The Phase Structure and Properties of Polyurethanes Based on Polycaprolactone Block Copolymers Dangling Side-Chain PEG

Qing Tang,Hengyu Yan,Shunlan Chen,Li Ma,Nijia Song,Hong Tan,Jiehua Li

Polycaprolactone (PCL)-based polyurethanes are widely used in biomedical applications. A common strategy to enhance their performance is modification with hydrophilic polyethylene glycol (PEG). Moreover, side-chain PEG incorporation enhances molecular mobility, thereby promoting phase separation and enabling the tuning of polyurethane properties from a phase structure perspective. This study compared the effects of side-chain PEG located in the soft segment versus hard segment on the structure and properties of polyurethanes. We designed and synthesized a series of PCL copolymer soft segments containing side-chain PEG, using them to prepare the corresponding polyurethanes. In this structure, the distanced side-chain PEG from the urethane bond reduced interference with hard segment aggregation, promoting a more continuous hard phase and leading to enhanced overall performance. Further research revealed that these polyurethanes exhibit both one- and two-way shape memory properties driven by the human body temperature (37 °C). This work expands the applicability of PCL-based polyurethanes in the biomedical field.

Synthesis of PAMAM-g-PEG-g-DS-g-RB@IO towards dual-modal imaging in atherosclerosis

Qi Hu,Jia Wei,Chao Fang,Ya-Peng Li,Jing-Yuan Wang

This study aimed to synthesize a dual-modal magnetic resonance imaging (MRI)/fluorescence imaging (FLI) nanoprobe in order to detect atherosclerosis. Herein, we had prepared the polyamidoamine-graft-poly (ethylene glycol) (PAMAM-g-PEG) as the carrier. Dextran sulfate (DS) and Rhodamine B (RB) were grafted to PAMAM-g-PEG continuously through reductive amination and amidation reaction to synthesize PAMAM-g-PEG-g-DS-g-RB. The structure of PAMAM-g-PEG-g-DS-g-RB was characterized through nuclear magnetic resonance (1H-NMR) and Fourier transform infrared spectroscopy (FTIR). A new water-soluble superparamagnetic iron oxide nanoparticles (IONPs) has been synthesized through simple ligand exchange between the iron oxide nanaparticles and PAMAM-g-PEG-g-DS-g-RB. The analysis of transmission electron microscopy (TEM) indicated that micelles were well dispersed in water and had uniform sizes. The result of thermogravimetric analysis (TGA) proved that about 83% (mass fraction) polymers were coated on the surface of IONPs. The MRI in vitro evaluation demonstrated a high R2 value (130.8 mM−1s−1) to be served as a T2-weighted contrast agent. The cell counting kit (CCK) assay showed no significant toxicity in RAW264.7. The above results confirmed that PAMAM-g-PEG-g-DS-g-RB@IO could play an significant role of MRI and FLI in the atherosclerosis.

Hierarchical MXene-PEG/Fluroresin Composite Coating: An Interfacial Self-Adaptive Strategy for Lubrication and Corrosion Inhibition

Kang Liu,Li Cheng,Wanting Xu,Weixiang Sun,Shuoqi Li,Chengbao Liu

Polymer-based nanocomposite coatings capable of reducing abrasion and inhibiting corrosion simultaneously have great potential in marine metallic equipment. However, realizing composite coatings interfacial adaptability to external dynamic load and saltwater erosion still faces significant challenge. Herein, a marine environment self-adaptive nanocomposite coating was developed, leveraging frictional heat-driven solid-liquid phase transition of hierarchical MXene-polyethylene glycol (PEG)/fluroresin. The coating takes fluorocarbon resin (FR) as the matrix, incorporating MXene nanosheets chemically grafted with polyethylene glycol (PEG) of different molecular weights. Notably, this coating maintains exceptional lubricity across varying external loads (3N ∼ 5N), achieving a low friction coefficient of 0.047 and wear rate of 11.8×10 − 5 mm 3 /N·m. The low-friction behavior is primarily attributed to the hierarchical self-adaptive lubrication strategy: MXene deal with low-load friction, while higher loads generated friction is resolved through the phase transition capability of PEG. Theoretical calculation results also proved that frictional heat drives the directional migration of MXene-PEG towards the frictional interface, ensuring self-adaptive interface. Meanwhile, electrochemical impedance spectroscopy (EIS) measurements confirmed that the coating exhibits outstanding anti-corrosion performance. This work develops a hierarchical self-adaptive coating based on MXene interlayer sliding and PEG solid-liquid phase transition, enabling the coating to provide reliable anti-corrosion and wear-resistant for equipment under complex alternating load conditions.

