BACKGROUND Bark beetles rely on detoxifying enzymes to resist the defensive terpenoids of their host trees. Research on carboxylesterases (CarEs) has focused on their multiple functions in the metabolic detoxification of pesticides and plant allelochemicals, drug resistance, and juvenile hormone and pheromone degradation. RESULT We identified eight new CarE genes in the Chinese white pine beetle ( Dendroctonus armandi ) and carried out bioinformatics analysis on the deduced full-length amino acid sequences. Differential transcript levels of CarE genes were observed between sexes; within these levels, significant differences were found among the different development stages, and between insects fed on the phloem of Pinus armandi and exposed to five stimuli [(−)-α-pinene, (−)-β-pinene, (+)-3-carene, limonene and turpentine] at 8 and 24 h. CONCLUSION Transcription levels of CarE genes suggest some relationship with the detoxification of terpenoids released by host trees. The functions of bark beetle esterase are mainly in hydrolyzing the host chemical defense and degrading odorant molecules during host selection and colonization. © 2018 Society of Chemical Industry

BACKGROUND Thrips Anaphothrips obscurus are one of the cosmopolitan major pests feeding on cereals and other grasses. In order to develop alternatives of chemical insecticides for thrips control, based on fumigant activity screening and evaluation of 22 essential oil (EO) compounds against the thrips, the binary interactions of methyl salicylate and carvacrol (MS-C) with high fumigant toxicity were studied systematically by bioassay and field trials. RESULTS The bioassay results showed that six in 22 EO compounds had high fumigant toxicity against both the second-instar nymphs and adults of A. obscurus , including methyl salicylate, carvacrol, thymol, trans -cinnamaldehyde, diallyl trisulfide, and L-perillaldehyde. Furthermore, the combination of methyl salicylate mixed with carvacrol at a volume ratio of 5:5 exhibited the most significant synergism against A. obscurus , with a poison ratio value of 1.32 and a co-toxicity coefficient of 151.15. The optimal formulation of microemulsion (ME) was composed of 5% methyl salicylate, 5% carvacrol, 46% adjuvant and 44% deionized water. The result of dynamic light scattering and stability showed that MS-C 10% ME was a transparent, single-phase and homogeneous liquid system. Field trials indicated that the ME displayed a significant control efficacy of about 89.17% on thrips in peppers, and 82.59% in broad bean on the seventh day post application with a dosage of 600.0 g A.I hm –2 , respectively. CONCLUSION The binary combination of MS-C possesses strikingly synergistic action against thrips A. obscurus , and the MS-C 10% ME has the potential to be developed as a botanical pesticide product for thrips control. © 2019 Society of Chemical Industry

BACKGROUND Bismerthiazol is a commonly used bactericide against rice bacterial leaf blight in China. Although previous research determined that bismerthiazol is susceptible to photolytic degradation, the photodegradation pathway and degradation products, except for 2-amino-5-mercapto-1,3,4-thiadiazole, have remained unknown. RESULTS The photodegradation of bismerthiazol was investigated after 4 and 8 h of irradiation in a solar simulator. Inhibition of Xanthomonas oryzae pv. oryzae ( Xoo ) was greater with a photolysed solution than with a non-photolysed solution of bismerthiazol. Six photoproducts of bismerthiazol were characterised by liquid chromatography coupled with mass spectrometry, and, based on these products, a photodegradation pathway was inferred. Inhibition of Xoo was significantly greater with bismerthiazol and 2-amino-5-mercapto-1,3,4-thiadiazole than with 5-amino-1,3,4-thiadiazole. In addition, Xoo strain 2-1-1 was bismerthiazol- and 2-amino-5-mercapto-1,3,4-thiadiazole resistant in vivo . CONCLUSION Photodegradation increased the inhibitory activity of bismerthiazol against Xoo . The photodegradation pathway was inferred on the basis of the photoproducts of bismerthiazol. In vitro assays indicated that the sulfhydryl group was crucial for the inhibition of Xoo by bismerthiazol and its photoproducts. Bismerthiazol and 2-amino-5-mercapto-1,3,4-thiadiazole might have a similar mode action in vivo and in vitro . © 2015 Society of Chemical Industry

