An electrochemiluminescence biosensor based on Graphitic carbon nitride luminescence quenching for detection of AFB1

Dongyan Tian,Jie Wang,Qiandong Zhuang,Songmei Wu,Yu Yu,Kejian Ding

Based on graphite-like carbon nitride (g-CN) nanocomposites coupled with aptamer, a regenerable electrochemiluminescence (ECL) biosensor is developed for the quantitative detection of aflatoxin B 1 (AFB 1 ). In the existence of AFB 1 , the structure of the aptamer changed into a loop, and the original ECL intensity was reduced owing to the enhancement of luminescence quenching between the ferrocene modified at the end of the aptamer and the luminescent substrate g-CN. Moreover, AFB 1 with oxidation state could also react with high energy state g-CN, leading to further reduction of the electrochemiluminescence signal. At optimum conditions, ECL intensity was decreased in linearity with an AFB 1 concentration range from 0.005 ng/mL to 10 ng/mL, and the minimum detectable concentration was down to 0.005 ng/mL, which realized trace detection demand with high sensitivity. It was selective for AFB 1 and its performance had been verified on rice samples, which indicated a promising applying prospect of non-enzymatic electrochemiluminescence AFB 1 detection.

Nitrogen-doped carbon dot and DNA tetrahedron nanostructure based electrochemiluminescence aptasensor for AFB1 detection

Ying Li,Pengyue Song,Qingbin Xu,Wenjuan Wu,Nan Long,Jiabo Wang,Lidong Zhou,Ruile Pan,Weijun Kong

An electrochemiluminescence (ECL) aptasensor with aptamer as recognition element, N-doped carbon dots (N-CDs) as luminophore, and DNA tetrahedral nanostructure (DTN) as carrier to loading abundant N-CDs for signal amplification was developed for ultra-sensitive aflatoxin B 1 (AFB 1 ) detection. The complementary DNA (cDNA) was firstly immobilized on the Au electrode (AuE) surface to hybridize with the AFB 1 aptamer. In the presence of AFB 1 , it would competitively with cDNA for binding the aptamer, causing the exposure of cDNA. DTN modified with AFB 1 aptamer on top and N-CDs on the four vertices would combine with cDNA onto the AuE surface to generate ECL signals due to electron transfer between co-reactants (K 2 S 2 O 8 ) and N-CDs, which was positively related to the concentration of AFB 1 . Due to the high specificity of aptamer, the outstanding ECL performance of N-CDs and the enhanced signal output of DTN/N-CDs probes, the as-constructed ECL aptasensor realized excellent sensitivity with a low detection limit of 77 pg/mL and a wide linear concentration of 1.0 × 10 −1 -1.0 × 10 4 ng/mL for AFB 1 , as well as fascinating accuracy with acceptable recoveries of 89.54–111.0% in the spiked Coix seed matrices. Successful application for AFB 1 detection in actual samples confirmed the extensive practicability of the ECL aptasensor, providing a universal platform for more trace contaminants by altering the corresponding aptamers.

Synthesis and Characterization of Different Crystalline Calcium Silicate Hydrate: Application for the Removal of Aflatoxin B1 from Aqueous Solution

Lu Zeng,Ligang Yang,Shuping Wang,Kai Yang

Different crystalline calcium silicate hydrates (CSH) were synthesized under specific hydrothermal conditions and several methods were used to analyze samples. Amorphous calcium silicate hydrates (ACSH) mainly consists of disordered calcium silicate hydrate gel (C-S-H gel) and crystalline calcium silicate hydrates (CCSH) consists of crystallized tobermorite. The adsorption of carcinogenic aflatoxin B1 (AFB1) onto ACSH and CCSH was investigated. The adsorption kinetics was studied using pseudo-first-order and pseudo-second-order kinetic models and intraparticle diffusion model. The pseudo-second-order model provided the best correlation and the intraparticle diffusion controlled the adsorption process of AFB1 onto CCSH. Adsorption isotherm parameters were obtained from Langmuir and Freundlich and the adsorption data fitted to Freundlich much better. Based on the results of N2 adsorption/desorption, adsorption kinetics, and adsorption isotherms, the adsorption mechanism of AFB1 onto CCSH was developed. All results indicate that CCSH has a great potential to be a safe, easy-made, and cost-effective material for the control of AFB1 contamination.

