Parkinson's disease (PD) is characterized by the progressive loss of dopaminergic neurons in the midbrain, and the stem cell transplantation method provides a promising strategy for the treatment. In these studies, tracking the biological behaviors of the transplanted cells in vivo is essential for a basic understanding of stem cell function and evaluation of clinical effectiveness. In the present study, we developed a novel ultrasmall superparamagnetic iron oxide nanoparticles coating with the polyacrylic acid (PAA) and methoxypolyethylene glycol amine (PEG) by thermal decomposition method and a two-step modification. The USPIO-PAA/PEG NPs have a uniform diameter of 10.07 ± 0.55 nm and proper absorption peak of the corresponding ligands, as showed by TEM and FTIR. MTT showed that the survival of cells incubated with USPIO-PAA/PEG NPs remained above 96%. The synthesized USPIO-PAA/PEG had a good relaxation rate of 84.65 s −1 Mm −1 , indicating that they could be efficiently uptake and traced by MRI. Furthermore, the primary human adipose-derived stem cells (HADSCs) were characterized, labeled with USPIO-PAA/PEG and transplanted into the striatum of 6-hydroxydopamine (6-OHDA)-induced PD rat models. The labeled cells could be traced by MRI for up to 3 weeks after the transplantation surgery; moreover, transplantation with the labeled HADSCs significantly attenuated apomorphine-induced rotations in PD models and increased the number of the dopaminergic neurons in the substania nigra . Overall, the development of USPIO-PAA/PEG NPs provides a promising tool for in vivo tracing technique of cell therapy and identifies a novel strategy to track stem cells with therapeutic potential in PD.

Bone tissue plays an important role in supporting and protecting the structure and function of the human body. Bone defects are a common source of injury and there are many reconstruction challenges in clinical practice. However, 3D bioprinting of scaffolds provides a promising solution. Hydrogels have emerged as biomaterials with good biocompatibility and are now widely used as cell-loaded materials for bioprinting. This study involved three steps: First, sodium alginate (SA), gelatin (Gel), and nano-hydroxyapatite (na-HA) were mixed into a hydrogel and its rheological properties assessed to identify the optimum slurry for printing. Second, SA/Gel/na-HA bioscaffolds were printed using 3D bioprinting technology and their physical properties characterized for surface morphology, swelling, and mechanical properties. Finally, human periodontal ligament stem cells (hPDLSCs) were mixed with SA/Gel/na-HA printing slurry to create a “bioink” to prepare SA/Gel/na-HA/ hPDLSCs cell bioscaffolds. These were tested for biocompatibility and osteogenic differentiation performance using live/dead cell staining, cell adhesion, cell proliferation, and alkaline phosphatase activity. The SA/Gel/na-HA hydrogel exhibited shear-thinning behavior. The equilibrium swelling of the bioscaffold was 125.9%, the compression stress was 0.671 MPa, and the compression elastic modulus was 8.27 MPa. The SA/Gel/na-HA/hPDLSCs cell bioscaffolds caused effective stimulation of cell survival, proliferation, and osteoblast differentiation. Therefore, the SA/Gel/na-HA/hPDLSCs cell bioscaffolds displayed potential as a material for bone defect reconstruction.

Human mesenchymal stem cells (hMSCs) are promising in the treatment of pulmonary fibrosis (PF). However, the behavior of hMSCs after transplantation, including dynamic translocation, location and survival, impeding the clinical application of hMSCs in PF is still ambiguous. Herein, we report an effective dual-labeling strategy combining endogenous bioluminescence imaging (BLI) and exogenous near-infrared-persistent luminescence (NIR-PL) imaging for in situ visualization of the transplanted stem cells. The long persistent luminescence nanoparticles (LPLNPs), Zn1.1Ga1.8Ge0.1O4:Cr3+,Eu3+, were developed to track the dynamic translocation, position and distribution of the transplanted hMSCs, taking advantage of their long-lasting NIR-PL imaging ability and minimal autofluorescence background interference. Moreover, in virtue of their ability to express red-emitting firefly luciferase (RfLuc), the living stem cells after transplantation could be discriminated from the dead cells by BLI. This facile pattern contributes to the in situ monitoring of stem cells regarding their spontaneous behavior in vivo and therefore deepening our knowledge in the role played by the transplanted hMSCs in PF therapy.

