Preparation of Ta(Ph)-Fe/AP Composite Microspheres by Ultrasonic Spray Drying and Characterization of Their Catalytic Properties

Naimeng Song,Li Yang,Pingan Liu

In this study, the ultrasonic spray drying technology was applied to prepare composite microspheres of catalyst (Ta−Fe, Ph−Fe) and AP. The surface morphology of the two composite microspheres was characterized by scanning electron microscopy (SEM). Their particle size distribution characteristics were tested by a nano laser particle size analyzer. The results showed that the spray drying technology can uniformly compound the AP and the catalyst into spherical particles. The thermal decomposition process of Ta(Ph)-Fe/AP composite microspheres were tested by differential scanning calorimetry (DSC). The DSC data demonstrated that Ta(Ph)-Fe/AP composite microsphere had a very good catalytic effect and significantly promoted the thermal decomposition process of AP. In addition, the catalytic mechanism was discussed by calculating the change in activation energy. Compared with the raw AP, the activation energies of the Ta−Fe/AP and Ph−Fe/AP composite microspheres were reduced by 18.5 kJ/mol and 28.1 kJ/mol, respectively.

Applying modified hyperbranched polyester in hydroxyl-terminated polyether/ammonium perchlorate/aluminium/cyclotrimethylenetrinitramine (HTPE/AP/Al/RDX) composite solid propellant

Xiaomu Wen,Keke Chen,Chao Sang,Shen Yuan,Yunjun Luo

Composite solid propellants demand fine and stable mechanical properties, creep resistance and stress relaxation performance during their long storage and usage time. In this study, modified hyperbranched polyester (MHBPE) was prepared and introduced into HTPE/AP/Al/RDX (HTPE, hydroxyl-terminated polyether; AP, ammonium perchlorate; RDX, cyclotrimethylenetrinitramine) solid propellant as an effective additive. The static tensile and dynamic mechanical properties of this propellant before and after the introduction of MHBPE were evaluated. The elevated interfacial interaction by using MHBPE between the binder and RDX fillers improved the toughness and elasticity of the propellant. The enhancement mechanisms were also confirmed by the influence on the fracture surface morphology of the binder which was investigated by SEM. In addition, some influence on the dynamic mechanical properties of HTPE/AP/Al/RDX propellant caused by MHBPE was investigated by dynamic mechanical analysis. The creep behaviors of the HTPE/AP/Al/RDX propellants with and without MHBPE were also investigated at different stresses and temperatures. Reduced creep strain rate and strain were obtained for the modified propellant, implying enhanced creep resistance performance. The creep properties were quantitatively evaluated using a six-element model and the long-term creep performance of the propellant was predicted using the time–temperature superposition principle. A creep behavior of nearly 10 6 s at 30 °C could be acquired in a short-term experiment (800 s) at 30–70 °C. Moreover, the stress relaxation investigation of the propellants with and without MHBPE at −40 °C, 20 °C and 70 °C suggested that MHBPE/HTPE/AP/Al/RDX propellant possessed better response ability to deformation. Thus, the application of MHBPE provides an efficient route of reinforcement to enhance the creep resistance and stress relaxation properties. © 2020 Society of Chemical Industry

Hepatocytic AP-1 and STAT3 contribute to chemotaxis in alphanaphthylisothiocyanate-induced cholestatic liver injury

Yishuang Luo,Jinyu Kang,Jia Luo,Zheng Yan,Shengtao Li,Zhuoheng Lu,Yufei Song,Xie Zhang,Julin Yang,Aiming Liu