A novel COOH–GO–COOH–MWNT/pDA/AuNPs based electrochemical aptasensor for detection of AFB1

Pengfei Wang,Bin Luo,Ke Liu,Cheng Wang,Hongtu Dong,Xiaodong Wang,Peichen Hou,Aixue Li

Aflatoxin B1 (AFB1), one of the most common mycotoxins in food matrixes, has been identified as the most toxic contaminant with mutagenic, teratogenic, immunosuppressive, and carcinogenic effects. In this study, an electrochemical aptamer sensor was developed for the on-site detection of AFB1. Carboxylated graphene oxide (COOH–GO) and carboxylated multi-walled carbon nanotubes (COOH–MWNT) nanocomposites, dopamine polymers (pDA) and gold nanoparticles (AuNPs) were used to enhance the electrochemical activity and the biocompatibility of the screen-printed electrodes (SPE). Once AFB1 was captured by the aptamer immobilized on the electrode surface, the redox current of [Fe(CN)6]3−/4− decreased. Therefore, the binding of aptamer (Apt) and AFB1 can be reflected by the change of the peak current. The as-prepared sensor showed a wide detection range of 0.1 fg ml−1–100 pg ml−1 and a low detection limit of 15.16 ag ml−1. It is also simple and low-cost, which shows great potential in practical application.

A novel SERS/electrochemical dual-channel aptasensor for the detection of aflatoxin (AFB1) in cereal foods

Xinran Yang,Chuanjin Cui,Haibin Liu,Hongshuo Chen,Xueli Zhang,Zonglin Li,Xuechao Zhang,Qian Li

This study developed a novel SERS/electrochemical dual-channel aptasensor for the highly sensitive detection of Aflatoxin B1 (AFB 1 ). Firstly, Fe₃O₄@Au@Ag nanoparticles, synthesized by a seed-mediated growth method, were functionalized with AFB1 aptamers to serve as capture probes. Secondly,ssDNA (AFB 1 aptamer complementary chain) was modified on the AuNs@4-MBA@Ag surface for obtaining the signal probes. Then, the competitive biosensors were constructed based on competition between capture probe and AFB 1 . The sediment separated under neodymium magnet influence was employed for surface-enhanced Raman spectroscopy (SERS) testing, whilst the supernatant (dissociation signal probe) was placed upon a modified gold electrode for electrochemical analysis. The synthesized capture and signal probes were characterized through SEM, TEM, and XPS, thus confirming their successful synthesis. More importantly, the sample results showed that the detection limit (LOD) of this biosensor was 2 × 10 −6  ng/mL, the sample recovery rate was 91–117 %, which is sufficient for trace detection of AFB 1 .

A fluorescent aptasensor for highly sensitive detection of OTA and AFB1 and constructing their logical operations

Qiu-Long Zhang,Xue Yang,Xuan-Ting Lin,Sha-Sha Chen,Zhu,Guang-Xiang Luan,Kun-Quan Chen,Zhi Zeng,Jian-Min Chen

In this study, a universal fluorescent aptasensor was developed for the sensitive detection of ochratoxin A (OTA) and aflatoxin B1 (AFB1). The design integrated a non-competitive transduction platform with highly efficient RNA-cleaving DNAzymes, forming the core of the sensing system. When OTA or AFB1 is present in the sample, the target mycotoxin specifically recognizes and binds to its corresponding aptamer. The strand displacement reaction is significantly hastened by this binding event, and in response, the DNAzyme is released as a consequence. The activated DNAzyme then triggers an amplified fluorescent signal, enabling quantitative detection of the target. The proposed aptasensor displayed sensitivity with a detection limit of 0.26 nM for AFB1 and 0.17 nM for OTA respectively. Beyond single-target detection, the platform also enabled simple and flexible implementation of logical operations between the two mycotoxins, along with the design of orthogonal experiments for multi-target analysis. Furthermore, specificity tests confirmed that other co-existing mycotoxins had negligible interference on the detection of AFB1 or OTA, verifying the sensor's high selectivity. To assess practical applicability, the aptasensor was utilized for the detection of AFB1 and OTA in actual samples, yielding recoveries of 93 %–101 % in milk and 101 %–107 % in corn flour, demonstrating the sensor's reliability in complex matrices. Collectively, this developed fluorescent aptasensor features portability, simplicity, cost-effectiveness, and broad adaptability, making it a promising alternative tool for detecting various mycotoxins in food safety monitoring and related fields.