BACKGROUND Application of a matrix-immobilised target enzyme for screening inhibitors is widely used in drug development, but there are few studies in insecticide discovery. In this paper, an economical and effective immobilised acetylcholinesterase (AChE) column was prepared using the sol–gel embedment method, which was further combined with high-performance liquid chromatography for screening the AChE inhibitors and insecticidal compounds from complex natural products. RESULTS AChE inhibitory constituents magnolol and honokiol were isolated from the ethanol extract of Magnolia officinalis , with IC 50 values of 0.069 and 0.057 mM respectively. In an in vivo bioassay, magnolol and honokiol showed insecticidal activity against Nilaparvata lugens , with LC 50 values of 0.324 and 0.137 mM, which are comparable with that of commonly used insecticide chlorpyrifos (0.233 mM). Moreover, molecular docking was carried out against a homology model of N. lugens AChE. The complexes showed that magnolol and honokiol placed themselves nicely into the active site of the enzyme and exhibited an interaction energy that was in accordance with our activity profile data. CONCLUSION These results demonstrate that magnolol and honokiol have great applied potential to be developed as natural insecticides, and an immobilised AChE column is very useful as a rapid screening tool for target enzymes towards potent inhibitors. © 2014 Society of Chemical Industry

BACKGROUND Maleimides, both natural and synthesised, have good biological activities. In a continuous effort to discover new maleimides with good antifungal activities, the authors have synthesised a series of 3,4-dichloro-, 3-methyl and non-substituted maleimides based on previous studies. The compounds were biologically evaluated against the fungal pathogen Sclerotinia sclorotiorum. RESULTS Of the 63 compounds evaluated, 25 compounds had interesting inhibitory potency with EC 50 < 10 µg mL −1 . N -(3,5-Dichlorophenyl)-3,4-dichloromaleimide (EC 50 = 1.11 µg mL −1 ) and N -octyl-3-methylmaleimide (EC 50 = 1.01 µg mL −1 ) were more potent than the commercial fungicide dicloran (EC 50 = 1.72 µg mL −1 ). The results showed that compounds exhibiting log P values within the range 2.4–3.0 displayed the best results in terms of fungicidal activity, and this seemed, therefore, to be the optimum range for this physicochemical parameter. CONCLUSION The present work demonstrates that some maleimides can be used as potential lead compounds for developing novel antifungal agents against S. sclerotiorum . © 2014 Society of Chemical Industry

BACKGROUND Bark beetles rely on their detoxifying enzymes to resist the defensive terpenoids of host trees. Glutathione S -transferases (GSTs) conjugate xenobiotic compounds with a glutathione moiety (GSH) and often work in tandem with cytochromes P450 or other enzymes that aid in the detoxification, sequestration or excretion of toxic compounds. RESULT We identified nine new GST genes in the Chinese white pine beetle ( Dendroctonus armandi ) and carried out a bioinformatics analysis on the deduced full-length amino acid sequences. These genes belong to four different classes (epsilon, sigma, omega and theta). Differential transcript levels of each class of GST genes were observed between sexes, and, within these levels, significant differences were found among the different adult substages that were fed phloem of Pinus armandi and exposed to six stimuli [(±)- α -pinene, (−)- α -pinene, (−)- β -pinene, (+)-3-carene, (±)-limonene and turpentine] at 8 and 24 h. CONCLUSION The increased transcription levels of GST genes suggested that they have some relationship with the detoxification of terpenoids that are released by host trees. The mediating oxidative stress that is caused by monoterpene might be the main role of the bark beetle GSTs. © 2015 Society of Chemical Industry

BACKGROUND The rice stem borer, Chilo suppressalis Walker (Lepidoptera: Crambidae), is one of the most economically important pests of rice in Asia. Chemical control remains the most efficient primary means for controlling this pest. RESULTS Significant variations among field populations in their resistance to seven insecticides were observed. The populations exhibited LC 50 values that ranged between 0.605 and 108.088 mg AI L −1 for chlorantraniliprole and between 0.046 and 3.919 mg AI L −1 for flubendiamide. The YY14 population collected from Yuyao in Zhejiang Province in 2014 showed a moderate resistance level to the two diamides, i.e. up to 77.6-fold and 42.6-fold for chlorantraniliprole and flubendiamide respectively. Synergism tests and biochemical assays showed no obvious correlations between diamide resistance and three detoxifying enzymes. Sequence comparison of the ryanodine receptor gene between the YY14 resistant population and susceptible population revealed that a glycine to glutamic acid substitution (G4910E) was presented in the YY14 population. CONCLUSION G4910E mutation might be involved in the resistance evolution of C. suppressalis to the diamides. An appropriate insecticide resistance management programme should be established to maintain the effectiveness of the insecticides and to ensure sustainable management. © 2016 Society of Chemical Industry

