Enhanced degradation of post-consumer polyethylene terephthalate (PET) wastes by fusion cutinase: Effects of anchor and linker peptides

Qingjun Jia,Zixuan Zhang,Lankai Su,Shu Bai,Di Cai,Changjing Chen,Linling Yu,Yan Sun

Enzymatic degradation of poly(ethylene terephthalate) (PET) into its corresponding monomers provides an attractive route for green plastic recycling and environmental remediation applications. However, the binding of PET hydrolases toward the PET surface is a rate-limiting step in PET degradation. Hence, we selected three anchor peptides and four linker peptides to fuse the previously engineered leaf-branch compost cutinase for facilitating enzyme binding and improving degradation of real post-consumer PET wastes of Coca Cola and C′eastbon PET bottles, with using the amorphous PET as a control. The systematical characterization of the seven cutinases revealed that the anchor peptide with moderate hydrophobicity plus chaperone activity and the flexible linker peptide with small hydrophobicity but long length were more suitable for each domain to serve its own role, affording the facilitated PET binding affinities and enhanced PET degradation performance. Thus, the optimal fusion cutinase possessed up to 3.3 times higher catalytic efficiency ( k cat / K m ) and 76 % increased long-term degradation performance than the parental cutinase, respectively. Further, complete degradation of C′eastbon PET wastes and 77 % degradation of Coca Cola PET wastes were achieved by the optimal cutinase of 33.3 nmol·g −1 PET in 96 h, proving its superiority against the real post-consumer PET wastes.

High-purity H2 production from mixed PVC/PET plastic wastes through tandem hydrothermal depolymerization and aqueous phase reforming

Hongcai Su,Yuheng Wu,Jie Pan,Lingjun Zhu,Shurong Wang,Yanjun Hu

Mixed plastic wastes are posing substantial challenges to the environment due to their mass production and difficulty in separation, especially mixed polyvinyl chloride/polyethylene terephthalate (PVC/PET) plastic wastes. In this work, a novel tandem thermochemical process including hydrothermal depolymerization of mixed PVC/PET plastic wastes, and subsequent catalytic aqueous phase reforming (APR) of hydrolysate solution was developed. The activated carbon derived from PVC (PVC-AC) supported Ru catalysts were prepared and successfully used in the catalytic APR of hydrolysate solution for high-purity H 2 production. Besides, the presence of NaOH realized in-situ capture of CO 2 in APR process, which promoted the water-gas shift reaction and inhibited methanation reactions. As a result, a high H 2 yield of 12.23 mol/kg Plastic with H 2 concentration of ca. 98% was obtained via tandem hydrothermal conversion of mixed PVC/PET plastic wastes. The by-products from the entire tandem process including the terephthalic acid (TPA) and sodium chloride (NaCl) were separated and purified for further industrial application. This work provided a sustainable route for conversion of mixed plastic wastes into value-added fuel.

Flow reactor-based batch production of direct radiolabelable superparamagnetic iron oxide for PET/MRI dual-modality contrast agent

Jie Zhong,Shuang Yan,Yanling Yu,Guihao Hu,Wei Chen,Haiyu Wang,Wenxue Li,Jinping Yuan,Suping Li,Xinghui Li,Xiaoming Zhang,Changqiang Wu

