Background Sweet sorghum ( Sorghum bicolo r (L.) Moench) is a popular forage crop in arid and semi-arid areas due to its high drought tolerance, rapid growth, and low production costs. In addition, sweet sorghum, has relatively specific ensiling characteristics because of its high moisture content and sufficient amount of water soluble carbohydrates. Therefore, it is crucial to investigate the raw material characteristics and exogenous additive pretreatment for the regulation of silage quality. This study aimed to evaluate the effects of Lactobacillus plantarum (LP) and cellulase (CEL) on the fermentation profile, carbohydrate composition, in vitro rumen fermentation and microbiota communities of sweet sorghum silages with two different raw material characteristics (fresh sweet sorghum material (MC1); sweet sorghum material wilted outdoors for 6 h after chopping (MC2)). Results In this study, the sweet sorghum treatments were: control (without additives), LP, CEL, or a combination of LP and CEL (LP_CEL). All treated sweet sorghum samples were ensiled for 30 d. A higher content of lactic acid, propionic acid, D-fructose, glucose, sucrose and a lower content of structural carbohydrates were observed in MC1 silage than in MC2 silage. In MC2 silage, the addition of CEL or LP_CEL decreased the content of structural carbohydrates, while it increased the content of D-fructose, glucose, D-arabinose, xylitol, maltose and trehalose ( P < 0.05). The in vitro gas production at 48 h was greater in MC1 silage than in MC2 silage, and the addition of CEL or LP_CEL increased the in vitro dry matter digestibility in MC2 silage ( P < 0.05). After 30 d of ensiling, disaccharides such as sucrose, maltose and alginate were almost entirely utilized by the microorganisms, while more-consumed monosaccharides included D-fructose, glucose and L-rhamnose. Lactobacillus was the dominant genus (> 80% relative abundance) in all silage samples. Conclusions Raw material characteristics determine carbohydrate composition, in vitro digestibility, and microbial community of sweet sorghum silage. For wilted sweet sorghum with relatively low moisture content, pretreatment with CEL or LP_CEL reduced the structural carbohydrate content, increased the nonstructural carbohydrate content, and improved the digestibility of the silage. However, additives had no obvious impact on enhancing the fermentation quality of sweet sorghum silage for two raw material characteristics. Graphical abstract

In order to increase the stability and the release time of nisin, nisin/ Tremella fuciformis polysaccharide nanoparticles (NTNs) were prepared and evaluated based on their uniform particle size dispersion (168–356 nm) and high loading rate (70–86%). The condensation reaction between 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride and hydroxysuccinimide (EDC/NHS) amide was successfully used to prepare novel live particles (NTN@LP). When the mass ratio of NTN and LP was 2:1, the bacterial viability (85.6 ± 2.11%) and grafting efficiency (68.8 ± 1.96%) of the NTN@LP were both relatively high. The hemolytic activity and in vitro cytotoxicity indicated that NTN@LP is safe to use in food. The NTN@LPs were then used as natural microbial preservatives in sausages. The pH value, water activity, thiobarbituric acid-reactive substances value, and sensory evaluation of the sausages were used to assess the effect of NTN@LPs on the quality of the sausages during fermentation. NTN@LPs played an important role in reducing and decomposing biogenic amines (BAs) throughout the sausage fermentation process and displayed a strong inhibitory effect on histamine (>95%). Compared to that of the control group, the total amount of BAs in the 2:1 group was reduced by 67.90%. Tyramine content in the 2:1 NTN@LP group decreased by 78.16% compared to that in the control group, while tyramine content in the pure LP group was decreased by only 38.25%. These results indicate that NTN@LP can be potentially applied as a natural microbial preservative in the food industry.

