Impact of ripening on the distribution of jackfruit polyphenol fractions and their inhibitory activities against metabolic enzymes

Liru Ma,Zhen Feng,Chao Zhang,Haode Chang,Wenjing Zhang,Quanmiao Zhang,Yihong Bao,Chunhe Gu

This study evaluated polyphenol fractions from unripe (J1), underripe (J2), and ripe (J3) jackfruit to explore stage-dependent industrial potential. Free (FP), conjugated (CP), and bound phenolics (BP) were isolated and assessed for antioxidant and enzyme inhibitory activities. Ripening significantly altered composition and bioactivity. CP antioxidant capacity and inhibition of α-glucosidase, pancreatic lipase, and xanthine oxidase increased progressively from J1 to J3, while FP and BP showed fluctuating trends. Ripe fruit (J3) exhibited the strongest CP-related bioactivities and a more diverse conjugated phenolic profile, whereas early stages had greater FP compositional complexity. Molecular docking supported these findings, identifying cyclomorusin as a potent ligand for α-glucosidase (−9.6 kcal/mol) and xanthine oxidase (−11.8 kcal/mol), and cyclokievitone for pancreatic lipase (−6.7 kcal/mol). These results demonstrate stage-specific functional differentiation and support targeted industrial utilization for health promotion.

Bioinspired hydrogel-engineered bioprosthetic valve via metal-polyphenol-polysaccharide coordinated assembly for thromboresistance, accelerated endothelialization, and immunoregulation

Cheng Zheng,Bangquan Wei,Lepeng Chen,Xueyu Huang,Yunbing Wang

Valvular heart disease (VHD) presents a growing global health burden, with bioprosthetic heart valves (BHVs) emerging as preferred clinical solutions due to superior hemodynamics and reduced anticoagulation requirements. However, BHVs suffer from structural valvular degeneration caused by cytotoxic glutaraldehyde residuals, thrombogenicity of exposed collagenous matrices, poor endothelialization and immune responses. To address these limitations, a metal-polyphenol-polysaccharide composited glycocalyx-mimetic hydrogel-engineered BHV (AACE-PP) was engineered through integrated hierarchical biomimetic design. A glycocalyx-mimetic hydrogel layer was welded onto BHVs via UV-initiated polymerization of methacrylated alginate, providing a hydrophilic barrier to shield thrombogenic surfaces, while EGCG-Cu metal-phenolic networks (MPNs) were incorporated to confer enzyme-mimetic and immunoregulation functionality. This dual-interfacial architecture synergistically combines physical shielding with biochemical regulation: the hydrogel coating effectively resists plasma protein adsorption, platelet adhesion and thrombosis, whereas the MPNs enable reactive oxygen species (ROS) scavenging, sustained nitric oxide release and immunoregulation. Biological evaluations demonstrated that AACE-PP significantly enhanced endothelialization, suppressed inflammatory activation, and reduced calcification compared to conventional BHVs. By emulating vascular homeostasis mechanisms through coordinated material integration, this biomimetic strategy establishes a multifunctional platform to improve BHV hemocompatibility and mitigate structural valvular degeneration.

Mitochondria-Targeted Metal Polyphenol Networks Inhibit Crystalline Nephropathy by Modulating SerpinE1 and Remodeling the Pathological Mineralization Microenvironment

Xue-Wu Chen,Jun Long,Xiao-Peng Zheng,Bang-Xian Yu,Guang-Zhao Wang,Wen-Qi Wu,Yong-Da Liu,Guo-Hua Zeng,Ling-Hong Huang,Xin-Yuan Sun

