Changes of Potent Odorants in Salted Duck Egg Yolk before and after Roasting

Xiaofan Hao,Miao Liang,Runhu Xin,Yuping Liu

As the second most widely consumed eggs, duck eggs are made into preserved eggs, salted duck eggs, and roasted duck eggs to extend their shelf-life. To investigate the differences in potent odorants (POs) between salted duck egg yolk (SDEY) and roasted duck egg yolk (RDEY), the volatiles in SDEY and RDEY were extracted through solvent extraction coupled with solvent-assisted flavor evaporation and were assayed with gas chromatography-mass spectrometry-olfactometry. A total of 45 volatiles were identified in two samples, 24 odor-active compounds (OACs) were screened, and more OACs were in RDEY. The flavor-dilution (FD) factors of OACs were obtained by aroma extract dilution analysis and ranged from 3 to 6561. Twenty-two OACs with FD factors ≥ 9 were quantitated, and the results indicated the concentrations of OACs in yolk increased greatly after salted duck eggs were roasted. Based on the concentrations and thresholds, odor activity values (OAVs) were determined; 17 odorants with OAVs ≥ 1 were determined as POs. Acetoin was the most PO in SDEY; there were more POs in RDEY, including 2-ethyl-3,6-dimethylpyrazine, acetoin, 2-acetyl-3-methylthiophene, dihydro-4-hydroxy-2(3H)-furanone, etc. The outcomes obtained have reference values for making better use of duck eggs in the food industry.

Prevalence of fluoroquinolone resistance and mutations in the gyrA, parC and parE genes of Riemerella anatipestifer isolated from ducks in China

Zhu Dekang,Zheng Mingyu,Xu Jinge,Wang Mingshu,Jia Renyong,Chen Shun,Liu Mafeng,Zhao Xinxin,Yang Qiao,Wu Ying,Zhang Shaqiu,Huang Juan,Liu Yunya,Zhang Ling,Yu Yanling,Pan Leichang,Chen Xiaoyue,Cheng Anchun

Background Riemerella anatipestifer is one of the most serious infectious disease-causing pathogens in the duck industry. Drug administration is an important method for prevention and treatment of infection in duck production, leading to widespread drug resistance in R. anatipestifer . Methods For a total of 162 isolates of R. anatipestifer, the MICs were determined for a quinolone antimicrobial agent, namely, nalidixic acid, and three fluoroquinolones, namely, ciprofloxacin, enrofloxacin and ofloxacin. The gyr A, par C, and par E gene fragments were amplified by PCR to identify the mutation sites in these strains. Site-directed mutants with mutations that were detected at a high frequency in vivo were constructed (hereafter referred to as site-directed in vivo mutants), and the MICs of these four drugs for these strains were determined. Results In total, 100, 97.8, 99.3 and 97.8% of the 137 R. anatipestifer strains isolated between 2013 and 2018 showed resistance to nalidixic acid, ciprofloxacin, enrofloxacin, and ofloxacin, respectively. The high-frequency mutation sites were detected in a total of 162 R. anatipestifer strains , such as Ser83Ile and Ser83Arg, which are two types of substitution mutations of amino acid 83 in GyrA; Val799Ala and Ile811Val in ParC; and Val357Ile, His358Tyr, and Arg541Lys in ParE. MIC analysis results for the site-directed in vivo mutants showed that the strains with only the Ser83Ile mutation in GyrA exhibited an 8–16-fold increase in MIC values, and all mutants showed resistance to ampicillin and ceftiofur. Conclusions The resistance of R. anatipestifer to quinolone agents is a serious problem. Amino acid 83 in GyrA is the major target mutation site for the fluoroquinolone resistance mechanism of R. anatipestifer .

Assessment of the Effect of Baicalin on Duck Virus Hepatitis

Chen Yun,Yao Fangke,Ming Ke,Shi Jintong,Zeng Ling,Wang Deyun,Wu Yi,Hu Yuanliang,Liu Jiaguo

Background: Duck virus hepatitis (DVH) caused by duck hepatitis A virus type 1 (DHAV-1) is a malignant disease in ducklings, causing economic losses in the duck industry. However, there is still no antiviral drug against DHAV-1 in the clinic. Objective: Our aim is to investigate the anti-DHAV-1 effect of baicalin, which is a flavonoid derived from the Chinese medicinal herb huangqin (Scutellaria baicalensis Georgi). Methods: Here, we first detected its anti-DHAV-1 ability in vitro and in vivo. At the same time, the inhibition of baicalin on DHAV-1 reproduction was determined. Finally, we tested and verified the anti-oxidative and immuno-enhancing roles of baicalin on its curative effect on DVH. Results: Baicalin possessed anti-DHAV-1 effect. It improved the cytoactive of DEH which was infected by DHAV-1 as well as reduced the DHAV-1 reproduction in DEH. Under baicalin treatment, mortality of ducklings infected by DHAV-1 decreased, additionally the DHAV-1 level and liver injury in such ducklings were significantly reduced or alleviated. The in vitro mechanism study indicated baicalin inhibited DHAV-1 reproduction via interfering the viral replication and release. Furthermore, the in vivo mechanism study manifested both the anti-oxidative and immuno-enhancing abilities of baicalin, which played crucial roles in its curative effect on DVH. Conclusion: This study may provide a scientific basis for developing baicalin as one or a part of the anti-DHAV-1 drugs.

