CHA-driven SERS-nanoenzyme sensor for ultra-sensitive diagnosis of gastric precancerous lesions

Ya Huang,Yanwen Zhuang,Miao Zhu,Yangfan Zhou,Miaowen Xu,Zixun Wang,Jingwen Zhu,Jinxin Sheng

Early identification and timely intervention of precancerous lesions of gastric cancer are the core strategies to block their malignant transformation into gastric cancer (GC). This study developed a dual functional sensor that combined surface-enhanced Raman scattering (SERS) activity and nanoenzyme catalytic properties. The sensor was driven by catalytic hairpin self-assembly (CHA) technology triggered by the target microRNA-21 (miR-21), promoting the conversion of 3,3′,5,5′-tetramethylbenzidine (TMB) and signal amplification of TMB oxidation (ox-TMB). This work used Au–platinum nanoparticles (Au–Pt NPs) as nanozyme probes and silica-encapsulated ferrite (Fe 3 O 4 @SiO 2 ) as capture probes. In the presence of target miR-21, CHA technology induced Au–Pt NPs to assemble on the Fe 3 O 4 @SiO 2 surface, endowing the sensor with SERS enhancement capability and peroxidase-like (POD-like) simulated catalytic performance. This catalyzed the conversion of TMB and hydrogen peroxide (H 2 O 2 ) into the oxidized product ox-TMB with characteristic absorption. CHA technology, acting as an intrinsic driving force for sensor assembly, further facilitated exponential amplification of the ox-TMB detection signal. This SERS sensor demonstrated outstanding detection performance, achieving a detection limit as low as 20.24 aM for miR-21. It also exhibited excellent clinical applicability and accuracy in clinical sample testing. Therefore, the proposed SERS sensor holds significant clinical potential for diagnosing gastric precancerous lesions. Early identification and timely intervention of precancerous lesions of gastric cancer are the core strategies to block their malignant transformation into gastric cancer (GC).

Tailoring Defects in Highly Siliceous CHA-Type Zeolite for Enhanced CO2 Hydrogenation to Light Olefins

Jiejie Ling,Jilong Wang,Yao Xiao,Yan Gao,Xudong Tian,Jie Tuo,Chuang Liu,Dunru Zhu,Jingang Jiang,Shipeng Ding,Jian Li,Zhendong Wang,Anmin Zheng,Peng Wu,Le Xu

The conversion of CO2 to hydrocarbons via the methanol-mediated pathway represents a crucial route for carbon neutrality, yet preferable production of more value-added light olefins remains constrained by the fundamental activity-selectivity trade-off in zeolite catalysts. While previous studies established an acidity regulation mechanism based on balancing acid density with acid strength, the conversion of methanol intermediates is also critically governed by their intracrystalline diffusion, which can be severely hindered by structural defects particularly prevalent in highly siliceous zeolites. This work demonstrates a different approach that synergistically integrates zeolite acidity and defect engineering. A hydrothermal synthesis strategy with a selected inorganic source precisely controls aluminum incorporation into the highly siliceous CHA zeolite framework (Si/Al > 800). Subsequently, a postsynthetic fluorination followed by calcination effectively heals silanol defects. The resulting HS-CHA-F zeolite features a hydrophobic framework that facilitates rapid methanol diffusion while utilizing precisely tuned ultraweak acidity to steer selective C–C coupling. The optimized OXZEO bifunctional catalyst ZnZrOx/HS-CHA-F achieves 38.5% CO2 conversion and 91.3% light olefin selectivity in hydrocarbons at 653 K and 4 MPa, substantially outperforming conventional SSZ-13 and SAPO-34 benchmarks. This study establishes a generalizable method for overcoming long-standing limitations in CO2 hydrogenation through coupled zeolite acidity and defect engineering.