Enhanced Protection Against Toxicity of Nemopilema nomurai Venom Using a PEG-EGCG/Tetracycline Hydrochloride Micellar Nanocomplex

Jie Li,Yanan Hu,Yunfeng Qian,Sai Luo,Juxingsi Song,Shaoqian Zhu,Minglei Wang,Huiliang Gan,Qianqian Wang,Liming Zhang

Jellyfish stings are the most common type of marine life injuries. However, at present, the treatment measures against jellyfish stings are mostly empirical and supportive, with uncertain therapeutic outcomes, and there is a lack of specific antidotes based on the toxic mechanism of jellyfish venom in clinical practice. In our previous study, polyphenol epigallocatechin-3-gallate (EGCG) was found to neutralize the toxicity of jellyfishNemopilema nomuraivenom (NnV) in vivo and in vitro. Herein we further demonstrated that EGCG exerted its antagonistic effect against NnV through inhibiting the oxidative stress, pro-apoptotic proteins, and systemic inflammatory responses. Subsequently, we constructed a polyethylene glycol (PEG)-EGCG/tetracycline hydrochloride (HTC) co-loaded micellar nanocomplex in order to enhance the stability and bioavailability of EGCG in vivo, which successfully integrated the membrane-repair function of PEG, the enzyme inhibitory effect of HTC and the antioxidant properties of EGCG. Notably, this micellar nanocomplex demonstrated significant protective effects against both functional damage and pathological alterations in a non-lethal NnV-envenomed mouse model. When administered 1 h after NnV envenomation, EGCG (40 mg/kg), HTC and PEG-EGCG (containing 40 mg/kg EGCG) only partially improved abnormal blood biochemical indicators and moderately alleviated histopathologic damage, and PEG-EGCG/HTC containing merely 8 mg/kg EGCG completely mitigated the toxic reactions in envenomed mice. In the preventive regimen, the administration of EGCG, HTC or PEG-EGCG 30 min before exposure showed no significant improvement in abnormal blood biochemical indicators and histopathologic damage, while PEG-EGCG/HTC could still significantly improve the functional impairments and histopathologic damage of the heart and liver in NnV-envenomed mice. These findings suggest the clinical translational potential of PEG-EGCG/HTC against jellyfish envenomation.

Dual-crosslinking strategies for advancing PVA/PEG composite films: Enhancing mechanical properties and thermal management capability

