Platinum Nanoparticles Regulated V2C MXene Nanoplatforms with NIR-II Enhanced Nanozyme Effect for Photothermal and Chemodynamic Anti-infective Therapy

Xiaojun He,Ya Lv,Yanling Lin,Hong Yu,Yipiao Zhang,Yuhua Tong,Chunwu Zhang

Given the challenge of multidrug resistance in antibiotics, non-antibiotic-dependent antibacterial strategies show promise for anti-infective therapy. V 2 C MXene-based nanomaterials have demonstrated strong biocompatibility and photothermal conversion efficiency (PCE) for photothermal therapy (PTT). However, the limitation of V 2 C MXene's laser irradiation to the near-infrared region I (NIR-I) restricts tissue penetration, making it difficult to achieve complete bacterial eradication with single-effect therapeutic strategies. To address this, Pt nanoparticles (Pt NPs) were attached to V 2 C, forming artificial nanoplatforms (Pt@V 2 C). Pt@V 2 C exhibited enhanced PCE (59.6%) and a longer irradiation laser (NIR-II) due to the surface plasmon resonance effect of Pt NPs and V 2 C. Notably, Pt@V 2 C displayed dual enzyme-like activity with chemodynamic therapy (CDT) and NIR-II enhanced dual enzyme-like activity. The biocatalytic mechanism of Pt@V 2 C was elucidated using density functional theory. In an in vivo animal model, Pt@V 2 C effectively eliminated methicillin-resistant Staphylococcus aureus from deep-seated tissues in subcutaneous abscesses and bacterial keratitis environments, accelerating abscess resolution and promoting wound and cornea healing through the synergistic effects of PTT/CDT. Transcriptomic analysis revealed that Pt@V 2 C targeted inflammatory pathways, providing insight into its therapeutic mechanism. This study presents a promising therapeutic approach involving hyperthermia-amplified biocatalysis with Pt NPs and MXene nanocomposites. This article is protected by copyright. All rights reserved

Noninvasive Detection and Monitoring of the Integrity of the Intestinal Barrier through NIR-II Fluorescence Imaging and Colorimetric Urinalysis

Li Zhang,Lei Feng,Lin Yang,Hao Wan

The variation of the integrity of the intestinal barrier is closely related to the occurrence and progression of various diseases, making its accurate detection and monitoring essential to provide reliable information for guiding the refinement of treatment scenarios. Herein, we developed a strategy utilizing glutathione-capped gold clusters (Au-GSH) to achieve simultaneous in vivo second near-infrared (NIR-II) fluorescence imaging and in vitro colorimetric urinalysis for noninvasive detection and monitoring of intestinal barrier integrity. After oral administration, Au-GSH can demonstrate different distribution behaviors in terms of the variation of intestinal barrier integrity. Specifically, Au-GSH could selectively permeate through compromised intestinal barrier to be distributed into the bladder in view of its ultrasmall size below the glomerular filtration cutoff, leading to the rapid excretion through urine. Such process can be visually profiled by collecting NIR-II fluorescence (>1100 nm) emitting from Au-GSH in a noninvasive real-time manner. By virtue of the peroxidase-like activity of Au-GSH, the simple colorimetric urinalysis was further established for evaluating the integrity of the intestinal barrier, advancing the detection and monitoring performance. As a result, our developed strategy successfully detected the damage of intestinal barrier integrity in ulcerative colitis (UC) mice, a common disease characterized by compromised intestinal barrier integrity. Excitingly, the monitoring of restoration of intestinal barrier integrity in both therapeutic and preventative modes for UC mice was also realized by our strategy, making it a promising diagnosis scenario for diseases related to the variation of intestinal barrier integrity.

NIR-II-amplify high-entropy MXene-based sonosensitizer as sonodynamic therapy promotes methicillin-resistant Staphylococcus aureus–Infected wound healing