Radiolabeled superparamagnetic iron oxide nanoparticles (SPIONs) have gained significant attention for their potential in dual-modality imaging, combining positron emission tomography (PET) and magnetic resonance imaging (MRI). The integration provides clinicians with enhanced and complementary imaging information, improving the accuracy of disease diagnosis. However, the complexity of ligand synthesis for functionalization and stable radiolabeling and the difficulty in achieving controlled large-scale production with consistent nanoparticle quality, both of which limit their potential in clinical applications. To overcome these limitations, we report a novel method for preparing SPIONs utilized a simple dual-chelation functional ligand and a continuous synthesis approach to facilitate their clinical translation. The functionalized SPIONs were successfully synthesized by using a flow synthesis technique and the dual-chelation functional ligand (PAsp- g -DA/EA). Compared with traditional synthesis methods, this approach enabled mass production with high reproducibility. The obtained SPIONs showed ultra-small sizes (5.1 ± 0.1 nm), good monodispersity, excellent relaxivity, and superior biocompatibility. The dual-chelation functional ligand enabled direct and efficient radiolabeling of SPIONs with [ 64 Cu]Cu 2+ achieving over 80 % efficiency and outstanding labeling stability. After intravenous injection into rats, high-quality PET and MRI images were obtained. Furthermore, both PET and MRI demonstrated significant nanoprobe accumulation in tumor-bearing mice. In this study, a dual-chelation functional ligand was employed to facilitate the batch and reproducible synthesis of directly radiolabelable SPIONs, thereby addressing key challenges associated with the clinical translation of PET/MRI dual-modality nanoprobes. The nano-probes developed demonstrate excellent biocompatibility, superior imaging efficacy in both PET and MRI, making them potential candidates for medical PET/MRI dual-modality probes.

Chen Lijuan,Li Xiaochen,Ge Yao,Li Huiqiang,Li Ruili,Song Xiaosheng,Liang Jianfei,Zhang Weifeng,Li Xiaona,Wang Xiaoqi,Wang Yunjuan,Wu Yaping,Bai Yan,Wang Meiyun

Background A novel positron emission tomography (PET) imaging tracer, [ 18 F] SynVesT-1, targeting synaptic vesicle glycoprotein 2 (SV2A), has been developed to meet clinical demand. Utilizing the Trasis AllinOne-36 (AIO) module, we’ve automated synthesis to Good Manufacturing Practice (GMP) standards, ensuring sterile, pyrogen-free production. The fully GMP-compliant robust synthesis of [ 18 F] SynVesT-1 boosting reliability and introducing a significant degree of simplicity and its comprehensive validation for routine human use. Results [ 18 F] SynVesT-1 was synthesized by small modifications to the original [ 18 F] SynVesT-1 synthesis protocol to better fit AIO module using an in-house designed cassette and sequence. With a relatively small precursor load of 5 mg, [ 18 F] SynVesT-1 was obtained with consistently high radiochemical yields (RCY) of 20.6 ± 1.2% (the decay-corrected RCY, n  = 3) at end of synthesis. Each of the final formulated batches demonstrated radiochemical purity (RCP) and enantiomeric purity surpassing 99%. The entire synthesis process was completed within a timeframe of 80 min (75 ± 3.1 min, n  = 3), saves 11 min compared to reported GMP automated synthesis procedures. The in-human PET imaging of total body PET/CT and time-of-flight (TOF) PET/MR showed that [ 18 F] SynVesT-1 is an excellent tracer for SV2A. It is advantageous for decentralized promotion and application in multi-center studies. Conclusion The use of AIO synthesizer maintains high production yields and increases reliability, reduces production time and allows rapid training of production staff. Besides, the as-prepared [ 18 F] SynVesT-1 displays excellent in vivo binding properties in humans and holds great potential for the imaging and quantification of synaptic density in vivo.

An unmodified recycled polyethylene terephthalate (PET) nanofibrous membrane for water-in-oil and oil-in-water emulsions separation

Panchan Dansawad,Yanxiang Li,Lixia Cao,Haigang Gao,Chaoyong Yang,Enming Huang,Siming You,Wangliang Li

