Impact of Bifidobacterium longum NSP001 on DSS-induced colitis in conventional and humanised mice

Menglin Chen,Hong Yao,Huizi Tan,Wenqi Huang,Quanyong Wu,Shaoping Nie

Most scientific investigations regarding inflammatory bowel disease (IBD) pathogenesis or therapeutic strategies use dextran sulfate sodium (DSS)-induced models performed on mice. However, differences between human and animal microbiota may confound the data reproducibility from rodent experiments to clinical trials. In this study, the intervention effects of Bifidobacterium longum NSP001 on DSS-induced colitis were investigated using mice colonized with either native or humanised microbiota. Disorders in disease activity index (DAI), morphology and histology of colon tissue, intestinal permeability, and secretion of MPO, TNF-α and IL-6 were ameliorated by daily intake of live B. longum NSP001 cells in both conventional and humanised colitis mice. But the abnormal thymus index, and colonic production of ZO-1 and iNOS were improved only in colitis mice treated with B. longum NSP001 and humanised microbiome. The accumulation of acetic acid and propionic acid in colon microbiome, and the optimization of primary bile acid biosynthesis and glycerophospholipid metabolism pathways in cecum commensals were likely to explain the beneficial effects of B. longum NSP001. These data revealed that intestinal microbiome baseline would possibly affect the manifestation features of interventions by probiotics or dietary components and highlighted the necessity to include humanised microbiome while investigating potential therapeutic strategies based on rodent models.

Protective effects of Bifidobacterium longum subsp. longum BL21 against cyclophosphamide-induced reproductive dysfunction in zebrafish

Dong Yao,Zhou Jiazhi,Tian Hairui,Gai Zhonghui,Zou Kang,Wei Quanwei,Han Mei

This study aimed to investigate the effects of probiotic Bifidobacterium longum subsp. longum BL21 on reproductive health in zebrafish ( Danio rerio ), focusing on hormonal modulation, sperm vitality, and overall reproductive function improvement. Adult male zebrafish of the wild-type AB strain were divided into four groups: control (CTL), model control with cyclophosphamide-induced oligospermia (CS), probiotic-treated (BL21), and a positive control treated with clomiphene citrate (CC). All groups, except CTL, were exposed to cyclophosphamide from day 1 to day 9 to induce reproductive dysfunction, with subsequent monitoring until day 16. Key metrics assessed included body and testicular weight, sperm vitality, male courtship behavior, hormone levels, sperm DNA fragmentation, and gene expression of PCNA, NANOG, ZBTB16, mTOR, DDX4, CYP26A1, and ALDH1A2. In this study, BL21 demonstrated significant therapeutic potential in treating reproductive dysfunction in zebrafish. Metabolomic analysis revealed that BL21 influenced crucial pathways, notably upregulating mTOR signaling and isoflavone biosynthesis, while downregulating the TCA cycle and pyruvate metabolism, suggesting a broad biochemical impact. The probiotic treatment notably improved testicular weight ( p  < 0.01) and sperm count and vitality ( p  < 0.001) compared to the cyclophosphamide-induced model control, underscoring its efficacy in enhancing reproductive parameters. Additionally, BL21 intervention led to a marked increase in chasing behavior and sex hormone levels, surpassing those of the positive control. The treatment also significantly reduced sperm DNA fragmentation ( p  < 0.001) and increased the expression of genes crucial for spermatogenesis and testicular function ( p  < 0.05), confirming its potential to restore reproductive health at multiple biological levels. These results highlight the promise of BL21 as a multi-faceted agent for improving reproductive health, warranting further investigation in clinical settings. Probiotic BL21 enhances reproductive parameters in zebrafish by modulating hormone levels, improving sperm quality, and positively affecting reproductive behavior. These findings suggest potential therapeutic applications of probiotics in managing reproductive health issues.

