A biosensor based on carbon dots-protein interactions for specific and sensitive detection of pepsin in saliva

Chenzhao Wang,Yuhao Jiang,Zhiyu Wu,Xiujuan Wang,Jiawei Yang,Junxian Li,Shanshan Wei,Guoying Sun,Zhibing Wang

Salivary pepsin has emerged as a promising biomarker for rapid screening and diagnosis of gastroesophageal reflux disease (GERD). Pepsin mainly exists in strongly acidic environments and exhibits its highest activity. However, the poor stability of most fluorescent sensors in strong acidic environments brings a significant challenge for pepsin detection. Herein, an innovative biosensor was developed for the highly specific and sensitive detection of pepsin on the basis of green-emitting ionic liquid-based carbon dots (G-IL-CDs) conjugated with whey proteins (WPs). The G-IL-CDs exhibited aggregation-induced fluorescence enhancement when interacting with WP, and the fluorescence intensity decreased after incubation with pepsin due to the disruption of the aggregation structure. This strategy is highly selective for pepsin due to the strongly acidic environment in which other proteases are inactivated. Under optimal experimental conditions, this biosensor successfully detected pepsin in real human saliva with a satisfying recovery. Furthermore, this study not only developed a CDs-based sensor for detecting pepsin but also laid a solid theoretical foundation for the future development of novel biosensors combining CDs and proteins.

Structural characteristics of wheat bran insoluble dietary fiber with various particle size distributions and their influences on the kinetics of gastrointestinal emptying in mice

Bingqian Yang,Kaikai Li,Meng Niu,Jianying Wei,Siming Zhao,Caihua Jia,Yan Xu

Wheat bran is an abundant yet underutilized agricultural byproduct. Herein, the insoluble dietary fiber from wheat bran (WBIDF) was ultra-milled to investigate its impact on physicochemical properties and gastrointestinal emptying. SEM and CLSM showed that the laminar structure of WBIDF was disrupted as the particle size was significantly reduced. In the similar characteristic peaks appearing at 3410, 2925, 1635, 1041, and 895 cm −1 in the FT-IR spectra and at 2940, 1593, 1080, and 526 cm −1 in the Raman spectra, the peak intensity was increased as the particle size decreased. It may be that the hydrogen bonding between cellulose, hemicellulose, or other macromolecules was enhanced. X-ray diffraction showed cellulose type I results for all five samples. Correspondingly, the water-holding, swelling, and oil-holding capacities increased by 75.33 %, 52.62 %, and 75.00 %, respectively, in WBIDF-CW 1.8 compared with WBIDF-CWy. Additionally, smaller particle sizes had lower viscosity, thereby enhancing intestinal propulsion and gastric emptying rates. Enhanced contact of the cecal tissue growth factor with the intestinal mucosa delayed ghrelin secretion and stimulated the secretion of motilin, gastrin, and cholecystokinin. In conclusion, the particle sizes of WBIDF were reduced through ultramicro-grinding, leading to altered structure, enhanced hydration and oil-holding capacities, decreased viscosity, and improved gastrointestinal emptying capacity.

CircMTA2 Drives Gastric Cancer Progression through Suppressing MTA2 Degradation via Interacting with UCHL3

Gengchen Xie,Bo Lei,Zhijie Yin,Fei Xu,Xinghua Liu

Our previous study has reported that metastasis-associated protein 2 (MTA2) plays essential roles in tumorigenesis and aggressiveness of gastric cancer (GC). However, the underlying molecular mechanisms ofMTA2-mediated GC and its upstream regulation mechanism remain elusive. In this study, we identified a novel circular RNA (circRNA) generated from theMTA2gene (circMTA2) as a crucial regulator in GC progression. CircMTA2was highly expressed in GC tissues and cell lines, and circMTA2promoted the proliferation, invasion, and metastasis of GC cells both in vitro and in vivo. Mechanistically, circMTA2interacted with ubiquitin carboxyl-terminal hydrolase L3 (UCHL3) to restrainMTA2ubiquitination and stabilizeMTA2protein expression, thereby facilitating tumor progression. Moreover, circMTA2was mainly encapsulated and transported by exosomes to promote GC cell progression. Taken together, these findings uncover that circMTA2suppressesMTA2degradation by interacting with UCHL3, thereby promoting GC progression. In conclusion, we identified a cancer-promoting axis (circMTA2/UCHL3/MTA2) in GC progression, which paves the way for us to design and synthesize targeted inhibitors as well as combination therapies.Keywords:CircRNAs;MTA2;gastric cancer;UCHL3;ubiquitination