A nanofibrous membrane made of unmodified recycled polyethylene terephthalate (rPET) was fabricated by electrospinning using plastic water bottle as raw materials. Using rPET polymer to fabricate membranes may help limit the discharge of plastic bottle waste into the environment and can treat oily wastewater simultaneously, especially emulsions. The variation in the concentration of rPET polymer and the properties of electrospun nanofibers , such as surface morphology and fiber diameter , had a significant impact on both the efficiency of oil droplet capturing and the surface wettability of the membrane. Water-in-oil emulsions and oil-in-water emulsions were separated using the nanofibrous membranes of different concentrations of rPET polymer, and the separation efficiencies at different concentrations of oil, and oil types were measured. The rPET nanofibrous membrane exhibited a water contact angle of 140.1° and was superoleophilic in air and underwater. The membranes exhibited excellent separation performances: >98% separation efficiency and a flux of more than 6500 L m -2 h -1 for water in hexadecane and octane emulsions; >95 % efficiency and a flux of 1509.70 L m -2 h -1 after three separation cycles of water-in-oil emulsions. For oil-in-water emulsions, the separation efficiencies were higher than 95 %, and flux was more than 3525.22 L m -2 h -1 for hexadecane octane and heptane in water emulsions, with an efficiency of >87.9 % and flux of 4186.20 L m -2 h -1 after three separation cycles. Moreover, the as-prepared membrane exhibited high performance of oil-water separation in harsh conditions, including strong acidic, strong basic, and strong salt solutions, with separation efficiencies of more than 98 % and flux of more than 12,700 L m -2 h -1 . However, as water accumulated on the membrane surface , absorption and separation efficiency and flux were lowered significantly. This suggests the need to enhance the efficiency of the membrane and reduce the chance of water accumulation on the membrane surface.

Preparation of flame retardant and anti-dripping PET fabric based on heme chloride dyeing and mussel-inspired strategy

Yongli Zhang,Chunlong Zuo,Xiaoyu Liu,Ziyin Ye,Wei Tan,Ping Li,Yuanlin Ren,Xiaohui Liu

Polyethylene terephthalate (PET) fibers are widely used in many fields, but their flammability and molten droplet characteristics during combustion pose a significant safety risk. To address the issue, a novel approach was developed to enhance the flame retardant and anti-dripping performances of PET fabric through a combination of dyeing and mussel-inspired strategy. First, PET fabric was directly dyed with heme chloride (HC) to produce dyed PET fabric (HC-PET), which was then treated by a mussel-inspired tannic acid-based flame retardant (TAPP) and an impregnation-drying-curing process to prepare flame retardant PET fabric (FR-PET). The results indicated that the total heat release (THR) and peak smoke production rate (PSPR) of FR-PET were decreased by 22.8 % and 49.7 %, respectively, and the limiting oxygen index (LOI) was increased from 21 % to 29.8 %, showing good flame retardant properties. Additionally, the UV resistance and antibacterial performances of FR-PET were also significantly improved. In summary, this study provided a feasible approach for the preparation of multifunctional PET textiles with dyeing, flame retardant, anti-dripping, UV resistance and antibacterial performances.

Synthesis of Polyethylene Terephthalate (PET) with High Crystallization and Mechanical Properties via Functionalized Graphene Oxide as Nucleation Agent

Yingdi Dan,Yao Wang,Miaorong Zhang,Linjun Huang,Quankai Sun,Pengwei Zhang,Zengkun Li,Wei Wang,Jiangguo Tang

In this work, a novel functionalized graphene oxide nucleating agent (GITP) was successfully synthesized using a silane coupling agent (IPTES), and polymer block (ITP) to efficiently improve the crystallization and mechanical performance of PET. To comprehensively investigate the effect of functionalized GO on PET properties, PET/GITP nanocomposites were prepared by introducing GITP into the PET matrix using the melt blending method. The results indicate that PET/GITP exhibits better thermal stability and crystallization properties compared with pure PET, increasing the melting temperature from 244.1 °C to 257.1 °C as well as reducing its crystallization half-time from 595 s to 201 s. Moreover, the crystallization temperature of PET/GITP nanocomposites was increased from 185.1 °C to 207.5 °C and the tensile strength was increased from 50.69 MPa to 66.8 MPa. This study provides an effective strategy for functionalized GO as a nucleating agent with which to improve the crystalline and mechanical properties of PET polyester.

