Self-Assembled Nanoparticles of Silicon (IV)–NO Donor Phthalocyanine Conjugate for Tumor Photodynamic Therapy in Red Light

Kadireya Aikelamu,Jingya Bai,Qian Zhang,Jiamin Huang,Mei Wang,Chunhong Zhong

The combination of photodynamic therapy (PDT) and pneumatotherapy is emerging as one of the most effective strategies for increasing cancer treatment efficacy while minimizing side effects. Photodynamic forces affect nitric oxide (NO) levels as activated photosensitizers produce NO, and NO levels in the tumor and microenvironment directly impact tumor cell responsiveness to PDT. In this paper, 3-benzenesulfonyl-4-(1-hydroxy ether)-1,2,5-oxadiazole-2-oxide NO donor–silicon phthalocyanine coupling (SiPc–NO) was designed and prepared into self-assembled nanoparticles (SiPc–NO@NPs) by precipitation method. By further introducing arginyl-glycyl-aspartic acid (RGD) on the surface of nanoparticles, NO-photosensitizer delivery systems (SiPc–NO@RGD NPs) with photo-responsive and tumor-targeting properties were finally prepared and preliminarily evaluated in terms of their formulation properties, NO release, and photosensitizing effects. Furthermore, high reactive oxygen species (ROS) generation efficiency and high PDT efficiency in two breast cancer cell lines (human MCF-7 and mouse 4T1) under irradiation were also demonstrated. The novel SiPc–NO@RGD NPs show great potential for application in NO delivery and two-photon bioimaging-guided photodynamic tumor therapy.

Photonic hydrogels combining the slow photon effect and NO gas therapy for synergetic enhanced photodynamic antibacterial therapy

Hui Wang,Duohang Bi,Bowen Yu,Qiang Chen,Shuo Du,Ge Xie,Jintao Zhu,Lianbin Zhang

Photodynamic therapy (PDT) offers potential for combating bacterial infections through the generation of reactive oxygen species (ROS). However, the antibacterial efficiency of PDT is largely impeded by the limited photon absorption of photosensitizers and the short diffusion length and lifespan of ROS. Herein, we present a light-harvesting platform based on l -arginine-modified photonic hydrogels loaded with new indocyanine green (PG@Arg/IR820) for synergizing the slow photon effect with NO gas therapy to enhance PDT antibacterial efficiency. Upon near-infrared (NIR) light irradiation, PG@Arg/IR820 can maximize the utilization of photons via the slow photon effect to generate sufficient ROS, which not only acts as the primary bactericidal agent in PDT but also triggers l -arginine to generate NO. NO exhibits a long diffusion distance and lifespan and can freely diffuse to inhibit distant bacterial growth, demonstrating a vital complementary advantage in bacterial inactivation by ROS. The synergistic effect of the slow photon effect combined with NO gas therapy allows PG@Arg/IR820 to intensify bacterial destruction and enhance PDT antibacterial efficiency. This antibacterial system sheds light on an advisable design principle for efficient antibacterial activities in photodynamic inactivation.

S-scheme Bi12TiO20/Bi4Ti3O12 heterojunction immobilized on 3D-printed support as a monolithic photocatalyst for NO removal

Hongxia Liu,Minggang Zhang,Zhipeng Jin,Hui Mei,Gangqiang Zhu,Longkai Pan,Laifei Cheng,Litong Zhang

Photocatalysis is recognized as one of the most promising techniques for environment purification. However, the inconvenient recovery of powder photocatalysts significantly impeded their practical applications. Immobilization of the photocatalysts on supports to form monolithic photocatalysts is an effective method to solve this problem. Herein, a monolithic photocatalyst Bi 12 TiO 20 /Bi 4 Ti 3 O 12 /SiOC is fabricated by growing optimal Bi 12 TiO 20 /Bi 4 Ti 3 O 12 S-scheme heterojunction onto 3D-printed porous SiOC support. On one side, S-scheme heterojunction facilitates the separation of photoinduced electrons and holes while retaining their strong redox ability. On the other side, the porous structure favors mass transport and improves light harvesting by reflecting the light several times. Under simulated solar light irradiation, the NO removal efficiency of the monolithic photocatalyst Bi 12 TiO 20 /Bi 4 Ti 3 O 12 /SiOC is 16.4 %. Furthermore, the composite has good stability and reusability, suggesting its great potential in practical application. The possible photocatalytic mechanism is proposed according to systematic experiments and density functional theory (DFT). This work provides a rational reference for constructing efficient photocatalysts for environmental purification.