Synergy Effects of Three Plant Extracts on Protection of Gastric Mucosa

Caihui Wang,Wen Su,Xingli Su,Guojun Ni,Tao Liu,Yi Kong

The gastric mucosa protection effect of three natural plant extracts, Hericium erinaceus (HE), Centella asiatica (CA) and Amomum villosum (AV), were evaluated using the indomethacin damage model. Compared with a single extract, a combination of HE/CA/AV, especially with the ratios of 80:10:10, 45:45:10 and 45:10:45, showed significant synergistic effects for protection of the gastric mucosa with gastric ulcer inhibition rates of 97.8 ± 0.7%, 86.5 ± 2.8% and 86.1 ± 3.6%, respectively. Microscopic appearances of the gastric mucosa were carried out to help confirm the results.

Effects of NaCl partially substituted with KCl or MgCl2 on the properties, oral mastication, in vitro gastric digestion, and pepsin diffusion of myofibrillar protein gel

Haibo Shi,Ruyi Zhang,Jiabao Zheng,Xianqi Yao,Wei Wang,Igor Tomasevic,Weizheng Sun

The effects of NaCl partially substituted by KCl, MgCl 2 , and KCl/MgCl 2 on the gel characteristics, oral mastication, and in vitro gastric digestion of myofibrillar protein (MP) gel were investigated. The results indicated that partial substitution of NaCl by KCl or MgCl 2 did not cause obvious deterioration of gel properties within 30 % substitution levels. MgCl 2 substitution significantly increased the surface hydrophobicity of MP, and formed homogeneous network structure for effective limitation of moisture loss. The MgCl 2 -substituted gel exhibited relatively lower viscosity and hardness for better swallowing. The high pepsin diffusivity in MgCl 2 -substituted gel (59.57 ± 4.28 μm 2 /s, 30 %-Mg group) improved protein hydrolysis, and produced more shorter peptides due to the loose and relatively uniform structure compared to those of the gels only containing NaCl and partial KCl. Therefore, the changes of textural parameters, microstructure, and water mobility in sodium-reduced gel with different substitution levels could influence the subsequent gastric digestion characteristics.

Controllable fabrication of alginate-based floating beads with core-shell architecture

Chunhuan Liu,Yumeng Shi,Yuchun Liu,Yan Liu,Chenjing Yang,Huan Chen,Cheng Yang,Rong Liang

Hydrogel beads have been explored to be normal platform for nutrition encapsulation and delivery. However, hydrogel beads are naturally denser than water, which make hydrogel beads limite in some floating applications such as gastric floating systems. Here, we designed alginate-based floating beads featuring a core-shell structure, which were composed of a vegetable oil core encapsulated within an alginate network shell, utilizing coaxial co-extrusion microfluidic devices. The shell thickness and core size of the core-shell beads (with the size of 2.80–3.40 mm) can be controlled by simply adjusting the inner phase flow rate ( Q in ) and outer phase flow rate ( Q out ). Notably, the fabrication of floating core-shell beads was achievable with Q in ≥ 40  μ L/min and Q out set at 300  μ L/min. The Hen's egg test – chorioallantoic membrane (HET-CAM) test showed the floating core-shell beads were non-irritant, indicating their safety and biocompatibility properties. Given their floating properties and biocompatibility, these floating core-shell beads hold great potential for applications in the fields of gastric-retentive nutrition delivery. Following that, the floating core-shell beads were further encapsuled with pepsin. They displayed possessed satisfactory pepsin activity (90%) even after 15 days, suggesting that floating core-shell beads can protect the enzyme ingredients against damage. Interestingly, the floating core-shell beads exhibited both efficient sustained-release of pepsin and floating ability in simulated gastric solution. This work sheds light on the design and development of controllable floating core-shell hydrogel beads for various floating system applications.

