The persistent challenges of multidrug resistance and systemic toxicity in cancer chemotherapy necessitate the development of intelligent, tumor-targeted co-delivery systems. Herein we report a novel nickel-based metal-organic framework (CUR + PTX@Ni-MOF) engineered with a phosphazene-derived ligand for the pH-responsive co-delivery of paclitaxel (PTX) and curcumin (CUR). The framework integrates a hexachlorocyclotriphosphazene (HCCP)-based linker featuring an intrinsically acid-labile P3N3 core enabling controlled degradation within the acidic tumor microenvironment (TME). The Ni-MOF exhibits a high BET surface area (177.118 m 2 /g) and a mesoporous architecture facilitating exceptional dual-drug loading capacities of 56.15% for CUR (96.3% encapsulation efficiency, EE) and 26.18% for PTX (43.01% EE). In vitro release studies demonstrated minimal drug leakage at physiological pH (7.4), while triggered, sustained release was achieved under acidic conditions (pH 6.5 and 5.0) with maximum cumulative releases of 75.42% for CUR and 49.35% for PTX at pH 5.0. Cellular assays confirmed high biocompatibility with normal LO2 hepatocytes (>80% viability) and potent, synergistic cytotoxicity against HeLa cancer cells driven by enhanced cellular uptake and apoptosis induction. This work establishes CUR + PTX@Ni-MOF as a promising, intrinsically pH-responsive platform for targeted and synergistic combination chemotherapy.

Green tea, a commonly consumed beverage worldwide, is extensively used as dietary supplements for cancer prevention. However, the interaction between green tea and paclitaxel is still unknown. This study aimed to systematically investigate the herb-drug interaction of green tea consumption on the pharmacodynamics and pharmacokinetics of paclitaxel. The ATP assay results indicated that green tea extract (GTE) significantly enhanced the cytotoxicity of paclitaxel in both human and murine breast cancer cells. Further, the 4T-1 tumor model testified that daily green tea consumption could improve the therapeutic effects of paclitaxel on breast carcinoma. The pharmacokinetic profiles revealed that in the group with short-term green tea consumption (0.5 h), there was a more than 55.87% increase in the maximum plasma concentration ( C max ) of paclitaxel and a 38.49% increase in the area under the plasma concentration curve (AUC). In the long-term green tea consumption group (2 weeks), there was a 23.80% increase in the C max of paclitaxel and a 28.29% increase in the AUC. Moreover, long-term green tea consumption significantly reduced hepatic microsomal protein level. Taken together, green tea consumption enhances the anticancer efficacy and increases the pharmacokinetic profile of paclitaxel, which provides information to study the interaction between herbs and chemotherapy.

Immunotherapy shows promise for tumor control but is limited by low response rates. Paclitaxel (PTX) induces immunogenic cell death (ICD), yet conventional delivery systems face challenges like low drug loading and insufficient intracellular accumulation, reducing ICD efficacy. Small-molecule self-assembled PTX nanoparticles offer a promising solution due to high drug loading and dose delivery. In this study, PTX was conjugated with phenylboronic acid (PBA) to form the derivative PTX-PBA, which spontaneously self-assembled with fructose into nanoparticles (PTX-PBA-Fru NPs). These nanoparticles exhibited a uniform size of 107.8 ± 2.9 nm, a PDI of 0.064 ± 0.042, and a zeta potential of -12.2 ± 0.9 mV, with spherical morphology. In 4T1 tumor-bearing mice, PTX-PBA-Fru NPs significantly enhanced tumor inhibition (p < 0.001) and increased body weight (p < 0.05). No allergic reactions in healthy Balb/c mice and the maximum tolerated intravenous dose reached 200 mg/kg, underscoring its favorable safety profile of PTX-PBA-Fru NPs. The ICD effects induced by PTX-PBA-Fru NPs, when combined with the immunomodulator resiquimod (R848), elicited a robust anti-tumor immune response. This combination therapy effectively remodeled the immunosuppressive tumor microenvironment and achieved a 37.5% tumor eradication rate. Moreover, it established long-term immune memory, providing protection against tumor re-challenge. This novel PTX formulation demonstrates strong anti-tumor effects, safety, and clinical potential in combination with R848-based immunotherapy.

Tumor-targeted drug delivery and microenvironment-responsive drug release are attractive strategies in cancer treatment. Our previous study demonstrated that redox-sensitive micelles based on hyaluronic acid-deoxycholic acid (HA-ss-DOCA) conjugates exhibited excellent drug-loading capacities (34.1%) for paclitaxel (PTX) and rapid drug release in response to reducing agent, glutathione . In the present study, the physicochemical and biological properties of PTX-loaded HA-ss-DOCA (PTX-HA-ss-DOCA) micelles were investigated further. The micelles have an average size of about 120 nm and a zeta potential of about −36 mV. Transmission electron microscopy and wide-angle X-ray diffraction analysis demonstrated redox-sensitive degradation of micelles in the presence of glutathione. Moreover, the encapsulated payload was effectively released from HA-ss-DOCA micelles into cytoplasm and then rapidly transported into nuclei. In vitro cytotoxicity and cell apoptosis assay further revealed that HA significantly improved the tumor-specific drug delivery of HA-ss-DOCA micelles via receptor-mediated endocytosis, while efficient intracellular drug release and transportation lead to marked inhibition of tumor cell growth, as compared to Taxol ® and insensitive micelles. More importantly, PTX-HA-ss-DOCA micelles demonstrated superior in vivo antitumor activity compared with Taxol ® and insensitive control, and decreased systemic toxicity. Herein we present data which provide valuable insight into the design and development of tumor-specific drug delivery systems .