A dual signal ratio electrochemical sensor based on DNA tetrahedrons and bimetallic nanocomposites for AFB1 analysis

Liang Guo,Liying Zhao,Shijin Zhou,Yanju Liu,Jinming Kong,Huaixia Yang

NGR-HP-AgPt and DNA tetrahedrons build a 3D track, avoiding probe entanglement. • Exo III-driven DNA walker achieves dual-signal ratiometric sensing of AFB 1 . • The built-in self-calibration effectively eliminates false-positive artifacts. • Ultrasensitive assay (LOD 73.99 fg/mL) successfully applied in complex matrices. Given the hepatotoxicity and widespread contamination of Aflatoxin B 1 (AFB 1 ), developing ultra-sensitive, anti-interference analytical platforms is paramount for public health. Herein, a dual signal ratio electrochemical sensing platform for accurate AFB 1 analysis was constructed integrating DNA tetrahedrons (TNDA), a dynamic DNA walker, and nitrogen-doped graphene oxide-supported hollow silver-platinum bimetallic nanospheres (NGR-HP-AgPt). The NGR-HP-AgPt cavity provides a uniform microenvironment, synergistically promoting catalytic effects to significantly accelerate interfacial electron transfer. Simultaneously, rigid 3D TNDA prevent spatial probe entanglement, providing a well-oriented track. To avoid false-positive artifacts in complex matrices, a competitive binding strategy is employed. Upon target recognition, AFB 1 displaces a complementary sequence (DNA1) from the aptamer. The released DNA1 acts as a walking strand, hybridizing with signal probes (Cd 2+ -DNA2). Subsequently, Exonuclease III initiates the DNA walker, continuously cleaving probes to amplify the ratiometric signal variation for reliable self-calibration. Under optimal conditions, this sensor exhibits a broad linear range from 2 × 10 −4 to 20 ng/mL, with an ultra-low detection limit of 73.99 fg/mL. Furthermore, its practical utility and high accuracy were successfully validated in complex food and medicinal matrices, yielding consistent results with the standard HPLC-FL method. This work broadens the robust design paradigm of anti-interference biosensing. Download: Download high-res image (296KB) Download: Download full-size image

Aptamer-based lateral flow strip for rapid detection of AFB1 in peanuts, soybeans, corn, and rice

Sixian Wang,Zhuoer Chen,Shuang Jiang,Xiaoling Zheng,Yueyang Ren,Xunqing Nie,Wenyu Xiang,Xianlu Lei,Qi Sun,Tao Le

Aflatoxin B1 (AFB1), a Group 1 carcinogenic mycotoxin, poses a significant threat to human health by contaminating crops. It is urgent to establish a simple and rapid detection method to promptly identify the risk of AFB1 residues, thereby mitigating its adverse health effects and reducing economic losses. In this study, a lateral flow strip (LFS) via a high-performance aptamer as the recognition element was developed for the detection of AFB1 in crops. Firstly, Aptamer7 was selected from 8 reported AFB1 aptamers employing molecular dynamics and the SYBR Green I assay, demonstrating good affinity and stability. Subsequently, gold nanoparticles (AuNPs)-based LFS was prepared with the application of Aptamer7. The results showed that the linear detection range was 1–200 ng/mL, with a limit of detection (LOD) of 0.23 ng/mL and strong anti-interference capability. Finally, the method's reliability was evaluated through AFB1-spiked samples. The average recoveries ranged from 92.51 to 119.21 %, with coefficients of variation (CV) between 1.47 and 6.32 % in peanuts, soybeans, corn, and rice. Moreover, the test results generated via the established method showed high consistency with those derived from high-performance liquid chromatography (HPLC), indicating that the LFS provides an alternative and promising strategy for monitoring AFB1 in crops.

Electrochemiluminescence Immunosensor Based on Platinum Electrode Modified with TiO2@ATO Nanocomposite for Sensitive Detection of AFB1

Shuting Cheng,Guanglei Chu,Zengning Liu,Yemin Guo,Xia Sun

A novel electrochemiluminescence (ECL) immunosensor was fabricated by using titanium dioxide (TiO2) and antimony tin oxide (ATO) nanocomposite as electrode modified material to detect aflatoxin B1 (AFB1). Luminol with ECL characteristics was modified on the surface of a platinum electrode to prepare the luminescent electrode. The buffer solution contained hydrogen peroxide (H2O2) was used to monitor the ECL signals owing to the chemiluminescence properties of luminol-H2O2 in alkaline solution. TiO2 and ATO were doped with chitosan to form TiO2@ATO nanocomposite, which was modified on the electrode to enhance the ECL intensity by accelerating the electron transfer and provided more binding sites for aflatoxin B1 antibody (Ab) and AFB1. An ECL immunosensor was prepared for AFB1 detection by modifying Ab and bovine serum albumin (BSA) successively. The specific combination of AFB1 and Ab weakened the ECL intensity, thus the concentration of AFB1 was determined according to the strength change of ECL signal. Under optimum conditions, the constructed immunosensor provided a wide linear range from 1.0 × 10−2 to 1.0 × 103 ng mL−1 with a detection limit of 0.03 ng mL−1 (S/N = 3) and showed great specificity and high stability. Furthermore, the proposed immunosensor successfully demonstrated its applicability for detecting AFB1 in peanut oil samples.

