhemin Shen,Fu Wang,Junle Qu

Ratiometric fluorescence imaging in the second near-infrared (NIR-II) region holds great promise for the accurate identification of metastatic lymph nodes (MLNs), but faces challenges in biocompatible probe design with simplified composition and broad spectral tunability. Here, a minimalist, self-calibrating ratiometric probe based on homologous Ag 2 Se quantum dots (QDs) is developed. Through rational redesign of tributylphosphine-mediated coordination kinetics, continuous size-dependent emission tuning across 935–1710 nm, with the synthetic mechanism and structure–property relationships systematically elucidated, is achieved. The inherently low-toxicity Ag 2 Se core, combined with poly(amino acid) coating, ensures excellent biosafety, as validated by in vitro cytotoxicity and in vivo biochemical assays. Two spectrally distinct QDs (emitting at 1040 and 1305 nm, with full width at half maximum of 176 and 119 nm) enable multiplexed anatomical imaging, allowing both real-time intraoperative detection and postoperative assessment of MLNs, with a diagnostic threshold ratio of ≈0.9. This work presents a scalable strategy for full-NIR-II monochromatic QD synthesis and establishes a minimal-component biocompatible platform for precision ratiometric diagnostics.

Aggregation-Enhanced Photophysical Performance of D-π-A Structured Hemicyanine for NIR-II Fluorescent and Photoacoustic Imaging-Guided Photothermal Therapy

Baoling Li,E Pang,Shaojing Zhao,Guowei Deng,Shuodong Wang,Benhua Wang,Jieyun Wu,Guangle Niu,Xiangzhi Song,Minhuan Lan

Near-infrared (NIR)-II fluorescence and photoacoustic (PA) dual-model imaging-guided photothermal therapy (PTT) can precisely diagnose and treat tumors and evaluate the therapeutic efficacy in real-time. Herein, we utilized a donor-π-acceptor (D-π-A) structured hemicyanine dye (named M1) with a large conjugated structure and strong intramolecular charge transfer effect and demonstrated that the aggregation of M1 could significantly enhance its photophysical performance by improving its photostability and photothermal conversion capability as compared with M1 in a single molecular state. Furthermore, we prepared water-dispersible NIR-II fluorescent nanoparticles (M1 NPs) by wrapping M1 with DSPE-PEG2000-NH2. The obtained M1 NPs exhibit strong NIR-I absorption and NIR-II fluorescence with their maxima at 734 and 1040 nm, respectively, with a fluorescence quantum yield of 2.84%. Moreover, M1 NPs also exhibit excellent biocompatibility, good photostability, and high photothermal conversion efficiency of 77.5%. In vitro and in vivo experiments reveal that M1 NPs can effectively image tumors through NIR-II fluorescence and PA signals, inhibit DNA replication, trigger cytoskeleton collapse, and eventually induce tumor cell apoptosis under 808 nm laser irradiation. Based on these outstanding properties, the application of M1 NPs in PA and NIR-II fluorescence imaging-guided PTT of tumors is demonstrated.

GSH-Activatable Rare-Earth Nanoprobe for NIR-II Fluorescence Imaging-Guided Surgery and Enhanced Chemotherapy/Chemodynamic Therapy of Breast Cancer

Lin-Ling Lin,Jing-Wen Bai,Liang Song,Kang-Liang Lou,Yi-Yang Gao,Hong-Yu Chen,Sheng-Jie Lin,Cheng-Xi Li,Yun Zhang,Guo-Jun Zhang

The precise intraoperative delineation of tumor margins and the controlled intratumoral release of chemotherapeutic agents remain major challenges in breast cancer therapy. Tumor microenvironment (TME)-responsive nanoplatforms provide a promising approach by utilizing the distinctive biochemical properties of tumors to achieve targeted imaging and treatment. Herein, we developed a glutathione (GSH)-activatable theranostic nanoprobe (DSM-cRGD) emitting fluorescence in the second near-infrared window (NIR-II). The probe is constructed from Nd-doped rare-earth nanoparticles coated with a manganese dioxide (MnO 2 ) shell and functionalized with cyclic RGD peptide for α v β 3 -integrin targeting. The MnO 2 shell quenches NIR-II fluorescence during systemic circulation and undergoes GSH-responsive degradation within the TME. In addition, the nanoprobe shows strong targetability within tumors via specific binding to α v β 3, thereby enabling real-time, high-contrast surgical navigation. Moreover, a newly developed therapeutic agent, DSM-cRGD@DOX, formed by loading doxorubicin (DOX) onto DSM-cRGD, enables the simultaneous release of DOX and Mn 2+ ions upon degradation of the MnO 2 shell in the tumor microenvironment (TME), thereby inducing synergistic chemodynamic and chemotherapeutic effects. This further induces immunogenic cell death, activating anti-tumor immune responses and suppressing tumor recurrence and metastasis. Collectively, this DSM-cRGD-based platform represents an integrated nanoplatform that combines precise tumor imaging with stimuli-responsive drug delivery, offering an advanced strategy for the precision management of breast cancer.

