Inhibition Mechanism of P-glycoprotein Mediated Efflux by mPEG-PLA and Influence of PLA Chain Length on P-glycoprotein Inhibition Activity

Wenjing Li,Xinru Li,Yajie Gao,Yanxia Zhou,Shujin Ma,Yong Zhao,Jinwen Li,Yan Liu,Xinglin Wang,Dongdong Yin

The present study aimed to investigate the effect of monomethoxy poly(ethylene glycol)-block-poly(d,l-lactic acid) (mPEG-PLA) on the activity of P-glycoprotein (P-gp) in Caco-2 cells and further unravel the relationship between PLA chain length in mPEG-PLA and influence on P-gp efflux and the action mechanism. The transport results of rhodamine 123 (R123) across Caco-2 cell monolayers suggested that mPEG-PLA unimers were responsible for its P-gp inhibitory effect. Furthermore, transport studies of R123 revealed that the inhibitory potential of P-gp efflux by mPEG-PLA analogues was strongly correlated with their structural features and showed that the hydrophilic mPEG-PLA copolymers with an intermediate PLA chain length and 10.20 of hydrophilic–lipophilic balance were more effective at inhibiting P-gp efflux in Caco-2 cells. The fluorescence polarization measurement results ruled out the plasma membrane fluidization as a contributor for inhibition of P-gp by mPEG-PLA. Concurrently, mPEG-PLA inhibited neither basal P-gp ATPase (ATP is adenosine triphosphate) activity nor substrate stimulated P-gp ATPase activity, suggesting that mPEG-PLA seemed not to be a substrate of P-gp and a competitive inhibitor. No evident alteration in P-gp surface level was detected by flow cytometry upon exposure of the cells to mPEG-PLA. The depletion of intracellular ATP, which was likely to be a result of partial inhibition of cellular metabolism, was directly correlated with inhibitory potential for P-gp mediated efflux by mPEG-PLA analogues. Hence, intracellular ATP-depletion appeared to be possible explanation to the inhibition mechanism of P-gp by mPEG-PLA. Taken together, the establishment of a relationship between PLA chain length and impact on P-gp efflux activity and interpretation of action mechanism of mPEG-PLA on P-gp are of fundamental importance and will facilitate future development of mPEG-PLA in the drug delivery area.

p-Phenylenediamines and p-phenylenediamine quinone derivatives in rubber consumer products and typical urban dust: Sources, transformation profiles, and health risks

Tianao Mao,Wenbin Liu,Jinglin Deng,Chunci Chen,Tianqi Jia,Haifeng Li,Fei Yin

N , N ′-Substituted p -phenylenediamines (PPDs) are widely used as antioxidants in the rubber industry and are released into the environment in large quantities during the production and use of rubber products. We quantified PPDs and PPD quinone derivatives (PPD-Qs) in rubber consumer products, including car tires, rubber belts, rubber gloves, rubber cables, and rubber hoses, to obtain information on the degree of weathering over time during their use. Additionally, we investigated the occurrences and sources of PPDs and PPD-Qs in dust samples collected from four typical urban environments (roads, parking lots, automotive repair shops, and residences). The detected compounds included the highly toxic N -(1,3-dimethylbutyl)- N ′-phenyl-1,4-phenylenediamine quinone, which can cause acute mortality of coho salmon ( Oncorhynchus kisutch ). Concentrations of PPDs in the automotive repair shops reached 56.0 μg/g, and were much higher than in the other environments, while the residential samples had the lowest contaminant concentrations. In road and residential samples, N , N ′-di-2-naphthyl- p -phenylenediamine accounted for 17 %–30 % of the PPDs, and may have originated from different sources. We preliminarily identified 32 transformation products, and 11 of these were N , N ′-di-2-naphthyl- p -phenylenediamine transformation products. The average daily intakes of PPDs and PPD-Qs were calculated to assess the health risks of dust exposure in each environment. Workers had high total intakes of PPD [60.3 ng/(kg day)] and PPD-Qs [20.1 ng/(kg day)], and were at some risk of occupational exposure. These results improve our understanding of the environmental occurrences, sources, transformation, and health risks of PPDs and PPD-Qs.

