Synthesis of a dual pH and temperature responsive star triblock copolymer based on β-cyclodextrins for controlled intracellular doxorubicin delivery release

Beibei Lu,Lei Li,Jianning Wu,Lulu Wei,Jun Hou,Zhiyong Liu,Xuhong Guo

Well-defined dual pH and temperature responsive triblock star-shaped amphiphilic copolymers of β-cyclodextrin-g-poly(2-hydroxyethylmethacrylate)-b-poly(N-isopropylacrylamide)-b-poly(2-(dimethylamino)ethylmethacrylate) (β-CD-g-(PHEMA-b-PNIPAM-b-PDMAEMA)) were synthesized by the combination of reversible addition–fragmentation chain transfer (RAFT) polymerizations. The chemical structures and compositions of these copolymers were characterized by Fourier transform infrared spectroscopy (FTIR), proton nuclear magnetic resonance (1H NMR), and gel permeation chromatography (GPC) measurements. Subsequently, the obtained triblock copolymers could self-assemble into micelles in aqueous solution owing to their amphiphilic property resulting from the hydrophobic β-CD core and the hydrophilic PHEMA, PNIPAM, and PDMAEMA segments, which were investigated by dynamic light scattering (DLS) and transmission electron microscopy (TEM). The triblock copolymer nanoparticles exhibited two separate lower critical solution temperatures (LCSTs) and these triblock copolymers in aqueous solutions were investigated at the different pH values of 5.0 and 7.4 as doxorubicin (DOX) delivery systems for controlled release. Polymeric micelles exhibited a pH controlled release performance. Investigation of DOX release from the micelles indicated that the release rate of the drug could be effectively controlled by altering the pH, encapsulating DOX in neutral surroundings (blood circulation or extracellular matrix) and triggering DOX release in acidic surroundings (intracellular endosomes). CCK-8 assays and confocal laser scanning microscopy (CLSM) against HeLa cells indicated that the polymeric micelles themselves had no associated cytotoxicity and possessed good biodegradability and biocompatibility and that the micelles identified the location of the DOX in the HeLa cells. On the other hand, the DOX-loaded micelles possessed a high cytotoxicity to HeLa cells and exhibited inhibition of the proliferation of HeLa cells. Hence, this type of biodegradable, biocompatible, and stimuli-responsive copolymer could serve as a promising material for drug delivery.

Electrochemical performances of a new solid composite polymer electrolyte based on hyperbranched star polymer and ionic liquid for lithium-ion batteries

Wang Ailian,Xu Hao,Zhou Qian,Liu Xu,Li Zhengyao,Gao Rui,Liu Xiangfeng,Zhang Liaoyun

A new composite polymer electrolyte membrane composed of hyperbranched star polymers (HBPS-(PMMA- b -PPEGMA) 30 (the hyperbranched star polymer with hyperbranched polystyrene as core and polymethyl methacrylate block poly(ethylene glycol) methyl ether methacrylate) as arms)), ionic liquid (1-butyl-3-methylimidazolium tetrafluoroborate (BMImBF 4 ) or 1-butyl-3-methylimidazolium hexafluorophosphate (BMImPF 6 )), and lithium bis(trifluoromethanesulfonyl) imide (LiTFSI) has been fabricated by solution casting method. This type of solid polymer electrolyte membrane shows an excellent flexibility, transparency, and free-standing properties. Room temperature ionic conductivity of composite electrolytes with 40 wt% BMImBF 4 or 40 wt% BMImPF 6 can reach 2.5 × 10 −4 and 4.1 × 10 −5  S cm −1 , respectively. The HBPS-(PMMA- b -PPEGMA) 30 /BMImPF 6 /LiTFSI (the composite polymer electrolyte with 1-butyl-3-methylimidazolium hexafluorophosphate) composite polymer electrolyte displays better performances including a good thermal stability with decomposition temperature of 350 °C, a wide electrochemical window with oxidation potential of 4.3 V, and the excellent interfacial compatibility with lithium electrode. Moreover, the Li/LiFePO 4 batteries based on HBPS-(PMMA- b -PPEGMA) 30 /BMImPF 6 /LiTFSI electrolyte retain about 96% of their highest discharge capacity (120.5 mAh g −1 ) after 100 cycles under a current density of 0.1 C at 60 °C, exhibiting the excellent reversible cyclability. The free-standing composite electrolyte films composed of ionic liquids, hyperbranched star polymers and lithium salts show the high ionic conductivities.

