P/N/S-containing cellulose nanofibrils enable curcumin encapsulation via Pickering emulsion based microcapsules

Qingtian Xu,Jierui Ye,Shuaibo Han,Yuebing Gao,Pan Chen,Siqun Wang,Qiang Wu,Qian Li

Renewable polymeric microcapsules hold promise for precisely controlled release of hydrophobic drugs (curcumin) in therapeutic treatment. However, there still remains challenges to stabilize the properties of microcapsules and to enhance encapsulation efficiencies of curcumin. By using an eco-friendly ternary deep eutectic solvent here, we successfully fabricated native cellulose nanofibrils, holocellulose nanofibrils, and lignocellulose nanofibrils containing both phytates and sulfate groups. In addition, a novel type of curcumin-loaded polymeric microcapsules with diameter of 10–20 μm were prepared by cellulose nanofibrils as stabilizing agents in Polylactide Pickering emulsion. The Pickering emulsions stabilized by P/N/S-containing cellulose nanofibrils, holocellulose nanofibrils, and lignocellulose nanofibrils are more stable than the unmodified cellulose nanofibrils-stabilized emulsion. The Pickering emulsion stabilized by 0.2 wt% P/N/S-containing lignocellulose nanofibrils exhibited the highest emulsion layer height and the zeta potential reached −39.74 ± 2.88 mV. Moreover, the highest encapsulation efficiency of curcumin can be up to 94.80%. Antimicrobial test revealed that curcumin in the polymeric microcapsules exhibited an excellent inhibitory effect on S. aureus and the antibacterial efficiency reached 98.66%. These results provided important information on the comparison of different cellulose nanofibrils-stabilized Pickering emulsions and the application of controlled release of curcumin incorporated polymeric microcapsules in drug delivery system.

Binding, stability, and antioxidant activity of curcumin with self-assembled casein–dextran conjugate micelles

Yue Wu,Xiaoyong Wang

This work studied the potential of self-assembled micelles of casein–dextran conjugates (CD) as a protective carrier for bioactive curcumin. CD was prepared through the Amadori rearrangement of the Maillard reaction. In casein micelles and CD micelles, the addition of curcumin can quench the intrinsic fluorescence of casein gradually. According to the double logarithm equation, the binding constants were determined as 3.9 × 104 and 1.8 × 105 M−1 for the binding of curcumin with casein micelles and CD micelles, respectively. The higher affinity of hydrophobic binding of curcumin with CD micelles was ascribed to the more hydrophobic and compact structure of casein core surrounded by the hydrophilic dextran shell, which was supported by the fluorescence spectrum and fluorescence anisotropy of curcumin. Furthermore, CD micelles were found to be superior to casein micelles for improving the stability and radical scavenging activity of curcumin.

Chitosan-curcumin conjugate prepared by one-step free radical grafting: Characterization, and functional evaluation

Xuerong Luo,Lingyu Zhao,Imran Mahmood Khan,Lin Yue,Yin Zhang,Zhouping Wang

Curcumin (Cur) is a naturally hydrophobic polyphenol, and it has a wide range of physiological functions. But the practical application of Cur is constrained by its low water solubility and poor stability. To improve these deficiencies of Cur, a novel Cur derivative (CS-Cur) was prepared by grafting chitosan (CS) with Cur through a one-step reaction of a free radical-mediated redox system. A series of characterizations provided evidence that the grafting of CS with Cur was successful. The obtained CS-Cur showed lower crystallinity and thermal properties than CS and Cur. After grafting, the water solubility of CS-Cur was found to be 9.76 ± 2.45 g/L and greatly improved. Meanwhile, the CS-Cur showed good photothermal stability, antioxidant activity, and photodynamic antibacterial activity in an aqueous solution, and it had good in vitro biosafety. This provides an idea for the design and synthesis of novel highly water-soluble Cur derivatives and also improves the practical application of Cur in aqueous systems.

Curcumin–polydopamine nanoparticles alleviate ferroptosis by iron chelation and inhibition of oxidative stress damage

Li Lei,Jiali Yuan,Qingqing Yang,Qiuxia Tu,Haijun Yu,Liangzhao Chu,Lei Tang,Chunlin Zhang

Ferroptosis, characterized by elevated iron levels and lipid peroxidation (LPO), is a recently identified regulatory mechanism of cell death. Its substantial involvement in ischemic tissue injury, neurodegenerative disorders, and cancer positions ferroptosis inhibition as a promising strategy for managing these diverse diseases. In this study, we introduce curcumin–polydopamine nanoparticles (Cur–PDA NPs) as an innovative ferroptosis inhibitor. Cur–PDA NPs demonstrate remarkable efficacy in chelating both Fe2+ and Fe3+ in vitro along with scavenging free radicals. Cur–PDA NPs were found to efficiently mitigate reactive oxygen species, reduce Fe2+ accumulation, suppress LPO, and rejuvenate mitochondrial function in PC12 cells. Thus, these NPs can act as potent therapeutic agents against ferroptosis, primarily via iron chelation and reduction of oxidative stress.