Crystalline nephropathy is driven by a self-perpetuating cycle in which crystal-induced tubular injury promotes further nucleation, growth, and aggregation, transforming the kidney into a pathological mineralization niche. Conventional therapies fail to disrupt this vicious loop, while nanomedicines are hindered by the glomerular filtration barrier (<10 nm), and natural antioxidants like protocatechuic acid (PCA) suffer from poor bioavailability. Here, we report a 7.3 nm mitochondria-targeted metal−polyphenol network (Fe-PCA@TPP) that overcomes both delivery and targeting challenges to directly intervene at sites of redox imbalance. Formed via PCA−Fe3+ coordination and functionalized with triphenylphosphonium (TPP), Fe-PCA@TPP efficiently traverses the glomerular barrier, accumulates selectively in mitochondria, scavenges free radicals broadly, restores redox homeostasis, and potently inhibits calcium oxalate (CaOx) crystal nucleation, growth, and aggregation. In vitro, Fe-PCA@TPP protects renal tubular epithelial cells from oxalate-induced oxidative stress by preserving mitochondrial bioenergetics and preventing apoptosis. In vivo, it preferentially accumulates in the kidney, reduces CaOx deposition, improves renal function, and downregulates markers of injury and inflammation. Mechanistically, Fe-PCA@TPP targets Plasminogen Activator Inhibitor Type 1 (SerpinE1) to block oxidative stress−apoptosis cascades, thereby remodeling the intrarenal pathological microenvironment. This work presents a size-compatible, mitochondria-targeted, and SerpinE1-specific nanoplatform that integrates crystal inhibition, redox regulation, and efficient renal clearance, offering a promising therapeutic strategy for crystalline nephropathy.

A biomimetic mitochondria-targeted cerium–polyphenol nanoplatform for modulating the cGAS–STING axis in acute kidney injury

Shichao Zhang,Quan Liao,Qidan Tan,Yuhan Xie,Tengfei Li,Zhouting Tuo,Rui Yao,Yuanjiong Qi,Shiyong Qi

A biomimetic ultrasmall cerium–polyphenol nanoplatform (CeSAB) is developed for precision therapy of acute kidney injury. • CeSAB enables efficient renal targeting and mitochondrial accumulation via albumin-mimicking surface engineering. • The nanoplatform modulates redox homeostasis and suppresses cGAS–STING–mediated inflammatory signaling. • CeSAB disrupts mitochondrial ROS-driven pathological cascades in AKI. • Significant renal protection with excellent biosafety and in vivo clearance is achieved in multiple AKI models. Acute kidney injury (AKI) remains a major clinical challenge due to the absence of effective therapeutic interventions. Mitochondrial dysfunction in proximal tubular epithelial cells (PTECs) induces excessive reactive oxygen species (ROS) accumulation, leading to mitochondrial DNA (mtDNA) release and activation of the cGAS–STING inflammatory pathway, thereby establishing a self-amplifying cycle of oxidative stress and inflammation. Herein, we develop a biomimetic ultrasmall cerium–polyphenol nanoplatform (CeSAB) to precisely disrupt this pathogenic cascade. CeSAB is constructed through coordination-driven assembly of Ce 3+ and salvianolic acid B (SAB), integrating polyphenol-derived antioxidant activity with the dynamic redox-buffering capability of cerium. Bovine serum albumin (BSA) functionalization endows CeSAB with albumin-mimetic renal-selective delivery through megalin-mediated uptake, enabling efficient mitochondrial localization. Upon mitochondrial accumulation, CeSAB restores redox homeostasis, preserves mitochondrial integrity, and prevents mtDNA release through SAB-mediated antioxidant activity, thereby suppressing cGAS–STING activation and downstream inflammatory responses. The Ce 3+ /Ce 4+ redox cycle further reinforces oxidative microenvironment regulation, while the pH-responsive coordination structure facilitates acid inflammatory microenvironment-triggered drug release, thereby alleviating oxidative stress and tissue hypoxia. In multiple AKI models, CeSAB significantly ameliorates renal dysfunction, oxidative injury, and inflammation with excellent biosafety. This work establishes a biomimetic mitochondria-targeted nanoplatform for interrupting ROS-driven inflammatory amplification, providing a promising approach for precision treatment of AKI. Polysaccharide-functionalized metal-polyphenol network-coated porous microspheres for improving the treatment of probiotics in ulcerative colitis

Congcong Xiao,Bohan Chen,Liming Gong,Jing Feng,MongHsiu Song,Xiaolong Li,Manru Zhang,Qiushuo Li,Mingji Jin,Liqing Chen,Zhonggao Gao,Qingyun Yang,Wei Huang