BACKGROUND Diverse lepidopteran insects cause serious damage to plants, and their larvae possess a crucial epidermal barrier against environmental stimuli. Their ultraviolet (UV) resistance is enhanced by accumulating uric acid granules in the epidermis, suggesting that genes involved in this process may be potential targets for lepidopteran pest management. RESULTS The silkworm pan-genome dataset is a valuable source for studying genomic mutations and phenotype–genotype associations. Hoarfrost translucent ( oh ) is a recessive silkworm mutant with a translucent larval integument. Using comparative genomic analysis, we found that the oh mutant has an 828-bp deletion in the BmBLOC1S6 genome. BmBLOC1S6 encodes a BLOC-1 complex subunit and is conserved during lepidopteran evolution. Knockout of BmBLOC1S6 replicated the oh phenotype. Furthermore, BmBLOC1S6 knockout and oh larvae are more sensitive to UV irradiation compared to the wild-type. These results revealed that BmBLOC1S6 is essential in forming uric acid granules for silkworm epidermal UV resistance. CONCLUSION Our results showed that BmBLOC1S6 is responsible for the oh phenotype in silkworms and is conserved during lepidopteran evolution. This study may help us better clarify uric acid granules formation in the epidermis, explore their function in UV resistance and identify a potential molecular target for lepidopteran pest control. © 2025 Society of Chemical Industry.

BACKGROUND Citrus huanglongbing (HLB) is a devastating disease in citrus, caused by Candidatus Liberibacter asiaticus (CLas), which primarily resides in the phloem where chemicals cannot effectively reach, posing a significant challenge in controlling HLB. To address these challenges, plant essential oils (EOs), widely used as transdermal enhancers and known for their benefits for plant tissues, were investigated for their potential to enhance chemical permeation. RESULTS In this study, seven EOs – eugenol, carvacrol, eucalyptol, geraniol, linalool, cinnamaldehyde, and d -limonene – were evaluated for their potential to enhance chemical penetration into citrus leaves. Preliminary screening with tracer methods revealed that geraniol provided the greatest enhancement of permeation, achieving the deepest and broadest distribution into the spongy tissue of the citrus leaf. The permeation levels of rhodamine B and crystal violet through the cuticular layer were elevated from 0.55 mg L −1 and 0.11 mg L −1 to 20.04 mg L −1 and 7.14 mg L −1 , respectively, by the addition of geraniol. Further validation through high-performance liquid chromatography–mass spectrometry (HPLC–MS) confirmed that the penetration of fludioxonil and tetracycline into citrus leaf tissues was enhanced by 15.07-fold and 20.43-fold, respectively. In planta experiments confirmed that adding geraniol to tetracycline and oxytetracycline via foliar application eliminated CLas bacteria in HLB-affected citrus more efficiently than controls. CONCLUSION Our study underscores the potential of geraniol as an effective EO-based permeation enhancer for combating HLB. By significantly improving the delivery and efficacy of bactericidal treatments, geraniol offers a promising strategy for managing this catastrophic citrus disease, potentially transforming the approach to controlling HLB. © 2024 Society of Chemical Industry.