A broad-spectrum antibiotic targets multiple-drug-resistant bacteria with dual binding targets and no detectable resistance

He Wenyan,Huan Xueting,Li Yinchuan,Deng Qisen,Chen Tao,Xiao Wen,Chen Yijun,Ma Lingman,Liu Nan,Shang Zhuo,Wang Zongqiang

The rapid emergence of difficult-to-treat multidrug-resistant pathogens, combined with the scarcity of antibiotics possessing novel mechanisms, poses a significant threat to global public health. Here, we integrate the synthetic-bioinformatic natural product approach with peptide optimization to unveil the antibiotic-producing potential of Paenibacillaceae bacteria. Our culture-independent approach led to the discovery of paenimicin, a novel 11-mer depsi -lipopeptide featuring an unprecedented dual-binding mechanism. By sequestering the phosphate and hydroxyl groups of lipid A in Gram-negative bacteria, as well as the phosphate groups of teichoic acids in Gram-positive bacteria, paenimicin exhibits potent and broad-spectrum efficacy against MDR pathogens in vitro and in vivo models. Paenimicin demonstrates no detectable resistance, favorable pharmacokinetics and low nephrotoxicity, positioning it as a promising candidate for treating severe and urgent MDR infections.

The lead-free halide perovskite/UiO-66 with dual Lewis acid sites for efficient photocatalytic NO removal

Zhijian Xiao,Wenda Yang,Jing Wang,Shuang Cao,Jingling Yang,Mingshan Zhu

Photocatalysis is a promising technology for removing low concentrations of nitric oxide (NO) from the atmosphere. However, the unsatisfactory photocatalytic NO removal efficiency and the high selectivity for NO 2 in catalysts limit the development of this technology. In this work, we used an in-situ synthesis method to incorporate Cs 3 Bi 2 Br 9 into UiO-66 for efficient and robust photocatalytic NO removal. The Cs 3 Bi 2 Br 9 /UiO-66 catalyst exhibits a 77.6 % NO removal efficiency and low NO 2 selectivity under visible light irradiation at a WHSV of 1,650,000 mL h −1  g −1 , exceeding the efficiencies of Cs 3 Bi 2 Br 9 (36.4 %) and UiO-66 (2.3 %), as well as those of most reported MOF-based photocatalysts. The synergy of weak Lewis acid sites from Cs 3 Bi 2 Br 9 in Cs 3 Bi 2 Br 9 /UiO-66 and the strong Lewis acid sites from UiO-66 promotes the enrichment of NO on the catalyst surface and act as electron traps to induce carrier separation, thereby promoting the generation of O 2 − to achieve high efficiency. This work provides a new idea for the development of catalysts in NO elimination.

Extraction, Characterization, and Impact of Polysaccharides From Zhonghuang No. 1 Etiolated-Green Tea on Human Fecal Microbiota

Baoming Tian,Xue Zhou,Shuo Pan,Wei Liu

Tea polysaccharides have attracted widespread attention in regulating gut microbiota. A highly efficient enzymolysis and ultrasonic-assisted extraction method was developed and optimized for the extraction of polysaccharides from Zhonghuang No. 1 etiolated-green tea (GTP) with a-single factor and response surface methodology, i.e, with a pH of 5.5, an enzyme concentration of 7%, an ultrasonic time of 2.5 h, a solid-liquid ratio of 1:20 g/mL, and an ultrasonic temperature of 80°C. GTP exhibited a triple helix structure and demonstrated the ability to bind with Congo red. Its surface was observed to be spherical with slender chain fragments. GTP remained intact throughout the simulated in vitro digestive process, which encompassed the oral, gastric, and small intestinal phases. The relative abundance of Bacteroidota was significantly increased, while the Firmicutes/Bacteroidota ratio was reduced following the administration of GTP. The contents of Faecalibacterium and Bacteroides were increased, while the contents of Escherichia_Shigella and Dorea were reduced after GTP treatment. Furthermore, GTP were degraded and utilized by gut microbiota to produce SCFAs and reducing pH value. GTP regulated the composition of the gut microbiota and has the potential to act as a prebiotic, with the possibility of its use as a functional food.

