Excessive high levels of reactive oxygen species (ROS) at the wound site are also one of the main reasons for difficulty in wound healing. Herein, a kind of antioxidant nanofiber membranes (NFMs) was fabricated by electrospinning sodium alginate (SA) and multi-ether fullerene derivative (mono-C60-O) solutions, which combined the good biocompatibility of SA and excellent antioxidant property of fullerene. The molecular structure of mono-C60-O was characterized by 1H NMR, 13C NMR, and MALDI-TOF mass spectrometry. The morphology of the produced SA/mono-C60-O-X NFMs was homogeneous and consistent, as characterized by scanning electron microscopy (SEM). The scavenging efficiencies of SA/mono-C60-O-25 NFMs were high, about 93.3%, 91.1%, and 92.2% for DPPH, ABTS, and ·OH, respectively. The efficiency value is also significantly higher than that of the commercial product Tegaderm. Good cell and blood compatibility exhibited a potential use of SA/mono-C60-O-25 NFMs in wound healing. In vivo wound healing studies showed that the wound of the SA/mono-C60-O-25 NFM-treated mice group was almost completely healed, while the SA NFM-treated and control groups still obviously healed on day 14. Hematoxylin and eosin (H&E), Masson, and immunohistochemical staining indicated that the SA/mono-C60-O-25 NFMs had a rapid wound healing effect due to the excellent antioxidant property of fullerene, which could reduce inflammatory factors, accelerate angiogenesis, increase collagen deposition, and exert anti-inflammatory effects.

Sepsis, a life-threatening organ dysfunction from a dysregulated infection response, causes many deaths, especially in the elderly. Aging population exacerbates the sepsis burden, requiring cost-effective prevention, detection, and treatment strategies. Hemoperfusion has emerged as an effective clinical treatment for sepsis. Herein, polystyrene-divinylbenzene microspheres functionalized with metal-phenolic networks and polyethylenimine (P-PEI Cu/TAn microspheres) with endotoxin and cell-free DNA (cfDNA) adsorption, oxidative stress relief and antibacterial activity were constructed as a multifunctional adsorbent for hemoperfusion therapy toward sepsis. The adsorbent exhibited good blood compatibility, high-efficiency adsorption of endotoxin (with an adsorption capacity of 1239.6 EU/g) and cfDNA (with a clearance ratio of 74.8%), robust scavenging of reactive oxygen/nitrogen species, and antibacterial activity against Escherichia coli (84.6%) and Staphylococcus aureus (68.1%) within 4 h. In simulated hemoperfusion, P-PEI Cu/TA0.5 microspheres achieved rapid endotoxin removal (75.3% clearance in 15 min) and effectively mitigated oxidative stress induced by endotoxin, as evidenced by preserved blood oxidoreductase activity. This work introduced a novel therapeutic approach that combines toxin adsorption, antioxidative protection, and antibacterial effects, holding significant promise for enhancing clinical sepsis management through extracorporeal blood purification.

In vitro fibrillation of food-derived proteins is increasingly accepted as a promising strategy to prepare new food ingredients, which have broader functional properties than natural protein. In current study, acidic-heating (90 °C, pH 2.0) was performed for perilla seed protein isolate (PSPI) derived from oilseed residues to evaluate the feasibility to produce protein fibrils. TEM images revealed that PSPI were hydrolyzed and reassembled a fundamental linear structure of fibrils after 6 h of heating, as reflected by the variation in particle size distribution. And the branching effects were observed with prolonged time. The conformational changes of PSPI were systematically assessed by electrophoresis, fourier transform infrared spectroscopy, fluorescence spectroscopy, circular dichroism, molecular dynamics simulations. Together with the results of surface hydrophobicity, zeta-potential confirmed that the self-assembly behavior of PSPI based fibrils was dominated by hydrophobic and electrostatic interactions. In addition, correlation analysis was combined to reflect the dynamical variation on emulsification, foaming, viscosity, interfacial and antioxidant properties during the structural evolution of PSPI. Overall, in-depth insights on the potential of perilla residues to convert into protein fibers will provide theoretical support for the regulation of functional properties of plant proteins and expand the potential applications of oilseed residues in the food field.