Effects of nonionic surfactant and salts on the interactions between oppositely charged star-shaped copolymer and ionic surfactant in aqueous solutions

Yi Guo,Jingwen Shen,Meng Li,Yazhuo Shang,Honglai Liu

The interaction between star -shaped copolymer poly(2-(dimethylamino)ethyl methacrylate-b-2-(2-methoxyethoxy)ethyl methacrylate) (CDPDPM) and anionic surfactant sodium dodecyl sulfate (SDS) in aqueous solutions are investigated by using fluorescence and dynamic light scattering (DLS) techniques. The effects of nonionic surfactant dodecyl β-D-maltopyranoside (C 12 G 2 ) and salts including inorganic salt ammonium bromide (NH 4 Br) and organic salt tetra-n-butylammonium bromide (Bu 4 NBr) on the interaction between CDPDPM and SDS have been explored systematically. The results have suggested that SDS molecules can bind on CDPDPM by electrostatic interaction between the positively charged CDPDPM and negatively charged SDS and thus weaken the binding ability of CDPDPM to water molecules. CDPDPM exhibits temperature-responsive aggregation behavior in aqueous solution gradually with the increase of SDS concentration and the lower critical solution temperature (LCST) decreases correspondingly. The introduction of nonionic surfactant C 12 G 2 changes the interaction between CDPDPM and SDS significantly by participate the formation of complexes and the stability of the CDPDPM/SDS complexes declines leading to the chance of forming larger complexes is deprived at higher temperature. The addition of NH 4 Br to the CDPDPM/SDS system can retard the binding of SDS on the polymer chains because of electrostatic shielding. Correspondingly, LCST is higher and the formed aggregates are smaller. For CDPDPM/Bu 4 NBr/SDS system, the phenomenon is similar to that of the CDPDPM/C 12 G 2 /SDS system. This should be attributed to the nature of Bu 4 N + , possesses not only positive charges but also certain hydrophobicity , and thus Bu 4 N + has the ability to participate and facilitate the formation of SDS micelles . Obviously, the LCST of the mixed system can be tailored by adjusting the system composition on purpose.

Thermosensitive four-arm star-shaped copolymers with phthalocyanine as core and porphyrin copolymer as arms was used as visible light photocatalyst

Shan Wu,Jian Shi,Qian Duan,Ludan Shang,Xiaodong Zhang,Yuhan Duan

Porphyrins and phthalocyanines are considered as visible light catalysts with great development potential due to their unique π-conjugated structure, strong visible light absorption and structural tunability. However, they are easy to aggregate and difficult to dissolve, and their application in the field of photocatalysis is limited. Based on the above problems, in this study, three four-arms star-shaped thermosensitive copolymers (PcZn-(PNIPAM n -co-PUMPP m ) 4 ) photocatalysts with zinc phthalocyanine as the core and porphyrin thermosensitive copolymer as the arms was constructed by using RAFT polymerization technology and the ’core first’ route. The visible light catalytic activity of the prepared star-shaped copolymers was evaluated by using 1,5-dihydroxynaphthalene (DHN) and methylene blue (MB) as model pollutants. The three copolymers were found to be effective in oxidizing DHN to juglone and degrading MB. Among them, PcZn-(PNIPAM 33.5 -co-PUMPP 1.6 ) 4 (P3) showed the highest DHN conversion efficiency (100.0 %), reaction yield (70.0 %) and MB degradation (96.9 %). 1 O 2 and O 2 · - are the main active substances in the visible photocatalytic process. Based on UV–vis absorption spectra, Fluorescence spectra, EIS Nyquist plots and transient photocurrents, it is confirmed that P3 has a wider range of visible light absorption, and there is effective energy and electron transfer between TTPcZn and UMPP units in P3, which makes it have higher catalytic activity. More importantly, the lower critical solution temperature (32.6 ∼ 32.9 °C) of P3 makes it possible to achieve homogeneous catalysis and heterogeneous recovery through simple temperature regulation in the catalytic process, which makes P3 have good reusability. This work paves a way for the construction of high-performance and recyclable visible polymer photocatalyst.

