Staphylococcus aureus (S. aureus) is one of the foodborne pathogens that can cause infectious diseases and food poisoning. Herein, colorimetric and fluorescent dual-mode nanoprobes were developed for ultrasensitive detection of S. aureus to immediately respond to public health emergencies, reduce false positives, and improve measurement accuracy and persuasiveness. The nanoprobe consists of aptamer-labeled magnetic nanoparticles (apt-MNPs) as the capture signal probe and horseradish peroxidase and complementary DNA-functionalized upconversion nanoparticles (HRP-UCNPs-cDNA) as the chromogenic signal probe. In the absence of S. aureus, the probe forms an immune complex through base complementation with an observable signal. When S. aureus is introduced to this system, it preferentially binds to the apt-MNPs, releasing HRP-UCNPs-cDNA from the apt-MNPs and restoring the chromogenic probe signal. Under optimum conditions, an ultrasensitive assay of S. aureus was obtained, with limits of detection of 22 CFU mL–1 for fluorescence and 20 CFU mL–1 for colorimetry in a linear range of 56–5.6 × 106 CFU mL–1. Additionally, the standard plate counting method confirmed the reliability and accuracy of the established nanoprobe with an insignificant difference. Hence, the developed dual-mode platform has extensive application prospects for speedy and specific determination of S. aureus in meat.

ABSTRACT Diseases caused by harmful microorganisms pose a serious threat to human health. Safe and environmentally friendly disinfectants are, therefore, essential in preventing and controlling such pathogens. This study aimed to investigate the antimicrobial activity and mechanism of a novel hydrogen peroxide and silver (H 2 O 2 -Ag + ) complex (HSC) in combatting Staphylococcus aureus ATCC 29213, Escherichia coli O157:H7 NCTC 12900, and Salmonella Typhimurium SL 1344. The MICs and MBCs against S. aureus were found to be 0.014% H 2 O 2 –3.125 mg/L Ag + , and for both E. coli O157:H7 and Salmonella Typhimurium they were 0.028% H 2 O 2 –6.25 mg/L Ag + . Results of the time-kill trial suggest that HSC could inhibit the growth of the tested bacteria, because 99.9% of viable cells were killed following treatment at 1 MIC for 3 h. The mechanism of antibacterial action of HSC was found to include the disruption of the bacterial cell membrane, followed by reduction of intracellular ATP concentration and inhibition of the activity of antioxidases, superoxide dismutase, and catalase. The enhanced bactericidal effect of hydrogen peroxide combined with silver indicates a potential for its application in environmental disinfection, particularly in the food industry.

Clinical wound management remains a major challenge due to massive bleeding, bacterial infection, and difficult wound healing after tissue trauma. To simultaneously address these issues, composite polymer sponges for accelerating drug-resistant bacterial infected wound healing are fabricated by facilely mixing sodium polyacrylate (PAAS), double quaternary ammonium salts-conjugated chitosan (QAS-CS), and collagen (COL) in aqueous solution, followed by lyophilization. Composite sponges (PAAS/QAS-CS/COL, PQC) show highly porous microstructures (porosity ≈90%) with moderate compress modulus (≈0.3 MPa), tensile strength (0.004 MPa), and high swelling ratio (≈3500%). Importantly, PQC sponge demonstrates superior hemostasis ability over commercially available CS sponge by inducing rapid hemagglutination, and exhibits significantly better antibacterial activity against both methicillin-resistant Staphylococcus aureus (MRSA) and Escherichia coli by destroying cell membrane and causing the leakage of bioactive components such as potassium ion and β -galactosidase from treated bacterial. Furthermore, PQC sponge can efficiently promote cell proliferation. Significantly, the sponge greatly expedites the regeneration of MRSA-infected full-thickness skin wound in rabbit by successfully eradicating bacterial infection, and reducing inflammation. PQC sponge also improves both early angiogenesis and blood vessel maturation at the wound site. Overall, this multifunctional sponge is a promising wound dressing for clinical use and holds great potential for rapid clinical translation.

In this study, we investigated the antibacterial activity of quinic acid (QA) and elucidated its membrane action mechanism against Staphylococcus aureus . In vitro antibacterial assay showed that QA effectively inhibited the growth of test bacteria. In model food systems of cabbage suspension and barley soup, QA possessed a great antibacterial capacity. The membrane hyperpolarization and a decrease of membrane fluidity demonstrated a significant effect of QA on damaging the normal functions of cell membrane, which was further validated by the observation of the transmission electron microscope. The interaction of QA with Phe residues in the membrane protein was revealed by fluorescence quenching. Overall, the present study illustrates QA as a potent antibacterial agent and its inhibition action on S . aureus by damaging the cell membrane. Practical applications In model food systems of cabbage suspension and barley soup, QA possessed a great antibacterial capacity to control the total counts of viable bacteria. This study provided the theoretical foundations for the application of QA in foods to prevent the growth of foodborne pathogens.

