Microcystin-LR (MC-LR), a toxic cyanotoxin, poses severe risks to environmental and human health with its efficient detection urgently required for water safety monitoring. Here, we reported a photoelectrochemical (PEC)-surface-enhanced Raman scattering (SERS) dual-mode aptasensor based on WS 2 /copper(II) meso-tetra(4-carboxyphenyl) porphyrin (WS₂/Cu-TCPP) MOFs Z-scheme heterojunction with a single laser source to produce photocurrent and SERS signals. To access interference between PEC and SERS signals, their correlation was systematically examined by applying a certain bias voltage. This bias voltage drives photogenerated electrons into external circuit, thereby enhancing carrier separation efficiency and suppressing the return of Raman photons to the ground state. In this way, a complementary “PEC-enhancement/SERS-attenuation” signal pattern can be achieved to allow better reliability for biosensing. The presence of ascorbic acid (AA) replicated an identical response to confirm competitive coupling between PEC and SERS derived from WS₂/Cu-TCPP heterojunction. Based on this signal transduction mechanism and localized generated AA by magnetic separation, a dual-mode aptasensor was developed for MC-LR detection with superior sensitivity and interference resistance. It allowed a linear range of 0.01–50 ng mL −1 and a detection limit of 0.14 pg mL −1 for MC-LR as well as high selectivity and stability for real water samples analysis. This work should offer an efficient paradigm for precise analysis of hazardous pollutants in complex matrices.

The high toxicity and bioaccumulation of heavy metal ion pollutants pose severe threats to the environment and public health. Tilted fiber Bragg grating-based surface plasmon resonance (TFBG-SPR) sensors have emerged as effective tools for heavy metal ion sensing owing to their portability and rapid response. These sensors typically feature a single plasmonic structure. And they are constrained by hardware restrictions and complex spectral analyses. Thus, these conventional designs render multicomponent detection and precise analysis challenging. To address these limitations, this study introduces a lightweight multichannel sensing platform based on a composite plasmonic TFBG structure. A two-dimensional metal–organic framework (2D MOF Cu-TCPP(Pt)) and silver nanoparticles (AgNPs) are integrated onto a gold (Au)-coated TFBG surface. This configuration enables strong coupling between the SPR and localized SPR (LSPR). Hence, the signal sensitivity is enhanced. The molecular sieve effect of the Cu-TCPP(Pt) improves the target analyte enrichment and suppresses the background interference, thereby ensuring a stable signal output. Additionally, a DNA aptamer conformational switching strategy ensures selective target analyte recognition. This platform integrates an AgNPs@Cu-TCPP(Pt)/Au/TFBG sensor, optical channel array, and real-time signal processing module and enables simultaneous detection of Pb 2+ , Cd 2+ , and Hg 2+ within 6 min, with detection limits of 0.007, 0.012, and 0.005 nM, respectively. The proposed sensor effectively mitigates background interference in complex matrices, demonstrating excellent linearity and recovery. Compared with conventional TFBG-based methods, this platform offers high sensitivity, short detection times, and enhanced robustness, and presents a novel solution for real-time multicomponent detection in complex matrices.

The optical fiber sensor, which exploits surface plasmon resonance (SPR), finds extensive application in biological detection owing to its miniaturized and real-time measurement capabilities. Nevertheless, SPR sensors alone exhibit limited sensitivity when detecting small biological molecules. This paper presents a novel gold nanoparticle (AuNPs)/two-dimensional metal–organic framework (2D MOF Cu-TCPP)/Au/D-shaped fiber SPR sensor for dopamine detection. The composite structure of AuNPs/2D MOF/Au was designed to facilitate the coupling of SPR and local surface plasmon resonance (LSPR), resulting in a substantial enhancement of the intensity of the surface electric field. A 104.61 % increase in sensitivity and a 73.87 % increase in figure of merit (FOM) were achieved by optimizing the optimal deposition concentration of AuNPs/2D MOF . The finite element method was utilized in COMSOL software to demonstrate that the coupling of SPR and LSPR can significantly enhance the sensitivity value (10,000 nm/RIU). Additionally, the sensitization mechanism of the sensor is elucidated by the energy band theory. Moreover, probe DNA was utilized as a linker to capture dopamine molecules, achieving a low limit of detection (LOD) of 1.07 ± 0.07*10 −14 M. These results demonstrated that the proposed sensor enables accurate and highly sensitive detection of dopamine molecules.