Efficient electrochemical synthesis of green fluorescent carbon dots for curcumin detection in common foods

Chuntong Liu,Haiyan Qi,Tao Jing,Jun Li,Ming Zhao,Lixin Qiu,Qiuying Li

Curcumin (Cur) is commonly employed as a food additive in the pharmaceutical and food industries. However, improper use of curcumin can lead to adverse effects on human health. In this research, we developed an electrochemical approach for the synthesis of green fluorescence nitrogen and sulfur co-doped carbon dots (G-CDs) specifically designed for curcumin detection. The prepared G-CDs displayed strong fluorescent emissions at 540 nm, along with notable attributes such as excellent fluorescence stability against photobleaching, and resilience to changes in ionic strength. The G-CDs demonstrated selective recognition of curcumin, resulting in fluorescence quenching in a linear range of 0–140 μmol/L, with an impressive detection limit of 46.4nmol/L. Five distinct food samples were effectively detected for curcumin using the developed G-CDs nanoprobe, yielding satisfactory outcomes. This newly proposed approach offers simplicity, outstanding sensitivity, and selectivity, showcasing its promising capabilities as an alternative method for Cur detection without the need for specialized instruments.

Enhanced bioavailability of curcumin amorphous nanocomposite prepared by a green process using modified starch

Changying Chen,Zhixing Wang,Hongliang Fu,Guoqi Yu,Xiang Luo,Kewu Zhu

Curcumin (Cur), a bioactive compound extracted from plants, has attracted widespread attention due to its multiple pharmacological activities . However, the low bioavailability due to the inherent limitations in water solubility, chemical stability, and permeability poses great challenges for realizing its clinical potentials. In the current study, octenyl succinic anhydride-modified starch (OSA-S), a renewable and biodegradable biopolymer , was employed to fabricate Cur amorphous composite nanoparticles (Cur/OSA-S NPs) through a solvent-free pH-driven method with the aim to enhance Cur's bioavailability by improving its solubility and stability. Cur/OSA-S NPs, with mean sizes of about 128.9 ± 8.6 nm, encapsulation efficiencies of about 90.0 %, and the drug loading capacities around 51.0 ± 0.2 %, were successfully prepared. Cur was found to be dispersed within the composite nanoparticles in amorphous state as confirmed by the XRD and DSC characterizations. In addition, Cur/OSA-S NPs offers excellent storage, thermal and light stability, excellent re-dispersibility, and approximately 92 times better solubility than the original Cur. Furthermore, studies of dissolution and the parallel artificial membrane permeability assay (PAMPA) confirmed enhanced dissolution rates and in vitro permeabilities of Cur/OSA-S NPs. Cancer cell viability and uptake experiments revealed that Cur/OSA-S NPs possessed more potent inhibitory effects on cancer cell proliferation compared to the raw Cur. The results obtained from the current study demonstrated the effectiveness of OSA-S for manufacturing Cur amorphous composite nanoparticles with enhanced solubility, stability, and permeability, which might be valuable for further development of Cur based products for treatment of various diseases.

An Asymmetric Bacterial Cellulose Membrane Incorporating CuPt Nanozymes and Curcumin for Accelerating Wound Healing

