CaO2/Fe3O4 nanocomposites for oxygen-independent generation of radicals and cancer therapy

Marhaba Mamat,Xiaofeng Wang,Lin Wu,Rong Zhao,Jin Cao,Xueyong Qi,Song Shen

The hypoxic tumor environment prevents the generation of reactive oxygen species (ROS), reducing the therapeutic efficiency. We construct oleylamine (OA) coated CaO 2 /Fe 3 O 4 nanocomposites to realize oxygen-independent generation of ROS and high efficient treatment of cancer. In the tumor site, CaO 2 reacts with water to generate H 2 O 2 , which can be catalized by Fe 2+ that is produced by Fe 3 O 4 , to form highly toxic hydroxyl radicals (∙OH). To inhibit the premature reaction, CaO 2 /Fe 3 O 4 nanoparticles were coated with pH sensitive OA. The nanocomposites exhibited remarkable tumor growth inhibition ability and favorable biocompatibility, holding a great potential for hypoxic tumor therapy.

Green Glyphosate Treatment with Ferrihydrite and CaO2 via Forming Surface Ternary Complex

Nuanqin Zhang,Hongwei Sun,Guangming Zhan,Junning Zu,Lizhi Zhang

Glyphosate (PMG) is a globally used broad-spectrum herbicide and receives environmental concerns because of its moderate persistence and potential carcinogenicity. Traditional PMG treatment methods often suffer from the generation of a more toxic and persistent aminomethylphosphonic acid (AMPA) intermediate. Herein, we develop a green method with ferrihydrite (FH) and CaO2 (FH/CaO2) via regulating the coordination of PMG with FH and Ca2+, where the phosphonate group of PMG preferentially binds to FH and its carboxylate side complexes with Ca2+ released by CaO2, forming a FH-PMG-Ca ternary surface complex. This unique ternary complex can redistribute electrons within the PMG molecule for its C–P activation and C–N bond stabilization, favoring the selective C–P bond attack of superoxide radical produced by the Fenton reaction between CaO2-derived H2O2 and FH, thus generating environment-friendly glycine instead of AMPA. The FH/CaO2 process realizes over 99% PMG degradation in industrial wastewater within 1 h, with residual PMG < 0.1 ppm and AMPA < 40 ppb. More importantly, the CaO2 consumption was as low as 3.1 mg of CaO2/mg of PMG, one-fifth those of previously reported CaO2-based counterparts. This study provides an effective and environment-friendly PMG treatment strategy and highlights the importance of surface coordination modes on the degradation pathway of PMG.

Improved humification and Cr(VI) immobilization by CaO2 and Fe3O4 during composting

Li Chen,Zhigang Yi,Yaoning Chen,Yuanping Li,Hongjuan Jiang,Jun Wang,Yanrong Chen,Yaoqin Nie,Mengwei Luo,Qianruyu Wang,Wei Zhang,Yanting Wu

The current research studied how Fe 3 O 4 nanomaterials (NMs) and CaO 2 affect humification and Cr(VI) immobilization and reduction during the composting of oil-tea Camellia meal and Cr-contaminated soil. The results showed that Fe 3 O 4 NMs and CaO 2 successfully construct a Fenton-like reaction in this system. The excitation-emission matrix-parallel factor (EEM-PARAFAC) demonstrated that this Fenton-like treatment increased the generation of humic acids and accelerated the humification. Meantime, RES-Cr increased by 5.91 % and Cr(VI) decreased by 16.36 % in the treatment group with CaO 2 and Fe 3 O 4 NMs after 60 days. Moreover, the microbial results showed that Fe 3 O 4 NMs and CaO 2 could promote the enrichment of Cr(VI) reducing bacteria, e.g., Bacillus , Pseudomonas , and Psychrobacter , and promote Cr(VI) reduction. This study gives a novel view and theoretical reference to remediate Cr(VI) pollution through composting.

Fabricating 3D Printable Self-Oxygenating Hybrid Bioactive Hydrogel Scaffolds for Vascularized Bone Regeneration

Qianrui Zeng,Yang Yang,Jie Shi,Qingxin Guan,Jia Li,Wei Li

Limited cells and growth factors, insufficient mechanical properties, and the hypoxic status in defect areas are the severe challenges faced by bone tissue engineering (BTE) scaffolds, currently. In this work, a 3D printable composite scaffold (Gel-MCM-41-CaO 2 ) without cells or growth factors is designed to promote vascularized bone regeneration through sustained release of oxygen and bioactive ions, utilizing a 1D tube encapsulation release technology to achieve slow release of oxygen from both ends of MCM-41-CaO 2 . Calcium silicate is used as a cross-linking agent and a bioactive agent to cross-link with sodium alginate, combined with polyacrylamide to form a hybrid double network, which endows the hydrogel with excellent compressive strength and 3D printable properties. Based on amounts of data in vitro and in vivo, the oxygen and silicon ions released from this hydrogel scaffold can promote cell proliferation and differentiation, upregulate the expression of osteogenic and angiogenic-related genes, and promote angiogenesis and osteogenesis. This work presents a strategy for fabricating BTE scaffolds without the need for cells or growth factors, with the potential to be applied in clinical bone regeneration and repair.

