NIR-II-driven and glutathione depletion-enhanced hypoxia-irrelevant free radical nanogenerator for combined cancer therapy

Zhang Li,Fan Yadi,Yang Zhe,Yang Mo,Wong Chun-Yuen

Background Though the combination of photodynamic therapy (PDT) and chemodynamic therapy (CDT) appears to be very attractive in cancer treatment, hypoxia and overproduced glutathione (GSH) in the tumor microenvironment (TME) limit their efficacy for further application. Results In this work, a smart hypoxia-irrelevant free radical nanogenerator (AIPH/PDA@CuS/ZIF-8, denoted as APCZ) was synthesized in situ via coating copper sulphide (CuS)-embedded zeolitic imidazolate framework-8 (ZIF-8) on the free radical initiator 2,2′-azobis[2-(2-imidazolin-2-yl)propane]-dihydrochloride (AIPH)-loaded polydopamine (PDA). APCZ showed promising GSH-depleting ability and near-infrared (NIR)-II photothermal performance for combined cancer therapy. Once internalized by 4T1 cells, the outer ZIF-8 was rapidly degraded to trigger the release of CuS nanoparticles (NPs), which could react with local GSH and sequentially hydrogen peroxide (H2O2) to form hydroxyl radical (·OH) for CDT. More importantly, the hyperthermia generated by APCZ upon 1064 nm laser excitation not only permitted NIR-II photothermal therapy (PTT) and promoted CDT, but also triggered the decomposition of AIPH to give toxic alkyl radical (·R) for oxygen-independent PDT. Besides, the PDA together with CuS greatly decreased the GSH level and resulted in significantly enhanced PDT/CDT in both normoxic and hypoxic conditions. The tumors could be completely eradicated after 14 days of treatment due to the prominent therapeutic effects of PTT/PDT/CDT. Additionally, the feasibility of APCZ as a photoacoustic (PA) imaging contrast agent was also demonstrated. Conclusion sThe novel APCZ could realize the cooperative amplification effect of free radicals-based therapies by NIR-II light excitation and GSH consumption, and act as a contrast agent to improve PA imaging, holding tremendous potential for efficient diagnosis and treatment of deep-seated and hypoxic tumors.Graphic abstract

NIR-II Luminescent Nanothermometer for Precise Assessment of Ischemic Stroke

Xiaochen Qiu,Wei Yuan,Chen Jiang,Wei Feng,Fuyou Li

Ischemic stroke, an acute cerebral local blood circulation disorder caused by neurological deficit syndrome, has emerged as a prominent ailment that can affect the well-being and quality of life of the elderly. The difficulty in early assessment and narrow therapeutic time window can increase the risks of mortality and disability. Monitoring the temperature of brain microregions is a promising strategy for assessing the onset of an early stroke, which is positively correlated with the intricate immune response. In this study, an innovative hybrid nanothermometer combining lanthanides and quantum dots was established for contactless monitoring of cerebral lesion temperatures in mice with ischemic stroke. The output of ratio optical signals in the near-infrared-II window (1000–1700 nm) guaranteed deep penetration, high accuracy, and reduced tissue damage. An obvious temperature increase was detected in the minor middle cerebral artery occlusion model mice (4 h after reperfusion), indicating the occurrence of a minor stroke even in the absence of abnormal behaviors. Moreover, a further increase in the brain temperature of the ischemic region resulted in a more severe cerebral lesion, suggesting the potential advancement of the treatment window for early ischemic stroke. The proposed approach can enable precise assessment of ischemic stroke, with significant implications for the incidence and progression of the illness, ultimately enhancing the quality of life of affected patients.