Micro/nanobubbles for use as ultrasound contrast agents have been fabricated with different shell materials. When various biomedical nanoparticles have been embedded in the shells of bubbles, the composite structures have shown promising applications in multi-modal imaging, drug/gene delivery, and biomedical sensing. In this study, we developed a new gas-liquid interface self-assembly method to prepare magnetic nanobubbles embedded with superparamagnetic iron oxide nanoparticles (SPIONs). The diameter of the generated assembled nanobubbles was 227.40 ± 87.21 nm with a good polydispersity index (PDI) of 0.29. Under the condition of 150 compression cycles, the nanobubble concentration could reach about 6.12 × 10 9 /mL. Transmission electron microscopy (TEM) and scanning electronic microscopy (SEM) demonstrated that the assembled nanobubbles had a hollow gas core with SPIONs adsorbed on the surface. Ultrasound (US) imaging and magnetic resonance imaging (MRI) experiments indicated that the assembled magnetic nanobubbles exhibited good US and MR contrast capabilities. Moreover, the assembled magnetic nanobubbles were used to label neural stem cells under ultrasound exposure. After 40 s US exposure, the magnetic nanobubbles could be delivered into cells with 2.80 pg Fe per cell, which could be observed in the intracellular endosome by TEM. Compared with common incubation methods, the ultrasound exposure method did not introduce the potential cytotoxicity of transfection reagents and the efficiency was about twice as high as the efficiency of incubation. Therefore, the assembled magnetic nanobubbles prepared through the pressure-driven gas-liquid interface assembly approach could be a potential US/MRI dual model imaging nanocarrier for regenerative applications.

Cancer stem cells (CSCs) are considered as the origin and driving cells of cancer, and play a key role in the progress of cancer. Studies have shown that capsaicin exerted inhibitory effect on prostate cancer cells, however, the effects of capsaicin on prostate CSCs remain undefined. In the present study, we showed that capsaicin could downregulate prostate CSCs markers and inhibit the growth of PC-3 and DU145 prostate cancer stem cells. Further, we found capsaicin suppressed the expression of Wnt-2, p-GSK3β and β-catenin, along with downregulation of Wnt/β-catenin pathway target genes c-myc and cyclinD1. Using LiCl, a activator of Wnt/β-catenin pathway, we found activation of Wnt/β-catenin pathway could ameliorate the downregulation of prostate CSCs markers and the growth inhibition induced by capsaicin in prostate cancer stem cells. Those data suggested that the inhibition effect of capsaicin on prostate cancer stem cells and the anti-cancer effect of capsaicin on prostate cancer stem cell may be mediated by Wnt/β-catenin pathway. Findings from this study reveals for the first time the potential role and mechanisms of capsaicin on prostate cancer stem cells.

Adult stem cells, a class of cells that possess self-renewal and differentiation capabilities, modulate tissue regeneration, repair, and homeostasis maintenance. These cells undergo functional degeneration during aging, resulting in decreased tissue regeneration ability and increased disease incidence. Thus, it is essential to provide effective therapeutic solutions to preventing the aging-related functional decline of stem cells. Quercetin (Que) is a popular natural polyphenolic flavonoid found in various plant species. It exhibits many beneficial effects against aging and aging-related diseases; however, its efficacy against adult stem cell aging remains largely unclear.Drosophilapossesses a mammalian-like intestinal system with a well-studied intestinal stem cell (ISC) lineage, making it an attractive model for adult stem cell research. Here, we show that Que supplementation could effectively prevent the hyperproliferation of ISCs, maintain intestinal homeostasis, and prolong the lifespan in agedDrosophila. In addition, we found that Que could accelerate recovery of the damaged gut and improve the tolerance ofDrosophilato stressful stimuli. Furthermore, results demonstrated that Que prevents the age-associated functional decline of ISCs via scavenging reactive oxygen species (ROS) and inhibiting the insulin signaling pathway. Overall, our findings suggest that Que plays a significant role in delaying adult stem cell aging. Keywords:intestinal stem cell;aging;quercetin;insulin signaling;ROS;Drosophila

