A series of novel C15 urushiol derivatives were designed by introducing a pechmann structure and F-, Cl-, and Br-nitro substituents with different electronic properties into its alkyl side chain, as well as a triazolyl functional group in its aromatic oxide. Their chemical structures were determined based on the analysis of the NMR (nuclear magnetic resonance) spectroscopic and mass spectrometric data. The results showed that compound4exhibited a strong inhibition of the HepG2 cell proliferation (half maximal inhibitory concentration (IC50): 2.833 μM to human hepatocellular carcinoma (HepG2), and 80.905 μM to human normal hepatocytes (LO2)). Furthermore, it had an excellent synergistic effect with levopimaric acid. The nitrogen atom of the triazole ring formed a hydrogen-bonding interaction with Gly103, Gly154, and Tyr308, which made compound4bind to histone deacetylase (HDAC)2 more tightly. One triazole ring and His33 formed a π–π stacking effect; the other, whose branches were deep into the pocket, further enhanced the interaction with HDAC2. Meanwhile, compound4involved a hydrophobic interaction with the residues Phe210 and Leu276. The hydrophobic interaction and π–π stacking provided powerful van der Waals forces for the compounds.Keywords:urushiol;triazole;diels-alder;pechmann;anti-tumor;HDAC2

The economical production of biodiesel from lignocellulose hydrolysates requires a robust D-xylose fermentation path. The present work found that Lipomyces starkeyi AS 2.1560 and Rhodosporidium toruloides AS 2.1389 could produce micro-lipid effectively from D-xylose solution by a two-stage culture mode. Results demonstrated that both L. starkeyi and R. toruloides could achieve high lipid production in xylose solution. High cell density fermentation was carried out by a feed-batch fermentation mode. After fermentation for 48 h in a bioreactor using unsterile xylose solution by L. starkeyi, the biomass, lipid and lipid content reached 97.4 g L−1, 63.9 g L−1, and 65.5%, respectively. Accordingly, the lipid productivity reached 1.3 g L−1 h−1 in the second stage. And the overall lipid productivity was 0.55 g L−1 h−1. Therefore, L. starkeyi and R. toruloides are highly effective D-xylose-fermenting oleaginous yeasts for lipid production in xylose solution. The two-stage culture method should provide a new opportunity to make the industrial production of biodiesel possible in the future.

The solid-liquid equilibrium (SLE) of d -sorbitol in (ethanol + acetonitrile) binary solvent mixtures was explored at temperatures ranging from 278.15 K to 333.15 K under 0.1 MPa. For the temperature range investigated, the equilibrium solubility varies with the composition of the solvent and temperature. The equilibrium solubility data were then modelled using modified Apelblat equation, a variant of the combined nearly ideal binary solvent/Redich-Kister (GSM) model and Jouyban-Acree model. Computational results showed that the modified Apelblat equation and CNIBS/R-K model have the low mean deviation ( MD ). In addition, the thermodynamic properties of the solution process, including the Gibbs energy , enthalpy, and entropy were calculated by the van't Hoff analysis.

Polymerization-driven removal of pollutants in advanced oxidation processes (AOPs) offers a sustainable way for the simultaneous achievement of contamination abatement and resource recovery, supporting a low-carbon water purification approach. However, regulating such a process remains a great challenge due to the insufficient microscopic understanding of electronic structure-dependent reaction mechanisms. Herein, this work probes the origin of catalytic pollutant polymerization using a series of transition metal (Cu, Ni, Co, and Fe) single-atom catalysts and identifies the d -band center of active site as the key driver for polymerization transfer of pollutants. The high-valent metal-oxo species, produced via peroxymonosulfate activation, are found to trigger the pollutant removal via polymerization transfer. Phenoxyl radicals, identified by the innovative spin-trapping and quenching approaches, act as the key intermediate in the polymerization reactions. More importantly, the oxidation capacity of high-valent metal-oxo species can be facilely tuned by regulating their binding strength for peroxymonosulfate through d -band center modulation. A 100% polymerization transfer ratio is achieved by lowering the d -band center. This work presents a paradigm to dynamically modulate the electronic structure of high-valent metal-oxo species and optimize pollutant removal from wastewater via polymerization.