Baoyi Wu,Yingying Li,Tingli Liu,Chunhua Ge,Lixia Wang,Xiangdong Zhang

Dual-crosslinked PVA/PEG films suppress PEG leakage via interpenetrating network. • Synergistic enhancement of mechanical strength and cycling reliability. • Maintained high phase-change enthalpy with reduced crosslinker content. • Effective as external thermal buffers for food temperature maintenance. Poly(ethylene glycol) (PEG), a widely used phase-change material, offers high latent heat but suffers from leakage during the solid-liquid transition. To address this issue, phase-change composite films were fabricated by solution casting using poly(vinyl alcohol) (PVA) as the matrix and PEG6000 as the energy storage component. Tetraethyl orthosilicate (TEOS) and formaldehyde were used as single crosslinkers, and a dual-crosslinking system combining both agents was further constructed. The effects of single and dual-crosslinking strategies on the overall film performance were systematically compared. TEOS formed an inorganic Si-O-Si network by sol-gel reaction, while formaldehyde constructed an organic network through acetalization with PVA hydroxyl groups. The combination of these two networks formed a complementary organic-inorganic dual-network structure, which restricted PEG chain mobility and enhanced the structural stability of the films. Compared with the uncrosslinked film, the tensile strength and elongation at break of the dual-crosslinked film increased by 60.2% and 122.1%, respectively. After 50 thermal cycles, it showed only a 1.4 J/g loss in phase-change enthalpy, while the leakage ratio remained below 7% after 50 heating-cooling cycles. Notably, this balanced mechanical and thermal performance was achieved with approximately 50% less crosslinker than that used in the single-crosslinked systems. These results demonstrate a low-crosslinker-content strategy for developing flexible external thermal-buffering layers for short-term food-temperature maintenance. Hydrophobic Tail and Linker Diversification of PEG-Lipids Unlocks Precise Muscle Tropism of mRNA-Lipid Nanoparticles

Bingqian Ou,Xuying Tang,Sunxiang Qian,Bo Chen,Sisi Fang,Jinshuang Su,Xiaopeng Sheng,Jie Feng,Zhefan Yuan,Jing Zhang

Intramuscular mRNA lipid nanoparticles (LNPs) often exhibit undesirable liver accumulation, compromising safety and efficacy. While optimization efforts focus on ionizable and helper lipids, the role of trace PEG-lipids remains underexplored. To address this, we constructed a 45-member PEG-lipid library via Ugi/Passerini reactions, keeping the PEG chain constant at 2 kDa while systematically diversifying the hydrophobic tail and linker structures. High-throughput screening identified Pr-182-BNT (P1B LNP) as a lead candidate. P1B LNPs enable highly efficient and selective mRNA delivery to skeletal muscle while drastically minimizing hepatic off-targeting. This precise tropism originates from enhanced muscle cell uptake and optimized membrane interactions, driven by the lipid's unique branched, asymmetric tail. In Ai9 reporter mice, P1B LNPs drive potent muscle-specific gene editing and reduce off-target recombination. As an RSV mRNA vaccine, they elicit robust antigen-specific IgG titers and expands polyfunctional CD8 + IFN-γ + T cells, indicating a Th1-skewed response. By re-engineering only the trace PEG-lipid, this work overcomes a key targeting limitation of classical LNPs and establishes a translatable platform for safer, more effective mRNA vaccines and therapeutics.

Enhanced Pore Size in Solid Amine Adsorbent Pellets with PEG Porogen for High-Performance CO2 Capture

Haixin Liu,Jintao Xu,Luming Qi,Zhenyang Rong,Jiaxuan Wang,Xiaoyang Tang,Zhaoyang Fei,Xian Chen,Qing Liu,Xu Qiao

The introduction of a porogen into a solid amine adsorbent was effective in enhancing the CO2 capture performance, structural stability, and long-term cycling stability. This study selected polyethylene glycol (PEG) as a porogen and prepared a series of CSP-nP-mT pellets, in which ZSM-5 served as the support, chitosan (CS) as the binder, and tetraethylenepentamine (TEPA) as the amine source. The results demonstrated that PEG effectively optimized the adsorbent pore structure, which enhanced the CO2 diffusion and improved accessibility to active amine sites. The structural optimization resulted in a significant increase in the CO2 adsorption capacity by 48.8% (from 1.74 to 2.59 mmol·g–1) compared to the sample without PEG. In addition to the improved adsorption performance, the compressive strength of CSP-1P-0.2T was 23% higher than that of CSP-0.2T. For the characterization of the adsorbent, N2 adsorption–desorption tests were employed to evaluate the pore structure of adsorbent pellets. SEM was used to measure the morphology of adsorbent pellets with different amounts of porogens. FT-IR and TGA were utilized to analyze the variation of functional groups and weight loss during the adsorption process. Furthermore, the adsorbents exhibited excellent cycling stability, retaining 87.2% of their initial capacity after 30 adsorption–desorption cycles. These results demonstrated that the incorporation of PEG in the adsorbent can effectively enhance the adsorption performance of solid amine adsorbents, which provided a strategy for developing high-performance adsorbents.