Wei Song,Danyan Wang,Shuai Xiao,Xiaojun He,Wei Xiong,Jianliang Shen

The growing pace of two-dimensional (2D) transitional metal carbides and nitrides (MXenes) has become a spotlight for recent research. Similarly, the need to close the gap left by conventional antibiotics, fighting off bacterial infections, and drug resistance has become eminent. Especially, MXenes are widely used as excellent photothermal agents. However, there are limited reports on MXenes as sonosensitizer. Herein, a high-entropy MXene (HEM), TVNMC was fabricated for the generation of reactive oxide species (ROS) against Methicillin-resistance Staphylococcus aureus (MRSA). Interestingly the energy storage and conversion from the diverse composition of the TVNMC MXenes allowed the generation of ROS under US irradiation and even enhanced under Photothermal therapy. This combinatorial therapy undertook a near-total kill and eradication of MRSA with its biofilm. Moreover, TVNMC MXene simultaneously eliminated MRSA abscesses while promoting wound healing with negligible anomaly to murine tissues under sonodynamic and photothermal therapy, making the TVNMC MXene a promising sonosensitizer for possible biomedical usage. Overall, this system provides great potential for the use of high-entropy MXenes as a new type of sonosensitizer in anti-infection therapy.

Phototheranostic LPP-QDs-IR-820 Nanocomposites for Specific NIR-II Imaging of Lymphatic and Photothermal Therapy of Cervical Tumors

Lijun Zhu,Zhong Du,Jiabao Xiong,Hui Li,Chi Zhang,Xueliang Zhang,Nuernisha Alifu,Biao Dong

Precise theranostics of tumors is intricately linked to the early detection and monitoring of lymph nodes (LN) and metastases, making the targeted localization of LNs essential for tumor identification. However, designing LN-targeting probes remains a significant challenge due to issues such as lymphatic uptake, biocompatibility, and fluorescence stability. To address these challenges, near-infrared II (NIR-II) fluorescence probes are developed through meticulous analysis of LN physiological structure and passive targeting strategy for LN detection and tumor therapy. An LPP-QDs-IR-820 nanocomposite (NCs) is engineered, comprising the IR-820 molecules and ultrabright PbS@CdS quantum dots (QDs), which are encapsulated within a liposome-SH-mPEG 2000 polymer matrix. These NCs demonstrates remarkable lymphatic enrichment, facilitating real-time tracking of LN via electrostatic repulsion and extracellular matrix effects. Importantly, the NCs exhibit negligible in vivo toxicity and high biocompatibility. The intense NIR-II fluorescence emissions of IR-820 and PbS@CdS QDs confer upon the NCs a high NIR-II fluorescence quantum yield (6%). The cervical tumors and their deep microvessels are clearly observed via NIR-II fluorescence imaging. Moreover, the photothermal properties of IR-820 enable the NCs to achieve a photothermal conversion efficiency of 36.56%, leading to effective photothermal therapy in cervical tumor mice.

NIR-II photothermal therapy combined with activatable immunotherapy against the recurrence and metastasis of orthotopic triple-negative breast cancer

Wentao Lei,Yinghui Wang,Tangyue Zheng,Qihang Wu,Hui Wen,Tingting Sun,Jun Liu,Zhigang Xie

It remains a clinical challenge to treat triple-negative breast cancer due to its aggressiveness and metastasis. Photothermal therapy (PTT) in combination with checkpoint blockade immunotherapy offers a promising strategy for such intractable tumors. In this study, a second near-infrared (NIR-II) photothermal agent (PCD NPs) and a tumor microenvironment-activatable nanoprodrug (NLG NPs) for indoleamine 2,3-dioxygenase 1 (IDO-1) blockade have been designed for the therapy of orthotopic triple-negative breast cancer. The NIR-II absorption of PCD NPs can guarantee the high penetration depth of the laser during PTT. At the same time, NLG NPs can be decomposed into the NLG919 monomer in the tumor microenvironment, which can effectively strengthen the immunogenic cell death-induced immune response. NIR-II PTT in synergy with IDO-1 blockade can effectively inhibit tumor growth and prevent tumor recurrence and metastasis. This work thus provides a safe, efficient and feasible method for the treatment of malignant tumors.

