AchMYC2 mediates ABA-induced suberin deposition by activating AchABCG6 during wound healing in harvested kiwifruit

Ziang Zhou,Chenggui Yang,Xiaopeng Wei,Yaoran Li,Huiyan Liu,Haitian Fang,Xiaobo Wei

Abscisic acid (ABA) is known to promote suberization, but its role in coordinating transmembrane transport processes has not been fully elucidated. In this study, wounded kiwifruit was treated with 0.5 mM ABA to investigate the regulatory mechanism underlying ABA-mediated suberin transport during wound healing. The results showed that ABA treatment significantly upregulated the expression of the AchMYC2 , AchABCG6-1 , AchABCG6-2 and increased ABCG transporter protein accumulation. Transient overexpression of AchMYC2 in kiwifruit significantly upregulated the transcription of AchABCGs and increased their protein abundance, concomitant with a marked enhancement of suberin deposition at wound sites. Subcellular localization analyses further revealed that AchABCG6-1 and AchABCG6-2 are predominantly localized to the plasma membrane. Consistently, transient overexpression of either AchABCG6-1 or AchABCG6-2 significantly promoted suberin accumulation, indicating that these transporters play essential roles in the transmembrane transport of suberin precursors and thereby facilitate wound-induced suberization. DNA affinity purification sequencing (DAP-seq) identified “CACGTG” as the conserved binding motif of AchMYC2, while yeast one-hybrid (Y1H), Dual-luciferase reporter (Dual-luc), and molecular docking (MD) further confirmed the direct binding of AchMYC2 to G-box motifs in the promoters of AchABCGs and the potential interaction of ABCG transporters with suberin monomers. Collectively, these results demonstrate that ABA activates an AchMYC2–ABCG regulatory module to enhance transmembrane transport of suberin monomers, thereby promoting suberin deposition and accelerating wound healing in kiwifruit.

Boric acid-functionalized PANI-ABA/PA hydrogel-based flexible pH sensor for real-time sweat monitoring

Meihui Gao,Xianjing Zhang,Shiqi Li,He Gao,Xinyu Li,Dongyuan Xu,Longbin Xu

Background Wearable pH sensors have emerged as promising tools for real-time health monitoring, particularly for sports and clinical applications, due to their ability to monitor key physiological indicators such as sweat pH. Sweat analysis provides valuable insights into metabolic processes and physical exertion. However, the development of flexible, stable, and highly sensitive sensors for accurate sweat pH detection remains a challenge. Conductive polymer such as polyaniline (PANI), have gained attention due to their unique properties, but further improvements in mechanical flexibility and sensor performance are needed to fully realize their potential for wearable biosensing applications. Results We present a boric acid-functionalized PANI-ABA/PA hydrogel, synthesized via electrochemical deposition, where boric acid plays a crucial role in crosslinking the rigid PANI material into a flexible, porous grid structure. This crosslinking improves the mechanical properties of the hydrogel while enhancing its ion-exchange capacity, leading to greater sensitivity and selectivity for H + ions. PANI-ABA/PA hydrogel exhibits super-Nernstian behavior (77.80 mV pH −1 ) across the physiological pH range (3.00–10.00), with rapid response times, minimal temperature drift, and excellent repeatability. Moreover, the hydrogel exhibits outstanding antifouling properties, ensuring consistent performance even in dynamic environments. Real-time human subject testing confirms the sensor's ability to accurately monitor sweat pH during physical activity, providing reliable and real-time data. These results highlight the promising potential of the PANI-based hydrogel for wearable biosensors in health monitoring. Significance and novelty This study highlights the role of boric acid in facilitating the formation of a crosslinked PANI-based grid structure, which improves the sensor's performance in wearable applications. Enhanced sensitivity, stability, and mechanical flexibility make this smart wearable sensor a promising candidate for continuous, non-invasive health monitoring. It opens new opportunities for real-time athletic performance tracking, personalized healthcare, and disease diagnostics, positioning it as a leading technology for future wearable health-monitoring systems.

