6′-O-Galloylpaeoniflorin attenuates Helicobacter pylori-associated gastritis via modulating Nrf2 pathway

Yun Zhang,Maihuan Wang,Kebin Zhang,Junze Zhang,Xinpu Yuan,Guijun Zou,Zhen Cao,Chaojun Zhang

As a common disease of the digestive system, chronic gastritis is inflammation of the gastric mucosa caused by various factors. Helicobacter pylori (H. pylori) is one of the main causes of chronic gastritis, which can lead to gastric mucosal damage and gland atrophy, thereby promoting gastrocarcinogenesis. Oxidative stress and the inflammatory response are important mechanisms of H. pylori -induced gastritis. 6′-O-Galloylpaeoniflorin (GPF) is a substance isolated from peony root with antioxidant and anti-inflammatory activities. However, its role and mechanism in the pathogenesis of H. pylori -induced chronic gastritis remain unclear. This study explored the effects of GPF on H. pylori -induced gastric mucosal oxidative stress and inflammation using flow cytometry, western blotting, real-time quantitative PCR, and immunohistochemistry. We found that H. pylori infection increased oxidative stress and expression of inflammatory cytokines in vitro and in vivo and that these outcomes were inhibited by GPF. Furthermore, GPF activated nuclear factor erythroid-related factor-2 (Nrf2) and its downstream target genes in H. pylori -infected GES-1 cells and mice. The anti-inflammatory and antioxidant effects of GPF on H. pylori -infected cells were attenuated by an Nrf2 inhibitor. Taken together, these data suggest that GPF reduces H. pylori -induced gastric mucosa injury by activating Nrf2 signaling and that GPF is a potential candidate for the treatment of H. pylori -associated gastritis.

Inducing large birefringence by enhancing asymmetric electron distribution of Y–O polyhedra

Mengen Gao,Qiang Bian,Hongping Wu,Hongwei Yu,Zhanggui Hu,Jiyang Wang,Yicheng Wu

Birefringent crystals, which can modulate the polarization of light, are particularly important in optical communication and the laser industry. Here we report the synthesis, structures, optical properties, and theoretical analyses of four rare-earth borates containing BO3 groups, i.e. LiRb2YB2O6, CaRbYB2O6, and Li2MY4B5O15 (M = Rb, Cs). Their structures are not all typical BO3 group distributions with large birefringence values, e.g., being parallel with each other's distribution or being a regular polygon distribution with a specific form that is parallel with an optical axis. However, their birefringence values exhibit an obvious increase from LiRb2YB2O6 (0.038@532 nm) to Li2MY4B5O15 (M = Rb, Cs) (0.103@532 nm). The origin of this birefringence increase was studied by structural analysis and theoretical calculation. First-principles calculations and structure comparisons indicate that the non-symmetry localization of electron density of the Y–O polyhedra has an important contribution to the large birefringence of Li2MY4B5O15 (M = Rb, Cs).

Preparation and properties of W/O emulsion bacteriostatic microcapsules loaded with KDF

Zhang XiaoNan,Yang Zhongxin,Xu Wenqin,Li Xing,Pan Guanghua,Chen Nan-Chun,Xie Qing-Lin,Wang Xiu Li

Potassium diformate (KDF) has broad prospects in replacing common antibiotics in animal feed; however, the utilization rate of KDF in the animal intestine is low. Therefore, it is necessary to construct a drug delivery system for its targeted release in the colon. When Zeolite P was used as a drug-binding effector, KDF was uniformly dispersed in the pectin matrix, and the pectin bacteriostatic microspheres loaded with KDF were prepared using the emulsion cross-linking method. The optimized pectin bacteriostatic microspheres were systematically characterized, we observed that the drug loading rate and stability of pectin bacteriostatic microspheres improved significantly after adding Zeolite P and the utilization rate of KDF in the simulated intestine was higher than that in the stomach. The pectin bacteriostatic microspheres exerted significant bacteriostatic effect on Escherichia coli and Staphylococcus aureus , indicating that they may be widely used in livestock feed as bacteriostatic agents. Graphical Abstract In order to improve the antibacterial effect of KDF in intestinal tract, W/O emulsion microcapsules were prepared by emulsification method. When Zeolite P was used as a drug-binding effector, KDF was uniformly dispersed in the pectin matrix, and the pectin bacteriostatic microspheres loaded with KDF were prepared using the emulsion cross-linking method. Moreover, the bacteriostatic microspheres possessed significant growth inhibitory effects on E. coli, S. aureus and Bacillus subtilis.