Probiotic therapy offers a new approach to the treatment of ulcerative colitis (UC), but oral drug delivery still faces challenges such as low gastrointestinal survival rates, insufficient intestinal retention, and poor inflammatory targeting. In this study, a layer-by-layer encapsulation probiotic delivery system ( HGM-L.re PM) was developed. Lactobacillus reuteri ( L.re ) was loaded onto poly-L-lactic acid porous microspheres, followed by in situ assembly of a tea polyphenol/Fe 3+ metal-polyphenol network (MPN) coating, and then layer-by-layer electrostatic deposition of an ethylene glycol chitosan/ hyaluronate (GCS/HA) polysaccharide gel layers via electrostatic interactions. The porous microspheres provide efficient probiotic loading and physical protection; the MPN coating confers transferrin-responsive degradability, enabling on-demand release at sites of inflammation; the GCS/HA outer layer synergistically forms a dense gel network via acid-triggered electrostatic cross-linking to resist gastric erosion, while the terminal HA moiety specifically binds to CD44 to enhance colonic adhesion. Compared to unencapsulated probiotics, HGM-L.re PM demonstrated significantly enhanced viability protection throughout all simulated stages of gastrointestinal digestion and achieved retention for up to 72 h in mice with colitis. In a DSS-induced acute UC model, this formulation effectively alleviated weight loss and colonic shortening, reduced mucosal damage, downregulated IL-6, IL-1β, and TNF-α levels, upregulated IL-10, restored the expression of mucin and tight junction proteins, and reshaped the gut microbiota composition by enriching beneficial bacteria such as Lactobacillus species. The underlying mechanism involves metabolic reorientation and stress resolution. The formulation also demonstrated good in vivo safety. This layer-by-layer encapsulated probiotic delivery platform offers a new strategy for enhancing the efficacy of oral probiotic therapy for UC. In this study, we developed an oral probiotic multi-layer-coated microsphere formulation capable of protecting the viability of L.re in the gastrointestinal tract, releasing the probiotic in response to an inflammatory environment in the colon, and significantly alleviating inflammation, repairing the intestinal barrier and regulating the gut microbiota in a mouse model of colitis. Multifunctional hydroxyapatite/tea polyphenol composite coatings on magnesium alloy surfaces for enhancing corrosion resistance and antibacterial properties

Jihan Liao,Xiaohong Li,Guoqiang Li

The deposition of hydroxyapatite coatings on the surface of magnesium alloys has proven to be an effective option for efficiently reducing their corrosion resistance and increasing their bioactivity. However, the challenge of implant-associated infections due to inadequate antimicrobial properties remains. In this work, a hydroxyapatite/tea polyphenol (HA/TP) composite coating was prepared on a magnesium alloy (AZ31B) substrate by a one-step hydrothermal method. The aim was to retard the corrosion rate of the magnesium alloy while synergistically combining the bioactivity of HA with the antimicrobial properties of TP to realize the multifunctionality of the material. The optimum reaction conditions for the precursor solution at pH 8.5 were determined by the morphology of the coating surface and the crystallographic phase analysis. Fourier transform infrared spectroscopy analysis experiments showed that chemical bonds were formed between HA and TP, which improved the stability of the structure. Antimicrobial experiments showed that the addition of TP exhibited broad-spectrum antibacterial activity against Escherichia coli and Staphylococcus aureus . Electrochemical experiments showed that the HA/TP coating reduced the corrosion rate of magnesium alloys to 0.349 mm/y. In addition, the coating exhibited excellent biocompatibility, promoted the proliferation of MC3T3-E1 osteoblasts and had the ability to rapidly induce mimetic mineralization. This study presents a novel strategy for the preparation of multifunctional coatings with both corrosion resistance, osteogenic activity and antimicrobial properties, providing a potential solution for degradable magnesium-based bone implants.

Preparation of gastrointestinal pH-responsive zein coated tea polyphenol-heparin hydrocolloids using for inflammatory bowel disease therapy