Engineering Mg-doped CoMo-LDH as an efficient sensitizer for NIR-II light-enhanced photo-sonodynamic therapy and immune activation

Zhang Huifeng,Li Mengyang,Wang Tao,Zhu Huifen,Deng Lin,Guo Fulian,Zhang Zhihua,Hu Tingting,Liang Ruizheng,Cao Min,Li Zhangping

Cancer remains a critical global health burden, posing a severe threat to human life. Photo-sonodynamic therapy (PSDT), which combines photodynamic (PDT) and sonodynamic (SDT) modalities, has emerged as a promising strategy to overcome limitations in tissue penetration and therapeutic precision. Despite progress, the broader application of PSDT is hindered by PDT’s potential phototoxicity/limited therapeutic penetration depth and SDT’s thermal/mechanical damage under high ultrasound (US) power, as well as the complex synthesis of sensitizers and their suboptimal reactive oxygen species (ROS) generation. Here, we engineer Mg-doped defect-rich CoMo-LDH (a-CoMgMo-LDH) nanosheets via a doping-etching strategy to address these challenges, enabling on-demand treatment from superficial to deep-seated tumors. This sensitizer exhibits exceptional dual responsiveness to NIR-II light and US. It achieves a relative singlet oxygen quantum yield (1.53) superior to most reported photosensitizers, while its sonodynamic ROS yield surpasses that of commercial TiO 2 by 22.3 times and outperforms most reported sonosensitizers, ranking among the highest reported. After bovine serum albumin (BSA) functionalization, a-CoMgMo-LDH-BSA triggers ROS-driven apoptosis and immunogenic cell death under dual irradiation, effectively suppressing tumor growth and metastasis. By integrating NIR-II-enhanced PDT/SDT into a single-component platform, this work advances precision oncology with deep-tumor targeting and immune modulation, offering a transformative strategy for clinical translation. Graphical Mg-doped defect-rich CoMo-LDH (a-CoMgMo-LDH) nanosystem is engineered through doping-etching strategy as a new photo-sonosensitizer for highly efficient photo-sonodynamic therapy, which exhibits remarkable ROS generation efficiency under NIR-II/US stimulation, achieving a 1 O 2 quantum yield of 1.53 under NIR-II light irradiation and 22.3 times ROS yields that of commercial TiO 2 under US irradiation, respectively. After bovine serum albumin (BSA) modification, a-CoMgMo-LDH-BSA efficiently kills cancer cells and eliminates tumors by inducing dual-modal ROS generation and immunogenic cell death

Magnetically navigated and PSMA/STEAP1 dual-targeted nanoliposomes for NIR-II/MRI imaging with photothermal-chemo-immunotherapy for prostate cancer

Jian-Xuan Sun,Ye An,Si-Han Zhang,Jin-Zhou Xu,Abudureyimu Maimaiti,Jing-Yu Xu,Yi-Fan Huang,Yu-Xuan yang,Mei-Cheng Liu,Shao-Gang wang,Qi-Dong Xia

Precision theranostics for castration-resistant prostate cancer remain a major challenge because of tumor heterogeneity, drug toxicity, and resistance to conventional therapies. Here, we report a multifunctional theranostic nanoplatform that integrates multimodal imaging guidance, dual-targeted delivery, photothermal therapy, and immune activation for the precision treatment of prostate cancer. By encapsulating aggregation-induced emission luminogens and magnetic nanoparticles, this system achieves superior second near-infrared fluorescence and magnetic resonance imaging fusion for deep-tissue visualization. The dual-targeting strategy, which integrates magnetic navigation with simultaneous engagement of six-transmembrane epithelial antigen of the prostate 1 and prostate specific membrane antigen (PSMA), effectively addresses tumor heterogeneity by enabling externally guided and biologically specific delivery to both PSMA+ and PSMA− prostate cancer cells. Upon targeted delivery, the synergistic combination of photothermal therapy and chemotherapy achieves potent tumor growth inhibition, activates CD8+ T cells and macrophages to promote robust antitumor immune responses, and elicits durable immunological memory to prevent tumor recurrence. In vitro and in vivo studies demonstrate superior tumor targeting specificity, enhanced imaging contrast, and minimal off-target toxicity. This study provides novel insights into the construction of a multifunctional theranostic nanoplatform in precision oncology, with promising translational potential for clinical management of castration-resistant prostate cancer.

Preparation of an activatable benzothiadiazole-based nanoprobe for multispectral optoacoustic and NIR-II fluorescence dual-mode imaging of liver injury

Wu Yinglong,Zhang Chaobang,Chen Junjie,Zeng Fang,Wu Shuizhu,Zhao Yanli

Light-excited dual-mode imaging that integrates multispectral optoacoustic tomography (MSOT) and near-infrared region II fluorescence (NIR-II FL) imaging allows complementary deep-tissue visualization with high anatomical resolution and molecular sensitivity, thereby enhancing the accuracy of biomedical diagnostics. Activatable probes further advance this approach by producing signals upon recognition of disease-related biomarkers, which reduces background interference and improves imaging specificity. BTPE-NO 2 @F127, a benzothiadiazole-based nanoprobe, exhibits selective activation of both optoacoustic and NIR-II FL signals in response to hydrogen peroxide (H 2 O 2 ), a crucial early stage biomarker of liver injury, thus permitting cross-validated detection of hepatic damage in vivo with excellent signal-to-background ratio contrast. Here we present detailed procedures for preparation of the BTPE-NO 2 @F127 nanoprobe and its applications in MSOT/NIR-II FL dual-mode imaging of trazodone- or ischemia–reperfusion-induced liver injury in mice. Compared to conventional liver injury diagnostic methods, such as invasive tissue biopsy, ex vivo blood analysis and previously reported fluorescent/optoacoustic probes, BTPE-NO 2 @F127 offers real-time, in situ monitoring with high sensitivity and signal-to-background ratio, as well as mutually corroborating signals for increased reliability. The fabrication of BTPE-NO 2 @F127, including the chemical synthesis and characterization, requires ~17 d, while the in vitro validation of its H 2 O 2 responsiveness takes ~5 d. Notably, the complete workflow of data acquisition and analysis for MSOT/NIR-II FL dual-mode imaging of liver injury in mice using BTPE-NO 2 @F127 can be accomplished within 10 h. The protocol is easy to follow and suitable for clinicians and researchers with a basic understanding of chemistry and bioimaging techniques.
