Background Since the pathogenesis of depression is complex, antidepressant therapy remains unsatisfactory. Recent evidence suggests a link between depression and lipid metabolism. Saikosaponin (SS) exhibits antidepression and lipid-regulating effects in modern pharmacology. However, it is unknown whether lipid regulation is the key mechanism of the SS antidepressant effect and how it works. Purpose In this study, we investigated the relationship between the antidepressant activity of SS and the regulation of lipid metabolism and explored potential mechanisms. Methods APOE -/- mice, in combination with the chronic unpredictable mild stress (CUMS) model, were used to study the relationship between SS antidepressant activity and lipid metabolism through behavioral, electrophysiological techniques, and non-targeted lipidomics. Western blot, primary cell culture technology, and laser speckle cerebral blood flow imaging were employed to elucidate potential mechanisms. GraphPad Prism was used for statistical analysis, and p < 0.05 was considered statistically significant. Results APOE -/- mice exhibit more severe depressive-like behavior and dysregulation of sphingolipid metabolism in CUMS. SS alleviates depressive behavior and cortical sphingolipid metabolism disorder caused by CUMS, but has no effect on APOE -/- mice. SS alleviates the imbalance between ceramide (Cer) and sphingomyelin (SM) through acidic sphingomyelinase (AMSase). In addition, SS regulates neuronal glutamate release via sphingolipid metabolism, thereby alleviating the CUMS-induced inhibition of neurovascular coupling (regulates metabotropic glutamate receptor and IP3 receptor), which ameliorates the reduction of cerebral blood flow in depressed mice. Conclusion Our study highlights the role of lipid metabolism in the antidepressant activity of SS and explores its underlying mechanisms. This study provided new insights into the better understanding of the antidepressant mechanisms of phytomedicine while increasing the possibility of lipid metabolism as a therapeutic strategy for depression.

While extensive research has focused on the flavor of Fu brick tea (FBT), the perceptual interactions among its aromatic components remain unclear. To address this, twenty representative aroma compounds were selected from FBT to determine their thresholds and evaluate their interactions by analyzing threshold changes before and after mixing. The results revealed discrepancies between the determined thresholds and literature values, particularly for α -terpineol, methyl heptenone, and geranylacetone, which demonstrated ratios of 73.54, 6.62, and 6.41, respectively. Mixtures with similar aromas or structures exhibited synergy, whereas those with differing aromas or structures displayed masking effects. Additionally, the odor compound ( E )-nerolidol was added to the tea infusion at a concentration below its threshold level to investigate its impact on the overall aroma perception of FBT through sensory analysis and electronic nose assessments. The findings indicated that sub-threshold compounds could significantly influence the overall aroma profile of FBT.

Flexible electronic skin ( E -skin) demands materials with skin-like softness, self-healing capability, and antibacterial properties for long-term wearable applications. Here, we developed a series of self-healing, antibacterial zwitterionic polydimethylsiloxane (PDMS) elastomers via a facile photo-polymerization strategy. The elastomer was synthesized by crosslinking acrylate-terminated PDMS (PDMS-IU-MA) with zwitterionic sulfobetaine methacrylate (SBMA) using reactive hyperbranched polysiloxane (HPBSi) containing methacrylate groups as a multifunctional mediator. The resulting material exhibited remarkable antibacterial effects against E. coli and S. aureus , attributed to the zwitterionic SBMA moieties, while the dynamic hydrogen-bonding network formed by urethane/urea groups enabled self-healing ability. The mechanical properties, self-healing efficiency, and antibacterial performance were systematically tuned by varying the SBMA content. Furthermore, a conductive E -skin capable of precisely monitoring human motion through resistance changes was fabricated by integrating liquid metal (LM) microdroplets into an optimized PDMS matrix. This work provides a scalable strategy to integrate antimicrobial protection, self-healing, and strain sensing into E -skin for next-generation wearable electronics.