Effective dopants in p-type elementary Te thermoelectrics

Xin Qian,Yu Xiao,Lei Zheng,Bingchao Qin,Yiming Zhou,Yanling Pei,Bifei Yuan,Shangkai Gong,Li-Dong Zhao

Elementary Te is predicted to be a high-performance thermoelectric material through theoretical calculation, and the experimental results have confirmed this prediction through optimizing carrier concentrations. In this work, we investigate the thermoelectric transport properties in undoped Te at 300–673 K, revealing a low thermal conductivity ∼1.8 W m−1 K−1 at room temperature and a ZT ∼ 0.2 at 653 K. To further improve the thermoelectric performance in the Te system, we choose Sn, Pb, Bi, and Sb as dopants in Te. We find Bi and Sb can effectively enhance the electrical conductivity by two orders, from ∼2 S cm−1 in undoped Te to ∼300 S cm−1 and ∼550 S cm−1 at 300 K in Bi-doped and Sb-doped samples respectively. Meanwhile, these dopants can simultaneously significantly suppress the lattice thermal conductivity, decreasing from 1.22 W m−1 K−1 in undoped Te to 0.68 W m−1 K−1 and 0.73 W m−1 K−1 at 673 K in Bi-doped and Sb-doped samples, respectively. Maximum ZT values of ∼0.8 in Te0.9925Bi0.0075 and ∼1.1 in Te0.985Sb0.015 at 673 K can be achieved. The results indicate that Sb is an effective dopant in the Te system.

P-Doped Carbon Quantum Dots with Antibacterial Activity

Shuiqin Chai,Lijia Zhou,Shuchen Pei,Zhiyuan Zhu,Bin Chen

It is a major challenge to effectively inhibit microbial pathogens in the treatment of infectious diseases. Research on the application of nanomaterials as antibacterial agents has evidenced their great potential for the remedy of infectious disease. Among these nanomaterials, carbon quantum dots (CQDs) have attracted much attention owing to their unique optical properties and high biosafety. In this work, P-doped CQDs were prepared by simple hydrothermal treatment of m-aminophenol and phosphoric acid with fluorescence emission at 501 nm when excited at 429 nm. The P-doped CQDs showed effective antibacterial activity againstEscherichia coli(E. coli) andStaphylococcus aureus(S. aureus). The minimal inhibitory concentrations (MICs) of P-doped CQD were 1.23 mg/mL forE. coliand 1.44 mg/mL forS. aureus. Furthermore, the morphologies ofE. colicells were damaged andS. aureusbecame irregular when treated with the P-doped CQDs. The results of zeta potential analysis demonstrated that the P-doped CQDs inhibit antibacterial activity and destroy the structure of bacteria by electronic interaction. In combination, the results of this study indicate that the as-prepared P-doped CQDs can be a promising candidate for the treatment of bacterial infections. Keywords:carbon quantum dots;biocompatibility;antibacterial activity

Asymmetric forward osmosis membranes from p-aramid nanofibers

Lei Miao,Tingting Jiang,Shudong Lin,Tao Jin,Jiwen Hu,Min Zhang,Yuanyuan Tu,Guojun Liu

p -Aramid is an ideal building block for forward osmosis (FO) membranes due to its extraordinary thermal resistance, chemical stability, and mechanical properties. However, existing aramid membranes have certain limitations such as large pore diameters and low salt rejection rates. In this work, we describe a facile solvent exchange-delay phase inversion strategy to prepare p -aramid nanofibrous membranes that would be suitable for FO applications. In this strategy, p -aramid nanofibers with an average diameter of 16 ± 4 nm and an average length of 382 ± 89 nm were employed as membrane matrices. Prior to the immersion of the cast film into a coagulation bath, a pre-evaporation protocol was carefully designed and introduced to provide a slower exchange rate between the good solvent and the non-solvent, which delayed the demixing process between p -aramid nanofibers and thus yielded an asymmetric membrane with a denser active layer as well as a loose substrate layer. The resultant membrane showed excellent FO water flux, NaCl rejection ratios, tensile strength, thermal properties, and solvent resistance. The membrane reported in this work may provide a promising candidate for separation applications and the results reported herein will facilitate the development of high-performance nanofibrous membranes.

Yao Feichong,Xie Wenhao,Zhao Xiqiu,Gu Hongbo

With the advent of the information age, wearable thermoelectric fabric devices have garnered significant attention for their ability to harness environmental waste heat and body heat to supply convenient, reliable, and environmentally friendly electricity for next-generation wearable electronics. However, the complex production process and unstable power supply restrict their development. Here, we report a high-performance flexible, dependable, and wearable p-type thermoelectric device consisting of silk threads/polyaniline/amino multiwalled carbon nanotubes through dyeing process. By optimizing the weaving structure and simple assembly, this p-type fabric device generates a voltage of 0.749 ± 0.003 mV, a maximum power of 0.326 ± 0.007 nW, and a power density of 1087.532 ± 22.985 nW·m −2 at a temperature difference of 90 ℃. Besides, this device also possesses an excellent photo-thermoelectric conversion capability and the assembled fabric bracelet can generate an output voltage of approximately 6.1 mV outdoors when worn on the hand of the experimental personnel. This fabric device also exhibits a superb reliable property even after 5000 times of folding. This strategy makes it easy to manufacture a thermoelectric fabric device on a large scale and provides a promising way for wearable electronics.