Functions of Pugionium cornutum (L.) Gaertn Extracts: Investigating the Mechanism of Gastroparesis Amelioration from the Perspective of the Gut Microbiota and Its Metabolites

Yangzu Gao,Haoyu Li,Qian Wu,Bang Chen,Kangzhen Xu,Cong Li,Yehua Shen

The functional exploration of natural foods, coupled with the increasing prevalence of gastrointestinal motility disorders and the associated therapeutic challenges, has generated significant interest in this field. This study aims to investigate the ameliorative effects of the extract fromPugionium cornutum(L.) Gaertn (EAEPC), a traditional edible vegetable in northwest China’s desert region, on atropine-induced gastroparesis in mice, as well as to elucidate its mechanism in terms of the gut microbiota and major metabolites. The findings indicate that EAEPC effectively reduces the rate of pigment residual in the stomach while shortening the gastrointestinal transit time and alleviating other symptoms associated with atropine-induced gastroparesis. These effects may be mediated through modulation of the expression levels of major intestinal metabolites, such as short-chain fatty acids (SCFAs), bile acids (BAs), and L-tryptophan, alongside remodeling of both the diversity and relative abundance of the gut microbiota. Furthermore, correlation analyses were conducted on significantly altered strains and metabolites to clarify their interactions. Moreover, the chemical constituents of EAEPC were identified by UPLC-Q-TOF-MS/MS, and the key active components responsible for improving gastroparesis were predicted through network pharmacology approaches and validated experimentally. These results provide a foundation for further research into the functions ofPugioniumand offer scientific support for developing natural plant-based strategies aimed at treating gastrointestinal motility disorders.

Tea Polyphenol Liposomes Overcome Gastric Mucus to Treat Helicobacter Pylori Infection and Enhance the Intestinal Microenvironment

Guiyun Deng,Yang Wu,Zhiyong Song,Shuojun Li,Moqing Du,Jiamin Deng,Quan Xu,Liu Deng,Hagar Shendy Bahlol,Heyou Han

Infection with Helicobacter pylori (Hp) is one of the leading causes of stomach cancer. The ability to treat Hp infection is hampered by a lack of stomach gastric acid environment. This work introduces a nanoliposome that can rapidly adjust the gastric acid environment to ensure a drug’s optimal efficacy. We introduce CaCO3@Fe–TP@EggPC nanoliposomes (CTE NLs) that are composed of Fe3+ and tea polyphenols (TPs) forming complexes on the surface of internal CaCO3 and then with lecithin producing a phospholipid bilayer on the polyphenols’ outer surface. Through the action of iron–TP chelate, the phospholipid layer can fuse with the bacterial membrane to eliminate Hp. Furthermore, CaCO3 can promptly consume the excessive gastric acid, ensuring an ideal operating environment for the chelate. TPs, on the other hand, can improve the inflammation and gut microbes in the body. The experimental results show that CTE NLs can quickly consume protons in the stomach and reduce the bacterial burden by 1.2 orders of magnitude while reducing the inflammatory factors in the body. The biosafety evaluation revealed that nanoliposomes have good biocompatibility and provide a new strategy for treating Hp infection.

LncRNA POU6F2-AS2 contributes to malignant phenotypes and paclitaxel resistance by promoting SKP2 expression in stomach adenocarcinoma

Yanzhao Jin,Jiaqing Cao,Hua Cheng,Xiaoyun Hu

This study aimed to investigate the role and mechanism of POU6F2-AS2 in the development of gastric cancer. POU6F2-AS2 expression was considerably higher in clinical stomach adenocarcinoma (STAD) tissues and gastric cancer cell lines (MKN-28 and MGC-803) than in neighbouring normal tissues and gastric mucosa epithelial cells (GES-1). POU6F2-AS2 overexpression resulted in a low overall survival probability, progression-free survival probability and post progression survival probability, as well as increased cell viability, migration and invasion of gastric cancer cells, thereby inhibiting apoptosis. Based on RNA pull-down, cycloheximide and MG132 incubation experiments, POU6F2-AS2 promoted SKP2 by stabilizing NONO expression. In addition, in vivo silencing of POU6F2-AS2 in gastric cancer cells can inhibit tumour progression and produce a synergistic antitumour effect when combined with paclitaxel. POU6F2-AS2 is overexpressed in STAD, which is attributed to a bad prognosis. In vitro and in vivo experiments have confirmed that the POU6F2-AS2/NONO/SKP2 axis promotes STAD progression, and that the silencing of POU6F2-AS2 plays a synergistic antitumour effect when combined with paclitaxel. Therefore, POU6F2-AS2 may be potentially developed as a target to inhibit STAD and reduce chemoresistance.