An electrochemical aptasensor based on Ti 3 C 2 T x (MXene) and cDNA-PAA was established for sensitive detection of aflatoxin B1 (AFB1) in peanuts. The high specific surface area of Ti 3 C 2 T x (MXene) contributed to improve electrochemical efficiency and enhancing sensor stability which were characterized by the Randles−Sevcik equation. The self-made polyacrylic acid (PAA) nanogel was added with aptamer complementary chain (cDNA) to form cDNA-PAA nano gel composite, which could realize the synergistic amplification of the difference between the electrochemical signals before and after the addition of AFB1. The sensing effect of cDNA-PAA was validated using DPV. Methylene blue (MB) was modified at the proximal 3′ termini of the aptamer (Apt-MB), meanwhile AuNPs was used to immobilize Apt-MB and improve the efficiency of electrochemical reactions. The competition between AFB1 and cDNA-PAA combined with Apt MB resulted in a significant change in the electrochemical signal current. In addition, the constructed electrochemical aptasensor had a lower detection limit of 1.0 × 10 −3 ng/L for AFB1 under optimal detection conditions, and a detection range of 1 to 1000 ng/L. Moreover, peanuts with different concentrations of AFB1 were used as actual detection samples. Through the constructed sensor detection, the spiked recovery rates within the spiking range of 50 to 500 ng/mL were 97.76% to 101.84% ( n = 3).

Klebsiella pneumoniae is a globally recognized microbial pathogen with significant clinical impact. The bacterium harbors the clustered regularly interspaced short palindromic repeats (CRISPR)-Cas systems, which provide adaptive immunity against invading foreign nucleic acids. Recent studies suggest that certain CRISPR-Cas systems can regulate endogenous genes, influencing bacterial virulence. However, their role in regulating pathogenicity in K. pneumoniae remains poorly understood. This study investigates the regulatory role of the type I-E* CRISPR-Cas system in a hypervirulent K. pneumoniae strain, focusing on its impact on histidine metabolism and pathogenicity. Transcriptome analyses identified differentially expressed genes (DEGs) between the casABECD-deletion and wild-type strains, including significant upregulation of the histidine utilization (Hut) operon and downregulation of biofilm-related genes. These molecular changes resulted in enhanced histidine metabolic activity, reduced biofilm formation, attenuated virulence in A549 lung epithelial cells, and improved survival of Galleria mellonella, as validated through phenotypic and virulence assays. Our bioinformatic analysis indicated that the CRISPR-Cas system in K. pneumoniae targets the hutT sequence, which is part of the Hut operon. Furthermore, the overexpression of hutT mitigated CRISPR-Cas-mediated repression of the Hut operon, as observed in virulence assays, while simultaneous deletion of hutH and casABECD restored the reduced virulence in the ΔcasABECD strain. Additionally, deletion of casABECD significantly enhances the growth of the strain in medium with histidine as the sole carbon source, highlighting the intricate regulatory role of the CRISPR-Cas system in metabolic adaptation. Collectively, these findings uncover a novel role for the CRISPR-Cas system in regulating metabolic pathways and virulence in hypervirulent K. pneumoniae.

The complex synthesis, purification, and delivery of photosensitizers remain major bottlenecks for clinical photodynamic therapy (PDT). Here, we develop PCB@Ecoli@CaP, an engineered Escherichia coli –based living therapeutic that autonomously biosynthesizes the natural photosensitizer phycocyanobilin (PCB) and is encapsulated within a calcium phosphate (CaP) shell to form a self-contained photodynamic system. Under 660 nm laser irradiation, PCB@Ecoli@CaP efficiently generates reactive oxygen species (ROS), which induce potent oxidative stress to eradicate 4T1 tumor cells and simultaneously trigger bacterial self-killing, thereby establishing a self-limiting therapeutic platform. The CaP coating enhances biocompatibility and stability while modulating light penetration and ROS release, as confirmed by DPBF photobleaching, intracellular ROS imaging, and CCK-8 viability measurements. This dual-function system integrates in situ photosensitizer biosynthesis, on-demand photodynamic activation, and built-in safety through self-elimination into a single, programmable microbial platform, offering a simplified, scalable, and safer strategy for next-generation PDT.

