Canagliflozin is a potential cardioprotective drug but exerts no significant effects on pirarubicin‑induced cardiotoxicity in rats

Hongwei Shi,Qingfu Zeng,Yunjie Wei,Hong Yang,Heng Tang,Dan Wang,Peng Pu,Rui Feng

Pirarubicin (THP), one of the anthracycline anticancer drugs, is widely used in the treatment of various types of cancer, but its cardiotoxicity cannot be ignored. Canagliflozin, the first sodium‑glucose co‑transporter‑2 inhibitor approved by the USA FDA, has been shown to have a significant effect on cardiovascular damage caused by diabetes. However, it has not been reported whether it can resist THP‑induced cardiotoxicity. The aim of the present study was to investigate the effect of canagliflozin on THP‑induced cardiotoxicity and its mechanism. A rat model of cardiotoxicity induced by THP was established and canagliflozin treatment was performed at the same time. The changes of electrocardiography, cardiac coefficient and echocardiogram were observed. The levels of lactate dehydrogenase, brain natriuretic peptide, creatine kinase MB, cardiac troponin T, superoxide dismutase (SOD) and malondialdehyde were detected. The expression of SOD2, NADPH oxidase 2, pro/cleaved‑caspase‑ and Bcl‑2/Bax were evaluated by western blotting. The primary culture of cardiomyocytes was prepared to explore the effect in vitro. After eight weeks, a series of cardiotoxicity manifestations were observed in THP rats. However, canagliflozin treatment had no significant effect on the above adverse reactions. Similarly, further studies showed that canagliflozin had no significant effect on THP‑induced cardiomyocyte injury in vitro. The present study showed that there was no significant protective effect of canagliflozin on THP‑induced cardiotoxicity and cardiomyocyte injury.

Structure of Cardamine hupingshanensis No. 2 Polysaccharide (CHP-2) and Its Effect on Streptozotocin-induced Diabetic Rats

Meidong Li,Xiufang Huang,Qin Zhang,Yifeng Zhou,Kai Luo

Four new types of water-soluble polysaccharides are isolated from Cardamine hupingshanensis . Herein, this study examines C. hupingshanensis polysaccharide no. 2 (CHP-2) with a molecular weight of 21.75 kDa. Analysis revealed that CHP-2 is composed of d -glucuronic acid, d -galacturonic acid, glucose, galactose, and L-arabinose. It is speculated that the main connecting bond is →4)-α-D-Glc(p)-(1→), →t)-α-D-Glcp-(1→). Through perfusion experiments using streptozotocin-induced diabetic rats, oral CHP-2 solution is found to significantly reduce food intake, water intake, and fasting blood glucose levels, while increasing the weights of rats. In addition, CHP-2 significantly reduces the serum and liver malondialdehyde contents and increases the activity of the corresponding antioxidant enzymes. Moreover, the effect elicited by the CHP-2 solution is similar to that of phenformin hydrochloride solution. Hence, the results of this study suggest that CHP-2 exhibits blood sugar-lowering and anti-oxidation effects.

Bifunctional SnO2 Colloid Offers No Annealing Effect Compact Layer and Mesoporous Scaffold for Efficient Perovskite Solar Cells

Liangbin Xiong,Jiashuai Li,Feihong Ye,Haibing Wang,Yaxiong Guo,Xing Ming,Qingyun Chen,Shaoan Zhang,Ruihao Xie,Zhanxu Chen,Yang Lv,Guangqi Hu,Yingji He,Guojia Fang

SnO 2 compact layer (c-SnO 2 ) frequently suffers from degradation in high temperature processes (HTP) such as crack, worse interfacial contact, and electrical properties, that is, annealing effect. To solve this problem, a kind of bifunctional SnO 2 colloid is developed by using small molecular oxalate whose organic components can be removed clearly at a low temperature process (LTP). The c-SnO 2 and SnO 2 mesoporous layer (m-SnO 2 ) derived from the fresh and aged sols with the same colloid show no annealing effect, decreasing oxygen vacancy, and adsorbing water on increasing annealing temperature. The champion devices of LTP and HTP SnO 2 planar perovskite solar cells (PSCs) achieve, respectively, stabilized photoelectric conversion efficiencies (PCEs) of 20.74% and 20.70%. In contrast, the performance of champion devices of their mesoporous counterparts is significantly improved, showing nearly hysteresis free character with stabilized PCEs of 22.40% and 22.37%, respectively. The inclusion of m-SnO 2 plays a role of an energy bridge, improving electrons collection efficiency, which is supported by photoluminescence and transient photoluminescence characterizations. HTP SnO 2 mesoporous PSCs can preserve 97.6% and 80% of their initial PCEs after aging for 25 weeks and 8-h irradiated/16-h dark cycle within 104 h. The high stability of HTP SnO 2 PSCs may ascribe to low oxygen vacancy and adsorbed water of HTP SnO 2 .

