A Novel Gelatin-Based Sustained-Release Molluscicide for Control of the Invasive Agricultural Pest and Disease Vector Pomacea canaliculata

Jie Wang,Yuntian Xing,Yang Dai,Yingnan Li,Wenyan Xiang,Jianrong Dai,Fei Xu

Pomacea canaliculata, one of the 100 most destructive invasive species in the world, and it is an important intermediate host ofAngiostrongylus cantonensis. The molluscicides in current use are an effective method for controlling snails. However, most molluscicides have no slow-release effect and are toxic to nontarget organisms. Thus, these molluscicides cannot be used on a large scale to effectively act on snails. In this study, gelatin, a safe and nontoxic substance, was combined with sustained-release molluscicide and was found to reduce the toxicity of niclosamide to nontarget organisms. We assessed the effects of gelatin and molluscicide in controllingP. canaliculatasnails and eggs. The results demonstrated that the niclosamide retention time with 1.0% and 1.5% gelatin sustained-release agents reached 20 days. Additionally, the mortality rate ofP. canaliculataand their eggs increased as the concentration of the niclosamide sustained-release agents increased. The adult mortality rate ofP. canaliculatareached 50% after the snails were exposed to gelatin with 0.1 mg/L niclosamide for 48 h. The hatching rate ofP. canaliculatawas only 28.5% of the normal group after the treatment was applied. The sustained-release molluscicide at this concentration was less toxic to zebrafish, which means that this molluscicide can increase the safety of niclosamide to controlP. canaliculatain aquatic environments. In this study, we explored the safety of using niclosamide sustained-release agents with gelatin againstP. canaliculata. The results suggest that gelatin is an ideal sustained-release agent that can provide a foundation for subsequent improvements in control ofP. canaliculata. Keywords:invasive species;biological material carrier for sustained drug;Pomacea canaliculatacontrol;environmental safety

Polyphenol–Metal Functionalized Hydrogel Dressing with Sustained Release, Antibacterial, and Antioxidant Properties for the Potential Treatment of Chronic Wounds

Zhiwen Zeng,Cuiping Guo,Daohuan Lu,Zhijie Geng,Dating Pei,Shan Yu

Infection and chronic inflammation caused by oxidative stress are the main challenges in chronic wound healing. In this study, a silk fibroin/kappa-carrageenan ( k CA)-based hydrogel functionalized with epigallocatechin-3-gallate (EGCG) and Cu 2+ is fabricated for chronic wound healing. Polyphenol–metal functional complexes and ionic-covalent entanglement networks are prepared by introducing Cu 2+ . Cu 2+ coordinates with k CA to enhance the mechanical properties and chelates with EGCG to form an EGCG-Cu 2+ complex that continuously releases EGCG. The sustained release of EGCG enhances the antibacterial properties by synergizing with the effects of Cu 2+ . Furthermore, the prepared hydrogel exhibits a potent ability to scavenge excessive intracellular reactive oxygen species, controllable antioxidant bioactivity, and reliable biocompatibility. Ultimately, the resulting multifunctional hydrogel dressing provides a potential and effective strategy for chronic wound care management with increased mechanical strength and antibacterial and antioxidant properties.

Effects of different pore structures on loading and sustained-release of mitomycin C by hollow mesoporous Fe(0)@mSiO2

Chang Ziling,Li Huafei,Yang Yuxiang,Yuan Hongming,Huang Yan,Yao Pingping,Ni Chaoying

Herein, we developed the dual-function template method to fabricate hollow magnetic nano-spheres (denoted as HMFe–Si–C n , n = 16, 18) with a mesoporous shell and hollow interior structure using alkyl chain trimethoxysilane templating. The shorter chain template directed formation of HMFe–Si–C 16 with size of 119 nm, having disordered inkbottle type mesopores and saturation magnetization of 50.01 emu/g higher than that of HMFe–Si–C 18 with cylindrical type mesopores. In addition, the HMFe–C n loaded with MMC diaplays a pH and magnetic dual responsive drug release behavior. By contrast, Mitomycin C (MMC) loading efficiency of HMFe–Si–C 16 was higher owing to the fact that the pore size, surface area, and pore volume of HMFe–Si–C 16 were larger than those of HMFe–Si–C 18 . Besides, MMC loaded HMFe–Si–C n hollow spheres showed a clear pH-dependent drug release behavior, having a higher release rate in acidic environments of pH 5.7. For the pH 5.7 and 7.4 release, the release kinetic for HMFe–Si–C 16 –MMC composites follows pseudo-first-order attributable to its special pore structure. With Fe(0) as a core, HMFe–Si–C n is distinguished for higher magnetic properties, and it is more conducive to magnetic targeted treatment of cancer cells and can release drugs through external magnetic field. In addition, HMFe–Si–C n has lower cytotoxicity and better biocompatibility. For this reason the inner cavity of HMFe–Si–C 16 could be labeled with radioisotope 99 Tc m to study the magnetic targeting distribution of HMFe–Si–C 16 in vivo, and its cytotoxicity against in vitro HeLa cells was also studied. These results indicate the potential of HMFe–Si–C 16 in the magnetic targeted drug delivery system.

