The bone immune response dominated by macrophages plays an indispensable role in the osteogenesis of bone defects. Moreover, moderate polarization of macrophages against inflammatory M2 has been proved to promote osteogenesis. Therefore, the addition of anti-inflammatory agents to bioactive bone repair materials facilitates efficient bone regeneration by regulating the polarization of macrophages. Bioactive glass (BG) has been widely used for bone defect repair. However, BG alone cannot effectively inhibit the inflammatory response caused by in vivo biomaterial implantation, and finally cannot achieve satisfactory bone repair effect. Herein, the design of mesoporous bioactive glass nanoparticles (MBG) modified with β-cyclodextrin (CD-MBG) is reported. Our research shows that the anti-inflammatory drug naringin (NG) is loaded into CD-MBG (NG@CD-MBG), which achieves a sustained release within 6 days. In vitro studies reveal that NG@CD-MBG promotes better transformation of macrophages to the M2 phenotype than MBG inhibiting macrophage inflammatory responses, while the induced local immune microenvironment synergistically facilitates osteogenesis and inhibits osteoclastogenesis. Furthermore, in vivo high expression of osteogenesis-related genes in the microenvironment stimulated by NG@CD-MBG significantly promotes new bone formation in the femoral defect model of rats. The results indicate that the combination of MBG and NG has a synergistic effect on immunomodulating osteogenesis and osteoclastogenesis, providing a novel idea for the development of bone biomaterials with favorable bone immunomodulatory properties.

The flaky carbon nitride containing nitrogen defects (NDCN) could effectively perform the photocatalytic reduction of carbon dioxide (CO 2 ) due to its abundant active sites. Reducing the recombination of electrons and holes was also a method of semiconductor photocatalyst design. A nanosphere ball-flower Indium sulfide (In 2 S 3 ) was synthesized via a simple hydrothermal approach, and then calcined to obtain the β-In 2 S 3 /NDCN heterojunction photocatalyst and applied for CO 2 photocatalytic reduction. The best total yield (carbon monoxide, CO: 20.32 μmol·g −1 ·h −1 ; methane, CH 4 : 2.12 μmol·g −1 ·h −1 ) could be obtained at the optimized 20% β-In 2 S 3 /NDCN under near room temperature and pressure and without using any sacrificial agents or promoters, almost 1.7 times higher compared with NDCN. The composite catalyst still exhibited excellent stability after four cycles. The improvement of excellent performance was due to not only the enhancement of fine CO 2 adsorption/activation and the light absorption ability, but also attributed to the formation of heterojunction, which accelerated the effective separation of electrons and holes. This work might provide a novel approach to design carbon nitride heterojunction photocatalysts with nitrogen defects for CO 2 utilization.

Enantioseparation has always been one of the research hotspots and difficulties in the field of modern separation science . In this work, a binary chiral electrophoretic separation system was constructed using deep eutectic solvents (DESs) coupled with beta-cyclodextrin derivatives based on capillary electrophoresis with amperometric detection system, and five groups of typical adrenergic receptor agonists (adrenaline, salbutamol , isoproterenol , norepinephrine and terbutaline) were selected as the model enantiomers . The effects of additive types and contents of DESs and cyclodextrins , and the pH value and concentration of the running buffer on the resolution of the selected chiral compounds were investigated in detail. The mechanism of DESs improving separation was explored preliminarily by means of UV spectrophotometry , which was further verified based on the comparison of single and mixed components of choline chlorine-urea DES. Under the optimum conditions, the relative standard deviations for inter-day and intra-day repeatability of the migration time, peak area and resolution for adrenaline and salbutamol were within 8.7%, and the limits of detection reached 0.030 μg mL −1 (S/N = 3). The recovery data were in the range of 96.3–118.7%. The developed methods have been applied for the analyses of (+)-adrenaline hydrochloride injection and (±)-salbutamol aerosol. This binary chiral electrophoretic separation system by CE-AD has high detection sensitivity and low analytical cost, providing an alternative for the separation and analysis of chiral drugs.

Amyloid-beta (Aβ) oligomers have been become believable biomarkers for predicting and diagnosing Alzheimer’s disease (AD). Herein, an ultrasensitive electrochemical assay for detection of Aβ oligomers was built based on the Au nanoparticles-modified vertical graphene/carbon cloth (Au-VG/CC) as substrate electrode and in-situ synthesized copper nanoparticles (CuNPs) as signal probes. For the biosensor preparation, the VG/CC film was firstly treated by the electrodeposition of Au nanoparticles, and the Au-VG/CC electrode provided large surface area and high conductivity. Then the residues 95–110 of cellular prion protein (PrP C ) as the specific receptor for Aβ oligomers were immobilized on the Au-VG/CC electrode surface by the Au-S bond. After the selective recognition to Aβ oligomers, the poly(thymine)-templated CuNPs as the electrochemical probes were introduced by the design of aptamers. This assay for Aβ oligomer detection showed a good performances including ultrahigh sensitivity with a low detection limit of 3.5 pM, excellent specificity, and great stability. This biosensor also presented feasibility for sensing Aβ oligomers in serum samples of AD patients, and would contribute to understand the role of Aβ oligomer as a biomarker for the early diagnosis of AD.