Background Sarcandra glabra (Caoshanhu) is a traditional Chinese herbal medicine used for treating various oxidative-stressed diseases. The present work evaluated its protective effect on mesenchymal stem cells (MSCs) from oxidative stress and then discussed possible mechanisms underlying this observation. Methods Ethanolic extract of S. glabra (ESG) was investigated by chemical methods for its content of total phenolics, rosmarinic acid, and astilbin. ESG, along with rosmarinic acid and astilbin, was investigated for the effect on the viability of Fenton-treated MSCs using 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyl (MTT) assay. The observed cell protective effect was further explored by mechanistic chemistry using various antioxidant assays, including DNA protection, •OH-scavenging, •O 2 − -scavenging, FRAP (ferric ion reducing antioxidant power), ABTS + •-scavenging, DPPH•-scavenging, and Fe 2+ -chelating assays. Results Analysis of ESG revealed a content of 46.31 ± 0.56 mg quercetin/g total phenolics, 0.78 ± 0.01 % rosmarinic acid, and 3.37 ± 0.01 % astilbin. Results from the MTT assay revealed that three compounds (rosmarinic acid>astilbin>ESG) could effectively increase the survival of Fenton-treated MSCs. Similarly, in •O 2 − -scavenging, DPPH•-scavenging, and Fe 2+ -chelating assays, rosmarinic acid exhibited more activity than astilbin; while in FRAP, ABTS + •-scavenging assays, astilbin was stronger than rosmarinic acid. Conclusion S. glabra can prevent MSCs from •OH-induced oxidative stress. Such protective effect can be attributed to its antioxidant ability and the presence of two kinds of phytophenols, i.e. caffeoyl derivatives and flavonoids. As the respective representatives of caffeoyl derivatives and flavonoids, rosmarinic acid and astilbin may exert the antioxidant action via direct ROS-scavenging and indirect ROS-scavenging (i.e. Fe 2+ -chelating). The direct ROS-scavenging ability involves hydrogen atom transfer (HAT) and/or electron transfer (ET) pathway. Astilbin engages the latter pathway more, which can be attributed to the larger planar conjugation in A/C fused rings. Rosmarinic acid, on the other hand, shows more HAT and Fe 2+ -chelating potential, which may be due to rosmarinic acid bearing one more catechol moiety whereas astilbin has steric-hindrance from 3-α-L-rhamnose and an H-bonding between 4,5 sites. The antioxidant features of rosmarinic acid can be generalized to other caffeoyl derivatives, while that of astilbin cannot be generalized to other flavonoids because of the difference in chemical structures.

Realization of clinical potential of human pluripotent stem cells (hPSCs) in bone regenerative medicine requires development of simple and safe biomaterials for expansion of hPSCs followed by directing their lineage commitment to osteoblasts. In the present study, a chemically defined peptide-decorated polycaprolactone (PCL) nanofibrous microenvironment was prepared through electrospinning technology and subsequent conjugation with vitronectin peptide to promote the culture and osteogenic potential of hPSCs in vitro. The results indicated that hPSCs successfully proliferated and maintained their pluripotency on the biointerface of peptide-conjugated nanofibers without Matrigel under defined conditions. Moreover, the prepared niche exhibited an appealing ability in promoting directed differentiation of hPSCs to osteoblastic phenotype without embryoid body formation step, determined from the cell morphological alteration, alkaline phosphate activity, and osteogenesis-related gene expression, as well as protein production. Such well-defined, xeno-free, and safe nanofiber scaffolds that allow the survival and facilitate osteo-differentiation of hPSCs provide a novel platform for hPSCs differentiation via cell-nanofiber interplay, and possess great value in accelerating the translational perspectives of hPSCs in bone tissue engineering.