The development of cholestatic liver injury (CLI) involves inflammation, but the dominant pathway mediating the chemotaxis is not yet established. This work explored key signaling pathway mediating chemotaxis in CLI and the role of Kupffer cells in the inflammatory liver injury. Probe inhibitors T-5224 (100 mg/kg) for AP-1 and C188–9 (100 mg/kg) for STAT3 were used to validate key inflammatory pathways in alpha-naphthylisothiocyanate (ANIT, 100 mg/kg)-induced CLI. Two doses of GdCl 3 (10 mg/kg and 40 mg/kg) were used to delete Kupffer cells and explore their role in CLI. Serum and liver samples were collected for biochemical and mechanism analysis. The liver injury in ANIT-treated mice were significantly increased supported by biochemical and histopathological changes, and neutrophils gathering around the necrotic loci. Inhibitor treatments down-regulated liver injury biomarkers except the level of total bile acid . The chemokine Ccl2 increased by 170-fold and to a less degree Cxcl2 by 45-fold after the ANIT treatment. p-c-Jun and p-STAT3 were activated in the group A but inhibited by the inhibitors in western blot analysis . The immunofluorescence results showed AP-1 not STAT3 responded to inhibitors in ANIT-induced CLI. With or without GdCl 3 , there was no significant difference in liver injury among the CLI groups. In necrotic loci in CLI, CXCL2 colocalized with hepatocyte biomarker Albumin, not with the F4/80 in Kupffer cells. Conclusively, AP-1 played a more critical role in the inflammation cascade than STAT3 in ANIT-induced CLI. Hepatocytes, not the Kupffer cells released chemotactic factors mediating the chemotaxis in CLI.

Combustion and energy release characteristics of LiF- and AP-modified B/JP-10 suspension fuels prepared using a two-solvent method

LongJin Du,Jianzhong Liu,Binghong Chen,ZhiHao Sun,WeiJuan Yang

B/JP-10 suspension fuel has very high energy density and high application prospects, but energy release during its actual combustion is incomplete. In this study, the surfaces of boron nanoparticles were modified by coating the particles with LiF and AP using a two-solvent method. Thermogravimetric experiments show that the ignition temperature of boron decreases from 772°C to 599°C when the LiF coating amount is 20 wt%. In a laser ignition experiment, the AP-modified B/JP-10 suspension fuel displayed a relatively obvious "microburst" phenomenon at the end of the combustion process . The "microburst" intensifies as the amount of AP coating increases; this behavior has positive implications for the release of energy from the boron in the fuel. The oxidation of boron in 10 wt% LiF-modified fuel increased from 26.35% to 36.22% without an increase in the ambient oxygen partial pressure . AP modification increases not only the oxidation rate of boron in the fuel but also the amount of oxidizing gas produced by decomposition, and this increases the burning intensity of JP-10. LiF and AP modification can destroy the shell layer of the agglomerates formed by the combustion of suspended fuels. LiF makes it difficult to close the shell layer by reacting with B 2 O 3 to form easily sublimated substances and flaky crystals. AP breaks the shell structure due to the high pressure generated by the "microburst", thus forming a porous configuration on the surface of the agglomerate.

Control of burning rate pressure sensitivity for solid propellants by changing the interfacial contact of Al/HMX/AP with precisely located graphene-based energetic catalysts

Ruixuan Xu,Zhihua Xue,Haorui Zhang,Hongqi Nie,Qi-Long Yan

The control of burning rate pressure sensitivity is essential for stable and reliable combustion of solid propellants. It has been proved that the combustion behavior of propellants can be effectively tuned by interfacial control of Al/oxidizers. In this work, core-shell Al-based composites, using oxidizers such as ammonium perchlorate (AP) and cyclotetramethylene tetranitramine (HMX) as the shell layer, have been prepared with a well-controlled location of graphene-based carbohydrazide complexes (GO-CHZ-M, M = Co 2+ or Ni 2+ ) as the catalysts. The catalysts can be located inside HMX or embedded on the surface of AP particles. The decomposition of AP and HMX could be greatly promoted with a trace amount of catalyst. Under the synergistic effects of Al/oxidizer interfacial control and GO-CHZ-M, the ignition delay time is shortened by >48 % with increased in flame radiation. More importantly, the burning rate ( r ) and pressure exponent ( n ) of the solid propellants are effectively regulated in a wide range. In particular, the use of Al@HMX/Co results in a lowered n of 0.29 within 1∼20 MPa compared to 0.79 for the blank reference sample. The agglomeration of Al particles during combustion was also inhibited due to the micro-explosion effect of Al@HMX composites. Novelty and Significance statement The innovative approaches of integrating Al/HMX/AP composites and precise catalysis of graphene-based energetic catalysts in solid propellants have successfully achieved, regarding the modulation of the burning rates and pressure sensitivity. Moreover, the effect of catalyst location on the ignition delay and burning rates has also been studied thoroughly and clarified. This paper provides new insight into the regulation of combustion performance of solid propellants, with improved reaction efficiency and maintained energy density.

