Structural analysis and in vitro fermentation characteristics of an Avicennia marina fruit RG-I pectin as a potential prebiotic

Jinwen Huang,Huiqi Wang,Huaqun Chen,Zidong Liu,Xuedong Zhang,Hao Tang,Shiying Wei,Wangting Zhou,Xinzhou Yang,Yonghong Liu,Longyan Zhao,Qingxia Yuan

Avicennia marina (Forssk.) Vierh. is a highly salt-tolerant mangrove, and its fruit has been traditionally used for treating constipation and dysentery . In this study, a pectin (AMFPs-0-1) was extracted and isolated from this fruit for the first time, its structure was analyzed, and the effects on the human gut microbiota were investigated. The results indicated that AMFPs-0-1 with a molecular weight of 798 kDa had a backbone consisting of alternating →2)-α-L-Rha p -(1→ and →4)-α-D-Gal p A-(1→ residues and side chains composed of →3-α-L-Ara f -(1→-linked arabinan with a terminal β-L-Ara f , →5-α-L-Ara f -(1→-linked arabinan, and →4)-β-D-Gal p -(1→-linked galactan that linked to the C-4 positions of all α-L-Rha p residues in the backbone. It belongs to a type I rhamnogalacturonan (RG-I) pectin but has no arabinogalactosyl chains. AMFPs-0-1 could be consumed by human gut microbiota and increase the abundance of some beneficial bacteria, such as Bifidobacterium , Mitsuokella , and Megasphaera , which could help fight digestive disorders . These findings provide a structural basis for the potential application of A. marina fruit RG-I pectic polysaccharides in improving human intestinal health.

Physical interaction between gallic acid and Monascus pigments enhances mechanical strength and pigment protection of pectin-gelatin hydrogel carriers

Chaoran Yang,Caoyu Guo,Boyan Gao,Marcin A. Kurek,Yuge Niu,Liangli Yu

Natural polymer hydrogels formed by polysaccharides and proteins can be protective carriers for Monascus pigment (MPs) in the food industry. In this study, in order to enhance the mechanical properties of the hydrogel and its ability to protect MPs, we introduced polyphenols as bifunctional components into the hydrogel carrier. This hydrogel carrier consisted of gallic acid (GA) grafted pectin (LPG) and gelatin. The results showed that the grafting of GA enhanced the affinity between MPs and pectin. A more compact network structure formed by pectin under the interaction of MPs with GA improved the mechanical properties of the hydrogel carriers. With the combination of GA with MPs, the gelation rate of the hydrogel carrier was accelerated, the gel strength increased from 550.07 g to 937.67 g and its water swelling rate decreased significantly from 447.87 % to 311.76 %. Moreover, the multiple pigment protection produced by the GA in the hydrogel greatly reduced the degradation rate of MPs under continuous light expose conditions. The findings provide new insights into developing hydrogel carriers for pigment protection with ideal mechanical properties.

Fabrication and characterization of citrus pectin/Dictyophora Rubrovalvata polysaccharide films: pH-cycle treatment and Penaeus vannamei shelf-life extension

Lansha Song,Keting Li,Lian Shuai,Sha Li,Xinyao Wei,Xingyi Chen,Lei Wang

Novel bioactive films were formulated via optional novel pH-cycle treatment using pectin with its weight basis of 0–8% polysaccharide from fungi Dictyophora rubrovalvata (DRP), known for its antioxidant and hypoglycemic properties. Characterizations were performed by microscopic techniques, Fourier transform infrared, X-ray diffraction, and thermal gravimetric analysis (TGA), as well as correlation analysis, etc. It was found that pectin integrated with DRP (especially those after pH-cycle treatment) exhibited significantly improved functionalities, including light barrier resistance, mechanical flexibility, thermal stability and antioxidant capacity. Notably, the elongation at break of film by incorporating 8% DRP into pectin via pH cycle reached the highest value of 20.25% as well as the strongest scavenging rates for DPPH∙ + and hydroxyl radicals at the respective rate of 75.52% and 85.71%, indicating the highest preservation efficacy on fresh Penaeus vannamei ( P <0.05). Correlational analysis offers theoretical information for the fabrication of pectin-based films as natural active packaging.

