Broad-specific immunochromatography for simultaneous detection of various sulfonylureas in adulterated multi-herbal tea

Haihuan Xie,Yingying Li,Jin Wang,Yi Lei,Anastasios Koidis,Xiangmei Li,Xing Shen,Zhenlin Xu,Hongtao Lei

Sulfonylureas (SUs) are a series of anti-diabetic drugs widely used for type 2 diabetes mellitus for clinic treat. However, it is often illegally adulterated in multi-herbal tea to improve the claimed anti-diabetic activity in recent years. In this study, a novel hapten was rationally designed, and a broad-specific monoclonal antibody (anti-SUs mAb) recognizing nine SUs was developed. This mAb was used to develop a colloidal gold lateral flow immunochromatographic assay (CG-LFIA). The anti-SUs mAb demonstrated half inhibitory concentration (IC 50 ) ranged from 0.15 ng/mL to 3.25 ng/ mL for nine SUs by ELISA. The cut-off value of developed CG-LFIA for nine SUs was from 3 to 100 ng/ mL for the spiked samples. LC-MS/MS confirmed the reliability of the new CG-LFIA. The results indicated that the proposed CG-LFIA could be an ideal method in on-site screening surveillance assay for SUs illegally adulterated in multi-herbal tea products.

Novel active starch films incorporating tea polyphenols-loaded porous starch as food packaging materials

Zhikun Miao,Yanfei Zhang,Panfang Lu

A novel active food packaging film was developed by casting a corn starch/tea polyphenol (TP)-loaded porous starch (PS, obtained by enzymatic hydrolysis) film forming solution, with the latter helping to regulate the slow release of TP. Results showed that PS had a favorable TP adsorption capacity, and the casted films had a homogeneous distribution of the formulation components. Likewise, the active films had good mechanical properties, UV barrier properties, thermal stability, and excellent antioxidant properties. The slow release of TP from the films was sustained, which is a desired characteristic for extending the protection afforded by the active film to the food under consideration.

Synthesis of cationic branched tea polysaccharide derivatives for targeted delivery of siRNA to hepatocytes

Shuyun Wu,Na Li,Chuan Yang,Li Yan,Xuan Liang,Meng Ren,Liqun Yang

The cationic branched tea polysaccharide (CTPSA) derivative bearing N -acylurea and 3-(dimethylamino)-1-propylamine residues was synthesized and characterized using FTIR and 1 H NMR spectroscopy. A nonspecific siRNA (NsiRNA) was used as a model molecule of functional siRNA that could downregulate over-expressed glycometabolism enzymes in the liver. The result from the agarose gel electrophoresis confirmed that the CTPSA and NsiRNA could form stable complexes when their weight ratio was larger than 18. The zeta potentials and sizes of the complexes were in the range of +8–+15 mv and 120–150 nm, respectively. The CTPSA/NsiRNA complex was observed as nanoparticles with a spherical shape of approximately 100 nm using scanning electron microscopy. The CTPSA derivative and the CTPSA/NsiRNA complexes exhibited lower cytotoxicity in HL-7702 cells when compared with the branched PEI (bPEI) and bPEI/NsiRNA complexes assessed by the Cell Counting Kit-8 assay. The results of flow cytometric analysis and laser confocal microscopy indicated that the CTPSA derivative could effectively target the transfer of the NsiRNA to HL-7702 cells. This work provides a potential approach to promote the CTPSA derivative as a nonviral vector for targeted delivery of functional siRNA to hepatocytes.