Enhanced wound healing in diabetic mice by hyaluronan/chitosan multilayer-coated PLLA nanofibrous mats with sustained release of insulin

Xiang Gao,Rui Huang,Yanpeng Jiao,Thomas Groth,Wufeng Yang,Chenlin Tu,Hailong Li,Fan Gong,Jiaqi Chu,Mingyan Zhao

The healing of wounds in diabetes mellitus patients remains a critical clinical challenge globally. To promote healing, the development of bioactive dressings that not only provide physical protection but also supply a favorable microenvironment for healing at the wound interface is urgently required. Herein, Michael addition reaction was first used for intrinsic-crosslinking of bioactive maleilated chitosan/thiolated hyaluronan (mCH/tHA) multilayer coatings on poly (L-lactic acid) (PLLA) nanofibrous mats (P-mCH/tHA) for a sustained release of insulin (IN) and compared that with pure PLLA and PLLA mats coated with native CH/HA (P-CH/HA) multilayers. The physicochemical studies showed that P-mCH/tHA have higher tensile strength, an accelerated degradation rate, more antibacterial activity and sustained release of insulin when compared to P-CH/HA and pure PLLA mats. Studies on bioactive properties of PLLA mats coated with either mCH/tHA or CH/HA indicated an improved adhesion, spreading and proliferation of mesenchymal stem cells on P-mCH/tHA_IN in comparison to the other PLLA mats. Most interestingly, in vivo investigations with a diabetes mouse model demonstrated fastest wound closure when using P-mCH/tHA_IN among all groups with facilitated re-epithelization, angiogenesis, and increased collagen deposition in the wounds, which makes this coating in combination with IN promising for development of new bioactive wound dressings.

Exploitation of enrofloxacin-loaded docosanoic acid solid lipid nanoparticle suspension as oral and intramuscular sustained release formulations for pig

Yanfei Tao,Fei Yang,Kuiyu Meng,Dongmei Chen,Yujuan Yang,Kaixiang Zhou,Wanhe Luo,Wei Qu,Yuanhu Pan,Zonghui Yuan,Shuyu Xie

In our previous study, enrofloxacin-loaded docosanoic acid solid lipid nanoparticles (SLNs) could be effectively delivered to cells in vitro. In this study, its properties and exploitation as possible oral and intramuscular sustained release formulations for pigs were studied after being made into suspension. The re-dispersed time and sedimentation rate of the nanosuspension were 55 s and 1, respectively. It showed good stability when stored away from light and sustained release in pH = 7.4 PBS buffer. The suspension exhibited no irritation at the injection site and good palatability. Compared with commercial injection and soluble powder, the nanosuspension increased the bioavailability of enrofloxacin by 1.63 and 2.38 folds, and extended the mean residence time (MRT) of the drug from 11.27 and 12.33 to 37.76 and 35.15 h after intragastric and intramuscular administration, respectively. These results suggest that docosanoic acid SLN suspension (DAS) might be a promising oral and intramuscular sustained release formulation to enhance the pharmacological activity of enrofloxacin.