The Development of a 3D PET Fibrous Scaffold Modified with an Umbilical Cord dECM for Liver Tissue Engineering

Yang Li,Yang Zhang,Kebo Zhong,Shuguang Liao,Guifeng Zhang

Organ and tissue dysfunction represents a clinically significant condition. By integrating cell biology with materials science, tissue engineering enables the reconstruction and restoration of damaged tissues or organs, offering a noninvasive repair approach. In our study, we replicated the cellular growth environment by utilizing a human umbilical cord-derived decellularized extracellular matrix (dECM) as a modifying agent for the polyethylene terephthalate (PET) polymeric fiber scaffold. This allowed us to create a dECM-coated polyester fiber-based scaffold, PET-dECM, tailored for liver tissue engineering purposes. We effectively produced a decellularized human umbilical cord-derived ECM through a combined decellularization process involving trypsin/EDTA, TritonX-100, and sodium deoxycholate. The application of the dECM coating onto the PET material was accomplished through several steps, such as ester hydrolysis, EDC/NHS-activated crosslinking, and dECM conjugation. The biological performance of the PET-dECM was validated using RG cell culture assays. Notably, the dECM coating significantly improved PET’s hydrophilicity and biocompatibility, thereby aiding cell adhesion, proliferation, and functional differentiation (p< 0.05). It was further found that the hepatocyte function of HepaRG was significantly enhanced on the PET-dECM, which may be attributed to the dECM’s ability to facilitate the restoration of cell polarity. The PET-dECM holds promise as an effective hepatocyte culture carrier and could potentially find application in liver tissue engineering.

Phosphorus-functionalized Schiff base flame retardant for PET and its filaments: enhancing fire resistance, anti-dripping, and strength

Zhenhui Bai,Rong Zhou,Qun Wang,Mao Li,Wenjun Yan,Qiaoli Xu,Hengshu Zhou,Xiaoqiang Li,Ruchao Yuan,Jinping Guan,Faxue Li

The combination of enhanced char formation capacity and radical quenching effect in polymer materials at elevated temperatures is crucial for improving flame inhibition and anti-dripping performance. This study developed a phosphorus (P)-functionalized Schiff base flame retardant (HSCP) specifically for poly(ethylene terephthalate) (PET). Notably, incorporation of merely 4 wt% of HSCP effectively enhanced the anti-dripping performance of PET. The PET composites containing 8 wt% of HSCP (PET/HSCP8) achieved a limiting oxygen index of 30.0% with complete anti-dripping performance, attaining a V-0 rating in vertical burning. Furthermore, PET/HSCP8 exhibited a remarkable 16.4% reduction in total smoke production compared to unmodified PET. The enhanced fire safety of PET/HSCP was achieved through the dual-phase mechanism of HSCP, which combines the char-forming effect with the quenching effect. Importantly, the PET/HSCP8 composites maintain excellent interfacial compatibility, enabling the successful fabrication of filaments through melt-spinning that simultaneously exhibit enhanced fire safety and preserved mechanical properties.

Engineering of 2D/2D MoS2/CdxZn1−xS Photocatalyst for Solar H2 Evolution Coupled with Degradation of Plastic in Alkaline Solution

Yiqun Li,Shipeng Wan,Cheng Lin,Yujie Gao,Yuan Lu,Luyang Wang,Kan Zhang

Sufficiently utilizing photogenerated electron/hole pairs for a coupled redox enables green, sustainable, and low-cost photocatalysis. Herein, a series of MoS 2 /Cd x Zn 1− x S photocatalysts with uniform 2D/2D structure are reported for solar water–reductive H 2 products coupled with the degradation of polyethylene terephthalate (PET) plastic. The MoS 2 /Cd 0.5 Zn 0.5 S heterojunction with a loading mass of 4.3 wt% MoS 2 exhibits the best H 2 evolution rate of 15.90 mmol g −1 h −1 in an aqueous solution due to its appropriate redox ability, great light absorption, and carrier separation abilities. Then, the oxidation products of PET are analyzed by 1 H-NMR spectroscopy, which demonstrates the PET being efficiently degraded to molecule compounds, such as formate and acetate. The work provides a feasible solution to the problem of plastic pollution and meets the demand for sustainable clean energy simultaneously.