As the main bioactive component of Chinese herbal medicine Danshen, salvianolic acid B (Sal B, or lithospermic acid B) was observed to maintain the viability of mesenchymal stem cells (MSCs) treated by Fenton's reagent in the study. Interestingly, at a higher concentration, Sal B could even increase the viability rate to 175.1%. In mechanistic analysis experiments, Sal B was found to resist DNA destruction by •OH radical, scavenge various radicals in vitro , reduce Cu 2+ → Cu + , chelate Fe 2+ to yield an absorption maximum at 710 nm. Based on these results, we concluded that, (1) Sal B can not only protect MSCs against •OH-induced damages, but also promote their proliferation. This extraordinary capacity makes Sal B an ideal candidate in MSCs transplantation especially when MSCs are polluted by iron-overload or other oxidative stress factors and, can partly be responsible for the versatile properties of Sal B in pharmacology. The possible mechanisms of its protective effect are hypothesized to include Fe 2+ chelating, and direct radical scavenging which is involved in electron transfer (ET) or hydrogen atom transfer (HAT) from the catechol moieties. Its proliferation-promoting effect is presumed to be from its ester group, carboxylic group, or benzofuran ring.

Regeneration of large bone defects is a common clinical problem. Recent studies have shown that mesenchymal stem cells (MSCs) have emerged as a promising alternative to traditional surgical techniques. However, it is still a key question how to enhance the osteogenic potential of MSCs for possible clinical trials. The aim of the present study was to investigate the effect of adenovirus-mediated erythropoietin (Ad-EPO) transfer on BMSCs, we performed extensive in vitro/in vivo assays in this study. Flow cytometry analysis and the result of MTT showed that EPO could promote BMSCs proliferation. QPCR data demonstrated that EPO increased expressions of Runx2, Sp7, and Col1 in osteoblast at various time points and also increased alkaline phosphatase activity and the calcium deposition. These results indicate that EPO can increase the differentiation of osteoblast. Importantly, in vivo assays clearly demonstrate that EPO can efficiently induce new bone formation in the bone defect model. Our results strongly suggest that EPO can affect osteoblast differentiation and play important roles in bone regeneration leading to an increase in bone formation.

Surface plasmon resonance imaging (SPRi) has emerged as a versatile biosensor to detect a wide range of biomolecular interactions with divergent potential applications. However, the use of this advanced-level technology for stem cell lysate study is still not much explored. Cell lysates are significant biological analytes used for disease diagnostics and proteomic studies, but their complex nature limits their use as an analyte for SPRi biosensors. Here, we review the problems associated with the use of SPRi for stem cell lysate study and examine the role of surface chemistry, running buffer, and blocking solution in order to minimize nonspecific adsorption (NSA). We detect the expression of Oct4, Sox2, Nanog , Rex1 , and Lin28 biomarkers present in mouse embryonic stem cell (mESC) lysate against their corresponding antibodies immobilized on the sensor surface with reduced NSA. The current study shows that the conjunction of SPRi and microarray can be used as a label-free, high-throughput, and rapid technique for detection of biomarkers and their relative abundance in stem cell lysate study.

Polydatin, a glucoside of resveratrol, has been reported to possess potent antioxidative effects. In the present study, we aimed to investigate the effects of polydatin in bone marrow-derived mesenchymal stem cells (BMSCs) death caused by hydrogen peroxide (H2O2), imitating the microenvironment surrounding transplanted cells in the injured spinal cord in vitro. In our study, MTT results showed that polydatin effectively prevented the decrease of cell viability caused by H2O2. Hochest 33258, Annexin V-PI, and Western blot assay showed H2O2-induced apoptosis in BMSCs, which was attenuated by polydatin. Further studies indicated that polydatin significantly protects BMSCs against apoptosis due to its antioxidative effects and the regulation of Nrf 2/ARE pathway. Taken together, our results indicate that polydatin could be used in combination with BMSCs for the treatment of spinal cord injury by improving the cell survival and oxidative stress microenvironments.