Reversible Shape-Color Synchronous Dual-Response Flexible Poly(vinyl chloride)-Based Composite Film via Star-Shaped Diacetylene-Modified Poly(ε-caprolactone)

Yunpeng Zhang,Peng Gao,Rui Guan,Haoran Zheng,Chuanren He,Longxiang Tao,Bingjie Zhao,Jun Yin

Shape-color dual-responsive polymers triggered by thermal stimuli have garnered significant attention due to their potential to impart a variety of functionalities within a single material. Nevertheless, developing a nontoxic and durable color-shape dual-responsive polymer system through a cost-effective and feasible strategy remained a challenge. In this study, a star-shaped poly(ε-caprolactone) modified by diacetylene (s-DA-PCL31) was synthesized and blended with poly(vinyl chloride) (PVC) via solution casting followed by in situ photopolymerization, resulting in a nontoxic, reversible shape-color dual-responsive flexible PVC-based composite film. The s-PDA-PCL31@PVC composite film exhibited enhanced mechanical properties, including an elongation at break of approximately 447.1% and toughness of 67.1 MJ/m3, which were ∼94 and ∼60 times greater than those of pure PVC, respectively. Furthermore, the composite film demonstrated excellent durability, with only ∼10.1% volatility and ∼2.0% degree of migration. The PVC-based composite film also displayed shape-memory properties, which can be attributed to the formation of stable physical cross-links through hydrogen bonding interactions and chain entanglements. Due to the feasible conformational transition of PDA enabled by the star-shaped PCL with symmetrical chains, the composite film exhibited reversible thermochromism, with an obvious color transition from blue–purple–red–purple–blue with a temperature cycle from 25–80–120–80–25 °C. More importantly, by fine-tuning the appropriate shape transfer temperature (Ttr), the composite exhibited both shape recovery and color change in response to thermal stimuli within a temperature range of 0 to 80 °C. This dual-responsive smart material demonstrated great potential for a variety of applications, including camouflage, anti-counterfeiting, and temperature monitoring.

Highly porous star-crosslinked POSS-based single-ion conducting hybrid electrolytes enabling fast ion transport for dendrite-free lithium metal batteries

Yu Ding,Liya Chen,Xiao Wang,Puyan Huang,Guorong Chen,Jifang Fu

High-performance lithium metal batteries demand polymer electrolytes materials possessing excellent electrochemical performance, mechanical properties and interfacial compatibility. However, the low ionic conductivity, significant polarization and parasitic side reactions observed in conventional polymer electrolytes hinder their practical application. In this work, novel POSS-based single-ion conducting hybrid electrolytes (PSCHEs) with a star -crosslinked network were synthesized by introducing multifunctional POSS into single-ion conducting polymer chains through epoxy ring-opening polymerization. These PSCHEs exhibit a high porosity exceeding 90 %, a room-temperature ionic conductivity of 1.15 × 10 −4  S cm −1 , a high lithium-ion transference number of 0.80 and a wide electrochemical window of 4.8 V. Furthermore, the formation of a stable LiF and Li x SiO y -rich SEI layer at the lithium metal anode interface and excellent interfacial compatibility between the electrolytes and the electrodes were demonstrated, enabling stable lithium plating/stripping for over 1400 h at a current density of 0.1 mA cm −2 . The Li|PSCHEs|LiFePO 4 cells exhibit good cycling and rate performance at room temperature. This work offers a promising strategy for designing polymer electrolytes with a range of desirable properties for high-performance lithium metal batteries.

