Controlling the dissemination of antibiotic-resistant bacteria (ARB) and antibiotic resistance genes (ARGs) is a global concern. While commonly used chlorine disinfectants can damage or even kill ARB, dissolved oxygen (DO) may affect the formation of reactive chlorine species. This leads to the hypothesis that DO may play roles in mediating the effectiveness of chlorine disinfection for antibiotic resistance. To this end, this study investigated the impacts of DO on the efficiency of chlorine disinfection for antibiotic resistance. The results revealed that DO could increase the inactivation efficiency of ARB under chloramine and free chlorine exposure at practically relevant concentrations. Reactive species induced by DO, including H 2 O 2 , O 2 − , and OH, inactivated ARB strains by triggering oxidative stress response and cell membrane damage. In addition, the removal efficiency of extracellular ARGs (i.e. tetA and bla TEM ) was enhanced with increasing dosage of free chlorine or chloramine under aerobic conditions. DO facilitated the fragmentation of plasmids, contributing to the degradation of extracellular ARGs under exposure to chlorine disinfectants. The findings suggested that DO facilitates disinfection efficiency for antibiotic resistance in water treatment systems.

Tetracyclines (TCs) have been widely detected in agricultural soil due to their widespread use in animal husbandry. The impact of low-generation TCs, i.e., the first- and second- generations, on soil ecosystem has attracted widespread attention. However, the dynamic response of soil microbial community to high-generation TCs, i.e., the third- and fourth- generations, remains largely unknown. Herein, we characterized the variations in the composition, diversity and succession of microbial community and the proliferation of antibiotic resistance genes (ARGs) under the stress of four generations of TCs in brown soil and red soil. The results demonstrated that the exposure of low- and high- generation TCs consistently decreased the alpha diversity and stimulated the succession rate of microbial community in soil. High-generation TCs strongly shifted microbial community composition by reducing community resilience. The complexity of microbial networks and cross-module associations were strengthened to cope with the stress of high-generation TCs in soil. The abundance of ARGs was exacerbated by 1.75 times in response to the fourth-generation TCs compared to control in brown soil. The potential bacterial hosts of ARGs were more diverse in brown soil exposed to high-generation TCs, but the dominant hosts were not changed. These results highlight the potential ecological risk of the newly developed antibiotics, which is helpful for a comprehensive risk assessment of emerging contaminants.

The prevalence of antibiotic resistance genes (ARGs) among bacteria has become a global environmental issue. There is a lack of research on whether nano/microplastics could affect ARG transfer between bacteria. In this study, 20, 100 nm, 1 μm, and 1 mm polystyrene (PS) plastic particles were investigated to determine if nano/microplastics could affect the ARG transfer. Nanoplastics and 1 μm microplastics significantly enhanced the transfer efficiency, whereas 1 mm microplastics had no significant effect. Exposure to 0.1 mg/L 20 nm nanoplastics resulted in the highest transfer frequency, which was 8.9-fold greater than that of the control. Exposure to nanoplastics increased the expressions of genes related to outer membrane porin proteins and the key genes of conjugative transfer. Exposure to 1 μm microplastics had no significant effect on the genes related to transfer. After exposure to 1 μm microplastics, the amounts of extracellular polymeric substances (EPS) were increased, which affected the transfer efficiency. The addition of nanoplastics also could stimulate the production of EPS, and the increase of EPS content in the nanoplastic treatment group was also an important factor that affected the transfer frequency. This work provides important insights into the evaluation of the environmental and health risks of ARGs caused by microplastics.