New features of edge-selectively hydroxylated graphene nanosheets as NIR-II photothermal agent and sonothermal agent for tumor therapy

Wenqian Zhang,Man Fan,Ruchao Yang,Zhihao Li,Yuzhi Qiu,Mengna Dong,Peng Song,Nan Wang,Yajiang Yang,Qin Wang

Second near-infrared (NIR-II) laser-mediated photothermal therapy and sonothermal therapy using low-intensity focused ultrasound exposure for tumors have attracted increasing attention owing to their ability to penetrate deep tissues and provide noninvasive ablation with high therapeutic efficacy. However, their applications were limited by the shortness of optimal NIR-II photothermal agents and sonothermal agents. In this study, we discovered that the edge-selectively hydroxylated graphene nanosheets (EHG NSs) with excellent water dispersibility and an “intact conjugated plane” were not only an outstanding NIR-II photothermal agent but also an effective sonothermal agent for tumor therapy. EHG NSs were incorporated into an injectable adhesive thermosensitive hydrogel with a characteristic sol–gel phase transition behavior. EHG NSs endowed the injectable hydrogel with an exceptional photothermal effect under the laser irradiation (1064 nm, 1.0 W cm−2) as well as an effective sonothermal effect under ultrasonic exposure (3.0 MHz, 2.1 W cm−2), effectively killing tumor cells in vitro and inhibiting tumor growth after intratumoral injection. Especially, the NIR-II photothermal therapy based on the hybrid hydrogel completely ablated the primary tumors and effectively activated systemic anti-tumor immune responses benefiting from the protein adsorption capacity of the injectable hydrogel, significantly inhibiting the growth of the distal tumors. Collectively, EHG nanosheets loaded in the injectable hydrogel will be a promising “all-rounder” for noninvasive deep penetrating thermotherapy and a potent platform that integrates various therapies.

Defect-engineered PdOv-TiOX nanozymes with NIR-II responsiveness for synergistic photothermal–chemodynamic therapy of drug-resistant bacterial infections

Youpei Wang,Xiang Zheng,Shiyun Liang,Danyan Wang,Xiaojun He,Lingxia Pang

Traditional treatments for drug-resistant bacterial keratitis and pneumonia often prove ineffective due to the presence of biofilm barriers and multidrug resistance. Existing nano-antibacterial systems face several limitations, including insufficient tissue penetration depth in the first near-infrared (NIR I) window, the inability of a single treatment modality to simultaneously disrupt biofilms and eradicate residual bacteria, and a lack of efficacy validation in cross-organ (ocular-pulmonary) infection models. In this study, we employed a defect engineering strategy, to construct a palladium-doped TiO 2 nanozyme with oxygen vacancies (PdO V -TiO X ). This nanozyme achieves multiple antibacterial effects through the synergistic combination of second near-infrared (NIR-II) laser-activated photothermal therapy and chemodynamic therapy. Under 1064 nm laser excitation, the system utilizes oxygen vacancy-enhanced peroxidase-like activity to continuously generate hydroxyl radicals (•OH), resulting in a 5.2-fold increase in radical production compared to dark conditions. Additionally, it induces a local temperature rise (ΔT = 28.2 °C). These combined effects—physical disruption of the biofilm matrix and ROS-mediated oxidative stress—effectively dismantle the defense mechanisms of drug-resistant bacteria. In models of drug-resistant Staphylococcus aureus keratitis and pneumonia, PdO V -TiO X demonstrated therapeutic efficacy in both the eye and lung, indicating the broad cross-organ applicability of this treatment. Under NIR-II irradiation, the PdO V -TiO X nanozyme significantly elevated ROS levels at the infection site, achieving efficient bactericidal action while effectively penetrating and disrupting mature biofilms. This work presents a novel defect-engineering-regulated nanozyme synergistic therapeutic strategy for the treatment of multidrug-resistant bacterial infections.

Engineering Ho3+-Mediated Energy Transfer for Enhanced Pr3+ NIR-II Luminescence in Core–Shell Nanocrystals toward Ultrasensitive Temperature Sensing