Pyrrole-Containing ABA Triblock Brush Polymers as Dual Functional Molecules to Facilely Access Diverse Mesostructured Materials

Guokang He,Peng Wang,Ning Gao,Xianpeng Yin,Fuwei Sun,Wenyun Li,Hongwei Zhao,Chen Wang,Guangtao Li

We report a new type of ABA-type brush-like triblock copolymer (PEGMA-b-PPHMA-b-PEGMA), which is characterized by two hydrophilic end segments (A) and a hydrophobic midblock (B) bearing polymerizable pyrrole moieties. This kind of symmetric triblock copolymers can not only serve as a structure-directing agent for the formation of mesoporous silicates but also function as a polymerizable monomer or carbon source for further chemical conversion to deliver mesostructured polymer or carbon materials within the preformed silicate framework. By utilizing such dual functional macromolecules, rich morphologies of mesostructures are accessible through microphase separation, and the order–order transition among cubic, lamellar, 2D hexagonal, and bicontinuous mesostructures is sensitively achievable by varying the hydrophilic volume fraction of copolymers and TEOS dosage. These features enable facile access to diverse mesostructured silica, polymer, and carbon materials. Remarkably, due to the brush-like polymer architecture and the formation of loop configuration, a widely tunable pore size from mesoscale to macroscale is realizable through using a relatively lower molecular weight copolymer. Importantly, it is found that the brush-like hydrophilic segments of our triblock copolymers can firmly anchor into the silica walls without appreciable escape of copolymer template in commonly used solvents even under reflux extraction, providing a prerequisite for reliable confined chemical transformation into mesostructured polymer and carbon materials. Our work demonstrates that the design and synthesis of functional amphiphilic block copolymers will bring great opportunity for the development of novel mesostructured materials.

R2R3-MYB Transcription Factor NtMYB330 Regulates Proanthocyanidin Biosynthesis and Seed Germination in Tobacco (Nicotiana tabacum L.)

Zhao Lu,Song Zhongbang,Wang Bingwu,Gao Yulong,Shi Junli,Sui Xueyi,Chen Xuejun,Zhang Yihan,Li Yongping

Proanthocyanidins (PAs) are important phenolic compounds and PA biosynthesis is regulated by a ternary MBW complex consisting of a R2R3-MYB regulator, a bHLH factor and a WDR protein. In this study, a tobacco R2R3-MYB factor NtMYB330 was characterized as the PA-specific regulator in which the PA biosynthesis was promoted in the flowers of NtMYB330-overexpressing lines while decreased in the flowers of ntmyb330 mutants. NtMYB330 can interact with flavonoid-related bHLH partner NtAn1b and WDR protein NtAn11-1, and the NtMYB330-NtAn1b complex is required to achieve strong transcriptional activation of the PA-related structural genes NtDFR1, NtANS1, NtLAR1 and NtANR1. Our data reveal that NtMYB330 regulates PA biosynthesis in seeds and affects seed germination, in which NtMYB330-overexpressing lines showed higher PA accumulations in seed coats and inhibited germination, while ntmyb330 mutants had reduced seed coat PAs and improved germination. NtMYB330 affects seed germination possibly through two mechanisms: modulating seed coat PAs to affect coat-imposed dormancy. In addition, NtMYB330 regulates the expressions of abscisic acid (ABA) and gibberellin acid (GA) signaling-related genes, affecting ABA-GA crosstalk and seed germination. This study reveals that NtMYB330 specifically regulates PA biosynthesis via formation of the MBW complex in tobacco flowers and affects germination through adjustment of PA concentrations and ABA/GA signaling in tobacco seeds.

