Curcumin is a dominating active component of Curcuma longa and has been studied widely because of its prominent biological activities. The extremely low aqueous solubility, stability, and bioavailability of curcumin limit its application in the field of medicine. In this study, we developed pectin–curcumin (PEC–CCM) conjugates that could self-assemble water-soluble nanomicelles in aqueous solution. The structure of PEC–CCM conjugates was characterized by ultraviolet–visible spectra, fluorescence spectra, Fourier transform infrared spectroscopy, and 1H nuclear magnetic resonance spectroscopy. The thermal property of PEC–CCM conjugates was investigated by thermogravimetric analysis. It was found that PEC–CCM conjugates had formed nanomicelles in aqueous medium via self-assembly. These nanomicelles were observed as small spheres or ellipsoids and aggregated with a size range of 70–190 nm by transmission electron microscopy analysis. In a solution of nanomicelles, the stability of curcumin was improved, and its antioxidant property was preserved. The anticancer activity of PEC–CCM conjugates was quantified by the MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] assay using a hepatic cancer cell line (HepG2), a breast cancer cell line (MCF-7), a cervical cancer cell line (HeLa), and a human normal kidney cell line (293A). It was found that the curcumin of PEC–CCM conjugates had a more significant inhibitory effect on cancer cells and was less cytotoxic to normal cells than free curcumin was. PEC–CCM conjugates have great potential for some food and pharmaceutical applications.

Thermo-responsive hydrogels with biodegradability have great application in biomedical areas, while the Poly(N-isopropylacrylamide) (PNIPAM) based hydrogels exhibiting lower critical solution temperature (LCST) around body temperature have shown promising applications in diverse areas such as drug delivery, tissue engineering, and wound repairing. In this study, PNIPAM based thermo-responsive copolymer with ketone functional group was polymerized and the LCST was regulated to body temperature, then biodegradable self-healing hydrogels with injectability were fabricated by cross-linking the thermo-responsive copolymers with dopamine functionalized pectin hydrazide (PDAH). The hydrogel exhibited good mechanical strength, excellent biocompatibility and thermo-responsive sustained drug release process. The nano-graphene oxide (GO) was incorporated to design near-infrared (NIR) sensitive nanocomposite hydrogel to serve as photothermal enhanced wound dressing with vancomycin loading. The composite hydrogel also showed pH-responsive drug release behavior and the NIR irradiation also accelerated the release rate based on photothermal induced phase transition. More importantly, the nanocomposite hydrogel with vancomycin loading and photothermal property can promoted burn wound repairing rate by inhibiting the bacterial growth. As a result, this GO composite hydrogel (GO/hydrogel) could act as NIR triggered photo-thermal enhanced drug delivery platform for promoted wound repairing in the near future.

Background The prevalence of obesity has raised global concerns. Environmental pollutants are one of the main causes of obesity. Many studies have demonstrated that dietary fiber could reduce obesity induced by high-fat diets, but whether environmental pollutant-induced obesity can be reversed is still unknown. Objectives This study aimed to investigate the effects of pectin on obesity induced by a typical environmental pollutant p,p'-dichlorodiphenyldichloroethylene (p,p′-DDE) and explore the underlying mechanism by which pectin reversed p,p′-DDE-induced obesity. Methods p,p′-DDE was used to induce obesity in C57BL/6J mice and pectin was supplied during and after cessation of p,p'-DDE exposure. Body and fat weight gain, plasma lipid profile and insulin resistance of mice were assessed. Gut microbiota composition and the levels of short-chain fatty acids (SCFAs) as well as the receptor proteins and hormones in the SCFAs-related signaling pathway were analyzed. Moreover, p,p′-DDE levels in various tissues of mice were detected. Results Pectin supplementation reversed body and fat weight gain, dyslipidemia, hyperglycemia and insulin resistance in p,p′-DDE-exposed mice. Furthermore, pectin apparently altered the p,p′-DDE-induced microbial composition and then promoted the levels of SCFAs in colonic feces as well as the expression of G-protein coupled receptors and the concentration of hormone peptide YY (PYY) and glucagon like peptide-1 (GLP-1). Pectin treatment also significantly reduced p,p′-DDE accumulation in mice tissues during p,p′-DDE exposure but did not change p,p′-DDE metabolism after termination of p,p′-DDE exposure. Conclusions Pectin had a good effect on reducing p,p′-DDE-induced obesity through regulating gut microbiota and provided a potential strategy for the treatment of environmental pollutant-caused health problems.