Disrupting calcium homeostasis and glycometabolism in engineered lipid-based pharmaceuticals propel cancer immunogenic death

Qiuxia Peng,Xiaolong Li,Chao Fang,Chunyan Zhu,Taixia Wang,Binxu Yin,Xiulin Dong,Huaijuan Guo,Yang Liu,Kun Zhang

Homeostasis and energy and substance metabolism reprogramming shape various tumor microenvironment to sustain cancer stemness, self-plasticity and treatment resistance. Aiming at them, a lipid-based pharmaceutical loaded with CaO 2 and glucose oxidase (GOx) (LipoCaO 2 /GOx, LCG) has been obtained to disrupt calcium homeostasis and interfere with glycometabolism. The loaded GOx can decompose glucose into H 2 O 2 and gluconic acid, thus competing with anaerobic glycolysis to hamper lactic acid (LA) secretion. The obtained gluconic acid further deprives CaO 2 to produce H 2 O 2 and release Ca 2+ , disrupting Ca 2+ homeostasis, which synergizes with GOx-mediated glycometabolism interference to deplete glutathione (GSH) and yield reactive oxygen species (ROS). Systematical experiments reveal that these sequential multifaceted events unlocked by Ca 2+ homeostasis disruption and glycometabolism interference, ROS production and LA inhibition, successfully enhance cancer immunogenic deaths of breast cancer cells, hamper regulatory T cells (Tregs) infiltration and promote CD8 + T recruitment, which receives a considerably-inhibited outcome against breast cancer progression. Collectively, this calcium homeostasis disruption glycometabolism interference strategy effectively combines ion interference therapy with starvation therapy to eventually evoke an effective anti-tumor immune environment, which represents in the field of biomedical research.

Synergistic antifouling coating based on PS-b-PDMS/CaO2 for marine aquaculture nets

Li Zhu,Sun Ningxin,Li Shiwei,Yang Jiaao,Guo Yi,Dong Lijie

Biofouling on marine aquaculture nets poses significant challenges to the economic viability and environmental sustainability of mariculture. This study developed a novel composite coating PS- b -PDMS/CaO 2 on the net surface by incorporating oxygen-releasing calcium peroxide nanoparticles (CaO 2 NPs) into a polystyrene-polydimethylsiloxane block copolymer (PS- b -PDMS). This coating synergistically combines low surface energy with a sustained oxygen-enriched microenvironment and enhanced reactive oxygen species (ROS) generation, resulting in significantly enhanced antifouling performance. Experiments demonstrated that the coating achieved outstanding antifouling efficacy, with antibacterial rates exceeding 99% against Escherichia coli, Staphylococcus aureus, and the marine bacterium Pseudomonas aeruginosa, anti-diatom adhesion rates of 92.18%, and anti-protein adhesion rates of 96.82%, with an optimal loading of 0.6 wt%. Moreover, the coating improved the mechanical properties (~ 35.53 MPa) while preserving the low-surface-energy characteristics and sustained oxygen release for over 60 h. A 90-day marine field test verified minimal biological fouling on the PS- b -PDMS/CaO 2 -0.6 coating surface, showing excellent real-sea antifouling performance and structural stability, indicating the long-term serviceability of aquaculture nets in marine environments. This study proposes a novel technical pathway for the development of environmentally friendly, long-term stable, and high-performance antifouling and antibacterial coatings that can effectively adapt to complex marine environments. MnO2-coated CaO2 nanoparticles as pH-responsive oxygen self-sufficient carriers to potentiate radioimmunotherapy

Wei Li,Chunjie Wang,Yujie Zhu,Yangguang Zhang,Hengze Ding,Linqi Zhu,Wei Zhou,Yongyou Wu,Liangzhu Feng