Fabricating multifunctional lyocell fabric with cationic nanospheres: enhanced anti-fibrillation, with no strength loss, and green dyeability

Ge Xinchang,Fang Kuanjun,Fang Lei,Zhang Longxue,Li Hanyu,Gao Hongguo,Bao Wei

Sustainable lyocell fiber is widely used to manufacture various high-end low carbon and green textile products. However, fibrillation usually occurred during wet processing, causing severe pilling and poor appearance. The current anti-fibrillation methods still suffer from strength loss or unsatisfactory anti-fibrillation performance, and the functional modification of lyocell fibers is generally a distinct processing stage, which causes more water consumption. The present study reported a new strategy for fabricating multifunctional lyocell textiles using cationic P(St-BA-DMC) nanospheres with an average diameter of 90.9 nm and zeta potential of + 49.4 mV. The nanospheres were adsorbed on the negatively charged lyocell fiber surfaces through electrostatic forces when the fibers were treated with the nanosphere suspension. FTIR, SEM, and XPS revealed that when cured at 150 °C, the core–shell-structure nanospheres on the fibers spread into dot membranes like frying eggs, which made the fiber much more hydrophobic through the lotus effect. As a result, the fiber swelling rate reduced from 23.7% to 7%, while the bursting strength of the knitted fabric was not changed significantly. The air permeability at the wet state increased by 30.5% compared to the original fabric. Additionally, the treated fabric’s antibacterial rate against S. aureus reached 97%. The lyocell fabric was endowed with anti-fibrillation, higher air permeability, and anti-bacterial functions without significant strength loss. Moreover, the dyeability of treated fabric was greatly enhanced. This work provides a new strategy for the anti-fibrillation and functionalization of low-carbon lyocell textiles.

A Self-Catalytic NO/O2 Gas-Releasing Nanozyme for Radiotherapy Sensitization through Vascular Normalization and Hypoxia Relief

Shuyu Wang,Miaomiao Cheng,Shenghui Wang,Wei Jiang,Feifei Yang,Xiaomei Shen,Lirong Zhang,Xiyun Yan,Bing Jiang,Kelong Fan

Radiotherapy (RT), essential for treating various cancers, faces challenges from tumor hypoxia, which induces radioresistance. A tumor-targeted “prosthetic-Arginine” coassembled nanozyme system, engineered to catalytically generate nitric oxide (NO) and oxygen (O 2 ) in the tumor microenvironment (TME), overcoming hypoxia and enhancing radiosensitivity is presented. This system integrates the prosthetic heme of nitric oxide synthase (NOS) and catalase (CAT) with NO-donating Fmoc-protected Arginine and Ru 3+ ions, creating HRRu nanozymes that merge NOS and CAT functionalities. Surface modification with human heavy chain ferritin (HFn) improves the targeting ability of nanozymes (HRRu-HFn) to tumor tissues. In the TME, strategic arginine incorporation within the nanozyme allows autonomous O 2 and NO release, triggered by endogenous hydrogen peroxide, elevating NO and O 2 levels to normalize vasculature and improve blood perfusion, thus mitigating hypoxia. Employing the intrinsic O 2 -transporting ability of heme, HRRu-HFn nanozymes also deliver O 2 directly to the tumor site. Utilizing esophageal squamous cell carcinoma as a tumor model, the studies reveal that the synergistic functions of NO and O 2 production, alongside targeted delivery, enable the HRRu-HFn nanozymes to combat tumor hypoxia and potentiate radiotherapy. This HRRu-HFn nanozyme based approach holds the potential to reduce the radiation dose required and minimize side effects associated with conventional radiotherapy.

Pore structure regulation of nano-CdSnO3 based on particles adhesion and mass transfer for enhanced NO-sensing

Yinghao Jia,Xin Hu,Hengjie Huang,Yangcan Deng,Wenning Cao,Ying Zhou,Xinghui Hou

CS- x ( x = 1, 2, 4, 6) nanoparticles have been successfully prepared by simple hydrothermal method combined with calcination process, and the calcination time is adjusted to optimize micro-structure characterization for their enhanced NO-sensing. The techniques of XRD, Raman, SEM, TEM, TGA, DSC, BET, and XPS are used to study their phase structure, micro-morphology, thermal decomposition process and surface states, then the possible gas-sensitive mechanism is analyzed. Results show that the typical CS-2 sensor has the maximum response of 130–50 ppm NO at 90 ℃, the short response/recovery time of 40/45 s, and the low detection limit of 0.1 ppm (1.18). It also has the reliable NO selectivity against to other interfering gases, good repeatability and high stability. Its enhanced NO-sensing performance may be due to the optimization of pore structure by adjusting the calcination process based on particles adhesion and mass transfer, which increases the pore size and specific surface area that could provide the channel and active sites for gas diffusion and adsorption, thus further promoting the gas-sensing reaction. The experimental methods and ideas of pore structure regulation provide a new way for the design of high-performance sensitive materials.

