Wenwen Yi,Qiao Li,Tengfei Song,Lei Chen,Xing-Cong Li,Zhizhen Zhang,Xiao-Yuan Lian

New indole alkaloids streptoprenylindoles A–C ( 1 – 3 ) and diterpenoids 18-acetyl-cyclooctatin ( 8 ), 5,18-dedihydroxy-cyclooctatin ( 9 ), and 5-dehydroxy-cyclooctatin ( 10 ) were isolated from the culture of marine-derived Streptomyces sp. ZZ820, along with known 3-cyanomethyl-6-[3-methyl-2-butenyl]indole ( 4 ), N -(2-(1 H -indol-3-yl)ethylacetamide ( 5 ), 1-acetyl-β-carboline ( 6 ), indole-3-methylethanoate ( 7 ), cyclooctatin ( 11 ), and chromomycin A 3 ( 12 ). Their structures were elucidated by a combination of extensive spectroscopic analyses, ECD calculation, and the Mosher's method. Streptoprenylindoles A ( 1 ) and B ( 2 ) are enantiomers that were separated through the preparation of their Mosher esters. Three new diterpenoids ( 8 – 10 ) showed antibacterial activities against methicillin-resistant Staphylococcus aureus (MRSA) and Escherichia coli with MIC values of 24.11–55.12 μM, while chromomycin A 3 ( 12 ) showed potent antibacterial activities against MRSA (MIC: 0.59 μM) and E. coli (MIC 0.04 μM).

Transcriptomic Mechanism of the Phytohormone 6-Benzylaminopurine (6-BAP) Stimulating Lipid and DHA Synthesis in Aurantiochytrium sp.

Xin-Jun Yu,Hong Chen,Chang-Yi Huang,Xiao-Yu Zhu,Zhi-Peng Wang,Dong-Sheng Wang,Xiao-Yan Liu,Jie Sun,Jian-Yong Zheng,Hui-Juan Li,Zhao Wang

The phytohormone 6-benzylaminopurine (6-BAP) significantly improves lipid synthesis of oleaginous microorganisms with the great potential applied in lipid production. In the current study, the lipid and DHA productions in oleaginous Aurantiochytrium sp. were found to be improved by 48.7% and 55.3%, respectively, induced by 6-BAP treatments. Then, using high-throughput RNA-seq technology, the overall de novo assembly of the cDNA sequence data generated 53871 unigenes, and 15902 of these were annotated in at least one database. The comparative transcriptomic profiles of cells with and without 6-BAP treatments revealed that a total of 717 were differently expressed genes (DE), with 472 upregulated and 245 downregulated. Further annotation and categorization indicated that some DE genes were involved in pathways crucial to lipid and DHA productions, such as fatty acid synthesis, central carbon metabolism, transcriptional factor, signal transduction, and mevalonate pathway. A regulation mode of 6-BAP, in turn, perception and transduction of 6-BAP signal, transcription factor, expression regulations of the downstream genes, and metabolic changes, respectively, was put forward for the first time in the present study. This research illuminates the transcriptomic mechanism of phytohormone stimulation of lipid and DHA production in an oleaginous microorganism and provides the potential targets modified using genetic engineering for improving lipid and DHA productivity.

Tetrahydropalmatine alleviates cancer induced bone pain by inhibiting TRPV1-SP-mediated macrophage recruitment and promoting M2 polarization

Qing Zhang,Ziyun Chen,Qingyong Yu,Hanwen Wang,Yucui Jiang,Lan Zhou,Guang Yu,Zongxiang Tang,Changming Wang