Novel Silicoaluminophosphate Molecular Sieves with CHA/GME Intergrowth Structures: Synthesis, Structural Elucidation and Catalytic Application

Dehua Wang,Beibei Gao,Dong Fan,Miao Yang,Peng Tian,Zhongmin Liu

Two novel SAPO molecular sieves, detonated as DNL-8 and DNL-9, were synthesized by an aminothermal co-templating strategy. This approach employed diisopropanolamine (DIPA) as both the solvent and the primary organic structure-directing agent (OSDA), with benzyltrimethylammonium chloride (BTACl) or trimethylamine (TMA) serving as a co-OSDA. 13C MAS NMR results demonstrate that the organics occluded in the crystals are the combination of TMA & DIPA for both samples. Three-dimensional (3D) rotation electron diffraction (RED) was used to elucidate their structures, revealing that both the DNL-8 and DNL-9 are the intergrowth of CHA/GME framework. In combination with the simulated XRD patterns of the intergrowths, the GME/CHA ratio is estimated to be 80%GME/20%CHA for DNL-8 and 20%GME/80%CHA for DNL-9. Interestingly, DNL-9 exhibits remarkable catalytic performance in the methanol amination reaction with a super-high selectivity of monomethylamine (MMA) plus dimethylamine (DMA) (~ 96 wt%), suggesting its potential application for the synthesis of MMA and DMA.

A dual-position nucleic acid coding strategy for one-pot recognition of a pathogen and multiple drug-resistant mutations

Chunxu Yu,Baiyang Lu,Yidan Tang,Weilan Zhuang,Xin Zhao,Lulu Guo,Fanzheng Meng,Bingling Li

Timely and accurate detection of a pathogen and its drug-resistance mutations is crucial for guiding clinical treatment and reducing therapy duration. Traditional methods such as PCR, NGS, and microbial culture, though commonly used, have limitations like slow processing, high costs, and limited sensitivity, making them less suitable for urgent clinical needs. Here we propose a dual-position nucleic acid coding strategy, termed Iso-D-nCir, which can achieve one-pot detection of both conserved region and drug-resistant mutations of a pathogen rapidly and precisely. This approach integrates loop-mediated isothermal amplification (LAMP) and catalytic hairpin assembly (CHA) as the isothermal amplification (Iso) and circuit (Cir) to form the LAMP-D-nCir platform as a model, which is low cost, flexibility of use and short detection time. Using Mycoplasma pneumoniae (MP) and its drug-resistant mutations (A2063G and A2064G) as model targets, LAMP-D-nCir demonstrates ultrahigh sensitivity (200 copies/mL) and specificity. Clinical validation across 126 samples demonstrated 100 % concordance with NGS, PCR, and microbial culture. Additionally, the platform exhibits high-resolution discrimination of low-frequency mutations, 1 % mutation abundance from the mixtures can be detected. Furthermore, CHA enables transduction of analyte into diverse signal outputs, such as electrochemical and colorimetric readouts, thereby rendering Iso-D-nCir particularly well-suited for on-site detection in resource-limited environments.

CHA-mediated split G-quadruplex assembly: a ratiometric AIE strategy for label-free detection of APE1

Yang Chan,Yan Zihan,Qin Yanru,Wei Ying,He Ziyi,Mo Liuting,Lin Weiying

Apurinic/apyrimidinic endonuclease 1 (APE1), a critical enzyme in the base excision repair pathway, is a potential biomarker for numerous cancer types. Traditional APE1 activity detection methods are hindered by complex protocols, expensive labeling requirements, and vulnerability to environmental factors. Herein, we develop a label-free ratiometric fluorescent biosensor (TCHG) that integrates APE1-mediated catalytic hairpin assembly (CHA) amplification with split G-quadruplex/malachite green (MG)–mediated aggregation-induced emission (AIE) signaling for sensitive activity analysis of APE1. Our system operates through APE1’s specific cleavage of apurinic/apyrimidinic (AP) site-containing double-stranded substrates, which releases trigger strands that initiate a CHA cascade. This cascade drives the assembly of split G-quadruplex structures. Upon reconstruction, the complete G-quadruplex binds MG, generating amplified near-infrared fluorescence with AIE characteristics. Simultaneously, Hoechst 33258 (HOE) embedded within double-stranded DNA provides a stable reference signal, enabling self-calibrated ratiometric output that effectively minimizes signal fluctuations from instrumental and environmental variations. The TCHG biosensor demonstrates high selectivity for APE1 with a detection limit of 2.4 × 10 –3 U/mL. Importantly, it maintains reliable analytical performance in complex biological matrices, including fetal bovine serum and various cell lysates, successfully differentiating endogenous APE1 activity between normal and cancer cells. The TCHG system offers a tool for APE1 activity analysis in biological samples, with potential applications in biomedical research. Graphical abstract

