Visual Analysis of Carbendazim Residues in Carrot Tubers via Postionization Mass Spectrometry Imaging

Tianyu Wang,He Zhang,Yongjun Hu

Carbendazim (CBZ) residues in food are a severe threat to food safety, and their detection is a challenging problem in food science. We introduce here a new method based on laser desorption postionization mass spectrometry imaging (LDPI-MSI) for detecting CBZ residues in carrots. In the novel LDPI-MSI method, two distinct laser beams simultaneously exert dissociation and ionization, which offers several advantages over traditional techniques based on single-photon matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI), including simplified sample preparation, streamlined operation workflow, and a lower limit of detection (LOD). The LOD, in the proposed method, has been lowered to 0.019 ppm. Coupled with mass spectrometry imaging (MSI), the LDPI-MS method enabled in situ detection of small molecular compounds, such as chemical pesticides, and provided comprehensive and accurate results. The image obtained from the characteristic mass spectrometric signature of CBZ at m/z 191 illustrated that most of the CBZ could not enter the carrot tubers directly, but a small amount of CBZ entered the carrot root and was mainly concentrated in the central xylem. The results suggest that the proposed method could potentially be used in pesticide analysis.

Differential ratiometric molecularly imprinted electrochemical method for methyl-parathion and carbendazim in foods

Yue Sun,Xifeng Yu,Qiao Zhang,Haiyan Song,Qi Kang,Dazhong Shen

Non-specific adsorption of interferents on molecularly imprinted polymer (MIP) membranes bottlenecks their selective recognition to templates. Herein, we reported an improved differential MIP strategy to correct such adverse effect for determining carbendazim (CBZ) and methyl-parathion (MP) in food samples. MIP membranes of CBZ and MP were prepared using mixed functional monomers of β-cyclodextrin and thionine with synergistic recognition selectivity. Co 3 O 4 NPs@CNTs nanocomposites were used for signal amplification with the oxidation currents of CBZ and MP enhanced 16.5-fold and 14.9-fold, respectively. With the current of poly-thionine (at −0.292 V) in MIP as reference signal, the ratiometric current differences between MIP -MP and MIP -CBZ at 0.223 and 0.611 V were used in the determination of MP and CBZ with the detection limits of 1.6 and 2.5 nM, respectively. The interference levels in the improved differential MIP mode were − 13 % ∼15 % of those obtained in the standard MIP mode, strengthening the anti-interference ability of MIP-based sensors.

Complementary Strand-Free Persistent Luminescence Ratiometric Aptasensor for Ultrasensitive and Selective Carbendazim Assay

Xu Zhao,Xue-Bing Qin,Zhu-Ying Yan,Li-Jian Chen,Xiu-Ping Yan

Carbendazim (CBZ) poses significant health risks, necessitating its selective and sensitive detection. Herein, we report a complementary strand (cDNA)-free ratiometric aptasensor based on persistent luminescence nanoparticles (PLNP) that operates without in-situ excitation for the accurate quantification of trace CBZ in complex matrices. The sensor integrates an aptamer to enhance affinity and specificity, yet eliminates the conventional quencher-labelled cDNA. Methylene blue (MB) is innovatively introduced as a multifunctional substitute: it self-adsorbs onto the aptamer-modified PLNP through π–π stacking, electrostatic attraction, and hydrogen bonding, serving simultaneously as the resonance-energy-transfer acceptor and the second signaling channel. MB quenches the PLNP phosphorescence at 542 nm while generating its own 700 nm emission. Upon CBZ addition, CBZ-aptamer binding releases MB, disrupts the energy-transfer pathway and switches the 542 nm signal “on” while turning the 700 nm signal “off”. This anti-directional dual-channel read-out efficiently eliminates background fluorescence and external interference, enhancing sensitivity. The sensor exhibits a wide linear range of 2.27–500 ng mL−1, a detection limit of 0.682 ng mL−1, and quantitative recovery (90.1–107.4%). The proposed strategy offers superior selectivity, sensitivity, and accuracy. The developed sensor was validated by analyzing a certified reference materials and comparing with an HPLC method and successfully applied to the determination of CBZ in real food samples (cherry tomatoes, oranges, lettuce, green peppers, strawberries, cucumbers, apples, and Chinese chives).

The Effects of Carbendazim on Acute Toxicity, Development, and Reproduction in Caenorhabditis elegans

Jie Li,Xinghua Zhou,Caiqin Zhang,Yansheng Zhao,Ying Zhu,Jiayan Zhang,Juan Bai,Xiang Xiao

Carbendazim, as a fungicide, was commonly used to control fungal diseases in agriculture, forestry, and veterinary medicines. In this study, the acute and reproductive toxicity of carbendazim was assessed using Caenorhabditis elegans (C. elegans) as a model in order to preliminarily evaluate the potential risks of this fungicide in agricultural production and application. The results showed that the growth of C. elegans was inhibited by 0.01 μg/L carbendazim. The treatment of 0.1 μg/L carbendazim caused a significant decrease in locomotion behavior and significant damage to the reproductive and antioxidant system, causing the lifespan of nematodes to be drastically shortened. These results provide a better understanding of the environmental risk of carbendazim and raise new concerns about safety.