Determination of phthalate esters in teas and tea infusions by gas chromatography–mass spectrometry

Liping Du,Lijuan Ma,Yang Qiao,Yan Lu,Dongguang Xiao

Phthalate esters (PAEs), a group of environmental pollutants which are carcinogenic to human body, have been detected in teas. In this work, five PAEs in teas and tea infusions were quantitatively determined by a modified simultaneous distillation extraction (SDE) coupled with gas chromatography–mass spectrometry. After the optimization of SDE, the proposed method afforded a wide range of linearity and high linear regression coefficients with the limits of detection range of 0.24–3.72   μg/kg. The average recoveries were 79.83–116.67% for tea samples and 78.22–101.64% for tea infusions with all the relative standard deviations below 20%. The total content of five PAEs in teas was 1.135–3.734   mg/kg and the total dissolving ratio of five PAEs from tea to infusion was 19.05–28.07% for the selected tea samples. The risk assessment result of all the selected tea samples demonstrated that the population with the habit of drinking tea won’t cause risk to human health.

Quantum-Dot-Based Lateral Flow Immunoassay for Detection of Neonicotinoid Residues in Tea Leaves

Shuangjie Wang,Ying Liu,Shasha Jiao,Ying Zhao,Yirong Guo,Mengcen Wang,Guonian Zhu

Neonicotinoid insecticides are commonly used for pest control on tea plantations as a result of their broad-spectrum activity. However, neonicotinoid residues released from tea leaves into tea infusions pose a dietary risk to consumers. Therefore, a rapid, sensitive, and reliable on-site detection method for neonicotinoids is needed. We developed a quantum-dot-based fluorescent lateral flow immunochromatographic strip (LFICS) combined with a broad-specific antibody for detection of typical neonicotinoids (imidacloprid, imidaclothiz, and clothianidin), with sensitivities [50% inhibitory concentration (IC50)] of 0.104–0.33 ng/mL and visual detection limits of 0.5–1 ng/mL. The strip assay could be completed in less than 30 min. Using the LFICS to analyze spiked tea samples (green tea, black tea, and oolong tea), the average recovery of the three neonicotinoids ranged between 71 and 111%, with the coefficient of variation below 12%. The results from the LFICS tests for field samples were consistent with results from ultraperformance liquid chromatography–tandem mass spectrometry. The newly developed strip is a useful tool for the on-site detection of neonicotinoid residues in tea.

Rapid determination of flavonoids in green tea by synchronous fluorescence spectra coupled with chemometrics

Jiajia Shan,Xue Wang,Mohammad Russel,Junbo Zhao

Flavonoids are one of the most important antioxidant compounds of tea. It is necessary to determine flavonoid concentration of tea infusions and to understand how the storage condition and tea brewing processing impact on flavonoid content. The aim of this study is to analyze flavonoids’ properties and predict flavonoid concentration using synchronous fluorescence spectra coupled with chemometrics. Different wavelength intervals were measured to develop an optimal model. Successive projection algorithms and interval partial least square regression were applied to extract informative wavelengths. The optimal interval partial least square regression model was obtained with the determination coefficient of calibration of 0.95, determination coefficient of cross-validation of 0.90.

Determination of Bioactive Components in Chrysanthemum Tea (Gongju) Using Hyperspectral Imaging Technique and Chemometrics

Yunpeng Wei,Huiqiang Hu,Minghua Yuan,Huaxing Xu,Xiaobo Mao,Yuping Zhao,Luqi Huang

The bioactive components of chrysanthemum tea are an essential indicator in evaluating its nutritive and commercial values. Combining hyperspectral imaging (HSI) with key wavelength selection and pattern recognition methods, this study developed a novel approach to estimating the content of bioactive components in chrysanthemums, including the total flavonoids (TFs) and chlorogenic acids (TCAs). To determine the informative wavelengths of hyperspectral images, we introduced a variable similarity regularization term into particle swarm optimization (denoted as VSPSO), which can focus on improving the combinatorial performance of key wavelengths and filtering out the features with higher collinearity simultaneously. Moreover, considering the underlying relevance of the phytochemical content and the exterior morphology characteristics, the spatial image features were also extracted. Finally, an ensemble learning model, LightGBM, was established to estimate the TF and TCA contents using the fused features. Experimental results indicated that the proposed VSPSO achieved a superior accuracy, with R2scores of 0.9280 and 0.8882 for TF and TCA prediction. Furthermore, after the involvement of spatial image information, the fused spectral–spatial features achieved the optimal model accuracy on LightGBM. The R2scores reached 0.9541 and 0.9137, increasing by 0.0308–0.1404 and 0.0181–0.1066 in comparison with classical wavelength-related methods and models. Overall, our research provides a novel method for estimating the bioactive components in chrysanthemum tea accurately and efficiently. These discoveries revealed the potential effectiveness for constructing feature fusion in HSI-based practical applications, such as nutritive value evaluation and heavy metal pollution detection, which will also facilitate the development of quality detection in the food industry.