Hepatocellular carcinoma (HCC) is one of the greatest public health problems worldwide, and chemotherapy remains the major approach for the HCC treatment. Doxorubicin (DOX) is one of the anthracycline antibiotics but its clinical use is limited due to its severe cardiotoxicity. In this study, novel hybrid nanoparticles by self-assembling based on pectin-doxorubicin conjugates (PDC-NPs) were fabricated for HCC treatment. The stabilized structure of the PDC-NPs was characterized by methylene blue absorption, the size, zeta potential and the morphology, which was investigated by Zetasizer nanoparticle analyzer and transmission electron microscope (TEM), of nanoparticles. The PDC-NPs achieved a sustained and prolonged release ability, which was illustrated with in vitro drug release profiles, anti-cell proliferation study, cellular uptake assay and in vivo pharmacokinetics analysis. Biocompatibility of the PDC-NPs was assessed with bovine serum albumin (BSA) adsorption test, hemolysis activity examination and viability evaluation of human umbilical vein endothelial cells. Importantly, in vivo studies of the PDC-NPs, which were performed in the athymic BALB/c nude mice, demonstrated that the PDC-NPs significantly reduced the lethal side effect of DOX. Additionally, the H&E staining and serum biochemistry study further confirmed the excellent biological security of the PDC-NPs.

Summary To improve long-term stability of the liposomes, resveratrol (RES) liposomes, epigallocatechin gallate (EGCG) liposomes, RES+EGCG liposomes, RES+EGCG liposomes coated with low methoxyl pectin (LMP) were prepared, and their physical properties were evaluated. The prepared liposomes have a particle sizes of about 110 – 160 nm. The LMP-coated composite liposomes have the largest particle size and zeta potential, the latter indicating higher system stability. Liposome systems were pasteurised and placed in an orange juice system as to study liposome stability in a real the food environment. LMP bridge with metal ions forming a network-like gel in the LMP-coated composite liposomes, thereby enhancing the stability of liposomes. LMP-decorated liposomes could be developed as a formulation for encapsulating and delivering functional ingredients.

The development of environmentally benign, transparent, durable, and healable anti-fogging coating is vital for automobile windshields and food packaging in our daily production and life. The fabrication of existing anti-fogging coatings is often time-consuming in addition to potential toxicity issues. Here, we have reported a highly transparent and healable anti-fogging coating on the surface of poly(ethylene terephthalate) (PET) film by using bioderived pectin and tannic acid (TA) via a facile dip-coating method. The results show that the pectin/TA composite coating endows the PET film with excellent anti-fogging property without sacrificing the transparency of the PET films. This coating can maintain exceptional anti-fogging capabilities in both hot-vapor and cold-warm conditions. In addition, such anti-fogging coatings also exhibit good durability as the anti-fogging capability can be well-retained even after spray rinsing 5 times with high-speed water faucet. Moreover, the scratches or cuts on the coating can be completely healed within 4 minutes of triggering with water. The study provides an innovative methodology for the design of highly transparent, durable, and healable anti-fogging coatings, and offers promising value-added utilization of abundant and renewable biomass.

In the present study, acoustic cavitation assisted extraction (ACAE) which is a green approach was used for two-stage extraction of pectin from waste grapefruit peels regarding the specific two stages and different peel particle sizes. The yields, physicochemical properties and structure prediction of pectin from different peel particle sizes and different stages by two methods were compared. Highest pectin yield of 23.49% was achieved at the peel size of 0.9 mm and was slightly higher than the yield of 23.44% at finer particle size. ACAE achieved four times higher yield compared with conventional heating method (CHE) at the second stage. ACAE pectin which had lower molecular weight and degree of methoxylation was richer in rhamnogalacturonan-I (RG-I) region with long side chains compared with CHE pectin from chemical and FT-IR analysis. Based on the results, a potential general mechanism for ACAE of plant cell wall viscous polysaccharides was discussed in terms of the effects of cavitation on the swelling index and morphological study of residue to reveal the “barrier effect” during the process. Furthermore, the energy consumption for ACAE was significantly lower than the conventional method, indicating its promising application in industrial scale.

The development of biodegradable bone scaffolds that align with the growth cycle of new bone has become a prominent focus in the field of bone defect repair. The sintered porous hydroxyapatite scaffolds exhibit a high degree of crystallization and demonstrate a slow degradation rate following implantation. The apatite phase with low crystallinity can be obtained following the hydration reaction of the non-sintered calcium phosphate bone cement (CPC) scaffolds. However, this leads to the formation of a weak alkaline environment during degradation, ultimately resulting in unsustainable degradation. The present study focuses on the fabrication of porous pectin/CPC composite scaffolds through two approaches during the preparation process of non-sintered porous CPC scaffolds: uniform composite and surface modification. The degradation performance of porous composite scaffolds exhibited an increase with the rise in pectin content. At a pectin content of 70 mg/mL, the scaffolds demonstrated a degradation rate of 13.79% within a span of 30 days. The mechanical properties of the porous scaffolds were enhanced with an increase in pectin concentration through surface coating. When the pectin concentration was 5 wt%, the scaffold exhibited a compressive strength of 8.72 MPa, an elastic modulus of 0.82 GPa, and experienced a degradation rate of 12.57% after a period of 30 days. The local acidic environment not only facilitates the dissolution of bone-like apatite but also enhances cell proliferation and adhesion. Pectin/apatite porous scaffolds exhibit excellent biocompatibility, offering a novel approach for the development of biodegradable bone replacement scaffolds.