Kejie Li,Haikuo Liu,Zhiying Wang,Mengmeng Dai,Jiaqi Zhao,Zuoling Fu

Near-infrared II (NIR-II, 1000–1700 nm) luminescence has emerged as a powerful tool for biomedical applications, including sensing, imaging, and drug delivery, owing to its superior tissue penetration depth and high spatial resolution. Lanthanide-based NIR-II nanoprobes are particularly promising due to their outstanding photostability and minimal cytotoxicity. However, two critical challenges impede their widespread implementation: severe thermal quenching at elevated temperatures and inefficient downshifting processes. Herein, a novel strategy is reported to address these limitations by precisely modulating energy transfer dynamics between Ho 3+ and Pr 3+ ions in NaYF 4 core–shell engineering. By carefully optimizing the concentration ratio and spatial distribution of these lanthanide ions in NaYF 4 core–shell nanocrystals, the population of 1 G 4 (Pr 3+ ) is significantly increased, effectively suppressing thermal quenching while boosting downshifting efficiency. The optimized system leverages dual NIR-II emission channels ( 1 G 4 and 5 I 6 ) in NaYF 4 :Yb 3+ /Ho 3+ @NaYF 4 :Yb 3+ /Pr 3+ nanocrystals, enabling ultrasensitive temperature sensing with a maximum relative sensitivity ( S r ) of 2.03% K −1 . Furthermore, the synergistic up/down-conversion properties of Ho 3+ and Pr 3+ facilitate multimodal anti-counterfeiting, offering a robust platform for advanced information encryption and storage. This work not only establishes a new paradigm for engineering NIR-II luminescence but also underscores their vast potential in advanced applications, from precision biosensing to high-security encoding technologies.

Self-Calibrated Identification of Metastatic Lymph Nodes Using Full-NIR-II Tunable Ag2Se Quantum Dots Engineered by Short-Chain Phosphines

Wei Ge,Beibei Gao,Shuxian Li,Guangbo Yu,Lu Gao,Zhemin Shen,Fu Wang,Junle Qu

Ratiometric fluorescence imaging in the second near-infrared (NIR-II) region holds great promise for the accurate identification of metastatic lymph nodes (MLNs), but faces challenges in biocompatible probe design with simplified composition and broad spectral tunability. Here, a minimalist, self-calibrating ratiometric probe based on homologous Ag 2 Se quantum dots (QDs) is developed. Through rational redesign of tributylphosphine-mediated coordination kinetics, continuous size-dependent emission tuning across 935–1710 nm, with the synthetic mechanism and structure–property relationships systematically elucidated, is achieved. The inherently low-toxicity Ag 2 Se core, combined with poly(amino acid) coating, ensures excellent biosafety, as validated by in vitro cytotoxicity and in vivo biochemical assays. Two spectrally distinct QDs (emitting at 1040 and 1305 nm, with full width at half maximum of 176 and 119 nm) enable multiplexed anatomical imaging, allowing both real-time intraoperative detection and postoperative assessment of MLNs, with a diagnostic threshold ratio of ≈0.9. This work presents a scalable strategy for full-NIR-II monochromatic QD synthesis and establishes a minimal-component biocompatible platform for precision ratiometric diagnostics.

Aggregation-Enhanced Photophysical Performance of D-π-A Structured Hemicyanine for NIR-II Fluorescent and Photoacoustic Imaging-Guided Photothermal Therapy

Baoling Li,E Pang,Shaojing Zhao,Guowei Deng,Shuodong Wang,Benhua Wang,Jieyun Wu,Guangle Niu,Xiangzhi Song,Minhuan Lan

Near-infrared (NIR)-II fluorescence and photoacoustic (PA) dual-model imaging-guided photothermal therapy (PTT) can precisely diagnose and treat tumors and evaluate the therapeutic efficacy in real-time. Herein, we utilized a donor-π-acceptor (D-π-A) structured hemicyanine dye (named M1) with a large conjugated structure and strong intramolecular charge transfer effect and demonstrated that the aggregation of M1 could significantly enhance its photophysical performance by improving its photostability and photothermal conversion capability as compared with M1 in a single molecular state. Furthermore, we prepared water-dispersible NIR-II fluorescent nanoparticles (M1 NPs) by wrapping M1 with DSPE-PEG2000-NH2. The obtained M1 NPs exhibit strong NIR-I absorption and NIR-II fluorescence with their maxima at 734 and 1040 nm, respectively, with a fluorescence quantum yield of 2.84%. Moreover, M1 NPs also exhibit excellent biocompatibility, good photostability, and high photothermal conversion efficiency of 77.5%. In vitro and in vivo experiments reveal that M1 NPs can effectively image tumors through NIR-II fluorescence and PA signals, inhibit DNA replication, trigger cytoskeleton collapse, and eventually induce tumor cell apoptosis under 808 nm laser irradiation. Based on these outstanding properties, the application of M1 NPs in PA and NIR-II fluorescence imaging-guided PTT of tumors is demonstrated.