Background Cancer-induced bone pain (CIBP) remains a debilitating clinical challenge due to its complex pathogenesis and limited therapeutic options. Tetrahydropalmatine (THP), an active alkaloid from  Corydalis tuber , has shown analgesic potential, but its specific mechanisms in mitigating CIBP- especially its interactions with neural factors and immune cells-remain incompletely understood. This study aimed to investigate the efficacy of THP in alleviating CIBP and clarify its underlying mechanisms, with a focus on the roles of transient receptor potential vanilloid 1 (TRPV1), substance P (SP), and macrophage dynamics in a mouse model of CIBP. Methods Using a multidisciplinary approach, we established a CIBP model in male C57BL/6 mice (and TRPV1-knockout mice) via intramedullary injection of lung cancer cells. Behavioral assessments were performed to evaluate mechanical, thermal, and cold allodynia, as well as spontaneous pain, following daily oral administration of THP (80 mg/kg) from day 7 post-modeling. Molecular and cellular analyses included immunofluorescence staining, real-time PCR, ELISA (to quantify SP, cytokines, and Tac1 expression), calcium imaging (to measure TRPV1-mediated calcium influx in DRG neurons), scratch assays (to assess macrophage migration), and flow cytometry (to analyze macrophage polarization in RAW 264.7 cells). Results THP significantly alleviated CIBP-related allodynia and spontaneous pain in mice. Mechanistically, THP directly inhibited TRPV1 function in the primary phase (≤ 14 days post-modeling) (with an IC 50 of 77.9 µM), reducing SP release from DRG neurons and suppressing macrophage recruitment to DRG and sciatic nerve via the TRPV1-SP pathway. Importantly, THP directly promoted the polarization of recruited macrophages toward the anti-inflammatory M2 phenotype, as evidenced by downregulated iNOS, TNF-α, IL-1β and upregulated CD206, IL-4, IL-10 in vitro (RAW264.7 cells) and in vivo. These effects were abrogated in TRPV1-knockout mice, confirming TRPV1 as a critical mediator in the primary phase. Conclusion In conclusion, THP mitigates CIBP through a dual mechanism: regulating macrophage recruitment via the TRPV1-SP pathway in the early stage to inhibit CIBP initiation and directly modulating the migrated macrophage polarization. This study provides novel insights into THP’s analgesic mechanisms and supports its potential as a preclinical candidate for CIBP treatment. Identification of an efficient phenanthrene-degrading Pseudarthrobacter sp. L1SW and characterization of its metabolites and catabolic pathway

Junlan Li,Wanli Peng,Xianqi Yin,Xiaozheng Wang,Zhixiang Liu,Qinchen Liu,Zixin Deng,Shuangjun Lin,Rubing Liang

Phenanthrene, a typical chemical of polycyclic aromatic hydrocarbons (PAHs) pollutants, severely threatens health of wild life and human being. Microbial degradation is effective and environment-friendly for PAH removal, while the phenanthrene-degrading mechanism in Gram-positive bacteria is unclear. In this work, one Gram-positive strain of plant growth-promoting rhizobacteria (PGPR), Pseudarthrobacter sp. L1SW, was isolated and identified with high phenanthrene-degrading efficiency and great stress tolerance. It degraded 96.3% of 500   mg/L phenanthrene in 72   h and kept stable degradation performance with heavy metals (65   mg/L of Zn 2+ , 5.56   mg/L of Ni 2+ , and 5.20   mg/L of Cr 3+ ) and surfactant (10 CMC of Tween 80). Strain L1SW degraded phenanthrene mainly through phthalic acid pathway, generating intermediate metabolites including cis -3,4-dihydrophenanthrene-3,4-diol, 1-hydroxy-2-naphthoic acid, and phthalic acid. A novel metabolite ( m/z 419.0939) was successfully separated and identified as an end-product of phenanthrene, suggesting a unique metabolic pathway. With the whole genome sequence alignment and comparative genomic analysis, 19 putative genes associated with phenanthrene metabolism in strain L1SW were identified to be distributed in three gene clusters and induced by phenanthrene and its metabolites. These findings advance the phenanthrene-degrading study in Gram-positive bacteria and promote the practical use of PGPR strains in the bioremediation of PAH-contaminated environments.

Efficient biosynthesis of (R)-3-amino-1-butanol by a novel (R)-selective transaminase from Actinobacteria sp.

