Construction and application of sustained-release active intelligent packaging incorporating phenolic acid copigmented Aronia melanocarpa anthocyanins and microencapsulated carvacrol

Liu Yang,Xue Ding,Rui Xiao,Xinyu Zhang,Zhipeng Jiang,Wenwen Chen,Shengyu Xu,Hongyuan Zhang

Active intelligent packaging often suffers from insufficient indicator sensitivity and limited antibacterial duration. To overcome these limitations, the system incorporates Aronia melanocarpa anthocyanins (AMA) copigmented by phenolic acid, carvacrol (CL) encapsulated by hydroxypropyl-β-cyclodextrin (HP-β-CD), and integrates these components into potato starch (PS) to produce a multifunctional film with enhanced indicator sensitivity and sustained release properties. The results revealed that ferulic acid (FA) provided notable stabilizing effect on AMA, maintaining ΔE < 1 after two cycles of acid-base alternation. The incorporation of HP-β-CD allowed for efficient encapsulation of CL through hydrogen bonding, achieving sustained release over 96 h. The incorporation of FA-AMA and CL@HP-β-CD system improved the roughness, UV-blocking, and antioxidant performance of the film. Additionally, the film extended shrimp shelf life by 12 h, with a visible transition from pink to blue upon spoilage. This research offers a novel approach to the advancement of active intelligent packaging.

Preparation and in vitro/in vivo evaluation of a clonidine hydrochloride drug–resin suspension as a sustained-release formulation

Hongfei Liu,Xiaoya Xie,Chao Chen,Caleb Kesse Firempong,Yingshu Feng,Limin Zhao,Xuezhi Yin

Objective The purpose of the present study was to prepare a clonidine hydrochloride (CH) sustained-release suspension. Methods The processes involved in the drug formulation included drug loading, impregnating, and suspension preparation. Clonidine hydrochloride drug–resin complexes (CH-DRC) were prepared using the bath method and the CH-DRC impregnated before the microencapsulation process. Based on the bottom spray fluidized bed coating method, the CH microencapsulated drug–resin complexes (CH-MC) were also prepared using Surelease® (the suspension of ethyl cellulose aqueous dispersion) as the coating material. The effects of coating (process/formulation) on the in vitro release of coating microcapsule were evaluated via single factor investigation and orthogonal design optimization. The CH-MC with optimized formulation was further dispersed in a suitable medium to obtain a sustained-release suspension. Rats were given commercial CH ordinary tablets and the CH sustained-release suspension via intragastric administration. The plasma concentration–time curve and related pharmacokinetic parameters were investigated using the non-compartment model. Results The Tmax of the CH sustained-release suspension was delayed from 2 h to 5 h compared with the CH ordinary tablets. Similarly, the Cmax was reduced from 32.138 µg·mL−1 to 18.150 µg·mL−1 with the concentration–time curve being more gentle compared with the commercially CH ordinary tablets. After oral administration, the relative bioavailability of CH sustained-release suspension (AUC0–24 of 137.703 µg·h·mL−1) to its CH ordinary tablets (AUC0–24 of 123.337 µg·h·mL−1) was 111.65%.Conclusions The findings showed that the CH sustained-release suspension for oral administration was successfully formulated.

Hyaluronic Acid-Modified Celecoxib-Loaded Nanostructured Lipid Carriers Combined with Dissolving Microneedles for Targeted Sustained-Release Therapy in Osteoarthritis

Junfeng Ban,Yichao Li,Yanhua Guo,Huashen He,Chuangzan Yang,Peining Li,Jiawei Liang,Shirui Li,Xueyuan Luo,Yu Li,Yi Xie,Xiangmei Liu,Xiaofang Li