Uptake, Translocation, Metabolism, and Distribution of Glyphosate in Nontarget Tea Plant (Camellia sinensis L.)

Mengmeng Tong,Wanjun Gao,Weiting Jiao,Jie Zhou,Yeyun Li,Lili He,Ruyan Hou

The uptake, translocation, metabolism, and distribution behavior of glyphosate in nontarget tea plant were investigated. The negative effects appeared to grown tea saplings when the nutrient solution contained glyphosate above 200 mg L–1. Glyphosate was highest in the roots of the tea plant, where it was also metabolized to aminomethyl phosphonic acid (AMPA). The glyphosate and AMPA in the roots were transported through the xylem or phloem to the stems and leaves. The amount of AMPA in the entire tea plant was less than 6.0% of the amount of glyphosate. The glyphosate level in fresh tea shoots was less than that in mature leaves at each day. These results indicated that free glyphosate in the soil can be continuously absorbed by, metabolized in, and transported from the roots of the tea tree into edible leaves, and therefore, free glyphosate residues in the soil should be controlled to produce teas free of glyphosate.

Flavor characteristic and characterization of key sweet aroma compounds in Camellia nanchuanica black tea

Fang Wei,Liyong Luo,Fanwei Sun,Bowen Zhang,Jie Yang,Yan Liu,Liang Zeng

Camellia nanchuanica (NC) black tea has garnered attention for it's appealing sweet taste and aroma . Previously, phloridzin was identified as the key sweet taste compound in NC black tea. To investigate the aroma characteristic of NC black tea, a flavor wheel containing 54 flavor attributes, was developed using 30 samples of NC black tea. Sweet aroma was identified the most prominent characteristic aroma attribute of NC black tea. To further reveal the key sweet aroma compounds, 112 volatile compounds were totally detected using gas chromatography–mass spectrometry. Among them, β -ionone, linalool, phenylacetaldehyde, octanal, and geraniol were the potential key sweet aroma compounds in NC black tea because their relative odorant activity values and odorant activity values were exceeded 1. Notably, when β -ionone, linalool, and geraniol concentrations were 47.17–94.35, 168.02–336.03, 376.79–753.58 μg/L primarily with floral and sweet aroma, significantly enhanced the sweet aroma of NC black tea. Generally, our work not only revealed the flavor characteristics of NC black tea but analyzed its key sweet aroma compounds, which provided a theoretical basis for improving its higher sweet aroma.

Konjac glucomannan/zein active film loaded with tea polyphenol–ferric nanoparticles for strawberry preservation

Zhenzhen Wu,Lin Wang,Zhiwen Hu,Xiuqing Guan,Yibin Chen,Mingming Xu,Xianrui Chen,Nitong Bu,Jie Duan,Wei Liu,Chen Ma,Jie Pang