Background. Perioperative topical tranexamic acid as antifibrinolytic agent is often used for total joint replacement to reduce bleeding currently. Staphylococcus aureus was the most common isolates from perioperative infection of prosthetic joint. The influence of topical application with tranexamic acid on the incidence of acute prosthetic joint infection of Staphylococcus aureus has not been clarified. Methods. Mouse model of Staphylococcus aureus knee prosthesis infection was constructed. Tranexamic acid was intra-articular injected during the perioperative period. CFU counting from tissue and implant sample was evaluated 3 days and 7 days after inoculating of Staphylococcus aureus. Bacterial growth curve, biofilm formation, aggregation, and plasmin inhibition of Staphylococcus aureus were tested with tranexamic acid added to the synovial culture medium. Results. There were no significant differences of CFU counting from tissue and implant samples in knee prosthesis infection after a single local injection of tranexamic acid at the postoperative 3 or 7 days. The amount of bacterial colonization on the surface of implant increased after 3 days’ continuous local injection of tranexamic acid. Tranexamic acid has no effect on bacterial growth at the concentration (10 mg/ml) of clinical application, but it can inhibit bacterial aggregation and mildly inhibit biofilm formation. Plasmin can significantly inhibit biofilm formation which can be revised by adding tranexamic acid. Conclusion. Although continuous local injection of tranexamic acid can promote the biofilm formation of Staphylococcus aureus on the surface of articular implant, it has clinical safety for using one single local injection of tranexamic acid during the perioperative period.

Sapogenin is the main block of Camellia oleifera saponin, which was purified and structurally modified by the C28 acylation reaction to synthesize 19 new derivatives. The growth and biofilm inhibition of Staphylococcus aureus and Escherichia coli was measured to evaluate their antibacterial effects. A three-dimensional quantitative structure–activity relationship (3D-QSAR) assay indicated that the antibacterial activities were significantly enhanced after sapogenin was modified with an aromatic ring or heterocyclic ring and electron-withdrawing substituents at the meta or para position. Among them, the derivative of sapogenin with a 2-mercapto-4-methyl-5-thiazolyl acetyl group obviously destroyed bacterial biofilm and made bacteria lysis. 3D-QSAR provides practical information for the structural design of sapogenin derivatives with strong antibacterial activity, and the C. oleifera sapogenin derivative 28-O-(2-mercapto-4-methyl-5-thiazolyl)-3β,16α,21β,22α-O-tetrahydroxy-oleantel-2-ene-23-aldehyde (S-16) is an effective candidate as an antibacterial agent for the prevention of bacterial resistance against antibiotics.

Phytochemicals are promising antibacterials for the development of novel antibiofilm drugs, but their antibiofilm activity in physiologically relevant model systems is poorly characterized. As the host microenvironment can interfere with the activity of the phytochemicals, mimicking the complex environment found in biofilm associated infections is essential to predict the clinical potential of novel phytochemical-based antimicrobials. In the present study, we examined the antibiofilm activity of borneol, citral, and combinations of both as well as their Pickering emulsions against Staphylococcus aureus and Pseudomonas aeruginosa in an in vivo-like synthetic cystic fibrosis medium (SCFM2) model, an in vitro wound model (consisting of an artificial dermis and blood components at physiological levels), and an in vivo Galleria mellonella model. The Pickering emulsions demonstrated an enhanced biofilm inhibitory activity compared to both citral and the borneol/citral combination, reducing the minimum biofilm inhibitory concentration (MBIC) values up to 2 to 4 times against P. aeruginosa PAO1 and 2 to 8 times against S. aureus P8-AE1 in SCMF2. In addition, citral, the combination borneol/citral, and their Pickering emulsions can completely eliminate the established biofilm of S. aureus P8-AE1. The effectiveness of Pickering emulsions was also demonstrated in the wound model with a reduction of up to 4.8 log units in biofilm formation by S. aureus Mu50. Furthermore, citral and Pickering emulsions exhibited a significant degree of protection against S. aureus infection in the G. mellonella model. The present findings reveal the potential of citral- or borneol/citral-based Pickering emulsions as a type of alternative antibiofilm candidate to control pathogenicity in chronic infection.IMPORTANCE There is clearly an urgent need for novel formulations with antimicrobial and antibiofilm activity, but while there are plenty of studies investigating them using simple in vitro systems, there is a lack of studies in which (combinations of) phytochemicals are evaluated in relevant models that closely resemble the in vivo situation. Here, we examined the antibiofilm activity of borneol, citral, and their combination as well as Pickering emulsions (stabilized by solid particles) of these compounds. Activity was tested against Staphylococcus aureus and Pseudomonas aeruginosa in in vitro models mimicking cystic fibrosis sputum and wounds as well as in an in vivo Galleria mellonella model. The Pickering emulsions showed drastically increased antibiofilm activity compared to that of the compounds as such in both in vitro models and protected G. mellonella larvae from S. aureus-induced killing. Our data show that Pickering emulsions from phytochemicals are potentially useful for treating specific biofilm-related chronic infections.