The excessive application of antibiotics and surfactants resulted in their massive accumulation in waste activated sludge (WAS), but the co-occurrent impacts of antibiotics and surfactants on the antibiotic resistant genes (ARGs) fates have seldom reported. This work mainly revealed the roles and critical mechanisms of sodium dodecyl benzene sulfonate (SDBS) on the sulfadiazine (SDZ) stressing for ARGs distribution during WAS anaerobic fermentation. High-throughput qPCR and metagenomic analysis revealed that SDBS aggravated the SDZ selective pressure, and accelerated the proliferation of ARGs. The total abundance of ARGs was increased from 8.81 × 10 10 in SDZ to 1.17 × 10 11 copies/g TSS in the SDBS/SDZ co-occurrence system. Specifically, the absolute abundances of ermF (MLSB), mefA (MLSB), tetM-01 (tetracycline), tetX (tetracycline), sul2 (sulfonamide) and strB (aminoglycoside) were risen from 4.60 × 10 8 –7.44 × 10 9 copies/g TSS in the SDZ reactor to 1.02 × 10 9 –4.63 × 10 10 copies/g TSS in SDBS/SDZ reactor. SDBS was contributed to the SDZ solubilization and simultaneously effective in disintegrating extracellular polymeric substances and improving cell membrane permeability, which would facilitate the SDZ transport and its interactions with ARGs hosts. Consequently, the microbial community structure was evidently altered, and the typical ARGs hosts ( i.e. , Saccharimonadales and Ahniella ) were greatly enriched. Also, the cell signal transduction systems ( i.e. , glnL , glrK and pilG ), oxidative stress response ( i.e. , frmA and recA ) and bacterial secretion systems ( i.e. , VirB4 ), which were related with ARGs propagation, were all provoked in the co-occurred SDBS/SDZ reactor compared with that of sole SDZ. PLS-PM analysis suggested that the bacterial community was the predominant factor that determined the ARGs fates, followed by mobile genetic elements and metabolic pathway. This work demonstrated the interactive effects of surfactants and antibiotics on the ARGs fates in WAS fermentation systems and gave insightful implications on the ecological risks of different exogenous pollutants.

Antibiotic resistance genes (ARGs) and antibiotic resistant bacteria (ARB) are widespread in urban wastewater treatment plants (UWTPs). In this research, a horizontal transfer model of recipient ( Pseudomonas . HLS-6) and donor ( Escherichia coli DH5α carries RP4 plasmid) was constructed to explore the effect of sulfidated nanoscale zerovalent iron (S-nZVI) on the efficiency of plasmid-mediated horizontal transfer. When the S/Fe was 0.1, the inactivation efficiency of 1120 mg/L S-nZVI on the donor and recipient bacteria were 2.36 ± 0.03 log and 3.50 ± 0.17 log after 30 min, respectively (initial ARB concentration ≈ 5 ×10 7 CFU/mL). Effects of treatment time, S/Fe molar ratio, S-nZVI dosage and initial bacterial concentration were systemically studied. S-nZVI treatment could increase the extracellular alkaline phosphatase and malondialdehyde content of the ARB, cause oxidative stress in the bacteria, destroy the cell structure and damage the intracellular DNA. This study provided evidence and insights into possible underlying mechanisms for reducing conjugative transfer, such as hindering cell membrane repair, inducing the overproduction of reactive oxygen species , inhibiting the SOS response, reducing the expression of ARGs and related transfer genes. S-nZVI could inhibit the gene conjugative transfer while inactivating the ARB. The findings provided an alternative method for controlling antibiotic resistance.

Microplastics (MPs) are becoming ubiquitous in environments and viewed as carriers of antibiotic resistance genes (ARGs). Rivers connecting differently urbanized areas contribute a significant input of MPs and ARGs to the environment. However, a systematic study assessing the role of urbanization in shaping antibiotic resistome and mobilome in riverine MPs is lacking. Here, we conducted a large-scale study by placing five types of MPs (polyethylene, polypropylene, polystyrene, polyethylene-fiber, and polyethylene-fiber-polyethylene) into Beilun River with an urbanization gradient. A total of 314 ARGs and 57 mobile genetic elements (MGEs) were detected in MPs by high-throughput quantitative polymerase chain reaction (PCR). The ARGs in MPs showed a clear spatial distribution with the abundance increased by 2 orders of magnitude from rural to urban regions. A holistic analysis of 13 socioeconomic and environmental factors identified that urbanization predominantly contributed to both the abundance and potential MGE-mediated dissemination of ARGs in riverine MPs. Notably, MPs types were found to significantly affect the resistome and dissemination risk of ARGs, with polypropylene being the preferred substrates to acquire and spread ARGs. This work highlights the necessity of controlling MPs and ARGs pollution in urban areas and provides an important guide for the future usage and disposal of plastics.

