Effects of curcumin plus Soy oligosaccharides on intestinal flora of rats with ulcerative colitis

G. Huang,L. Ye,G. Du,Y. Huang,Y. Wu,S. Ge,Z. Yang,G. Zhu

To explore the therapeutic effect of curcumin (Cur) and soybean oligosaccharides (SBOS) on ulcerative colitis (UC) through testing the intestinal flora and ulcerative colitis (UC). 80 male SD rats were selected divided into four groups with 20 rats in each group: normal group, sulfasalazine (SASP) group, model group and group of curcumin plus soy oligosaccharide. All animals were treated for 4 weeks. In the fifth week rats were decapitated. Macroscopic damage scores of colonic mucosa were calculated. A 4mL blood sample was taken to detect the contents of serum tumor necrosis factor -α (TNF-α) and interleukin 8 (IL-8) by the double antibody sandwich ABC-ELISA method (enzyme-linked immunosorbent assay). Colonic tissues with the most obvious lesions were obtained using a surgical scissor. A routine hematoxylin-eosin (HE) staining method was used to stain pathological specimens and images of staining results were obtained. Histological injury scores of colonic mucosa were calculated. Ulcerative colitis model rats had the highest macroscopic damage scores and histological injury scores of colonic mucosa. After treatment the contents of TNF-α and IL-8 decreased significantly in the group of curcumin plus soy oligosaccharide compared with the model group with statistical significance (P <0.01) while the contents were close to those in the SASP group. There was no statistical significance (P> 0.05). The treatment could decrease TNF-α and IL- 8 expression and reduce colonic mucosa inflammation and tissue damage.

Effects of surfactants on the formation of gelatin nanofibres for controlled release of curcumin

Lingli Deng,Xuefan Kang,Yuyu Liu,Fengqin Feng,Hui Zhang

This work studied the effects of non-ionic Tween 80, anionic sodium dodecyl sulfonate (SDS) and cationic cetyltrimethyl ammonium bromide (CTAB) surfactants on the morphology of electrospun gelatin nanofibres, and on the release behaviour, antioxidant activity and antimicrobial activity of encapsulated curcumin. Scanning electron micrographs showed that addition of SDS significantly increased the nanofibre diameter. Fourier transform infrared and differential scanning calorimetry analysis indicated that gelatin and SDS intimately interacted via electrostatic and hydrophobic interactions. However, these interactions inhibited the release of curcumin from the nanofibres with SDS, while CTAB and Tween 80 both facilitated the release. SDS and Tween 80 showed protective effects on curcumin from the attack of 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) radicals, and the increased release of curcumin from nanofibres with CTAB or Tween 80 resulted in a higher reducing power. The antimicrobial activity results suggested that the curcumin encapsulated gelatin nanofibres with CTAB exhibited effective inhibition against Staphylococcus aureus .

Epigallocatechin-3-gallate stabilizes aqueous curcumin by generating nanoparticles and its application in beverages

Ruge Lin,Yi Wang,Huan Cheng,Xingqian Ye,Shiguo Chen,Haibo Pan

In this study, we addressed the limited water solubility of curcumin by utilizing epigallocatechin-3-gallate to form nanoparticles through self-assembly. The resulting particles, ranging from 100 to 150 nm, exhibited a redshift in the UV–visible spectrum, from 425 nm to 435 nm, indicative of potential π-π stacking. Molecular docking experiments supported this finding. Curcumin loaded with epigallocatechin-3-gallate showed exceptional dispersibility in aqueous solutions, with 90.92 % remaining after 60 days. The electrostatic screening effect arises from the charge carried by epigallocatechin-3-gallate on the nanoparticles, leading to enhanced retention of curcumin under different pH, temperature, and ionic strength conditions. Furthermore, epigallocatechin-3-gallate can interact with other hydrophobic polyphenols, improving their dispersibility and stability in aqueous systems. Applying this principle, a palatable beverage was formulated by combining turmeric extract and green tea. The nanoparticles encapsulated with epigallocatechin-3-gallate show potential for improving the applicability of curcumin in aqueous food systems.