Background Rapid local drug clearance and poor sustained retention pose challenges to the treatment of osteoarthritis. Therefore, a local delivery system that prolongs intra-articular drug retention and achieves efficient targeted delivery holds significant clinical promise. Methods Hyaluronic acid-modified celecoxib nanostructured lipid carriers (HA-Cxb-NLCs) were prepared and encapsulated in dissolving microneedles to form a composite delivery system (HA-Cxb-NLCs-DMNs). The system was characterised in terms of its physicochemical properties, in vitro drug release profiles, transdermal permeation behaviour, cellular compatibility, and uptake mechanisms. Subsequently, a sodium iodoacetate-induced rat osteoarthritis model was used to evaluate the therapeutic efficacy in vivo. Results HA-Cxb-NLCs-DMNs exhibit a uniform particle size (113.93 ± 1.66 nm) and a zeta potential of (−39.16 ± 3.00 mV). In vitro release persisted for 72 hours, with a cumulative permeation of 62.74 ± 1.40 μg·cm−2 and a skin retention of 3.20 ± 0.62 μg·cm−2. HA modification enhances cellular uptake of the nanocarriers via the CD44 receptor. Animal studies demonstrated that it reduces joint swelling, improves cartilage damage, and inhibits MMP-13 expression. Conclusion This study presents a promising microneedle-nanoparticle synergistic delivery strategy for the local targeted therapy of osteoarthritis.

Transforming sustained release into on-demand release: self-healing guanosine–borate supramolecular hydrogels with multiple responsiveness for Acyclovir delivery

Cheng-guang Wu,Xiang Wang,Yun-feng Shi,Bin-cheng Wang,Wei Xue,Yi Zhang

Supramolecular hydrogels derived from natural biomolecules have promising applications for drug delivery due to their inherent biocompatibility and tunable responsiveness to various stimuli. However, conventional hydrogels only modulate the release kinetics roughly to achieve sustained drug release, exhibiting fast-then-slow release behavior without on/off control. Herein, a guanosine (G)-quartet·Na+-borate supramolecular hydrogel (GB hydrogel) cross-linked via a guanosine–borate diester and intertwined by G4-nanofibres formed by π–π stacking of G4-quartets stabilized by Na+ is developed for on-demand release of Acyclovir (Acv). This GB hydrogel is facilely prepared by a one-pot hierarchical assembly involving hydrogen bonds, dynamic borate ester bonds and cation coordination, which endow it with tunable mechanical properties, excellent self-healing properties and reversible degradation behavior in response to pH, glucose and ion concentration. Benefiting from that the guanosine analog Acv is able to assemble into a G4-quartet by replacing guanosine via reversible hydrogen bonding, the Acv-loaded GB hydrogel showed favorable stability in physiological medium without undesired release and achieved external stimulus-triggered on-demand release with switchable on/off control and tunable release kinetics. Moreover, the GB hydrogel also exhibited excellent in vitro and in vivo biocompatibility. Such a natural nucleoside-based supramolecular hydrogel with on-demand drug release, self-healing property, biodegradability and biocompatibility provides a precisely controlled paradigm to overcome early burst release behavior of conventional hydrogels for the development of injectable hydrogel delivery systems.

Sustained Release of BMSC-EVs from 3D Printing Gel/HA/nHAP Scaffolds for Promoting Bone Regeneration in Diabetic Rats

Tingting Yang,Yunsheng Dong,Jinpeng Wan,Xiangsheng Liu,Yufei Liu,Jiaxing Huang,Jie Zhou,Hui Xiao,Lizong Tang,Yanying Wang,Shufang Wang,Hong Cai