Zero-valent iron (ZVI) activated Fenton-like processes have been proposed to inactivate ARB that carry diverse antibiotic resistance genes (ARGs) in wastewater, whereas they exhibit very low capacity of degrading ARB-carrying ARGs. Our previous study has revealed that pre-acidification can make cell inactivation and lysis. Thus, the present study investigated the effect of pre-acidification on the inactivation of a multi-drug resistant bacterium Pseudomonas putida MX-2 and the simultaneous degradation of carried seven ARGs by ZVI/H 2 O 2 and ZVI/PDS processes. Results showed that the pre-acidification of bacterial solution to acidic pH greatly promoted the degradation of carried ARGs from 0.14 to 0.17 logs to 2.48–2.97 logs at the meanwhile of almost completely inactivating P. putida MX-2 in both ZVI/H 2 O 2 and ZVI/peroxydisulfate (PDS) processes. Moreover, the pre-acidification seriously lysed the bacterial cells, released the carried ARGs into the extracellular environment, and facilitated the degradation of extracellular ARGs (e-ARGs) by the generated radicals. During the degradation of the resultant e-ARGs, they were fragmented to short fragments by ZVI/H 2 O 2 and ZVI/PDS processes. The treatment of real wastewater containing P. putida MX-2 further validated the promoting effect of pre-acidification on the degradation of carried ARGs. Therefore, it is extremely crucial to lyse the cells of ARB efficiently via acidification or other methods to promote the degradation of carried ARGs greatly, which may also be applicable for other advanced oxidation processes to reduce antibiotic resistance in wastewater effectively.

Waste activated sludge properties vary widely with different regions due to the difference in living standards and geographical distribution, making a big challenge to developing a universally effective sludge dewatering technique. The Fe(II)-activated persulfate (S 2 O 8 2− ) oxidation process shows excellent ability to disrupt sludge cells and extracellular polymeric substances (EPS), and release bound water from sludge flocs. In this study, the discrepancies in the physicochemical characteristics of sludge samples from seven representative cities in China (e.g., dewaterability, EPS composition, surface charge, microbial community, relative abundance of antibiotic resistance genes (ARGs), etc.) were investigated, and the role of Fe(II)-S 2 O 8 2− oxidation in enhancing removal of antibiotic resistance genes and dewatering ability were explored. The results showed significant differences between the EPS distribution and chemical composition of sludge samples due to different treatment processes, effluent sources, and regions. The Fe(II)-S 2 O 8 2− oxidation pretreatment had a good enhancement of sludge dewatering capacity (up to 76 %). Microbial analysis showed that the microbial community in each sludge varied significantly depending on the types of wastewater, the wastewater treatment processes, and the regions, but Fe(II)-S 2 O 8 2− oxidation was able to attack and rupture the sludge zoogloea indiscriminately. Genetic analysis further showed that a considerable number of ARGs were detected in all of these sludge samples and that Fe(II)-S 2 O 8 2− oxidation was effective in removing ARGs by higher than 90 %. The highly active radicals (e.g., SO 4 − ·, ·OH) produced in this process caused drastic damage to sludge microbial cells and DNA stability while liberating the EPS/cell-bound water. Co-occurrence network analysis highlighted a positive correlation between population distribution and ARGs abundance, while variations in microbial communities were linked to regional differences in living standards and level of economic development. Despite these variations, the Fe(II)-S 2 O 8 2− oxidation consistently achieved excellent performance in both ARGs removal and sludge dewatering. The significant modularity of associations between different microbial communities also confirms its ability to reduce horizontal gene transfer (HGT) by scavenging microbes.

Disinfectants and antibiotics are widely used for the prevention and control of bacterial infectious diseases. Frequent disinfection is thought to exacerbate antibiotic resistance. However, little is known about how disinfectants and antibiotics co-induce changes in the soil antibiotic resistance genes (ARGs). This study determined the ARG profiles and bacterial community dynamics between unamended soil and manure-amended soil exposed to benzalkonium chloride (C 12 ) (BC, 10 mg kg −1 ) disinfectant and sulfamethazine (SMZ, 1 mg kg −1 ), using high-throughput quantitative PCR and 16 S rRNA gene sequencing. Manure application enriched the soil in terms of ARGs abundance and diversity, which synergistically amplified the co-selection effect of BC and SMZ on soil antibiotic resistome. Compared with the control treatment, BC and SMZ exposure had a smaller impact on the bacterial infectious diseases and antimicrobial resistance-related functions in manure-amended soil, in which bacterial communities with greater tolerance to antimicrobial substances were constructed. Manure application increased the proportion of rank I ARGs and potential human pathogenic bacteria, while BC and SMZ exposure increased the drug-resistant pathogens transmission risk. This study validated that BC and SMZ aggravated the antimicrobial resistance under manure application, providing a reference for managing the spread risk of antimicrobial resistance in agricultural activities.

