Ultrafast microwave-assisted multicomponent tandem polymerization for rapid fabrication of AIE-active fluorescent polymeric nanoparticles and their potential utilization for biological imaging

Ruming Jiang,Long Huang,Meiying Liu,Fengjie Deng,Hongye Huang,Jianwen Tian,Yuanqing Wen,Qian-yong Cao,Xiaoyong Zhang,Yen Wei

The fabrication and biomedical applications of fluorescent polymeric nanoparticles (FPNs) with aggregation-induced emission (AIE) feature has attracted the most intensive research interest since the first discovery of AIE phenomenon by Tang' group. Although great attention has been devoted to preparation of AIE-active FPNs, an efficient, facile and versatile strategy is still highly desirable to advance their biomedical applications. In this work, a one-pot microwave-assisted multicomponent tandem polymerization was proposed to fabricate AIE-active FPNs based on a microwave-assisted Kabachnik–Fields (KF) reaction, which involves the conjugation of aldehyde group containing polyethylene glycol (CHO-PEG-CHO) and amino-group terminating AIE dye (H 2 N-PhE-NH 2 ) using diethyl phosphate as the lock. The KF reaction can occur under rather facile and mild experimental conditions (e.g. absent of catalyst and solvents, air atmosphere) with the assistance of microwave irradiation in 5   min. The resultant (PEG-DP-PhE) copolymers would self-assemble into FPNs that showed high water dispersibility and enhanced fluorescence intensity. The desirable cytocompatibility and cell uptake efficiency of PEG-DP-PhE FPNs endow their great potential for biomedical applications. Considering the convenience and effectiveness, the method should be promising for fabrication of many AIE-active functional materials with great application potential.

Atomic Engineering and Aggregation Effect to Regulate Synergistically Type I Reactive Oxygen Species of AIE-Active Deep Red/Near Infrared Red Photosensitizer

Yaoqiu Zheng,Yin Li,Xue Bai,Muzhou Teng,Yiwen Tang,Shuo Zhao,Zihang Ma,Hongbo Liang,Yili Xie,Qing Wan

“Molecular science” has long been regarded as the golden rule to guide the design of organic materials’ performances in the past many years, but some interesting phenomena of conventional aggregation-caused fluorescence quenching and new aggregation-induced emission reflect that materials’ properties would be changed from “molecule” to “aggregate”. Therefore, “molecular science” theory faces certain limitations to guide regulating the performance of materials at aggregation. In this work, it is discovered that the photosensitizer's performances contain fluorescence and reactive oxygen species, which could be affected by changing molecular atoms and aggregation form. The introduction of oxygen and selenium atoms could redshift fluorescence and improve reactive oxygen species (ROS) efficiency. In addition to the atomic effect, the ROS efficiency of photosensitizers could be affected after coating a polymeric shell, that is, the production of type II ROS singlet oxygen ( 1 O 2 ) is suppressed, while the type I ROS of superoxide anion (O 2 −• ) is improved. This work discovers that the fluorescence and ROS efficiency of photosensitizers are relevant to the atomic effect and polymeric aggregation effect, and discussing deeply the influence mechanism, which has important research significance for modulating precisely the performances of photosensitizers and promoting the development of type I photodynamic therapy.

Biomimetic androgen receptor-based AIE biosensor for detecting bisphenol analogues: An integrating in silico topological analysis, molecular docking, and experimental validation study

Ning Li,Xiaoxiao Liu,Canfeng Bian,Chenxi Ren,Qin Hu,Zhenquan Yang,Lixia Xiao,Tianzhu Guan