Eight kinds of chiral diacid monomers were prepared with amino acids with different side groups or configurations. Polyester-imides (PEIs) were synthesized from these diacid monomers and diphenol monomers through polycondensation reaction, and the performances and properties were compared with the chiral polyamide-imides (PAIs) previously synthesized by our work group. Their thermal properties were analyzed by thermo gravimetric analysis (TGA) and dynamic thermomechanical analysis (DMA), and it was found that the glass transition temperature (Tg) of PEI was mainly affected by the volume of side groups. Their degradability was studied through buffer degradation experiments, and the changes in their water contact angle, molecular weight, structure and appearance during the degradation process were characterized by contact angle tester, gel permeation chromatography (GPC), Fourier transform infrared spectroscopy (FTIR), and scanning electron microscopy (SEM). With degradation, the hydrophilicity of PEI was improved, and when amino acids with larger side groups or D configuration were introduced into the backbone of PEI, the degradability decreased. Keywords:chiral amino acids;polyester-imides (PEIs);degradation;thermal property

As an advanced mode of coaxial 3D printing, its outer layer can be designed to shape a soft 2D inner core to a strong stereoscopic structure. Here, we used oil-based Pickering emulsions stabilized by starch nanoparticle as the inner core, with carrageenan gel as the outer layer. Emulsion cores with different rheological properties greatly influence the overall printability (especially 70% oil content), which have been complemented by fluid dynamics simulation of pressure and velocity distributions (at the ratio of inner core to outer layer of 2:3). Vitamin D (V D ) was used as a functional additive embedded into different emulsions of inner cores and protected both by the inner and the outer layers. Particle size and stability analyses demonstrated that emulsions loaded with V D (even higher than 90% with the protection of outer layer after 15 days) still had a stabilized system.

Post-stroke cognitive impairment is a severe sequela of cerebral ischemia, with its underlying mechanisms remaining elusive and specific diagnostic biomarkers currently lacking. Growing evidence suggests that secondary neurodegeneration is closely associated with post-stroke cognitive impairment, although its metabolic basis has not been fully elucidated. Therefore, this study aimed to investigate the spatiotemporal changes and metabolic characteristics of secondary neurodegeneration and cognitive function after cortical stroke. We established a photothrombotic mouse model for post-stroke cognitive impairment research and conducted longitudinal assessments with final endpoints at 14, 32, and 84 days postsurgery. Voxel-based morphometry analysis of whole-brain regions using magnetic resonance imaging revealed that only the hippocampus exhibited gray matter alterations consistent with secondary neurodegeneration pathology. Morris water maze and open field tests demonstrated persistent impairments in recent and remote memory, along with anxietylike behaviors in photothrombotic mice. Untargeted metabolomic and lipidomic analyses were established to comprehensively characterize secondary neurodegeneration-related metabolic disturbances in the hippocampus, highlighting pathophysiological mechanisms involving oxidative stress, lipid peroxidation, neurotransmitter dysregulation, and disrupted energy metabolism. These important mechanisms were verified by immunohistochemistry, immunofluorescence staining, and real-time polymerase chain reaction. The screened potential biomarker, N -acetylneuraminic acid, was validated via targeted metabolomics in both photothrombotic mouse serum and 148 clinical samples, showing significant elevation in both cohorts. Receiver operating characteristic curve analysis and decision curve analysis confirmed the clinical utility of N -acetylneuraminic acid in diagnosing post-stroke cognitive impairment (area under the curve = 0.951, 95% confidence interval: 0.903–0.980). Flow cytometry and immunofluorescence staining revealed that N -acetylneuraminic acid activates microglia-driven neuroinflammation and oxidative stress. Our findings elucidate a potential pathological mechanism of post-stroke cognitive impairment: cortical stroke induces hippocampal accumulation of N -acetylneuraminic acid, which promotes microglial oxidative stress and inflammation, thereby triggering hippocampal secondary neurodegeneration and leading to persistent cognitive deficits. Importantly, N -acetylneuraminic acid serves as a dual-functional biomarker capable of predicting post-stroke cognitive impairment progression while dynamically tracking secondary neurodegeneration.