To utilize thinned young kiwifruit for potential industrial applications, this study compared structural properties and in vitro bioactivities of pectic polysaccharides isolated from three thinned young red-fleshed kiwifruit cultivars, including Actinidia chinensis cv. ‘Donghong’, ‘Hongshi’, and ‘Hongyang’. The results demonstrated that these young kiwifruit byproducts were rich in pectic polysaccharides, with extraction yields of 90.7–101.3 mg/g dry weight. Although their bound polyphenols (8.02–8.93 mg GAE/100 mg), esterification degrees (50.45–61.32 %), and molecular weights (7.44 × 10 4 –8.51 × 10 4 Da) varied across cultivars, the glycosidic linkage patterns showed remarkable consistency, dominated by homogalacturonan (12.07–13.84 mol%) and rhamnogalacturonan-I (70.25–75.09 mol%) regions. Notably, all these pectic polysaccharides, particularly those isolated from the ‘Donghong’ cultivar, demonstrated notable antioxidant, antiglycation, prebiotic, and immunoenhancing activities in vitro . These findings indicate that thinned red-fleshed kiwifruit byproducts are rich sources of RG-I enriched pectic polysaccharides, with significant potential as functional food ingredients.

Goji ( Lycium barbarum L.) leaves are widely processed as a healthy tea (GT) in China for their multiple biological activities. In this study, different polar segments from GT ethanolic extract were obtained using the liquid-liquid fractionation method. n -Butanol fraction (GN) had the highest antioxidant activity while ethyl acetate fraction (GE) showed the highest inhibitory effect on the α -glucosidase enzyme. The anti-diabetic effect of GE was further investigated in type 2 diabetic mellitus (T2DM) mice. The oral administration of GE (300 or 600 mg/kg) reduced fasting blood glucose (FBG) levels, improved glucose tolerance ability, and ameliorated insulin resistance (IR) in T2DM mice. GE intervention could enhance the cholesterol transportation in the serum in coincidence with decreased total cholesterol, triglyceride levels, and non-esterified fatty acid. Transcriptomic, immunofluorescence and western blotting analysis revealed that GE significantly up-regulated insulin receptor substrate 1 (IRS1), phosphatidylinositol-3-kinase (PI3K), protein kinase B (AKT), glycogen synthesis (GS), and glucose transporter 4 (GLUT4). AMP-activated protein kinases (AMPK)-mediate gluconeogenesis and glycogenolysis, such as phosphoenolpyruvate carboxy kinase (PEPCK) and glucose-6-phosphatase (G6Pase) in the liver and kidney tissues of GE groups were decreased. Thus, GE alleviated the liver, kidney, and pancreatic tissue damage. The phytoconstituents of GE were characterized, and rutin, neochlorogenic acid, and its acylated derivatives have hypoglycemic effects. Furthermore, these findings suggested that Goji tea might be used as a promising hypoglycemic candidate.

The high metastatic rate of breast tumor is the prominent reason of its poor prognosis, while the inflammatory microenvironment of tumor tissues further promoted tumor metastasis. Although photothermal therapy (PTT) displays high antitumor efficacy, the rise of inflammation-induced reactive oxygen species (ROS) during PTT exacerbate tumor metastasis. To prevent breast tumor metastasis and relieve inflammation-induced oxidative stress during PTT, herein, we developed self-assembled nanodrugs (FCP) consisting of carminic acid, iron ion, and polyvinylpyrrolidone, demonstrating photoacoustic imaging-guided PTT and anti-inflammatory activity to restrict the growth of both primary breast tumor and metastatic tumor. The as designed self-assembled spherical FCP nanoparticles (NPs, 41 nm) exhibited good light to heat conversion and broad-spectrum multienzyme (superoxide dismutase, etc.) mimetic activity to scavenge excess ROS and alleviate oxidative stress. Meanwhile, FCP NPs showed a positive correlation between the photothermal heating and ROS scavenging, allowing the continuous consumption of ROS during PTT. Importantly, owing to the intrinsic bimodal photothermal and photoacoustic imaging abilities, FCP NPs effectively guided and monitored the in vivo treatment process, which facilitated to restrict the growth of both primary and metastatic breast tumor in vivo due to coordinated PTT and anti-inflammation as confirmed by TUNEL, ki67, and matrix metalloproteinase-9 stainings. Whereas FCP NPs did not pose any potential damage to the vital organs, presenting good biosafety in vivo . We envision that self-assembled nanodrugs with concurrent anti-inflammatory and photothermal activities may have great clinical prospects in the treatment of metastatic cancers.

