Dual-emission fluorescence biosensing of vancomycin based on AIEgen–peptide conjugates and aptamer-modified Au nanoclusters

Fangya Mu,Junqing He,Fang Fan,Guoyue Shi

Precise doses of antibiotics are necessary to prevent bacterial drug resistance . Although fluorescent sensors are promising for quantitative analyses of antibiotics, improvements in feasibility, selectivity , and sensitivity are needed. In this study, a dual-emission fluorescence biosensor platform was developed for simple, selective, and sensitive determination of vancomycin (Van) based on a peptide conjugated with blue-emitting aggregation-induced emission luminogens (AIEgen) and aptamer-modified red-emitting gold nanoclusters (AuNCs-apt). The peptide and aptamer together recognized Van with high affinity, thus changing the fluorescence intensity at 470 nm and 650 nm, respectively. This platform displayed excellent linear correlation between the fluorescence response and a Van concentration ranging 0.01–100 μg mL −1 , and the limit of detection (LOD) was 2.79 ng mL −1 . In addition to the ability to accurately distinguish Van from glycopeptide antibiotics , the newly developed biosensor allowed for naked-eye detection of 1 μg mL −1 Van. These results and those of serum samples and microdialysate samples support the application of this newly developed method for Van monitoring and clinical diagnosis.

Joint Synovial Fluid Metabolomics Method to Decipher the Metabolic Mechanisms of Adjuvant Arthritis and Geniposide Intervention

Xiang Zhan,Huan Wu,Hong Wu

Rheumatoid arthritis (RA), a chronic systemic autoimmune disease, is mainly characterized by joint lesions and permanent loss of joint function. To discover the metabolic characteristics of RA and the underlying mechanisms in treatment with geniposide (GE), untargeted metabolomic analysis based on hydrophilic interaction liquid chromatography coupled to high-resolution mass spectrometry (HILIC–HRMS) was performed using the joint synovial fluid samples from adjuvant arthritis (AA) rats. Microdialysis (MD) was utilized to collect the dialysate samples precisely from the articular cavity of AA rats. Multivariate statistical analysis was then conducted to discover the metabolite changes induced by AA and to differentiate GE-related biomarkers. The mass spectrometry data are available on the Chorus website (https://chorusproject.org/pages/index.html) with the data set identifier 1680. The results showed that 20 metabolites differed significantly between AA rats and normal rats. GE treatment recovered the altered levels of the 13 metabolites mentioned above, such as palmitoylethanolamide (PEA), Cer (d18:0/22:0), and PC (18:1(11Z)/16:1(9Z)), and normalized glycerophospholipid metabolism. As evidenced by western blotting, the changes in PEA levels adjusted by GE were associated with the down-regulated expression of N-acylethanolamine-hydrolyzing acid amidase (NAAA) in synovial tissues. Taken together, the elucidation of metabolic changes of joint synovial fluid and how this is influenced by GE will promote future therapeutic interventions of RA.

Development of a dual-mode Ru-UiO-TPEA MOF probe for highly sensitive and accurate detection of Cu2+ in biological systems

Qian Long,Hanfeng Cui,Jing Zhang,Qiqi Fan,Ting Huang,Hai Shuai,Xiaocui Yin,Guobing Wei,Huanwen Chen,Hao Fan

Cu 2+ serves as a significant biomarker in various physiological processes. This study presents the development of a novel Ru-UiO-TPEA functionalized metal-organic framework (MOFs) probe for Cu 2+ detection, employing both electrochemiluminescence (ECL) and electrochemical (EC) methods. Employing a dual ligand-assisted energy transfer approach, we synthesized Ru-UiO with optimal ECL and EC properties by adjusting the synthesis ratio of biphenyldicarboxylic acid to Ru(dcbpy) 3 2+ . This was followed by its conjugation with N-(2-aminoethyl)-N, N′, N′-tris(pyridin-2-ylmethyl) ethane-1,2-diamine (AE-TPEA) to form the highly conjugated Ru-UiO-TPEA probe for Cu 2+ detection. As the Cu 2+ concentration increased, the probe exhibited a diminished ECL signal and an enhanced differential pulse voltammetry (DPV) signal. The dual-mode detection method enabled cross-validation of the ECL and EC signals, enhancing accuracy and effectively minimizing errors. The ECL and EC methods had detection limits of 0.26 nM (range: 1 nM to 10 μM) and 0.65 nM (range: 2 nM to 80 μM), respectively. Meanwhile, these two methods can effectively detect Cu 2+ in rat brain microdialysis fluid. Notably, the measured concentrations correlated well with those determined by Inductively Coupled Plasma Atomic Emission Spectroscopy (ICP-AES), highlighting its potential in biological and clinical bioelectronic sensing applications.

Study of the Neurotransmitter Changes Adjusted by Circadian Rhythm in Depression Based on Liver Transcriptomics and Correlation Analysis

Peng Wang,Xiaoxia Gao,Fang Zhao,Yao Gao,Kexin Wang,Jun-Sheng Tian,Zhenyu Li,Xue-Mei Qin

Depression has drawn increasing attention from the public around the world in recent years. Studies have shown that liver injury caused by chronic stress is relevant to depression and neurotransmitter changes. It is essential to clarify the relationship between neurotransmitter changes and hepatic gene expression in depression. In this study, we used the chronic unpredictable mild stress (CUMS) model combined with UHPLC–MS to explore the changes of neurotransmitters in serum and hippocampus and to decipher the differential gene expression in the liver by using the RNA-Seq combined with multivariate statistical analysis. Compared with the control group, the levels of neurotransmitters including 5-hydroxytryptamine (5-HT), acetylcholine, glutamate (Glu), and dopamine (DA) in the hippocampus and 5-HT, norepinephrine, γ-aminobutyric acid (GABA), and 5-hydroxyindoleacetic acid in serum were significantly changed in the CUMS rats. The results of liver transcriptomic analysis and correlation analysis showed that the Glu, DA, 5-HT, and GABA were impacted by 68 liver genes which were mainly enriched in three pathways including circadian rhythm, serotonergic synapse, and p53 signaling pathway. The expressive levels of clock genes and serotonergic synapse genes were validated by using q-PCR, and the diurnal rhythms of neurotransmitters were validated by in vivo hippocampus microdialysis. The CUMS stressors might cause phase advance of Glu and GABA by adjusting clock genes. The transcriptomic technique combined with correlation analysis and in vivo microdialysis could be used to discover comprehensive pathways of depression. It provides a new strategy for the rational assessment of the mechanism of disease.