PEG-engineered semi-interpenetrating network hydrogel with superior deformability and robustness for ultra-sensitive pressure sensing

Shuxian Sun,Guoxu Zhang,Xiaolong Li,Tao Li,Mengyuan Fu,Qiming Han,Hanna He,Xin Tang,Chuhong Zhang

Conductive hydrogels have garnered substantial attention in wearable flexible electronics owing to their exceptional mechanical flexibility and comfort, yet the development of durable hydrogels with high sensitivity under low pressure remains a critical challenge. Here, we propose a facile one-pot in-situ polymerization strategy for the creation of a semi-interpenetrating network hydrogel pressure sensor, integrating polyethylene glycol (PEG) embedded in a crosslinked polyacrylamide (PAM) matrix, with MXene serving as the conductive filler. The resulting semi-interpenetrating network, formed by PEG through its flexible chains and hydrogen bonding interactions with both PAM and MXene, endows the hydrogel with excellent deformation resistance and superior mechanical properties, without compromising its electrical conductivity. Notably, the hydrogel demonstrates a remarkable peak tensile strength of 762 kPa at a strain of up to 1226 %, with full recovery after 70 % compression, and the assembled hydrogel sensor achieves a sensitivity of 6.69 kPa −1 within a rather small pressure range from 0 to 10 kPa. Owing to its ultra-sensitivity, biocompatibility and flexible wearability, the assembled sensor reveals significant medical application potentials, such as aiding deaf- mutes through handwriting recognition and throat vibration–speech translation, detecting unconsciously subtle eye or limb movement for accurate epilepsy diagnosis, as well as expediting the wound healing by further optimizing its composition.

A study on the PEG-assisted stability of spherical nucleic acid constructed by the freezing method

Han Dong,Letian Zhong,Yuliang Cheng,Hang Yu,Yunfei Xie,Weirong Yao,Yahui Guo,Hideya Kawasaki

The freezing method used to prepare DNA-modified gold nanoparticles (SNA) has the advantages of being fast and simple, but the prepared SNA is easy to aggregate. In this paper, the effects of polyethylene glycol (PEG) as an organic macromolecular crowding agent on the single-strand polyadenylate DNA functionalized gold nanoparticles (polyA-SNA) and the single-strand thiolated DNA functionalized gold nanoparticles (HS-SNA) were systematically studied. The experimental results showed the molecular weight and concentration of PEG significantly affect the dispersion and DNA loading of the two kinds of SNA . An increase in DNA strand length inhibits the adsorption of thiolated DNA and polyA-DNA, and the hydrated particle size of SNA increases at first and then decreases with the length of the DNA strand. These phenomena lay on the speculative principle that PEG improved the energy barrier for gold nanoparticles (AuNPs) aggregation through the long chain of macromolecules , and the modification mode and length of surface DNA also play an auxiliary role. This work provided a systematic study and helpful results on improving the stability of SNA prepared by the freezing method.

Tunable Crosslinked Polyvinyl Alcohol/Polyethylene Glycol (cPVA/PEG) Nanofiber Membranes with Enhanced Mechanical and Hydrophilic Balance