Near-Infrared Ii Hemicyanine Dye with Large Stokes Shift Designed by Tict Regulation for Boosting Imaging-Guided Photothermal Therapy

Liang Zhao,Han Zhu,You-Yang Duo,Dai-Wen Pang,Zhi-Gang Wang,Shu-Lin Liu

The serious threat that cancer poses to human health highlights the significance of early detection and effective treatment. The integration of fluorescence diagnosis and photothermal therapy in NIR-II has gained attention due to its high sensitivity, fast response, and non-invasiveness. Fluorescence, produced by the radiative relaxation process of electrons in a molecule, and photothermal, generated by the non-radiative relaxation process of electrons in a molecule, are competing photophysical processes. Hence, it is a challenge for the molecule to balance between the properties of fluorescence and photothermal. In this study, we designed a NIR-II hemicyanine with TICT character to obtain molecules with both better fluorescence and photothermal properties, utilizing positively charged pyridine salt and triphenylamine as electron acceptor and donor, respectively, and oxole as the conjugated π-bridge. HCY-995 , one of the synthesized compounds, had a quantum yield of 0.09%, photothermal conversion efficiency of 54.90%, and a significant Stoke shift of 232 nm, which made it appropriate for the integration of photothermal therapy and high-resolution imaging. This study provides new insights into the development of NIR-II molecules with fluorescent and photothermal integrated properties. This article is protected by copyright. All rights reserved

Visible-near infrared-II skull optical clearing window for in vivo cortical vasculature imaging and targeted manipulation

Dong-Yu Li,Zheng Zheng,Ting-Ting Yu,Ben-Zhong Tang,Peng Fei,Jun Qian,Dan Zhu

Skull optical clearing window permits us to perform in vivo cortical imaging without craniotomy, but mainly limits to visible (vis)-near infrared (NIR)-I light imaging. If the skull optical clearing window is available for NIR-II, the imaging depth will be further enhanced. Herein, we developed a vis-NIR-II skull optical clearing agents with deuterium oxide instead of water, which could make the skull transparent in the range of visible to NIR-II. Using a NIR-II excited third harmonic generation microscope, the cortical vasculature of mice could be clearly distinguished even at the depth of 650 μm through the vis-NIR-II skull clearing window. The imaging depth after clearing is close to that without skull, and increases by three times through turbid skull. Furthermore, the new skull optical clearing window promises to realize NIR-II laser-induced targeted injury of cortical single vessel. This work enhances the ability of NIR-II excited nonlinear imaging techniques for accessing to cortical neurovasculature in deep tissue.

InSe Nanosheets for Efficient NIR-II-Responsive Drug Release

Chi Huang,Zhengbo Sun,Haodong Cui,Ting Pan,Shengyong Geng,Wenhua Zhou,Paul K. Chu,Xue-Feng Yu

Near-infrared-II (NIR-II) biowindow is appealing from the perspectives of larger maximum permissible exposure in comparison with the near-infrared-I biowindow, so the NIR-II-responsive drug-delivery nanoplatform is highly desirable. In this work, two-dimensional InSe nanosheets (InSe NSs) are modified with poly(ethylene glycol) and evaluated as an effective NIR-II-responsive cancer treatment nanoplatform. The InSe NSs synthesized by liquid exfoliation exhibit prominent NIR-II-responsive photothermal conversion efficiency (39.5%) and photothermal stability. Moreover, the InSe NSs have a doxorubicin (DOX) loading capacity as high as 93.6%, along with excellent NIR-II-responsive DOX release characteristic. The superior synergistic chemo/photothermal effects have also been demonstrated by the in vitro experiments in killing cancer cells. In combination with good biocompatibility, the InSe NSs have great potential in therapeutic applications.

Luminescence Nanoprobe in the Near-Infrared-II Window for Ultrasensitive Detection of Hypochlorite