Hyaluronic acid-decorated curcumin-based coordination nanomedicine for enhancing the infected diabetic wound healing

Shuai Wu,Li Zhu,Sheng Ni,Yuan Zhong,Kai Qu,Xian Qin,Kun Zhang,Guixue Wang,Da Sun,Wuquan Deng,Wei Wu

Persistent over-oxidation, inflammation and bacterial infection are the primary reasons for impaired wound repairing in diabetic patients. Therefore, crucial strategies to promote diabetic wound repairing involve suppressing the inflammatory response, inhibiting bacterial growth and decreasing reactive oxygen species (ROS) within the wound. In this work, we develop a multifunctional nanomedicine (HA@Cur/Cu) designed to facilitate the repairing process of diabetic wound. The findings demonstrated that the synthesized infinite coordination polymers (ICPs) was effective in enhancing the bioavailability of curcumin and improving the controlled drug release at the site of inflammation. Furthermore, in vitro and in vivo evaluation validate the capacity of HA@Cur/Cu to inhibit bacterial growth and remove excess ROS and inflammatory mediators, thereby significantly promoting the healing of diabetic wound in mice. These compelling findings strongly demonstrate the enormous promise of this multifunctional nanomedicine for the treatment of diabetic wound.

Introduction Alzheimer’s disease (AD), a neurodegenerative condition, stands as the most prevalent form of dementia. Its complex pathological mechanisms and the formidable blood-brain barrier (BBB) pose significant challenges to current treatment approaches. Oxidative stress is recognized as a central factor in AD, underscoring the importance of antioxidative strategies in its treatment. In this study, we developed a novel brain-targeted nanoparticle, Ce/Zr-MOF@Cur-Lf, for AD therapy. Methods Layer-by-layer self-assembly technology was used to prepare Ce/Zr-MOF@Cur-Lf. In addition, the effect on the intracellular reactive oxygen species level, the uptake effect by PC12 and bEnd.3 cells and the in vitro BBB permeation effect were investigated. Finally, the mouse AD model was established by intrahippocampal injection of Aβ1-42, and the in vivo biodistribution, AD therapeutic effect and biosafety of the nanoparticles were researched at the animal level. Results As anticipated, Ce/Zr-MOF@Cur-Lf demonstrated efficient BBB penetration and uptake by PC12 cells, leading to attenuation of H2O2-induced oxidative damage. Moreover, intravenous administration of Ce/Zr-MOF@Cur-Lf resulted in rapid brain access and improvement of various pathological features of AD, including neuronal damage, amyloid-β deposition, dysregulated central cholinergic system, oxidative stress, and neuroinflammation. Conclusion Overall, Ce/Zr-MOF@Cur-Lf represents a promising approach for precise brain targeting and multi-target mechanisms in AD therapy, potentially serving as a viable option for future clinical treatment.

P/N/S-containing cellulose nanofibrils enable curcumin encapsulation via Pickering emulsion based microcapsules

Qingtian Xu,Jierui Ye,Shuaibo Han,Yuebing Gao,Pan Chen,Siqun Wang,Qiang Wu,Qian Li

Renewable polymeric microcapsules hold promise for precisely controlled release of hydrophobic drugs (curcumin) in therapeutic treatment. However, there still remains challenges to stabilize the properties of microcapsules and to enhance encapsulation efficiencies of curcumin. By using an eco-friendly ternary deep eutectic solvent here, we successfully fabricated native cellulose nanofibrils, holocellulose nanofibrils, and lignocellulose nanofibrils containing both phytates and sulfate groups. In addition, a novel type of curcumin-loaded polymeric microcapsules with diameter of 10–20 μm were prepared by cellulose nanofibrils as stabilizing agents in Polylactide Pickering emulsion. The Pickering emulsions stabilized by P/N/S-containing cellulose nanofibrils, holocellulose nanofibrils, and lignocellulose nanofibrils are more stable than the unmodified cellulose nanofibrils-stabilized emulsion. The Pickering emulsion stabilized by 0.2 wt% P/N/S-containing lignocellulose nanofibrils exhibited the highest emulsion layer height and the zeta potential reached −39.74 ± 2.88 mV. Moreover, the highest encapsulation efficiency of curcumin can be up to 94.80%. Antimicrobial test revealed that curcumin in the polymeric microcapsules exhibited an excellent inhibitory effect on S. aureus and the antibacterial efficiency reached 98.66%. These results provided important information on the comparison of different cellulose nanofibrils-stabilized Pickering emulsions and the application of controlled release of curcumin incorporated polymeric microcapsules in drug delivery system.