Chloroplast vesiculation fine-tunes seed germination by antagonizing ABA signaling through dual degradation of ABI5 and XIW1

Xiaoyu Cui,Liqing Feng,Jiayin Li,Shan Gao,Shunping Yan,Ting Pan

SUMMARY Abscisic acid (ABA) plays a crucial role in enforcing seed dormancy and suppressing germination under stress conditions by stabilizing key transcriptional repressors. However, the mechanisms by which plants actively attenuate this repression to resume developmental growth remain largely unclear. In this study, we identify chloroplast vesiculation (CV) as a negative regulator of ABA signaling in Arabidopsis. Silencing of CV resulted in hypersensitivity to ABA during seed germination and stomatal closure, whereas its overexpression conferred ABA insensitivity. We demonstrate that CV physically interacts with and directly promotes the vacuolar degradation of two key positive regulators of ABA signaling—the transcription factor ABI5 and the WD40-repeat protein XIW1. Genetic epistasis analyses confirmed that ABI5 and XIW1 function downstream of CV, as loss-of-function mutations in either ABI5 or XIW1 suppressed the ABA-hypersensitive phenotype of CV-silenced plants. Furthermore, we found that ABA itself negatively regulates CV at both the transcriptional and post-translational levels, forming a feedback loop that fine-tunes ABA sensitivity. Our study thus uncovers a previously unknown regulatory module in which CV attenuates ABA responses by targeting ABI5 and XIW1 for vacuolar degradation, thereby facilitating the transition from seed dormancy to germination.

Global hypermethylation of the N6-methyladenosine RNA modification associated with apple heterografting

Xu Jidi,He Jieqiang,Hu Bichun,Hou Nan,Guo Junxing,Wang Caixia,Li Xuewei,Li Zhongxing,Zhai Jingjing,Zhang Ting,Ma Chuang,Ma Fengwang,Guan Qingmei

Grafting can facilitate better scion performance and is widely used in plants. Numerous studies have studied the involvement of mRNAs, small RNAs, and epigenetic regulations in the grafting process. However, it remains unclear whether the mRNA N6-methyladenosine (m6A) modification participates in the apple (Malus x domestica Borkh.) grafting process. Here, we decoded the landscape of m6A modification profiles in ‘Golden delicious’ (a cultivar, Gd) and Malus prunifolia ‘Fupingqiuzi’ (a unique rootstock with resistance to environmental stresses, Mp), as well as their heterografted and self-grafted plants. Interestingly, global hypermethylation of m6A occurred in both heterografted scion and rootstock compared with their self-grafting controls. Gene Ontology (GO) term enrichment analysis showed that grafting-induced differentially m6A-modified genes were mainly involved in RNA processing, epigenetic regulation, stress response, and development. Differentially m6A-modified genes harboring expression alterations were mainly involved in various stress responses and fatty acid metabolism. Furthermore, grafting-induced mobile mRNAs with m6A and gene expression alterations mainly participated in ABA synthesis and transport (e.g. carotenoid cleavage dioxygenase 1 [CCD1] and ATP-binding cassette G22 [ABCG22]) and abiotic and biotic stress responses, which might contribute to the better performance of heterografted plants. Additionally, the DNA methylome analysis also demonstrated the DNA methylation alterations during grafting. Downregulated expression of m6A methyltransferase gene MdMTA (ortholog of METTL3) in apples induced the global m6A hypomethylation and distinctly activated the expression level of DNA demethylase gene MdROS1 (REPRESSOR OF SILENCING 1) showing the possible association between m6A and 5mC methylation in apples. Our results reveal the m6A modification profiles in the apple grafting process and enhance our understanding of the m6A regulatory mechanism in plant biological processes.

