Ultrasensitive hairpin mediated upconversion fluorescence biosensor for Staphylococcus aureus detection in foods and waters exploiting g–C3N4–assisted catalysis

Yi Xu,Waqas Ahmad,Md Mehedi Hassan,Huanhuan Li,Qin Ouyang,Quansheng Chen

A novel g-C 3 N 4 nanosheets (g-C 3 N 4 NSs)-assisted upconversion fluorescent aptasensor was proposed for Staphylococcus aureus ( S. aureus ) detection by adopting hybridization chain reaction (HCR) as a sensitizer. Two hairpin (H1 and H2) structured DNA probes were engineered predicated on the partial complementary sequence (cDNA) of S. aureus aptamer and modified on the exterior of the upconversion nanoparticles (UCNPs), respectively. The presence of S. aureus initiated the HCR system and activated H1 and H2 probes to form a double-helix away from the g-C 3 N 4 NSs vicinity. This led to the decrease in peroxidase-like activity (PA) of the g-C 3 N 4 NSs and corresponding fluorescence recovery proportional to the concentration of S. aureus (10-10 6  cfu mL −1 ). The method was applied to real food samples with acceptable recoveries (91.1–101.6%) and further validated by traditional plate counting method (p > 0.05).

NIR-II photo-amplified peroxidase-like activity using biocatalytic copper-based nanoplatform against subcutaneous methicillin-resistant Staphylococcus aureus infection

Danyan Wang,Xiaojun He,Qiang Wang,Pingping Li,Enoch Obeng,Dongyang Zheng,Jianliang Shen,Zi Jin

Methicillin-resistant Staphylococcus aureus (MRSA) has been associated with significant mortality and longer hospital stays. Artificial nanozymes not requiring antibiotics can generate enough reactive oxygen species (ROS) to kill bacteria. In this study, we synthesized hexagonal copper sulfide nanosheets (CuS NSs) using a modified one-pot synthesis. The CuS NSs demonstrated photothermal stability and superior photothermal conversion efficiency due to large surface area and existence in the second near-infrared (NIR-II) window (1270 nm), promoting the production of ROS. Upon NIR-II irradiation, CuS NSs displayed an excellent biocatalytic antibacterial role in vitro and accelerated abscess resolution, and enhanced the recovery of MRSA-infected wounds in vivo . Moreover, CuS NSs demonstrated negligible toxicity following an amplifying peroxidase-like activity through hyperthermia. Overall, CuS NSs provide a feasible strategy for using the synergistic therapeutic platform for effective therapy of deep-seated MRSA-infected wounds.

Bio-interface engineering of MXene nanosheets with immobilized lysozyme for light-enhanced enzymatic inactivation of methicillin-resistant Staphylococcus aureus

Daorui Zhang,Lunjie Huang,Da-Wen Sun,Hongbin Pu,Qingyi Wei

Being a unique biocatalyst to hydrolyse structural polysaccharides on the cell wall of bacteria, lysozyme can slow down bacterial resistance caused by antibiotic overdose treatments, however, efficient activation of lysozyme activities under pathogenic microenvironments remains challenging. Herein, a bio-interface engineering strategy was proposed for the remote modulation of thermoresistant lysozyme upon rationally designed photothermal nanoplatforms. For this, Ti 3 C 2 T X MXene nanosheets were functionalized by polydopamine (PDA) surface chemistry to enhance photothermal effects and performance durability, during which lysozyme biomacromolecules were immobilized at such a two-dimensional hybrid interface via intermolecular electrostatic affinity. The integrated nanoplatform (denoted as [email protected] @Lyso), with an optimal high light-to-heat conversion efficiency of 46.88%, realized not only precision control of local heat but also photo-responsive up-regulation for bio-catalysis of laden lysozyme. As a result, in vitro and in vivo antibacterial experiments revealed that [email protected] @Lyso could effectively inhibit the proliferation of methicillin-resistant Staphylococcus aureus and accelerated wound disinfection of mice with negligible biological toxicities. The outstanding antibacterial activities of [email protected] @Lyso were attributed to the photo-enhanced lysozyme activity, assisted by bacterial death caused by the mild local hyperthermia and the physical destruction derived from the [email protected] This work exemplified a solution to the bacterial resistance threats via stimuli-responsive enzymatic nanoplatforms.