Fabrication of self-cleaning photocatalytic durable building coating based on WO3-TNs/PDMS and NO degradation performance

Guanyu Liu,Huiyun Xia,Yanhui Niu,Xu Zhao,Gengtong Zhang,Lifang Song,Huaxin Chen

Practical significance in terms of the long-term effective application of photocatalytic coatings is greatly challenged by its easy pollution and poor self-cleaning performance. In this paper, a self-cleaning photocatalytic WO 3 -TiO 2 nanorod (MWT) /PDMS building coating (MWT coating) was prepared by spraying methods. The surface hierarchical structure fabricated by MWT and PDMS was a crux to construct a superhydrophobic surface, which endowed MWT coating with self-cleaning performance. Besides, the MWT coating exhibited prominent durability and antifouling property after 450 d practical application of the external wall. The excellent self-cleaning property of the coating was confirmed by the removal and resistance to the adhesion of powder particles. Meanwhile, the photocatalytic properties of MWT endowed MWT coating with the ability to degrade organic pollutants on the surface. The color difference of the coating polluted by methyl red was only 1.18 and 4.77 after 24 h UV and visible irradiation, respectively. This mutual benefit effect of self-cleaning and photocatalysis was also reflected in the degradation of NO. In the first five cycles, the degradation efficiency of NO decreased by 4.74% gradually under visible light due to the adhesion of oxidation products, and most of the MWT on the coating surface was regenerated after flushing, the PCD efficiency increased by 3.71%. Furthermore, after 30 days of outdoor weathering test, the PCD efficiency of MWT coating decreased by 11.74% and 12.05% under UV and visible light, respectively, which lays a foundation for the long-term effectiveness of the MWT coating in the process of use.

A superior Fe-Zr mixed oxide catalyst for the simultaneous reduction of NO and SO2 with CO

Liam John France,Wan Li,Yanke Zhang,Wentao Mu,Zhihang Chen,Junjun Shi,Qiang Zeng,Xuehui Li

A series of Fe-Zr mixed-metal oxides were prepared and examined for the simultaneous reduction of NO and SO 2 with CO. The catalyst containing 40 mol% exhibited near-complete conversion of both pollutants at a minimum temperature of 350 °C and a GHSV of 90,000 h −1 . The addition of Fe 3+ to ZrO 2 decreased the crystallinity of the latter phase and significantly enhanced surface area. Concentrations of 30 mol% or greater produced segregated FeO x phases, which affected catalytic performance in distinctly different ways: supported Fe 3+ (30 mol%) possessed similar activity to doped Fe 3+ ; α-Fe 2 O 3 (50 mol%) inhibited activity due to its’ lower reducibility; an amorphous phase (40 mol%) promoted iron-weighted areal activity. A complete identification of the ambiguous component was not successfully established, but the obtained properties revealed that it exhibited weaker interaction with the support and had a size greater than that of oligomeric-Fe 2 O 3 yet smaller than 3 nm.

SO2-tolerant and H2O-promoting Pt/C catalysts for efficient NO removal via fixed-bed H2-SCR

Tu Baosheng,Shi Nian,Sun Wei,Cao Limei,Yang Ji

In this paper, Pt supports on carbon black powder (Vulcan XC-72) were synthesized via a hydrothermal method for selective catalytic reduction (SCR) of NO with H 2 in the presence of 2 vol% O 2 over a wide temperature of 20–300 °C. The results showed that the 3 and 5 wt% Pt/C catalysts resulted in high NO conversion (>90 %) over a temperature range of 120 to 300 °C, and the maximum NO conversion of 98.6 % was achieved over 5 wt% Pt/C at 120 °C. Meanwhile, the influence of SO 2 and H 2 O on the catalyst performance of 3 wt% Pt/C was investigated. The catalysts exhibited good SO 2 poisoning resistance when the SO 2 concentration was lower than 260 ppm. Moreover, a positive effect on NO conversion was detected with the addition of 3 and 5 vol% H 2 O in the feed gas stream. TEM image and good NO conversion performance of the Pt/C catalysts

Influence of calcination temperature on CeO2-CuO catalyst for the selective catalytic reduction of NO with NH3