Background The detection and quantification of glucose in human biofluids are critical for the early diagnosis and management of metabolic disorders such as hypoglycemia and diabetes. Non-invasive wearable sensors for sweat glucose monitoring have emerged as promising alternatives to traditional blood glucose testing due to their ease of use, rapid response, and portability. However, the development of stable, cost-effective, and enzyme-free wearable platforms capable of reliable sweat glucose detection remains a significant challenge. Current enzyme-based sensors often suffer from poor stability under ambient conditions, complex fabrication processes, and high costs, limiting their widespread applicability in resource-constrained settings. Results Au NPs/Cu-TCPP(Fe) nanozymes exhibiting dual peroxidase-like and glucose oxidase-like catalytic activities were synthesized and characterized. These nanozymes catalyze a cascade reaction: glucose oxidation generates H 2 O 2 , which is then decomposed to produce hydroxyl radicals that oxidize 3,3′,5,5′-tetramethylbenzidine (TMB), yielding a blue-colored product for colorimetric detection. A flexible, multi-layer hydrophilic-hydrophobic array patch was fabricated by integrating the nanozymes and TMB into a paper-based matrix. This design enables efficient in situ sweat collection, concentration, and colorimetric analysis. The patch exhibits a linear detection range of 20–200 μM with a detection limit of 12.72 μM, suitable for physiological sweat glucose levels. Selectivity tests confirmed negligible interference from common sweat components. A smartphone application was developed to enable real-time, quantitative analysis of colorimetric signals. Clinical validation on multiple volunteers across different skin locations (chest, back, forearm, forehead) showed strong correlation with reference methods (HPLC-MS, commercial glucose detection kit), with relative errors <10 %. Significance This study presents a low-cost and enzyme-free sweat glucose detection platform based on Au NPs/Cu-TCPP(Fe) nanozymes. The integration of smartphone technology enables point-of-care testing, making the platform particularly suitable for personalized health monitoring. The successful validation in human subjects demonstrates its potential for real-world applications in diabetes management and early disease detection. This work bridges the gap between nanozyme catalysis and wearable sensor technology, providing a versatile platform for non-invasive biomarker detection.

Controlling the morphology of the metal–organic framework (MOF) for nanosheets is beneficial for understanding their crystal growth kinetics and useful for extending these MOF nanosheets to advanced applications, in particular for gas separation and device integration. However, synthesizing MOF nanosheets with uniform thickness or desirable size still remains challenging. Herein, we provide a crystal dissolution–growth strategy for fabricating dispersible porphyrin MOF nanosheets with lateral dimensions and nanometer thickness. A morphological transition (bulk crystals–nanosheets–bulk crystals) in Zn-TCPP was observed when controlling the crystal growth kinetics by adjusting the reaction parameters (temperature and acidity). These findings encouraged the synthesis of other types of nanosheets (Cu-TCPP, Zn-TCPP (Pd), and Cu-BDC nanosheets). Zn-TCPP (Pd) nanosheets were applied in field-effect transistors and exhibited photoresponse characteristics. This work demonstrates a new strategy for obtaining MOF nanosheets and casts a new light upon fabricating two-dimensional inorganic–organic hybrid materials with controlled thickness.

A self-cleaning electrochemical biosensor based on two-dimensional Cu-porphyrin (Cu-TCPP) metal-organic framework nanofilms , novel super G-quadruplex (G4), and DNA nanomotors was developed for the cyclic detection of Pb 2+ ions. The Cu-TCPP framework with inherent peroxidase activity can create an ultra-thin nanofilm that functioned as a carrier to support the metastable G4 comprising four individual DNA strands. The introduction of Pb 2+ and the intercalation of hemin can help it to form stable G4-hemin DNAzymes, which exhibits strong catalytic H 2 O 2 reduction activity, and its number will be directly related to the amount of the introduced Pb 2+ . Moreover, a DNA nanomotor system is introduced to achieve cyclic detection, and the addition of the fuel DNA strands enables G4 to perform a “complete-dissociation-complete” process for achieving self-cleaning of the electrode interface and the cycle detection of Pb 2+ . The synergistic effects of Cu-TCPP and G4-hemin DNAzymes, which exhibits efficient and catalytic H 2 O 2 reduction, enhance the performance of the electrochemical sensing system. The linear range of this sensor to Pb 2+ is 5 nM–5 μM, and the detection limit is 1.7 nM. Compared with the best system in reported studies, its linear range is five times wider and its detection limit is lower than the previously lowest one. Taking advantage of the Pb 2+ stabilized G4, the proposed sensor can selectively detect Pb 2+ in the presence of other metal ions . The results presented herein comprise a valuable reference for constructing DNA nanoelectronic devices and establish sensitive and cyclic detection of the target and preparing of self-cleaning electrode interfaces.