Ratiometric fluorescent 3D DNA walker and catalyzed hairpin assembly for determination of microRNA

Li Qing,Liang Xuehua,Mu Xiaomei,Tan Li,Lu Jiangnan,Hu Kun,Zhao Shulin,Tian Jianniao

Using 6-carboxyfluorescein (FAM) and tetramethyl rhodamine (TAMRA) as fluorescent signals a ratiometric fluorescent three-dimensional (3D) DNA walker based on a catalytic hairpin assembly (CHA) reaction for microRNA-122 detection was constructed. This method uses CHA reaction triggered indirectly by the target to mediate the 3D DNA walker operation to amplify the signal. The dual emission ratio fluorescent signal with a single excitation wavelength was used as the signal output. This strategy combines DNA walker with CHA reaction and proportional fluorescence signal output methods, which can effectively reduce the background fluorescence signal and the risk of generating false-positive signals. Thus, the impact of environmental factors on the experiment is reduced, thereby obtaining reliable and stable experimental results. It uses the fluorescence excitation wavelength of 488 nm and the maximum fluorescence emission wavelength of 520 nm and 580 nm, respectively. It has a good linear response at a microRNA concentration range of 156.0 pM ~ 7.00 nM and a detection limit of 42.94 pM. This strategy has been successfully applied to detect microRNAs in spiked serum samples. Green Synthesis of CHA Zeolite from Expanded Perlite Waste for Rapid and Selective Pb2+ and Cd2+ Removal

Changchang Fan,Binyu Wang,Pan Xu,Jiaojiao Lv,Haoyang Zhang,Zixuan Liang,Wenfu Yan

The increasing release of non-biodegradable heavy metals, particularly lead (Pb2+) and cadmium (Cd2+), poses severe risks to ecosystems and human health. Herein, we present a sustainable “treating-waste-with-waste” strategy that simultaneously addresses heavy-metal contamination in water and the accumulation of expanded perlite waste. Expanded perlite waste was directly converted into a high-purity, low-silicaCHAzeolite via a simple, one-pot, template-free hydrothermal conversion. The resulting sodium-exchanged material (Na-CHA-p) demonstrated excellent Pb2+and Cd2+removal performance, featuring ultrafast adsorption kinetics (reaching equilibrium within 5 min for both ions), high adsorption capacities (555.6 mg·g−1for Pb2+and 211.0 mg·g−1for Cd2+), and superior selectivity. This study demonstrates an efficient pathway for the high-value utilization of perlite waste and highlights the strong potential of waste-derivedCHAzeolites as advanced adsorbents for heavy-metal wastewater remediation.

Aptamer-gated self-feedback CHA/DNAzyme molecular circuit for enzyme-free ultrasensitive fluorescent sensing of P-tau231

Fang Yang,Tingting Gong,Xia Li,Bingying Jiang,Ruo Yuan,Yun Xiang

A highly sensitive fluorescent biosensor for P-tau231 detection is developed. • Aptamer-gated self-feedback CHA/DNAzyme circuit enables drastic amplification. • 0.45 pg mL −1 detection limit with high specificity against interferents is achieved. • The detection of trace P-tau231in diluted human serums is validated. Phosphorylated tau at threonine 231 (P-tau231) is a clinically informative blood-based biomarker for early Alzheimer's disease (AD), yet its extremely low abundance demands robust signal amplification for sensitive detection of P-tau231. Here, we develop an enzyme-free, target-activated fluorescence assay based on a continuous self-feedback DNA molecular circuit that integrates aptamer recognition, catalytic hairpin assembly, and a Mg 2+ -assisted DNAzyme. P-tau231 binding to an aptamer/ssDNA duplex releases a trigger strand that unlocks a DNAzyme-containing hairpin, initiating an autocatalytic amplification cascade. The activated DNAzyme repeatedly cleaves a fluorophore/quencher-labeled substrate hairpin, converting molecular recognition into a turn-on fluorescence output with substantial signal gain. Under optimized conditions, the proposed strategy enables highly sensitive P-tau231 assay (0.45 pg mL −1 detection limit), while maintaining high specificity against potential interferents. Feasibility in complex matrices is further supported by analysis in diluted human serum. This self-feedback CHA-DNAzyme circuit provides a modular framework for ultrasensitive fluorescence biosensing of low-abundance protein biomarkers relevant to early AD screening. Xiaoer Qixing Cha alleviates functional constipation via regulating gut microbiota and short-chain fatty acids in mice