Nitration of o-xylene in the microreactor: Reaction kinetics and process intensification

Shuai Guo,Le-wu Zhan,Bin-dong Li

Nitration reactions are extensively studied in organic synthesis despite their notorious dangers, high reaction rates, and heterogeneous reaction systems. To date, most industrial nitration reactions rely on conventional batch reactors characterized by low mass transfer rates and limited reaction conditions. In contrast, microreactors represent a valuable technology for intensifying such nitration reactions. In this work, we established a continuous flow system of o -xylene nitration and determined the kinetics and mass transfer. The effects of mixing performance, nitric acid concentration, molar ratio, reaction temperature, and residence time on the process safety were investigated separately. The experimental data is supported by dimensionless number calculations, providing a more comprehensive understanding of the nitration reaction's transport and reaction performance in a heart-shaped microreactor. Remarkably, the residence time of the heart-shaped microreactor was reduced by an order of magnitude, while the volumetric mass transfer coefficient was improved by several orders of magnitude compared to conventional stirred tank reactors. Moreover, the spent nitric acid could be efficiently recovered and reused after concentration, further enhancing the sustainability and cost-effectiveness of the process. Our findings demonstrate the significant potential of microreactors for intensifying nitration reactions and paving the way for the design and optimization of more efficient and safer synthetic processes.

A new synthesis of O/N-doped porous carbon material for supercapacitors

Huizi Li,Tingyao Du,Qin Wang,Jianyu Guo,Siyong Zhang,Yan Lu

In this work, a novel carbonization method using H 2 SO 4 as dehydrating agent to prepare graphitized carbon material with O doping is firstly reported. Starch is used as the carbon source. After dehydration in H 2 SO 4 at 100 °C for 6 h, an O-enriched carbon material (OSC) is prepared. It exhibits a specific capacitance of 354 F g −1 at the current density of 1 A g −1 in 1 M H 2 SO 4 solution. After 20,000 cycles, the capacitance retention remains at 110.2 %. It is proved that appropriate amounts of doping O can provide a certain pseudocapacitance , and also improve the wettability of the material, which therefore improve the electrochemical performance of the carbon material. Further, a vapor deposition method using dicyandiamide is applied to grow N-doped carbon nanotubes on the smooth surface of the OSC, which form a three-dimensional conductive network structure on the surface and get a suitable porous structure with the specific surface area of 355 m 2 /g (NOSC). The specific capacitance of the NOSC is 759 F g −1 at the current density of 1 A g −1 in 1 M H 2 SO 4 solution. After 10,000 cycles, the capacitance retention still remains at 95.8 %. The assembled all-solid symmetric supercapacitor based on the NOSC can be operated in a 0–1.8 V voltage window. The energy density is 41.12 Wh kg −1 with a power density of 449.95 W kg −1 .

Role of Catechol-O-Methyltransferase in the Disposition of Luteolin in Rats

Zhongjian Chen,Meng Chen,Hao Pan,Siyuan Sun,Liping Li,Su Zeng,Huidi Jiang

Luteolin is mainly metabolized by phase II enzymes in animals and humans with glucuronidation and sulfation as the two known metabolic pathways. Although methylation of luteolin was reported previously, the structure of the methylated metabolites and the enzymes involved in the process have not been clarified. In our study, two methylated metabolites, M1 (chrysoeriol) and M2 (diosmetin), were identified in the urine after intravenous administration of luteolin to rats, and the data suggested that the methylation was mediated by catechol- O -methyltransferase (COMT). When luteolin was coadministered with a specific COMT inhibitor , entacapone , the formation of M1 and M2 was significantly reduced, whereas the plasma concentration of luteolin increased. Methylation of luteolin was also studied in vitro using rat tissue homogenates. The apparent kinetic parameters associated with the formation of M1 and M2 in vitro were estimated, and regioselectivity of methylation of luteolin was observed. In the in vitro experiment, there was a preference for the formation of M2 over M1. In contrast, accumulation of M1 was preferred in vivo in both rat plasma and urine after an intravenous dose of luteolin. In conclusion, COMT played a crucial role in the disposition of luteolin in rats. Our results indicated that the methylation pathway in rats was significantly reduced when luteolin was coadministered with a specific COMT inhibitor. Therefore, COMT-associated drug-drug interactions need be considered in the future in luteolin clinical trials because the plasma concentrations and related therapeutic effects may be altered in vivo in the presence of a COMT inhibitor.