The human gastrointestinal system contains the conditional pathogen Escherichia coli ( E. coli ), which is a significant cause of vomiting and diarrhea—both of which can be fatal in extreme cases. Given the surge in hospital infections and water pollution, the development of early detection and diagnosis procedures is imperative. This study proposes an electrochemical biosensor for E. coli detection, utilizing nucleic acid aptamers as recognition elements and using E. coli-2571 as an example. It is based on the CRISPR/Cas gene-editing technology. Specifically, the collateral cleavage activity of Cas12a shifts the electrochemical signal after loading the DNA sequence tagged with an electrochemical signal onto the gold electrode, which ultimately enables the identification of E. coli . The biosensor has a detection limit of 13.1 CFU/mL and exhibits a good linear relationship in the concentration range of 10 2 ––10 7 CFU/mL. It was effectively used for the quantitative detection of E. coli in milk and pure water, with a recovery rate ranging from 93.12 % to 98.93 %. The biosensor offers a rapid, precise, affordable, and sensitive tool for E. coli detection. It is anticipated to become a powerful instrument for identifying microorganisms that pose a risk to public health and human health.

Flexible pressure sensors with porous structures have a wide range of applications in many fields, including in flexible electronic skin, medical diagnosis, and human–computer interaction. As such, the fabrication of pressure-sensitive layers with porous structures capable of superior performance has become a significant research hotspot. Herein, we report a mechanically-sensitively enhanced structure of surface patchy protrusions and porous composites which can be used to accurately monitor limb movement, pulse, and breathing. Notably, the flexible pressure sensor exhibits high sensitivity (21.457 kPa −1 ), wide detection range (0.11–11.022 kPa), rapid response/recovery time (41.84 ms/20.82 ms), and excellent stability and cycle repeatability (up to 6000 cycles). Further, the pressure sensor array can act as an electronic skin to detect the pressure distribution. We further propose a newly developed miniaturized flexible wireless feedback system equipped with five pressure sensors. Using the detected pressure as input, the accurate recognition of nine gestures (98.22 %) was achieved with the help of a convolutional neural network (CNN) model.

A colorimetric assay based on the Fe/Al bimetallic organic framework NH 2 -MIL-53(Fe, Al) was developed for the sensitive detection of Escherichia coli O157:H7 ( E. coli O157:H7), a critical pathogen threatening food safety. The framework’s high specific surface area facilitated efficient antibody immobilization, whereas the incorporation of Al 3+ enhanced electron transfer between Fe 2+ /Fe 3+ , thereby significantly boosting peroxidase activity. The optimized NH 2 -MIL-53(Fe, Al) exhibited 1.4-fold and 4.9-fold higher relative absorbance than NH 2 -MIL-53(Fe) and NH 2 -MIL-53(Al), respectively. And it demonstrated 40-fold and 2-fold greater affinity for TMB and H 2 O 2 , respectively, compared to the natural enzyme. By integrating a dual-recognition system based on antibody and aptamer, the method enabled rapid detection with a limit of detection of 10 1 CFU/mL in colorimetric mode. Its successful application in complex matrices, including lettuce, juice, pool water, and chocolate milk, had yielded satisfactory recovery and precision, confirming its promising potential for food safety monitoring.