Bifidobacterium adolescentis CCFM1386 relieved prediabetes symptoms by modulating gut microbiota and metabolic profiles in mice

Xihua Yu,Yurong Zhao,Yi Wang,Liuruolan Li,Wenwei Lu,Wenjun Wu,Hongchao Wang

Prediabetes, a hyperglycaemic state preceding pathologic diabetic hyperglycaemia, is a prevalent condition worldwide. In our preliminary population study, we identified Bifidobacterium adolescentis as a key bacterial strain potentially associated with prediabetes alleviation, by analyzing gut microbiota changes in obese subjects with pre- and post-intervention with hypocaloric balanced diet (HBD). To verify its effects, we administered B. adolescentis CCFM1386 via gavage to prediabetic model mice. The strain significantly mitigated weight gain, improved glucose tolerance, especially decreased serum insulin concentrations by 43.8%, and lowered insulin resistance (IR) compared to the model group; and reduced low-density lipoprotein cholesterol (LDL-C) levels in prediabetic mice. Furthermore, it significantly decreased aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels, demonstrating protective effects on liver and pancreatic tissues as confirmed by histological analysis. To elucidate the underlying mechanisms, we conducted metagenomic sequencing of gut microbiota and non-targeted metabolomics of faecal and serum samples. Our findings revealed that B. adolescentis CCFM1386 helped restore gut microbiota structure, promoted beneficial bacterial growth, and increased the production of beneficial metabolites such as Nicotinate and phenylalanine, thereby influencing multiple metabolic pathways (including protein digestion and absorption, and biosynthesis of amino acids etc.).

Bifidobacterium longum subsp. Infantis modulates intestinal immunity in growing mice in a strain-specific manner

Qing Zhang,Mengfan Ding,Zheng Huang,Shilong Jiang,Jianxin Zhao,Catherine Stanton,R. Paul Ross,Wei Chen,Bo Yang

The early life period is a critical phase for the colonization of initial gut microbes in infants, during which the immune system develops and matures. These processes have long-lasting implications that may extend into adolescence and even adulthood. Bifidobacterium longum subsp. infantis , recognized as an ideal probiotic during infancy, has the potential to modulate intestinal immunity and mitigate immune-mediated diseases. This research explored the potential of various B. longum subsp. infantis strains to modulate the balance between T helper (Th)1 and Th2 responses, immune cell populations, Immunoglobulin A (IgA), and the related genes expression in mice. Notably, B. longum subsp. infantis FHNFQ4M11 and CCFM1269 demonstrated more robust regulatory capabilities. These two strains, which exhibited higher ILA production, activated the aryl hydrocarbon receptor (AHR) signaling pathway, upregulated the levels of galectin-1 (Gal-1) and galectin-3 (Gal-3), and modulated Th1/Th2 differentiation markers in the colon. Additionally, these strains significantly elevated IgA concentrations in both serum and colon and modulated the gut microbiota composition. These findings underscored the potential of specific B. longum subsp. infantis strains as targeted probiotic interventions in early life, providing a promising strategy for promoting immune homeostasis and gut health in infants.

Bifidobacterium breve CCFM1310 enhances immunity in immunosuppressed mice via modulating immune response and gut microbiota

Huiting Fang,Huizhen Li,Yang Chen,Xiaoming Liu,Jianxin Zhao,Paul Ross,Catherine Stanton,Wei Chen,Bo Yang

There has been a rising interest in studying how Bifidobacterium breve can affect the immune system over the years. The aim of this study was to evaluate the effect of B. breve on the immune system and to explore the mechanism in a mouse model of cyclophosphamide (CTX)-induced immunosuppression. The results of the study showed that B. breve CCFM1310 significantly increased the thymus index (p < 0.05), promoting the secretion of immunoglobulins and the number of immune cells in the blood. Moreover, B. breve CCFM1310 could repair the spleen injury. It increased the mRNA expression level of T-bet (p < 0.05) and decreased the mRNA expression level of TGF-β and FOXP3 (p < 0.05). B. breve CCFM1310 alleviated oxidative stress by increasing enzymatic activities of SOD, CAT and T-AOC and reducing the concentration of MDA. Moreover, it repaired intestinal damage by increasing the number of goblet cells, upregulating mRNA expression of tight junction proteins, and promoting the production of short-chain fatty acids (SCFA). B. breve CCFM1310 could regulate the gut microbiota by increasing the relative abundance of Lactobacillus (p < 0.05) and decreasing the relative abudance of Lachnospiraceae NK4A136 group . Overall, our study suggests that B. breve CCFM1310 could enhance immunity in CTX-induced immunosuppressed mice by affecting immune responses, oxidative stress and gut microbiota.