Understanding the Photo- and Electro-Carboxylation of o-Methylbenzophenone with Carbon Dioxide

Keyi Tian,Ruonan Chen,Jiafang Xu,Ge Yang,Xintong Xu,Yanhua Zhang

The lack of understanding of the radical reaction mechanism of Carbon dioxide (CO2) in photo- and electro-catalysis results in the development of such applications far behind the traditional synthesis methods. Using methylbenzophenone as the model, we clarify and compare the photo-enolization/Diels−Alder (PEDA) mechanism for photo-carboxylation and the two-step single-electron reduction pathway for electro-carboxylation with CO2through careful control experiments. The regioselective carboxylation products,o-acylphenylacetic acid and α-hydroxycarboxylic acid are obtained, respectively, in photo- and electro-chemistry systems. On the basis of understanding the mechanism, a one-pot step-by-step dicarboxylation ofo-methylbenzophenone is designed and conducted. Both the experimental results and related density functional theory (DFT) calculation verify the feasibility of the possible pathway in which electro-carboxylation is conducted right after photo-carboxylation in one vessel. This synthesis approach may provide a mild, eco-friendly strategy for the production of polycarboxylic acids in industry.Keywords:photocatalysis;electrocatalysis;photoelectro-chemistry;CO2reduction;methylbenzophenone

Unveiling the Actual Catalytic Sites in Nanozyme-Catalyzed Oxidation of o-Phenylenediamine

Lu Cheng,Fengxia Wu,Haibo Bao,Fenghua Li,Guobao Xu,Yongjun Zhang,Wenxin Niu

Nanozymes have offered remarkable advantages over natural enzymes and found widespread applications including biosensors, immunoassays, nanomedicines, and environmental remediation. Oxidation of o -phenylenediamine (OPD) by nanozymes has been listed as a standard protocol for determining nanozyme activities. Given the complexity of OPD oxidation processes, however, the mechanism of nanozyme-catalyzed oxidation of OPD remains elusive. In this report, mechanistic studies of nanozyme-catalyzed oxidation of OPD are performed and a distinguishably different mechanism from that of natural enzymes is found. A combination of Fourier transform infrared spectroscopy, nuclear magnetic resonance, electrospray ionization mass spectrometry, and electron microscopic studies provides compelling evidence that polymerization of OPD occurs on the surface of several different nanozymes. The unexpected polymerization causes a dense coating layer of poly( o -phenylenediamine) (POPD) on nanozymes renders the intrinsic properties of nanozymes. Therefore, this fundamental discovery raise serious concerns using OPD-based colorimetric method for determining nanozyme activities. Without examining the surface change of nanozymes after catalytic reactions, the use of OPD-based colorimetric method for determining nanozyme activities is strongly discouraged. Furthermore, POPD is discovered as a new oxidase mimic, and this new mechanism also provides a general and robust method to coat nanomaterials with POPD polymers of enzyme-mimicking properties.

Cyanidin-3-O-glucoside ameliorates cadmium induced uterine epithelium proliferation in mice

Dacheng Yang,Yanhong Ran,Xusheng Li,Xinwei Jiang,Jiali Chen,Jianxia Sun,Lingmin Tian,Katia Teerds,Weibin Bai

Cadmium (Cd) is an environmental pollutant and endocrine disrupter , abundantly present in water, food, and soil. Accumulation of Cd in the body can negatively affect female reproduction; especially the uterus is exceptionally sensitive to the toxic actions of Cd. The anthocyanin cyanidin-3- O -glucoside (C3G) is a naturally occurring phenolic compound in fruits and plants that can antagonize the toxic effects of Cd. This capacity makes C3G a possible candidate to prevent Cd-induced female infertility. The present study aimed to investigate: 1) whether C3G intake could prevent Cd-induced female reproductive toxicity, and 2) the underlying mechanisms responsible for this protective effect. The results of our study indicated that Cd exposure did not affect ovarian function, but induced hypertrophy of the uterine endometrium. Oral intake of C3G markedly reduced the effects of Cd exposure on the thickness of the uterine epithelium cells. Transcriptomic analysis of the endometrium revealed that C3G intake had anti-estrogenic effects, attenuating Cd-induced endometrial epithelial cell proliferation by inhibiting estrogen-responsive genes, enhancing epithelial progesterone receptor expression, and regulating Klf4 expression. The current findings implicate that C3G has the potential to be used as a dietary supplement based on its capacity to intervene in Cd-induced female reproductive toxicity.