Vascular calcification (VC), a major complication in chronic kidney disease (CKD), is predominantly driven by osteoblastic differentiation. Recent studies have highlighted the crucial role of microRNAs in CKD’s pathogenesis. Here, our research focused on the effects of miR-204-5p and its molecular mechanisms within VC. We initially found a notable decrease in miR-204-5p levels in human aortic vascular smooth muscle cells stimulated with inorganic phosphate, using this as a VC model in vitro. Following the overexpression of miR-204-5p, a decrease in VC was observed, as indicated by alizarin red S staining and measurements of calcium content. This decrease was accompanied by lower levels of the osteogenic marker, runt-related transcription factor 2, and higher levels of α-smooth muscle actin, a marker of contractility. Further investigation showed that calcium/calmodulin-dependent protein kinase 1 (CAMK1), which is a predicted target of miR-204-5p, promotes VC. Conversely, overexpressing miR-204-5p reduced VC by suppressing CAMK1 activity. Overexpressing miR-204-5p also effectively mitigated aortic calcification in an in vivo rat model. In summary, our research indicated that targeting the miR-204-5p/CAMK1 pathway could be a viable strategy for mitigating VC in CKD patients.

Long-term levodopa administration can lead to the development of levodopa-induced dyskinesia. Gamma oscillations are a widely recognized hallmark of abnormal neural electrical activity in levodopa-induced dyskinesia. Currently, studies have reported increased oscillation power in cases of levodopa-induced dyskinesia. However, little is known about how the other electrophysiological parameters of gamma oscillations are altered in levodopa-induced dyskinesia. Furthermore, the role of the dopamine D3 receptor, which is implicated in levodopa-induced dyskinesia, in movement disorder-related changes in neural oscillations is unclear. We found that the cortico-striatal functional connectivity of beta oscillations was enhanced in a model of Parkinson’s disease. Furthermore, levodopa application enhanced cortical gamma oscillations in cortico-striatal projections and cortical gamma aperiodic components, as well as bidirectional primary motor cortex (M1) ↔ dorsolateral striatum gamma flow. Administration of PD128907 (a selective dopamine D3 receptor agonist) induced dyskinesia and excessive gamma oscillations with a bidirectional M1 ↔ dorsolateral striatum flow. However, administration of PG01037 (a selective dopamine D3 receptor antagonist) attenuated dyskinesia, suppressed gamma oscillations and cortical gamma aperiodic components, and decreased gamma causality in the M1 → dorsolateral striatum direction. These findings suggest that the dopamine D3 receptor plays a role in dyskinesia-related oscillatory activity, and that it has potential as a therapeutic target for levodopa-induced dyskinesia.

Spinal cord injury represents a severe form of central nervous system trauma for which effective treatments remain limited. Microglia is the resident immune cells of the central nervous system, play a critical role in spinal cord injury. Previous studies have shown that microglia can promote neuronal survival by phagocytosing dead cells and debris and by releasing neuroprotective and anti-inflammatory factors. However, excessive activation of microglia can lead to persistent inflammation and contribute to the formation of glial scars, which hinder axonal regeneration. Despite this, the precise role and mechanisms of microglia during the acute phase of spinal cord injury remain controversial and poorly understood. To elucidate the role of microglia in spinal cord injury, we employed the colony-stimulating factor 1 receptor inhibitor PLX5622 to deplete microglia. We observed that sustained depletion of microglia resulted in an expansion of the lesion area, downregulation of brain-derived neurotrophic factor, and impaired functional recovery after spinal cord injury. Next, we generated a transgenic mouse line with conditional overexpression of brain-derived neurotrophic factor specifically in microglia. We found that brain-derived neurotrophic factor overexpression in microglia increased angiogenesis and blood flow following spinal cord injury and facilitated the recovery of hindlimb motor function. Additionally, brain-derived neurotrophic factor overexpression in microglia reduced inflammation and neuronal apoptosis during the acute phase of spinal cord injury. Furthermore, through using specific transgenic mouse lines, TMEM119, and the colony-stimulating factor 1 receptor inhibitor PLX73086, we demonstrated that the neuroprotective effects were predominantly due to brain-derived neurotrophic factor overexpression in microglia rather than macrophages. In conclusion, our findings suggest the critical role of microglia in the formation of protective glial scars. Depleting microglia is detrimental to recovery of spinal cord injury, whereas targeting brain-derived neurotrophic factor overexpression in microglia represents a promising and novel therapeutic strategy to enhance motor function recovery in patients with spinal cord injury.