Xiao-Ling Tang,Nan-Nan Zhang,Guo-Yan Ye,Yu-Guo Zheng

( R )-3-amino-1-butanol is a key intermediate of Dolutegravir for the treatment of HIV/AIDS and its green and efficient biosynthesis has attracted a great deal of attention. Transaminases are currently used as promising biocatalyst for the synthesis of chiral amines. However, many transaminases have ( S )-specificity and ( R )-selective transaminases were less exploited and studied, making the production of ( R )-amines remain challenging. In this study, a novel transaminase from Actinobacteria sp. ( As -TA) was obtained and applied for the biosynthesis of ( R )-3-amino-1-butanol by transferring the amino group from isopropylamine to 4-hydroxy-2-butanone. After optimization of the reaction condition and using a substrate fed-batch strategy, the conversion of 100, 200, 300, 400 and 500 mM 4-hydroxy-2-butanone reached 100%, 94.9%, 86.1%, 76.1% and 70.9%, respectively. ( R )-3-amino-1-butanol with a maximum yield of 29.6 g/L and 99.9% e.e. value was obtained. This was the first time demonstrating the successful biosynthesis of ( R )-3-amino-1-butanol with transaminase as biocatalyst and the obtained As -TA enriched the enzyme pool of transaminase with ( R )-specificity.

Enantioselective resolution of (±)-1-phenylethyl acetate by extracellular proteases from deep-sea bacterium Bacillus sp. DL-2

Lu Dong,Yongkai Xu,Yun Zhang,Aijun Sun,Yunfeng Hu

Chiral 1-phenylethanol and its ester derivative are important chiral chemicals in diverse industries and the preparation of those optically pure enantiomers is of great importance. One bacterium, Bacillus sp. DL-2, whose extracellular proteases could efficiently asymmetrically hydrolyse (±)-1-phenylethyl acetate, was isolated from the deep sea of the Western Pacific. After the immobilization of extracellular proteases and the optimization of enzymatic reactions, (R)-1-phenylethanol was prepared with the enantiomeric excess (e.e.) being 97% and the yield being 41%, respectively. The optimal resolution reaction condition for the preparation of (R)-1-phenylethanol using immobilized extracellular proteases was found to be 5-mM (±)-1-phenylethyl acetate, 360 mg/mL immobilized extracellular proteases, pH 6.5, and 20 °C for 2 h. (S)-1-phenylethyl acetate was generated through enzymatic kinetic resolution with the e.e. being as high as 99% and the yield being 71%, respectively. The optimal resolution reaction condition for the preparation of (S)-1-phenylethyl acetate was found to be 2.5-mM (±)-1-phenylethyl acetate, 440 mg/mL immobilized extracellular proteases, pH 7.5, and 35 °C for 10 h. Our report is the first report about the kinetic resolution of (±)-1-phenylethyl acetate using proteases and the enantio-preference of the proteases was found to be the same as those of most other esterases/lipases. Also notably, the optical purity of (S)-1-phenylethyl acetate generated through kinetic resolution using the proteases of Bacillus sp. DL-2 was the highest report so far. Proteases from deep-sea Bacillus sp. DL-2 are new contributions to the biocatalyst library for the preparation of valuable chiral alcohols and chiral esters through kinetic resolution.

Characterization of a novel aryloxyphenoxypropionate herbicide-hydrolyzing carboxylesterase with R-enantiomer preference from Brevundimonas sp. QPT-2

Xinyun Xu,Jiahao Wang,Ting Yu,Haohan Nian,Hui Zhang,Guangli Wang,Feng Li

Aryloxyphenoxypropionate (AOPP) herbicides with a chiral center are widely used to selectively remove annual and perennial grasses. However, the study of the enzymes involved in enantioselective degradation of AOPP herbicides is limited. A novel family VIII carboxylesterase gene, estwx , hydrolyzed the ester bond cleavage of AOPP herbicides to form the corresponding acid and alkyl side chain alcohol, was cloned from Brevundimonas sp. strain QPT-2 and overexpressed in E. coli BL21. The purified recombinant EstWX was shown to hydrolyze a wide range of AOPP herbicides with various catalytic efficiencies. The enantioselectivity assay indicated that EstWX preferentially catalyzed the hydrolysis of the R -enantiomer of AOPP herbicides. The S73, K76, Y196 together with W368 residues, especially S73 and Y196, were essential for the catalytic function of the carboxylesterase. EstWX is a promising candidate for the bioremediation of multiple AOPP herbicide-contaminated environments and for future mechanism studies on the enantioselective degradation of chiral AOPP herbicides.