Eco-friendly antioxidant carboxymethyl cellulose /pectin films incorporating oregano essential oil and vanillic acid for tomato shelf-life extension

Qiaoqiao Liu,Yangcheng Xu,Hang Su,Zheliang Nie,Xin Shi,Bingcheng Ge,Shuyan Gao

The widespread use of conventional non-degradable packaging films for fruit preservation has led to serious environmental concerns, highlighting the urgent need for sustainable, biodegradable, and multifunctional packaging materials. Herein, we developed a carboxymethyl cellulose/pectin/oregano essential oil/vanillic acid (CMC/pectin/OEO/VA) film, in which hydrogen bonding between CMC and pectin hydroxyl groups enhances structural integrity and mechanical properties, further strengthened by VA, while phenolic compounds such as carvacrol and thymol in OEO provide antioxidant and antimicrobial functions. The optimized CMC/Pectin/OEO/VA0.06 (CPOV0.06) films demonstrate outstanding comprehensive performance, including excellent mechanical properties (tensile strength: 49.24 MPa), remarkable antioxidant capacity (DPPH radical scavenging rate: 85.87 %), significant antimicrobial effects ( E. coli : 6.33 mm, S. aureus : 6.00 mm), and complete biodegradability (soil assimilation time: 10 days). In addition, the blend film effectively prolonged the shelf life of cherry tomatoes up to 19 days under ambient storage conditions. This film offers a sustainable packaging approach, effectively addressing both food preservation and plastic pollution challenges.

Extraction of Citrus maxima (Burm.) peel oil and pectin via acidic deep eutectic solvents: Storage effects and RSM optimization

Manmin Zhang,Yan Huang,Hongzi Lai,Hongming Zhang,Guanle Li,Xiaoxiao Dai,Qihe Yang,Cuihua Liu,Lubin Zhang,Xiaonan Zhang

Landmark fruit Citrus maxima (Burm.) is highly productive, but by-products can cause environmental pollution. This study simulates the storage conditions of Citrus maxima at various stages after harvest and models the nutrient content and changes in different by-products. An organic acid deep eutectic solvent (OADES) was configured using six organic acids as hydrogen bond donor (HBD) in combination with choline chloride (ChCl); and the yields of essential oils and pectin were optimized using response surface methodology. The study showed that the OADES configured with oxalic acid as the HBD had the highest extraction rate. The results of electronic nose (E-nose) combined with GC-MS showed that the volatile odors of the essential oils were mainly from β-Myrcene (38 %), D-Limonene (37 %), and β-Pinene (4.3 %). When the raw materials were stored for more than 30 days, the yields of essential oils were decreased by 27.68 %, and the composition was mainly a decrease of 4.3 % in the proportion of sesquiterpenes and an increase of 7 % in alcohols. After 60 days of storage, the yield decreased by 27.68 %, the composition was mainly mono sesquiterpenoids decreased by 21.8 % and alcohols increased by 20.8 %. In addition, throughout the storage period (0–60 d), the pectin yield increased and then decreased, with a denser structure and more thermally stable and methylated pectin compared to the pre-extraction grapefruit peel. The research results not only provide new methods for green and energy-saving extraction technologies, but also offer valuable insights for optimizing the storage and reproduction practices in the citrus industry.

Protective Effects of Zein/Ferulic Acid (FA)–Pectin (PEC)/Chitosan (CS) Nanocomplexes on DSS-Induced Ulcerative Colitis in Mice

Yifei Guo,Xinyu Yu,Rongrong He,Jianfei Pei,Haiming Chen,Weijun Chen

Ferulic acid (FA) exhibits beneficial properties in ulcerative colitis (UC) pathogenesis, while sensitivity to the environment and enzymes limits its use in UC therapy. Therefore, this study aims to develop a colon-targeted nanocomplex delivery system using FA and investigate its protective effects and underlying regulatory mechanisms in UC mice. A novel Zein/FA–pectin (PEC)/chitosan (CS) nanocomplex was successfully fabricated in this study. Through systematic adjustment of the PEC/CS-to-Zein/FA ratio, optimal encapsulation efficiency (60.1%) and loading capacity (26.2%) were achieved. The characterized data indicated that hydrogen bonds, electrostatic interactions, and hydrophobic forces were the main driving forces maintaining the formation of the nanocomplexes, accompanied by alterations in the secondary structure of Zein. The Zein/FA–PEC/CS nanocomplexes demonstrated excellent thermal/storage particle size stability and exhibited both protective and sustained-release effects of FA during simulated gastrointestinal digestion. Furthermore, the results demonstrated that the nanocomplexes potentially alleviate UC by regulating inflammatory cytokines, oxidative stress, and gut microbiota. Compared to unencapsulated FA, the nanocomplexes have a better effect on alleviating UC symptoms. In summary, Zein/FA–PEC/CS nanocomplexes have promising prospects in alleviating colitis in UC mice.

