Efficient hydration of 2-amino-2,3-dimethylbutyronitrile to 2-amino-2,3-dimethylbutyramide in a biphasic system via an easily prepared whole-cell biocatalyst

Shujing Zhu,Xiaoqiang Ma,Erzheng Su,Dongzhi Wei

From an environmental perspective, utilizing nonconventional solvents (i.e. green solvents) with low ecological footprints is a highly beneficial alternative to using conventional organic solvents to form a reaction system. The nitrile hydratase (NHase, EC 4.2.1.84) catalyzed hydration of 2-amino-2,3-dimethylbutyronitrile (ADBN) to 2-amino-2,3-dimethylbutyramide (ADBA) in various green solvent–aqueous reaction systems was investigated in this study. After systematically optimizing the reaction conditions, the HFE-7100/H2O (v/v, 10%) biphasic system was ultimately identified as a promising reaction system for reducing product inhibition, avoiding substrate hydrolysis, and facilitating product separation and solvent recovery. The average ADBA yield of an entire batch reaction was 97.3%, which is obviously higher than those obtained with previously reported chemical or enzymatic methods. This is the first attempt to apply a fluorous solvent–aqueous biphasic system to a biocatalytic process, and the results suggest that the fluorous solvent employed in the biphasic system satisfies the requirements for green chemistry.

Sodium-Alginate-Modified Electrospun Cod-Collagen-Peptide Nanofibers: Box–Behnken Process Optimization and Structure–Hydration–Property Correlations in a Chitosan-Supported Composite Mat

Han Jiawei,Wang Yi,Deng Kehui

Electrospun protein–polysaccharide nanofibers are promising biocompatible substrates for skin-contact materials, yet controlling their morphology and hydration behavior in ternary systems remains challenging. In this work, the modification of a cod collagen peptide/poly(ethylene oxide) (CCP/PEO) system with sodium alginate (SA) was investigated as a model polyelectrolyte interaction. A three-factor Box–Behnken design was applied to optimize the spinning-solution preparation for the liquid absorption capacity (LAC), yielding an optimum at a 12.5 h reaction time, 30 °C, and a 30:1 CCP/SA mass ratio. SEM, FTIR, and XRD analyses indicated that electrostatic association and hydrogen bonding between the protonated amino groups of CCP and carboxylate groups of SA facilitated the formation of uniform nanofibers (458 ± 114 nm) with reduced crystallinity. These interactions significantly enhanced the mechanical performance ( p  < 0.01), with a 13.1% increase in tensile strength and a 27.3% increase in elongation at break. Compared to unmodified controls, the SA-modified composite mats exhibited a ~ 27% higher LAC (1063.9%) and improved water retention after simulated skin contact, despite a moderate reduction in permeability. A three-stage hydration mechanism—comprising capillary substrate uptake, osmotic swelling within the proposed polyelectrolyte network, and nanofiber dissolution—is proposed to explain the enhanced hydration capacity. This study provides a quantitative process map for the design of high-performance dry-state carriers for functional facial masks.

Polyethyleneimine/nano-silica core–shell materials with different degrees of branching for improving shale hydration inhibition and plugging in water-based drilling fluids