A Beta-KIT-6 (BK) micro-mesoporous composite material was assembled using zeolite Beta seed solution as raw material. BK composite contained both zeolite Beta units and three-dimensional cubic Ia3d mesostructure of KIT-6, which served as a catalyst support for hydroisomerization of n-heptane. The composite was characterized by a variety of techniques. Results show that catalyst Pt/HBK using the composite as the support exhibited the highest isoheptanes yield among all catalysts, which is attributed to both appropriate surface acidity and excellent mass transfer properties of Beta-KIT-6 composite. Experimental results verified that the indexed isomerization factor as a general performance descriptor could accurately predict the hydroisomerization performance of support materials.

Beta-MCM-41(BM) composites consisting of Beta zeolite grown on MCM-41 type mesoporous structure were prepared. The composite materials were used as a support for stabilization of Pt. The Pt containing catalysts (Pt/HBM) were applied in n-heptane hydroisomerization reaction. The Pt/HBM catalysts showed higher n-heptane conversion and higher selectivity to isoheptanes in comparison to pure Beta and MCM-41. The results were explained by the accelerated diffusion of isoheptanes in the Pt/HBM composites, and the high concentration of acid sites with a moderate strength in the composites, which reduced the cracking of reactants and products. Based on the experimental results, an indexed isomerization factor (IIF) was deduced, which is correlated with the physical properties of the supports. The results indicated that the isoheptane yield of the catalysts is strongly dependent on the support (IIF). The general IIF descriptor for predicting the isomerization performance of support materials can provide precise guidance for the design of new micro-mesoporous composites.

Beta-site amyloid precursor protein cleaving enzyme 1 (BACE1) is an aspartic protease that promotes the production of beta-amyloid (Aβ) peptides from beta-site amyloid precursor protein. Pharmacological reduction of Aβ production by inhibiting BACE1 has emerged as a promising target for the treatment of Alzheimer's disease (AD). In this work, we reported a simple electrochemical method for detection of BACE1 and screening of its inhibitor. This method made use of an unlabeled peptide that comprises a segment of beta-site amyloid precursor protein with the Swiss mutation. The peptide–heme complex formed on the electrode surface showed strong electrocatalytic O 2 reduction. Cleavage of the peptide by BACE1 detached the heme-binding fragment from the electrode surface, leading to a remarkable decrease in the electrochemical signal. Suppression of BACE1 by the inhibitor yielded a larger voltammetric signal. The IC 50 of the inhibitor tested by this method (15.7 nM) was in agreement with that reported. The proposed method could serve as a promising alternative to the traditional methods for probing of BACE1 activity and screening of BACE1 inhibitor owing to its simplicity, high sensitivity and low assay cost.

Lactic acid is an important platform molecule in the synthesis of a wide range of chemicals. However, in aqueous solutions without alkali, its efficient preparation via the direct catalysis of sugars is hindered by a side dehydration reaction to 5-hydroxymethylfurfural due to Brønsted acid, which originates from organic acids. Herein, we report that a previously unappreciated combination of common two metal mixed catalyst (Zn-Sn-Beta) prepared via solid-state ion exchange synergistically promoted this reaction. In water without a base, a conversion exceeding 99% for sucrose with a lactic acid yield of 54% was achieved within 2 hours at 190 °C under ambient air pressure. Studies of the acid and base properties of the Zn-Sn-Beta zeolite suggest that the introduction of Zn into the Sn-Beta zeolite sequentially enhanced both the Lewis acid and base sites and the base sites inhibited a series of side reactions related to fructose dehydration to 5-hydroxymethylfurfural and its subsequent decomposition.

In this study, an effective and precise fluorescent molecularly imprinted polymers (FMIPs) for the determination of Beta-Cyfluthrin (BC) was synthesized via precipitation polymerization with SiO 2 as the carrier, BC as the target molecule, ally fluorescein as the functional monomer, trimethylolpropane trimethacrylate (TRIM) as the crosslinker. Moreover, the characteristic of material has been measured by FTIR, TEM, SEM, TGA, LSCM and fluorescence spectrophotometer. Average diameter and shell thickness of as-synthesized microspheres were 300 nm and 50 nm, respectively. An excellent linear relationship of SiO 2 -MPTMS@FMIPs with a correlation coefficient of 0.9919 could be gained covering a wide concentration range of 10.11–80 nM described by the Stern-Volmer equation. The limit of detection (LOD) was evaluated with the equation LOD = 3σ/S and was found to be 10.11 nM. The study demonstrated that SiO 2 -MPTMS@FMIPs could improve the determination for BC and illustrated the good prospects of SiO 2 -MPTMS@FMIPs for BC detection in agricultural products.