Lei Li,Haoze Li,Junrong Wang,Yulin Xie,Minghong Gao,Zhongjun Yang,Chunxia Li

Damaged skin compromises its ability to effectively prevent the invasion of harmful bacteria into the tissue, leading to bacterial infection of the wound and hindering the healing process. To address this challenge, we have developed a multifunctional asymmetric wound dressing (CuPt-Cur-ABC) that effectively addresses the lack of bactericidal activity and the release of active ingredients in conventional bacterial cellulose (BC), which can be employed to create a barrier of defense between the wound and its surrounding environment. Compared with BC, asymmetric bacterial cellulose (ABC) used starch as a pore-causing agent, forming holes of different sizes at the top and bottom, which enhanced the ability of ABC to load and moderate-release drugs. First, as-synthesized CuPt nanozymes with an octopod nanoframe structure had multiple enzymatic activities including peroxidase-like, catalase-like, and glutathione peroxidase-like activities. Then, CuPt and curcumin (Cur) were loaded into ABC under ultrasound. Under 808 nm laser irradiation, the nanocomposites possessed good photothermal properties. So the photothermal therapy combined with chemodynamic therapy and inherent antibacterial performance of Cur achieved 99.3% and 99.6% in vitro bactericidal efficacy against Staphylococcus aureus and Escherichia coli, respectively. Moderate release of Cur can clear the excess reactive oxygen species and promote the polarization of macrophages toward the M2 type. In vivo experiments additionally confirmed that the constructed wound dressing achieved multiple functions, including effective antibacterial activity, reversing the inflammatory microenvironment, and promoting wound healing.

A metal mediated specific poly(cyclotriphosphazene-co-curcumin) fluorescent micro-probe for tetracycline sensing

Xiqi Su,Le Wang,Xiao Zhang,Jin-Hua Xie,Yi Qu,Di Zhang,Li-June Ming

The extensive use of tetracycline antibiotics (TCs) renders their widespread in the environment that may cause bacterial resistance. Thus, detection of residual TCs and understanding of their broader roles and impacts in the environment and soil ecology are is of critical significance. This paper describes the investigation of a Mg (II)-specific fluorescent micro-probe (PCC–Mg) based on cyclotriphosphazene (Cpz) and curcumin for classical TC•HCl sensing with a low detection limit of 29 nM and a fast response time of less than 10 s. The sensing mechanism is presumably attributed to specific Mg 2+ binding with TCs since the fluorescence enhancement is not observed in the absence of Mg 2+ and not influenced by the presence of several metal ions and anions. The binding of Mg 2+ to curcumin/TC and the formation of TC-metal-curcumin ternary complex can be concluded with NMR and mass spectrometry . Moreover, for the purpose of easy applications, test strips are made to render visual detection of TC with noticeable and selective fluorescence enhancement. The formation of the ternary complex between Cur from the popular herb Curcuma longa and TC from Streptomyces spp. mediated by metal ions also hints a potential role of microbiolochemical-phytochemical interactions in maintaining biodiversity and equilibria in soil microecology.

Protective effects and mechanisms of curcumin on podophyllotoxin toxicity in vitro and in vivo

Juan Li,Cai-Xia Dai,Hua Sun,Lu Jin,Chong-Yi Guo,Wei Cao,Jie Wu,Hai-Yan Tian,Cheng Luo,Wen-Cai Ye,Ren-Wang Jiang

Podophyllotoxin (POD) is a naturally occurring lignan with pronounced antineoplastic and antiviral properties. POD binds to tubulin and prevents the formation of mitotic spindle . Although cases of overdose or accidental ingestion are quite often, no specific therapy is currently available to treat the POD intoxication. In the current investigation, the protective effects and mechanisms of curcumin (CUR) on podophyllotoxin toxicity were evaluated in vitro and in vivo . The results showed that CUR could protect POD-induced cytotoxicity by recovering the G2/M arrest and decrease the changes of membrane potential and microtubule structure in Vero cells. A significant decrease of mortality rates was observed in Swiss mice treated by intragastrical administration of POD   +   CUR as compared with POD alone. The POD   +   CUR group also exhibited decreases in plasma transaminases , alkaline phosphatase , lactate dehydrogenase , plasma urea , creatinine and malondialdehyde level but elevated superoxide dismutase and glutathione levels as compared to the POD group. Histological examination of the liver and kidney demonstrated less morphological changes in the treatment of POD   +   CUR as compared with POD alone. The mechanism of the protective effects might be due to the competitive binding of CUR with POD in the same colchicines binding site as revealed by the tubulin polymerization assay and the molecular docking analysis, and the antioxidant activity against the oxidative stress induced by POD. In summary, both in vitro and in vivo data indicated the promising role of CUR as a protective agent against the POD poisoning.