Ferroptosis, which relies on iron (Fe) ions for the production of reactive oxygen species (ROS), is an effective approach for treating liver cancer. Cells will activate several mechanisms, such as decreasing Fe absorption and sequestering surplus Fe in ferritin, to diminish intracellular Fe levels. This study presents a straightforward, one-pot approach for the fabrication of bovine serum albumin (BS)-stabilized MnFe2O4 (MnFe@BS NPs) intended for liver cancer treatment in ferroptosis therapy. Cabozantinib (CA) and paclitaxel (PA), which can promote ferritin breakdown and augment uptake by elevating transferrin receptor and binding protein IRP2 levels, were encapsulated in MnFe@BS NPs to improve the liver cancer cytotoxic properties. The fabricated MnFe@BS-CA/PA NPs substantially elevate cellular Fe concentration, hence augmenting ROS production within HepG2 cells. Thus, the bimetallic nanozyme demonstrated a synergistic effect of MnFe, CA, and PA, providing a foundation for improving ferroptosis therapy for liver cancer.

Introduction: Breast cancer is one of the most common cancers in the world. Paclitaxel (PTX), a potent chemotherapeutic agent, is widely used in cancer treatment but suffers from poor aqueous solubility and systemic toxicity. Methods: In this study, we developed a thermo-sensitive peptide, TSP-5, which forms a stable hydrogel at 37 °C and dissociates upon heating to 45 °C to facilitate drug release. The hydrogel was co-loaded with PTX and photothermal agent antimony nanosheets (AM) for targeted delivery to tumor sites. Under near-infrared (NIR) laser irradiation, AM rapidly elevated the local temperature due to its high photothermal conversion efficiency, triggering hydrogel dissolution and subsequent PTX release. Simultaneously, AM-mediated photothermal therapy (PTT) contributed to tumor cell ablation. Results: Both in vitro and in vivo studies demonstrated that the TSP-5 hydrogel significantly enhanced PTX solubility, reduced systemic toxicity, and improved anti-tumor efficacy.Discussion: The findings highlight its potential as a promising platform for localized drug delivery.

This study investigated the rapid drug delivery capabilities of neutrophil-like cells using a microfluidic chip-based mechanical deformation approach, with an emphasis on glioblastoma treatment at the cellular level. We designed a microfluidic chip comprising multiple contraction gaps and parallel microchannels to enable efficient drug loading into HL-60 cells and neutrophil-like cells derived from differentiated HL-60 cells (dHL-60 cells). Optimization was performed using dye molecules, including FITC-dextran (4 kDa, 20 kDa) and FITC-BSA, with delivery efficiency and the cell recovery rate serving as critical evaluation parameters. The optimal performance was achieved at a gap size of 8 µm and a flow rate of 150 µL/min; for FITC-BSA, a concentration of 300 µg/mL was deemed suitable. Under these conditions, neutrophil-like cells loaded with albumin-bound paclitaxel (PTX-ALB) were successfully and rapidly prepared, yielding a delivery efficiency of 55.93 ± 19.7% and a drug loading of 784.20 ± 74.6 ng/105 cells, with a throughput of up to ~ 107 cells per hour. The chemotactic performance of PTX-ALB-loaded neutrophil-like cells did not change significantly, and these cells could cross the endothelial barrier constructed in vitro and exerted antitumour effects on U87-eGFP cells. The antitumour effects could be further strengthened by increasing the dosage of drug-loaded cells (from 4× 105 cells to 1× 106 cells) and extending the treatment duration (from 48 hours to 72 hours), which reduced U87-eGFP cell viability to 80±7% and 62±3%, respectively. This microfluidic-based mechanically mediated cargo delivery platform represents a rapid, high-throughput strategy for cell-based drug loading, with broad potential in cellular therapy and immunotherapy.