Binding, stability, and antioxidant activity of curcumin with self-assembled casein–dextran conjugate micelles

Yue Wu,Xiaoyong Wang

This work studied the potential of self-assembled micelles of casein–dextran conjugates (CD) as a protective carrier for bioactive curcumin. CD was prepared through the Amadori rearrangement of the Maillard reaction. In casein micelles and CD micelles, the addition of curcumin can quench the intrinsic fluorescence of casein gradually. According to the double logarithm equation, the binding constants were determined as 3.9 × 104 and 1.8 × 105 M−1 for the binding of curcumin with casein micelles and CD micelles, respectively. The higher affinity of hydrophobic binding of curcumin with CD micelles was ascribed to the more hydrophobic and compact structure of casein core surrounded by the hydrophilic dextran shell, which was supported by the fluorescence spectrum and fluorescence anisotropy of curcumin. Furthermore, CD micelles were found to be superior to casein micelles for improving the stability and radical scavenging activity of curcumin.

Chitosan-curcumin conjugate prepared by one-step free radical grafting: Characterization, and functional evaluation

Xuerong Luo,Lingyu Zhao,Imran Mahmood Khan,Lin Yue,Yin Zhang,Zhouping Wang

Curcumin (Cur) is a naturally hydrophobic polyphenol, and it has a wide range of physiological functions. But the practical application of Cur is constrained by its low water solubility and poor stability. To improve these deficiencies of Cur, a novel Cur derivative (CS-Cur) was prepared by grafting chitosan (CS) with Cur through a one-step reaction of a free radical-mediated redox system. A series of characterizations provided evidence that the grafting of CS with Cur was successful. The obtained CS-Cur showed lower crystallinity and thermal properties than CS and Cur. After grafting, the water solubility of CS-Cur was found to be 9.76 ± 2.45 g/L and greatly improved. Meanwhile, the CS-Cur showed good photothermal stability, antioxidant activity, and photodynamic antibacterial activity in an aqueous solution, and it had good in vitro biosafety. This provides an idea for the design and synthesis of novel highly water-soluble Cur derivatives and also improves the practical application of Cur in aqueous systems.

Curcumin–polydopamine nanoparticles alleviate ferroptosis by iron chelation and inhibition of oxidative stress damage

Li Lei,Jiali Yuan,Qingqing Yang,Qiuxia Tu,Haijun Yu,Liangzhao Chu,Lei Tang,Chunlin Zhang

Ferroptosis, characterized by elevated iron levels and lipid peroxidation (LPO), is a recently identified regulatory mechanism of cell death. Its substantial involvement in ischemic tissue injury, neurodegenerative disorders, and cancer positions ferroptosis inhibition as a promising strategy for managing these diverse diseases. In this study, we introduce curcumin–polydopamine nanoparticles (Cur–PDA NPs) as an innovative ferroptosis inhibitor. Cur–PDA NPs demonstrate remarkable efficacy in chelating both Fe2+ and Fe3+ in vitro along with scavenging free radicals. Cur–PDA NPs were found to efficiently mitigate reactive oxygen species, reduce Fe2+ accumulation, suppress LPO, and rejuvenate mitochondrial function in PC12 cells. Thus, these NPs can act as potent therapeutic agents against ferroptosis, primarily via iron chelation and reduction of oxidative stress.