A New γ-Glutamyltranspeptidase-Based Intracellular Self-Assembly of Fluorine-18 Labeled Probe for Enhancing PET Imaging in Tumors

Siqin Ye,Shijie Wang,Dingyao Gao,Ke Li,Qingzhu Liu,Bainian Feng,Ling Qiu,Jianguo Lin

γ-Glutamyltranspeptidase (GGT) is a cell -membrane-associated enzyme which has been recognized as a promising biomarker for the diagnosis of many malignant tumors. Herein, we rationally designed a fluorine-18 labeled small-molecule probe, [18F]γ-Glu-Cys(StBu)-PPG(CBT)-AmBF3 (18F-1G), by applying a biocompatible CBT-Cys condensation reaction and ingeniously decorating it with a GGT-recognizable substrate, γ-glutamate (γ-Glu), for enhancing PET imaging to detect GGT level of tumors in living nude mice. The probe had exceptional stability at physiological conditions, but could be efficiently cleaved by GGT, followed by a reduction-triggered self-assembly and formation of nanoparticles (NPs) progressively that could be directly observed by transmission electron microscopy (TEM). In in vitro cell experiments, 18F-1G showed GGT-targeted uptake contrast of 2.7-fold to that of 18F-1 for the detection of intracellular GGT level. Moreover, the higher uptake in GGT overexpressed HCT116 tumor cells (∼4-fold) compared to GGT-deficient L929 normal cells demonstrated that 18F-1G was also capable of distinguishing some tumor cells from normal cells. In vivo PET imaging revealed enhanced and durable radioactive signal in tumor regions after 18F-1G coinjecting with 1G, thus allowing real-time detection of endogenous GGT level with high sensitivity and noninvasive effect. We anticipated that our probe could serve as a new tool to investigate GGT-related diseases in the near future.

Quantitative assessment of renal functions using 68Ga-EDTA dynamic PET imaging in renal injury in mice of different origins

Ding Ying,Liu Yu,Zhang Li,Deng Yinqian,Chen Huanyu,Lan Xiaoli,Jiang Dawei,Cao Wei

Background: Early detection of kidney diseases can be challenging as conventional methods such as blood tests or imaging techniques (computed tomography (CT), magnetic resonance imaging (MRI), or ultrasonography) may be insufficient to assess renal function. A single-photon emission CT (SPECT) renal scan provides a means of measuring glomerular filtration rates (GFRs), but its diagnostic accuracy is limited due to its planar imaging modality and semi-quantification property. In this study, we aimed to improve the accuracy of GFR measurement by preparing a positron emission tonometry (PET) tracer 68Ga-Ethylenediaminetetraacetic acid (68Ga-EDTA) and comprehensively evaluating its performance in healthy mice and murine models of renal dysfunction. Methods: Dynamic PET scans were performed in healthy C57BL/6 mice and in models of renal injury, including acute kidney injury (AKI) and unilateral ureter obstruction (UUO) using 68Ga-EDTA. In a 30-min dynamic scan, PET images and time-activity curves (TACs) were acquired. Renal function and GFR values were measured using renograms and validated through serum renal function parameters, biodistribution results, and pathological staining. Results: 68Ga-EDTA dynamic PET imaging quantitatively captured the tracer elimination process. The calculated GFR values were 0.25 ± 0.02 ml/min in healthy mice, 0.01 ± 0.00 ml/min in AKI mice, and 0.25 ± 0.04, 0.29 ± 0.03 and 0.24 ± 0.01 ml/min in UUO mice, respectively. Furthermore, 68Ga-EDTA dynamic PET imaging and GFRPET were able to differentiate mild renal impairment before serum parameters indicated any changes. Conclusions: Our findings demonstrate that 68Ga-EDTA dynamic PET provides a reliable and precise means of evaluating renal function in two murine models of renal injury. These results hold promise for the widespread clinical application of 68Ga-EDTA dynamic PET in the near future.