Hui Li,Shuangshuang Chen,Yumei Li,Zelin Sang,Zhenhua Chen,Xifan Mei,Xiuli Ren

In the pathogenesis of inflammatory bowel disease (IBD), overstimulation of inflammatory factors can trigger a coagulation cascade, increase the risk of intestinal micro-thrombosis and lead to microcirculation disorders. However, prevention of microcirculation pathways has not received enough attention. Heparin is commonly used in anticoagulant therapy, but oral delivery does not have an excellent anticoagulant effect. To improve the stability of heparin (HEP) in the gastrointestinal tract, zein/tea polyphenol nanospheres with a core-shell structure (EGNs@Z) were developed for oral administration of heparin (HEGNs@Z). The Zein shell has pH-responsive properties and is effective in preventing premature dissolution of heparin. At the same time, EGCG nanospheres (EGNs) play an anti-inflammatory role, jointly improve the vicious cycle between inflammation and microthrombosis. The results of SEM, TEM and FTIR showed that EGNs successfully encapsulated heparin and formed zein shells on the surface of microspheres with a thickness of 50-100 nm. In vitro simulated digestion experiments showed that zein shells prevented the breakdown of microspheres and heparin in a simulated gastric environment, whereas EGNs and HEP were slowly degraded and released in a simulated intestinal environment. Coagulation analysis showed that HEGNs@Z was effective in delaying clotting time. A mouse model of acute colitis has also shown that HEGNs@Z robust promotes colonic epithelial regeneration, inhibits malignant microcirculation, and reduces bleeding risk. These findings reveal that this orally bioavailable multifunctional material may provide a novel, effective and convenient treatment for inflammatory bowel disease.

Enabling multifunctional bio-based waterborne polyurethane through polyphenol-mediated assembly of zirconium phosphate nanoplatelets for patternable coating

Jiabo Shi,Xiaoyi Wu,Li Sheng,Meng Chen,Lei Shi,Yi Zhang,Jingyi Yang,Shumin Mao,Qiang Liu,Zhijun Zhou

Sustainable bio-based waterborne polyurethane (WPU) has been widely used in a wide range of industrial applications owing to its favorable environmental safety and excellent properties. However, achieving superior mechanical properties and favorable multifunctionalities in bio-based WPU remains a considerable challenge. In this work, we proposed a facile and effective interfacial engineering strategy which was synergistically enabled via phosphoric acid-assisted exfoliation and polyphenol-mediated assembly of zirconium phosphate nanoplatelets (ZrP NPs) to prepare multifunctional castor oil-based supramolecular WPU composites for patternable leather coating. Tunable ZrP based supramolecular assemblies with the weight ratio of TA to pZrP NPs of 2.0 wt.% and pH > 5.0 were fabricated to manipulate the assembly of the WPU molecules. Active TA molecules bound on the interfaces of ZrP NPs can function as organic junctions through the formation of hydrogen-bonding interactions between the phenolic groups of TA molecules and the functional groups of WPU molecules. Benefiting from this hydrogen-bonding-driven co-assembly, the resultant supramolecular WPU composite films not only exhibited excellent transparency and reinforced mechanical strengths, thermal stability, and flame-retardant properties, but also possessed photoluminescent properties suitable for patternable leather coating. For example, the peak heat release rate (PHRR) was decreased from 389.45 kW/m 2 to 199.75 kW/m 2 , and the total heat release (THR) was from 15.49 MJ/m 2 to 13.58 MJ/m 2 . We envision that this study contributes to the development of sustainable multifunctional bio-based WPU coatings with significant potential for advanced applications in information encryption and displays, etc.

Metal-polyphenol networks hardened coacervate microcapsules for encapsulation of piperine: Crosslinking mechanism, stability, and in vitro digestion

Maye Li,Yuanyang Song,Mi Tian,Xinyang Huang,Jing Xu,Zengwang Guo,Zhongjiang Wang,Lechuan Wang,Zhaoxian Huang,Lianzhou Jiang

Although metal polyphenol networks (MPNs) have been widely investigated in the field of multifunctional surface modification, the exploration of their application in the hardening of microcapsules is relatively scarce. In this study, piperine (PIP)-loaded microcapsules were hardened and modified using MPNs formed by tannic acid (TA) and Fe 3+ (TA-Fe 3+ ) to enhance the stability of the microcapsules and the bioaccessibility of PIP. The results showed that TA-Fe 3+ constructed a network-modified coating with a supramolecular structure on the surface of microcapsules. The TA-Fe 3+ -hardened microcapsules exhibited excellent environmental stability and illustrious encapsulation efficiency (97.7 %). In vitro digestion demonstrated that TA-Fe 3+ delayed the release of PIP in the gastrointestinal tract and improved the bioavailability of the microcapsules. This research established a vital basis for the creation of novel microcapsule systems based on MPNs and demonstrated the potential for applications in food science for the delivery and controlled release of bioactive ingredients.