N-methyl-2-pyrrolidone exfoliated graphene as highly sensitive analytical platform for carbendazim

Piaopiao Wei,Tian Gan,Kangbing Wu

Graphene nanosheets (GS) with approximately 10–15 layers was prepared via ultrasonic exfoliation of graphite powder in N -methyl-2-pyrrolidone (NMP), and confirmed using transmission electron microscopy (TEM) and atomic force microscopy (AFM). X–ray photoelectron spectra (XPS) and Raman spectra were employed to get further insights into the structural characteristics of exfoliated GS. On the surface of the GS modified glassy carbon electrode (GS/GCE), oxidation signals corresponding to carbendazim, a widely used pesticide, were greatly improved due to the higher accumulation ability, as determined by chronocoulometry. The oxidation mechanism of carbendazim was studied using a rotating disk electrode, and two electrons were transferred. Depending on the signal amplification of GS, a highly sensitive analytical platform for carbendazim was constructed. The linear range was from 1.0 to 300.0 μg L −1 (0.005–1.57 μM), and the detection limit was 0.15 μg L −1 (0.78 nM). Finally, this newly developed sensor was used to detect carbendazim in ground water, soil, and cucumber samples with satisfactory accuracy.

Development of an immunoassay for carbendazim based on a class-selective monoclonal antibody

Huijuan Yan,Liqiang Liu,Naifeng Xu,Hua Kuang,Chuanlai Xu

In the present study, a class-selective monoclonal antibody (mAb) for carbamate fungicides was obtained using two haptens. One hapten was designed as immunogen and the other one for coating. Under the optimum assay conditions of coating antigens H2-ovalbumin, in the assay buffer of sodium chloride 1.6%, no methanol, and pH 6.5, the assay system was more sensitive. The linear range was from 0.06 to 7.47 ng/mL. The cross-reactivity test was carried out with IC50 values of 0.45 ng/mL for carbendazim, 0.57 ng/mL for benomyl, and 9.70 ng/mL for thiabendazole. Other related compounds thiophanate methyl, fenbendazole, mebendazole, methyl paraoxon, methyl parathion, chlorpyrifos, and chlorothalonil did not cross-react significantly in this assay. To evaluate the proposed immunoassay, real samples such as apples, tomatoes, and cucumbers were subjected to recovery test with carbendazim. The recovery and coefficient variation range from 73% to 125% and 0.36% to 11.80%, respectively. The developed method was also compared with the commercial kit for assaying carbendazim in the fruits and vegetables and the results were consistent. All results indicated that this developed enzyme-linked immunosorbent assay (ELISA) is useful for the detection of carbendazim residues in fruits and vegetables.

Detection of carbendazim in oranges with metal grating integrated microfluidic sensor in terahertz

Yanqun Tong,Liuxin Ding,Kuiguo Han,Xiaobo Zou,Shiyan Wang,Zhang Wen,Yunxia Ye,Xudong Ren

A novel microfluidic metal grating integrated terahertz sensor has been designed, which is composed of a metal microstructure array-dielectric layer-metal layer, where the dielectric layer is the microfluidic channel carrying the analyte. By adjusting the structural parameters of the metal grating sensor, a highly confined electromagnetic field can be obtained in the microfluidic channel, thereby significantly enhancing the interaction between the analyte and the terahertz wave and improving the terahertz detection sensitivity. The metal grating described in this paper is composed of an array of square holes, which is manufactured using laser micromachining technology, so that the measurement method is simplified and improved. The results show that the addition of different concentrations of carbendazim solution resulted in a redshift of the overall spectrum, with the highest sensitivity reaching 8.773 GHz/mg L−1, which is about eight times more sensitive than the traditional terahertz transmission sensor. The relative error of using this method to determine carbendazim levels in orange juice samples was less than 5.3%. The terahertz time-domain spectroscopy technology combined with the metal grating integrated microfluidic sensor can improve the sensitivity of sample detection and realize the rapid detection and analysis of trace elements.

Rapid detection of carbendazim residue in tea by machine learning assisted electrochemical sensor

Tang Man,Guo Junying,Shen Zhen

The presence of pesticide residues in agricultural products, such as tea, poses significant health risks to consumers and has become a major concern for the food industry. Carbendazim, a widely used fungicide, has been identified as one of the harmful contaminants in tea. Rapid and accurate detection of carbendazim residues is therefore essential for ensuring food safety and quality. In this study, we propose a novel machine learning-assisted electrochemical sensor based on a CuO/reduced graphene oxide (rGO)/poly N-phenylglycine (NPAN) composite electrode for the rapid detection of carbendazim residues in tea. The CuO/rGO/NPAN electrode offers enhanced selectivity and sensitivity, enabling reliable detection of carbendazim even in the presence of other pesticides. By systematically evaluating various machine learning models and preprocessing techniques, we optimize the sensor’s performance and achieve accurate classification and identification of carbendazim residues. Our research contributes to the field of food safety assessment by introducing a novel electrode design and demonstrating the effectiveness of machine learning-assisted electrochemical sensors for the detection of carbendazim residues in tea.