Solidification of plant essential oils by electrostatic adsorption and Ca2+ crosslinking for pot-stewed duck neck preservation

Junping Liu,Yuhao Huang,Xing Lu,Wenzhi Yu,Kaibo Yu,Shengfeng Peng,Lei Zhou,Wei Liu

Pot-stewed duck neck (PSDN) is a popular leisure food in China. However, PSDN is rich in nutrients and has high water activity, which can easily cause the oxidation of fat and protein, and microbial growth, resulting in a short shelf life. As a potential preservative, plant essential oils have restricted application due to their poor water solubility, effumability, and easy oxidizability. In this study, the porous starch that had absorbed cinnamon and zanthoxylum essential oils (EOs) was encapsulated by electrostatic adsorption between chitosan and sodium alginate, and solid preservative (SP) was further formed by crosslinking of Ca 2+ with sodium alginate. Results showed that SP exhibited good encapsulation property and sustained-release effect on EOs. Cinnamon EO showed antibacterial properties and the main component was cinnamaldehyde. Antioxidant property was observed in zanthoxylum EO and the main components were linalool and D-limonene. Subsequently, SP was used to preserve PSDN and stored in modified atmosphere packaging (MAP). Results showed that the SP-MAP treatment provided at least double-time extension of the PSDN shelf life for 3 and 11 days at 25 and 4 ○ C, respectively. The lower values of volatile base nitrogen and thiobarbituric acid indicated that the lipid oxidation of PSDN was delayed. Besides, microbial growth of PSDN with SP-MAP was inhibited with the lowest total viable and fungal counts, and the results of incubation and enumeration of pathogenic bacteria in PSDN samples indicated that Escherichia coli , Bacillus subtilis and Staphylococcus aureus were effectively inhibited during the storage time. Therefore, solidifying essential oils combined with MAP was a promising method to preserve pot-stewed meat products.

Effects of galangal extract on lipid oxidation, antioxidant activity and fatty acid profiles of salted duck eggs

Harlina Putri Widyanti,Shahzad Raheel,Ma Meihu,Wang Ning,Qiu Ning

In the present study, we investigated the effect of galangal extract on the lipid oxidation, antioxidant activity and fatty acid composition of salted duck eggs. Three groups of salted duck eggs were comparatively assessed: salted duck egg enriched with 0.1% and 0.5% (w/v) of galangal extract and salted duck egg without galangal extract as a control. Results showed that galangal extract significantly reduced 2-thiobarbituric acid reactive substances from 1.60 mg MDA kg −1 to 1.07 mg MDA kg −1 and anisidine values from 67.55 to 53.64 at the end of salting time, moreover, decrease in the levels of conjugated dienes were also noticed from 0.018 to 0.007% during the salting time compared to the control group. Furthermore, galangal extract along with salting time also had significant effects on the fatty acid composition. Oleic acid, followed by palmitic acid and margaric acid, were the predominant fatty acids found in salted duck eggs supplemented with galangal extract after 14 days of salting process. Results of DPPH-scavenging activity indicated that salted duck eggs supplemented with 0.5% (w/v) galangal extract had significantly higher antioxidant activity ( p  < 0.05) than control at 28th day of salting. Further, analysis of electronic nose revealed that the addition of galangal extract could provide unique flavor to salted eggs during the curing process.