Yingxin Wang,Lei Tang,Dongyu Qiao,Yuhao Liu,Xuecheng Zheng

Clay mineral hydration dispersion and pressure transfer are the main factors leading to well wall instability during shale development. In this study, the surface of nano-SiO2 was modified with silane coupling agent KH-550, polyethyleneimine PEI30 000 and PEI300, and three kinds of end-amidated nano-SiO2 core–shell structural materials with different degrees of branching were successfully prepared. The results show that –NH2 is truly present on the surface of three modified SiO2 nanostructures. Additionally, the modified nanosilica nanoparticles show better dispersion in water and enhanced thermal stability compared with the unmodified sample. Linear swelling and drum recycling were used to investigate the inhibitory effect of sodium bentonite on water sensitivity, with BPEI@SiO2 exhibiting the strongest inhibition against clay hydration expansion. At a 2.0 wt% concentration, it reduced the 16-hour expansion rate of sodium bentonite by 30.77%. Pressure transmission and microporous filtration membrane blocking demonstrated that the amine-terminated nano-SiO2 exhibits a smaller particle size than the other samples, enabling effective blocking of pore channels within the rock core. The zeta potential and the clay-layer spacing test were used to thoroughly analyze the mechanism of action of the end-amidated nano-SiO2. The results showed that all three types of modified end-amidated nano-SiO2 exhibited effective chemical inhibition and physical sealing, as evidenced by a pronounced increase in the zeta potential of bentonite suspensions from −32.6 mV to −18.95 mV and a significant reduction in clay interlayer spacing after treatment with BPEI@SiO2. The interactions between modified silica nanoparticles and shale showed different mechanisms of action for the modified nano-SiO2 with different chain-length shells. KH550@SiO2 neutralises the negative charge on the clay surface by protonating –NH3+. BPEI@SiO2 inserts into the bentonite crystal layer and reduces its crystal layer spacing to inhibit its hydration and swelling. In contrast, HPEI@SiO2 features a larger and more distinctive hyperbranched polymer shell, which extends outward from the nanoparticle surface and enables simultaneous electrostatic adsorption onto the external surfaces of multiple bentonite grains.

Approaches to scarless burn wound healing: application of 3D printed skin substitutes with dual properties of anti-infection and balancing wound hydration levels

Shuying Chen,Yahui Xiong,Fan Yang,Yanke Hu,Jinghao Feng,Fei Zhou,Zhonghua Liu,Hengdeng Liu,Xiaogang Liu,Jingling Zhao,Zhaoqiang Zhang,Lei Chen

Background Severe burn wounds face two primary challenges: dysregulated cellular impairment functions following infection and an unbalanced wound hydration microenvironment leading to excessive inflammation and collagen deposition. These results in hypertrophic scar contraction, causing significant deformity and disability in survivors. Methods A three-dimensional (3D) printed double-layer hydrogel (DLH) was designed and fabricated to address the problem of scar formation after burn injury. DLH was developed using methacrylated silk fibroin (SFMA) and gelatin methacryloyl (GelMA) for the upper layer, and GelMA and hyaluronic acid methacryloyl (HAMA) for the lower layer. To combat infection, copper-epigallocatechin gallate (Cu-EGCG) was incorporated into the lower layer bioink, collectively referred to as DLS. To balance wound hydration levels, HaCaT cells were additionally encapsulated in the upper layer, designed as DLS/c. Findings DLH demonstrated suitable porosity, appropriate mechanical properties, and excellent biocompatibility. DLS exhibited potent antimicrobial properties, exerted anti-inflammatory effects by regulating macrophage polarisation, and may enhance angiogenesis through the HIF-1α/VEGF pathway. In the DLS/c group, animal studies showed significant improvements in epidermal formation, barrier function, and epidermal hydration, accompanied by reduced inflammation. In addition, Masson’s trichrome and Sirius red staining revealed that the structure and ratio of dermal collagen in DLS/c resembled that of normal skin, indicating considerable potential for scarless wound healing. Interpretation This biomimetic matrix shows promise in addressing the challenges of burn wounds and aiming for scarless repair, with benefits such as anti-infection, epidermal hydration, biological induction, and optimised topological properties.

Injectable strontium-containing tricalcium silicate biocements reinforced by nano HA/F127 hydrogel: Enhanced hydration kinetics, anti-washout, and dentinogenesis for vital pulp preservation

Meng Wu,Yucheng Zhou,Sichao Huang,Bixin Jiang,Peng Huang,Nan Liang,Laijun Xu

Preserving the vital pulp poses significant challenges in clinical pulp injury repair. Tricalcium silicate (C3S)-based cement has emerged as a successful pulp capping biomaterial. However, its efficacy in inducing pulp-dentin complex regeneration requires enhancement, and handling difficulties, poor anti-collapsible ability and inadequate self-setting performance remain significant obstacles, restricting its broader application in endodontics. In this study, a multifunctional SrCO 3 -doped C3S biocement (C3S-Sr/F127-nHA) reinforced by Pluronic® F127 thermoreversible hydrogel and nano hydroxyapatite (nHA) was developed to overcome the limitations of C3S-based cements. The thermoresponsive characteristic of F127 conferred the C3S pastes smart response behavior, allowing for optimal operability at room temperature and maintaining excellent anti-collapse properties at body temperature. C3S-Sr/F127-nHA exhibited an injectability rate of up to 93 %. nHA facilitated the hydration kinetics of C3S, resulting in reduced setting time and enhanced compressive strength. Furthermore, C3S-Sr/F127-nHA demonstrated excellent apatite mineralization ability, cytocompatibility, and potential for osteo/odontogenic induction. The inclusion of Sr 2+ ions and nHA promoted osteo/odontogenic differentiation of stem cells isolated from human exfoliated deciduous teeth (SHED). In vivo findings indicated that C3S-Sr/F127-nHA could stimulate the formation of a homogeneous dentin bridge. Consequently, C3S-Sr/F127-nHA is a promising biocement for use in regenerative endodontics.