Bisphenol analogues are the typical class of endocrine disrupting chemicals (EDCs) that interfere with binding of endogenous hormones to androgen receptor (AR). With the expansion of industrial activities and the intensification of environmental pollution, an increasing array of bisphenol analogues is being released into the environment and food chain. This highlights the urgency to develop sensitive methods for the detection of bisphenol analogues. Here, we propose a biomimetic AR-based biosensor platform for detecting bisphenol analogues (BPF, TBBPA, and TBBPS) by binding with Aggregation-Induced Emission (AIE) probes. Following a comparison of the PROSS and ABACUS methods, biomimetic AR was designed using the ABACUS approach and subsequently expressed in vitro via the E. coli expression system. Through molecular docking and the observation of fluorescence changes upon binding with biomimetic AR, BS-46006 was selected as the AIE probe for the biosensor. The biomimetic AR-based biosensor showed sensitive detections of BPF, TBBPA, and TBBPS within a range of 0–50 mM. To further elucidate the multi-residue recognition mechanism, molecular orbitals, Electron Localization Function (ELF), and Localized Orbital Locator (LOL) were systematically calculated in this study. Lowest unoccupied molecular orbital and highest occupied molecular orbital indicated the energy gap of BPF, TBBPA, and TBBPS, which correspond to 0.12812, 0.19689, and 0.18711 eV, respectively. ELF and LOL offered clearer perspective through heat maps to visually represent the electron delocalization in BPF, TBBPA, and TBBPS. The matrix effect analysis suggested that the responses of bisphenol analogues in soil matrices could be effectively mitigated through sample pretreatment. The analysis of spiked soil samples showed the acceptable recoveries ranged from 91 % to 105 %. Additionally, the biomimetic AR-based AIE biosensor, which combines multi-residue detection with Tolerable Daily Intakes, shows great promise for the risk assessment of bisphenol analogues. This research may present a viable approach for the analysis of environmental pollutants.

A powerful “one-pot” tool for fabrication of AIE-active luminescent organic nanoparticles through the combination of RAFT polymerization and multicomponent reactions

Qing Wan,Ruming Jiang,Liucheng Mao,Dazhuang Xu,Guangjian Zeng,Yingge Shi,Fengjie Deng,Meiying Liu,Xiaoyong Zhang,Yen Wei

Multicomponent reactions (MCRs) have recently received increasing attention for the synthesis of structural complexity in a single step from three or more reactants. They have also been considered as a powerful tool for the construction of sequence-controlled multifunctional polymers owing to their good substrate adaptability, simple operation and high efficiency. In this work, we reported methods that are a combination of the three-component mercaptoacetic acid locking imine (MALI) reaction and reversible addition fragmentation chain transfer (RAFT) polymerization in one pot to form luminescent organic nanoparticles (LONs) with aggregation-induced emission (AIE) features, high-brightness, great water dispersibility, ultra-small nanoscale size and excellent biocompatibility. In the reaction system, the MALI reaction and RAFT polymerization happened simultaneously in a “one-pot” route. On the one hand, the AIE-active organic dye with one amino group ((Z)-3-(4-aminophenyl)-2-(10-hexadecyl-10H-phenothiazin-3-yl)acrylonitrile) (named as Phe-NH2) was conjugated with an aldehyde-containing monomer (10-undecenal) by the MALI reaction, while the aldehyde-containing monomer was copolymerized with the hydrophilic monomer polyethylene glycol methyl methacrylate (PEGMA) through RAFT polymerization at the same time. Compared with other fabrication strategies, “one-pot” strategies possess some advantages such as high efficiency, simplicity, and atom economy. On the other hand, due to the good applicability of RAFT polymerization and the MALI reaction, many other multifunctional AIE-active LONs could also be fabricated via adjusting the function of the substrates. Therefore, this strategy should be a general and important route for fabrication of AIE-active materials for different applications.

Encapsulating Pt NCs with an AIE effect into a metal–organic framework as a novel fluorescence nanoprobe for discriminatively detecting bisphosphonate drugs