Jiang He,Huilong Lian,Jia Wang,Bojian Wu,Xiaoying Liu,Zhongyu Zhang,Wenxin Yin,Pengwei Gao,Xu Wu,Jiliang Cao,Haitao Li,Xiaodan Huang,Shengpeng Wang

Background Constipation is a prevalent gastrointestinal disorder associated with gut microbiota dysbiosis. Conventional pharmacological treatments, while effective for short-term symptom relief, are limited by adverse effects, high recurrence rates after discontinuation, and risks of dependency with prolonged use. Xiaoer Qixing Cha (XQC), a well-established Chinese herbal prescription, is widely utilized in pediatric care for addressing dyspepsia, diminished appetite, and constipation. However, its therapeutic mechanisms remain poorly elucidated. Purpose This study was designed to comprehensively explore the therapeutic efficacy and elucidate the mechanistic basis of XQC in murine constipation models. Study design and methods The chemical composition of XQC was comprehensively characterized using high-resolution mass spectrometry. The therapeutic efficacy of XQC (200, 400, and 800 mg/kg) was investigated in loperamide-induced constipated mice. Changes in fecal microbiota were evaluated via 16S rRNA sequencing. Both untargeted and targeted metabolomic analyses were carried out to explore the metabolomic profiles of mice treated with XQC. Additionally, fecal microbiota transplantation (FMT) was performed to evaluate the functional role of gut microbiota mediated by XQC in constipation. Results High-resolution mass spectrometry identified 141 distinct compounds in XQC. Oral administration of XQC significantly alleviated loperamide-induced constipation in mice, as evidenced by improved fecal output, increased fecal water content, enhanced intestinal motility, accelerated whole-gut transit, and modulated inflammatory cytokine levels and mucin 2 expression. Furthermore, XQC selectively restored the gut microbiota, reversing loperamide-induced microbial dysbiosis and enriching beneficial bacterial taxa, including norank_f__Muribaculaceae and Bacteroides . FMT experiments confirmed that the therapeutic effects of XQC were at least partially mediated by gut microbiota. Notably, XQC restored short-chain fatty acid (SCFA) levels, particularly butyrate and isobutyrate. Exogenous butyrate supplementation further alleviated constipation symptoms in mice, supporting its role in XQC’s mechanism of action. Conclusion These findings underscore XQC’s dual therapeutic role in mitigating constipation and restoring gut microbial balance, positioning it as a viable alternative to conventional therapies. Furthermore, this study establishes a theoretical foundation and clinical framework for the evidence-based application of XQC in constipation management, advancing its rational integration into pediatric and gastroenterological practice.

A dual-locked and self-feedback CHA-DNAzyme nanomachine for tumor-specific molecular imaging in living cells

Baizong Fang,Wei Guo,Zhenyu Liu,Huakui Huang,Xiaohong Zhong

Background The detection and imaging of intracellular biomarkers can provide important information for the early diagnosis of cancer. Leveraging the inherent advantages of DNA such as biocompatibility, ease of synthesis, programmability, and sequence-specific recognition, DNA-based probes have been widely employed for biomarker analysis. Currently, a variety of DNA probes combined with signal amplification methods have been widely used to visualize biomarkers in living cells. However, conventional single-locked DNA probes cannot ensured the accurate diagnosis. Furthermore, the lack of efficient cascade signal amplification strategies makes it challenging to detect and image low-abundance biomarkers. Results In this work, we report a dual-locked, self-feedback catalytic hairpin assembly (CHA)-DNAzyme cascade amplification platform, termed hMNS@SCD. This platform utilizes MnO 2 nanoflowers as nucleic acid carriers and DNAzyme cofactor supplement, employing apurinic/apyrimidinic endonuclease 1 (APE1) and miR-21 as two molecular keys. When the APE1 and miR-21 existence, hairpin probe carry out the CHA cycle, leading to the assembly of a Y-shaped DNAzyme complex. When Mn 2+ is present, the Y-shaped DNAzyme can undergo a hybridization-cleavage-release cycle on the MB, leading to a gradual restoration of Cy5 fluorescence. In addition, the released target analogue from MB can trigger a new CHA cycle, generating more Y-shaped DNAzyme complexs and cleaving more MB, further amplifying the fluorescence signal. Through the self-feedback CHA-DNAzyme cascade amplification mechanism, hMNS@SCD effectively enhances the detection sensitivity, with a detection limit for miR-21 reaching 34.6 fM. Experimental data show that hMNS@SCD has high sensitivity, selectivity, and biocompatibility. Significance we constructed a dual-locked, self-feedback CHA-DNAzyme cascade nanomachine, which enables tumor-specific molecular imaging and effectively distinguishes between normal cells and cancer cells. In addition, due to the programmability of nucleic acid probes, this strategy can be readily adapted for detection and imaging of other low-abundance biomarkers, offering a promising way forward for tumor-specific molecular imaging and early cancer diagnosis.