AmyZ1: a novel α-amylase from marine bacterium Pontibacillus sp. ZY with high activity toward raw starches

Fang Wei,Xue Saisai,Deng Pengjun,Zhang Xuecheng,Wang Xiaotang,Xiao Yazhong,Fang Zemin

Background Starch is an inexpensive and renewable raw material for numerous industrial applications. However, most starch-based products are not cost-efficient due to high-energy input needed in traditional enzymatic starch conversion processes. Therefore, α-amylase with high efficiency to directly hydrolyze high concentration raw starches at a relatively lower temperature will have a profound impact on the efficient application of starch. Results A novel raw starch digesting α-amylase (named AmyZ1) was screened and cloned from a deep-sea bacterium Pontibacillus sp. ZY. Phylogenetic analysis showed that AmyZ1 was a member of subfamily 5 of glycoside hydrolase family 13. When expressed in Escherichia coli , the recombinant AmyZ1 showed high activity at pH 6.0–7.5 and 25–50 °C. Its optimal pH and temperature were 7.0 and 35 °C, respectively. Similar to most α-amylases, AmyZ1 activity was enhanced (2.4-fold) by 1.0 mM Ca 2+ . Its half-life time at 35 °C was also extended from about 10 min to 100 min. In comparison, AmyZ1 showed a broad substrate specificity toward raw starches, including those derived from rice, corn, and wheat. The specific activity of AmyZ1 towards raw rice starch was 12,621 ± 196 U/mg, much higher than other reported raw starch hydrolases. When used in raw starch hydrolyzing process, AmyZ1 hydrolyzed 52%, 47% and 38% of 30% (w/v) rice, corn, and wheat starch after 4 h incubation. It can also hydrolyze marine raw starch derived from Chlorella pyrenoidosa , resulting in 50.9 mg/g DW (dry weight of the biomass) of reducing sugars after 4 h incubation at 35 °C. Furthermore, when hydrolyzing raw corn starch using the combination of AmyZ1 and commercial glucoamylase, the hydrolysis rate reached 75% after 4.5 h reaction, notably higher than that obtained in existing starch-processing industries. Conclusions As a novel raw starch-digesting α-amylase with high specific activity, AmyZ1 efficiently hydrolyzed raw starches derived from both terrestrial and marine environments at near ambient temperature, suggesting its application potential in starch-based industrial processes.

Fabrication of CNT/Cu composites with enhanced strength and ductility by SP combined with optimized SPS method

Liang Liu,Rui Bao,Jianhong Yi,Dong Fang

Mono-dispersed and homogeneous carbon nanotube (CNT)/copper (Cu) composite powders were fabricated by spraying pyrolysis (SP) method successfully. Subsequently, CNT/Cu composites were obtained by spark plasma sintering (SPS) at sintering temperature ranged from 1023 to 1223 K (70–90% melting point of pure Cu). The mechanical properties , micro-morphology and interface microstructure of CNT/Cu composites were measured and characterized. The results reveal that Cu nano-particles (NPs) coated on the CNT surface are conducive to improving the wettability of CNT within matrix and reducing the interface energy between matrix and CNT, and finally stable interface structure is established. The traditional trade-off tendency between strength and ductility is balanced in the CNT/Cu composites, which are sintered at 1223 K. The ultimate tensile strength (UTS) shows 344 MPa and the elongation is 19.6%, which owing to the improvement of CNT–Cu interface bonding. At last, the role of interfacial bonding extent played in determining the load transfer efficiency of CNT is discussed.