Radioimmunotherapy represents a clinically available combination strategy for treating a broad range of solid tumors, however, its therapeutic efficacy remains significantly compromised by tumor hypoxia due to the inhibition of cytotoxicity and the suppression of antitumor immunity activation. Herein, we synthesized a pH-responsive oxygen-releasing nanomedicine (denoted as an oxygen nanotank) by growing a thin manganese dioxide (MnO 2 ) layer onto calcium hyperoxide (CaO 2 ) nanoparticles through an in-situ oxidation process to enhance radioimmunotherapy. Using a two-step liposomal coating strategy, we obtained CaO 2 @MnO 2 -PEG, which exhibited excellent physiological stability, with MnO 2 functioned as a catalase-like nanozyme to enable pH-dependent oxygen generation. This nanomedicine significantly enhanced the X-ray irradiation induced cytotoxicity against hypoxic tumor cells by amplifying intracellular reactive oxygen species. After intravenous injection, CaO 2 @MnO 2 -PEG specifically accumulated in tumor tissues and substantially alleviated hypoxia, resulting in enhanced therapeutic outcomes of X-ray-mediated external radiotherapy in a subcutaneous CT26 tumor model. Furthermore, combining CaO 2 @MnO 2 -PEG with immune checkpoint blockade therapy achieved synergistic tumor suppression by concurrently activating innate and adaptive immune responses. This study establishes a controllable synthesis method for pH-responsive oxygen nanotanks, effectively addressing hypoxia-induced therapeutic resistance in radioimmunotherapy.

H2O2 Self-Supplying CaO2/POM@MOF Bimodal Nanogeneration Materials for Photothermal and Chemodynamic Synergistic Antimicrobials

Na Chen,Yuan Li,Yang Pan,Haozhe Wang,Hao Gu,Yuan Sun,Tiedong Sun

The emergence of bacterial resistance has a negative impact on the conventional antimicrobial treatments, and research into the development of the new antimicrobial materials is of great significance. Multimodal synergistic antimicrobial materials exert better antimicrobial effects compared with a single modality. In recent years, Polyoxometalate (POM) has shown great potential in the biomedical field due to its high catalytic activity and high photothermal conversion ability. However, owing to its small surface area, its applications have been greatly limited. Herein, we designed a H 2 O 2 self-supplied CDT and PTT bimodal nanogeneration material (i.e., CaO 2 /POM@MOF) possessing a larger surface area for the treatment of the bacterial infections, in which CaO 2 could release O 2 and H 2 O 2 in the weakly acidic microenvironment of bacterial infection and the dual catalytic site of POM@MOF could enhance the CDT reaction to generate ROS, resulting in the bacterial oxidative stress and the leakage of the bacterial contents, and the exposure to the NIR light generates localized high temperature that cause the rupture of bacterial cell membranes and the denaturation of their proteins. Meanwhile, the superior stability of POM@MOF can release fewer metal ions to improve its biocompatibility. The in vitro antimicrobial experiments demonstrated that CaO 2 /POM@MOF possessed the combined CDT and PTT effect and exhibited excellent antimicrobial efficacy. This work presents a promising strategy to combat the bacterial infections.

Remediation of 2,4-dichlorophenol-contaminated groundwater using nano-sized CaO2 in a two-dimensional scale tank

Li Tianyi,Zhang Chengwu,Zhang Jingyi,Yan Song,Qin Chuanyu

This study evaluates the applicability of nano-sized calcium peroxide (CaO 2 ) as a source of H 2 O 2 to remediate 2,4-dichlorophenol (2,4-DCP) contaminated groundwater via the advanced oxidation process (AOP). First, the effect and mechanism of 2,4-DCP degradation by CaO 2 at different Fe concentrations were studied (Fenton reaction). We found that at high Fe concentrations, 2,4-DCP almost completely degrades via primarily the oxidation of ·OH within 5 h. At low Fe concentrations, the degradation rate of 2,4-DCP decreased rapidly. The main mechanism was the combined action of ·OH and O 2 ·− . Without Fe, the 2,4-DCP degradation reached 13.6% in 213 h, primarily via the heterogeneous reaction on the surface of CaO 2 . Besides, 2,4-DCP degradation was significantly affected by solution pH. When the solution pH was>10, the degradation was almost completely inhibited. Thus, we adopted a two-dimensional water tank experiment to study the remediation efficiency CaO 2 on the water sample. We noticed that the degradation took place mainly in regions of pH < 10 (i.e., CaO 2 distribution area), both upstream and downstream of the tank. After 28 days of treatment, the average 2,4-DCP degradation level was ≈36.5%. Given the inadequacy of the results, we recommend that groundwater remediation using nano CaO 2 : (1) a buffer solution should be added to retard the rapid increase in pH, and (2) the nano CaO 2 should be injected copiously in batches to reduce CaO 2 deposition.