A biosensor using semi-DNA walker and CHA -FRET loop for ultrasensitive detection of single nucleotide polymorphism

Yunshan Zhang,Shijie Xu,Jian Chen,Lanyue Wang,Lina Bian,Jing ye,Lin Weng,Xiaoyu Zhao,Cheng-Te Lin,Shuang Li,Diming Zhang

Single nucleotide polymorphisms (SNPs) are emerging as important biomarkers for disease diagnosis, prognostics and disease pathogenesis. However, it is still challenging to make highly specific and sensitive SNPs detection to distinguish target DNA with single-base difference, so that flap endonuclease 1 (FEN 1) manipulated semi-DNA walker reaction and catalytic hairpin assembly (CHA)-FRET loop for ultrasensitive SNP detection has been proposed. Herein, FEN 1 was employed to specifically recognize and cut the 5′ flap of three-base overlapping structure formed by only hybridizing with mutant target (MT). Simultaneously, FEN 1 drove MT hybridization to autonomously walk along downstream probe (DP) and the cut sequence (CS) from semi-DNA walker reaction was collected by magnetic bead-based separation to avoid false-positive signal. Next, the cleaved CS triggered CHA and generated fluorescence resonance energy transfer (FRET) via Cy3 and Cy5. FEN 1 manipulated semi-DNA walker reaction to specifically recognize the target DNA and combined CHA-FRET loop to dually amplify signal. Thus, the SNP biosensor demonstrated superior performance for ultrasensitive detection of mutant KRAS gene with a low detection limit as low as 80 aM and was successfully applied to monitor the expression of mutant KRAS gene in human cancer cell lysates. This strategy provides an ultrasensitive way for the detection of biomolecules and reveals an effective avenue for diseases diagnosis.

An ultrasensitive aptamer-based fluorescent biosensor for luteinizing hormone with mutually orthogonal DNAzyme and self-replication CHA amplification

Li Yuhao,Wang Yanni,Pan Linwei,Li Xiaolong,He Tingting,Tang Yaqin,Xie Jing,Li Daxiu

Sensitive and specific detection of luteinizing hormone (LH) is critical for the early diagnosis of diseases and cancers, offering valuable insights for clinical strategies aimed at regulating physiological functions to maintain health. To this end, we developed a novel sensing platform for the ultrasensitive detection of LH, harnessing a mutually orthogonal cascade amplification composed of DNAzyme and self-replicating catalytical hairpin assembly (SR-CHA). Upon binding of LH to its aptamer, an LH/aptamer complex is formed, leading to the release of an initiator strand (“strand A”) that triggers the assembly of Y-shaped DNA structures. Each Y-shaped structure comprises a modular DNAzyme-based amplification unit, which generates a one-to-many fluorescence signal, and an initiator mimic sequence capable of self-replicating to produce numerous new initiators for accelerating the formation of additional Y-shaped structures, thereby amplifying the detection signal. This synergistic design resulted in outstanding analytical performance, achieving a detection limit as low as 0.00038 mIU/mL in buffer and 0.0006 mIU/mL in real samples, with a broad dynamic range from 0.001 to 100 mIU/mL. In addition, the proposed method also demonstrated excellent selectivity, and a strong correlation with the standard ELISA method, which reveals its promising potential as a universal simple and sensitive methodology for the construction of various aptamer-based bioassays.