Extracellular vesicles (EVs) play an important role in intercellular communication, and the function of EVs mainly depends on the state of source cells. To determine the effect of diabetic microenvironment on EVs secreted by bone marrow mesenchymal stem cells (BMSCs), this work explores the effect of normal glucose (5.5 m m ) cultured BMSCs derived EVs (NG-EVs) and high glucose (30 m m ) cultured BMSCs derived EVs (HG-EVs) in regulating the migration, proliferation and osteoblastic differentiation of BMSCs in vitro. In order to improve the bioavailability of EVs, this work constructs a sustained release system of polydopamine (PDA) functionalized 3D printing gelatin/hyaluronic acid/nano-hydroxyapatite (Gel/HA/nHAP) scaffolds (S/PDA) and verifies its function in the calvarial defect model of diabetic rats. This work confirms that both NG-EVs and HG-EVs can promote proliferation and migration, inhibit apoptosis and promote osteogenic differentiation, but the function of HG-EVs is weaker than that of NG-EVs. Therefore, EVs secreted by autologous cells of diabetic patients are not suitable for self-repair. This work hopes that the 3D printing scaffold designed for sustained-release EVs will provide a new strategy for acellular tissue engineering bone repair in diabetic patients.

Characterization and antivibrio activity of chitosan-citral Schiff base calcium complex for a calcium citrate sustained release antibacterial agent

Ran Tao,Ning Zhang,Liangliang Zhang,Théogène Habumugisha,Yicun Chen,Yin Lu,Yinjuan Wang,Kui Wang,Yangdong Wang,Jianchun Jiang

Vibrio parahemolyticus is the “Number one killer” of seafood products. Anti-vibrio agents having low cost and high-safety are urgently needed to supplement the application needs. This work attempted to prepare CS-CT-CCa complex with citral (CT), chitosan (CS) and calcium citrate (CCa) as raw material by microwave-assisted high-pressure homogenization. Additionally the coordination structure and morphology of Bridge-CS-CT-Schiff base/OH-CCa were verified. The prepared CS-CT-CCa had a well-dispersed property (the size: 3.55~9.33 μm and the zeta potential: +38.7~+67.5 mV) and an excellent sustained released ability (sustained release up to 180 min). MIC, Glucose assay, MDA assay, biofilm formation inhibition assay, SEM, swimming and swarming motility assay demonstrated that CS-CT-CCa had strong (MIC of 128 μg/mL) and sustained (more than 12 h) inhibitory effects against V. parahaemolyticus. Meanwhile, CS-CT-CCa could increase the membrane permeability of V. parahaemolyticus and inhibit their biofilm-forming ability in a dose-dependent manner. It could be inferred that the antibacterial activities against V. parahaemolyticus caused inhibition of biofilm formation, swimming and swarming motilities. This study provided necessary data for the further design and development of chitosan antibacterial agents, food and feed additives.

Copolymer intercalated hydrotalcite sustained release-type fluid loss additive for water-based drilling fluid: Synthesis, characterization, and performance evaluation

Pengpeng Li,Ming Liu,Jie Feng,Baotong Hui,Miaomiao Hu,Jintang Guo

The polyanionic copolymer fluid loss additive (PSADM) intercalated hydrotalcite sustained release-type fluid loss additive (PSADM–LDH) was fabricated through in situ intercalation polymerization to alleviate the deterioration of filtration and rheological properties of water-based drilling fluid (WBDF) under extreme geological conditions (e.g., high-temperature and alt-gypsum formations). The PSADM between layers of LDH can be slowly exchanged by anions in WBDF to give full play to its original efficiency. Results of FT–IR, TG and XRD demonstrate that the PSADM successfully intercalated the layers of LDH, and the intercalation rate was approximately 14 wt%. In addition, results of filtration measurements of WBDF demonstrate that the PSADM–LDH significantly decreased the filtration volume, even though after thermal rolling at 200 °C the filtration volume of brine drilling fluids contained 2 wt% PSADM–LDH was reduced by approximately 50% compared with PSADM. Moreover, the introduction of PSADM–LDH enhanced the thixotropy of WBDF because of forming a dynamic reversible gel structure by copolymer and charged clay colloidal particles (LDH and bentonite), which is conducive to giving full play to the water power of the bit, and carrying and suspending the drill cutting well in the annulus. Therefore, PSADM–LDH has broad application prospects for exploring and developing super-deep reservoirs.