Preparation of graphene oxide/polydopamine-curcumin composite nanomaterials and its antibacterial effect against Staphylococcus aureus induced by white light

Hongjun Yan,Peiyuan Li,Xiantao Jiang,Xiaoxun Wang,Yuting Hu,Ying Zhang,Rixiang Su,Wei Su

Curcumin (Cur) plays a key role in photodynamic antibacterial activity as a photosensitizer. On the other hand, the antimicrobial potential of graphene oxide (GO) has been reported controversially, and how to improve its antimicrobial ability has become an meaningful study. In this study, we prepared polydopamine-curcumin (PDA-Cur) by pi-pi stacking and loaded it onto the GO surface to obtain GO/PDA-Cur composite nanomaterials. GO/PDA-Cur was characterized by physical and optical means, and GO/PDA-Cur possessed good dispersion and stability in water. In vitro antibacterial results showed that GO/PDA-Cur mediated photodynamic therapy significantly reduced Gram-positive Staphylococcus aureus ( S. aureus ) by 4 orders of magnitude with a bactericidal rate of 99.99 %. The antibacterial mechanism stems from the fact that GO/PDA-Cur can generate reactive oxygen species (ROS) under white light irradiation (405–780 nm), which causes bacterial outer membrane breakage and cellular deformation. In addition, GO/PDA-Cur has good biocompatibility. The antibacterial ability of graphene oxide was significantly improved by combining it with PDA-Cur, which allows it to be used as a photodynamic antibacterial material.

A novel SERS method for the detection of Staphylococcus aureus without immobilization based on Au@Ag NPs/slide substrate

Xiaoyuan Ma,Shan Xu,Liangyu Li,Zhouping Wang

Here, an aptamer-based SERS method for the detection of Staphylococcus aureus ( S. aureus ) without immobilization using [email protected] NPs/slide as enhanced substrate was constructed. ROX-aptamer of S. aureus was modified on the surface of [email protected] NPs/slide through electrostatic interaction. Based on the specific binding effect of the aptamer to S. aureus , the ROX-aptamer fell off from the surface of the substrate, resulting in a decrease of the SERS signal intensity of the substrate. Under the optimal experimental conditions, a good linear relationship was found between SERS intensity at 1500 cm −1 and the logarithm of concentration of S. aureus in the range of 10 2  cfu/mL-10 7  cfu/mL (y = 6623-796lgx, R 2  = 0.994) with a detection limit of 6 cfu/mL. The selectivity analysis revealed that the method had higher selectivity toward the corresponding target. The results for milk sample using the developed SERS method for the detection of S. aureus were similar to those of the plate counting method. The recovery ratio was from 90.60% to 107.26%, indicating the accuracy and reliability of the developed method. This method eliminates the need for bacterial immobilization and improves the convenience and efficiency of detection.

Guo Xin,Deng Xin-Chun,Zhang Ya-Qing,Luo Qin,Zhu Xiao-Kang,Song Yang,Song Er-Qun

Rapid and accurate detection of pathogenic bacteria of Staphylococcus aureus ( S. aureus ) is of importance in the field of clinical research and practical application. Herein, we report a magnetic relaxation switching (MRS)-based strategy for detecting S. aureus in complex samples via the assistance of signal amplification system of liposome. In this design, vancomycin (Van) modified 200 nm magnetic beads (MB 200 -Van) and biotinylated immunoglobulin (Biotin-IgG) were employed as capture probes for S. aureus which following labelled by biotinylated liposomes loading with glucose molecule (Biotin-liposome@Glu, BLGs) based on the affinity of biotin with streptavidin. The released glucose molecules combining with glucose oxidase generated Fe 3+ , causing transverse relaxation time ( T 2 ) signal change. The combination of dual recognition of S. aureus , liposome assisted signal amplification, and the MRS-based assay achieved direct and specific detection of S. aureus in blood and abscess samples. The as-proposed method for S. aureus assay in this study shows potential application for infectious diagnosis in clinical.