CKF ameliorates intestinal dysfunction and neurobehavioral abnormalities in IBS-D mice. • CKF boosts L. johnsonii and tryptophan-derived indoles, notably ILA. • L. johnsonii acts as a psychobiotic for IBS-D and comorbidities. • ILA improves IBS-D intestinal and behavioral deficits mainly via AhR activation. • CKF targets the L. johnsonii -ILA-AhR/IL-22 gut-brain axis. Background Gut dysbiosis contributes to irritable bowel syndrome (IBS) by disrupting intestinal barrier functions and promoting inflammation. Chang‑kang‑fang (CKF), a Chinese herbal formula proven effective for diarrhea‑predominant IBS (IBS‑D) in a phase III trial, is thought to modulate the gut microbiota, yet the underlying mechanism remains unclear. Purpose To elucidate how CKF ameliorates IBS‑D by modulating the gut microbiota. Methods IBS‑D was induced by restraint stress, tail pinch, and senna gavage. Intestinal function, inflammation, and behavior were evaluated. Gut microbiota and metabolites were profiled via 16S rRNA sequencing and metabolomics. FMT confirmed the causal involvement of the microbiota. The key microbe and metabolite were validated in vivo . Mechanistic analyses evaluated intestinal barrier integrity, systemic inflammation, hippocampal blood‑brain barrier (BBB) function, neuroinflammation, and synaptic plasticity. Results CKF alleviated diarrhea, visceral hypersensitivity, inflammation, barrier dysfunction, and anxiety‑ and depression‑like behaviors. These benefits were associated with gut microbiota remodeling, featuring enrichment of Lactobacillus johnsonii and enhanced tryptophan metabolism. FMT confirmed that this microbial remodeling mediates the therapeutic effects of CKF. Supplementation with L. johnsonii or its metabolite indole‑3‑lactic acid (ILA) largely recapitulated these gut-brain benefits via AhR/IL‑22 activation. Mechanistically, CKF‑induced L. johnsonii enrichment activated the ILA-AhR-IL‑22 pathway, which enhanced intestinal and BBB integrity, thereby attenuating systemic and neuroinflammation, and restoring hippocampal synaptic plasticity. Conclusion This study positions CKF as a microbiota‑modulating therapy for IBS‑D that restores gut‑brain homeostasis via the L. johnsonii -ILA-AhR-IL‑22 axis, highlighting L. johnsonii and ILA as potential therapeutics for IBS and its comorbidities. The beneficial effects of probiotics on ameliorating ulcerative colitis (UC) have attracted much attention in recent years. Nevertheless, the number of these identified probiotics is still limited. In addition, the adhesion abilities of probiotics are considered to be a key determinant for probiotic efficacy. However, the relationship between the adhesion abilities of probiotics and their role in ameliorating UC has been poorly studied to date. This study measured the adhesion abilities of four Lactobacillus strains to Caco-2 cells and their anti-adhesion effects on Caco-2 cells against pathogenic bacteria, as well as their application in ameliorating the symptoms of dextran sulfate sodium-induced UC, and further illustrated the relationship between these two potential probiotic properties of probiotics and their beneficial effects on UC. Results suggested that the adhesion abilities of the four tested Lactobacillus strains exists highly strain-specific and the mechanisms of their anti-adhesion effect on Caco-2 cells against Escherichia coli may be different. Moreover, all these strains had promising effects on ameliorating UC by reducing inflammatory response and improving the intestinal mucosal barrier function, as well as promoting the production of SCFAs. In conclusion, the four tested Lactobacillus strains can be considered as alternative dietary supplements in alleviating UC. In addition, it could be concluded that there is no significant correlation between the adhesion abilities of probiotics and their role in ameliorating UC, which further illustrated that the adhesion properties of probiotics in vitro may not be suitable as the key criterion for screening potential strains with UC-alleviating effects.

A novel bacteriocin DY4-2 was produced by strain DY4-2 which was isolated from cutlassfish ( Trichiurus lepturus ) and identified as Lactobacillus plantarum . This bacteriocin reached the maximum production at stationary phase after 24 h incubation. It was purified by ethyl acetate extraction followed by ultrafiltration, Sephadex G25 chromatography and high-performance liquid chromatography. The molecular mass of bacteriocin DY4-2 was determined as 1465 Da by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) analysis. The amino acid sequence of bacteriocin DY4-2 was predicted to be HAQIGMTMNGSFR by MALDI-TOF-MS/MS. Bacteriocin DY4-2 was heat-stable (30 min at 121 °C) and kept active at the pH range of 2.5–5.5, but was sensitive to proteolytic enzymes. The mode of action of bacteriocin DY4-2 was bactericidal. It exhibited broad spectrum antimicrobial activity against fish pathogenic and spoilage bacteria, such as Pseudomonas fluorescens , P. aeruginosa , Vibrio parahaemolyticus , Aeromonas sobria and Listeria monocytogenes . The addition of partially purified bacteriocin DY4-2 in turbot fillets reduced the number of P. fluorescens by 2.7 log units at 4 °C storage for 12 days. Moreover, compared to the control, a significant decline in total viable counts and total volatile base nitrogen was observed in bacteriocin DY4-2 treated samples. Theses results suggested that bacteriocin DY4-2 had potential to act as seafood bio-preservative.

Probiotics and prebiotics have received attention in alleviating neurodegenerative diseases. Lactobacillus plantarum (L. plantarum) 69-2 was combined with galactooligosaccharides (GOS) and supplemented in a d-galactose (d-gal)-induced neurodegeneration and memory impairment mice model to explore its effects on the brain and the regulation of short-chain fatty acids. The results showed that the L. plantarum-GOS supplementation inhibited d-gal-induced oxidative stress and increased the brain’s nuclear factor erythroid 2-related factor 2 (Nrf2) levels. Butyrate, a metabolite of the gut microbiota regulated by L. plantarum combined with GOS, inhibits p-JNK expression, downregulates pro-apoptotic proteins expression and the activation of inflammatory mediators, and upregulates synaptic protein expression. This might be a potential mechanism for L. plantarum 69-2 combined with GOS supplementation to alleviate d-gal-induced neurodegeneration and memory impairment. This study sheds new light on the development of aging-related neuroprotective dietary supplements based on the gut–brain axis.