Fabrication of biodegradable hollow microsphere composites made of polybutylene adipate co-terephthalate/polyvinylpyrrolidone for drug delivery and sustained release

Chuan Xie,Qinqin Xiong,Yuanzhi Wei,Xin Li,Jiajun Hu,Min He,Shinan Wei,Jia Yu,Sha Cheng,Mashaal Ahmad,Yufei Liu,Sihai Luo,Xi Zeng,Jie Yu,Heng Luo

Sustained drug release has attracted increasing interest in targeted drug therapy. However, existing methods of drug therapy suffer drug action time, large fluctuations in the effective concentration of the drug, and the risk of side effects. Here, a biodegradable composite of polybutylene adipate co-terephthalate/polyvinylpyrrolidone (PBAT/PVP) consisting of electrospun hollow microspheres as sustained-released drug carriers is presented. The as-prepared PBAT/PVP composites show faster degradation rate and drug (Erlotinib) release than that of PBAT. Furthermore, PBAT/PVP composites loaded with Erlotinib provide sustained release effect, thus achieving a better efficacy than that after the direct injection of erlotinib due to the fact that the composites allow a high drug concentration in the tumor for a longer period. Hence, this work provides a potential effective solution for clinical drug therapy and tissue engineering using drug microspheres with a sustained release.

The incorporated hydrogel of chitosan-oligoconjugated linoleic acid vesicles and the protective sustained release for curcumin in the gel

Huan Liu,Xinyu Meng,Lei Li,Yongmei Xia,Xueyi Hu,Yun Fang

Assemblies of as called “chitosan hydrogel-liposome” are expected for overcoming the burst effect in drug release from chitosan (CS) hydrogels. Herein, a hydrogel delivery system made of chitosan incorporated fatty acid vesicles was constructed for protective sustained release of curcumin (Cur). The curcumin was encapsulated in the prepared oligo-conjugated linoleic acid vesicles (OCLAVs), and then the drug-embedded vesicles were constructed to Cur-OCLAVs-CS hydrogels with CS solution. The fabricated Cur-OCLAVs-CS hydrogel was fluidic at room temperature and could be rapidly gelled at 37 °C. Morphology study proves that the OCLAVs stayed as nano-vesicles in the gel. The Cur-OCLAVs-CS hydrogels effectively declined the burst effect with enhanced antioxidant activity. The Cur (400 μM)-OCLAVs-CS gel presented a cumulative release rate of 51.23 % of curcumin in 96 h, comparing to 93.37 % of that from the Cur-CS gel. Moreover, the corporation of OCLAVs and CS made the gel exhibited strong synergistic effect on the antioxidant activity, with an enhancement of up to 148.1 % on the ferric reducing power. Therefore, the hydrogel carrier made of incorporated fatty acid vesicles-chitosan can be served as an injectable or 3D printable drug delivery system, which may provide a hint to overcome the burst effect that existed in chitosan and other polysaccharide-based gels.

Promotion of the genipin crosslinked chitosan-fiber hydrogel loaded with sustained release of clemastine fumarate in diabetic wound repair

Rongtai Zuo,Jingjing Shi,Susu Jiang,Min Chu,Qiyang Wang,Lingchi Kong,Qinglin Kang,YaPing Guo,Junjie Guan