Abscisic acid activates transcription factor module MdABI5–MdMYBS1 during carotenoid-derived apple fruit coloration

Jia Dongjie,Li Yuchen,Jia Kun,Huang Benchang,Dang Qingyuan,Wang Huimin,Wang Xinyuan,Li Chunyu,Zhang Yugang,Nie Jiyun,Yuan Yongbing

Carotenoids are major pigments contributing to fruit coloration. We previously reported that the apple (Malus domestica Borkh.) mutant fruits of ‘Beni Shogun’ and ‘Yanfu 3’ show a marked difference in fruit coloration. However, the regulatory mechanism underlying this phenomenon remains unclear. In this study, we determined that carotenoid is the main factor influencing fruit flesh color. We identified an R1-type MYB transcription factor, MdMYBS1, which was found to be highly associated with carotenoids and abscisic acid (ABA) contents of apple fruits. Overexpression of MdMYBS1 promoted, and silencing of MdMYBS1 repressed, β-branch carotenoids synthesis and ABA accumulation. MdMYBS1 regulates carotenoid biosynthesis by directly activating the major carotenoid biosynthetic genes encoding phytoene synthase (MdPSY2-1) and lycopene β-cyclase (MdLCYb). 9-cis-epoxycarotenoid dioxygenase 1 (MdNCED1) contributes to ABA biosynthesis, and MdMYBS1 enhances endogenous ABA accumulation by activating the MdNCED1 promoter. In addition, the basic leucine zipper domain transcription factor ABSCISIC ACID-INSENSITIVE5 (MdABI5) was identified as an upstream activator of MdMYBS1, which promotes carotenoid and ABA accumulation. Furthermore, ABA promotes carotenoid biosynthesis and enhances MdMYBS1 and MdABI5 promoter activities. Our findings demonstrate that the MdABI5-MdMYBS1 cascade activated by ABA regulates carotenoid-derived fruit coloration and ABA accumulation in apple, providing avenues in breeding and planting for improvement of fruit coloration and quality.

Transcription factor Constans-like 4 may regulate tomato fruit ripening by interacting with abscisic acid and ethylene signaling

Qiong Wu,Xiaoya Tao,Chunxiao Cui,Yanan He,Dongdong Zhang,Yurong Zhang,Li Li

Phytohormones are vital for tomato ripening, and abscisic acid (ABA) is speculated to induce ethylene biosynthesis at the initiate of tomato fruit ripening; however, its interaction sites remain unclear. In the present study, Constans-like 4 (SlCOL4), a CO/COL transcription factor, was cloned from tomato fruit. Amino acid sequence and subcellular location analysis proved that SlCOL4 was a typical transcription factor of the COL family. Gene expression analysis showed SlCOL4 expressed in different tissues of tomato plants and its transcription abundance declined at the onset of fruit ripening, coincidentally it was found to be down-regulated in ABA-treated tomato fruit and responded to exogenous ABA within 6 h after treatment. Meanwhile, yeast one-hybrid and dual-luciferase analysis showed that the ABA response factor SlAREB1 inhibited the transcription of SlCOL4 , indicating it was directly regulated by ABA at the mature green stage of tomato fruit . In addition, the pericarp color turned red faster than that of control fruit, and the key ethylene biosynthesis genes, ACS2, ACS4, and ACO1 , were up-regulated in SlCOL4 silenced tomato fruit, suggesting that SlCOL4 may be a negative regulator of ripening. Furthermore, several genes associated with the ethylene signaling pathway were identified downstream of SlCOL4 by chromatin immunoprecipitation sequencing (ChIP-Seq) analysis. Overall, the transcription factor SlCOL4 may interact with ABA and ethylene signals and negatively regulate tomato fruit ripening. The results in this study provide new insights for elucidating the molecular mechanism of ABA-ethylene interaction in regulating tomato fruit ripening, and provide references for further research on the role of COLs transcription factors in tomato fruit ripening as well.