Enhanced Immunogenic Cell Death and Antigen Presentation via Engineered Bifidobacterium bifidum to Boost Chemo-immunotherapy

Tingting He,Lu Wang,Shuangquan Gou,Lu Lu,Genhua Liu,Kui Wang,Yulu Yang,Qiaojian Duan,Wenbo Geng,Peng Zhao,Zhong Luo,Kaiyong Cai

The immunogenic cell death (ICD) of tumor cells has aroused great interest in the field of immunotherapy, mainly due to the production of plentiful tumor-associated antigens (TAAs) and damage-associated molecule patterns. However, doxorubicin (DOX)-induced tumor-specific T-cell-mediated immune response is usually very weak because of antigen presentation deficiency and the immunosuppressive tumor microenvironment (ITME). Herein, the probiotic Bifidobacterium bifidum (Bi) was covalently modified with DOX-loaded CaP/SiO2 nanoparticles (DNPs@Bi) for tumor therapy. On one hand, the pH-responsive release of DOX could induce chemotherapy and ICD in the ITME. On the other hand, tumor-targeting Bi is able to significantly enhance the presentation of TAAs from B16F10 cells to DCs via Cx43-dependent gap junctions. Due to the combination of enhanced ICD and TAAs presentation, the maturation of DCs and the infiltration of cytotoxic T lymphocytes in the ITME were stimulated. As a result, in vivo antitumor experiments demonstrated that DNPs@Bi prolonged the survival rate and significantly inhibited the tumor progression and metastasis. This strategy of bacterial-driven hypoxia-targeting delivery systems offers a promising approach to tumor chemo-immunotherapy.

Pollen Polysaccharides Drive the Strain-Level Assembly and Competitive Dynamics of Bifidobacterium in the Honeybee Gut

Yiyuan Li,Fangyuan Ye,Jiaqiang Wu,Yujie Meng,Haoyu Lang,Shuo Meng,Fuqian Li,Xiaofei Wang,Xue Zhang,Hao Zheng,Peijiong Wang

The gut microbiota orchestrates host health by influencing nutrition, immunity, and behavior. Bifidobacterium species are early colonizers maintaining gut homeostasis in honeybees. Despite these critical roles, strain-level dynamics and driving forces behind microbial competition during social transmission remain poorly understood. Here, we established six field-mimicking colonies to track Bifidobacterium communities across successive generations of newly emerged workers. Social transmission markedly reshaped community structure, generating distinct trajectories among phylotypes. OTU10 consistently dominated, reaching a median relative abundance of 77%, whereas OTU61 and OTU43146 were progressively outcompeted or lost. Comparative genomics of five representative species identified 69 carbohydrate-active enzyme families, with GH43 glycosyl hydrolases driving genomic divergence. Bifidobacterium polysaccharolyticum encoded an expanded CAZyme repertoire, supporting rapid growth on glucose and arabinan and indicating an r-selected strategy. In vitro and in vivo competition assays demonstrated that nutrient availability and priority effects act as key ecological filters. Pollen favored Bifidobacterium apousia , whereas sucrose-only diets promoted B. polysaccharolyticum . Moreover, early colonizers excluded later arrivals to some extent, highlighting priority effects. Together, these findings reveal how pollen polysaccharides and transmission bottlenecks interact to structure the strain-level landscape of the social microbiome.

Enzymatic hydrolysis of crayfish shell protein: Unveiling peptide structures for superior prebiotic promotion of Bifidobacterium animalis

Lei Dong,Yongmin Wu,Hao Wang,Qingqing Xiao,Zexin Zhao,Jun Cai

Crayfish shell protein is an abundant but underutilized resource for developing peptide-based prebiotics to support gut health. However, systematic evidence linking protease selection to peptide structure and subsequent probiotic responses remains limited. This study evaluated five hydrolysis treatments (papain, trypsin, pepsin, alkaline protease, and neutral protease) to identify structure–function determinants for promoting Bifidobacterium animalis subsp. lactis 3296. Papain and trypsin hydrolysates (CSPe-P and CSPe-T) achieved moderate hydrolysis (16.87–22.24% and 17.14–20.39%), balanced molecular weights (7409–7445 Da), semicrystalline organization (48.12–50.60%), and relatively high β-turn and random-coil proportions. These hydrolysates also contained higher total amino acid contents (610.39–625.34 mg/g) and were enriched in Glu, Asp, Arg, and Lys. Functionally, CSPe-P and CSPe-T increased Bifidobacterium animalis subsp. lactis 3296 with final OD 600 values of approximately 1.8–1.9 and maximum specific growth rates at 12 h of 5.14 h −1 and 4.83 h −1 compared with 4.00 h −1 in control. At 12 h, viable counts in the fructooligosaccharides (FOS), CSPe-P, and CSPe-T reached 59.94–64.19 × 10 8  CFU/mL, whereas the control reached 50.0 × 10 8  CFU/mL. CSPe-P and CSPe-T performed comparably to commercial FOS in this in vitro growth assay. In contrast, alkaline and pepsin hydrolysates presented disordered structures and inferior probiotic promotion. Overall, this study provides preliminary in vitro evidence that structure-guided enzyme selection can identify crayfish shell peptides with probiotic-supporting potential.