Role of PLC/IP3/IP3R axis in excess molybdenum exposure induced apoptosis in duck renal tubular epithelial cells

Huiling Guo,Caiying Zhang,Wenjing Pu,Xuesheng Chu,Gang Huang,Ting Cui,Bingyan Huang,Hang Liu,Xueyan Dai

Excess molybdenum (Mo) is harmful to animals, but its nephrotoxicity has not been comprehensively explained. To appraise the influences of excess Mo on Ca homeostasis and apoptosis via PLC/IP 3 /IP 3 R axis, primary duck renal tubular epithelial cells were exposed to 480 μM and 960 μM Mo, and joint of 960 μM Mo and 10 μM 2-APB or 0.125 μM U-73122 for 12 h (U-73122 pretreated for 1 h), respectively. The data revealed that the increment of [Ca 2+ ] c induced by Mo mainly originated from intracellular Ca storage. Mo exposure reduced [Ca 2+ ] ER , elevated [Ca 2+ ] mit , [Ca 2+ ] c , and the expression of Ca homeostasis-related factors (Calpain, CaN, CRT, GRP94, GRP78 and CaMKII). 2-APB could effectively reverse subcellular Ca 2+ redistribution by inhibiting IP 3 R, which confirmed that [Ca 2+ ] c overload induced by Mo originated from ER. Additionally, PLC inhibitor U-73122 remarkably mitigated the change, and dramatically reduced the number of apoptotic cells, the expression of Bak-1, Bax, cleaved-Caspase-3/Caspase-3, and notably increased the expression of Bcl-xL, Bcl-2, and Bcl-2/Bax ratio. Overall, the results confirmed that the Ca 2+ liberation of ER via PLC/IP 3 /IP 3 R axis was the main cause of [Ca 2+ ] c overload, and then stimulated apoptosis in duck renal tubular epithelial cells.

Influence of salting processes on water and lipid dynamics, physicochemical and microstructure of duck egg

Shasha Cheng,Tan Zhang,Xiaohui Wang,Yukun Song,Huihui Wang,Haitao Wang,Peiqiang Yang,Mingqian Tan

Water and lipid changes play important roles for the quality formation of salted duck eggs . In the present study, low-field nuclear magnetic resonance (LF-NMR) and magnetic resonance imaging (MRI) were introduced to monitor water and lipid dynamics of duck egg during salting as nondestructive methods . Four proton pools with different mobilities in fresh duck egg matrix were identified by LF-NMR transverse relaxation, and salting significantly changed the proton mobility due to the gelation of egg yolk, release of lipid and watery of egg white . MRI showed high density of proton with lower mobility and higher mobility in egg yolk and white, respectively, and the brighter region of egg yolk in T 1 images continuously decreased from vitelline membrane to center. The variations in the relaxation parameters and MRI images have a close relationship with other quality characteristics of duck eggs induced by salt. Therefore, LF-NMR and MRI could provide information about water and lipid dynamics of duck eggs during salting, and have great potential for quality assurance in salted duck eggs as a fast and non-destructive method.

Mass spectrometry differentiation of sciadonic acid isomers and estimate of long-chain polyunsaturated fatty acid intake from fresh and processed duck eggs

Chuanshun Dai,Lerong Qi,Tingxiang Yang,Zhen Wang,Dong Hao Wang

Sciadonic acid, or 20:3(5Z,11Z,14Z), is an unusual polyunsaturated fatty acid (PUFA) found in certain gymnosperm nuts and marine invertebrates. However, no study has detected it in commonly consumed food items. This study analyzed the fatty acid compositions of fresh, salted, and preserved duck eggs thoroughly via gas chromatography coupled with covalent adduct chemical ionization mass spectrometry (GC/CACI-MS). A multiple reaction monitoring method was developed to specifically characterize three 20:3 isomers found in duck eggs. Sciadonic acid was quantified at 0.13 % and 0.07 % (wt/wt of total fatty acids) in fresh and processed duck eggs, respectively. The major n-3 PUFA is DHA, with average concentrations of 1.44 %, 0.79 %, and 0.98 % in fresh, preserved, and salted duck eggs, respectively. For an average Chinese individual, the daily intake of DHA and total long-chain n-3 PUFA from all types of duck eggs is estimated to be 7.6 mg/day and 10.6 mg/day, respectively.

Plasma technology modifies the surface properties of desalted duck egg white and applied as a novel oleogel stabilizer

Xiaohan Dai,Bo Song,Xiaoqing Xiang,Bin Jiang,Dongmei Li,Chunhong Liu,Zhibiao Feng

Salted duck egg white (SDEW) is often discarded due to its high salt content. In this work, low-temperature plasma (LTP) technology was used to modify the functional properties of gelatinized desalted SDEW (gd-SDEW) through physical and chemical changes. After LTP treatment at 35 v for 3 min, gd-SDEW showed an increase in oil holding capacity. The analytical results indicated that the increase of β -sheets in the secondary structure, the surface hydrophobicity and the carbonyl content, the significant increase of BET surface area and decrease of adsorption mean pore size, and the increase of surface roughness were responsible for the increase of its oil holding capacity. LTP-treated gd-SDEW microgels can be used to stabilize the oleogels with excellent oil holding capacity, higher viscosity and gelation characteristics. This study offers the possibility of LTP as a novel technique to modify the surface properties of gd-SDEW for the reuse of SDEW.

