Molecular Electronic Coupling-Induced Photoacoustics for NIR-I/II Duplex in Vivo Imaging

Zong Chang,Liangjian Liu,Jingying Zhai,Chenchen Liu,Xiao Wang,Chengbo Liu,Xiaojiang Xie,Qinchao Sun

Molecular electronic coupling has been found to play a crucial role in the light-induced processes of natural and artificial systems. The light-induced acoustic process (photoacoustic) is also considered to be one of the most essential characters of the light-absorbing molecules. Herein, we report an interesting observation of the molecular electronic coupling-induced photoacoustic effect (MECPA) in which the PA intensity is dominated by the molecular electronic coupling, rather than the absorption intensity as usually concerned. The molecular electronic coupling effect results in a significant exciton absorption suppression from the monomer to pentamer. However, the corresponding PA efficiency was found to be approximately 2 orders of magnitude greater for the latter. A well-behaved linear correlation between the molecular exciton absorbance and PA efficiency was observed. The surprisingly significant MECPA effect was realized via novel NIR-II squaraine-benzothiopyrylium dyes. Excellent photophysical properties of the novel NIR-II dyes were achieved, such as a large absorption extinction coefficient and high photostability. In vivo duplex PA imaging was demonstrated for tumor tissue and macrophage cells and blood and lymphoid vessels. The MECPA effect would open a new window for the design of PA contrast agent systems for in vivo imaging.

NIR Activated Upper Critical Solution Temperature Polymeric Micelles for Trimodal Combinational Cancer Therapy

Yuejia Ji,Yuxin Sun,Mingyang Hei,Di Cheng,Bin Wang,Yao Tang,Yun Fu,Weiping Zhu,Yufang Xu,Xuhong Qian

The balance between drug efficiency and its side effects on normal tissues is still a challenging problem to be solved in current cancer therapies. Among different strategies, cancer therapeutic methods based on nanomedicine delivery systems have received extensive attention due to their unique advantages such as improved circulation and reduced toxicity of drugs in the body. Herein, we constructed dual-responsive polymeric micelles DOX&ALS@MFM based on an upper critical solution temperature (UCST) polymer to simultaneously combine chemotherapy, photothermal therapy (PTT), and photodynamic therapy (PDT). Amphiphilic block copolymer P(AAm-co-AN)-b-PEI-ss-PEG-FA with a critical point of 42 °C was able to self-assemble into polymeric micelles under physiological conditions, which further encapsulated anticancer drug doxorubicin (DOX) and photosensitizer ALS to obtain drug-loaded micelles DOX&ALS@MFM. Micelles aggregated at tumor sites due to folate targeting and an enhanced permeability retention (EPR) effect. After that, the high intracellular concentration of glutathione (GSH) and near-infrared (NIR) light prompted disassembly of the polymer to release DOX and ALS. ALS not only plays a role in PTT but also produces singlet oxygen, therefore killing tumor cells by PDT. Both in vitro and in vivo studies demonstrated the photothermal conversion and reactive oxygen species generation ability of DOX&ALS@MFM micelles, at the same time as the excellent inhibitory effect on tumor growth with NIR light irradiation. Thus, our research substantiated a new strategy for the biomedical application of UCST polymers in the cited triple modal tumor therapy.

UV–NIR Dual-Responsive Nanocomposite Coatings with Healable, Superhydrophobic, and Contaminant-Resistant Properties

Wei Li,Xingxing Ni,Xinhai Zhang,Yang Lei,Jie Guo,Jie Jin,Bo You

Self-healing superhydrophobic coatings are attracting interest, but it still remains a great challenge to develop facile and fast self-healing strategies for superhydrophobic coatings. In this work, a novel environmentally friendly self-healing superhydrophobic coating based on an ultraviolet (UV)/near-infrared light (NIR) dual-responsive action was fabricated by blending UV-responsive microcapsules with NIR-responsive carbon nanoparticles (NPs), hydrophobic silica NPs, and waterborne silicone latex. The UV-responsive microcapsules were simply prepared through the electrostatic adsorption of negatively charged TiO2 NPs onto a positively charged microcapsule surface. The UV–NIR dual-responsive properties were mainly reflected in the healing of the superhydrophobic property for coatings through NIR or UV light irradiation. The self-healing process could be repeated many times, which can be attributed to the continued release of fluorine silane (FAS-13) loaded in the UV-responsive microcapsules to the coating surface. The combinations of NIR and UV responses endow the coating with the characteristics of fast healing and large-area healing when damaged by the external environment. In addition, the self-healing superhydrophobic coating film has excellent oil, corrosion, and wear resistance, satisfying the requirements for realistic outdoor applications.