Xiao Fu,Jinmei Wu,Huajian Xu,Pingping Wan,Huimin Fu,Qingsong Mei

Sensitive and selective detection of hypochlorite is in great demand for food safety, especially in fresh cold chain products. However, the detection limit of traditional visible emission-based strategies cannot satisfy the requirement of ultrasensitive analysis in practical applications. In this work, we explored a novel luminescent nanoprobe in the near-infrared-II (NIR-II) window to greatly improve the hypochlorite detection limit for analysis of real milk samples, which was based on the fluorescence resonance energy-transfer process between the hypochlorite-responsive dye (FD1080) and the lanthanide-doped downconverted nanoparticles. Specifically, the NIR-II luminescence from Yb ions was first suppressed by FD1080 due to the energy-transfer mechanism. In the presence of hypochlorite, FD1080 was bleached to recover the luminescence. As a proof-of-concept, the optimal nanoprobe exhibited a linear luminescence recovery in the range of 0.1–1 nM with the detection limit of 0.0295 nM for hypochlorite. Real milk sample detection experiments showed that the probe had good accuracy and precision.

Amplification of oxidative damage using near-infrared II-mediated photothermal/thermocatalytic effects for periodontitis treatment

Xinyu Dai,Yiping Liu,Fanrong Meng,Qiqing Li,Fengxia Wu,Jianguo Yuan,Haoran Chen,Huixin Lv,Yanmin Zhou,Yulei Chang

Periodontitis is a biofilm-related disease characterized by damage to the periodontal tissue and the development of systemic diseases. However, treatment of periodontitis remains unsatisfactory, especially with deep-tissue infections. This study describes rationally designed multifunctional photothermocatalytic agents for near-infrared-II light-mediated synergistic antibiofilm treatment, through modification of Lu-Bi 2 Te 3 with Fe 3 O 4 and poly(ethylene glycol)-b-poly(l-arginine) (PEG- b -PArg). Notably, 1064-nm laser irradiation led to photothermal/thermocatalytic effects, resulting in the synergistic generation of reactive oxygen species (ROS) and reactive nitrogen species (RNS) and consequent damage to the biofilm. This treatment was based on the thermoelectric and photothermal conversion properties of Lu-Bi 2 Te 3 , the peroxidase-like catalytic capacity of Fe 3 O 4 , and the guanidinium polymer, PEG- b -PArg. Oxidative damage to biofilm was further enhanced by H 2 O 2 , resulting in the effective elimination of biofilm both in vitro and in vivo. These findings suggest that this synergistic therapeutic strategy is effective for the clinical treatment of periodontitis. Statement of Significance : The current treatment for periodontitis involves time-consuming and labor-intensive clinical scaling of the teeth. The present study is the first to assess the efficacy of a photothermal catalyst for periodontitis treatment. This used near-infrared-II light at 1064 nm to induce oxidative damage in the biofilm, resulting in its degradation. The synergistic photothermal/thermoelectric effect produced deep tissue penetration and was well tolerated, and can kill the biofilm formed by periodontitis pathogens up to 5 orders of magnitude, effectively treating the biofilm-induced periodontitis.

An Implant-Free Nanosystem Enabling Synergistic Oxidative Damage Mitigation and Deep Brain Stimulation for Alleviating Parkinsonian Symptoms

Chen Zhou,Feifei Wu,Liqing He,Huijie Yan,Zhaowei Zhang,Shuang Zhao,Wenyi Huang,Yuhao Luo,Wei Zhao,Bulong Gao,Zherui Zhang,Yaofeng Zhou,Baiheng Wu,Junqiu Liu,Dingcheng Zhu

Deep brain stimulation (DBS) effectively alleviates motor symptoms in Parkinson’s disease (PD) patients; however, it necessitates permanent invasive implantation of conduits, and its therapeutic effects diminish as PD progresses. Herein, an implant-free NIR-II laser-activated intervention nanosystem that combines wireless DBS with antioxidative neuroprotection is developed to overcome the above challenges. Enzyme-like 2-(phenylselanyl)ethan-1-amine (SePh) and manganese dioxide (MnO2) and NIR-II absorber IR-1048 (IR) are integrated onto the mesoporous polydopamine (mPDA) core to form mPDA-SeMn-IR. Upon NIR-II laser irradiation, intraventricularly injected mPDA-SeMn-IR nanoparticles effectively activate endogenously expressed inositol 1,4,5-trisphosphate receptors, leading to Ca2+ outflux from the endoplasmic reticulum. Ca2+ signaling enhances tyrosine hydroxylase expression and activity and triggers dopamine release to enhance dopaminergic function. Meanwhile, mPDA-SeMn-IR rapidly eliminates excessive reactive oxygen species, synergistically enhancing long-term DBS efficiency by increasing neuron survival and restoring neuronal function for improved dopamine secretion. In vivo, mPDA-SeMn-IR regulates midbrain neurons and coiling behaviors in zebrafish and considerably alleviates motor symptoms in the PD model, proposing a promising therapeutic strategy to address existing challenges in PD therapies.