Yawen Chang,Zijia Wang,Fujuan Liu

In recent years, membrane separation technology has undergone continuous advancements. Microfiltration (MF) membranes, as an important type, are usually prepared by electrospinning—a simple and efficient method. This study reports the development of crosslinked polyvinyl alcohol/polyethylene glycol (cPVA/PEG) nanofiber membranes through a combination of electrospinning and chemical crosslinking, investigating the effects of different crosslinking concentrations on the membrane morphology, surface wettability, and tensile properties. Comprehensive characterization was carried out by using scanning electron microscopy (SEM), a Fourier-transform infrared spectrometer (FTIR), an X-ray diffractometer (XRD), a thermogravimetric (TG) analyzer, differential scanning calorimetry (DSC), a contact angle tester, a universal testing machine, etc. The results showed that at the crosslinking concentration of 15%, the cPVA/PEG fiber membrane achieved a breaking stress of 29.07 ± 2.60 MPa, a breaking strain of 77.60 ± 6.02%, and a porosity exceeding 43%. SEM, FTIR, XRD, TG, and DSC analyses collectively confirmed the occurrence of chemical crosslinking within the membrane structure. The cPVA/PEG-15 membrane exhibited no observable shrinkage or curling upon water contact, combined with excellent hydrophilicity and lipophilicity in the air. These properties indicate that the membrane can serve as a novel functional membrane substrate (e.g., as hydrophilic separation layers) and is expected to play an important role in fields such as seawater desalination and wastewater treatment, demonstrating significant application potential.

Anti-fouling MOF/APT/PEG composite membranes with excellent durability for oil-in-water emulsion separation

Jingling Gong,Ruirui Jin,Bin Xiang,Fuli Du,Weimin Liu,Jian Li

Membrane technology has become one of the most effective methods for treating oily wastewater, which is harmful to the survival of ecosystems. However, separation membranes often suffer from serious fouling problems. Therefore, in this work, MOF-303 nanoparticles and attapulgites (APT) were loaded on the surface of polyvinylidene fluoride (PVDF) membrane based on the layer-by-layer self-assembly to construct MOF/APT/PEG composite membrane with excellent superhydrophilicity/underwater superoleophobicity, anti-fouling ability and outstanding emulsion separation performance, durability. It should be noted that a unique “absorption-storage” strategy for water molecules has been creatively adopted to realize anti-fouling and promote permeability. The separation efficiency of MOF/APT/PEG composite membrane was as high as 99.6 %, and permeability was above 1316 LMH/bar. Particularly, separation performance still remained stable after 10 cycles and 60 min of cross-flow separation. This study introduces a comprehensive and straightforward methodology for the development of membranes with anti-fouling properties aimed at the highly efficient treatment of oily wastewater.

Targeted multidrug-resistance reversal in tumor based on PEG-PLL-PLGA polymer nano drug delivery system

Liting Guo,Haijun Zhang,Fei Wang,Ping Liu,Yonglu Wang,Guohua Xia,Ran Liu,Xueming Li,Haixiang Yin,Hulin Jiang,Baoan Chen

The study investigated the reversal of multidrug resistance (MDR) and the biodistribution of nanoparticles (NPs) that target leukemia cells in a nude mice model via a surface-bound transferrin (Tf). The cytotoxic cargo of daunorubicin (DNR) and tetrandrine (Tet) was protected in the NPs by an outer coat composed of polyethylene glycol (PEG)-poly-l-lysine (PLL)-poly(lactic-co-glycolic acid) (PLGA) NPs. Injection of DNR-Tet-Tf-PEG-PLL-PLGA NPs into nude mice bearing MDR leukemia cell K562/A02 xenografts was shown to inhibit tumor growth, and contemporaneous immunohistochemical analysis of tumor tissue showed the targeted NPs induced apoptosis in tumor cells. Targeted tumor cells exhibited a marked increase in Tf receptor expression, with noticeable decreases in P-glycoprotein, MDR protein, and nuclear factor κB, as assessed by quantitative real-time polymerase chain reaction and Western blot analysis. Moreover, the concentration of DNR was shown to increase in plasma, tumor tissue, and major organs. Flow cytometry analysis with a near-infrared fluorescent (NIRF) dye, NIR797, was used to study the effectiveness of Tf as a targeting group for leukemia cells, a finding that was supported by NIRF imaging in tumor-bearing nude mice. In summary, our studies show that DNR-Tet-Tf-PEG-PLL-PLGA NPs provide a specific and effective means to target cytotoxic drugs to MDR tumor cells.