Construction and Performance Evaluation of a Bifidobacterium Delivery System Based on TBP-SA Pickering Emulsion Gels

An emulsion gel (EG-B) was fabricated through in situ calcium crosslinking of Pickering emulsions stabilized by tartary buckwheat protein–sodium alginate (TBP-SA) nanocomplexes for the encapsulation and delivery ofBifidobacterium longum. Control samples, including free bacteria (Free-B), Pickering emulsions (PE-B), and oil-free hydrogels (HG-B), were used to assess encapsulation efficiency, microstructure, rheological properties, thermal stability, dilution stability, gastrointestinal resistance, storage stability, and colonic adhesion of EG-B. The EG-B exhibited a dual structure comprising a compact Pickering interfacial membrane and a continuous calcium alginate gel network with solid-like rheology (G′>>G″) and viscosity of approximately 100 Pa·s, achieving 92.66% encapsulation efficiency. Viable counts exceeded 6 log CFU/g after pasteurization (72 °C, 15 s) and reached 6.12 log CFU/g post-simulated gastrointestinal digestion, significantly surpassing those of PE-B (5.12 log CFU/g) and HG-B (4.45 log CFU/g). Furthermore, EG-B displayed the highest viable retention during 14-day storage at 4 °C and 25 °C, along with a greater colonic adhesion efficiency (55.64%) compared to PE-B (47.09%) and HG-B (44.65%). The combination of interfacial stabilization and gel network formation provided by the TBP-SA complex presents a straightforward, food-grade strategy for probiotic delivery.

Effects of Bifidobacterium and rosuvastatin on metabolic-associated fatty liver disease via the gut–liver axis

Xue Ran,Ying-jie Wang,Shi-gang Li,Chi-bing Dai

Background/aims Research has indicated that treatment with rosuvastatin can improve liver pathology in metabolic-associated fatty liver disease (MAFLD) patients and that treatment with Bifidobacterium can improve MAFLD. Therefore, the effects of Bifidobacterium , rosuvastatin , and their combination on related indices in a rat model of diet-induced MAFLD need to be investigated. Methods Forty rats were divided into five groups: the normal diet group (N), high-fat diet (HFD) model group (M), HFD + probiotic group (P), HFD + statin group (S), and HFD + probiotic + statin group (P-S). To establish the MAFLD model, the rats in Groups M, P, S, and P-S were fed a HFD for 8 weeks. The treatments included saline in Group N and either Bifidobacterium , rosuvastatin , or their combination in Groups P, S, and P-S by intragastrical gavage. After 4 weeks of intervention, the rats were euthanized, and samples were harvested to analyze gastrointestinal motility and liver function, pathological changes, inflammatory cytokine production, and the expression of proteins in key signaling pathways. Results HFD feeding significantly increased the body weight, liver index, and insulin resistance (IR) index of the rats, indicating that the MAFLD model was successfully induced. Bifidobacterium reduced the liver of MAFLD rats, while Bifidobacterium with Rosuvastatin decreased the liver index, IR index, and levels of aspartate aminotransferase and alanine aminotransferase in MAFLD rats. The MAFLD model showed altered expression of proteins in signaling pathways that regulate inflammation, increased production of inflammatory cytokines, an elevated MAFLD activity score (MAS), and pathological changes in the liver. The MAFLD model also showed reduced relative counts of intestinal neurons and enteric glial cells (EGCs), altered secretion of gastrointestinal hormones, and slowed gastrointestinal emptying. Bifidobacterium , rosuvastatin , or their combination inhibited these various changes. HFD feeding changed the rats’ gut microbiota, and the tested treatments inhibited these changes. These results suggest that the gastrointestinal motility disorder and abnormal liver function in MAFLD rats may be related to a reduction in Escherichia-Shigella bacteria and an increase in Asticcacaulis bacteria in the gut microbiota and that the improvement in liver function induced by Bifidobacterium plus rosuvastatin may be related to increases in Sphingomonas and Odoribacter bacteria and a decrease in Turicibacter bacteria in the gut microbiota. Conclusions The combined use of Bifidobacterium and rosuvastatin could better regulate the gut microbiota of MAFLD model rats, promote gastrointestinal emptying, and improve liver pathology and function than single treatment with Bifidobacterium or rosuvastatin . This provides a better strategy for the treatment of MAFLD.