Intestinal anastomosis is indispensable for treating inflammatory bowel disease and colorectal cancer, yet anastomotic leakage (AL) remains a frequent, life-threatening complication that markedly prolongs hospitalization. Conventional suturing affords limited protection against leakage and infection. Here, we report an injectable, multifunctional hydrogel (PGOT) composed of γ-poly(glutamic acid) conjugated with L-cysteine and dopamine (γ-PGA-Cys-DA), oxidized konjac glucomannan (OKGM), and tannic acid (TA). Dynamic thiol-aldehyde cross-linking affords rapid gelation (~12 s) and repeatable self-healing, while catechol and polyphenol moieties confer strong wet-tissue adhesion and mechanical resilience that tolerates intestinal peristalsis and luminal pressure. In vitro and in vivo studies show that PGOT exhibits broad-spectrum antibacterial activity, robust pro-angiogenic properties, and effectively reprograms macrophages toward an M2 reparative phenotype. Application of PGOT to a rat colorectal anastomosis significantly reduces the incidence of AL and accelerates regenerative healing. These findings demonstrate that PGOT provides immediate mechanical sealing combined with bioactive regulation of the wound microenvironment, offering a promising strategy to improve postoperative outcomes after intestinal anastomosis.

Aminophylline (AMP), a xanthine-derived bronchodilator with a narrow therapeutic window, requires timely plasma concentration monitoring and pharmacokinetic (PK) studies for safe medication. However, efficient and sensitive detection methods remain limited. In this study, high quantum yield carbon dots (CA/EDA-CDs) were synthesized via a hydrothermal method using riboflavin as the carbon source, citric acid and ethylenediamine as functionalizing agents and dopants. The luminescent structure of CA/EDA-CDs, featuring a π-conjugated system (aromatic C=C and graphite N) and surface chromophores (C=N/C=O) connected via C-N, C-O, and hydrogen bonds, was controllably modulated by hydroxyl radical (·OH). Upon ·OH exposure, the emission band gaps of CA/EDA-CDs at 3.52 eV and 5.21 eV disappeared, while a new level at 2.63 eV emerged, leading to a quantitative and selective fluorescence quenching of CA/EDA-CDs to ·OH. Leveraging xanthine oxidase (XOD)-mediated AMP oxidation to hydrogen peroxide (H 2 O 2 ), which subsequently reacted with Fe 2+ to generate ·OH, a sensitive and accurate fluorescence method using CA/EDA-CDs as probe was fabricated for monitoring AMP with excellent performance. Furthermore, the developed fluorescence strategy was controllably applied to PK studies in rats administered intravenous AMP (11.25 mg/kg and 22.50 mg/kg), yielding parameters consistent with clinical data. These findings underscore the potential of CA/EDA-CDs for rapid and accurate AMP detection, addressing the clinical need for a simple and rapid monitoring method.

Saltiness–enhancing peptides are a novel class of salt substituents capable of partially replacing sodium chloride (NaCl) in food products. The current study describes the isolation and identification of saltiness–enhancing peptides from chicken skin collagen hydrolysate. Six of the peptides, including GERGPKGPKGETGPT (GT15), GPAGPPGARGQAG (GG13), GPPGPTGPA (GA9), GPPGPTGPAGER (GR12), VGPTGPAGPR (VR10), and VGSP (VP4), were found to enhance the saltiness intensity of a 0.35 % ( w / v ) solution of NaCl. These six peptides exhibited an umami taste, with GT15 and GR12 additionally having a salty taste. Molecular dynamics simulation revealed that these peptides can stably bind to the salty taste receptor transmembrane channel–like 4 via hydrogen bonds. In vitro studies revealed that these peptides exhibited antioxidant activities. These six saltiness–enhancing peptides can therefore be potentially applied for augmenting the saltiness of NaCl solution and partially replacing salt in food products while preventing the oxidative damage typically caused by high amounts of salt.