Aggregation relations and flotation behavior of the depressant pectin on talc, chlorite, and serpentine and its mechanism of action

Xusheng Yang,Bo Feng,Ziming Wang,Longxia Jiang,Jefferson Bayoundoula

In this study, pectin was used as an environmentally friendly depressant to study its flotation behavior and aggregation effect on key magnesium silicate gangue minerals commonly found in nickel ore, such as talc, chlorite, and serpentine. The interaction mechanisms between pectin and minerals were explored using a series of techniques, including IC-FBRM particle size analysis, adsorption tests, scanning electron microscopy (SEM) tests, FTIR analysis, XPS analysis, and zeta potential measurements. Results from flotation experiments, IC-FBRM tests, and SEM observations indicate that under natural conditions, serpentine tends to aggregate with talc, but the three mixed minerals still exhibit floatability. At low dosages of pectin, serpentine and chlorite tend to aggregate together. Further increasing the pectin dosage reduces the recovery rate to 0%. FTIR and XPS analyses reveal that the –OH groups in pectin chemically interact with Mg atoms in chlorite. Zeta potential results show that pectin causes a significant downward shift in the zeta potential of chlorite.

Impact of fatty acid unsaturation levels on the physicochemical properties and oxidative/digestive stability of pectin/LDL-based oleogels

Mahmoud Abou-Elsoud,Ibrahim Khalifa,Mohamed Salama,Zuyue Li,Hamdy Zahran,Zhaoxia Cai,Dong Uk Ahn,Xi Huang

This work aimed to determine the impact of fatty acid composition on the physicochemical, oxidative, and digestive properties of emulsion-templated oleogels structured using pectin (PE) and egg yolk low-density lipoprotein (LDL). Four oils with varying degrees of unsaturation, including olive oil (OO), soybean oil (SO), linseed oil (LO), and fish oil (FO), were incorporated into PE/LDL-based oleogels. Results showed that oleogels formulated with polyunsaturated fatty acid (PUFA)-rich oils (FO and LO) exhibited higher viscosity (4856 and 4456 Pa s), hardness (745 and 721 g), oil-binding capacity (98.85 and 97.69 %), and thermal stability than others, due to stronger intermolecular bonds in the gel matrix. Oxidative stability assessments revealed that oleogelation significantly suppressed both primary and secondary lipid oxidation in PUFA-rich systems, with further improvements when BHA was added. The in vitro digestion showed that oleogels effectively reduced free fatty acids release and fat bioaccessibility compared to bulk oils, with FO-based oleogels showing the lowest. Furthermore, correlation analyses establish clear relationships between fatty acid profiles and oleogel performance, providing a scientific basis for optimizing formulations. These findings provide valuable insights for tailoring functional oleogels with improved oxidative stability and modulated lipid digestion.

Solubility and bioavailability enhancement of baicalin by modified citrus pectin: preparation, in silico, in vitro and in vivo evaluation

Wenxin Ma,Fangling Du,Tongcheng Xu,Xiuxiu Wang,Baorui Li,Hao Yue,Min Jia,Aizhen Zong

Baicalin, a flavonoid compound with various health benefits, faces significant limitations in its application due to poor solubility. Identifying a nontoxic, biodegradable, and readily available solubilizer is therefore critical for improving the solubility of baicalin and related analogues. This study aimed to evaluate the potential of citrus pectin and its acid-hydrolyzed derivatives to improve baicalin solubility, elucidating the underlying mechanisms of solubilization. Three fractions, MCP-1, MCP-2, and MCP-3, were obtained via partial acid hydrolysis followed by anion exchange chromatography. The results demonstrated that MCP-3, composed of 91.2 % galacturonic acid and 8.8 % rhamnose, exhibited the highest efficacy in enhancing baicalin solubility. Fourier transform infrared (FTIR) spectroscopy, X-ray diffraction (XRD), thermodynamic, molecular dynamics (MD) simulation confirmed that MCP-3 adopted a helical structure in solution, encapsulating baicalin within its helical cavity through hydrogen bonding between carboxyl and hydroxyl groups, as well as hydrophobic interactions and van der Waals forces. The MCP-3/baicalin complex exhibited good pH and temperature stability. Pharmacokinetic studies demonstrated that MCP-3 significantly enhanced the in vivo bioavailability of baicalin by 1.95-fold, and the complex exhibited low cytotoxicity. This study offers a promising strategy for the development of more effective flavonoid-based products.