A gold/Fe3O4 nanocomposite for use in a surface plasmon resonance immunosensor for carbendazim

Li Qian,Dou Xiaowen,Zhao Xiangsheng,Zhang Lei,Luo Jiaoyang,Xing Xiaoyan,Yang Meihua

A surface plasmon resonance (SPR) biosensor for the pesticide carbendazim is described that has enhanced performance due to the use of a Au/Fe 3 O 4 nanocomposite as an amplifying label on the surface the carboxymethyldextran-coated gold layer of the sensor. The surface was further modified with monoclonal antibody to obtain a sensor for real-time detection of carbendazim. Binding of carbendazim results in a change in refractive index. SPR detection in the absence of Au/Fe 3 O 4 nanocomposite and by UPLC-MS analysis demonstrated the improved performance to be due to the use of the Au/Fe 3 O 4 nanocomposite. Response is linear in the 0.05 to 150 ng·mL −1 carbendazim concentration range, and the limit of detection is 0.44 ng·mL −1 . This is more than 1 order of magnitude lower than that of the conventional SPR assay. The recoveries from spiked medlar are between 102.4 and 115.0%. The selectivity was tested by using the pesticides benzimidazole, 2-mercaptobenzimidazole, 2-benzimidazole propionic acid, and 2-(2-aminoethyl) benzimidazole as potential interferents. Conceivably, this Au/Fe 3 O 4 nanocomposite-based method has a large potential for the detection of other small analytes at trace concentrations. Schematic presentation of the SPR sensor for the detection of the fungicide carbendazim (methyl 2-benzimidazole carbamate; MBC) based on the use of a gold/Fe 3 O 4 nanocomposites.

Fe-Doped ZnMOF-Derived Heterocarbon Nanotubes for Electrochemical Sensing of Carbendazim Residues in Food

Yicheng Yao,Liudi Ji,Peng Hu,Xiaoming Zhu,Junxing Hao,Kangbing Wu

Background Carbendazim (CBZ), is a benzimidazole fungicide and widely used for controlling fruit and vegetable diseases, which poses significant risks to human health due to its neurotoxicity, endocrine disruption, and reproductive toxicity. Sensitive and rapid monitoring of carbendazim residues in food is essential for ensuring food safety. Among the numerous developed methods, electrochemical sensing stands out due to its simplicity of operation, low cost, and capability for on-site detection. Its key point is to construct a high-performance sensing interface. Nevertheless, these interfaces still face some limitations, such as inadequate conductivity, relatively low sensitivity, and narrow linear range. Results Herein, graphene supported iron nanoparticles encapsulated nitrogen-doped carbon nanotubes (Fe@N-CNTs) were constructed. The MOF-derived “tube-bridge” CNTs architecture, enhanced by N-doping, ensures high conductivity and optimized electron transfer pathways. Encapsulated iron nanoparticles act as shielded yet accessible redox-active centers, enhancing catalytic specificity. Graphene prevents CNTs stacking, maximizing active site exposure and interfacial charge transfer. This synergistic design significantly boosts the electrochemical active surface area, electron transfer kinetics, and electrocatalytic activity. As a result, a highly sensitive electrochemical sensing platform for CBZ has been successfully constructed. An irreversible two-electron and two-proton process takes place on the surface of Fe@N-CNTs/Graphene/GCE. The composite demonstrated exceptional performance for CBZ detection, achieving a broad linear range (0.001-7.0 μM), and an ultralow detection limit (0.56 nM). Furthermore, the sensor exhibited superior repeatability, stability, and selectivity. Its practical applicability was successfully validated through the accurate quantification of CBZ residues in medicinal herbs and vegetables ( recoveries of 98.58%-101.7%). Significance This work offering a robust solution for monitoring ultra-low concentrations CBZ residues in complex food matrices. This innovative technology provides essential support for combating pesticide misuse while safeguarding public health and preserving ecological integrity related to CBZ contamination.

Highly sensitive quantitative detection of carbendazim residue based on terahertz metamaterial enhancement and chemometrics

Jun Hu,Xiaodong Mao,Zhikai Huang,Shimin Yang

Objective Carbendazim is widely used as an effective fungicide in agriculture, but its residues on crops pose potential health risks to consumers. This study aims to develop a rapid, non-destructive, and highly sensitive method for detecting carbendazim residues. Methods Based on electromagnetic theory, this paper presented a terahertz metamaterial sensor incorporating a “cross” compound double-peak structure. Terahertz transmission spectra were collected from 21 different concentration gradients of carbendazim solutions. The spectral response showed a clear decreasing trend in transmission peak amplitude with increasing concentrations. By comparing the results of data preprocessing and feature extraction, the optimal model of terahertz metamaterial detection of carbendazim residue was established. Result The related coefficient of prediction set (R P ) and root mean square error of prediction set (RMSEP) of this model are 0.9825 and 0.2001, and the Limit of Detection (LOD) is 0.672 μg/mL. Conclusion The results demonstrate the feasibility of using terahertz metamaterial sensors combined with spectral analysis for high-sensitivity, non-destructive detection of carbendazim, offering a promising approach for food safety monitoring.