Wei-guo Pan,Yue Zhou,Rui-tang Guo,Wen-long Zhen,Jie-nan Hong,Hong-jian Xu,Qiang Jin,Cheng-gang Ding,Shi-yi Guo

CeO-CuO catalysts were prepared by citric acid method at different calcination temperatures. The purpose of this study is to investigate the influence of calcination temperature on the catalytic activity of CeO 2 -CuO catalyst for the selective catalytic reduction of NO with NH 3 . As can be seen from the results, Ce-Cu(450) is of the best low temperature selective catalytic reduction (SCR) activity among the three catalysts, which may be resulted from its large surface area, good redox ability, strong NH 3 adsorption capacity, and high concentration of Cu 2+ and active oxygen species on the surface. © 2013 American Institute of Chemical Engineers Environ Prog, 33: 385–389, 2014

Epitaxial growth of Bi4Ti3O12-BiPO4 Z-scheme heterojunction to promote carrier transfer for photocatalytic oxidation of NO

Zhinian Liu,Guojun Li,Mingjia Zhang,Chen Zhang,Wangsheng Zheng,Xiangting You,Shule Zhang,Qin Zhong

The lack of compactness in heterojunction interfaces and poor charge separation is a great challenge in developing high-efficiency heterojunction photocatalysts . Herein, a novel Bi 4 Ti 3 O 12 -BiPO 4 heterojunction was successfully prepared for the first time by epitaxial growth of BiPO 4 on the surface of Bi 4 Ti 3 O 12 nanosheets . The optimized Bi 4 Ti 3 O 12 -BiPO 4 -0.5 increased the NO oxidation efficiency to 73.05%, surpassing pure Bi 4 Ti 3 O 12 (63.45%) and BiPO 4 (8.35%). Experiments and theoretical calculations indicated that the closely contacted heterointerface between BTO and BPO promoted the generation of the built-in electric field, which led to the formation of the Z- scheme transfer pathway for the photogenerated carriers . Therefore, the separation of photogenerated carriers was facilitated while retaining high redox potential , generating more ·O 2 – and ·OH to participate in NO oxidation. Furthermore, the adsorption of NO and O 2 was enhanced by introducing BiPO 4 , further improving the photocatalytic NO oxidation performance. This work emphasizes the critical role of heterointerface in accelerating charge transfer, providing a basis for the design and construction of tightly contacted heterojunction photocatalysts .

Efficient NO reduction by CO on Cu/Ce co-modified CaO in calcium looping CO2 capture process

Yi Fang,Shoubing Chai,Zhiwei Chu,Zirui He,Yingjie Li

In the calcium looping CO 2 capture process, using CO in the flue gas to reduce NO at a high molar ratio of CO to NO (e.g., above 10:1) can achieve efficient removal of NO. However, the lots of unreacted CO causes secondary pollution. In this study, a novel Cu/Ce co-modified CaO was proposed for cyclic CO 2 capture and the catalytic NO reduction at a low CO:NO molar ratio (e.g., 2:1). The experimental findings indicate that Cu/Ce modification improves the performance of CaO in both catalytic NO reduction and carbonation. The Cu/Ce co-modified CaO contained the optimal Cu/Ce/Ca molar ratio of 1:5:100 possesses both NO removal and CO conversion efficiencies of above 99 %, along with CO 2 removal efficiency exceeding 88%. Microscopic analysis determines that the electron transfer between Cu and Ce facilitates the conversion of lattice oxygen into surface oxygen, mitigating the competition between CO 2 and CO for surface oxygen. Furthermore, density functional theory calculations demonstrate that CO/NO/CO 2 exhibits non-competitive adsorption behavior on Cu/Ce co-modified CaO. Consequently, Cu/Ce-modified CaO possesses superior catalysis for efficient NO reduction by CO at low CO:NO molar ratio in calcium looping process.