Effects of nisin loaded chitosan-pectin nanoparticles on shelf life and storage stability of room temperature stored processed cheese

Biqi Liu,Qinggang Xie,Yang Liu,Shihang Li,Kai Dong,Yue Su,Yujun Jiang,Xinyan Yang,Ling Guo

Processed cheese faces challenges related to short shelf life and susceptibility to microbial contamination during room temperature storage. Nisin, a natural antimicrobial peptide used for food preservation, exhibits limited sustained activity and a narrow antimicrobial spectrum, making its enhancement essential. To address these issues, this study employed electrostatic self-assembly technology to develop chitosan-pectin nanoparticles loaded with nisin (CNP) to improve processed cheese stability at room temperature. The CNPs formed through electrostatic interactions and hydrogen bonding, exhibited ellipsoidal or polyhedral shapes. The encapsulation efficiency and loading capacity of CNPs were 73.93 % and 34.48 %. The CNPs provided sustained nisin release, significantly inhibiting the growth of Gram-negative bacteria such as Escherichia coli and Salmonella enterica . Notably, the addition of CNP, extended the shelf life of processed cheese by at least 21 days at room temperature, maintaining product stability. This study presents an innovative and efficient solution for processed cheese preservation at room temperature.

The Mechanism of Pectin-Mediated Thiamethoxam and Clothianidin Residues on Eggplant Peel and Enhanced Detection via Pectinase-Assisted Extraction

Yulun Chen,Linquan Wang,Aying Wen,Hang Yu,Yahui Guo,Yuliang Cheng,Weirong Yao,Shaofeng Yuan

Matrix binding poses a challenge to accurate pesticide residue detection in fruits and vegetables. This study investigated the distribution, binding status, and detection accuracy of neonicotinoid insecticides thiamethoxam (TMX) and clothianidin (CLO) in eggplant. To address the limitations posed by matrix binding, a pectinase-assisted extraction method was developed, which significantly enhanced the detectable levels of pesticides compared to the standard method and cellulase-assisted extraction. In situ Raman mapping and scanning electron microscopy revealed a strong correlation between the pectin distribution and the pesticide residues observed near the epidermal surface. Spectroscopic, nuclear magnetic resonance, and computational chemistry analyses demonstrated that TMX formed covalent bonds with pectin, while CLO interacted through hydrogen bonding. Notably, thermal processing promoted the formation of amide bonds, reducing detectable TMX levels. In summary, the optimized pectinase-assisted extraction method increased detectable TMX and CLO levels in positive samples by 25–40%, providing an effective approach for overcoming matrix binding effects.

Dual–responsive microgel beads prepared from pectin–stabilized emulsions for the controlled release of synbiotics to ameliorate ulcerative colitis

Xian He,Wenxin Dai,Shaoxian Liang,Yunyun Qin,Haoyue Liu,Kang Cheng,Mingyu He,Wanshui Yang,Xinsheng Qin

The benefits of synbiotics in ulcerative colitis are now supported by a growing amount of evidence. However, achieving its targeted delivery in vivo and reaching effective doses for therapeutic effects remains to be explored. To further explore the potential role of synbiotics in alleviating ulcerative colitis, this study designed a dual-response “egg–box” shaped microgel microsphere for targeted delivery and precise release of synbiotics. The internal “egg” structure was constructed using an apple pectin–zein hybrid particles (AZHPs) stabilized W 1 /O/W 2 double emulsions system, while a dense network formed by poly–L–lysine (PLL) and calcium alginate constituted the protective “box” shell. We investigated the physicochemical characteristics of W 1 /O/W 2 double emulsions prepared with different concentrations of AZHPs. The synbiotics–loaded W 1 /O/W 2 double emulsions were coated with PLL and calcium alginate to prepare microgel beads. In vitro digestion results demonstrated that the survival rate of probiotics within the microgel beads was significantly higher than that in a free state. Oral microgel beads alleviate colitis by modulating tight junction proteins (ZO–1, Claudin 1 and Occludin), reducing inflammatory factors (IL–1β, IL–6 and TNF–α), promoting the production of short–chain fatty acids. After treatment with EPMB, the number of Firmicutes in the intestinal flora increased while the number of Bacteroidetes decreased, and synbiotics intervention successfully restored the abundance of gut microbiota. This indicated that the loaded synbiotics “egg–box” shaped microgel beads offer a new option for the current treatment of colitis.