Du Juan,Chen Jinwen,Tong Huixiao,Liao Shenghua

Fluorescent metal organic frameworks (FMOFs) constructed by encapsulating aggregation-induced emission luminogens (AIEgens) into frameworks have garnered increasing attention due to their ability to overcome the aggregation-caused quenching (ACQ) effect of traditional fluorescent materials such as organic dyes, quantum dots, and up-conversion nanoparticles. However, conventional AIEgens usually contain aromatic groups that may pose potential health risks. Additionally, the quality of bisphosphonates (BPs), first-line treatment drugs in skeletal-related diseases, is critical to assure their effectiveness and safety. In this work, a new nanocomposite, Pt NCs@ZIF-8, was designed by encapsulating platinum nanoclusters (Pt NCs) with aggregation-induced emission (AIE) properties into zeolitic imidazolate framework-8 (ZIF-8) through self-assembly between Zn 2+ and Pt NCs. Owing to the spatial confinement provided by ZIF-8 and the AIE effect of Pt NCs, the resulting Pt NCs@ZIF-8 nanocomposite presents excellent photostability and anti-photobleaching. Furthermore, the framework of Pt NCs@ZIF-8 collapses in the presence of BP drugs due to the strong coordination interaction between the P = O groups in BPs and the Zn 2+ in ZIF-8, leading to the release of Pt NCs. This release of Pt NCs causes effective fluorescence quenching of the Pt NCs@ZIF-8 nanocomposite, resulting from the increasing intramolecular motion of surface ligands, glutathione (GSH), on Pt NCs. Therefore, a new Pt NCs@ZIF-8-based nanoprobe has been rationally designed for BP detection. More importantly, our designed Pt NCs@ZIF-8-based nanoprobe also presents excellent discrimination capability toward four structurally similar BPs—Etidronate, Clodronate, Alendronate, and Risedronate, as confirmed by statistical techniques such as principal component analysis (PCA) and hierarchical clustering analysis (HCA). Finally, the proposed Pt NCs@ZIF-8 nanoprobe was successfully applied to quantify the labeled amount percentage of commercially available Sodium Risedronate and Sodium Alendronate tablets, achieving satisfactory limits of detection (LOD) spanning from 1.2 to 8.8 µM and excellent recovery ranging from 97.50% to 102.6%. This work not only provides a novel and convenient strategy for detecting and distinguishing BPs but also demonstrates the potential application of metal–organic frameworks encapsulating metal nanoclusters with AIE properties in pharmaceutical quality control. A ratiometric fluorescent sensor based on AIE nanoparticles and nitrogen-doped carbon dots for rapid and sensitive detection of acetamiprid in rice

Mingsha Jie,Amei Zhu,Bing Zhu,Jiaxin He,Shengkai Lan,Xingang Lang,Yanhong Bai

Sensitive detection of acetamiprid (ACE) is valuable for human health and food security. Herein, a ratiometric fluorescent sensor based on AIE nanoparticles and nitrogen-doped carbon dots (luminol–Eu–NCDs) can sensitively and selectively detect ACE. luminol–Eu–NCDs were synthesized by the self-assembly of luminol–Eu nanoparticles (luminol–Eu NPs) by Eu3+ and luminol using the aggregation-induced emission (AIE) strategy while covalently linking to nitrogen-doped carbon dots (NCDs) synthesized using Listeria monocytogenes. In the presence of ACE, the fluorescence intensity at 542 nm decreased, with a slight decrease also observed at the 430 nm reference signal. Thus, the concentration of ACE can be accurately monitored by measuring ratiometric fluorescence emitted at 430 nm (F430) and 542 nm (F542) wavelengths. The luminol–Eu–NCD ratiometric fluorescent sensor design reduces errors stemming from environmental influences, thereby improving the sensor's overall accuracy. Optimal conditions yielded a fluorescence intensity ratiometric assay (F430/F542) with a detection limit of 89 ng mL−1 for ACE (S/N = 3), exhibiting a good linear response over the concentration range of 0.1–100 μg mL−1. The present method has been effectively used for the quantitative determination of ACE in rice and has shown remarkable anti-interference and reliability, which has important significance to ensure the safety of rice. This work presents a quick and efficient method for the sensitive detection of ACE contamination in grain food.

Metal-Enhanced Aggregation-Induced Emission Strategy for the HIV-I RNA-Binding Ligand Assay

Jun Li,Yao-Yao Fan,Jie Wen,Jing Zhang,Zhi-Qi Zhang

The HIV-Ι trans-activation responsive (TAR) RNA–trans-activator of transcription (Tat) protein complex is crucial for the efficient transcription of the integrated human immunodeficiency virus-I genome and is an established therapeutic target for AIDS diagnosis and treatment. Developing a sensitive strategy for the TAR RNA-binding ligand assay could provide antiviral leads with a radically new mechanism for the treatment of AIDS. Herein, a new TAR RNA-binding ligand assay platform was established using a signal amplification strategy that combines aggregation-induced emission (AIE) with a metal-enhanced fluorescence (MEF) concept. The tetraphenylethylene (TPE) derivative was labeled on the Tat peptide as a fluorescent molecule, while the TAR RNA was immobilized on the surface of the Fe3O4@Au@Ag@SiO2 nanoparticles (NPs) to specifically bind the TPE-Tat peptide. The TPE-Tat peptide was weakly emissive itself while emitting strongly in the NP–TAR–TPE-Tat complex by the AIE and MEF signal amplification effect. It was confirmed by known Tat peptide competitors that this system could be applied to the screening and detection of TAR RNA-binding ligands because they could replace the TPE-Tat peptide from the complex and make the system fluorescence decrease. When this system was adopted to test four candidate ligands, it was found that bisantrene had a favorable TAR RNA-binding ability. The proposed AIE–MEF strategy not only provides a sensitive and reliable method for the TAR RNA-binding ligand assay but also can avoid the influence of ligands on fluorescent detection in the conventional displacement assay.