Effects of Dietary Microcapsule Sustained-Release Sodium Butyrate on the Growth Performance, Immunity, and Gut Microbiota of Yellow Broilers

Zhenglie Dai,Xiuxi Wang,Yulan Liu,Jinsong Liu,Shiping Xiao,Caimei Yang,Yifan Zhong

Simple SummaryThe supplementation of sodium butyrate (SB) has proven to be an effective way to provide extra butyrate to improve the homeostasis and gut health of broilers and consequently enhance their performance. However, the rapid release of SB in the gastrointestinal system leads to lower efficiency in poultry production. Sustained-release sodium butyrate (MSSB) has shown potential, with a lower dosage required and a better effect on performance. A comparison of rapid SB absorption and MSSB in terms of performance, serum parameters, and gut microbiota is poorly demonstrated in the literature, especially in yellow broilers, a primary source of the live poultry trade in China. Our study found that, overall, MSSB contributes to improved performance, immunity, and gut health. Specifically, the supplementation of MSSB lowered the feed:gain ratio and increased the IgA, isovalerate, and villus height of yellow broilers. Alterations in gut microbiota, including the predicted composition and function, have also been observed, namely, inClostridiaUCG-014,BacilliRF39, andOscillospiraceaeUCG-005. The mediation analysis revealed a causal effect betweenClostridiaUCG-014 in the gut and serum IgA, with the tryptophan metabolism appearing to be a key mediator in this relationship. This study shows that MSSB can improve the performance and enhance the immunity of yellow broilers. Our results are significant because they reveal the beneficial effects together with the potential mechanism of MSSB in yellow broilers.AbstractThe beneficial effects of butyric acid in poultry production are well documented, while the relationship between sodium butyrate (SB) and microcapsule sustained-release sodium butyrate (MSSB), especially in yellow broilers, remains poorly investigated. This study was designed to elucidate the function as well as the potential mechanisms of SB and MSSB in enhancing health in yellow broilers. In total, 360 one-day-old yellow broilers were allocated to three treatment groups. The control group (CON) received a basic diet, while the SB group was provided with 1000 mg/kg of sodium butyrate (SB), and the MSSB received microcapsule sustained-release sodium butyrate (MSSB), all over a period of 56 days. Compared to the CON group, the dietary supplementation of both SB and MSSB showed a lower feed:gain ratio (p< 0.01). No significant (p> 0.05) difference in antioxidant capacity was observed between the three groups. We observed significantly higher levels (p< 0.05) of immunoglobulins and a reduction in concentrations in both the SB and MSSB groups compared to the CON group. Furthermore, both SB and MSSB induced alterations in the diversity, structure, and function of gut microbiota. MSSB demonstrated even more pronounced beneficial effects than SB, particularly in regard to the serum IgA level (p= 0.05), cecal isovalerate concentration (p< 0.05), and villus height (p< 0.01). The sequencing of the gut microbiota revealed that MSSB led to a significant increase in the relative abundance ofClostridiaUCG-014,BacilliRF39, andOscillospiraceaeUCG-005. Predictions of bacterial function indicated changes in KEGG pathways, including an enrichment of tryptophan metabolism (ko00380), and a reduction in fructose and mannose metabolism (ko00051), chloroalkane and chloroalkene degradation (ko00625), and naphthalene degradation (ko00626) in yellow broilers fed with MSSB. Among these, the mediation analysis revealed a causal effect between theClostridiaUCG-014 in the gut and serum IgA, with tryptophan metabolism being a key mediator in this relationship. Our results suggest that dietary MSSB can improve the growth performance, immunity, and gut microbiota of yellow broilers. MSSB increased the abundance ofClostridiaUCG-014 and activated the tryptophan metabolism pathway (ko00380), contributing to IgA levels in yellow broilers through this mechanism. Keywords:microcapsule sustained-release sodium butyrate;yellow broiler;gut health;immunity;gut microbiota

Apigenin@ZIF-8 with pH-responsive sustained release function added to propolis-gelatin films achieved an outstanding antibacterial effect

Lihan Wang,Shasha Cheng,Kaiwen Qin,Xinyan Yang,Hao Wang,Chaoxin Man,Qianyu Zhao,Yujun Jiang

Packaging films containing antibacterial substances play a crucial role in controlling contamination caused by food-borne pathogens. Herein, apigenin (APG) was encapsulated in zeolitic imidazolate frameworks 8 (ZIF-8) to achieve a synergistic antibacterial effect and prolonged drug release, aiming to extend the shelf life of food products. Under acidic condition, the release rate of APG reached 73.70 % after 12 h. Subsequently, propolis-gelatin (PG) films were prepared for food packaging. The addition of APG@ZIF-8 enhanced the hydrophobicity of PG films, making them more suitable for food packaging. However, the composite film exhibited reduced damage resistance and flexibility. The composite films demonstrated 100 % inhibition against Staphylococcus aureus and Escherichia coli under appropriate amount of APG@ZIF-8. The composite film coating significantly reduced the bacteria count on apple surfaces by 99.95% during storage, while maintaining their appearance. This study provides an effective way to develop novel antibacterial composite films.