Green synthesis characterization and antimicrobial activity against Staphylococcus aureus of silver nanoparticles using extracts of neem, onion and tomato

Kishore Chand,M. Ishaque Abro,Umair Aftab,Ahmer Hussain Shah,Muhammad Nazim Lakhan,Dianxue Cao,Ghazanfar Mehdi,Abdalla Mohamed Ali Mohamed

Recently, it has been shown that silver nanoparticles (AgNPs) exhibit great potential for different applications, including food storage, cosmetic products, electronic components, biosensor materials, cryogenics, dental materials and especially for drug-delivery activities. In this study, we synthesized AgNPs with neem extract (NE) alone and mixed plant extracts of neem, onion and tomato (NOT) as a combined reducing and stabilizing agent by a green synthesis method at different pHs. The synthesized products were characterized by ultraviolet-visible spectroscopy (UV-vis), X-ray diffraction (XRD), dynamic light scattering (DLS), atomic force microscopy (AFM), scanning electron microscopy (SEM) and transmission electron microscopy (TEM). The antibacterial effects of the synthesized products were studied by the Kirby disk diffusion method. It was confirmed that the AgNPs work effectively as a drug material against Gram-positive bacteria Staphylococcus aureus in nutrient agar. In addition, it was seen that the reducing and stabilizing agent NOT could work effectively with six medicines with a different nature at the maximum addition of 15 μg. However, the synthesized product with NE alone only worked for four of the medicines. Therefore, it was evident that the AgNPs synthesized with NOT extract were more susceptible to the Gram-positive bacteria Staphylococcus aureus. We believe that this new route for synthesizing AgNPs with NOT extract could be more beneficial in comparison to NE alone for improved antibacterial properties in drug-delivery applications.

The preservative potential of Octopus scraps peptides−Zinc chelate against Staphylococcus aureus: Its fabrication, antibacterial activity and action mode

Zheng Fang,Liangzong Xu,Yanlan Lin,Xixi Cai,Shaoyun Wang

A new type of zinc-chelating peptide was fabricated from Octopus scraps protein hydrolysate and its antibacterial activity against Staphylococcus aureus was characterized. Results of Fluorescence spectra and Fourier Transform Infrared Spectrometer (FT-IR) demonstrated that the amino nitrogen atoms and the oxygen atoms belonging to the carboxylate groups were the primary chelating sites for zinc ions. The zinc-peptide chelate exerted remarkably antimicrobial ability towards S. aureus , whose minimum inhibitory concentration (MIC) was 1.56 mg/mL. To investigate the antibacterial mechanism, changes of bacterial morphology, cell constituents, membrane potential, intracellular reactive oxygen species (ROS) level and intracellular enzyme activity were measured. Results revealed that Zn-peptide exerted its bactericidal activity by damaging bacterial cell membranes and cell walls, increasing the cell permeability which resulted in structural lesions and release of cell components. Besides, Zn-peptide induced S. aureus cell death via ROS triggered pathway. These findings further promote the development of Zn-peptide complexes, suggesting that Zn-peptide chelate has potential as natural bacteriostatic agent in food processing industry.