An AIE-based ratiometric fluorescent probe for reversible detection of bisulfite and hydrogen peroxide and its application in food samples and biological imaging

Xinya Yao,Ying Li,Yuetian Guo,Keli Zhong,Lijun Tang,Xiaomei Yan

Reversible detection of bisulfite (HSO 3 - ) and hydrogen peroxide (H 2 O 2 ) is highly desirable in biology and food safety owing to their redox pair function in biological systems and their utilization in food industry. In this work, a ratiometric fluorescent probe ( TPA-Y ) with aggregation-induced emission (AIE) properties based on triphenylamine fluorophore was designed and synthesized for reversibly monitoring of HSO 3 - /H 2 O 2 . Reaction of HSO 3 - with TPA-Y can result in ratiometric fluorescence changes ( I 444 / I 629 ), and the emission spectrum blue shift reaches 185 nm, which can effectively avoid spectral overlap. Meanwhile, the solution color changed obviously from orange to colorless, enabling naked eye detection of HSO 3 - . TPA-Y was capable of quantitatively detect HSO 3 - in sugar and wine samples with the recovery rates ranging from 94.52 to 102.18%. In addition, TPA-Y is suitable to prepare paper strips and anti-counterfeiting ink for HSO 3 - detection. In addition, the original TPA-Y can be released after the addition product ( TPA-Y-HSO 3 - ) is oxidized by H 2 O 2 , and the HSO 3 - /H 2 O 2 redox cycle can repeat for at least six times. The probe TPA-Y can be applied to fluorescence imaging of reversible and dynamic interconversion between HSO 3 - and H 2 O 2 in living MCF-7 cells.

Self-assembled nanoparticles based on cationic mono-/AIE tetra-nuclear Ir(III) complexes: long wavelength absorption/near-infrared emission photosensitizers for photodynamic therapy

Ziwei Wang,Lijuan Li,Weijin Wang,Runlin Wang,Guangzhe Li,Hang Bian,Dongxia Zhu,Martin R. Bryce

Cyclometalated Ir(III) complexes as photosensitizers (PSs) have attracted widespread attention because of their good photostability and efficient 1O2 production ability. However, their strong absorption in the UV-vis region severely limits their applications in photodynamic therapy (PDT) because the short wavelength illuminating light can be easily absorbed by the skin and subcutaneous adipose tissue causing damage to the patient's normal tissue. Herein, mono- and tetra-nuclear Ir(III) complex-porphyrin conjugates are rationally designed and synthesized, especially [TPP-4Ir]4+ exhibits obvious aggregation-induced emission (AIE) characteristics. PSs comprising Ir(III) complex-porphyrin conjugates self-assembled as nanoparticles (NPs) are successfully achieved. The obtained [TPP-Ir]+ NPs and [TPP-4Ir]4+ NPs exhibit long wavelength absorption (500–700 nm) and near-infrared emission (635–750 nm), successfully overcoming the inherent defects of short wavelength absorption of traditional Ir(III) complexes. Moreover, [TPP-4Ir]4+ NPs exhibit good biocompatibility, high 1O2 generation ability, low half-maximal inhibitory concentration (IC50) (0.47 × 10−6 M), potent cytotoxicity toward cancer cells and superior cellular uptake under white light irradiation. This work extends the scope for transition metal complex PSs with promising clinical applications.

A dibenzothiophene core-based small-molecule AIE probe for wash-free and selective staining of lipid droplets in live mammalian and fungal cells

Sayed Mir Sayed,Xiang-Fei Li,Hao-Ran Jia,Samran Durrani,Fu-Gen Wu,Xiaolin Lu

Lipid droplets (LDs) are spherical dynamic subcellular organelles, and play crucial roles in a number of cellular functions, such as lipid metabolism, protein degradation, membrane formation, energy storage, and signal transduction. It is widely recognized that the dysfunction of LDs can lead to many diseases, and hence it is important to monitor the size, distribution, and movement of LDs in living cells. Herein, we report a dibenzothiopene core-based fluorescent probe named triphenylamine dibenzothiophene-S,S-dioxide (TPA-DBTD), which can exhibit aggregation-induced emission (AIE) characteristic and realize selective and wash-free imaging of LDs in live cells. Most importantly, this probe can efficaciously track the dynamic changes of LDs including lipophagy (an autophagic process that is responsible for the degradation of lipids) in live mammalian cells and stain LDs in fungi. Hence, we believe that TPA-DBTD with high photostability, good biocompatibility, AIE characteristic, and LD staining ability in diverse biological systems (mammalian cells and microorganisms) can find practical applications in the fields of biomedical science, cell biology, and diagnosis of LDs-associated diseases.

