It is urgently needed to develop novel adjuvants for improving the safety and efficacy of vaccines. Metal-organic frameworks (MOFs), with high surface area, play an important role in drug delivery. With perfect biocompatibility and green preparation process, the γ-cyclodextrin metal-organic framework (γ-CD-MOF) fabricated with cyclodextrin and potassium suitable for antigen delivery. In this study, we modified γ-CD-MOF with span-85 to fabricate the SP-γ-CD-MOF as animal vaccine adjuvants. The ovalbumin (OVA) as the model antigen was encapsulated into particles to investigate the immune response. SP-γ-CD-MOF displayed excellent biocompatibility in vitro and in vivo. After immunization, SP-γ-CD-MOF loaded with OVA could induce high antigen-specific IgG titers and cytokine secretion. Meanwhile, SP-γ-CD-MOF also significantly improved the proliferation of spleen cells and activated and matured the bone marrow dendritic cells (BMDCs). The study showed the potential of SP-γ-CD-MOF in vaccine adjuvants and provided a novel idea for the development of vaccine adjuvants.

Molecular imaging of disease with multifunctional nanoparticles has improved specificity and sensitivity but also raises the complexity, potential toxicity, and cost. Here, we show a facile and degradable self-assembly β-cyclodextrin metal-organic framework (β-CD-MOF) nanoplatform for customizable multifunctional imaging. These β-CD-MOF nanoparticles were obtained with favorable morphology and size by controlling the degradation time. The β-CD-MOF were used as nanoplatforms for facile functionalization with adamantane (Ad)-modified probes through host-guest interactions between the surface β-CD units and Ad molecules. We demonstrated the method's feasibility and capability by developing various contrast agents for multiple biomedical imaging, including fluorescence imaging, magnetic resonance imaging (MRI), and computed tomography (CT) imaging. The nanoprobes showed superior performance compared to the corresponding small molecular probes, including better physio-chemical properties ( e.g. , about 5 times of T 1 relaxivity for MRI, 1.2 times of Hounsfield units for CT), improved pharmacokinetics, effective tissue imaging capability, and low safety concerns. These β-CD-MOF-based nanoparticles are promising host-guest nanoplatforms for developing multifunctional and safe imaging probes. Statement of Significance Molecular imaging of disease with multifunctional nanoparticles has improved specificity and sensitivity but also raises the complexity, potential toxicity, and cost. Here, we introduce facile and degradable self-assembly β-cyclodextrin metal-organic framework (β-CD-MOF) nanoplatforms for customizable multifunctional imaging. The significance of this work includes: 1) This work reports the tailoring of MOFs nanoparticles with suitable sizes and shapes for biomedical applications through controllable morphological transition and degradation; 2) The β-CD-MOF-based host-guest nanoplatforms are facile and feasible for developing multifunctional nanoparticular contrast agents for effective tissue imaging; 3) The nanoparticular contrast agents show low safety concerns with a long-term tissue deposition similar to the small molecular probes.

Due to the complexity of wastewater pollutants and their adverse effects on human health and the environment, there is an urgent need for the development of a recyclable adsorbent capable of removing both dyes and heavy metals. In this study, citric acid modification β-cyclodextrin metal-organic framework (CA-β-CD-MOF) was synthesized by chemical grafting followed by vapor diffusion methods. The concentration, dosage, and pH of citric acid had a certain impact on the adsorption effect of the adsorbent. The adsorption capacities for the anionic dye Congo red (CR) and copper ions (Cu 2+ ) were as high as 900.9 and 287.4 mg·g −1 , respectively. The adsorption process could be well described by the Langmuir isothermal model. The adsorption kinetics of CR and Cu 2+ conformed to the pseudo-second-order kinetic model. The adsorption process was a heat absorption and spontaneous process. The combination of density functional theory (DFT) calculations and experimental results proves that electrostatic interaction is the adsorption mechanism of CR. The adsorption of Cu 2+ was attributed to the complexation between the carboxyl group on citric acid (CA) and Cu 2+ . The CA-β-CD-MOF we prepared has good adsorption performance and recyclability and has great potential in removing Congo red and copper ions from wastewater.

A moisture-resistant and green cyclodextrin metal–organic framework (CD-MOF) nanosheet has been prepared via an one-pot antisolvent synthesis procedure. After the treatment of in situ chemical cross-linkage, the two-dimensional (2D) cross-linked CD-MOF exhibited both peroxidase (POD) and oxidase (OXD) enzymatic activities, as well as hydrolytic stability. On the basis of its POD mimics function, the proof-of-concept biosensors were constructed to realize the colorimetric detection for H 2 O 2 and glucose, respectively. In vitro cytotoxicity experiments showed that the 2D cross-linked CD-MOF nanozymes still maintained excellent biocompatibility even at a concentration reaching up to several mg/mL. The in situ colorimetric detection of H 2 O 2 secreted by HepG2 cells further confirmed its promising biocompatibility, showing its great promises as label-free colorimetric probe in early cancer detection and pathological process monitoring. Metal organic frameworks (MOFs), as a novel separation medium s , have in recent years attracted wide consideration in capillary electrochromatography (CEC). However, the instability of frameworks limited the application of the MOF-based chiral separation capillary. Herein, Iron-based γ-cyclodextrin metal-organic framework (Fe-CD-MOF) with mesoporous was developed as the chiral stationary phase in open tubular capillary electrochromatography column (OT-CEC) for the stereoisomer separation of fourteen chiral drugs and one chiral alcohol. The capillary column with MOF coating exhibited the best performance (resolution value: 17.07) for anisodamine in any MOF-based capillary column that has never been reported. Moreover, the effect of pH, buffer concentration, and methanol content has been investigated. The novel coated capillary was also applied for the quantitative analysis of a real sample. The Fe-CD-MOF coated capillary showed outstanding repeatability and stability, with the satisfactory relative standard deviations (RSDs) for intra-day, inter-day, and column-to-column. Finally, the enantioseparation mechanism of the Fe-CD-MOF coated capillary was evaluated by adsorption kinetic experiments. In summary, this work indicated the novel and renewable Fe-CD-MOF was a potential chiral stationary phase in CEC chiral separation.