Enhancing Cement Hydration and Mechanical Strength via Co-Polymerization of Sodium Humate with Superplasticizer Monomers and Sequential Blending with Aluminum Sulfate and Carbon Fibers

Zhiyuan Song,Sidra Chaudhary,Yan Ding,Yujiao Yan,Qinxiang Jia,Yong Wu,Xiaoyong Li,Yang Sun

This study presents a new ternary copolymer synthesized via aqueous free-radical polymerization from sodium humate, sodium 2-methylprop-2-ene-1-sulfonate (SMAS), and 2-acrylamido-2-methylpropane sulfonic acid (AMPS). The resulting highly water-soluble, three-dimensional porous copolymer is complexed with aluminum sulfate to form a composite admixture containing AlO(OH), which acts as a highly effective accelerator for cement hydration. This system significantly shortens the initial and final setting times to averages of 2.62 min and 4.53 min, respectively, and enhances early-age mechanical strength (1.7 MPa compressive, 1.4 MPa flexural at 6 h). These improvements are correlated with the formation of key crystalline phases, including Al2Si2O5(OH)4and Ca3Al2O6·xH2O gel. Incorporation of 50-mesh carbon fibers further reduces setting times (2.21 min initial, 3.93 min final) and increases 24 h strength (5.2 MPa compressive, 2.7 MPa flexural), despite a slight reduction in early strength (at 6 h). In contrast, 200-mesh carbon fibers extend the initial setting time and diminish early strength, associated with the formation of less effective gel phases such as Ca3Al2O6·xH2O, (CaO)x(Al2O3)11, and Ca4Al2O7·xH2O. Among these, the Al2Si2O5(OH)4phase demonstrates superior performance, while finer carbon fibers show limited effectiveness in bridging hydration products. Conventionally employed as retarders or reinforcing agents, humate-based polymers and carbon fibers are shown here to function as dual-functional admixtures—serving as efficient setting accelerators while enhancing mechanical properties through tailored material design. This strategy offers a promising pathway for developing advanced multifunctional cement admixtures.

Anti-fouling ternary zwitterionic coating membrane with strong surface hydration and salt resistant for efficient long-term emulsion separation in high salinity marine environment

Yan Zhang,Jianlin Yang,Yali Zhao,Zhecun Wang,Gwo Sung Lai

Membrane separation has been acknowledged as an advanced technology to handle the oil spills. However, most of the superhydrophilic/underwater superhydrophobic membranes currently applied for oil/water separation would lose their superwettability when exposed to salty marine environments, leading to serious membrane plugging and separation performance deterioration. Herein, inspired by seawater fish and mussels, a simple, green, and versatile one-step co-deposition strategy is proposed for fabricating a ternary zwitterionic coating with strong surface hydration on diverse membranes, showing excellent anti-fouling properties in high salinity environments, even in the saturated NaCl solution. Moreover, the modified membrane performs well in the long term when separating surfactant-stabilized oil-in-seawater emulsion and with impressive recyclability. The ternary zwitterionic coating membrane exhibits great potential in treating oily wastewater in marine.