PEGylated Doxorubicin Micelles Loaded with Curcumin Exerting Synergic Effects on Multidrug Resistant Tumor Cells

Huang Saiyan,Liu Jieying,Zhu Hao,Hussain Abid,Liu Qing,Li Jing,Shen Yuanyuan,Cheng Jiejun,Guo Shengrong

A novel folate-polyethylene glycol-succinic acid-doxorubicin conjugate (FA-PEG-SA-DOX) micelle loaded with curcumin (CUR) is designed and prepared to target tumor cells via folate-folate receptor interactions and co-deliver chemotherapeutic agent DOX and chemosensitizer CUR for treatment of multidrug resistant human breast cancer MCF-7/ADR cells. The CUR/FA-PEG-SA-DOX micelles are characterized with the average size of 90.64±0.15 nm and the zeta potential of −15.3±0.8 mV. The CUR and DOX contents of the micelles are 12.07 and 15.69% (w/w), respectively. The cytotoxicity, cellular uptake and effiux of the micelles are investigated by using MCF-7/ADR cells. The results demonstrate that the CUR/FA-PEG-SA-DOX micelles have greater toxicity (IC50 = 3.10 µg/ml) compared to the free DOX (IC50 = 31.99 µg/ml), moreover, the micelles present increased cellular uptake and reduced cellular effiux. The combination index of CUR/FA-PEG-SA-DOX micelles was 0.17, which indicate a strong synergistic antitumor efficacy against MCF-7/ADR cells. These results suggest that Curcumin-loaded FA-PEG-SA-DOX micelles may be a promising nanomedicine for co-delivery dual-drug treatment for cancer.

Curcumin-based antibacterial waterborne polyurethane/gelatin composite film for pork preservation and freshness indication

Yuye Zhong,Zhenhong Zhang,Zhengjie Chen,Wenshuo Zhang,Xinghai Liu,Pingping Zhao,Tao Zhang,Houbin Li

Gelatin possesses excellent film-forming ability, biodegradability, and biocompatibility, which makes it a promising alternative to petroleum-based plastics. However, its application in food packaging is limited by its brittleness, high water sensitivity, and insufficient microbial protection. In this study, curcumin was introduced as a chain extender into castor oil-based waterborne polyurethane (CWU), which was then blended with gelatin as a functional material to create reactive food composite films (CWGL). The effects of varying CWU concentrations on the microstructure and properties of the films were thoroughly investigated. The incorporation of CWU enhanced the mechanical properties, water solubility, water vapor barrier, oxygen barrier, and UV shielding capabilities of the gelatin-based composite films. Additionally, CWU imparted antioxidant, antibacterial, and pH-responsive properties to the CWGL. Packaging application tests demonstrated that CWGL could extend the shelf life of fresh pork up to 10 days under refrigeration. Furthermore, CWGL exhibited a visible color change (∆E > 5) during pork storage, suggesting its potential use as a freshness-indicating innovative packaging. This study presents a novel strategy to advance the application of gelatin in innovative food packaging.

Photothermal and Immunomodulatory Hydrogels Loaded with Black Phosphorus Nanosheets and Curcumin for Wound Healing

Rong Xiao,Longbao Feng,Xinyue He,Yifan Hao,Zhong Cao,Aliaksandr Astrowski,Yi Zhang,Rui Guo

In the field of skin wound treatment, traditional methods are often limited by the dual challenges of effectively clearing bacterial infections and accelerating wound healing. To address these issues, this study developed an innovative bioactive hydrogel that combines photothermally responsive antimicrobial and immunomodulatory effects. The hydrogel utilized methacrylated gelatin and oxidized dextran, forming a stable network structure through radical and Schiff bonds, loaded with black phosphorus (BP) nanosheets and curcumin (Cur). Characterization of the hydrogel’s physicochemical properties revealed not only its excellent swellability, biodegradability, mechanical strength, thermal stability, and antioxidative properties but also its ability to control the release of Cur through the photothermal effect. Under 808 nm NIR irradiation, the suitable photothermal effect was observed with BP nanosheets at a concentration of 250 μg/mL under a power density of 1.5 W/cm2, and the photothermal conversion efficiency (η) was 41.34%. In vitro experiments showed that the hydrogel exhibited a bactericidal rate of over 90% against E. coli, S. aureus, and P. aeruginosa and more than 80% antibiofilm formation activity. After coculturing with HUVEC cells for 24 and 48 h, cell viability remained above 80%. Cell migration assay confirmed the hydrogel’s effectiveness in promoting cell migration (closure rate reached 97.4% within 24 h). In a rat model with infected wounds, it was shown that on day 14, wounds were nearly healed, with an antibacterial rate of over 85%, the expression levels of proinflammatory factors TNF-α and IFN-γ were reduced, the expression of anti-inflammatory factors IL-4 and TGF-β1 was increased, re-epithelialization was observed, and collagen was deposited, which facilitated the rapid healing of the wounds. The hydrogel in this study with photothermal and immunomodulatory capabilities offers an efficient strategy for treating infected wounds.