Ethnopharmacological relevance Zhen-wu-tang ( ZWT ), composed of Radix Aconiti lateralis , Rhizoma Atractylodis macrocephalae , Poria , Radix Paeoniae alba and ginger, is a classic Chinese herbal formula for the treatment of chronic kidney diseases that may cause chronic renal failure (CRF). Aim of the study To better understand its clinical use, this study investigated the effects and underlying mechanisms of action of ZWT on CRF. Materials and methods CRF was induced by adenine . ZWT was given via an oral gavage method. The serum biochemical parameters were measured enzymatically or by ELISA . The kidneys were examined pathohistologically. The gene expression was analyzed by real time PCR and Western blot . Results Similar to the positive control losartan , ZWT extract inhibited adenine-induced increase in serum concentrations of creatinine, BUN and advanced oxidation protein products in rats. These effects were accompanied by attenuation of proteinuria and renal pathological changes and suppression of renal mRNA and protein overexpression of Collagen IV and fibronectin , two of the key components of fibrosis . Mechanistically, renal mRNA and protein expression of Wnt4, a Wnt signaling ligand, was increased in the adenine-treated group, compared to the vehicle-treated control. Consistently, Wnt4 downstream genes beta-catenin and Axin were also overexpressed. Treatment with ZWT extract and losartan suppressed adenine-stimulated overexpression of these mRNAs and proteins. Conclusions The present results demonstrate that ZWT extract ameliorates adenine-induced CRF in rats by regulation of the canonical Wnt4/beta-catenin signaling in the kidneys. Our findings provide new insight into the underlying renoprotective mechanisms of the ancient formula.

Herein, a sequentially responsive peptide DNA bilingual nanobiosensor is developed, which allows integrated quantification of amyloid signaling pathway. In this system, upstream beta-site amyloid precursor protein-cleaving enzyme 1 (BACE1) protease and downstream Aβ oligomer (AβO) are designed as two inputs for the AND DNA logic gate. In the existence of both inputs, peptide substrate with aptamer can be sequentially cleaved, reporting electrochemical and fluorescence dual-mode outputs. In comparison with conventional single protease activity assay based on peptide nanotechnology, this strategy permits accurate diagnosis of Alzheimer's disease (AD) from normal subjects. More importantly, it can achieve distinguished diagnosis between AD and type 2 diabetes mellitus patients. This bilingual nanobiosensor is successfully applied to detect BACE1 (1–100 U mL −1 ) and AβO (5–1000 pg mL −1 ) with limit of detections as low as 0.10 U mL −1 and 0.76 pg mL −1 , respectively. Furthermore, this strategy inspires advanced nanobiosensors to target the activation of other signaling pathways, which are potential tools for future biology and medicine investigation.

In view of the importance of quantification of beta-amyloid (Aβ), an “aptamer -target- peptide” sandwich electrochemical biosensor to assay Aβ42 was fabricated by using gold–palladium bimetallic nanoparticles covalent organic framework-carbon cloth (Au–Pd@COF TTF-Thi -CC) and prion protein (PrP C ) labeled with horseradish peroxidase (HRP) nanocomposites. The concentration of Aβ42 showed a positive correlation with the peak current resulting from the oxidation of hydroquinone (HQ) by HRP. In contrast, the reference current signal from thionin (Thi), derived from covalent organic framework (COF), showed a negative correlation. The current response of the electrochemical biosensor was proportional to the Aβ concentrations ranging from 0.01 to 2.5 nM with a detection limit (LOD) of 2.17 fM ( S / N = 3). The sensitivity of the sandwich electrochemical biosensor could be primarily amplified based on the substrates of Au–Pd@COF TTF-Thi -CC. The AuNPs and PdNPs not only facilitated the bonding of the linker based on the Au–S bond but also improved the performance of COF TTF-Thi -CC. Secondary sensitivity amplification of the biosensor could be achieved by using PrP C -labeled HRP to catalyze hydroquinone in solution. Furthermore, the biosensor showed excellent selectivity, anti-interference performance, and stability. The Aβ concentrations in cerebrospinal fluid (CSF) were successfully evaluated using the constructed biosensor. This assay provides a potential tool for early-stage Alzheimer's disease diagnosis.