Efficient energy transfer in NaBaScSi2O7: Cr3+, Er3+ phosphor enabling simultaneously strong NIR-II/III emissions for diversified applications

Sihan Yang,Xinyu Li,Qian Zhang,Meiting Li,Xuejiao Wang,Nan Wu

Studies have shown that emissions in the NIR-II and NIR-III regions exhibit high tissue penetration, along with superior performance in biological and medical imaging applications. Therefore, developing NIR phosphors capable of simultaneously show emissions in both the NIR-II and NIR-III regions holds significant value. Herein, we developed a novel Cr 3+ /Er 3+ co-doped NaBaScSi 2 O 7 phosphor that demonstrates efficient dual-band emission in both NIR-II and NIR-III regions under visible light excitation, enabled by highly effective Cr 3+ →Er 3+ energy transfer. The energy transfer efficiencies calculated by the emission intensity and fluorescence lifetime are 65.3% and 47.4%, respectively, with the energy transfer mechanism dominated by dipole-dipole interaction. The prepared phosphor can be effectively excited by multiple excitation sources (378 nm, 492 nm, 808 nm, and 980 nm), generating distinct emission responses, highlighting its potential for anti-counterfeiting applications. Additionally, the distinct temperature-dependent optical properties of Cr 3+ and Er 3+ offer a viable approach for non-contact temperature detection, achieving a relative sensitivity of 0.8% K -1 and a temperature resolution of 0.126 K at 498 K. The NIR-II/III pc-LED fabricated by NaBaScSi 2 O 7 : 1%Cr 3+ , 0.2%Er 3+ demonstrates outstanding non-invasive imaging capability and accurate predictions for organic solvents. This study presents a strategy for developing phosphors capable of simultaneous NIR-II/III emission, and significantly advances their multifunctional applications in NIR optical thermometry and NIR pc-LED technologies.

Protein-encapsulation-driven high-performance luminogen for three-photon imaging of deep tumor in the NIR-II biowindow

Mingdi Yang,Kaiwen Li,Yingcui Bu,Lixin Ma,Yingyong Ni,Jing Huang,Jingyan Zhang,Hongping Zhou

The development of organic molecules activated by near-infrared II (NIR-II) light holds considerable potential for deep image-guided disease diagnosis and therapeutic applications. However, the design and synthesis of such materials remain a significant challenge due to limitations in controllable molecular architecture. In this study, two small molecules, TPEPyC2 and DTPEPyC5 , were firstly synthesized. Subsequently, their respective assemblies, namely TPEPyC2@HSA and DTPEPyC5@HSA , were obtained through co-assembled with the hydrophobic human serum albumin (HSA). Comprehensive analyses demonstrated that only DTPEPyC5@HSA exhibited superior photostability, biosafety, and a large three-photon absorption cross-section (3.85 × 10 −80  cm 6  s 2 ). Notably, this probe enabled precise mitochondrial targeting in live HepG2 cells, as indicated by a co-localization coefficient of Pr  = 0.88, and achieved tumor tissue penetration of 143 μm using three-photon fluorescence imaging. This bio-protein encapsulation strategy provides a novel molecular design framework for developing NIR–II–excitable fluorescent probes, thereby significantly enhancing their imaging performance.