In recent years, the burden of disease caused by Staphylococcus aureus and Escherichia coli infections has been significant. Currently, the development of antibiotics faces severe challenges, as the long-term misuse of traditional antibiotics has accelerated the evolution of resistance, while the pace of novel drug development lags far behind the rate of bacterial evolution. Therefore, the development of new antibacterial agents is urgently needed. This study presents a metal-organic framework composite material loaded with zinc oxide and kaempferol (MIL-101(Fe)@Zn-KF). The prepared material was characterized using Fourier-transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), ultraviolet-visible spectrophotometry (UV-Vis), scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDS) and zeta potential analysis. Peroxidase-like activity, biosafety and antioxidant property of the composite were evaluated. The in vitro antibacterial effect of the composite against Staphylococcus aureus and Escherichia coli was investigated using the plate coating method and bacterial growth curves experiment. The synthesized material exhibited multifunctionality, such as enzyme-like activity, antibacterial property and antioxidant capacity. Compared to MIL-101(Fe)@Zn, MIL-101(Fe) and kaempferol alone, the MIL-101(Fe)@Zn-KF composite demonstrated superior antibacterial effect, with inhibition rate of 98.76% and 98.34% against Staphylococcus aureus and Escherichia coli , respectively. MIL-101(Fe)@Zn-KF was able to catalyze hydrogen peroxide to generate reactive oxygen species, which synergized with the composite itself to exert bactericidal effect. Systematic evaluation of the cytotoxicity and hemolytic effect of MIL-101(Fe)@Zn-KF revealed the excellent biosafety and clinical potential of the composite. In summary, the composite material synthesized in this study combined peroxidase-like activity, antibacterial performance, antioxidant capacity and excellent biocompatibility, providing new insights for the development of efficient and safe antibacterial materials.

Whey protein isolate (WPI), as a common emulsifier, is sensitive to environment conditions, reducing the multilayer emulsion stability. The covalent binding between WPI and polyphenols is commonly used to improve the physicochemical properties of WPI, and further improve the performance of multilayer emulsions. However, minimal studies are available that systematically examine the effect of different polyphenol-WPI complexes on the performance of multilayer emulsions. This study used three common polyphenols (i.e. luteolin, epigallocatechin gallate (EGCG), and rutin) for covalent coupling to WPI. The degree of covalent coupling of WPI and polyphenols was determined, suggesting the degree of covalent binding was EGCG > luteolin > rutin. Fourier transform infrared spectroscopy confirmed the potential coupling sites of WPI-polyphenol complexes. Fluorescence spectrum and circular dichroism spectrum determined changes in WPI structures. Among three WPI-polyphenol complexes, the WPI-EGCG (WPI-E) complexes displayed the best physicochemical characteristics, including emulsification property, antioxidant property, and thermal property. Then, the WPI-polyphenols complexes were used to fabricate multilayer emulsions for caffeic acid phenethyl ester (CAPE) encapsulation. The WPI-E complexes stabilized multilayer emulsion demonstrated the highest stability on pH, ionic strength, and temperature. The WPI-luteolin (WPI-L) stabilized multilayer emulsion presented the highest encapsulation rate, while the non-covalent complex WPI stabilized multilayer emulsion demonstrated the highest release rate during simulated in vitro digestion. This study helped us to better understand the structure-function relationships associated with the fabrication of multilayer emulsions from polyphenols.

The incorporation of antimicrobial functionalities into cellulose represents a significant advancement in biopolymer modification, enhancing its potential applications in biomedical and food sectors. In this study, we synthesized and characterized quaternary ammonium-functionalized cellulose (Q-Cell) and its intermediates, employing a three-step modification strategy (tosylation, amination, and quaternization). Structural and functional attributes of the derivatives were elucidated using SEM, TGA, FT-IR, and NMR ( 1 H and 13 C) spectroscopy. The optimized synthesis under an aqueous alkaline medium (H₂O/NaOH/triethylamine) at 5 °C for 5 h enabled selective functionalization while preserving the cellulose backbone integrity. A thorough assessment of the structural characterization and stability of the components revealed that the modified cellulose demonstrated superior antibacterial and antioxidant properties accompanied with enhanced thermal properties. The antimicrobial assessment revealed 100% inhibition of E. coli and S. aureus at 250 µg/L with MIC values of 4 and 8 µg/mL, respectively. Additionally, Q-Cell exhibited superior free radical-scavenging ability in superoxide, hydroxyl, and DPPH assays, attributed to electrostatic stabilization of radicals and charge-assisted hydrogen transfer mechanisms. These findings suggest that Q-Cell is a promising candidate for antimicrobial and antioxidant applications in biomedical, pharmaceutical, and food packaging industries.