With increasing global environmental awareness and concerns about food safety, biodegradable active packaging has garnered widespread attention. In this study, the stability and bioactivity of tea polyphenol (TP) were enhanced through the preparation of TP–ferric nanoparticles (TP–Fe NPs) using metal–polyphenol ion coordination. Moreover, the introduction of Fe ions can further enhance the antibacterial effects of TP–Fe NPs. Using the hydrogen bonding between konjac glucomannan (KGM) and zein to enhance the hydrophobicity and mechanical properties of the film. By employing KGM and zein as the matrix, we incorporated TP–Fe NPs as active fillers to create multifunctional active packaging films. This study aimed to meet the needs of food safety and sustainable development goals. The resulting film exhibited excellent water resistance (water contact angle: 117.73°), mechanical strength (tensile strength: 21.82 MPa, elongation at break: 94.30 %), ultraviolet-shielding ability (>99 %), biodegradability (5 days in soil), and antioxidant (>85 %) and antibacterial (>99 %) properties. Moreover, the film significantly reduced strawberry decay and extended its shelf life by 10 days. These findings provide new insights into the application of nanomaterials in active packaging, highlighting their potential and advantages in food preservation.

Integrative approaches unravelling tea drought alleviation mechanisms primed by carbonyl volatiles and signal peptide

Abdelkader Bassiony,Shujuan Sun,Kangni Yan,Qunhua Peng,Mengxue Zhou,Jianyu Fu,Susanne Baldermann,Jiang Shi,Haipeng Lv,Zhi Lin

Drought stress (DS) significantly hampers the growth and productivity of tea plants, necessitating effective strategies to enhance their resilience. This study comprehensively investigated the mechanisms of carbonyl volatiles-methyl jasmonate (MeJA) and cis-3-hexenyl acetate ( cis -3-HAC) and signal peptide CLAVATA3/EMBRYO-SURROUNDING REGION-RELATED 25 (CLE25) promotion DS resistance using integrative metabolomics and proteomics strategy. Total pigment content decreased, while soluble sugar and proteins increased significantly under DS and further increased after foliar inducement of CLE25, MeJA, and cis -3-HAC. Gallated catechins and amino acids exhibited apparent decreased under DS, especially EGCG (24.6–13.4 mg g −1 ) and theanine (10.66–3.78 mg g −1 ), but significantly mitigated by CLE25 inducement. Antioxidant enzymes activity, such as catalase (CAT), jumped from 23.1 to 48.2 and further boosted to 118.8 Ug −1 min −1 FW with CLE25. Proteomic analysis revealed massive increased in stress tolerance proteins, particularly dehydrins and heat shock proteins, rising by >50.0 % with CLE25 inducement and the expression levels of peroxidase (POX), superoxide dismutase (SOD), α-galactosidase (α-GAL), carboxypeptidases (CPs), and transaldolase (TAL) exhibited higher after inducement. Furthermore, stress signaling-related proteins were in-depth explored, especially thioredoxin proteins; sucrose non-fermenting 1-related protein kinase 2 (SnRK2) was novelly verified in activating abscisic acid (ABA) responding. Differences among drought resistance mechanisms after carbonyl volatile and CLE25 treatments were comprehensively studied. The integration of metabolite and protein levels provided a comprehensive illustration of tea DS tolerance mechanisms and offered promising promotion strategies through foliar application of MeJA, cis -3-HAC, and CLE25.

Development of antioxidant arabinoxylan-tea polyphenol composite films for enhanced preservation of fresh grapes

Rongrong Luo,Zimeng Peng,Na Wu,Li Zhang,Bin Peng,Rong Shao,Wei Xu,Lei Yang

In this study, arabinoxylan (AX) was used as a substrate, and tea polyphenol (TP) as a functional additive to create degradable, non-polluting, and antioxidant packaging materials. The effects of different TP concentrations on light transmittance and antioxidant activity of the AX-TP composite films were analyzed. The colors of the films gradually deepened with increasing TP content. The maximum scavenging rate of 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals using the AX-TP composite film solution of 59.13 ± 2.19 % was achieved at the TP concentration of 2.0 %. AX-TP composite films with different TP concentrations were applied to the surfaces of grapes, and the sensory quality, shriveling and decay rates, titratable acid, and weight loss rate of grapes during storage were evaluated using data from different experimental groups. The AX-TP composite films coated on grapes reduced the transpiration of water in the fruit and delayed grape spoilage, demonstrating an excellent preservation effect. These results show that AX-TP composite films increase the shelf life of fresh grapes.