Biobegradation and metabolic mechanism of cyprodinil by strain Acinetobacter sp. from a contaminated-agricultural soil in China

Xiaoxin Chen,Sheng He,Xiaolu Liu,Jiye Hu

Using sequential soil and liquid culture enrichments with cyprodinil as the sole carbon source, a Gram-negative cyprodinil-degrader from cyprodinil-polluted agricultural soil was isolated. The sequencing analysis of 16 S rRNA indicated that the strain showed 99% homology to Acinetobacter sp. The strain could effectively degrade cyprodinil at the neutral condition. At the initial concentrations of 10, 20, 50, 100, 150 and 200 mg L −1 in minimal medium, cyprodinil was degraded by 10, 20, 49.3, 64.2, 57 and 24 mg L −1 within 14 days, respectively. Two metabolites (4-cyclopropyl-6-methyl-2-pyrimidpyridine amine and monohydroxylated para-substitution) were identified using high performance liquid chromatography tandem quadrupole time-of-flight mass spectrometry (HPLC-QTOF-MS/MS). A biodegradation pathway involving imines hydrolysis and monohydroxyl substitution on benzene ring was proposed on basis of the identified metabolites. Acinetobacter sp. would have a potential application in bioremediation of cyprodinil-contaminated soil, and the strain might have important implications in detoxification and bioremediation of pyrimidine analogues.

Preparation of CNT/Cu nano composite powder with uniform dispersion and strong interface bonding by SP method

Lanbo Li,Rui Bao,Jianhong Yi,Liang Liu,Shuai Mao

Uniform dispersion of carbon nanotube (CNT) and strong interface combination between nano‑carbon reinforcement and metal matrix are of extremely importance to fabricate CNT/metals composite by powder metallurgy method due to the hereditary effects. Therefore, in this paper, CNT/Cu composite powder was fabricated through Spray Pyrolysis (SP) to improve the dispersity and interface property . Nucleation and growth of Cu grains on the CNT surface occurred in the ultrasonic atomization of acetate (Cu(CH 3 COO) 2 ·H 2 O) solution droplets with mono-dispersed CNT. The microstructures of composite powder were investigated by TGA , Raman, XRD, FESEM and TEM analysis methods. Results show that structural integrity and purity of CNT can be enhanced by SP process at 500   °C. And CNT/Cu composite powder with uniform dispersion and good combination between CNT and Cu particles could be obtained by SP even though high composition of CNT was added. At last, the mechanisms of nucleation and growth were estimated based on the microstructure analysis .

High Molecular Weight α-Galactosidase from the Novel Strain Aspergillus sp. D-23 and Its Hydrolysis Performance

Ke Chen,Jingyun Zhang,Xing Liu,Peiyuan Zhang,Wenlong Yue,Zhiqiang Cai

Aspergillussp. D-23 was obtained by ultraviolet-diethyl sulfate (UV-DES) compound mutagenesis fromAspergillussp. C18 that the α-galactosidase was purified from. According to sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and non-denaturing polyacrylamide gel electrophoresis (Native-PAGE), the purified enzyme demonstrated apparent homogeneity. The monomeric α-galactosidase’s native molecular weight was 125 kDa. The optimal temperature of α-galactosidase was 65 °C, and 75% of the initial enzyme activity could be maintained between 45 and 55 °C. Its optimal pH was 5.0 with good pH stability. After incubating for 2 h at pH 3.0–8.0, it could retain more than 80% of its original activity. Different concentrations of metal ions had different effects on the α-galactosidase activity. High concentrations of Cu2+could strongly inhibit enzyme activity and low concentrations of Fe2+could promote enzyme activity. Additionally, as shown by thin layer chromatography and high-performance liquid chromatography, the enzyme also had good hydrolysis ability, which could efficiently hydrolyze melibiose and raffinose by more than 95%. Therefore, these excellent characteristics could make α-galactosidase a good candidate for the food and feed industries.Keywords:α-galactosidase;enzymatic characterization;Aspergillussp.;enzymatic hydrolysis;higher molecular weight