Controlled Hydration, Transition, and Drug Release Realized by Adjusting Layer Thickness in Alginate-Ca2+/poly(N-isopropylacrylamide) Interpenetrating Polymeric Network Hydrogels on Cotton Fabrics

Pan Gu,Bing Li,Bisheng Wu,Jiping Wang,Peter Müller-Buschbaum,Qi Zhong

The controlled hydration, transition, and drug release are realized by adjusting layer thickness in thermoresponsive interpenetrating polymeric network (IPN) hydrogels on cotton fabrics. IPN hydrogels are synthesized by sodium alginate (SA) and poly(N-isopropylacrylamide) (PNIPAM) with a ratio of 1:5/% (w/v). The cotton-fabric-supported IPN hydrogels with a thickness of 1000 μm exhibit a transition temperature (TT) at 35.2 °C. When the hydrogel thicknesses are thinned to 500 and 250 μm, the TTs are reduced to 34.8 and 34.1 °C, respectively. Interestingly, the morphology of IPN hydrogels switches from a well-defined honeycomb-like network structure (1000 μm) to a densely packed layer structure (250 μm). The thinner layers not only present a smaller extent of hydration and collapse but also require longer time to reach an equilibrium state, which can be attributed to the more pronounced hindrance of the chain rearrangement by the cotton fabrics. To address the influence of layer thickness on the drug release, we compare the release rate and cumulative release percentage of the test drugs tetracycline hydrochloride (TCH) and levofloxacin hydrochloride (LH) between pure IPN hydrogels and cotton-fabric-supported IPN hydrogels (250, 500, and 1000 μm) at 25 °C (below the TT) and 37 °C (above the TT). Because of the compressive stress from the collapsed hydrogels, a higher release is observed in both hydrogels when the temperature is above TT. The cotton fabric induces a slower and less prominent drug release in IPN hydrogels. Thus, combining the obtained correlation between the transition and hydrogels layer thickness, the drug release in cotton-fabric-supported IPN hydrogels can be regulated by the layer thickness, which appears especially suitable for a controlled release in wound dressing applications.

A green and recyclable CuSO4·5H2O/ionic liquid catalytic system for the CO2-promoted hydration of propargyl alcohols: an efficient assembly of α-hydroxy ketones

Ye Yuan,Jia Hu,Yu Wang,Leilei Huang,Jiayun Zheng,Yufeng Zhao,Yiyun Chen,Cheng Chen,Francis Verpoort

α-Hydroxy ketones are important building blocks in biological, pharmaceutical and synthetic chemistry. In this work, diverse α-hydroxy ketones were efficiently constructed through the CO 2 -promoted hydration process of propargyl alcohols, which was catalyzed by a system consisted of economical CuSO 4 ·5H 2 O and a green 1-butyl-3-methylimidazolium acetate ionic liquid. Particularly, this catalytic system exhibited excellent activity under atmospheric CO 2 or even mimetic flue gas (20 vol% of CO 2 ). Moreover, this system employed the lowest metal loading ever reported (0.004–0.25 mol%) meanwhile reached the highest turnover number (11700) for the target hydration reaction. Additionally, this is the first reported Cu catalytic system with reliable recyclability, which could be easily reused at least 6 times with yields higher than 85%.

Regulation mechanism of selective adsorption-hydration effects in chalcopyrite-molybdenite flotation separation using 2-iminothiazolidin-4-one (ITO) as chalcopyrite depressant: Experimental and DFT studies

Xiangwen Lv,Anruo Luo,Xiong Tong,Jianhua Chen,Sheng Jian,Hejie Ouyang

The development of environmentally benign depressants with high selectivity remains a critical challenge in sustainable copper-molybdenum flotation separation. To overcome the limitations of conventional toxic reagents (e.g., cyanides and sulfides), this study introduces 2-iminothiazolidin-4-one (ITO) as a novel chalcopyrite depressant. Through integrated experimental characterization and density functional theory (DFT) simulations, we systematically investigated ITO's selective depression mechanism. Micro-flotation experiments confirmed ITO's ability to preferentially suppress chalcopyrite while maintaining molybdenite floatability. Surface analysis via Fourier-transform infrared spectroscopy (FT-IR), time-of-flight secondary ion mass spectrometry (TOF-SIMS), X-ray photoelectron spectroscopy (XPS), and density functional theory (DFT) demonstrated that the thiazolidine (-N-C S) and imino (=NH) groups coordinate with surface copper/iron (Cu/Fe) ions to form stable chelates on chalcopyrite, exhibiting stronger adsorption stability than ethyl xanthate. In contrast, only weak physical adsorption occurs on molybdenite surfaces. The carbonyl group (C O) was further found to enhance chalcopyrite hydrophilicity through hydrogen-bonding interactions with water molecules, as supported by contact angle measurements. This work establishes a molecular design strategy for green flotation reagents by leveraging functional group synergism, providing fundamental insights into mineral surface reactivity regulation for strategic metal resource recovery.