Characterization of Phenol Metabolization by P. stutzeri N2

Hongyun Nie,Maiqian Nie,Yongzhe Yang,Jing Zhao,Xinyi Zhang,Yutao Guo,Yi Wan,Jing Zi

Characterization of phenol metabolization by P. stutzeri N2 was proposed based on the identification of relevant metabolites and enzyme activities of catechol 1,2-oxygenase and catechol 2,3-dioxygenase. P. stutzeri N2 started to grow on 400 mg l−1 phenol with no obvious lag phase, and then phenol decomposition was rapidly completed within 32 h. Results of enzyme activities, UV spectrums of cultures, and the mass spectrum identification of cis, cis-muconic and 2-HMS, which showed that catechol C12O and C23O were acting simultaneously on the fission of the phenol ring. The existence of P-hydroxybenzoic acid and straight-chain saturated dicarboxylic acids in the culture medium showed that carboxylation and hydrogenation were involved during the phenol degradation. Metabolites containing five or four carbons indicated that catechol and its immediate ring-cleavage intermediates could be furthermore metabolized by P. Stutzeri N2. Therefore, P. stutzeri N2 which could degrade phenol with high efficiency and diverse pathways could have potential applications in the treatment of wastewater containing phenol.

Enhanced removal of p-fluoronitrobenzene using bioelectrochemical system

Huajun Feng,Xueqin Zhang,Yuxiang Liang,Meizhen Wang,Dongsheng Shen,Yangcheng Ding,Baocheng Huang,Jiali Shentu

p -Fluoronitrobenzene ( p -FNB) tends to accumulate in industrial effluents because of its recalcitrant properties. Approaches to the removal of p -FNB always encounter conflicts between treatment efficiency and economic efficiency. A bioelectrochemical system (BES) was established to facilitate the removal and mineralization of p -FNB. The treatment cost was reduced by using inexpensive electrode materials and reducing the electrical energy used. p -FNB was effectively removed using the BES, and the reaction rate was higher than the sum of the rates of two control systems, i.e., a biological system (BS) and an electrocatalytic system (ECS), by a maximum of 62.9% under a voltage of 1.4 V. The voltage is a crucial kinetic factor for the BES performance; as the voltage increased from 0 to 1.4 V, the reaction rate constants for p -FNB removal and defluorination increased from 0.0520 to 0.1811 h −1 and 0 to 0.0107 h −1 . The synergistic effect of multistrains gave a TOC removal efficiency in the BES of about 34.05%, yet the removal efficiencies were low for the two control. The defluorination reaction rate was significantly slower than the p -FNB removal rate, which indicated that defluorination lagged p -FNB removal, and p -FNB transformation to p -fluoroaniline ( p -FA) was the fastest step. The electrochemical assistance provided electrons and accelerated the electron transfer rate in the microbial reduction of p -FNB to p -FA. In this study, the critical voltage for defluorination in the BES was 0.8 V, which was approximately 0.2 V lower than that in the ECS. The decrease in the critical voltage for defluorination was based on the production of p -FA, which is more electrocatalytically activated. These results demonstrate the mechanism of efficient p -FNB removal and mineralization in a BES.

A molecular paradigm: “Plug-and-play” chemical sensing and crypto-steganography based on molecular recognition and selective response

Qing Yu Liu,Zhen Qi Bu,Min Xia Quan,Ying Wu,Xuezhi Ding,Li Qiu Xia,Jiao Yang Lu,Wei Tao Huang