Histone acetylation alteration by KAT6A inhibitor WM-1119 suppresses IgE-mediated mast cell activation and allergic inflammation via reduction in AP-1 signaling

Yu-Xin Jiao,Yan-Mei Zhou,Zi-Wen Zhou,Yong He,Shan Liu,Xue-Ting Xu,Kunmei Ji,Jia-Jie Chen

Activation of immunoglobulin E (IgE)-associated mast cells (MCs) triggers the onset of pro-inflammatory signals associated with type I allergic diseases. Although histone acetylation changes have been associated with inflammatory diseases, the impact of lysine-acetyltransferase (KAT) inhibitors on IgE-mediated MCs function is unclear. Potential anti-allergic effects of the KAT6A inhibitor WM-1119 on IgE-mediated MCs activation and allergic inflammation were examined in this study. WM-1119 was observed to reduce IgE-mediated degranulation in rat basophilic leukemia-2H3 cells (RBLs) and murine bone marrow-derived mast cells (BMMCs), as demonstrated by reduced the release of β-hexosaminidase (β-hex)   or histamine   (HA) and decreased inflammatory cytokines. Additionally, WM-1119 attenuated allergic responses in IgE-induced passive cutaneous anaphylaxis (PCA) and active systemic anaphylaxis (ASA) mice. No WM-1119 effects on histamine-induced hypothermia in mice were observed. Mechanically, WM-1119 reduced levels of histone H3 lysine 14 acetylation (H3K14ac) and H3K27ac, while also reducing IgE-induced MAPK or NF-κB activity. Moreover, WM-1119 reduced activator protein-1 (AP-1) activity in a manner involving inhibition of c-Fos transcription and translation together with decreased AP-1 binding of its downstream promoters. KAT6A knockdown in MCs also reduced AP-1 activity by inhibiting c-Fos expression. H3K14ac enrichment in the Fos promoter was observed, indicating that H3K14ac may regulate c-Fos expression. In conclusion, KAT6A inhibition or knockdown was shown to reduce IgE-mediated MCs activation and allergic inflammation through a mechanism involving changes in c-Fos expression and downstream AP-1 activity consequent to down-regulation of histone acetylation. KAT6A inhibition may represent a new treatment strategy for suppressing MCs in treating allergic diseases.

Kukoamine A Prevents Radiation-Induced Neuroinflammation and Preserves Hippocampal Neurogenesis in Rats by Inhibiting Activation of NF-κB and AP-1

Zhang Yaqiong,Gao Lingyue,Cheng Zhihua,Cai Jiayi,Niu Yixuan,Meng Weihong,Zhao Qingchun

Impaired hippocampal neurogenesis and neuroinflammation are involved in the pathogenesis of radiation-induced brain injury. Kukoamine A (KuA) was demonstrated to have neuroprotective effects through inhibiting oxidative stress and apoptosis after whole-brain irradiation (WBI) in rats. The aim of this study was to investigate whether administration of KuA would prevent radiation-induced neuroinflammation and the detrimental effect on hippocampal neurogenesis. For this study, male Wistar rats received either sham irradiation or WBI (30 Gy single dose of X-rays) followed by the immediate injection of either KuA or vehicle intravenously. The dose of KuA was 5, 10, and 20 mg/kg, respectively. The levels of pro-inflammatory cytokines were assayed by ELISA kits. The newborn neurons were detected by 5-bromo-2-deoxyuridine (BrdU)/neuronal nuclei (NeuN) double immunofluorescence. Microglial activation was measured by Iba-1 immunofluorescence. The expression of Cox-2 and the activation of nuclear factor κB (NF-κB), activating protein 1(AP-1), and PPARδ were evaluated by western blot. WBI led to a significant increase in the level of TNF-α, IL-1β, and Cox-2, and it was alleviated by KuA administration. KuA attenuated microglial activation in rat hippocampus after WBI. Neurogenesis impairment induced by WBI was ameliorated by KuA. Additionally, KuA alleviated the increased translocation of NF-κB p65 subunit and phosphorylation of c-jun induced by WBI and elevated the expression of PPARδ. These data indicate that KuA could ameliorate the neuroinflammatory response and protect neurogenesis after WBI, partially through regulating the activation of NF-κB, AP-1, and PPARδ.