Sacrificing Knight for Pawn-Queen Promotion: Sonosensitive Nano-Xanthiums Orchestrate NO to Retain H2O2 for Antibiofilm Therapy

Yuzheng Wu,Xiaoxue Ren,Dezhi Xiao,Pei Liu,You Ke,Wenqiang Cui,Paul K. Chu,Huaiyu Wang

Biofilm infections present escalating clinical challenges due to their intrinsic resistance to conventional treatments, underscoring the urgent need for innovative therapeutic strategies. Here, we show a pioneering approach that leverages sacrificial nitric oxide (NO) to enhance hydrogen peroxide (H 2 O 2 ) retention for antibiofilm therapy. H 2 O 2 has the ability to produce hydroxyl radicals (•OH), the most potent oxidants in nature, but it is often neutralized by H 2 O 2 scavengers before •OH formation. Our design utilizes ultrasound irradiation to release NO from the biofilm-anchoring nano-xanthiums (G-B@NXs), which subsequently interact with biofilm-associated H 2 O 2 scavengers, including catalase and glutathione, to suppress the decomposition ability toward H 2 O 2 . Additionally, glucose oxidase on the surface of G-B@NXs supplements H 2 O 2 production, facilitates its conversion into •OH, and exhibits effective biofilm sterilization in different infection models. Our strategy not only shows the promise for advancing antibiofilm applications but also provides novel insights into the development of NO-assisted biomaterials.

1, 8-Cineole Ameliorated Staphylococcus aureus-Induced Pneumonia through Modulation of TRP-KYN and Arginine-NO Reprogramming

Qianwen Cao,Luyao Liu,Xiaoge Ma,Chaomin Zhong,Mengqi Tang,Mengge Liu,Ling-Bo Qu,Bo Wei,Xia Xu

1, 8-Cineole (Cin), a cyclic monoterpenoid derived from tea trees and eucalyptus species, exhibits diverse pharmacological properties. Yet, its therapeutic impact and underlying mechanism against Staphylococcus aureus (S. aureus) pneumonia remain to be elucidated. In this study, metabolomics based on UPLC-MS/MS was integrated with network pharmacology, molecular biology, and molecular docking to investigate the effects of Cin. The findings demonstrated that Cin markedly reduced mortality and lung bacterial load, lessened pulmonary damage while suppressing the levels of proinflammatory factors, including tumor necrosis factor α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) in the bronchoalveolar lavage fluid (BALF) of infected mice. Additionally, 19 metabolites, primarily involved in tryptophan metabolism and arginine biosynthesis, were notably modified by Cin via suppressing the enzymatic activity of indoleamine 2, 3-dioxygenase 1 (IDO1) and inducible nitric oxide synthase (iNOS), thereby attenuating the inflammatory response. Notably, knockdown of IDO1 or iNOS significantly diminished the anti-inflammation effect of Cin. In conclusion, our study validates the therapeutic potential of Cin against S. aureus pneumonia via anti-inflammation by downregulating IDO1 and iNOS. Our results provide a theoretical basis of natural substances applied in bacterial pneumonia treatment.

Resolving NO-CO competitive adsorption in CO-SCR: The role of Ni doping in Mn-Ce catalysts

Shuya Rui,Na Liu,Lingwei Ren,Yang Liu,Zian Liu,Binghui Wu,Hua Pan,Zhiping Ye

Traditional Mn-Ce catalysts for selective catalytic reduction (SCR) of NO with CO (CO-SCR) in iron and steel flue gases are often hindered by competitive adsorption of NO and CO, which compromises catalytic efficiency. In this study, we present a Ni-doped Mn-Ce catalyst that overcomes these challenges, achieving 96 % NO conversion, 98 % N 2 selectivity, and 44 % CO conversion at 175 °C. Comprehensive characterization of the catalyst reveals that Ni doping significantly enhances Mn-Ce interactions, boosting oxygen vacancy density and promoting efficient redox cycling between Mn 3 + /Mn 4+ and Ce 3+ /Ce 4+ . Diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS) confirms preferential NO adsorption at oxygen vacancy sites, minimizing CO-NO competition and stabilizing the Langmuir-Hinshelwood mechanism, where both gases co-adsorb and react at oxygen-rich vacancies. Mechanistic insights suggest that Ni facilitates electron transfer, weakening the N–O bond in NO and accelerating its dissociation, resulting in faster NO reduction. This study establishes Ni doping as a promising strategy for improving the low-temperature performance of Mn-Ce-based CO-SCR catalysts, paving the way for more efficient catalytic systems.