BACKGROUND In recent years, pyridylpyrazole derivatives, such as pyridylpyrazole-containing anthranilic diamide have attracted much attention by virtue of their useful insecticidal properties and unique action mode. Moreover, some pyridylpyrazole-containing compounds have also been found to possess significant fungicidal activities. With the aim of discovering new bioactive agrochemicals for crop protection, a series of poly-heterocyclic compounds containing pyridylpyrazole and aziridine, or β-lactam, or thiazolinone moieties were synthesized. RESULTS A series of pyridylpyrazole-containing poly-heterocyclic compounds were obtained, and confirmed through IR, 1 H NMR, 13 C NMR, HRMS and elemental analysis. The crystalline structure of 4-(3-bromo-1-(3-chloropyridin-2-yl)-1 H -pyrazol-5-yl)-3-chloro-1-mesitylazetidin-2-one (compound 13f ) was determined to further illustrate a trans- configuration of the β-lactam motif. In addition, bioassays showed that most of these new compounds exhibited modest insecticidal activity towards Mythimna separate Walker at 200 µg mL −1 . Some of the compounds displayed excellent fungicidal activity towards some plant fungi, including Cercospora arachidicola ( 13j : EC 50 = 14.5 µg mL −1 ), Physalospora piricola ( 12d and 13d : EC 50 = 10.5 and 9.70 µg mL −1 ), Alternaria solani Sorauer ( 13j : EC 50 = 7.29 µg mL −1 ) , Puccinia sorghi Schw. ( 13d : control efficacy 99.0 ± 2.1% at 200 µg mL −1 ) and Erysiphe graminis ( 14d : control efficacy 95.0 ± 1.4% at 200 µg mL −1 ). CONCLUSION Compounds 12b – 12e , 13a , 13d , 13f , 13j , 13 k and 14d could be considered potential fungicidal lead compounds to do further structural optimization. The structure–activity relationship analysis in this study brings some new understanding to the biological activities of N -pyridylpyrazole-containing compounds, and provides important information for the research and development of novel agricultural fungicides with poly-heterocyclic structures. © 2017 Society of Chemical Industry

BACKGROUND Ralstonia solanacearum , a notorious and refractory bacterial plant pathogen, threatens multiple vegetable crops and causes significant economic loss in agriculture. Long-term use of traditional medicines not only increases the problem of drug resistance, but also causes great environmental pollution. Therefore, there is an urgent need to develop new agents with high efficacy and low toxicity. RESULTS In this study, we have synthesized and characterized graphitic carbon nitride incorporated copper oxide composite (g-C 3 N 4 @CuO), which showed higher antimicrobial effect than graphitic carbon nitride nanosheets (g-C 3 N 4 nanosheets) and copper oxide nanoparticles (CuONPs). Ralstonia solanacearum exposed to g-C 3 N 4 @CuO exhibited higher levels of oxygen toxicity, cell membrane damage, DNA damage, motility disruption and even cell death compared to g-C 3 N 4 nanosheets and CuONPs. In addition, g-C 3 N 4 @CuO was more effective in the control of tobacco bacterial wilt than g-C 3 N 4 nanosheets and CuONPs. CONCLUSION Thus, this study provides a new perspective on g-C 3 N 4 @CuO control of bacterial diseases in crops, and the mechanism is related to the destruction of cell membrane damage and motility disruption. © 2024 Society of Chemical Industry.

BACKGROUND The important wood-boring pest Batocera horsfieldi has evolved a sensitive olfactory system to locate host plants. Odorant-binding proteins (OBPs) are thought to play key roles in olfactory recognition. Therefore, exploring the physiological function of OBPs could facilitate a better understanding of insect chemical communications. RESULTS In this research, 36 BhorOBPs genes were identified via transcriptome sequencing of adults' antennae from B. horsfieldi , and most BhorOBPs were predominantly expressed in chemosensory body parts. Through fluorescence competitive binding and fluorescence quenching assays, the antenna-specific BhorOBP28 was investigated and displayed strong binding affinities forming stable complexes with five volatiles, including (+)-α-Pinene, (+)-Limonene, β-Pinene, (−)-Limonene, and (+)-Longifolene, which could also elicit conformation changes when they were interacting with BhorOBP28. Batocera horsfieldi females exhibited a preference for (−)-Limonene, and a repellent response to (+)-Longifolene. Feeding dsOBP19 produced by a bacteria-expressed system with a newly constructed vector could lead to the knockdown of BhorOBP28 , and could further impair B. horsfieldi attraction to (−)-Limonene and repellent activity of (+)-Longifolene. The analysis of site-directed mutagenesis revealed that Leu7, Leu72, and Phe121 play a vital role in selectively binding properties of BhorOBP28. CONCLUSION By modeling the molecular mechanism of olfactory recognition, these results demonstrate that BhorOBP28 is involved in the chemoreception of B. horsfieldi . The bacterial-expressed dsRNA delivery system gains new insights into potential population management strategies. Through the olfactory process concluded that discovering novel behavioral regulation and environmentally friendly control options for B. horsfieldi in the future. © 2024 Society of Chemical Industry.