Glutathione Depletion-Induced ROS/NO Generation for Cascade Breast Cancer Therapy and Enhanced Anti-Tumor Immune Response

Jing Wang,Yanxiang Sang,Weijian Chen,Liang Cheng,Wenxiang Du,Hongjie Zhang,Benyan Zheng,Lei Song,Yuan Hu,Xiaopeng Ma

Introduction As an effective alternative choice to traditional mono-therapy, multifunctional nanoplatforms hold great promise for cancer therapy. Based on the strategies of Fenton-like reactions and reactive oxygen species (ROS)-mediated therapy, black phosphorus (BP) nanoplatform BP@Cu2O@L-Arg (BCL) co-assembly of cuprous oxide (Cu2O) and L-Arginine (L-Arg) nanoparticles was developed and evaluated for synergistic cascade breast cancer therapy. Methods Cu2O particles were generated in situ on the surface of the BP nanosheets, followed by L-Arg incorporation through electrostatic interactions. In vitro ROS/nitric oxide (NO) generation and glutathione (GSH) depletion were evaluated. In vitro and in vivo anti-cancer activity were also assessed. Finally, immune response of BCL under ultrasound was investigated. Results Cu2O was incorporated into BP to exhaust the overexpressed intracellular GSH in cancer cells via the Fenton reaction, thereby decreasing ROS consumption. Apart from being used as biocompatible carriers, BP nanoparticles served as sonosensitizers to produce excessive ROS under ultrasound irradiation. The enhanced ROS accumulation accelerated the oxidation of L-Arg, which further promoted NO generation for gas therapy. In vitro experiments revealed the outstanding therapeutic killing effects of BCL under ultrasound via mechanisms involving GSH deletion and excessive ROS and NO generation. In vivo studies have illustrated that the nanocomplex modified the immune response by promoting macrophage and CD8+ cell infiltration and inhibiting MDSC infiltration.Discussion BCL nanoparticles exhibited multifunctional characteristics for GSH depletion-induced ROS/NO generation, making a new multitherapy strategy for cascade breast cancer therapy.

In Situ injectable hydrogel encapsulating Mn/NO-based immune nano-activator for prevention of postoperative tumor recurrence

Shengnan Huang,Chenyang Zhou,Chengzhi Song,Xiali Zhu,Mingsan Miao,Chunming Li,Shaofeng Duan,Yurong Hu

Postoperative tumor recurrence remains a predominant cause of treatment failure. In this study, we developed an in situ injectable hydrogel, termed MPB-NO@DOX + ATRA gel, which was locally formed within the tumor resection cavity. The MPB-NO@DOX + ATRA gel was fabricated by mixing a thrombin solution, a fibrinogen solution containing all-trans retinoic acid (ATRA), and a Mn/NO-based immune nano-activator termed MPB-NO@DOX. ATRA promoted the differentiation of cancer stem cells, inhibited cancer cell migration, and affected the polarization of tumor-associated macrophages. The outer MnO 2 shell disintegrated due to its reaction with glutathione and hydrogen peroxide in the cytoplasm to release Mn 2+ and produce O 2 , resulting in the release of doxorubicin (DOX). The released DOX entered the nucleus and destroyed DNA, and the fragmented DNA cooperated with Mn 2+ to activate the cGAS-STING pathway and stimulate an anti-tumor immune response. In addition, when MPB-NO@DOX was exposed to 808 nm laser irradiation, the Fe-NO bond was broken to release NO, which downregulated the expression of PD-L1 on the surface of tumor cells and reversed the immunosuppressive tumor microenvironment. In conclusion, the MPB-NO@DOX + ATRA gel exhibited excellent anti-tumor efficacy. The results of this study demonstrated the great potential of in situ injectable hydrogels in preventing postoperative tumor recurrence.

Dual-layer microneedles with NO/O2 releasing for diabetic wound healing via neurogenesis, angiogenesis, and immune modulation

Changjiang Liu,Kun Liu,Dong Zhang,Yuting Liu,Yifeng Yu,Haifei Kang,Xianzhen Dong,Honglian Dai,Aixi Yu

Diabetic wounds present multiple functional impairments, including neurovascular dysregulation, oxidative imbalance, and immune dysfunction, making wound healing particularly challenging, while traditional therapeutical strategies fail to address these complex issues effectively. Herein, we propose a strategy utilizing dual-layer microneedles to deliver therapeutic gases by modulating neurovascular coupling and immune functions for diabetic wound treatment. The microneedle can respond to reactive oxygen species (ROS) in the diabetic microenvironment and subsequently generate oxygen (O 2 ) and nitric oxide (NO). These gases comprehensively promote neuro-vascular regeneration, reduce oxidative stress levels, and attenuate inflammation. In vivo studies demonstrate that the microneedle can accelerate diabetic wound healing by modulating neurovascular regeneration and inflammatory processes. Transcriptomic analyses further validate the involvement of related advantageous signaling pathways. The potential mechanism involves the activation of the PI3K-AKT-mTOR pathway to facilitate autophagy, ultimately accelerating the healing process. Thus, our multifunctional dual-layer microneedles provide an effective strategy for treating diabetic wounds.