Efficient synthesis strategy of folate-modified carboxymethyl chitosan/CaCO3 hybrid nanospheres and their drug-carrying and sustained release properties

Jianfeng Guo,Juhui Liu,Haoran Qie,Feng Zhao,Catherine Hui Niu

Folate-modified carboxymethyl chitosan (FCMC) was made by folate acid as targeted group and attaching folate to carboxymethyl chitosan, and then, targeted FCMC/CaCO3 hybrid nanosphere were formed by self-assembly of calcium carbonate in FCMC solution. The physicochemical properties of the nanospheres were investigated by Fourier transform infrared spectroscopy, X-ray diffraction analysis, Brunauer–Emmett–Teller measurement and thermogravimetric analysis (TGA). The results showed that the FCMC/CaCO3 hybrid nanospheres were composed of calcite, vaterite and polysaccharides, and the content of organic compounds was 12.17%. Also, the structure performance of the hybrid nanospheres was analyzed. Besides, the effects of the hybrid nanospheres on the encapsulation efficiency, the drug loading content and the release behavior were also analyzed with the metformin (MET) as a model drug. Scanning electron microscope, Zeta potential analysis and UV–Vis were used to characterize the hybrid nanospheres. Under the conditions of FCMC/Ca2+ molar ratio of 4: 1 and reaction for 24 h, the achieved results showed that the spherical aggregates with regular morphology were obtained and the average particle size of the nanospheres was 207 nm. The specific surface area of the hybrid nanosphere is 27.06 m2·g−1 and the average pore diameter of the sample is 3.84 nm, indicating the presence of mesoporous structure in the sample. This mesoporous structure can supply potential space for adsorption of anticancer drugs. Additionally, the surface charge of the nanoparticles was positive and the entrapment efficiency was 83.32%. The hybrid nanospheres have a capability of effective pH-sensitivity controlled drug release. All the drug loaded hybrid nanospheres successfully sustained the release of MET at pH 7.4, only about 44.58% of the drug released in 6 days. While under acidic condition (pH 5.0) drug release was significantly accelerated, being over 98.85% of the drug released. The hybrid nanospheres demonstrated an excellent smart drug delivery behavior.

Long-term antibacterial composite via alginate aerogel sustained release of antibiotics and Cu used for bone tissue bacteria infection

Xia-Xiao Wu,Yu Zhang,Tian Hu,Wei-Xiong Li,Zeng-Lin Li,Han-Jian Hu,Shui-Rong Zhu,Wen-Zhao Chen,Chu-Song Zhou,Gang-Biao Jiang

Bone related-bacterial diseases including wound infections and osteomyelitis (OM) remain a serious problem accompanied with amputation in most severe cases. In this work, we report an exceptional effective antibacterial alginate aerogel, which consists of tigecycline (TGC) and octahedral Cu crystal as an organo-inorganic synergy platform for antibacterial and local infection therapy applications. The alginate aerogel could greatly prolong the release of copper ions and maintain effective antibacterial concentration over 18 days. The result of in-vitro experiments demonstrated that the alginate aerogel has an exceptional effective function on antibacterial activity . Cytotoxicity tests indicated that the alginate aerogel has low biological toxicity (average cell viability >75%). These remarkable results suggested that the alginate aerogel exhibits great potential for the treatment of OM, and has a prosperous future of application in bone tissue engineering.

Nanocellulose hyperfine network achieves sustained release of berberine hydrochloride solubilized with β-cyclodextrin for potential anti-infection oral administration

Lin Xiao,Abishek Jung Poudel,Lixia Huang,Yang Wang,Ahmed M.E. Abdalla,Guang Yang

Berberine hydrochloride (BBH) has been used to treat diarrhea and other gastrointestinal diseases, however its therapeutic efficacy is compromised because of poor aqueous solubility and dissolution. In this work, BBH was solubilized with β-cyclodextrin (β-CD) in aqueous solution through formation of the BBH/β-CD inclusion complex (IC), which was confirmed by the combination of different techniques including FT-IR, XRD , DSC , 1 H NMR and 1 H NOESY. The aqueous solubility of BBH at 25 °C was increased by ca. 102% in the presence of 16 mM β-CD. BBH/β-CD IC-loaded bacterial cellulose (BC) hydrogels (denoted as BC/IC) were prepared by physical absorption method, resulting in higher drug loading capacity (DLC) than BC/BBH hydrogels. In vitro drug release showed that sustained drug release was achieved at different pH conditions simulating the gastrointestinal fluids by BC/IC hydrogels due to the hyperfine network of BC matrix. Furthermore, in vitro anti-bacterial test demonstrated the BC/IC hydrogels induced effective bacterial inhibition. Together with the good biocompatibility and edibility of the BC matrix, these BC/IC hydrogels appear to be promising candidates of oral administration medicine against gastrointestinal infections.