Improvement of the Structure and Antioxidant Activity of Protein–Polyphenol Complexes in Barley Malts Using Roasting Methods

Guozhi Wu,Huiting Lin,Yongsheng Chen

Proteins and polyphenols are important components in barley malt. During the roasting process of barley malt, proteins and polyphenols interact and influence each other, ultimately altering the nutritional profile and functional properties of barley malt. In this research, polyphenol-free proteins and protein–polyphenol complexes were extracted from barley malt subjected to varying degrees of roasting. The antioxidant activity of protein–polyphenol complexes was assessed by ABTS, FRAP, and ORAC assays. The structural characteristics of the proteins were examined through UV, FL, CD, FTIR, and SEM. We found that roasting enhances the solubility of globulin, prolamin, and glutenin and facilitates the binding of these proteins with polyphenols. Conversely, the impact of roasting on albumin exhibits a trend opposite to that observed in the other three proteins. The antioxidant activity of protein–polyphenol complexes was significantly higher than that of polyphenol-free proteins. Additionally, the microenvironment of the amino acid residues of the four proteins exhibited increased polarity following the roasting process, and the structural conformation of albumin, globulin, and glutelin transitioned from an ordered to a disordered state. Our results indicate that roasting enhances the antioxidant activity of protein–polyphenol complexes by altering the secondary and tertiary structures of these proteins, thereby exposing more hydrophobic side-chain groups inside the proteins and offering more binding sites for polyphenols.

Key volatile off-flavor compounds identification in chicken meat and deodorizing effects of polyphenol and spice extracts

Houqi Ning,Aiping Yang,Wenjing Zhou,Songling Li,Yifang Tang,Wenbo Ma,Ping Liu

This study identified key volatile off-flavor compounds in chicken meat. Flavor compounds were analyzed using headspace solid-phase microextraction (HS-SPME), two-dimensional gas chromatography–mass spectrometry (GC × GC–MS) and gas chromatography–mass spectrometry-olfactometry (GC-O-MS). Eighteen volatile flavor compounds (odor activity values ≥1, flavor intensity ≥2) in various chicken meat were found, and green leg chicken has the highest number of selected flavor compounds (14). These 14 compounds were individually incorporated into a flavorless chicken matrix and quantitatively analyzed by GC × GC–MS. Thirteen key flavor compounds were determined and further validated through recombination and omission experiments. Six key off-flavor compounds, 1-octen-3-ol, heptanal, hexanal, nonanal, decanal, dodecanal, were identified. Besides, polyphenol and spice extracts significantly reduced off-flavors in chicken, particularly tea polyphenol. It decreased thiobarbituric acid (TBA) levels and off-flavor values of chicken meat by 90.6 % and 55.6 %, as well as the concentration of 6 key off-flavor compounds ( p  < 0.05).

Metal-polyphenol network-hardened coconut protein-carboxymethyl cellulose complex coacervates: Fabrication, characterization, environmental stability, and release kinetics

Maye Li,Sha Luo,Chunmei Cheng,Xun Liang,Jing Xu,Zengwang Guo,Zhongjiang Wang,Lechuan Wang,Zhaoxian Huang,Lianzhou Jiang

Metal-polyphenol networks (MPNs), as a novel surface structure modifier, whether it can have a hardening effect on complex coacervate microcapsules is still unknown. In this work, piperine (PIP)-loaded coconut protein (CCP)-carboxymethyl cellulose (CMC) complex coacervates (CP) microcapsules were hardened using tannic acid-Fe 3+ networks (TA-Fe 3+ ) to improve the stability of microcapsules. The results showed that the optimal coacervation conditions for CCP-CMC were at pH = 3.2 and a CCP: CMC ratio of 2:1 ( w /w). TA-Fe 3+ formed a network-decorated coating with a supramolecular structure on the CP surface. TA:Fe 3+ ratios significantly affected the structural cohesion of TA-Fe 3+ -hardened piperine-loaded CCP-CMC complex coacervates (TF) and thus the hardening effect. At low Fe 3+ levels, TF formed a dense and uniform structure, and at high Fe 3+ levels, the TF structure became looser. Besides, 2TA-1Fe 3+ -CP exhibited excellent encapsulation efficiency, desirable stability, and favorable antioxidant ability. After long-term storage, light irradiation, and pH treatment, the retention of PIP in 2TA-1Fe 3+ -CP was increased by 119 %, 133 %, and 111 % compared to CP, respectively. Moreover, the release kinetics results proved that TA-Fe 3+ delayed the release of CP in the food simulants. This study provided some valuable information for exploring a novel hardening microcapsule approach based on the complex coacervation.