Swine manure is an important reservoir of environmental antibiotic resistance genes (ARGs), and anaerobic digestion (AD) is a commonly used method for swine manure treatment. In this study, the optimized dosage of nano-magnetite to enhance methane production was figured out, the changes of the fate of ARGs response to nano-magnetite were investigated, and the microbial mechanisms were deciphered through the microbial community analysis and key functional genes quantification. Results showed that nano-magnetite could improve the methane production by maximum 6.0%, the maximum daily methane production could be increased by 47.8%, and the AD time could be shortened by above 20.0% at the addition of 75 mmol. The improved performance could be associated with the enhancement of direct interspecies electron transfer (DIET) and the inhibition release due to the formation of Fe-S precipitation not the nutrition elements role of nano-magnetite, and nano-magnetite did not significantly influence the dynamics of microbial community. Nano-magnetite could enhance the methanogenesis instead of the acetogenesis reflected by the functional genes analysis, and the limited effects of nano-magnetite on the fate of ARGs could be associated with its limited influence on the microbial community which determined the fate of ARGs during AD of swine manure.

Due to the widespread presence of antibiotic resistance genes (ARGs) in traditional biological wastewater treatment systems, electrochemical disinfection techniques have received extensive attention. The rTNA/Sb-SnO 2 /PbO 2 Reactive Electrochemical Ceramic Membrane (RECM) system has demonstrated outstanding performance in effectively removing refractory antibiotics. However, its capacity to remove antibiotic resistant bacteria (ARB) remains unclear. In this study, an antibiotic resistant strain of Escherichia coli (AR E. coli ) was employed to investigate the disinfection efficiency of the RECM system at a macroscopic level, along with its disinfection mechanism at a microscopic level. Furthermore, the secondary effluent from an antibiotic production wastewater treatment plant was collected to validate the disinfection performance of this system against ARB in actual wastewater. In the RECM system, the inactivation of AR E. coli and the removal of ARGs were primarily governed by the electrocatalytic process, with notable influences from critical factors such as current density, electrolyte type, and hydraulic retention time. Morphological observation and flow cytometry experiments confirmed that the AR E. coli in the RECM system was inactivated by alterations in cell morphology, leading to fragmentation, and by improvement in cell membrane permeability. More significantly, this system exhibited noteworthy efficiency in treating wastewater contaminated with antibiotic resistant pollutants, effectively eliminating both antibiotics and ARGs from the secondary effluent. Overall, our study comprehensively investigated the performance and underlying mechanisms of the RECM system in mitigating the dissemination of ARGs, as well as its potential feasibility of practical application.

Excess sludge, the primary by-product of wastewater treatment plants, is the source and sink of antibiotic resistance genes (ARGs). Sludge pretreatments are an indispensable pathway to improve the resource recovery and harmfulness for anaerobic digestion sludge. However, fewer studies have compared the effects of different pretreatment technologies on the distribution of ARGs during anaerobic sludge digestion. Here, this study established seven anaerobic digesters, and four typical ARGs and one integrase gene of class 1 integron ( intI1 ) regarded as the representative mobile genetic elements (MGEs) were examined during the whole anaerobic digestion process. It was found anaerobic digestion could effectively remove ARGs with about 70.86% removal rate of total ARGs. Among these pretreatments, the reduce efficiency of ARGs was the highest in 50 °C pretreatment, followed by oxidant, and the last was acid-alkaline. The microbial community analysis demonstrated the microbial community structure, including ARGs hosts and antibiotic resistant bacteria, was significantly changed and influenced by high temperature pretreatment. In addition, high temperature and K 2 S 2 O 8 observably decrease the level of ROS production. Macro transcriptome analysis indicated that sludge pretreatment, except for 50 °C pretreatment, up-regulated the genes relevant to lyases and transferase, but down-regulated the genes responsible for peroxidase, antioxidant enzymes and T4SS gene. This study emphasized and compared the different sludge pretreatments on the fate of ARGs in anaerobic sludge, and highlighted concerns regarding the environmental and health risks to our society.