Polar O–Co–P Surface for Bimolecular Activation in Catalytic Hydrogen Generation

Huanhuan Zhang,Ke Zhang,Saima Ashraf,Yanping Fan,Shuyan Guan,Xianli Wu,Yushan Liu,Baozhong Liu,Baojun Li

Boron hydrides release an abundant amount of hydrogen in the presence of a suitable catalyst. Accelerating bimolecular activation kinetics is the key to designing cost-effective catalysts for borohydride hydrolysis. In this study, the bimolecular activation of a polar O–Co–P site demonstrated superior hydrogen-generation kinetics (turnover frequency, TOF = 37 min −1 , 298 K) and low activation energy (41.0 kJ mol −1 ) close to that of noble-metal-based catalysts. Through a combination of experiments and theoretical calculations, it was revealed that the activated dangling oxygen atom in the Co–O precursor effectively replaced via surface-phosphorization because of strong electronic interactions between the dangling oxygen and P atoms. This substitution modulated the local coordination environment and electronegativity around the surface Co sites and formed a new polar O–Co–P active site for optimizing the activation kinetics of ammonia borane and water. This strategy based on bimolecular activation may create new avenues in the field of catalysis.

Biomass-based O, N-codoped activated carbon aerogels with ultramicropores for supercapacitors

Ye Zhengqing,Wang Feijun,Jia Chao,Shao Ziqiang

Three-dimensional (3D) O, N-codoped activated carbon aerogels with ultramicropores were prepared via oxidation polymerization, freeze-drying, and carbonization/activation of aniline and sodium alginate. All the O, N-codoped activated carbon aerogels exhibit 3D interconnected hierarchical porous network structures with high specific surface areas (1337–1695 m 2  g −1 ), ultramicropores (0.52–0.54 nm), and rich O (12.68–18.48%) and N (1.18–3.59%) doping. A typical 3D O, N-codoped activated carbon aerogel (ACA700) obtained at the activation of 700 °C exhibits excellent electrochemical performance. When ACA700 is utilized as electrode materials for supercapacitor, the highest specific capacitance of 342 F g −1 at a current density of 2 A g −1 in 3 M H 2 SO 4 electrolyte is achieved. Furthermore, ACA700//ACA700 all-solid-state supercapacitor device displays acceptable energy density (3.8 Wh kg −1 ) at a power density of 246.0 W kg −1 , extraordinary coulombic efficiency (95.8%), and good rate capability. Therefore, the sustainable 3D O, N-codoped activated carbon aerogels with ultramicropores demonstrate tremendous potential for energy storage devices.

In situ crosslinkable hydrogels formed from modified starch and O-carboxymethyl chitosan

Yangling Li,Ying Tan,Kun Xu,Cuige Lu,Xuechen Liang,Pixin Wang

An in situ hydrogel based on oxidation cholesterol starch (OCS) and O-carboxymethyl chitosan (CMCT) that is completely devoid of potentially cytotoxic small molecule cross-linkers and does not require complex manoeuvres or catalysis has been formulated and characterized. The network structure was created by Schiff base formation. The mechanical properties, internal morphology and swelling ability of the injectable hydrogel were examined. Rheological measurements demonstrated that increasing the concentration of the monomer improved the storage modulus. SEM showed that the hydrogel possessed a well-defined porous structure. In addition, the Schiff base reaction was acid sensitive. Under acid conditions, the hydrogel could hydrolyse quickly compared with high pH conditions. Doxorubicin (DOX) was used as a model drug to investigate the control and release properties of the hydrogel. The cytotoxic potential of the hydrogel was determined using an in vitro viability assay with L929 cells as a model and the results revealed that the hydrogel was non-cytotoxic.