A Novel NIR Fluorescent Nanoprobe Targeting HER2-Positive Breast Cancer: Tra-TTR-A

Meijuan Chen,Zhousheng Lin,Guangyu Yao,Xi Hong,Xiaolei Xue,Lujia Chen

TTRE, a photosensitizer molecule, has excellent biofluorescence imaging performance and effective antitumor properties for breast cancer. However, its application in breast cancer treatment is limited due to poor tumor selectivity and lack of targeting ability. In this study, TTRE and trastuzumab were combined to synthesize Tra-TTR-A, a novel near-infrared fluorescent nanoprobe for HER2 positive breast cancer. The targeting and antitumor abilities of Tra-TTR-A in breast cancer were also investigated. Like TTRE, Tra-TTR-A has a stable structure with remarkable optical properties and in vivo imaging capacity. However, Tra-TTR-A not only inhibits tumor growth by generating reactive oxygen species but also kills tumor cells by trastuzumab. In this study, Tra-TTR-A, a new type of near-infrared fluorescent nanoprobe that targets HER2-positive breast cancer, was successfully synthesized. Tra-TTR-A could be used in in vivo imaging, targeted photodynamic therapy, and diagnosis and treatment for breast cancer.

Hemin as a UCNP quencher for constructing a NIR-excitable nanoprobe for peroxynitrite

Yonggen Hong,Wenjie Yang,Yuxi Liu,Siyu Pan,Qiyuan He,Di Zhang,Yuxia Luo,Decai Yang,Lijun Jiang

The advancement of novel detection strategies, especially those activatable by near-infrared (NIR) light, is of great importance for advancing biomedical sensing. Herein, we report a rationally designed, NIR-excitable nanoprobe, UCNP‑hemin@F127, for the sensitive detection and imaging of peroxynitrite (ONOO - ). The central design feature is the dual role of the biological molecule hemin, which acts simultaneously as a luminescence resonance energy transfer (LRET) acceptor to quench the emission of core‑shell upconversion nanoparticles (UCNP) and as a specific recognition unit for ONOO - . Co‑encapsulated with the UCNP donor within an amphiphilic F127 matrix, hemin enables a highly efficient “turn‑on” sensing mechanism. Upon interaction with ONOO - , the porphyrin ring of hemin is cleaved, disrupting the LRET process and restoring the fluorescence. This nanoprobe allows rapid, selective, and sensitive detection of ONOO - in solution, achieving a detection limit of 37.41 nM and a signal enhancement of up to 7.7‑fold. It demonstrates good biocompatibility and has been successfully applied for visualizing both exogenous and endogenous ONOO - in live cells under 980 nm excitation. This integrated quencher‑sensor strategy, employing a native biomolecule as a key functional module, provides a reliable tool for monitoring ONOO - and offers a versatile design platform for the development of advanced LRET‑based bio-probes.

Non-Small Cell Lung Cancer Treatment via Cu9S8-Mediated NIR-II Photothermal Therapy

Huifang Hao,Zhe Yu,Cheng Cao,Yingao Jiao,Yu Cao,Xinyuan Cui,Yan Gao,Dengyao Gao,Shengsheng Cui,Chenyu Wang,Yanlei Liu

Non-small cell lung cancer (NSCLC) treatment is often compromised by acquired resistance and, in the context of photothermal therapy (PTT), by tumor thermotolerance─a protective stress response mediated by the molecular chaperone heat shock protein 90 (HSP90). To address this dual challenge, we developed a multifunctional nanoplatform (Cu9S8@Lum NPs) that synergistically integrates NIR-II PTT with HSP90 inhibition. This nanosystem is based on a Cu9S8 nanocore with high photothermal conversion efficiency, loaded with the HSP90 inhibitor Luminespib and designed for pH-responsive drug release. This spatiotemporally coordinated strategy enables concurrent tumor hyperthermia and molecular interference with the heat-shock defense mechanism. In vitro, the nanoparticles effectively suppressed HSP90 expression, exacerbated cellular oxidative stress, and induced mitochondrial apoptosis, resulting in markedly enhanced cytotoxicity compared to PTT alone. In vivo, the nanoplatform demonstrated effective tumor accumulation and, under mild NIR-II irradiation, achieved significant tumor growth inhibition in a syngeneic model. This work presents a mechanism-informed nanomaterial strategy to overcome thermotolerance in NSCLC, advancing the development of synergistic cancer nanotherapeutics.