Bioactive NIR-II gold clusters for three-dimensional imaging and acute inflammation inhibition

Huizhen Ma,Xiaoning Zhang,Ling Liu,You Huang,Si Sun,Ke Chen,Qi Xin,Pengfei Liu,Yuxing Yan,Yili Wang,Yuan Li,Haile Liu,Ruoli Zhao,Kexin Tan,Xinzhu Chen,Xun Yuan,Yonghui Li,Ying Liu,Haitao Dai,Changlong Liu,Hao Wang,Xiao-Dong Zhang

Strong fluorescence and high catalytic activities cannot be achieved simultaneously due to conflicts in free electron utilization, resulting in a lack of bioactivity of most near-infrared-II (NIR-II) fluorophores. To circumvent this challenge, we developed atomically precise Au22 clusters with strong NIR-II fluorescence ranging from 950 to 1300 nm exhibiting potent enzyme-mimetic activities through atomic engineering to create active Cu single-atom sites. The developed Au21Cu1 clusters show 18-fold higher antioxidant, 90-fold higher catalase-like, and 3-fold higher superoxide dismutase–like activities than Au22 clusters, with negligible fluorescence loss. Doping with single Cu atoms decreases the bandgap from 1.33 to 1.28 eV by predominant contributions from Cu d states, and Cu with lost electron states effectuates high catalytic activities. The renal clearable clusters can monitor cisplatin-induced renal injury in the 20- to 120-minute window and visualize it in three dimensions using NIR-II light-sheet microscopy. Furthermore, the clusters inhibit oxidative stress and inflammation in the cisplatin-treated mouse model, particularly in the kidneys and brain.

Tumor microenvironment-responsive nanohybrid for hypoxia amelioration with photodynamic and near-infrared II photothermal combination therapy

Pengli Zhang,Qinghe Wu,Jingxing Yang,Mengfei Hou,Benchao Zheng,Jia Xu,Yimin Chai,Liqin Xiong,Chunfu Zhang

Phototherapy, particularly photothermal therapy (PTT) and photodynamic therapy (PDT), has been widely investigated for tumor treatment. However, the limited tissue penetration depth of light in the near-infrared I (NIR-I) region and the hypoxic tumor microenvironment (TME) severely constrain their clinical applications. To address these challenges, in the present study, we developed a chlorin e6 (Ce6) and MnO 2 -coloaded, hyaluronic acid (HA)-coated single-walled carbon nanohorns (SWNHs) nanohybrid (HA-Ce6-MnO 2 @SWNHs) for PDT and PTT combination therapy of tumor. HA-Ce6-MnO 2 @SWNHs responded to the mild acidic TME to ameliorate tumor hypoxia , thus enhancing tumor PDT. Moreover, HA-Ce6-MnO 2 @SWNHs had a high photothermal conversion efficiency at 1064 nm (55.48%), which enabled deep tissue penetration (3.05 cm) and allowed for highly efficient tumor PTT in near-infrared II (NIR-II) window. PDT and PTT combination therapy with HA-Ce6-MnO 2 @SWNHs achieved a good therapeutic efficacy on 4T1 tumor-bearing mice, eradicating the primary tumors and suppressing cancer recurrence. Our study provides a promising strategy for developing a hypoxia relief and deep tissue penetration phototherapy platform by using SWNHs for highly effective tumor PDT and NIR-II PTT combination therapy. Statement of Significance The hypoxic tumor microenvironment (TME) and the limited penetration of the NIR-I light in biological tissues compromise the efficacy of photothermal therapy (PTT) and photodynamic therapy (PDT) on tumors. Here, we developed a chlorin e6 (Ce6) and MnO 2 -coloaded, hyaluronic acid (HA)-coated single-walled carbon nanohorns (SWNHs) nanohybrid (HA-Ce6-MnO 2 @SWNHs) for PDT and PTT combination therapy of tumors. The nanohybrid could efficiently accumulate in tumors through CD44-mediated active targeting. The sequential MnO 2 -enhanced PDT and efficient NIR-II PTT had a remarkable therapeutic effect by eliminating the primary tumor and simultaneously inhibiting tumor recurrence.