Development of ovalbumin-pectin nanocomplexes for vitamin D3 encapsulation: Enhanced storage stability and sustained release in simulated gastrointestinal digestion

Chuyue Xiang,Jian Gao,Haoxin Ye,Gerui Ren,Xiangjuan Ma,Hujun Xie,Sheng Fang,Qunfang Lei,Wenjun Fang

Vitamin D 3 (VD 3 ) is a fat-soluble vitamin and easily degraded under acidic conditions, which will greatly reduce its bioavailability. The use of complexes formed by protein-polysaccharide to control release and protect active compounds has become a feasible way in the food field. In this work, ovalbumin (OVA) and high methoxyl pectin (PEC) were applied to fabricate OVA-PEC nanocomplexes to encapsulate and protect VD 3 . The encapsulation efficiency of VD 3 by OVA-PEC complexes could reach to 96.37%, and the storage stability of VD 3 was obviously improved after encapsulation. The in vitro simulated gastrointestinal experiments combined with SEM and SDS-PAGE were employed to observe the digestive behavior of OVA-PEC-VD 3 nanocomplexes. A small amount of VD 3 is released from OVA-PEC-VD 3 nanocomplexes in simulated gastric fluid, while a large amount of VD 3 is released in simulated intestinal fluid. The sustained release kinetic of VD 3 from OVA-PEC-VD 3 nanocomplexes was consistent with the law of Higuchi model. The preliminary molecular mechanism for VD 3 encapsulation by OVA-PEC complexes was also investigated, and the formation of OVA-PEC-VD 3 nanocomplexes is ascribed to the electrostatic interactions, hydrogen bonding and hydrophobic interactions among OVA, PEC and VD 3 . The results demonstrated that protein-polysaccharide complexes can effectively encapsulate VD 3 and achieve the goal of sustained release in the simulated gastrointestinal tract, which have potential applications in food and medicine.

Development, Optimization, and Evaluation In Vitro/In Vivo of Oral Liquid System for Synchronized Sustained Release of Levodopa/Benserazide

Qu Yang,Lai Wen-Long,Xin Yuan-Rong,Zhu Fei-Qin,Zhu Yuan,Wang Li,Ding Yue-ping,Xu Ying,Liu Hong-Fei

To enhance efficiency, convenience, and safety of Parkinson’s disease (PD) treatment for geriatric patients, an advanced suspension of Levodopa/Benserazide hydrochloride (LD/BH) has been prepared by cation-exchange resin and used to synchronize sustained release of LD and BH by optimizing coating parameters and prescription. For the purpose, LD and BH were immobilized on the surface of cation-exchange resin, respectively. Based on HPLC results, the cation-exchange resin showed high loading capacity. The studies on drug loading mechanism indicated that both drugs were immobilized by electrostatic interaction rather than physical adsorption. After PEG modification, pretreated drug-resin complexes were coated by emulsion–solvent evaporation method. In order to control drug release in a sustained manner, coating parameters of drug-resin microcapsules were optimized respectively by single-factor analysis. Further, coating prescription of the microcapsules was optimized to synchronize sustained release of LD and BH in vitro by orthogonal design. Utilizing optimal LD-resin microcapsules and BH-resin microcapsules, LD/BH suspension, containing both of them, was prepared by an optimal formulation and characterized by accelerated test and pharmacokinetic study in vivo . The accelerated test confirmed high stability of LD/BH suspension. According to pharmacokinetic results in vivo , in contrast with LD/BH commercial tablets, LD/BH suspensions did not only synchronize sustained release of both drugs but also show good bioequivalence. As LD/BH sustained release suspension can synchronize sustained release of multiple active ingredients by oral administration, the suspension presents promising oral dosage forms for geriatric patients with PD. An advanced Levodopa/Benserazide hydrochloride (LD/BH) suspension, prepared by cation-exchange resin and optimized microencapsulation, synchronizes sustained releases of LD and BH in vivo to benefit Parkinson’s disease treatment for geriatric patients.