Self-Powered CaO2/Heme@Zn-MOF Enabled Fluorescence Immunosensing Strategy for Accurate and Selective Isoprocarb Monitoring

Yong Li,He Wang,Tianhu Long,Jing Liu,Yulun Nie,Chao Yang,Jianbo Shi

Accurate and sensitive detection of pesticide residues is crucial for environmental safety and human health. Herein, a self-powered metal–organic framework nanozyme-based immunoassay was developed for selective and sensitive detection of isoprocarb by encapsulating calcium peroxide and heme into a Zn-MOF to construct the CaO2/heme@Zn-MOF composite. The prepared nanozyme exhibits intrinsic peroxidase-like activity and enables in situ H2O2 generation under acidic conditions, effectively eliminating the need for external oxidants. By coupling antibody–antigen specific recognition with catalytic signal amplification of CaO2/heme@Zn-MOF, a competitive nanozyme-linked immunosorbent assay was established for selective detection of isoprocarb. The fluorescence signal originating from o-phenylenediamine oxidation gradually decreases with an increase in isoprocarb concentration, allowing quantitative analysis with a detection limit of 0.017 μM. The entire competitive immunoassay can be completed within an optimal detection time of 90 min. Furthermore, the proposed method demonstrates excellent practical reliability and anti-interference capability in real water and agricultural products, achieving quantitative recovery rates ranging from 92.85% to 107.14% with relative standard deviations of less than 5.91%. Furthermore, a portable immunomagnetic bead with paper devices was fabricated for on-site analysis. This work provides a general strategy for integrating self-powered nanozymes with immunoassays, thereby expanding their potential for portable environmental monitoring.

Modified steel slag/CaO2 enables magnetic separation and recovery of arsenic during hydrothermal liquefaction of hyperaccumulator

Ge Yan,Haiwei Jiang,Mingxiang Xie

Heavy metal separation and recovery from hyperaccumulators are crucial to promote the industrialization of phytoremediation and prevent secondary heavy metal pollution. In this study, we developed an innovative modified steel slag/CaO 2 technology for arsenic recovery and separation via hydrothermal liquefaction (HTL) of As-enriched Pteris vittata L. (PVL). Arsenic migrated from the aqueous phase to the solid phase, with a recovery rate as high as 99.96 %, and the oxidation of arsenite was achieved with an impressive efficiency of 97.64 %. Experiments with model compounds revealed that the As in the PVL was immobilized by metal oxides (CaO, MgO, Fe 2 O 3 and SiO 2 ) in the modified steel slag and deposited in the solid phase. The coprecipitation of arsenic and heavy metals was verified via Fourier transform infrared (FTIR) spectroscopy and X-ray diffraction (XRD). As(III) was converted into As(V) via the strong oxidability of •OH and H 2 O 2 from CaO 2 . XPS confirmed that the interconversion of Fe 3+ and Fe 2+ significantly contributed to the oxidation and removal of arsenic. The concentration of As(III) in the system decreased to 1.96 % with this method. To investigate heavy metal recovery from the solid phase, magnetic experiments were subsequently conducted to investigate the arsenic distribution in the solid phase after magnetic separation. Notably, the As could be recovered in the magnetic solid phase, whereas 0.05 % of the As(III) and 18.46 % of the As(V) of the total arsenic were distributed in the non-magnetic solid phase. This study demonstrated that modified steel slag/CaO 2 has great potential for the detoxification and recovery of heavy metals in hyperaccumulators.

Electrospun Nano-CaO2/Polycaprolactone/Gelatin Nanofibers and Their Wound Healing Application through In Situ Supplying H2O2

Lin Qi,Yong Huang,Zheng Liu,Yulin Jiang,Meixuan Du,Limin Liu,Ganjun Feng,Yubao Li,Li Zhang

Skin is highly susceptible to foreign pathogens when damaged. Infections usually lead to serious damage to wounds and hinder wound healing, which results in nonhealing and inflammation of the wounds. Herein, a kind of fibrous membrane incorporated with calcium peroxide nanoparticles (n-CaO 2 ) is fabricated by electrospinning the polymer blend solution of polycaprolactone and gelatin. The obtained membranes show a randomly distributed nanofibrous structure with n-CaO 2 particles embedded in each fiber. Such a structure endows the membranes with medium hydrophilicity and improves blood clotting in comparison with the commonly used gauze, while the loading of n-CaO 2 achieves the in situ generation and rapid release of hydrogen peroxide (H 2 O 2 ), enabling FM-10 to display a robust bacteria inhibition of over 90% elimination to E. coli . The in vitro cytocompatibility experiment on the fibrous membranes demonstrates low cytotoxicity to L929 fibroblasts. The in vivo assessment indicates that the fibrous membranes can distinctly accelerate the wound healing, of which FM-5 exhibits the highest wound closure rate (>95%) after 14 days. This study provides a versatile approach, simply by varying the loading dosage of n-CaO 2 into fiber matrix, to fulfill different functions to meet the multiple needs of wound care.