In the treatment of foodborne antibiotic-resistant bacterial infections, the use of bactericidal nanozymes is one of the most promising non-antibiotic therapeutic strategies. However, this approach faces problems such as the inability to monitor the bactericidal effect in real time. In this work, novel ZnGa 2 O 4 :Cr/Mn persistent luminescent nanoparticles (PLNPs) with dual functionality of afterglow and catalysis were directly synthesized. The ZnGa 2 O 4 :Cr/Mn PLNPs not only possess inherent near-infrared persistent luminescence but also exhibit peroxidase- and oxidase-like nanozyme activities, making them applicable to autofluorescence-free afterglow imaging and drug-resistant bacteria eradication. The nanozyme activity of PLNPs was optimized without compromising their afterglow properties, and the bactericidal efficacy against Staphylococcus aureus and MRSA under acidic conditions was validated. These results provide a foundation for potential infection therapies. This dual-functional platform offers a new strategy for afterglow imaging-guided treatment of drug-resistant foodborne infections, with promising applications in food safety control, particularly for real-time tracking and elimination of Staphylococcus aureus contamination.

IntroductionClerodendrum cyrtophyllum Turcz., a traditional Chinese medicinal plant, exhibits broad bioactivity. Nevertheless, the flavonoid constituents conferring anti-Staphylococcus aureus (S. aureus) activity and their mechanistic basis remain to be elucidated.MethodsTotal flavonoid extraction from Clerodendrum cyrtophyllum Turcz. (TFC) was first evaluated by single-factor experiments to screen key variables and subsequently optimized by a Box–Behnken design (BBD) within response surface methodology (RSM). followed by purification with D101 macroporous resin. Liquid chromatography–tandem mass spectrometry (LC–MS/MS) characterized the chemical profile. Antibacterial activity against S. aureus was assessed via inhibition zones, minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and growth curves. Mechanisms were explored by evaluating cell surface hydrophobicity (CSH), extracellular alkaline phosphatase (AKP), leakage of nucleic acids/proteins, membrane potential, ATP levels, reactive oxygen species (ROS) content, and morphological alterations.ResultsThe optimal extraction conditions for TFC were determined as 70% ethanol as the solvent, an extraction time of 102 min, a temperature of 64 °C, and a solid–liquid ratio of 1:25 (g: mL), resulting in a yield of 6.48% ± 0.58. After purification, the purity of TFC was increased to 76%, and preliminary analysis identified 85 flavonoid constituents. TFC exhibited inhibition zone diameters of 13 ± 0.45 mm, 16 ± 0.38 mm, and 20 ± 0.45 mm at concentrations of 50, 100, and 150 mg/mL, respectively, with MIC and MBC values of 0.781 mg/mL and 1.562 mg/mL. Notably, TFC effectively inhibited the growth of S. aureus within 24 h. Mechanistic investigations revealed that TFC treatment increased S. aureus CSH and decreased the membrane potential, leading to the leakage of intracellular nucleic acids and proteins, indicating damage to S. aureus cells, which was further confirmed by SEM. Furthermore, TFC induced significant intracellular ATP depletion, decreased intracellular nucleic acids and protein contents, and triggered excessive ROS production.ConclusionThis study optimized the extraction conditions of TFC and successfully obtained TFC rich in multiple flavonoid constituents. TFC exerts potent inhibitory effects on S. aureus by disrupting the integrity of cell walls and membranes, depleting intracellular energy reserves, decreasing the contents of intracellular nucleic acids and proteins, and inducing intracellular oxidative stress.