Colorimetric aptasensor based on peroxidase-mimetic metal–organic framework nanoparticles and magnetic carbon dots for visual detection of Staphylococcus aureus

Shanglin Li,Zhaomeng Sun,Ziyang He,Mei Liu

A colorimetric aptasensor based on the dual recognition of magnetic carbon dots (M-CDs) and copper-based metal–organic frameworks (Cu-MOFs) was constructed for the visual detection of Staphylococcus aureus (S. aureus). Aptamer-modified Cu-MOFs could specifically identify target bacteria and exhibited peroxidase-like activity by catalyzing the colorimetric reaction of the 3,3′,5,5′-tetramethylbenzidine–H2O2 (TMB–H2O2) system. M-CDs electrostatically attracted bacteria because of their strong positive electrical properties and offered better capturing performance compared with magnetic beads owing to their smaller size. Visual detection of the target was achieved using the color change of the reaction catalyzed by the resuspension of the sandwich-structured complex. The bacteria were sensitively detected with a limit of detection (LOD) of 1 CFU mL−1 using the peroxidase catalytic activity of Cu-MOFs. Non-target bacteria produced negative results, demonstrating the excellent selectivity of the proposed aptasensor. The strategy showed good recovery in real sample detection, showing great potential for visual monitoring of pathogenic bacteria, and could provide a reference for further development of novel sensing platforms for bacterial detection.

Antibiofilm Activities of Borneol-Citral-Loaded Pickering Emulsions against Pseudomonas aeruginosa and Staphylococcus aureus in Physiologically Relevant Chronic Infection Models

Wen Wang,Xuerui Bao,Mona Bové,Petra Rigole,Xiaofeng Meng,Jianyu Su,Tom Coenye

Phytochemicals are promising antibacterials for the development of novel antibiofilm drugs, but their antibiofilm activity in physiologically relevant model systems is poorly characterized. As the host microenvironment can interfere with the activity of the phytochemicals, mimicking the complex environment found in biofilm associated infections is essential to predict the clinical potential of novel phytochemical-based antimicrobials. In the present study, we examined the antibiofilm activity of borneol, citral, and combinations of both as well as their Pickering emulsions against Staphylococcus aureus and Pseudomonas aeruginosa in an in vivo-like synthetic cystic fibrosis medium (SCFM2) model, an in vitro wound model (consisting of an artificial dermis and blood components at physiological levels), and an in vivo Galleria mellonella model. The Pickering emulsions demonstrated an enhanced biofilm inhibitory activity compared to both citral and the borneol/citral combination, reducing the minimum biofilm inhibitory concentration (MBIC) values up to 2 to 4 times against P. aeruginosa PAO1 and 2 to 8 times against S. aureus P8-AE1 in SCMF2. In addition, citral, the combination borneol/citral, and their Pickering emulsions can completely eliminate the established biofilm of S. aureus P8-AE1. The effectiveness of Pickering emulsions was also demonstrated in the wound model with a reduction of up to 4.8 log units in biofilm formation by S. aureus Mu50. Furthermore, citral and Pickering emulsions exhibited a significant degree of protection against S. aureus infection in the G. mellonella model. The present findings reveal the potential of citral- or borneol/citral-based Pickering emulsions as a type of alternative antibiofilm candidate to control pathogenicity in chronic infection.IMPORTANCE There is clearly an urgent need for novel formulations with antimicrobial and antibiofilm activity, but while there are plenty of studies investigating them using simple in vitro systems, there is a lack of studies in which (combinations of) phytochemicals are evaluated in relevant models that closely resemble the in vivo situation. Here, we examined the antibiofilm activity of borneol, citral, and their combination as well as Pickering emulsions (stabilized by solid particles) of these compounds. Activity was tested against Staphylococcus aureus and Pseudomonas aeruginosa in in vitro models mimicking cystic fibrosis sputum and wounds as well as in an in vivo Galleria mellonella model. The Pickering emulsions showed drastically increased antibiofilm activity compared to that of the compounds as such in both in vitro models and protected G. mellonella larvae from S. aureus-induced killing. Our data show that Pickering emulsions from phytochemicals are potentially useful for treating specific biofilm-related chronic infections.