The Encapsulation of Illite Powders with Al2(SO4)3·18H2O and Hydrophilic Copolymers: Accelerating and Toughening Cement Hydration Through the Proliferation of 54CaO·MgO·Al2O3·16SiO2 Clinker

Zhiyuan Song,Sidra Chaudhary,Zainab Bibi,Yong Wu,Qinxiang Jia,Xiaoyong Li,Yang Sun

Two hydrophilic copolymers containing functional groups such as carboxyl, amido, and sulfonic acid are synthesized using ammonium persulfate-catalyzed free radical polymerization in water. Aluminum sulfate is then introduced, resulting in two polymer complexes that exhibit reduced cement setting times (initial, 1.16–2.44 min; final, 2.02–3.14 min) and improved compressive (24 h, 5.81–7.25 MPa) and flexural (24 h, 2.80–2.99 MPa) strengths compared to pure aluminum sulfate-facilitated cementing (initial, 19.11 min; final, 37.05 min; compressive, 24 h, 5.51 MPa; flexural, 24 h, 2.56 MPa). Following this, ball-milled illite powder is added, and the resulting admixtures further display slightly prolonged setting times (initial, 2.35–2.99 vs. 1.16–2.44 min; final, 3.98–4.35 vs. 2.02–3.14 min), along with comparable compressive strengths (5.85–7.11 vs. 5.81–7.25 MPa) and enhanced flexural strengths (3.92–5.83 vs. 2.80–2.99 MPa). Notably, a unique adhesive pozzolanic clinker, Ca54MgAl2Si16O90(54CaO·MgO·Al2O3·16SiO2), emerges in the presence of illite-based admixtures, contributing to the mechanical strength development of the hydrated mortars. Although illite itself is hydrophobic, the coating of ball-milled illite powder with aluminum sulfate and copolymers facilitates its dispersion into the gaps and pores of the cement matrix during setting, thereby increasing the flexural strength. This work presents an interesting approach to utilizing illite materials in cement applications, which is significant for reducing CO2emissions during cement production and use.

Understanding the acceleration effect of manganese and cerium doping on the hydration of CaO in CaO/Ca(OH)2 heat storage by density function theory

Zhiguo Bian,Yingjie Li,Feifei Wang,Yi Fang,Jianli Zhao,Jianhui Qi

CaO/Ca(OH) 2 heat storage system is one of the most promising candidates for large-scale thermochemical heat storage at temperature of 300–600 °C. In this work, the CaO/Ca(OH) 2 heat storage performances of the Ce- and Mn-doped CaO were investigated and the mechanisms of promoting hydration by Ce and Mn doping were determined via density functional theory (DFT) calculations. The Mn-doped CaO exhibits the higher hydration conversion than CaO and Ce-doped CaO. The structural parameters, partial density of states, electron differential densities and energy barriers during hydration stage on the CaO, Ce- and Mn-doped CaO surfaces were compared to clarify the effects of Ce and Mn on hydration of CaO. The results show the detailed hydration reaction pathways on the CaO, Ce- and Mn-doped CaO surfaces. During the H 2 O adsorption stage, H 2 O on the Ce-doped CaO surface approaches the Ca atom first, while H 2 O on the Mn-doped CaO surface approaches the O s atom first. The adsorption energy for Ce- and Mn-doped CaO are −1.612 and −0.896 eV, respectively, which are higher than that for CaO. The doping of Ce and Mn accelerates the H 2 O adsorption on the CaO surface. The Mn doping causes the 84.3 % reduction in the energy barrier for hydration of CaO and the reduction by Mn doping is larger than that by Ce doping. Thus, Mn-doped CaO exhibits the higher hydration reactivity and hydration conversion than CaO and Ce-doped CaO. The DFT calculations determined the promotion mechanism of Ce and Mn doping on the hydration of CaO in CaO/Ca(OH) 2 heat storage.