Inspired by information processing and communication in nature based on molecular recognition and structural diversity, ongoing efforts aim to development of artificial molecular or nano-systems for sensing, logic computing, and even data storage and safety. However, due to their preparation/functionalization shortcomings (laborious and time-consuming), poor flexibility and compatibility, and limited paradigm, it is still a big challenge whether simple molecules can be used to achieve comprehensive and universal applications from sensing to information storage and protection. Herein, we for the first time demonstrated a molecular paradigm—computer-like “basic input output system (BIOS)” which can realize “plug-and-play” sensing, information encoding, molecular cryptography, and steganography based on a simple artificial molecule (p-nitrophenol, PNP). Based on its molecular recognition and inherent chemical identities, PNP was utilized for colorimetric detection of multiple metal ions (Hg 2+ , Fe 3+ , Al 3+ , Cr 3+ ) and distinguishing their valence (like Cr 3+ /Cr 6+ , Fe 3+ /Fe 2+ ). Interestingly, PNP can achieve the “plug-and-play” fluorescent expansion of detection channels by directly mixing with fluorescent molecules, indicating that PNP molecule can be served as a molecular BIOS with flexibility and compatibility. Impressively, the selectivity embedded in PNP-based BIOS sensing system can be developed as molecular-level double cryptographic steganography to encode, encrypt and hide specific information (like the content of classical literature). This research not only provides a basic idea for building a molecular paradigm with “plug-and-play” flexibility and compatibility, but also provides ideas for the use of molecular sensing and informatization to open up the digitalization of the molecular world.

Oxidation-Shielded P(St-MMA)@Fe3O4@P(St-MMA) Mesoporous Magnetic Microspheres: A Robust Solid-Phase Carrier for Ultrasensitive CEA Chemiluminescence Immunoassay

Yu Chen,Lina Dong,Hengyan Tian,Fei Yang,Dengbang Jiang,Minglong Yuan

Magnetic polymeric microspheres are pivotal solid-phase carriers in chemiluminescence enzyme immunoassays (CLEIA). However, their practical clinical application is frequently hindered by non-specific adsorption, irreversible aggregation, and the intrinsic susceptibility of exposed outermost Fe3O4nanoparticles to oxidation. To overcome these critical bottlenecks, we rationally engineered highly original monodisperseP(St-MMA)@Fe3O4@P(St-MMA) sandwich-structured microspheres. The bespoke amphiphilic outer shell acts as an impenetrable shield against hydration and oxidation, while maintaining a topologically size-matched mesoporous network (average pore size of 13.11 nm) for optimal antibody anchoring. Strikingly, this architecture ensures exceptional long-term colloidal stability, completely preventing macroscopic agglomeration for over six months in buffer solutions. When evaluated in a carcinoembryonic antigen (CEA), CLEIA, our microspheres achieved an ultra-low limit of detection (LOD) of 0.055 ng·mL−1and high analytical recovery (93.37–108.25%). In a head-to-head comparison with industry-standard commercial magnetic beads, the engineered microspheres delivered stronger chemiluminescent signals and lower background noise, demonstrating excellent intra-assay (CV < 4.37%) and inter-assay (CV < 10%) precision. This work establishes a scalable, highly stable materials platform that effectively resolves the persistent oxidation limitations, holding immense practical importance for next-generation ultrasensitive clinical in vitro diagnostics.

Constructing abundant phase interfaces of the sulfides/metal-organic frameworks p-p heterojunction array for efficient overall water splitting and urea electrolysis

Tao Zhao,Dazhong Zhong,Lu Tian,Genyan Hao,Guang Liu,Jinping Li,Qiang Zhao

Designing efficient electrocatalysts to improve the overall water splitting and urea electrolysis efficiency for hydrogen generation can greatly solve the dilemma of energy shortage and environmental pollution. In this work, Co 8 FeS 8 @CoFe–MOF/NF heterojunction arrays were fabricated by embedding sulfides into the surface of metal-organic frameworks (MOFs) nanosheets as multifunctional electrocatalyst. The introduction of sulfide on CoFe–MOF/NF can not only adjust the electronic structure (electron-rich state) and change the surface properties (more hydrophilic), but also increase the active area to enhance the catalytic activity. The in situ Raman shows Co 8 FeS 8 @CoFe–MOF/NF is more easily to generate active species at low potentials and generates a higher content of active β-MOOH phase than CoFe–MOF/NF. Therefore, the Co 8 FeS 8 @CoFe–MOF/NF exhibits excellent oxygen evolution reaction (OER) performance with an overpotential of 213 mV at 10 mA cm −2 . Furthermore, when used as a urea oxidation reaction (UOR), only an ultralow potential of 1.311 V at 10 mA cm −2 . More importantly, the assembled two-electrode drives overall water splitting and urea electrolysis with cell voltages of 1.62 V and 1.55 V at 10 mA cm −2 , respectively. This work provides insights into the preparation of electrocatalysts with multifunctional heterostructure arrays for hydrogen production.