Interfacial decoration of desalted duck egg white nanogels as stabilizer for Pickering emulsion

Xiaojuan Chen,Yaqiong Pei,Bojia Li,Yueming Wang,Bin Zhou,Bin Li,Hongshan Liang

Pickering emulsions , stabilized by food-grade protein colloidal particles, have demonstrated potential in food , cosmetics, and medical applications, while their environmental instability remains serious challenges. Herein, in this study, we have prepared a simple but effective interfacial decoration method based on the noncovalent interaction between desalted duck egg white nanogels (DEWN) and natural polyphenol for enhancing the interfacial and environmental stability of Pickering emulsions. As demonstrated, the introduction of tannic acid (TA) tuned near neutral wettability (θ o/w ∼90°) of the DEWN-TA (named DEWN-Tn) colloidal particles and significantly lowered the interfacial tension. Subsequently, the emulsions with distinct TA concentrations were further investigated. The results presented a decrement in the droplet sizes by adding appropriate concentrations of TA and long-term storage stability. The rheological behavior explored that the addition of TA did increase in both G′ and G″ compared with a solely DEWN-based system. Furthermore, it was worth noting that the decoration of TA under optimized conditions could also increase heat and pH stability and effectively inhibit “salting-out” effects of the Pickering emulsions system. Our study implies a possibility of fabricating interfacial architecture on the oil-water interface, which possesses great potential to be used as excellent food-grade emulsifiers.

The crosstalk between fatty acid metabolism disorder and mitochondrial dysfunction is involved in hepatotoxicity co-induced by cadmium and molybdenum in ShaoXing duck (Anas platyrhyncha) livers

Shanxin Li,Gaohui Nie,Xueyan Dai,Jiamei Zhu,Li Xiao,Sunan Wang,Jirong Chen,Jianrong Ren,Caiying Zhang

Excessive molybdenum (Mo) and cadmium (Cd) exert negative influences on organism. Our prior experiments attested that Mo and Cd brought liver damage in ShaoXing ducks ( Anas platyrhyncha ); this injury mechanism has not yet been fully clarified. Hence, the hepatotoxicity mechanisms of Mo and Cd were roundly analyzed by network toxicology in the study, the analysis confirmed that Mo and Cd primarily induce liver injury through lipid metabolism dysregulation, oxidative stress, and mitochondrial dysfunction. Further validation demonstrated that Mo and/or Cd upregulated fatty acid synthesis, reduced PPARβ/δ-mediated fatty acid β-oxidation, promoted free fatty acids (FFA) content and mitochondrial reactive oxygen species (mtROS) level, impaired integrality of mitochondrial membrane and mitochondrial function, thereby causing fatty acid metabolism disarray and mitochondrial dysfunction. Both PPARβ/δ activator GW0742 and mtROS inhibitor ROS-IN-1 could alleviate the changes of above these indexes. Collectively, findings from this research showed that Cd and Mo additively caused crosstalk between fatty acid metabolism disarray and mitochondrial dysfunction, thus inducing liver injury in ducks.

Removal of Dinotefuran, Thiacloprid, and Imidaclothiz Neonicotinoids in Water Using a Novel Pseudomonas monteilii FC02–Duckweed (Lemna aequinoctialis) Partnership

Cai Xiao-Yu,Xu Man,Zhu Yu-Xuan,Shi Ying,Wang Hong-Wei

Neonicotinoids (NEOs) are the most widely used insecticides in the world and pose a serious threat to aquatic ecosystems. The combined use of free-floating aquatic plants and associated microorganisms has a tremendous potential for remediating water contaminated by pesticides. The aim of this study was to determine whether plant growth-promoting bacteria (PGPB) could enhance the phytoremediation efficiency of duckweed (Lemna aequinoctialis) in NEO-contaminated water. A total of 18 different bacteria were isolated from pesticide-stressed agricultural soil. One of the isolates, Pseudomonas monteilii FC02, exhibited an excellent ability to promote duckweed growth and was selected for the NEO removal experiment. The influence of strain FC02 inoculation on the accumulation of three typical NEOs (dinotefuran, thiacloprid, and imidaclothiz) in plant tissues, the removal efficiency in water, and plant growth parameters were evaluated during the 14-day experimental period. The results showed that strain FC02 inoculation significantly (p < 0.05) increased plant biomass production and NEO accumulation in plant tissues. The maximum NEO removal efficiencies were observed in the inoculated duckweed treatment after 14 days, with 92.23, 87.75, and 96.42% for dinotefuran, thiacloprid, and imidaclothiz, respectively. This study offers a novel view on the bioremediation of NEOs in aquatic environments by a PGPB–duckweed partnership.