Controllable Synthesis of Chain Center Dye-Labeled Star Polymers for Quantitative Examination of Interchain Conformation by Time-Resolved Fluorescence Resonance Energy Transfer

Zihao Meng,Ying Wang,Yanlong Luo,Zhenyang Luo,Linling Li,Ye Sha

Using a “core-first” approach with atom transfer radical polymerization, fluorescent center-functional star polymers of equivalent molecular weight but with varying numbers of arms (di-, tri-, and tetra-arm) were prepared. The sensitivity of fluorescence, combined with a dye-labeling technique introducing a fluorescent donor (carbazole) and an acceptor (anthracene) at the center of poly(methyl methacrylate) (PMMA) chains, enabled the application of time-resolved fluorescence resonance energy transfer to obtain quantitative insights into the conformation of the star polymer chains in the film state. When the results of star-branched polymers were compared with those of linear polymers of identical type and molecular weight, the impact of branching on polymer behavior was isolated for examination. Although the star topology does not alter the average intercoil distance, it affects the distance dispersity. Star polymers with higher arm numbers display decreased dispersity from distance due to reduced intermolecular aggregation at their geometric centers. This study presents the first spectroscopic evidence regarding the distribution of geometric centers in star polymers, offering a physical understanding of chain interpenetration and entanglement within star polymers.

Synthesis and Surface-Active Properties of a Homologous Series of Star-Like Triple-Chain Anionic Surfactants Derived from 1,1,1-Tris(hydroxymethyl)ethane

Li Xu,Xing Fenglan,Xu Qun,Sun Xiaolong,Wang Yuping,Wang Liyan,Wang Pinglang

A novel homologous series of trimeric anionic surfactants, 3C n TE3CNa (where n is a fatty acid chain length of 7, 10, or 12), with three hydrocarbon chains and three carboxylate heads connected via tri-etheric bonds were synthesized from long-chain α-bromo fatty acids and a triol, 1,1,1-tris(hydroxymethyl)ethane. The obtained trimeric carboxylic acids were esterified and purified by silica gel column chromatography, then hydrolyzed with dilute sodium hydroxide solution to form a series of trimeric carboxylate surfactant products. All prepared compounds were analyzed by IR, 1 H NMR, and 13 C NMR spectroscopy to confirm their chemical structures. Their surface-active properties were investigated. The critical micelle concentrations (cmc) of 3C n TE3CNa were in the range of 0.12–0.71 mmol/L, and the surface tensions at the cmc ( γ cmc ) were 29.3–34.8 mN/m.

Reduction-responsive amphiphilic star copolymers with long-chain hyperbranched poly(ε-caprolactone) core and disulfide bonds for trigger release of anticancer drugs

Shan Zhang,Yinglai Hou,Heng Chen,Zijun Liao,Jianxin Chen,Ben B. Xu,Jie Kong

In this contribution, the reduction-responsive star copolymers with long-chain hyperbranched poly(ε-caprolactone) (PCL) (HyperMacs) core and disulfide bonds were synthesized via Cu(I)-catalyzed azide-alkyne cycloaddition (CuAAC) reaction. The HyperMacs core was constructed from disulfide-containing AB 2 -type PCL macromonomers, which possesses length-adjustable chain segments between branching points, large cavities, low degree of crystallinity, and reduction-responsivity. After grafted with poly(ethylene glycol), the reduction-responsive star copolymers can self-assemble into micelles in aqueous solution. The obtained micelles exhibited much lower critical micelle concentration (CMC) than their linear analogues. The reduction-responsivity from disulfide bonds makes them a promising carrier candidate for trigger release of anticancer drugs. The in vitro release results confirmed that their doxorubicin (DOX)-loaded micelles exhibited desirable reduction-triggered release performance. The cellular proliferation inhibition against HepG2 cells demonstrated that the DOX-loaded micelles showed a comparable anticancer activity with free DOX. Therefore, it can be expected that the reduction-sensitive micelles may serve as smart vehicles for intracellular delivery of anti-cancer drugs in tumour therapy.