Reactive Oxygen Species-Responsive Ferrocene Nanoparticles Delivering Small Interfering RNA Targeting NOP2/Sun RNA Methyltransferase Family Member 2 for Gastric Cancer Therapy

Yunsheng Lu,Yibin Huang,Chenchen Mao,Pengfei Shan,Chenkang Wu,Jiongzhou Zhu,Yujie Lin,Zhongyu Li,Mingdong Lu

Silencing NOP2/Sun RNA methyltransferase family member 2 (NSUN2) effectively inhibits gastric cancer (GC) progression but is limited by RNase degradation, rapid renal clearance, and low uptake. Based on the characteristic high levels of reactive oxygen species (ROS) in the tumor microenvironment, this study designed and synthesized a novel ROS-responsive ferrocene nanoparticle loaded with siNSUN2 (PRPFc@siNSUN2). Under ROS conditions, the nanoparticle disintegrates to release siNSUN2. Characterization by proton nuclear magnetic resonance, transmission electron microscopy, dynamic light scattering, and ultraviolet–visible spectrophotometry revealed that PRPFc@siNSUN2 is spherical, with an average diameter of 88.79 ± 1.14 nm, an encapsulation efficiency of 83.10%, and a drug loading capacity of 13.85%. Moreover, these nanoparticles demonstrated excellent stability and, under hydrogen peroxide conditions, exhibited structural disruption leading to the release of siNSUN2, thereby confirming their high ROS responsiveness. In vitro, PRPFc@siNSUN2 markedly enhanced the inhibition of GC cell proliferation, migration, and invasion, and promoted apoptosis, accompanied by increased intracellular ROS and improved siNSUN2 uptake. In vivo studies further confirmed that PRPFc@siNSUN2 markedly enhanced the therapeutic efficacy of siNSUN2 against GC, while exhibiting low cytotoxicity and good biocompatibility. Overall, our findings indicate that PRPFc@siNSUN2, with its favorable morphology, stability, and ROS-triggered release, substantially improves the anti-GC effects of siNSUN2 by inhibiting GC cell proliferation, migration, and invasion, as well as by promoting apoptosis. These results support NSUN2 as a promising therapeutic target and underscore the potential of PRPFc@siNSUN2 nanoparticles in drug delivery, offering a novel strategy to improve clinical outcomes for GC patients.

BUB1B promotes cisplatin resistance in gastric cancer via Rad51-mediated DNA damage repair

Zhe Qin,Fangzhou Ye,Jiayi Wang,Jun Jiang,Xiaohong Zhang,Huanqing Li,Li Feng

Background Cisplatin resistance significantly impedes the treatment of gastric cancer (GC). This work examined the possible therapeutic target status and function of BUB1B in controlling cisplatin resistance. Methods Following the identification of differentially expressed genes (DEGs), protein-protein interaction (PPI) network analysis was conducted using datasets from the Cancer Genome Atlas-stomach adenocarcinoma (TCGA-STAD), GSE51575, and GSE79973. Functional tests assessed the effect of BUB1B overexpression and knockdown on the GC cells. Enrichment analysis and RNA-seq identified pathways linked to BUB1B . Additionally, the function of BUB1B in GC cells resistant to cisplatin in regulating DNA repair was examined, as its relationship with Rad51 inhibitor (B02) in regulating cell cycle, proliferation, and apoptosis. The combined effects of Rad51 suppression and BUB1B overexpression on tumor development in cisplatin-resistant GC cells were further validated in vivo xenograft models. Results Significant overexpression of six critical overlapping genes was seen in GC tissues. The GC cell invasion, migration, and proliferation processes were improved by BUB1B overexpression, whereas BUB1B knockdown prevented these outcomes. Genes involved in DNA repair were downregulated by BUB1B knockdown, according to an RNA-seq study. BUB1B overexpression boosted cell survival via modulating cell cycle proteins, but BUB1B knockdown hampered DNA repair and increased death in cisplatin-resistant GC cells. Overexpression of BUB1B enhanced tumor development in vivo and counteracted the inhibitory effects of B02 on cell growth. Conclusion BUB1B enhances cisplatin resistance in gastric cancer by regulating DNA repair and cell cycle progression, suggesting that targeting BUB1B may be a feasible therapeutic strategy.