The influence of temperature, light, solution pH, water stress, salt stress, and burial depth on seed germination and seedling emergence of catchweed bedstraw and the sensitivity of that weed to commonly available herbicides in China were studied in laboratory and greenhouse. Germination occurred at day/night temperatures from 5/0 C to 30/25 C, with optimum germination at 15/10 C. Catchweed bedstraw germinated equally well under a 12-h photoperiod and continuous darkness; however, a 24-h photoperiod inhibited seed germination. Catchweed bedstraw seed is moderately sensitive to osmotic potential and salt stress, with 15 and 3% germination rates at an osmotic potential of −0.5 Mpa and salinity level of 120 mM, respectively. Maximum seed germination was observed in near neutral pH; germination was greater than 80% over a broad pH range from 5 to 8. Seedling emergence of the seeds buried at a depth of 1 cm was higher (74%) than those placed on the soil surface (20%), but declined with burial depth increasing. Few (10%) seedlings emerged when seeds were placed at a depth of 5 cm. Bensulfuron-methyl, and ethametsulfuron-methyl applied PRE and tribenuron-methyl, fluroxypyr, and florasulam applied POST can be used to provide greater than 80% control of catchweed bedstraw. The results of this study have contributed to more complete understanding of the germination and emergence of catchweed bedstraw.

Hyperphosphorylation of tau is a key pathological hallmark of Alzheimer's disease and is closely associated with cognitive impairment. Passive immunotherapy targeting hyperphosphorylated tau is a promising approach to inhibit tau pathology. However, the blood-brain barrier restricts antibodies from reaching the central nervous system and exerting their therapeutic effects. Here, we developed an intranasal biomacromolecule delivery strategy for the treatment of Alzheimer's disease using dendritic mesoporous silica nanoparticles modified with hyaluronic acid as a drug carrier. Ten-month-old C57BL/6J mice, htau mice, and 5×FAD mice were intranasally administered 100 μg of hyaluronic acid-dendritic mesoporous silica nanoparticles@4B1 (antibody dose), and brain tissues were collected to evaluate antibody delivery efficiency. Using this delivery system, the anti-p-tau 396,404 antibody 4B1 was efficiently delivered to the brains of 5×FAD mice, with an enrichment of 7.31% ± 0.18% ID/g. Entry of the 4B1 antibody into the brain ameliorated tauopathy in Alzheimer's disease model mice, reduced neuronal loss and neuroinflammation caused by tau pathology, and reversed cognitive dysfunction. These findings suggest that the nasal delivery strategy based on hyaluronic acid-responsive release is an effective method for delivering antibody biomacromolecules to the central nervous system, showing high efficiency of antibody entry into brain tissue and intracellular delivery. Our findings also demonstrate the role of p-tau 396,404 in passive immunotherapy for Alzheimer's disease, suggesting that immunotherapy targeting p-tau 396,404 is a promising strategy for ameliorating Alzheimer's disease pathology, inhibiting the aggregation of hyperphosphorylated tau, and alleviating neurological damage and cognitive dysfunction.