Oral administration of large-leaf yellow tea protein-derived oligopeptides alleviates UV-induced skin photoaging through the inhibition of MAPK/AP-1/MMP9 signaling pathway

Ziyi Shen,Simeng Zhou,Yaru Zhang,Yuan Zhang,Hongyan Wang,Daxiang Li,Yijun Wang,Zhongwen Xie,Huifang Ge

Large-leaf yellow tea protein-derived oligopeptides (TPP) contain a substantial number of antioxidant peptides that facilitate skin damage repair and may be employed to mitigate skin photoaging. However, the potential alleviating mechanism by which TPP alleviates skin photoaging damage remains insufficiently understood. This study investigated the in vitro antioxidant capacity and cell migration-promoting ability of TPP. Additionally, it utilized ultraviolet (UV)-exposed and intrinsically aged ICR mice, which exhibit signs of skin aging, to assess the effects of an 8-week oral TPP intervention. The findings indicate that TPP exhibits potent free radical scavenging properties, which mitigate oxidative damage in Raw264.7 cells and promote the migration of NIH/3 T3 cells. TPP intervention significantly ameliorates oxidative stress and skin inflammation symptoms in UV-induced skin photoaging damage by restoring the activity of endogenous antioxidant enzymes and suppressing the mRNA expression of pro-inflammatory cytokines, including IL-1β , IL-6 , and TNF-α ., as well as inhibiting the MAPK/AP-1/MMP9 signaling pathways. This study proposes that TPP is a viable plant protein-derived nutritional supplement for the prevention of UV-induced skin photoaging damage.

UVB Induces Sympathetic Nervous System Activation and Norepinephrine Secretion to Regulate The Skin Color of Mice Through the β2-AR/AP-1 Pathway in Epidermal Keratinocytes

Deng Qirui,Liu Xinyan,Wen Xiujuan,Huang Hao,Tang Hongfeng

The aim of this study was to investigate how ultraviolet B (UVB) light regulates AP-1 expression via the β2-adrenergic receptor (β2-AR) in epidermal keratinocytes, which in turn regulates melanin synthesis in melanocytes, thereby modulating downstream melanin production in skin hair follicles and altering mouse skin color. We established a UV-irradiated mouse model to investigate the effects of UV radiation on changes in skin color. By measuring changes in the expression of genes related to cutaneous sympathetic nerves, norepinephrine synthesis and melanin synthesis, we investigated the relationship between β2-AR expression and cutaneous melanogenesis and determined the localization of β2-AR in cells. The results of the siRNA-mediated transfection of keratinized cells with downregulated β2-AR expression were further verified in vitro. Our results suggest that UVB alters the color of the dorsal skin in mice by activating the AP-1/IL-6 pathway, which triggers the sympathetic release of norepinephrine, thereby increasing β2-AR expression in keratinocytes. Overall, our study improves the current understanding of how UVB light influences skin color changes and highlights the complex interplay between ultraviolet radiation and skin physiology.

A novel near-infrared AP-M@AntiACLY detection probe based on SERS to trace detection of serum ACLY for early diagnosis of pediatric sepsis

Yilin Zhao,Amin Zhang,Shuyun Feng,Tiantian Liu,Jingjing Ning,Yucai Zhang,Jin Cheng,Qian Zhang,Chunxia Wang