Paclitaxel (PTX) is a potent chemotherapeutic agent for solid malignancies. However, its poor aqueous solubility necessitates toxic solubilizers in conventional formulations, which lead to adverse reactions and unpredictable pharmacokinetics. Liposomal encapsulation offers a biocompatible alternative to overcome these limitations. In this study, a sensitive ultrafiltration coupled with UPLC-MS/MS method was developed and validated for the simultaneous determination of free and total PTX in biological samples. This method was applied to compare the pharmacokinetic behavior of liposomal PTX (PTX-Lip) and conventional PTX injection in rats. Despite receiving a double dose (10 mg/kg), rats treated with PTX-Lip exhibited significantly lower dose-normalized initial concentration (C 0 /Dose) and systemic exposure (AUC (0-∞) /Dose) along with a prolonged elimination half-life (t 1/2 ) compared to the conventional formulation (5 mg/kg), indicating sustained release. Tissue distribution studies revealed extensive and persistent accumulation of PTX-Lip in the spleen and liver, consistent with reticuloendothelial system (RES) uptake, while distribution to other tissues was reduced. Excretion profiling demonstrated a fundamental shift in clearance pathways, from combined renal/hepatobiliary elimination for conventional PTX to predominantly hepatobiliary fecal excretion for PTX-Lip, with cumulative fecal recovery reaching 39.27% of the dose, a 2.4-fold increase over the conventional group (16.50%). Collectively, these findings illustrate how liposomal encapsulation modifies the in vivo disposition of PTX through RES-mediated sequestration, controlled drug release, and rerouted elimination. These descriptive observations, generated in healthy rats without pharmacodynamic or efficacy endpoints, provide a pharmacokinetic reference for the rational design of liposomal nanomedicines. However, these findings should be interpreted as a baseline pharmacokinetic characterization rather than as evidence of therapeutic benefit, and they are intended to generate hypotheses for validation in appropriate disease models with efficacy and safety endpoints.

Amphiphilic polymers (HA-ANI) were prepared by grafting hyaluronic acid (HA) and 6-(2-nitroimidazole)hexylamine (ANI) and then self-assemble in water to form nanoparticles (NPs) that could be loaded with paclitaxel (PTX) and gemcitabine (GEM) by dialysis. Infrared spectroscopy and 1 H-NMR indicated the successful synthesis of HA-ANI. Three different ratios of NPs were prepared by adjusting the ratios of hydrophilic and hydrophobic materials, and the particle size decreased as the ratio of hydrophilic materials increased. When HA:ANI = 2.0:1, the nanoparticles had the smallest size distribution, good stability and near spherical shape and had high drug loading and encapsulation rates. In vitro release experiments revealed that NADPH could accelerate the drug release from NPs. Cellular uptake rate reached 86.50% at 6 h. The toxic effect of dual drug-loaded nanoparticles (P/G NPs) on MDA-MB-231 cells at 48 h was stronger than that of the free drug. The AO/EB double-staining assay revealed that a large number of late apoptotic cells appeared in the P/G NPs group, and the degree of cell damage was significantly stronger than that of the free drug group. In the cell migration assay, the 24 h-cell migration rate of the P/G NPs group was 5.99%, which was much lower than that of the free group (13.87% and 17.00%). In conclusion, MDA-MB-231 cells could effectively take up P/G NPs, while the introduction of the nano-codelivery system could significantly enhance the toxicity of the drug to MDA-MB-231 cells as well as the migration inhibition effect.

In this study, a polymer with tumor targeting function and dual redox/pH sensitivity is designed and synthesized by integrating chitosan (CS), Poloxamer 407 (P407), cholesterol (CHO), folic acid (FA), and disulfide bonds into one molecule (P407-CS(FA)-SS-CHO). The polymer can self-assemble in water to form micelles and solubilize paclitaxel (PTX). The mean particle size of PTX-loaded P407-CS(FA)-SS-CHO micelles is 133 nm. In vitro drug release study demonstrates the acid and glutathione (GSH) triggered PTX release behavior of the micelles. In vitro and in vivo experiments demonstrate the excellent tumor growth inhibition efficacy and low systemic toxicity of the PTX-loaded micelles. The P407-CS(FA)-SS-CHO micelles will be potential intelligent drug carriers for PTX delivery.

Purpose Cervical cancer is one of the most common causes of death among women globally. Combinations of cisplatin, paclitaxel, bevacizumab, carboplatin, topotecan, and gemcitabine are recommended as first-line therapies. Methods This study focuses on the development of folate-decorated, pH-sensitive lipid-polymer hybrid nanoparticles (LPNs). Loading carboplatin (CBP) and paclitaxel (PTX), LPNs were expected to combine the therapeutic effects of CBP and PTX, thus show synergistic ability on cervical cancer. Results FA-CBP/PTX-LPNs showed the sizes of 169.9 ± 5.6 nm, with a narrow size distribution of 0.151 ± 0.023. FA-CBP/PTX-LPNs exhibited pH-responsive drug release, high cellular uptake efficiency (66.7 ± 3.1%), and prominent cell inhibition capacity (23 ± 1.1%). In vivo tumor distribution and tumor inhibition efficiency of FA-CBP/PTX-LPNs was the highest, with no obvious body weight lost. Conclusion High tumor distribution and remarkable antitumor efficiency obtained using in vitro as well as in vivo models further proved the FA-CBP/PTX-LPNs is a promising tool for cervical cancer therapy.