WEAK SEED DORMANCY 1, an aminotransferase protein, regulates seed dormancy in rice through the GA and ABA pathways

Yunshuai Huang,Jiawei Song,Qixian Hao,Changling Mou,Hongming Wu,Fulin Zhang,Ziyan Zhu,Ping Wang,Tengfei Ma,Kai Fu,Yaping Chen,Thanhliem Nguyen,Shijia Liu,Ling Jiang,Jianmin Wan

Seed dormancy is a critical trait that enhances plant survival by preventing seed germination at the wrong time or under unsuitable conditions. Lack of seed dormancy in rice can lead to pre-harvest sprouting on mother plants leading to reduced yield and seed quality. Although some genes have been identified, knowledge of regulation of seed dormancy is limited. Here, we characterized a weak seed dormancy mutant named weak seed dormancy 1 ( wsd1 ) that showed a higher seed germination percentage than the wild-type following the harvest ripeness. We cloned the WSD1 encoding an aminotransferase protein using a MutMap approach. WSD1 was stably expressed after imbibition and its protein was localized in the endoplasm reticulum. A widely targeted metabolomics assay and amino acid analysis showed that WSD1 had a role in regulating homeostasis of amino acids. PAC treatment and RNA-seq analysis showed that WSD1 regulates seed dormancy by involvement in the GA biosynthesis pathway. GA 1 content and expression of GA biosynthesis-related genes were increased in the wsd1 mutant compared with the wild-type. The wsd1 mutant had reduced sensitivity to ABA. Our overall results indicated that WSD1 regulates seed dormancy by balancing the ABA and GA pathways.

ABA-responsive transcription factor bZIP1 is involved in modulating biosynthesis of phenolic acids and tanshinones in Salvia miltiorrhiza

Deng Changping,Shi Min,Fu Rong,Zhang Yi,Wang Qiang,Zhou Yang,Wang Yao,Ma Xingyuan,Kai Guoyin

Phenolic acids and tanshinones are major bioactive ingredients in Salvia miltiorrhiza, which possess pharmacological activities with great market demand. However, transcriptional regulation of phenolic acid and tanshinone biosynthesis remains poorly understood. Here, a basic leucine zipper transcription factor (TF) named SmbZIP1 was screened from the abscisic acid (ABA)-induced transcriptome library. Overexpression of SmbZIP1 positively promoted phenolic acid biosynthesis by enhancing expression of biosynthetic genes such as cinnamate-4-hydroxylase (C4H1). Furthermore, biochemical experiments revealed that SmbZIP1 bound the G-Box-like1 element in the promoter of the C4H1 gene. Meanwhile, SmbZIP1 inhibited accumulation of tanshinones mainly by suppressing the expression of biosynthetic genes including geranylgeranyl diphosphate synthase (GGPPS) which was confirmed as a target gene by in vitro and in vivo experiments. In contrast, the phenolic acid content was reduced and tanshinone was enhanced in CRISPR/Cas9 [clustered regularly interspaced palindromic repeats (CRISPR)/CRISPR-associated protein 9]-mediated knockout lines. In addition, the previously reported positive regulator of tanshinone biosynthesis, SmERF1L1, was found to be inhibited in SmbZIP1 overexpression lines indicated by RNA sequencing, and was proven to be the target of SmbZIP1. In summary, this work uncovers a novel regulator and deepens our understanding of the transcriptional and regulatory mechanisms of phenolic acid and tanshinone biosynthesis, and also sheds new light on metabolic engineering in S. miltiorrhiza.

Integrated metabolome and transcriptome analyses reveal the seasonal dynamic accumulation and regulation network of benzylisoquinoline alkaloids in Stephania tetrandra roots

Jiqiang Hu,Nannan Tu,Cuiying Lin,Yuzhe Fan,Zhijian Xu,Jianbiao Yao,Su Zeng,Bingxian Yang,Lianli Sun