Bifidobacterium longum subsp. infantis NKU FB3-14 attenuates loperamide-induced constipation through regulation of gut microbiota

Fu Hanyue,Fan Dancai,Wang Jin,Kou Ruixin,Wang Yuanyifei,Wu Yuekun,Wang Shuo

ObjectivesConstipation is a prevalent gastrointestinal issue, and the efficacy of probiotics in alleviating constipation has been well proved. This study aimed to investigate the impact of Bifidobacterium longum subsp. infantis NKU FB3-14 on loperamide- induced constipation by focusing on improving intestinal barrier function and modulating gut microbiota compositionMaterials and MethodsThe constipated model mice induced by loperamide were treated with NKU FB3-14, and the laxative effect was assessed based on the fecal water content, first black stool time and gastrointestinal transit rate. Gastrointestinal regulatory peptides in serum, intestinal neurotransmitter and inflammatory cytokines in colon tissues were measured using ELISA kits. Changes of gut microbiota composition were analyzed through 16S ribosomal DNA sequencing identification. Additionally, HPLC detection was performed to quantify short chain fatty acids levels in fecesResultsTreatment with NKU FB3-14 led to increase the fecal water content, shorten the first black stool time and improve small intestine transit rate. Motilin and substance-P significantly decreased in the model group, and only motilin increased in FB3-14 group; somatostatin and vasoactive intestinal peptide were decreased in model mice and both increased in FB3-14 group; 5-HT levels in the colon tissue were upregulated following NKU FB3-14 treatment. Histological examination revealed thinner colonic mucosa in the model group along with significant in TNF-α, IL-1β and IL-17 levels form the colon tissues which were all relieved by NKU FB 3-14 treatment. Furthermore, NKU FB3-14 intervention resulted in reduced abundance of Desulfobacterota and Desulfovibrio while increasing the abundance of Ruminococcaceae and Eubacterium; higher level of butyric acid was observed in fecesConclusionsIn summary, our findings demonstrated that NKU FB3-14 treatment significantly enhanced the intestinal motility, regulated the expression levels of gastrointestinal regulatory peptides, prevented damage to colonic barriers as well as ameliorated gut microbiota imbalance associated with loperamide-induced constipation.

Bifidobacterium animalis F1-7 in combination with konjac glucomannan improves constipation in mice via humoral transport

Youyou Lu,Zhuang Yu,Zhe Zhang,Xi Liang,Pimin Gong,Huaxi Yi,Liuqing Yang,Tongjie Liu,Hanping Shi,Lanwei Zhang

Probiotics and natural products can promote humoral transport and effectively relieve intestinal motility. This study investigated the effects of probiotics in combination with konjac glucomannan (KGM) and an aqueous extract of Prunus persica on constipation. The growth promotion effect of these natural products on probiotics was investigated using co-culture in vitro. The combined effect of probiotics and natural products on constipation was observed in mice. The tryptophan, tryptamine and short-chain fatty acid levels were determined using enzyme-linked immunosorbent assay, reverse-phase high-performance liquid chromatography, and gas chromatography. The key genes and proteins involved in humoral transport were identified using real-time polymerase chain reaction, western blotting and fluorescence immunoassay. KGM promoted the growth of Bifidobacterium animalis F1-7 in vitro, and a mixture of KGM and B. animalis F1-7 effectively promoted defaecation in mice, increased the faecal water content, shortened the defaecation time and improved the gastrointestinal transit rate. In mice, the KGM + F1-7 mixture reduced the tryptophan level and increased the levels of tryptamine, acetic acid, propionic acid, butyric acid and valeric acid. In addition, the KGM + F1-7 mixture effectively increased the mRNA level of 5-HT4-G-protein-coupled receptors (5-HT4GPCR)/mucins-2 (MUC-2) and reduced the level of aquaporins (AQP3); furthermore, it upregulated the protein level of 5-HT4GPCR/MUC-2 and downregulated the protein level of AQP3. These findings indicated that the KGM + F1-7 mixture effectively improved intestinal motility and alleviated constipation through humoral transport-related pathways.