Targeted Inflammatory Microenvironment Remodeling and NIR-II Photothermal Therapy for Abdominal Aortic Aneurysm

Yijun Liu,Peng Qiu,Huifang Hao,Ang’ang Ding,Zhaoyu Wu,Jiahao Lei,Zhaoxi Peng,Hongji Pu,Chenlin Zeng,Hongbin Guo,Xiaodong Wu,Huaxiang Lu,Jinbao Qin,Ruihua Wang,Haoqi Liu,Kaichuang Ye,Xinwu Lu

Abdominal aortic aneurysm (AAA) is a degenerative aortic disease with high rupture-associated mortality. There is no effective medical therapy for early intervention, inflammatory microenvironment remodeling is a crucial therapeutic strategy. Herein, we designed a multifunctional nanoplatform, Cu 9 S 8 @MCC950-Belnacasan-cRGDfK (CS@MBR) for AAA treatment. This platform employed a Cu 9 S 8 core for near-infrared-II (NIR-II) photothermal therapy (PTT), encapsulated NLRP3 inflammasome inhibitors (MCC950 and Belnacasan), and coated with a cRGDfK peptide for active targeting of the aneurysm. In vitro, CS@MBR with mild PTT inhibited NLRP3 inflammasome activation, decreased pro-inflammatory macrophage polarization, and prevented the phenotype switching of vascular smooth muscle cells. In vivo, CS@MBR demonstrated excellent accumulation at the aneurysm site in an angiotensin-II-induced murine AAA model. Upon NIR-II irradiation, the treatment effectively inhibited AAA development, damage of elastic fibers, and imbalance of the inflammatory microenvironment. This study presents a promising, targeted nanotherapeutic strategy for the treatment of AAA and other inflammatory vascular diseases.

A NIR-driven green affording-oxygen microrobot for targeted photodynamic therapy of tumors

Lishan Zhang,Xiaoting Zhang,Hui Ran,Ze Chen,Yicheng Ye,Jiamiao Jiang,Ziwei Hu,Miral Azechi,Fei Peng,Hao Tian,Zhili Xu,Yingfeng Tu

Photodynamic therapy (PDT) is a light-activated local treatment modality that has promising potential in cancer therapy. However, ineffective delivery of photosensitizers and hypoxia in the tumor microenvironment severely restrict the therapeutic efficacy of PDT. Herein, phototactic Chlorella (C) is utilized to carry photosensitizer-encapsulated nanoparticles to develop a near-infrared (NIR) driven green affording-oxygen microrobot system (CurNPs-C) for enhanced PDT. Photosensitizer (curcumin, Cur) loaded nanoparticles are first synthesized and then covalently attached to C through amide bonds. An in vitro study demonstrates that the developed CurNPs-C exhibits continuous oxygen generation and desirable phototaxis under NIR treatment. After intravenous injection, the initial 660 nm laser irradiation successfully induces the active migration of CurNPs-C to tumor sites for higher accumulation. Upon the second 660 nm laser treatment, CurNPs-C produces abundant oxygen, which in turn induces the natural product Cur to generate more reactive oxygen species (ROS) that significantly inhibit the growth of tumors in 4T1 tumor-bearing mice. This contribution showcases the ability of a light-driven green affording-oxygen microrobot to exhibit targeting capacity and O2 generation for enhancing photodynamic therapy.

NIR-Actuated Targeted Janus Nanomotors Remodel Immunosuppressive Tumor Microenvironment for Augmented Cancer Immunotherapy

Xiaoyu Zhou,Enhui Ma,Yingying Zhang,Yujuan Xing,Wenbei Xu,Liang Chen,Hong Zhou,Xinran Zhang,Canran Jiang,Kai Xu,Hong Wang,Shaohui Zheng

Tumor-associated macrophages (TAMs) always display immunosuppressive M2 phenotype in the tumor microenvironment to facilitate tumor growth, invasion and metastasis. Ibrutinib (IBR), a novel irreversible Bruton's tyrosine kinase (BTK) inhibitor, has been employed to repolarize the BTK-overexpressed TAMs from M2 to M1 phenotype to remodel the immunosuppressive tumor microenvironment. However, the poor solubility of IBR extremely hinders its bioavailability, which results in low tumor accumulation and TAMs uptake in vivo. Herein, we proposed NIR laser-actuated Janus nanomotors for the effective and deep delivery of IBR to TAMs in solid tumor for targeted immunotherapy. Under NIR irradiation, the Janus nanomotors exhibited efficient photothermal conversion to produce powerful propulsion via self-thermophoresis with a speed of 12.15 μm −1 s. Combined with the salic acid targeting and IBR loading, the nanomotors significantly boosted their binding and uptake efficacy by M2-like macrophages during the active motion, which highly facilitated the reprogramming of M2 to M1 macrophages in vitro. Furtherly, the autonomous motion also validly improved in-vivo accumulation and penetration depth in tumors to alter the M1/M2 polarization balance and activate T cells. Overall, the synthesized IC@MSA JNMs would provide a promising strategy for the efficient delivery of immunological agents toward targeted cancer immunotherapy. Biomimetic submicromotor with NIR light triggered motion and cargo release inspired by cuttlefish