Efficient electrochemical synthesis of green fluorescent carbon dots for curcumin detection in common foods

Chuntong Liu,Haiyan Qi,Tao Jing,Jun Li,Ming Zhao,Lixin Qiu,Qiuying Li

Curcumin (Cur) is commonly employed as a food additive in the pharmaceutical and food industries. However, improper use of curcumin can lead to adverse effects on human health. In this research, we developed an electrochemical approach for the synthesis of green fluorescence nitrogen and sulfur co-doped carbon dots (G-CDs) specifically designed for curcumin detection. The prepared G-CDs displayed strong fluorescent emissions at 540 nm, along with notable attributes such as excellent fluorescence stability against photobleaching, and resilience to changes in ionic strength. The G-CDs demonstrated selective recognition of curcumin, resulting in fluorescence quenching in a linear range of 0–140 μmol/L, with an impressive detection limit of 46.4nmol/L. Five distinct food samples were effectively detected for curcumin using the developed G-CDs nanoprobe, yielding satisfactory outcomes. This newly proposed approach offers simplicity, outstanding sensitivity, and selectivity, showcasing its promising capabilities as an alternative method for Cur detection without the need for specialized instruments.

Enhanced bioavailability of curcumin amorphous nanocomposite prepared by a green process using modified starch

Changying Chen,Zhixing Wang,Hongliang Fu,Guoqi Yu,Xiang Luo,Kewu Zhu

Curcumin (Cur), a bioactive compound extracted from plants, has attracted widespread attention due to its multiple pharmacological activities . However, the low bioavailability due to the inherent limitations in water solubility, chemical stability, and permeability poses great challenges for realizing its clinical potentials. In the current study, octenyl succinic anhydride-modified starch (OSA-S), a renewable and biodegradable biopolymer , was employed to fabricate Cur amorphous composite nanoparticles (Cur/OSA-S NPs) through a solvent-free pH-driven method with the aim to enhance Cur's bioavailability by improving its solubility and stability. Cur/OSA-S NPs, with mean sizes of about 128.9 ± 8.6 nm, encapsulation efficiencies of about 90.0 %, and the drug loading capacities around 51.0 ± 0.2 %, were successfully prepared. Cur was found to be dispersed within the composite nanoparticles in amorphous state as confirmed by the XRD and DSC characterizations. In addition, Cur/OSA-S NPs offers excellent storage, thermal and light stability, excellent re-dispersibility, and approximately 92 times better solubility than the original Cur. Furthermore, studies of dissolution and the parallel artificial membrane permeability assay (PAMPA) confirmed enhanced dissolution rates and in vitro permeabilities of Cur/OSA-S NPs. Cancer cell viability and uptake experiments revealed that Cur/OSA-S NPs possessed more potent inhibitory effects on cancer cell proliferation compared to the raw Cur. The results obtained from the current study demonstrated the effectiveness of OSA-S for manufacturing Cur amorphous composite nanoparticles with enhanced solubility, stability, and permeability, which might be valuable for further development of Cur based products for treatment of various diseases.

An Asymmetric Bacterial Cellulose Membrane Incorporating CuPt Nanozymes and Curcumin for Accelerating Wound Healing

Lei Li,Haoze Li,Junrong Wang,Yulin Xie,Minghong Gao,Zhongjun Yang,Chunxia Li

Damaged skin compromises its ability to effectively prevent the invasion of harmful bacteria into the tissue, leading to bacterial infection of the wound and hindering the healing process. To address this challenge, we have developed a multifunctional asymmetric wound dressing (CuPt-Cur-ABC) that effectively addresses the lack of bactericidal activity and the release of active ingredients in conventional bacterial cellulose (BC), which can be employed to create a barrier of defense between the wound and its surrounding environment. Compared with BC, asymmetric bacterial cellulose (ABC) used starch as a pore-causing agent, forming holes of different sizes at the top and bottom, which enhanced the ability of ABC to load and moderate-release drugs. First, as-synthesized CuPt nanozymes with an octopod nanoframe structure had multiple enzymatic activities including peroxidase-like, catalase-like, and glutathione peroxidase-like activities. Then, CuPt and curcumin (Cur) were loaded into ABC under ultrasound. Under 808 nm laser irradiation, the nanocomposites possessed good photothermal properties. So the photothermal therapy combined with chemodynamic therapy and inherent antibacterial performance of Cur achieved 99.3% and 99.6% in vitro bactericidal efficacy against Staphylococcus aureus and Escherichia coli, respectively. Moderate release of Cur can clear the excess reactive oxygen species and promote the polarization of macrophages toward the M2 type. In vivo experiments additionally confirmed that the constructed wound dressing achieved multiple functions, including effective antibacterial activity, reversing the inflammatory microenvironment, and promoting wound healing.