Previous studies have shown that the organic solvent-free method has considerable potential for delivery systems in food applications. In this study, Nanocomplex of zein and pectin from apple (AP) or citrus (CP) was systemically fabricated and characterized by adjusting pectin pH (2.0–11.0) and mass ratios of zein:pectin (1:0, 1:0.5, 1:1, 1:2 and 1:3) via one-step pH-shifted process. Results showed that the size of zein/pectin nanocomplex remarkably decreased from >500 nm to ∼150 nm with the increasing pH level of pectin solution and proportion of pectin addition. Spherical shape was observed for nanocomplex formed by AP and CP in transmission electron microscopy images. Compared with zein/AP nanocomplex , zein/CP nanocomplex exhibited the smaller size, smoother surface and lower surface hydrophobicity, prompting us to conclude that CP, as compared to AP, had better performance. To further explore the mechanism, the behavior difference could be interpreted by the altered hydrophobic interaction between zein and pectin as revealed by fluorescence spectroscopy and Fourier transform infrared spectroscopy (FTIR). Findings in this study would facilitate the understanding of interaction between zein and pectin and provide new perspectives of pH-shift induced zein nanocomplex.

ABSTRACT Citri Reticulatae Pericarpium ‘Chachi’ ( Citrus reticulata ‘Chachi’) is a traditional Chinese medicine with dual medicinal and dietary uses. As its main component, pectin exhibited various biological activities. However, research and development on it, especially pectin oligosaccharides, remain limited. In this study, ‘Chachi’ pectin oligosaccharides (CPOS) was extracted and prepared for the first time, resulting in the purification of two oligosaccharide components, CPOS3 and CPOS4. Structural characterization of the predominant oligosaccharide, CPOS3, revealed it to be composed of a backbone α-D-Gal p A-(1→2)-α-L-Rha p -(1→4)-α-D-3-OAc-Gal p A-(1→4)-α-D-Gal p A-(1→4)-α-D-Gal p A and a side chain [→5)-α-L-Ara f -(1→] 4 attached at the C-4 of Rha p . Subsequent experiments have shown that CPOS3 can significantly promote the proliferation and migration of human immortalized keratinocyte cell. Further investigation revealed that it facilitated cell proliferation by stimulating DNA synthesis in the S phase of the cell cycle. Additionally, CPOS3 exhibited good thermal stability and rheological characteristics. Based on the good wound healing activity and favorable physicochemical properties of CPOS3, its prospects in the fields of wound dressings and tissue repair are promising.

Ulcerative colitis (UC) is a persistent intestinal disorder featuring periodic flare-ups of the colon's inner lining inflammation. Current therapeutic strategies, while effective in managing symptoms, are often limited by side effects and high costs. This study investigates the potential of pectin-loaded ferulic acid (PC-FA) nanoparticles as a novel therapeutic approach for UC, focusing on their ability to modulate the cGAS-STING pathway, a key mediator in the inflammation associated with UC. PC-FA nanoparticles were prepared and characterized for their physicochemical properties, antioxidant capacity, biocompatibility, and influence on the cGAS-STING pathway. In vitro experiments demonstrated that PC-FA nanoparticles enhanced the solubility and bioavailability of ferulic acid (FA), reduced oxidative stress, and protected colon epithelial cells from damage caused by the administration of dextran sulfate sodium (DSS). In vivo studies in a DSS-induced colitis mouse model showed that PC-FA nanoparticles mitigated weight reduction, lowered disease activity index (DAI) scores, and sustained colon length, and ameliorated histopathological changes. Additionally, PC-FA nanoparticles effectively targeted DNA damage and inhibited the cGAS-STING pathway, leading to a significant reduction in pro-inflammatory cytokines. Pharmacokinetic studies revealed rapid absorption of PC-FA in the bloodstream, with a predominant distribution in the intestines. The study concludes that PC-FA nanoparticles are a promising therapeutic strategy for UC, offering targeted drug delivery, enhanced bioavailability, and anti-inflammatory effects.

In this study, nitrogen-doped carbon dots (N-CDs) were synthesized via a one-step hydrothermal method using pectin polysaccharide as the carbon source, with 1-hexylamine, 1,2-diaminocyclohexane, and diethylenetriamine as individual nitrogen sources. The chemical composition, fluorescence properties, and morphology of the synthesized N-CDs were characterized. Their corrosion inhibition performance on N80 steel in 1 M H 2 SO 4 was evaluated using electrochemical analyses and weight loss experiments. Tafel and EIS analyses demonstrated that at a concentration of 200 mg/L, the maximum inhibition efficiencies of PHA-CDs, PCA-CDs, and PDA-CDs, synthesized using monoamine, diamine, and triamine as nitrogen sources, were 75.3 %, 90.1 %, and 95.4 %, respectively. The inhibition efficiency increased with the total nitrogen and pyrrolic N content in the N-CDs, corresponding to the choice of nitrogen source. Weight loss experiments further confirmed these trends. Additionally, the N-CDs were found to act as mixed-type inhibitors with a predominant anodic inhibition effect and are classified as predominantly chemisorptive. The adsorption behavior adhered to the Langmuir adsorption model, where coordination complexes formed between pyrrolic N and metal Fe increase the surface coverage of the adsorption film, thereby enhancing corrosion resistance.