Potential-resolved differential electrochemiluminescence immunosensor based on novel designed IBPHF for self-correctable detection of AFB1

Xiaoyi Lv,Fei Tan,Tian Miao,Jihui Zhang,Zhiwen Zhang,Bo Cui,Yishan Fang

Electrochemiluminophore system with potential-resolved has exhibited giant potentiality for the application of bioassays in recent years, however, rarely studies toward the novel luminophore with potential-resolved electrochemiluminescence (PRECL) have been reported. In this work, a novel organoboron complex microrod (IBPHF) with PRECL property was synthesized, which emitted two strongest electrochemiluminescence (ECL) signals in the anode region at around 1.5 V and cathode region at around −1.5 V by using tri- n -propylamine (TPA)/K 2 S 2 O 8 as coreactant. The relevant ECL properties and luminescence mechanisms have been discussed in detail. Moreover, electroactive substrate of three-dimensional porous starch carbon nanomaterials (3D-PSC NPs) were designed to capture antibodies. As a result, the competitive potential-resolved differential electrochemiluminescence immunosensor was fabricated, and greatly enhanced sensitivity was achieved by using IBPHF by using IBPHF as a carrier to label competing antigens. In the presence of aflatoxin B 1 (AFB 1 ), due to the competitive mechanism and the specific recognition of antigen–antibody, the well-prepared immunosensor performed a quantitative decreasement in the range from 50 fg/mL to 100 ng/mL with a minimum detectable limit of 16.44 fg/mL by using the intensity difference (△ECL) (ECL anode -ECL cathode ). The self-correction of △ECL extremely enhanced the accuracy and reliability of the sensor. Furthermore, the designed sensor achieved excellent detection towards AFB 1 in walnut samples, and performed a bright application for trace target detection in future.

A dual-modal biosensor coupling cooperative catalysis strategy for sensitive detection of AFB1 in agri-products

Wenting Li,Xinai Zhang,Yongqiang Shi,Xuetao Hu,Xin Wang,Nini Liang,Tingting Shen,Xiaobo Zou,Jiyong Shi

Herein, the cooperative catalysis effect between nanocomposite (AgPd NPs/POD-M/PEI-rGO) and horseradish peroxidase (HRP) was applied for the fast and sensitive detection of aflatoxin B1 (AFB 1 ). Upon specific and competitive binding of HRP@DNA and AFB 1 to cDNA, the working electrode presented different catalytic capacities for supporting electrolytes (TMB and H 2 O 2 ). In the redox mechanism of TMB and H 2 O 2 , HRP and nanocomposite effectively catalyzed the oxidization of TMB to form the one-electron oxidation intermediate TMB+, and contributed the electrical signals and absorbance signals. Electrochemistry and colorimetric analyses were successfully realized for AFB 1 detection with 0.2 pg/mL and 8 pg/mL of detection limits, respectively, which is much lower than that of traditional HPLC methods. Overall, this method had significant reliability and sensitivity, offering a promising potential for conveniently evaluating the quality of agri-products polluted with AFB 1 . Moreover, this approach provides a new idea for fast and accurate detection of mycotoxin.

Yiqian Kong,Yinghua Zhu,Juncheng Song,Qi Liu,Lili Song,Xiaowei Fei,Xiangyang Li

In this work, Ti 3 C 2 nano-enzymes (Ti 3 C 2 NEs) materials with simulated peroxidase activity and fluorescence quenching properties were prepared. Then Ti 3 C 2 NEs was functionalized using 6-carboxyfluorescein (FAM) labeled Aflatoxin B 1 (AFB 1 ) aptamers to construct a novel multimode nano enzyme biosensor for the detection of AFB 1 in peanuts. Based on the fluorescence quenching characteristics and the superior simulated peroxidase activity of Ti 3 C 2 NE S and the specific binding of the aptamer to AFB 1 , the sensitive and rapid fluorescence/colorimetric/smart phone detection of AFB 1 have been achieved, with detection limits of 0.09 ng mL −1 , 0.61 ng mL −1 and 0.96 ng mL −1 , respectively. The analytical method provided can not only detect AFB 1 in multiple modes, but also has a wider detection range, lower limit of detection (LOD) and better recovery rate, and can achieve on-site accurate detection of AFB 1 content in peanuts, which has great application potential in the field of food quality testing.