Presently, Cu(OH) 2 nanowire arrays@Cu Foam (Cu(OH) 2 –NWA@Cu Foam) nanostructure has been confirmed to possess an excellent capacity of nonenzymatical electroanalysis of histamine (His). Based on Cu(OH) 2 –NWA@Cu Foam electrode, electrochemical investigations elucidated electron transfer kinetics principles of electrooxidation of His. Substantially, it was realized that a four–electron–transferring dehydrogenation was involved in diffusion–controlled electrooxidation of His, contributing tremendously to establishing as–proposed mechanization of His electrooxidation. At optimal conditions, Cu(OH) 2 –NWA@Cu Foam electrode reflected excellent electrochemical performance such as sensitivity of 3.089 mA mM –1 cm –2 , limit of determination of 94 nM, respond time of 1.6 s and linear respond rang of 0.5–1250 μM. The outperformed electroanalytical performance can be physically explicated by the dedications from larger exposed surface area, bigger interface–hydrophilicity–deduced His diffusion coefficient and less charge transfer resistance. Moreover, the outstanding selectivity, stability and reproducibility of Cu(OH) 2 –NWA@Cu Foam electrode ensured its practical effectiveness in regulating real His levels in fermented mandarin fish. Compared with control group, the mandarin fish by inoculating Lactobacillus sakei subsp . sakei delivered less His contents, deducing that inoculation of Lactobacillus sakei subsp . sakei can effectively depress His generation during fermentation process. Eventually, the one–step as–established Cu(OH) 2 –NWA@Cu Foam nanostructure positioned itself as an upcoming possibility to pursue commercialized utilization.

In this work, we discovered a new fermentation broth that can prevent and regulate alcoholic liver disease (ALD) and intestinal flora, which fermented the mixture of Pueraria lobata, Lonicera japonica, and Crataegus pinnatifida by Lactobacillus rhamnosus 217-1. The contents of polyphenols, puerarin, total isoflavones, and amino acids were significantly increased. Animal experiments showed that the fermentation broth could improve the liver indexes of ALD mice model, increase the activity of superoxide dismutase and glutathione in liver tissue, and reduce the level of malondialdehyde (MDA). Furthermore, the fermentation broth can reduce the levels of serum lipopolysaccharide (LPS), inflammatory factors interleukin-6 (IL-6), and tumor necrosis factor-α (TNF-α). Importantly, intestinal flora analysis showed that the fermentation broth could increase the abundance of Lactobacillales and reduce the production of Gram-negative bacteria, thereby reducing the abnormal increase in bacterial diversity caused by alcohol. In conclusion, we may have discovered a new functional food raw material with great application potential. The above findings indicate that the fermentation broth can actively regulate the intestinal flora and improve liver inflammation. The underlying mechanism might be that the fermentation broth could enhance intestinal permeability and reduce the inflammatory signals and LPS transmitted through the gut-liver axis, thereby reducing the oxidative stress and inflammation of the liver caused by alcohol.

The 3,5-dicaffeoylquinic acid (3,5-DiCQA), a key component of Chuzhou chrysanthemum, one of the “Four Major Edible Chrysanthemums” in China, has been shown to enrich beneficial gut bacteria and reduce lipid synthesis based on its hydroxycinnamic acid structures. However, its low lipophilicity limits bioactivity, necessitating the development of more stable derivatives. Therefore, this study synthesized and characterized a novel esterified derivative 3,5-DiCQA dodecyl ester (3,5-DiCQAD) with markedly enhanced lipophilicity and bioactivity. In high-fat diet (HFD)-induced male C57BL/6J mice, 3,5-DiCQAD showed remarkable metabolic regulatory effects, including improvements in glucose metabolism, lipid homeostasis, BA signaling, and thermogenesis. Furthermore, 3,5-DiCQAD effectively reshaped the gut microbiota, enriching beneficial bacteria such as Lactobacillus plantarum . Moreover, the strain L. plantarum ZZ 240321 was isolated, and its fermentation of 3,5-DiCQAD generated metabolites that modulated bile acid (BA) metabolism through the FXR/SHP/LRH-1 pathway, alleviating obesity-related symptoms in antibiotic-treated mice. This research provides a novel perspective for developing plant-derived polyphenols as anti-obesity treatments. It also broadens the understanding of DiCQA's probiotic potential in the gut microbiome, highlighting its promising role in managing metabolism-associated disorders.