Natural products were extracted from traditional Chinese herbal emerging as potential therapeutic drugs for treating cardiovascular diseases. This study examines the role and underlying mechanism of dihydromyricetin (DMY), a natural compound extracted from Ampelopsis grossedentata , in atherosclerosis. DMY treatment significantly inhibits atherosclerotic lesion formation, proinflammatory gene expression and the influx of lesional macrophages and CD4-positive T cells in the vessel wall and hepatic inflammation, whereas increases nitric oxide (NO) production and improves lipid metabolism in apolipoprotein E-deficient ( Apoe − / − ) mice. Yet, those protective effects are abrogated by using NOS inhibitor L-NAME in Apoe − / − mice received DMY. Mechanistically, DMY decreases microRNA-21 (miR-21) and increases its target gene dimethylarginine dimethylaminohydrolase-1 (DDAH1) expression, an effect that reduces asymmetric aimethlarginine (ADMA) levels, and increases endothelial NO synthase (eNOS) phosphorylation and NO production in cultured HUVECs, vascular endothelium of atherosclerotic lesions and liver. In contrast, systemic delivery of miR-21 in Apoe − / − mice or miR-21 overexpression in cultured HUVECs abrogates those DMY-mediated protective effects. These data demonstrate that endothelial miR-21-inhibited DDAH1-ADMA-eNOS-NO pathway promotes the pathogenesis of atherosclerosis which can be rescued by DMY. Thus, DMY may represent a potential therapeutic adjuvant in atherosclerosis management.

ABSTRACT Dairy processing can alter the digestion stability and bioavailability of cow milk proteins in the gastrointestinal tract. However, analysis of stable linear epitopes on cow milk allergens that could enter into intestinal mucosal is limited. Thus, this study aimed to investigate the digestion and transportation properties and residual allergen epitopes entering into gastrointestinal mucosa of 3 commercial dairy products, including pasteurized milk (PM), ultra-heat-treated milk (UHTM), and dried skim milk (DSM). In this work, the digestive stability of the 3 kinds of dairy products has been performed in a standard multistep static digestion model in vitro and characterized by Tricine-SDS-polyacrylamide gel electrophoresis and reversed-phase HPLC. With respect to gastrointestinal digestion in vitro, the main allergens including β-lactoglobulin (β-LG), α-lactalbumin (α-LA), and caseins were degraded gradually, and the resistance peptides remained in the PM with a molecular weight of range from 3.4 to 5.0 kDa. Simultaneously, the potential allergenicity of the cow milk proteins was diminished gradually and is basically consistent after 60 min of gastrointestinal digestion. After gastrointestinal digestion, the remaining peptides were transported via an Ussing chamber and identified by liquid chromatography-MS/MS. By alignment, 10 epitopes peptides were identified from 16 stable peptides, including 5 peptides (AA 92–100, 125–135, 125–138, and 149–162) in β-LG, 2 peptides in α-LA (AA 80–93 and 63–79), 2 peptides in α S1 -casein (AA 84–90 and 125–132), and 1 peptide (AA 25–32) in α S2 -casein were identified by dot-blotting mainly exist in UHTM and PM. This study demonstrates dairy processing can affect the digestion and transport characteristics of milk proteins and in turn alter epitope peptides release.

With technological advancements, consumer demands for textile functionality and intelligence have increased substantially. Next-generation E-textiles should enable multidirectional force sensing while ensuring high levels of wearer comfort. However, existing electronic textiles are insufficient to simultaneously monitor the direction and degree of strain, and sweat accumulation can lead to poor comfort. Here, inspired by the asymmetric gradient structure of human skin, Janus double-layer woven electronic textile (JDET) was designed. Through material-structure-function biomimetic design, bidirectional bending recognition and moisture management function are integrated into the device. A high-sensitivity strain sensing unit (GF = 1402.94) was constructed using a single wrapped yarn and conductive materials. Combined with a double-layer fabric structure design, a plain weave layer woven with hydrophilic sensing yarn and a twill weave layer woven with hydrophobic polyester yarn formed a dual gradient structure of fabric wettability and porosity, resulting in excellent unidirectional moisture transport capability. The asymmetric design of the sensing layer enables JDET to selectively identify bending directions (−180°–180°), and has good stability (>8000 s) in bending cycle testing. In addition, JDET has been successfully applied to human motion monitoring and Morse code interaction systems. This asymmetric gradient structure design of textiles provides ideas for the design of the next generation of intelligent electronic textiles.