Diabetic skin disorders are lingering and refractory clinical diseases. In this study, a genipin-crosslinked porous chitosan fiber (CSF) hydrogel was fabricated to achieve rapid wound healing. By embedding clemastine fumarate (CF) in the CSF hydrogel pores, we synthesised a CSF/CF hydrogel for the treatment of diabetic wounds. The microstructure, chemical elements, spectral variation, mechanical properties, swelling ratios, degradability, and toxicity of the CSF/CF hydrogels were studied. Compared with the typical CS power hydrogel, the porous CSF hydrogel crosslinked with genipin possesses a stable structure and improved physicochemical properties. Moreover, CF was slowly released from the CSF hydrogel. Molecular simulation also showed that CF was evenly embedded inside the cavity formed by the novel CSF hydrogel. The results suggested that CF can resist damage from high glucose levels and promote proliferation, tube formation, and migration of endothelial cells (ECs) and fibroblasts. The CSF/CF hydrogel promoted wound healing in a rat model. Mechanistically, the beneficial effect of CF on wound healing may be related to activation of the MEK/ERK and PI3K/Akt signalling pathways. In conclusion, genipin-crosslinked CSF/CF hydrogel can accelerate wound healing and may be an effective therapeutic method for treating diabetic skin lesions.

Salicylaldehyde and D-(+)-galactose functionalized chitosan oligosaccharide nanoparticles as carriers for sustained release of pesticide with enhanced UV stability

Shaochun Su,Li Hao,Huayao Chen,Xinhua Zhou,Hongjun Zhou

Sustainable food consumption is intertwined with sustainable agriculture development, which focuses on the production and protection of agricultural crops in a safe and effective fashion. Herein, amphiphilic functionalized chitosan oligosaccharide nanoparticles (CSD) were formed by a Schiff base reaction grafted with salicylaldehyde and D-(+)-galactose ( D -Gal). Avermectin (AVM) was used as a model pesticide and self-assembled onto CSD to obtain AVM@CSD nanoparticles with an encapsulation efficiency of 74.94%. AVM@CSD nanocarrier exhibited excellent anti-UV property to retain more AVM, due to their Schiff base structure and salicylaldehyde for UV shielding. Compared with bare AVM and commercial AVM emulsifiable concentrates, AVM@CSD preserved high insecticidal activity after UV irradiation . Additionally, CSD carrier promoted cucumber seed germination and seedling growth. These data suggested the potential utilization of the amphiphilic chitosan oligosaccharide as carriers for pesticides to improve their delivery and release properties and opens a novel avenue towards the application of water-dispersible and biological CSD in the field of pest management and plant growth.

N-Nitrosodimethylamine formation in metformin hydrochloride sustained-release tablets: Effects of metformin and hypromellose used in drug product formulation

Gugu Hao,Rong Hu,Xiaoqin Wang,Pan Gao,Lin Wang,Manhua Jiang,Libo Xin,Guoying Tan,Yanping Zhao,Fuzhou Sun,Dexin Chu,Jinqian Lv,Jinsong You,Fangfang Huang,Xuezhi Song

In recent years, nitrosamines have been discovered in some types of drug products that becomes a current regulatory hotspot, and have attracted a lot attention from both regulatory authorities and industry. This manuscript provided an industry perspective on the nitrosamines research. A liquid chromatography coupled with tandem mass spectrometry(LC-MS/MS)method was developed and applied for the quantification of N-nitrosodimethylamine (NDMA) in metformin hydrochloride sustained-release tablets (MET). The key factors resulting in the NDMA formation in MET were identified through forced degradation and drug-excipient studies, which included high temperature, dimethylamine , strong alkali and oxidation conditions, peroxide and alkaline components contained in the formulation as well as the nitrite and nitrate impurities that might be presented in certain excipients. Further, API particle size and water content of the drug product would also affect the growth rate of NDMA. Therefore, the following mitigation strategies to reduce the risk of nitrosamines in the finished drug product are proposed in this manuscript: 1) avoid the use of excipients containing nitrite, nitrate and peroxide impurities; 2) avoid high temperature and strong alkaline environment in the production and storage condition; 3) maintain an appropriate water content level in the formulation. Based on the above principles, it was recommended to add antioxidant or incorporate excipient such as Na 2 CO 3 to modify the formulation pH to weak basic environment in the formulation of MET, which can could effectively prevent formation of NDMA in the stability process.