The emergence of drug-resistant bacterial infection and persistent biofilm colonization pose a rigorous challenge to effective wound healing and regeneration, necessitating the innovative therapeutic strategies to combat these pressing clinical crises. Herein, nortriptyline, a novel FDA-approved tricyclic antidepressant was uncovered to effectively potentiate bactericidal activities of β- lactam antibiotics against methicillin-resistant Staphylococcus aureus (MRSA). Mechanistically, nortriptyline functions by disrupting the microbial iron homeostasis and potentiation of Fenton chemistry-mediated oxidative stress, concomitant with metabolic reprogramming via TCA cycle dysregulation and membrane destabilization. To enhance combination therapy-mediated therapeutic potential in wound management, the dual-loaded self-healing hydrogel OHA-PLL@AN was engineered to exhibit excellent biocompatibility and antibacterial potentials through molecular cross-linking of oxidized hyaluronic acid (OHA) and ε-polylysine (PPL). The therapeutic efficacy of OHA-PLL@AN was further validated in a murine model with MRSA-infected cutaneous wounds. OHA-PLL@AN therapy significantly attenuated the inflammatory response, concurrently promoting angiogenesis and accelerating the cutaneous wounds healing. Collectively, these findings underscore the dual drug-loaded self-healing hydrogel OHA-PLL@AN with anti-infection and anti-inflammatory properties as a novel therapeutic strategy for drug-resistant bacterial infected wounds therapy.

Derazantinib (DZB), a pan-fibroblast growth factor receptor (FGFR) inhibitor, exhibits potent activity against FGFR1–3 kinases and has been clinically approved for antitumor therapy. However, its antibacterial properties remain unknown. Here, we demonstrated that DZB displays broad-spectrum activity against Staphylococcus aureus (S. aureus), with minimum inhibitory concentrations (MICs) ranging from 6.25 to 25 μM. DZB exhibited more rapid and stronger bactericidal activity against planktonic cells of both MSSA and MRSA compared to vancomycin. DZB at 6.25 μM robustly inhibited biofilm formation and even eradicated mature biofilms. Global proteomic profiling revealed that DZB’s antibacterial mechanism might involve disruption of microbial glycolysis/gluconeogenesis pathways. Furthermore, in vitro selection of DZB-induced resistant S. aureus resulted in a 2-fold increase in MIC, and whole-genome sequencing of this derivative isolate identified amino acid mutations in membrane-associated proteins. DZB was found to compromise bacterial membrane integrity, as evidenced by increased membrane permeability, and the membrane damage was also confirmed by scanning electron microscopy (SEM). The antibacterial activity of DZB was neutralized by the addition of exogenous phosphatidylglycerol and cardiolipin. Biolayer interferometry assays demonstrated a strong interaction between DZB and cardiolipin, suggesting membrane phospholipid targeting as a key mechanism. Lastly, DZB displayed a robust inhibitory effect against intracellular S. aureus SA113 and showed excellent in vivo anti-MRSA infection in both Galleria mellonella larvae and murine infection models. In summary, our findings established DZB as a promising anti-S. aureus agent with dual antibacterial and antibiofilm activities by disrupting the cell membrane through targeting membrane phospholipids.

Foodborne diseases caused by Staphylococcus aureus contamination on meat and meat products has gained increasing attention in recent years, while the pathogenicity of S. aureus is mainly attributed to its virulence factors production, which is primarily regulated by quorum sensing (QS) system. Herein, we aimed to uncover the inhibitory effects and mechanisms of citral (CIT) on virulence factors production by S. aureus , and further explore its potential application in pork preservation. Susceptibility test confirmed the antibacterial properties of CIT against S. aureus , the minimal inhibitory concentration (MIC) was 0.25 mg/mL. Treatment with sub-MICs of CIT reduced the hemolytic activity by inhibiting the production of α-hemolysin, and staphylococcal enterotoxins (SEs) production was significantly inhibited by CIT in both culture medium and pork without affecting bacterial growth. Transcriptomic analysis indicated that the differentially expression genes encoding α-hemolysin, SEs, and other virulence factors were down-regulated after treatment with 1/2MIC CIT. Moreover, the genes related to QS including agrA and agrC were also down-regulated, while the global transcriptional regulator sarA was up-regulated. Data here demonstrated that CIT could inhibited S. aureus virulence factors production through disturbing QS systems. In a challenge test, the addition of CIT caused a remarkable inhibition of S. aureus population and delay in lipid oxidation and color change on pork after 15 days incubation at 4 °C. These findings demonstrated that CIT could not only efficiently restrain the production of S. aureus virulence factors by disturbing QS, but also exhibit the potential application on the preservation of meat products.