Jiameng Feng,Junjie Zou,Xiaoyu Li,Xin Du

Biomimetic design is very helpful and significant for the smart construction of micro/nanomotors with artificial intelligence. In this work, inspired by cuttlefish, who can rapidly eject poisonous ink and are also capable of fast movement to escape, we designed and fabricated a biomimetic submicromotor with the ability of simultaneous quick movement and a temperature threshold caused explosive cargo release triggered by near infra-red (NIR) light irradiation, which was approximately equivalent to the action of cuttlefish when encountering a predator. The yolk@shell structured polydopamine@mesoporous silica (PDA@MS60) with immovable and asymmetric yolk distribution was employed as a platform, and this was followed by the simultaneous encapsulation of phase change materials (PCM) and cargo molecules. The NIR light irradiation could not only propel the direct motion of the submicromotor, but also caused the explosive release of the cargo loaded in the submicromotor when the temperature exceeded the melting point of the PCM.

Degradable PDA@PNIPAM-TA Nanocomposites for Temperature- and NIR light-Controlled Pesticide Release

Manting Wang,Rongrong Fan,Qingjian Yu,Jie-Xin Wang,Yuan Le,Jian-Feng Chen

Controlled pesticide delivery systems offer many distinctive advantages over conventional pesticide formulations. In this work, degradable poly(N-isopropylacrylamide) (PNIPAM)–tannic acid (TA) microgels and multifunctional PDA@PNIPAM-TA nanocomposites were prepared in a high-gravity rotating packed bed reactor (RPB) for smart pesticide delivery and release. The as-prepared microgels and nanocomposites showed reversible temperature-dependent swelling/deswelling behavior and irreversible pH-induced degradation. A dynamic contact angle test suggested that the introduction of TA and PDA into the PNIPAM matrix could enhance foliar adhesion and deposition efficiency. The nanocomposites were further used for the encapsulation and delivery of imidacloprid (IMI) to protect it from rapid photolysis and improve its pest-control efficiency. Their thermoresponsive behavior as well as pesticide loading capacity could be tuned by tailoring the PNIPAM-TA shell thickness, which could be varied by the NIPAM amount. The release rate of IMI from the core/shell nanocomposites was positively correlated with environmental temperature and near-infrared (NIR) light, which was adaptive to the positive temperature-dependent toxicity correlation of IMI and the increasing trend of pests under high temperature. The cumulative release of IMI was 23.5% at 25 °C, while it was 81.2% at 40 °C after 24 h of incubation, and the release rate was greatly enhanced under NIR irradiation. The results indicated that the facile control of pesticide release could be realized by regulating environmental conditions. This work also provides an idea for using high-gravity technology to conveniently construct a smart, effective, and environmentally friendly pesticide delivery system for sustainable crop protection.

Temperature-controlled, phase-transition ultrasound imaging-guided photothermal-chemotherapy triggered by NIR light

Wei Li,Wanqing Hou,Xiaomeng Guo,Lihua Luo,Qingpo Li,Chunqi Zhu,Jie Yang,Jiang Zhu,Yongzhong Du,Jian You

Recently, nano-sized ultrasound contrast agents encapsulating drugs for cancer diagnosis and therapy have attracted much attention. However, the ultrasound signal of these agents is too weak to obtain an ideal ultrasound imaging effect. Furthermore, conventional ultrasound contrast agents with strong echo signal are not suitable for drug delivery against cancer because of their large size. To circumvent this problem, phase-transition ultrasound contrast agents are believed to be an excellent choice. Methods: Liposomes co-encapsulating doxorubicin (DOX), hollow gold nanospheres (HAuNS), and perfluorocarbon (PFC) were synthesized by film dispersion method. The morphology, particle size, and stability of these liposomes (DHPL) were investigated. The photothermal effect, drug release, particle size change, cytotoxicity, and ultrasound imaging were studied by using the near infrared (NIR) light. Furthermore, tumor accumulation of DHPL was observed by in vivo fluorescence imaging and the antitumor effect was verified in a 4T1 tumor model. Results: The nanosystem displayed a homogeneous size distribution (~200 nm) and an efficient light-to-heat conversion effect under 808 nm NIR laser irradiation. The nanometer size enabled considerable accumulation of DHPL in the tumor sites. The localized hyperthermia resulting from the photothermal effect of HAuNS could trigger the size transformation of DHPL followed by significant DOX release. Due to the gasification of PFC, a remarkably enhanced ultrasound signal was detected. DHPL also exhibited a prominent photothermally reinforced chemotherapeutic effect under the control of NIR light both in vitro and in vivo . Importantly, no systemic toxicity was observed by DHPL treatment. Conclusion: In this study, we fabricated multi-functional perfluorocarbon liposomes for ultrasound imaging-guided photothermal chemotherapy which have the potential to serve as a prospective cancer treatment approach.