Stephania tetrandra is a medicinal plant that contains four main types of benzylisoquinoline alkaloids (BIAs): mono-BIAs, bis-BIAs, aporphine-type BIAs (APT-BIA), and tetrahydroprotoberberines (THBs). S. tetrandra is particularly rich in the bis-BIAs, tetrandrine and fangchinoline, which are critical quality markers and major components of the traditional Chinese medicine “Fangji”. However, changes in their BIA profiles and associated metabolic regulatory networks over time remain unclear. In this study, metabolomics and transcriptomics were integrated to investigate the dynamic accumulation patterns and underlying mechanisms of these alkaloids across three developmental stages: Autumnal Equinox (AuEq), Winter Solstice (WinSo), and Vernal Equinox (VeEq). Quantification revealed that both alkaloids reached their highest concentrations at VeEq, with tetrandrine levels more than doubling from WinSo. Untargeted metabolomics identified 61 differentially accumulated BIAs, displaying stage-specific accumulation patterns. Hierarchical clustering grouped these BIAs into four structural categories, predominantly enriched at VeEq. Transcriptome profiling showed significant upregulation of genes involved in isoquinoline alkaloid biosynthesis during VeEq, especially key enzymes such as TyrAT, TYDC, 4HPPDC, and NCS. Co-expression and protein interaction network analyses further identified abscisic acid (ABA) signaling pathways as major hormonal regulators. Notably, ABA levels and exogenous ABA application confirmed its pivotal role in promoting bis-BIA biosynthesis, acting in concert with reactive oxygen species (ROS) to fine-tune alkaloid production. Collectively, this study establishes a hormone–ROS–BIA regulatory framework that explains seasonal fluctuations in alkaloid content and provides new entry points for improving the medicinal quality and cultivation strategies of S. tetrandra .

Enhancing sweet sorghum emergence and stress resilience in saline-alkaline soils through ABA seed priming: insights into hormonal and metabolic reprogramming

Jianfeng Yang,Wenlan Zhang,Tianyu Wang,Jiawei Xu,Jinjing Wang,Jiahao Huang,Yingpeng Sun,Yu Ni,Yanjun Guo

Sweet sorghum ( Sorghum bicolor Moench) seedling emergence and growth are significantly impeded by physical soil crusts (PSCs) in saline-alkaline soils. Abscisic acid (ABA) is a potent seed priming agent known for modulating plant physiological and metabolic responses under salinity stress. However, the influence of ABA priming on seedling emergence in PSCs remains unclear. This study conducted both pot and field experiment to examine the effects of ABA priming on enhancing seedling emergence under PSC conditions. ABA priming altered the balance of at least 24 endogenous phytohormones, including abscisic acid, jasmonic acid, gibberellins, ethylene, auxins, and cytokinins. Additionally, it reprogrammed starch and sucrose metabolism, resulting in the differential expression of genes encoding key enzymes such as AMY, BAM, and INV, which are crucial for converting complex sugars into readily available energy sources, thereby supporting seedling growth. Furthermore, 52 differentially expressed metabolites (DEMs) of flavonoids were identified in germinating seedlings, including 15 anthocyanins, 3 flavones, 7 flavonols, 6 isoflavones, 7 flavanones, and 14 other flavonoids. Genetic and metabolic co-expression network analysis, along with flavonoid biosynthesis pathway exploration, revealed that the biosynthesis of 17 key DEMs—including liquiritigenin, apigenin, kaempferide, syringetin, phloretin, formononetin, dihydrokaempferol, and xanthohumol—was regulated by 10 differentially expressed genes (DEGs) associated with flavonoid biosynthesis. These DEGs encoded 7 enzymes critical for this pathway, including chalcone synthase, shikimate O-hydroxycinnamoyltransferase, bifunctional dihydroflavonol 4-reductase, naringenin 7-O-methyltransferase, and anthocyanidin reductase. This regulation, along with reduced levels of superoxide anion (O 2 − ) and malondialdehyde and increased antioxidant enzyme activities, suggested that flavonoids played a vital role in mitigating oxidative stress. These findings demonstrate that ABA priming can effectively enhance sweet sorghum seedling emergence in PSCs by accelerating emergence and boosting stress resistance.