Implant-associated infections (IAIs) are the main cause of prosthetic implant failure. Bacterial biofilms prevent antibiotic penetration, and the unique metabolic conditions in hypoxic biofilm microenvironment may limit the efficacy of conventional antibiotic treatment. Escaping survival bacteria may not be continually eradicated, resulting in the recurrence of IAIs. Herein, a sonosensitive metal-organic framework of Cu-TCPP (tetrakis(4-carboxyphenyl) porphyrin) nanosheets and tinidazole doped probiotic-derived membrane vesicles (OMVs) with high-penetration sonodynamic therapy (SDT), bacterial metabolic state interference, and bacterial cuproptosis-like death to eradicate IAIs is proposed. The Cu-TCPP can convert O 2 to toxic 1 O 2 through SDT in the normoxic conditions, enhancing the hypoxic microenvironment and activating the antibacterial activity of tinidazole. The released Cu(II) under ultrasound can be converted to Cu(I) by exogenous poly(tannic acid) (pTA) and endogenous glutathione. The disruption of the bacterial membrane by SDT can enhance the Cu(I) transporter activity. Transcriptomics indicate that the SDT-enhanced Cu(I) overload and hypoxia-activated therapy hinder the tricarboxylic acid cycle (TCA), leading to bacterial cuproptosis-like death. Moreover, the OMVs-activated therapy can polarize macrophages to a M2-like phenotype and facilitate bone repair. The sonodynamic biofilm microenvironment modulation strategy, whereby the hypoxia-enhanced microenvironment is potentiated to synergize SDT with OMVs-activated therapy, provides an effective strategy for antibacterial and osteogenesis performance.

Acrylamide (AA) is a carcinogen mainly ingested through food and drinking water, making its accurate determination crucial for both food safety and environmental protection. Herein, we proposed a derivatization-based ratiometric surface-enhanced Raman scattering (SERS) method for the quantification of AA. High density Au NPs were anchored to the surface of Cu-TCPP MOF nanosheets (MOFNs) to form the SERS sensor. The abundant Raman “hot spots” at the nanogaps generated by the Au NPs and the internal standard (IS) signal provided by Cu-TCPP MOFNs improved the sensitivity and quantitative accuracy of the method. Following the thiol-ene click derivatization reaction between p-aminothiophenol (PATP) and AA, the Raman peak intensity ratio (I 1080 /I 395 ) was employed to quantify AA. The linear range was 0.1 nM to 10 μM, and the limit of detection (LOD) was as low as 0.08 nM. Trace amounts of AA in food and water samples were successfully determined using this method.

A multiplex electrochemical immunosensor was developed for the simultaneous detection of two lung cancer markers, carcinoembryonic antigen (CEA) and carbohydrate antigen 125 (CA125), using electroactive probes. Cu-TCPP nanomaterials were synthesized by a hydrothermal method . Cu-TCPP–TB-anti-CEA and Cu-TCPP-PB-anti-CA125 immunosensor probes were prepared using toluidine blue (TB) and prussian blue (PB) as the indicators. The electrochemical immunosensor was constructed for the simultaneous detection of CEA and CA125. The linear range of CEA was 0.1–160 ng mL −1 with a low detection limit of 0.03 ng mL −1 . The linear range of CA125 was 0.5–200 U mL −1 with a low detection limit of 0.05 U mL −1 . Furthermore, the immunosensor was applied for the simultaneous determination of CEA and CA125 in real samples (human blood serum), with results comparable to the reference values obtained by the standard enzyme-linked immunosorbent assay. This study furthers knowledge on the construction of electrochemical immunosensors showing the simultaneous detection of two lung cancer markers.

Ferroptosis is a recently discovered route of regulated cell death that offers the opportunities for the treatment of chemotherapy-resistant tumor indications, but its efficacy can be affected by the glutathione peroxidase 4 (GPX4) and ferroptosis suppressor protein 1 (FSP1) antioxidant mechanisms, posing significant challenges for its clinical translation. In this study, we report a Cu-tetra(4-carboxyphenyl)porphyrin chloride(Fe(III)) (Cu-TCPP(Fe)) metal organic framework (MOF)-based nanosystem for the efficient incorporation of Au nanoparticles (NPs) and RSL3, which can demonstrate enzyme-like activities to universally suppress the antiferroptotic pathways in tumor cells for amplifying ferroptotic damage. Herein, Cu-TCPP(Fe) MOF nanosheets were integrated with Au NPs via in situ nucleation and loaded with RSL3 via π–π stacking, which were eventually modified with polyethylene glycol (PEG) and iRGD for tumor-targeted drug delivery. Specifically, the Au NPs can demonstrate glucose oxidase-like activities for efficient glucose depletion, thus disrupting the pentose phosphate pathway to impede reduced glutathione (GSH) biosynthesis and prevent the recycling of coenzyme Q10 (CoQ10) to CoQ10H2, while Cu species can oxidize GSH into oxidized glutathione (GSSG). These nanocatalytic activities can lead to the simultaneous inhibition of the GPX4/GSH and FSP1/CoQ10H2 pathways and cooperate with the GPX4-deactivating function of RSL3 to cause pronounced ferroptotic damage, thereby providing a strong rationale for the application of ferroptosis therapy in the clinic.