Sepsis is a critical condition characterized by high morbidity and mortality. The development of technologies for early recognition of sepsis is essential for enhancing patient outcomes. Our original finding indicates that ATP-citrate lyase (ACLY) holds promise as a diagnostic biomarker for sepsis; however, sensitive methods for detecting trace levels of ACLY in serum are currently lacking. We have therefore developed a novel AP-M@Anti ACLY detection probe based on surface-enhanced Raman scattering (SERS) for the detection of serum ACLY. The AuNBPs@MBA was synthesized using gold nanobipyramids (AuNBPs) as the base and 4-mercaptobenzoic acid (4-MBA) as a reflective indicator for ACLY activity. Specifically, the Raman signal of 4-MBA decreases proportionally with an increase in ACLY concentration. ACLY antibodies were subsequently linked to AuNBPs@MBA via the gold-sulfur bonds and blocked with bovine serum albumin (BSA). Under optimized conditions, the probe exhibits a strong linear correlation between Raman signal and ACLY concentrations, ranging from 1 pg/mL - 0.5 ng/mL (R 2 = 0.998) and 0.1–500 ng/mL (R 2 = 0.999), with a remarkable limit of detection (LOD) of 1.0 pg/mL. AP-M@Anti ACLY detection probe surpasses enzyme-linked immunosorbent assay (ELISA) when testing septic children's serum samples in terms of its detection range, accuracy, and analysis time. Recovery rates ranged from 96.80 % to 102.52 %. In conclusion, all results suggest that AP-M@Anti ACLY detection probe is a promising tool for the early diagnosis of sepsis and offers new insights into the development of preclinical approaches.

Inhibition of the AP-1/TFPI2 axis contributes to alleviating cerebral ischemia/reperfusion injury by improving blood–brain barrier integrity

Cao Yue,Xing Ruixian,Li Qiushi,Bai Yang,Liu Xuewen,Tian Buxian,Li Xin

Reperfusion after cerebral ischemia leads to secondary damage to the nervous system, called cerebral ischemia/reperfusion injury (CIRI). The blood–brain barrier (BBB) consists of endothelial cells and tight junction (TJ) proteins, and its disruption aggravates CIRI. Two GSE datasets identified Tissue Factor Pathway Inhibitor 2 (TFPI2) as a differentially upregulated gene (Log2FC > 1, p  < 0.01) in the cerebral cortex of ischemic rats, and TFPI2 affects angiogenesis of endothelial cells. Moreover, genes (c-Jun, c-Fos, FosL1) encoding subunits of Activator Protein-1 (AP-1), a transcription factor involved in IRI, were highly expressed in ischemic samples. Thus, the effects of the AP-1/TFPI2 axis on CIRI were explored. We determined increased TFPI2 expression in the cerebral cortex of rats receiving middle cerebral artery occlusion (MCAO) for 90 min and reperfusion (R) for 48 h. Then AAV2-shTFPI2 particles (5 × 10 10 vg) were injected into the right lateral ventricle of rats 3 weeks before MCAO/R. TFPI2 knockdown decreased infarct size and neuronal injury in ischemic rats. It improved BBB integrity, demonstrated by reduced FITC-dextran leakage in brain tissues of MCAO/R-operated rats. Furthermore, it increased the expression of TJ proteins (Occludin, Claudin-5, TJP-1) in the cerebral cortex of rats with CIRI. Consistently, we found that TFPI2 knockdown mitigated cell damage in mouse endothelial bEND.3 cells with oxygen and glucose deprivation (ODG) for 6 h and reoxygenation (R) for 18 h (OGD/R) treatment. High co-expression of c-Jun and c-Fos significantly elevated TFPI2 promoter activity. c-Jun knockdown inhibited TFPI2 expression in OGD/R-treated bEND.3 cell. Collectively, our findings demonstrate that inhibition of the AP-1/TFPI2 axis alleviates CIRI.

Bacterial identification by metabolite-level interpretable surface-enhanced Raman spectroscopy

Haoran Chen,Ruike Zhao,Xinyuan Bi,Nan Shen,Xi Mo,Yue Tao,Zhou Chen,Jian Ye

Surface-enhanced Raman spectroscopy (SERS), as a great potential label-free tool in metabolite detection, offers a strategy for rapid bacterial identification. However, it still lacks experimentally supported spectral interpretation at the metabolite level for complex biosamples. We present a SERS-based method for reliable bacterial intracellular metabolic profiling using plasmonic colloids with high rapidness and cost-efficiency. A convolutional neural network model was constructed to accurately classify eight types of bacteria with an overall accuracy as high as 90.44% and identify the key spectral features for classification by Shapley Additive Explanations. Molecule-level interpretation of the SERS metabolic profiles has been further realized in combination with laser desorption/ionization mass spectrometry, evidencing the primary metabolite contribution to the bacterial spectral signatures and molecule-level distinctions among different bacterial types. We provide insights into the mechanism of bacterial identification by label-free SERS and pave the way for interpretable SERS diagnostic tools for various diseases.