Directed Self-Assembly of Polystyrene-(Polymethyl acrylate)2 Miktoarm Star Copolymers on 193 nm Immersion Lithography-Defined Chemical Patterns via Thermal Annealing

Jin Zhang,Xiaoou Li,Yu Zhang,Xin Wang,Miaomiao Han,Yadong Liu,Shengxiang Ji

Directed self-assembly (DSA) of block copolymers in combination with 193 nm immersion lithography (193i) is one of the potential solutions for sub-10 nm patterning. Here we report the synthesis and DSA of a high-χ miktoarm star block copolymer, polystyrene-(poly(methyl acrylate)2 (PS–PMA2), for sub-10 nm nanolithography. Compared with diblock PS-b-PMA, PS–PMA2 has a higher effective χ value (0.095@150 °C) and a smaller period with the same molecular weights. The smallest achievable period (L0) was 12.8 nm for PS–PMA2, while a perpendicular orientation of lamellar domains was obtained in PS–PMA2 films with L0 down to 14.1 nm. DSA of PS–PMA2 with 5× density multiplication was successfully achieved on chemical patterns with the pattern period (LS) of 80–90 nm defined by 193i with a feature size down to 8 nm. In addition, DSA of PS-b-PMA was also demonstrated on 193i-defined patterns with a feature size of 9 nm.

Synthesis of Reduction-Sensitive Brush-Arm Star Polymers via Combined Ring-Opening Metathesis Polymerization–Atom Transfer Radical Polymerization and Controlled-Release Properties

Xuhui Liang,Yuping Liu,Zhen Dong,Nanting Qiu,Fuwei Kong,Kun Cao,Zhong-Ren Chen

Mild reaction conditions and excellent functional group tolerance make ring-opening metathesis polymerization (ROMP) a widely used technique to synthesize functional materials. In this work, we used ROMP and atom transfer radical polymerization (ATRP) to prepare a reduction-sensitive brush-arm star polymer (BASP) drug carrier by a one-pot method and further conducted in vitro studies. First, a bifunctional norbornene-based ATRP initiator NB-Br and a reduction-sensitive dinorbornene cross-linker (DNC) were synthesized. Subsequently, NB-Br was subjected to ATRP using the hydrophilic monomer oligoethylene glycol monomethyl ether methacrylate (OEGMA) at different polymerization times to obtain the norbornenyl macromonomer NB-P(OEGMA)n with different degrees of polymerization. In the same reaction flask, NB-P(OEGMA)n underwent an ROMP reaction to generate a brush-shaped polymer. Next, DNC was added to the reaction flask, and the reaction was continued to obtain BASP. This one-pot method could successfully be used to achieve the transformation of the macromonomer to the brush-shaped polymer and subsequently to the star polymer. In vitro biological experiments demonstrated that BASP micelles exhibited reduction-triggered cleavage of disulfide bonds and degradation of polymers after coculture with 10 mM glutathione, promoting the release of intracellular doxorubicin and achieving the controlled release of drugs. We expect the findings of our study to serve as an efficient synthetic strategy to prepare carriers for biomedical applications.

Shape-controlled synthesis of golf-like, star-like, urchin-like and flower-like SrTiO3 for highly efficient photocatalytic degradation and H2 production

Man Zhou,Jingwen Chen,Yakang Zhang,Mengting Jiang,Song Xu,Qian Liang,Zhongyu Li

As a typical perovskite-type metal oxide , SrTiO 3 has emerged as prospective candidate for many fields. However, the synthesis of SrTiO 3 with controlled morphology, high surface area, and enhanced photocatalytic activity are still lacking. Herein, a series of porous SrTiO 3 with well-controlled morphologies including assembled nanoparticles (ANPs), golf-like particles (GLPs), star-like microspheres (SLMs), urchin-like microspheres (ULMs), and flower-like microspheres (FLMs) were successfully prepared via an ethylene glycol-water mixed solvothermal route. The ratio of V EG /V H2O play an important role in the shape-evolution during the solvothermal reaction. A comparative study of photocatalytic H 2 production and photodegradation was performed, and a possible photocatalysis mechanism of SrTiO 3 has been proposed. Significantly, the ULMs and FLMs photocatalysts of SrTiO 3 with optimized low Pt loading amount (0.075  wt %) exhibited outstanding H 2 production rates (8.21 and 7.29 mmol g −1  h −1 ) due to its unique structure of high-surface area and defect-rich surface. The facile and shape-controlled synthesis of varied SrTiO 3 structures is believed to be useful for the design and application of perovskite .