Capsulation of AuNCs with AIE Effect into Metal–Organic Framework for the Marriage of a Fluorescence and Colorimetric Biosensor to Detect Organophosphorus Pesticides

Yue Cai,Hongshuai Zhu,Weichi Zhou,Ziyin Qiu,Congcong Chen,Aori Qileng,Kangshun Li,Yingju Liu

Organophosphorus pesticides (OPs) can inhibit the activity of acetylcholinesterase (AChE) to induce neurological diseases. It is significant to exploit a rapid and sensitive strategy to monitor OPs. Here, a metal–organic framework (MOF) acted as a carrier to encapsulate AuNCs, which can limit the molecular motion of AuNCs, trigger the aggregation-induced emission (AIE) effect, and exhibit a strong fluorescence with a fluorescence lifetime and quantum yield of 6.83 μs and 4.63%, respectively. Then, the marriage of fluorescence and colorimetric signals was realized on the basis of the dual function of the enzymolysis product from AChE and choline oxidase (CHO) on AuNCs@ZIF-8. First, it can decompose ZIF-8 to weaken the restraint on AuNCs, and thus the fluorescence receded. Second, it can be used as a substrate for the peroxidase mimics of the released AuNCs to oxidize 3,3′,5,5′-tetramethylbenzidine (TMB) and a visible blue appeared. Thus, on the basis of the inhibition of AChE activity by OPs, a fluorescence–colorimetric dual-signal biosensor was established. In addition, colorimetric paper strips were exploited to realize a visual semiquantitative detection, and a smartphone APP was developed to make the visualization results more precise and realize real-time supervision of pesticide contamination.

Ultrafast construction and biological imaging applications of AIE-active sodium alginate-based fluorescent polymeric nanoparticles through a one-pot microwave-assisted Döbner reaction

Ruming Jiang,Meiying Liu,Hongye Huang,Liucheng Mao,Qiang Huang,Yuanqing Wen,Qian-yong Cao,Jianwen Tian,Xiaoyong Zhang,Yen Wei

Aggregation-induced emission (AIE) is a unique fluorescence phenomenon that has attracted great interest for various applications especially for the fabrication of AIE-active polymeric nanoparticles . The AIE-active fluorescent polymeric nanoparticles (FPNs) showed attractive optical properties , which could elegantly overcome the lousy aggregation-caused quenching (ACQ) effect of FPNs based on conventional organic fluorogens. Although great advances and progress have been achieved for the fabrication of AIE-active FPNs, the preparation of AIE-active carbohydrate polymers is still challengeable for the poor solubility of carbohydrate polymers in organic solvents . In this work, we reported the sodium alginate-based fluorescent biopolymers (OSA-PA-PhE) with AIE characteristic could be facilely fabricated via an ultrafast one-pot multicomponent reaction (named as Döbner reaction) with the assistance of microwave irradiation. Among the Döbner reaction system, the oxidized sodium alginate (OSA) and AIE-active dye (named as PhE-NH 2 ) were conjugated using pyruvic acid (PA) as the molecular “bridge”. A number of characterization techniques, such as proton nuclear magnetic resonance ( 1 H NMR), fourier transform infrared (FT-IR) spectroscopy, UV–Vis absorption spectroscopy , fluorescent spectroscopy and transmission electron microscopy (TEM), have been utilized to confirm the successful preparation of OSA-PA-PhE biopolymers. The biological evaluation results confirmed that the OSA-PA-PhE FPNs possessed ultrahigh biocompatibility and desirable imaging properties. Moreover, compared with the previous work about FPNs, this work had at least two obvious advantages, one was the original materials of sodium alginate are resourceful, cheaper, and compatible for human tissue and cells, and another was the novel Döbner reaction is a facile conjugation reaction under mild experimental conditions (e.g. catalyst-free, air atmosphere and in the presence of water). Considered the features of microwave-assisted Döbner reaction and OSA, the OSA-PA-PhE FPNs should be of great potential for different biomedical applications .