GSH-Activatable Rare-Earth Nanoprobe for NIR-II Fluorescence Imaging-Guided Surgery and Enhanced Chemotherapy/Chemodynamic Therapy of Breast Cancer

Lin-Ling Lin,Jing-Wen Bai,Liang Song,Kang-Liang Lou,Yi-Yang Gao,Hong-Yu Chen,Sheng-Jie Lin,Cheng-Xi Li,Yun Zhang,Guo-Jun Zhang

The precise intraoperative delineation of tumor margins and the controlled intratumoral release of chemotherapeutic agents remain major challenges in breast cancer therapy. Tumor microenvironment (TME)-responsive nanoplatforms provide a promising approach by utilizing the distinctive biochemical properties of tumors to achieve targeted imaging and treatment. Herein, we developed a glutathione (GSH)-activatable theranostic nanoprobe (DSM-cRGD) emitting fluorescence in the second near-infrared window (NIR-II). The probe is constructed from Nd-doped rare-earth nanoparticles coated with a manganese dioxide (MnO 2 ) shell and functionalized with cyclic RGD peptide for α v β 3 -integrin targeting. The MnO 2 shell quenches NIR-II fluorescence during systemic circulation and undergoes GSH-responsive degradation within the TME. In addition, the nanoprobe shows strong targetability within tumors via specific binding to α v β 3, thereby enabling real-time, high-contrast surgical navigation. Moreover, a newly developed therapeutic agent, DSM-cRGD@DOX, formed by loading doxorubicin (DOX) onto DSM-cRGD, enables the simultaneous release of DOX and Mn 2+ ions upon degradation of the MnO 2 shell in the tumor microenvironment (TME), thereby inducing synergistic chemodynamic and chemotherapeutic effects. This further induces immunogenic cell death, activating anti-tumor immune responses and suppressing tumor recurrence and metastasis. Collectively, this DSM-cRGD-based platform represents an integrated nanoplatform that combines precise tumor imaging with stimuli-responsive drug delivery, offering an advanced strategy for the precision management of breast cancer.

Engineering Mg-doped CoMo-LDH as an efficient sensitizer for NIR-II light-enhanced photo-sonodynamic therapy and immune activation

Zhang Huifeng,Li Mengyang,Wang Tao,Zhu Huifen,Deng Lin,Guo Fulian,Zhang Zhihua,Hu Tingting,Liang Ruizheng,Cao Min,Li Zhangping

Cancer remains a critical global health burden, posing a severe threat to human life. Photo-sonodynamic therapy (PSDT), which combines photodynamic (PDT) and sonodynamic (SDT) modalities, has emerged as a promising strategy to overcome limitations in tissue penetration and therapeutic precision. Despite progress, the broader application of PSDT is hindered by PDT’s potential phototoxicity/limited therapeutic penetration depth and SDT’s thermal/mechanical damage under high ultrasound (US) power, as well as the complex synthesis of sensitizers and their suboptimal reactive oxygen species (ROS) generation. Here, we engineer Mg-doped defect-rich CoMo-LDH (a-CoMgMo-LDH) nanosheets via a doping-etching strategy to address these challenges, enabling on-demand treatment from superficial to deep-seated tumors. This sensitizer exhibits exceptional dual responsiveness to NIR-II light and US. It achieves a relative singlet oxygen quantum yield (1.53) superior to most reported photosensitizers, while its sonodynamic ROS yield surpasses that of commercial TiO 2 by 22.3 times and outperforms most reported sonosensitizers, ranking among the highest reported. After bovine serum albumin (BSA) functionalization, a-CoMgMo-LDH-BSA triggers ROS-driven apoptosis and immunogenic cell death under dual irradiation, effectively suppressing tumor growth and metastasis. By integrating NIR-II-enhanced PDT/SDT into a single-component platform, this work advances precision oncology with deep-tumor targeting and immune modulation, offering a transformative strategy for clinical translation. Graphical Mg-doped defect-rich CoMo-LDH (a-CoMgMo-LDH) nanosystem is engineered through doping-etching strategy as a new photo-sonosensitizer for highly efficient photo-sonodynamic therapy, which exhibits remarkable ROS generation efficiency under NIR-II/US stimulation, achieving a 1 O 2 quantum yield of 1.53 under NIR-II light irradiation and 22.3 times ROS yields that of commercial TiO 2 under US irradiation, respectively. After bovine serum albumin (BSA) modification, a-CoMgMo-LDH-BSA efficiently kills cancer cells and eliminates tumors by inducing dual-modal ROS generation and immunogenic cell death
